Abstract
Description
本願は、2016年5月2日に提出された米国特許仮出願第62/330,561号の優先権を主張するものであり、その全内容を参照により本明細書に組み込む。
Claims (21)
- 三次元形状を形成するように第一の材料から構築され、第一の生分解速度を持つ骨格、および
第二の材料から構築され、第二の生分解速度を持ち、前記第一の生分解速度がこの第二の生分解速度よりも小さく、前記骨格により形成された三次元形状を埋める母材を含む、骨組織工学に用いられる足場材料。 - 前記骨格と前記母材はユニット構造であり、前記母材の孔隙率は前記骨格の孔隙率よりも大きい、請求項1に記載の足場材料。
- 前記母材はさらに骨形成タンパク質を含む、請求項1または2に記載の足場材料。
- 前記第一の材料と前記第二の材料は異なる材料である、請求項1〜3のいずれか一項に記載の足場材料。
- 前記骨格は第一の孔隙率を持ち、前記母材は第二の孔隙率を持ち、前記第二の孔隙率は前記第一の孔隙率よりも大きい、請求項1〜4のいずれか一項に記載の足場材料。
- 前記骨格は、第一の平均径を持つ孔を含み、前記母材は第二の平均径を持つ孔を含み、前記第二の平均径は前記第一の平均径よりも大きい、請求項5に記載の足場材料。
- 前記骨格は複数の第一の支持体を含み、前記第一の支持体のそれぞれは他の第一の支持体に対して平行に配列されている請求項1〜6のいずれか一項に記載の足場材料。
- 前記骨格は複数の第二の支持体をさらに含み、前記第二の支持体のそれぞれは他の第二の支持体に対して平行に配列され、前記第一の支持体の角度とは異なる角度で配向されている、請求項7に記載の足場材料。
- 前記骨格は複数の第三の支持体をさらに含み、前記第三の支持体のそれぞれは他の第三の支持体に対して平行に配列され、前記第一の支持体の角度と第二の支持体の角度とは異なる角度で配向されている、請求項8に記載の足場材料。
- 前記第一の支持体と前記第二の支持体との角度は120°であり、前記第一の支持体と前記第三の支持体との角度は120°である、請求項9に記載の足場材料。
- 前記骨格は、前記母材と前記骨格との結合を促進するための係合構造をさらに含む、請求項1に記載の足場材料。
- 骨組織工学に用いられる足場材料を調製する方法であって、
患者の治療領域の3Dモデルを取得すること、
前記患者と前記治療領域に基づき前記足場材料の所望の分解性を決定すること、
前記所望の分解性を達成するように少なくとも1つの骨格特徴と少なくとも1つの母材特徴を選択すること、
前記治療領域に合うように骨格をデジタル設計すること、
前記少なくとも1つの骨格特徴を持つ前記骨格を製造すること、および
前記少なくとも1つの母材特徴を持つ母材を前記骨格に充填することを含む、方法。 - 前記少なくとも1つの骨格特徴は基材、材料処理技術、孔隙率、および孔径のうちの少なくとも1つを含み、前記少なくとも1つの母材特徴は基材、材料処理技術、孔隙率、および孔径のうちの少なくとも1つを含む、請求項12に記載の方法。
- 前記骨格を製造することは、前記骨格を3D印刷することを含む、請求項12または13に記載の方法。
- 前記骨格の3D印刷と同時に前記骨格と一体化するように前記母材を3D印刷することをさらに含む、請求項14に記載の方法。
- 前記母材は発泡体であり、前記母材を充填した前記骨格を冷凍乾燥して前記足場材料を形成することをさらに含む、請求項12〜15のいずれか一項に記載の方法。
- 前記母材の少なくとも1つの材料を選択すること、
前記母材の少なくとも1つの材料の溶液を調製すること、
前記溶液を撹拌して母材の発泡体を形成すること、
前記母材の発泡体を前記骨格内に成形すること、
前記母材の発泡体を充填した前記骨格を冷凍すること、および
前記骨格と前記母材を冷凍乾燥することをさらに含む、請求項12〜16のいずれか一項に記載の方法。 - 前記母材の少なくとも1つの材料は重合体またはタンパク質を含む、請求項17に記載の方法。
- 前記溶液を撹拌することは、100〜40000回転/分(RPM)の速度で1分間から60分間、前記溶液を撹拌することを含む、請求項17または18に記載の方法。
- 前記冷凍乾燥された骨格と前記母材を架橋剤で洗浄して、前記母材と前記骨格との架橋を促進すること、および
前記洗浄された母材と前記骨格を冷凍乾燥して、前記母材と前記骨格を架橋することをさらに含む、請求項16〜19のいずれか一項に記載の方法。 - 前記架橋剤は、グルタルアルデヒド、EDC/NHS、およびパラホルムアルデヒドのうちの少なくとも1つを含む、請求項20に記載の方法。
