本明細書に記載した融合タンパク質には、特に、介在性リンカーと一緒に融合されたIL2およびIL2Rβが含まれる。IL2についての配列は、当分野において公知である。典型的なヒトIL2前駆体配列は、配列番号34に示した。
アミノ酸1〜20は、シグナル配列であるので、所望であれば他のシグナル配列と置換することができる。ヒトIL2に対する典型的な核酸配列は、
当分野において公知のリンカー配列は、融合タンパク質の様々なドメイン間で使用できる;例えば、1つ、2つ、3つ、4つ、5つ以上のGGGS配列を使用できる。好ましい実施形態では、IL2とIL2RβのN末端との間のリンカーは、IL2Rαの細胞外ドメイン(EMETSQFPGEEKPQASPEGRPESETSC(配列番号28))を含む。タグ、例えばcMycタグ(EQKLISEEDL(配列番号29))は、さらに例えばIL2とリンカーとの間に追加することができる。IL2をコードする典型的な核酸配列は、GenBankにおいてアクセッション番号NM_000586.3で入手できる。
アミノ酸1〜26はシグナル配列であり、好ましくは本構築物中では欠失しており、例えばこの配列は、配列番号35のアミノ酸27〜551を含む。IL2Rβをコードする典型的な核酸配列は、GenBankにおいてアクセッション番号NM_000878.4(Var.1)、NM_001346222.1(Var.2);およびNM_001346223.1(Var.3)で入手できる。変異体1、2および3は、同一タンパク質をコードする。
インターロイキンIL4、IL7、IL9、IL15およびIL21は、IL2と同一ファミリーに属し、同一共通IL2Rgを使用する。それらは、それらの固有のα受容体に加えてIL2Rβを使用するIL2およびIL15を除いて、全部がそれらの固有のプライベート受容体を有する(図11)。可溶性IL2、IL4、IL7もしくはIL21がキメラNK92CIRBを発現するNK92細胞に加えられると、IL21だけがPC−3細胞に対する細胞傷害性を劇的に強化する。そこで、IL21Rの全細胞質ドメインをクローニングし、次にキメラCIRB内のIL2RβのC末端に頭−尾結合で加えた。これは、新規なIL2−IL2Rβ−IL21Rキメラ(図13に例示した、いわゆるCIRB21)を生じさせた。本明細書に示すように、唯一のリガンドおよびハイブリッド受容体を使用することによって、様々な受容体を介してNK細胞を活性化する複数のサイトカインから活性化シグナルを模倣することが可能であった。
好ましくは、これらの実施形態では、IL21R由来ドメインは、配列番号36のアミノ酸254〜538を含む。IL21Rをコードする典型的な核酸配列は、GenBankにおいてアクセッション番号NM_021798.3で入手できる。
NK細胞(およびその他)は、MHC−1分子の発現が形質転換細胞内(Algarra et al.,Hum Immunol 2000;61(1):65−73)およびウイルス感染中(Tortorella et al.,Annu Rev Immunol 2000;18:861−92)にダウンレギュレートされると、活性化される。しかし、腫瘍細胞による耐性表現型の獲得は、MHC−1からの阻害性シグナルの発現によって頻回に誘発される(Kochan et al.,Oncoimmunology 2013;2(11):e26491)。特にHLA−Gは、NK92媒介性腫瘍細胞溶解を阻害することが公知である(Lin et al.,Ann Oncol 2007;18(11):1804−9)。この問題に対する1つの可能性のある解決策は、これらの阻害性シグナルを相殺する複数の活性化シグナルの使用であろう。T細胞のために使用される最も効果的な共刺激性分子は、CD28および4−1BBである。CD28活性化は、腫瘍細胞上でのCD80およびCD86刺激性リガンド発現を必要とする。結果として、腫瘍内でのCD80発現は、それらの拒絶をもたらすことが証明されているが(Townsend et al.,Science 1993;259(5093):368−70)、これとは逆に、CD28−/−マウスでは、細胞性およびT細胞依存性免疫が完全に欠如する(Shahinian et al.,Science 1993;261(5121):609−12)。このため、低レベルのCD80は、数種の癌における腫瘍にとっての逃避機構であると考えられる(Tirapu et al.,Cancer Res 2006;66(4):2442−50;Hersey et al.,Int J Cancer 1994;58(4):527−32;Bernsen et al.,Br J Cancer 2003;88(3):424−31)。例えば、抗erbB2キメラ受容体内でのCD28活性化ドメインの使用は、MHC−1+リンパ腫のin vivoでの腫瘍進行の阻害を許容した(Pegram et al.,J Immunol 2008;181(5):3449−55)。CD28はNK92細胞によって発現するが(Gong et al.,Leukemia 1994;8(4):652−8)、その活性化は全ての癌によって媒介されない。
好ましくは、これらの実施形態では、CD28由来ドメインは、細胞内ドメイン、例えば配列番号38のアミノ酸180〜220、すなわち、RSKRSRLLHSDYMNMTPRRPGPTRKHYQPYAPPRDFAAYRS(配列番号39)を含む。CD28をコードする典型的な核酸配列は、GenBankにおいてアクセッション番号NM_006139.3で入手できる。
血液学的悪性腫瘍を有する患者は、一般に同種造血幹細胞移植片(AHSCT)から利益を得られる。この戦略では、ドナー免疫系は、移植片対腫瘍として公知の現象における治癒可能性を備えて患者の腫瘍細胞を攻撃するであろう。だが残念なことに、ドナー免疫細胞はさらに、直後またはその後の100日間のいずれかにレシピエント患者の健常組織を攻撃し、GVHDを誘発する可能性がある。これは、AHSCTの15%においては死亡および/または40〜60%において罹患率を引き起こすであろう。免疫抑制は、現在、GVHDを管理するための標準看護法である(Luznik and Fuchs,Immunol Res 2010;47(1−3):65−77;Storb et al.,Biol Blood Marrow Transplant 2010;16(1 Suppl):S18−27)。しかし、転写因子であるForkhead box P3(FOXP3)を発現するT−reg細胞(Roncador et al.,Eur J Immunol 2005;35(6):1681−91;Hall et al.,J Exp Med 1990;171(1):141−57)は、AHSCT中にGVHDを抑制もしくは軽減することが見いだされている(Beres et al.,J Immunol 2012;189(1):464−74;Brunstein et al.,Blood 2011;117(3):1061−70)。FOXP3発現の持続は、その第1イントロン内に位置する保存された非コーディング配列2(CNS2)内における11CpGモチーフの後成的脱メチル化によって維持される。この脱メチル化パターンはT−regの寿命にわたって持続し、DNAメチルトランスフェラーゼによる再メチル化からCNS2内のCpGモチーフを保護するために、IL2シグナル伝達によって活性化されたSTAT5によってCNS2に動員される(Nair et al.,Mol Cells 2016;39(12):888−97)Ten−Eleven−Translocation DNAジオキシゲナーゼによって保護される。同様に、T細胞活性化の重要なダウンレギュレーターであるCTLA−4は、T−regにおいてアップレギュレートされ、さらにIL2によって制御される(Wang et al.,Scand J Immunol 2001;54(5):453−8;Bell et al.,J Autoimmun 2015;56:66−80;Gasteiger et al.,Front Immunol 2012;3:179)。T−regは、それらの大規模なCD25発現(Dieckmann et al.,Exp Med 2001;193(11):1303−10)およびケモカイン受容体CCR7によってそれらがIL2源に達する能力(Smigiel et al.,J Exp Med 2014;211(1):121−36)に起因して、IL2に対して極度に応答性である。しかし、活性化T−regは、CD25発現を低下させ、それらのIL2シグナル伝達をICOSシグナル伝達経路にとって好都合に変化させることが証明されている。これは活性化されたが最終的には分化されなかったT−reg(Sharma et al.,Immunity 2010;33(6):942−54)から前炎症性T細胞エフェクターへの転位、またはIFN−γ−産生前炎症性Th1エフェクター細胞(Zhang et al.,J Immunol 2017;198(7):2612−25;Feng et al.,Gastroenterology 2011;140(7):2031−43;Takahashi et al.,J Exp Med 2011;208(10):2055−67)もしくはTh17さえへの発達を可能にするFOXP3発現の不安定性をもたらす(46)。手短には、T−regの長期活性化およびCNS2の脱メチル化ならびに増殖は、IL2およびCD28両方の共刺激を必要とする(Tang and Bluestone,Immunol Rev 2006;212:217−37;Chen et al.,J Immunol 2011;186(11):6329−37)。
本明細書に記載した組成物は、本明細書に記載したキメラをコードする核酸分子を含むことができる。発現ベクターを含む核酸分子は、例えば、本明細書に記載したNK細胞もしくはT−reg細胞内でキメラを発現させるために使用できる。
本方法には、安定性もしくは一過性のいずれかで本明細書に記載したキメラをNK細胞内、例えば、CD3−CD56+リンパ球内で発現させる工程が含まれる;Cheng et al.,Cellular & Molecular Immunology(2013)10,230−252を参照されたい。NK細胞は、一次細胞であってよく、例えば、被験者の末梢血由来であってよい、および体外で増殖させることができる、または培養NK細胞であってよい。
本明細書に記載した方法には、異常なアポトーシス性もしくは分化プロセスに関連する障害、例えば、細胞増殖障害もしくは細胞分化障害、例えば、固形腫瘍および血液学的癌の両方を含む癌を治療する方法が含まれる。一部の実施形態では、障害は、固形腫瘍、例えば、乳癌、前立腺癌、膵臓癌、脳腫瘍、肝臓癌、肺癌、腎臓癌、皮膚癌もしくは大腸癌である。一般に、本方法には、本明細書に記載したキメラを発現する治療有効量のNK細胞を、そのような治療を必要とする、または必要とすると決定されている被験者に投与する工程が含まれる。
本明細書に記載した方法には、異常な免疫応答に関連する障害、例えば、移植片対宿主病もしくはGVHDまたは自己免疫疾患を治療する方法が含まれる。本法は、それを必要とする被験者に本明細書に記載したIL2−IL2Rβ−CD28キメラを発現する制御性T細胞を投与する工程を含むことができる。
本方法は、好ましくは、本明細書に記載した治療有効量のNK細胞の静脈内注入による投与を含む。治療有効量は、経験的に、例えば、動物実験および臨床試験に基づいて決定することができる。一部の実施形態では、本方法は、投与1回当たり端点を含めて少なくとも細胞104個〜1×106個、5×106個、1×107個、5×107個、1×108個、3×108個、5×108個、1×109個もしくは5×109個、例えば細胞10億個〜30億個またはこれらの数の任意の2つの間の任意の範囲内の1回以上の注入を含む。細胞は、被験者に1回投与することができる、または複数回にわたり、例えば、1、2、3、4、5、6、7、8、9、10、11、12、13、14、15、16、17、18、19、20、21、22もしくは23時間毎に1回、または1、2、3、4、5、6もしくは7日間毎に1回、または治療期間中、1、2、3、4、5、6、7、8、9、10週間以上、または端点を含めてこれらの数の任意の2つの間の任意の範囲内に1回投与することができる。例えば、Ljunggren and Malmberg,Nat Rev Immunol.2007 May;7(5):329−39;Tonn et al.,J Hematother Stem Cell Res.2001 Aug;10(4):535−44;Klingemann,Cytotherapy.2005;7(1):16−22;Malmberg et al.,Cancer Immunol Immunother.2008 Oct;57(10):1541−52;Cheng et al.,Cellular & Molecular Immunology(2013)10,230−252を参照されたい。
一部の実施形態では、本明細書に記載したキメラを発現するNK細胞は、1種以上のチェックポイントブロック剤および/または抗腫瘍抗体を含む治療レジメンの一部として投与される。NK細胞は、チェックポイントブロック剤および/または抗腫瘍抗体と同時に、例えば、実質的に同時もしくは連続的に、例えばチェックポイントブロック剤および/または抗腫瘍抗体の投与の48、24、12、6、5、4、3、2もしくは1時間以内、または45、30、20もしくは15分間以内に投与することができる。
下記の実施例では、下記の材料および方法を使用した。
を使用して増幅させた。このプライマーは、リバースオリゴ
とともに使用した。増幅断片は、結果としてIL2−IL2Rαキメラを生じさせるIL2野生型を突然変異させるためのオリゴとして使用した。CIRB最終キメラ構築物を構築するために、IL2受容体αキメラを使用してC末端cMycタグ、その後にIL2Rαの細胞外ドメインのみ、次にフォワード5’−TGCAGGATCCACTCACAGTAACCTCAACTCC−3’(配列番号5)およびリバース5’−GGGAAGTGCCATTCACCGCGCAGGAAGTCTCACTCTCAGGA−3’(配列番号6)を使用するIL2RβのN末端断片によってIL2を増幅させた。これは後にIL2RβのN末端を導入する。生成物は、次に同一フォワードプライマーおよびリバース
を使用して再増幅させた。IL2中のXbaI部位は、最初にフォワードプライマー5’−CATCTTCAGTGCCTAGAAGAAGAACTC−3’(配列番号8)およびリバースプライマー5’−GAGTTCTTCTTCTAGGCACTGAAGATG−3’(配列番号9)を使用する突然変異により除去した。IL2Rβは、次に、フォワードプライマー
およびリバースプライマー
を使用して増幅させた。次に断片IL2−cMyc−IL2Rαは開放Xho−XbaIであったが、IL2Rβは、最終キメラCIRBを形成するためのSalI−XbaI断片として加えた。IL2およびCIRBはどちらもpcDNA4−TOからSpeI(鈍端)およびXhoIを使用して、BamHI(鈍端)およびXhoIにより消化されたCSCW−mcherryレンチウイルスベクターに移行させた。全構築物をシーケンシングし、レンチウイルス完全性について検証した。
標的乳癌BT474(細胞8×103個)は、96ウエルプレート内でプレーティングした。24時間後、細胞は、1μg/mLのトラスツズマブとともに室温で20分間インキュベートし、その後に2:1のE/T比でエフェクター細胞を添加した。3日間のインキュベーション後、癌細胞の生存率は、上記で説明したクリスタルバイオレット/アルコール抽出アッセイを使用して決定した。
IL2およびその受容体複合体の四次結晶構造(20)は、IL2のC末端およびIL2RβのN末端残基が41Åによって分離されることを示している。IL2とIL2RβのN末端との間のリンカーのために、本発明者らは、IL2Rαの細胞外ドメイン(EMETSQFPGEEKPQASPEGRPESETSC(配列番号28))を選択する。cMycタグ(EQKLISEEDL(配列番号29))は、IL2とリンカーとの間に添加した。完全成熟受容体IL2Rβタンパク質コーディング配列(シグナルペプチドを含まない)は、完全キメラCIRBを産生するためにリンカーの後に置いた(図1)。CIRBおよびIL2のどちらもmCherryを共発現するレンチウイルスベクター内でクローニングした。リンカーフォールドは、コンピューターを使用してヘリックス優勢構造であると予測された(図5A〜D)。リンカー柔軟性は、Karplus and Shultz法のコンピューターを使用する方法(47)を用いて評価し、全ペプチド結合で平均より優れた柔軟性(0〜2のスケール上で1以上)が示された。
46の同一MOIを備えるレンチウイルス媒介性安定性発現後、NK92IL2およびNK92CIRB細胞系は、IL2非依存性を獲得し、無限に増殖する。どちらの細胞系も6日間の期間中に類似の、およびまた別のIL2非依存性細胞系であるNK92−MIより高速の増殖を示し(図6A〜B)、親NK92細胞系およびNK92−MIと比較して、培養中での複数回の凍結およびプレーティングサイクルを受けた後に強固な生存率を示した。抗cMycモノクローナル抗体を使用して、本発明者らは、次に一過性でトレンスフェクトしたHEK293細胞(図2A)および安定性NK92CIRB細胞(図2B)の細胞表面でのCIRBの発現を試験した。本発明者らは、NK92CIRB細胞内ではcMycの表面発現の明確な証拠を見いだしたが、NK92IL2細胞内では見いだせなかった。CIRB発現は、さらに天然IL2Rβを認識する抗CD122モノクローナル抗体を使用してさらに確証され、ならびにキメラは、内因性CD122が、IL2RβおよびCIRBの両方を発現するNK92CIRB細胞系内より低いレベルであるが、NK92IL2細胞内で予想通りに存在することを示している。全長キメラCIRBの発現は、モノクローナル抗ヒトIL2を使用してウエスタンブロットによってさらに検出された。図2Dは、80kDaの予想サイズより高く、おそらくは翻訳後グリコシル化に起因すると思われる95kDaの全長サイズを示している。
本発明者らは、付着性の多発性骨髄腫細胞系U266GFPに対するNK92、NK92IL2およびNK92CIRB細胞の細胞傷害性を比較した。図3Aは、親NK92細胞系は、100IU/mLのIL2を用いて予備刺激されたにもかかわらず、NK92IL2もしくはNK92CIRBよりもはるかに低細胞傷害性であったことを示す。注目すべきことに、IL2は、4日間の共培養中に存在しなかった。NK92IL2およびNK92CIRB細胞は、U266GFPに向けての同等の細胞傷害性を示したが、これはin vitroでの匹敵するレベルの活性化を示唆していた。
トランスフォーミング成長因子TGFβ1は、TME内で過剰発現した免疫抑制剤であり、NK細胞内の数種の活性化シグナルを脱安定化させることによってNK細胞機能を阻害することが公知である(34)。グルココルチコイドデキサメタゾンは、CD4+T細胞からのIL2産生を抑制することによってリンパ球の機能を部分的に弱める(35)。IL−4は、NK細胞の増殖を阻害すると報告された(48)。本発明者らは、TGFβ1(10ng/mL)、IL4(10ng/mL)もしくはdex(1μM)の存在下で6日間にわたり細胞を培養することによってNK92IL2およびNK92CIRB細胞療法におけるこれらの免疫抑制剤の作用を試験した。図4Aは、NK92IL2細胞がdexへの曝露に耐えられなかったが、それらの増殖がTGFβ1によって強力に、およびある程度はIL4によって阻害されたことを示している。対照的に、NK92CIRB細胞の増殖はTGFβ1もしくはIL4による有意な影響を受けなかったが、dexによっては軽度にのみ阻害された(図4B)。
NK92細胞の最初の特性解析(43)によると、NK92細胞は、CD56+、CD3−、CD16−、CD25+、CD45+およびNKg2D−である(図5A)。これと比較して、NK92IL2細胞、NK92CIRB細胞および新しく単離したヒトNK(hNK)細胞は、様々なパターンのマーカー発現を提示する。NK92とは異なり、細胞系NK92IL2、NK92CIRBおよびhNK細胞は、全部がCD16+であり、発現レベルはhNK>NK92CIRB>NK92IL2であった(図5B)。本発明者らは、IL2非依存性NK92−MIについて試験し、以前の報告書にしたがうと(38)、CD16の発現を全く見いださなかった(図10A)。本発明者らは、グリコシル化もしくは非グリコシル化IL2を用いて処置されたNK92細胞内でのCD16発現を全く見いださなかった(図10B)。ヒトNK(hNK)細胞は、全NK92細胞系よりもはるかに高レベルのCD16およびNKG2Dを発現し、実質的にCD25陰性であるが(図5B)、NK92CIRB細胞は両方のIL2刺激NK92もしくはNK92IL2より低量のCD25を発現した。NK92細胞系中、CD25およびCD16に対するNK92CIRB発現はhNKと最も類似であった。
本発明者らは、次にHER2陽性乳癌細胞系BT474に対してトラスツズマブを使用してCD16発現がADCCに及ぼす影響を試験した。図5Cは、トラスツズマブの非存在下において2:1のE/T比を使用した場合に、BT474細胞がNK細胞の直接細胞傷害性による影響を受けなかったことを示している。しかし、1μg/mLのトラスツズマブの存在下では、NK92CIRBおよびNK92IL2はどちらもそれぞれ約60%および50%の実質的な細胞傷害性を与えた。1μg/mLでのトラスツズマブ単独は、BT474の生存率に有意には影響を及ぼさなかった。同様に、親NK92は、トラスツズマブの存在下でBT474に対する有意な細胞傷害性を全く誘発しなかった。
NK92CIRB細胞のqPCR分析は、IL2を用いて48時間にわたり刺激されたNK92IL2および親NK92と比較して、天然細胞傷害性受容体(NCR)であるNKP30(1.7倍)、NKP44(9倍)およびNKP46(1.4倍)の劇的な増加を明らかにした。NK92IL2内では、NKP44発現もまた増加した(3.3倍)。パーフォリン−1の発現は全細胞系内で類似であったが、NK92IL2内ではグランザイム−B発現が低下してTNF−αは増加し、NK92CIRB内ではどちらも辛うじて低下したが、IFNγは増加した。
NK92IL2およびNK92CIRBのin vivoの生存率および全身性循環を腫瘍担持動物の状況において評価した。U251MG腫瘍細胞は、Nod/scidマウスにおいて皮下増殖させた。腫瘍サイズが平均160mm3に達した時点に、動物には4日間以内に尾静脈を介して2回の非照射NK92IL2およびNK92CIRB細胞の注射を受けさせた。図6Aは、NK92CIRB生存細胞の初回注射から24時間以内に、迅速な46%の腫瘍容積後退が観察されたが、NK92IL2細胞は35%減少を誘発したことを示す。これとは対照的に、腫瘍は未処置動物では増殖し続け、消退する前にはほぼ200mm3の最大限度サイズに達した。このサイズ依存性の限定された増殖は、VEGF発現によって改善することのできる、これらの腫瘍の不良な血管形成に起因することが以前に証明された(49)。NK92CIRB処置群についての腫瘍消退は第2回注射後に持続したが、他方NK92IL2処置動物における腫瘍は増殖を再開し、第18日まで応答しなかった。3週間後、未処置およびNK92IL2群は、類似の腫瘍サイズを示した。これと比較して、NK92CIRB処置群は、86%の有意な腫瘍容積減少を提示した。NK細胞注射の17日後、血液(0.5mL)を3群のマウスから採取し、mCherryおよびヒトCD45の両方を発現する循環細胞に対してサイトメトリーにより分析した。図6Bは、循環中NK細胞がNK92CIRB処置群においてのみ検出できることを示している。この結果は、CIRB発現が腫瘍担持動物において存続する能力をNK細胞に授けるが、IL2分泌は授けないことを示唆している。
NK92細胞系は、アグレッシブ非ホジキンリンパ腫患者から単離した(43)。このため、FDAは、増殖を防止するための注入前の5〜10GyのNK92細胞照射を要求している。これらの条件下で、照射されたNK92細胞の生存率は、2日間以内に劇的に低下する。NK92IL2およびNK92CIRBは、10Gy(0.83gy、12分間)で照射し、その後、24時間毎にトリパンブルーを使用してそれらの生存率を決定するために完全NK92培地中でプレーティングした。図7Aは、照射24時間後、NK92CIRB細胞の57%およびNK92IL2細胞の45%が生存することを示している。NK92CIRBの生存率の利点は、第1日および第2日で統計的有意であった(*P<0.05)。
前立腺癌細胞系PC−3(50)は、アンドロゲン受容体およびPSA陰性であり、Nod/Scidマウスにおいて増殖させると極めてアグレッシブな腫瘍を形成する。腫瘍容積が約200mm3に達した時点に(第28日)、照射されたNK細胞(500cGy)を尾静脈を介して4週間毎の注射として投与した。図7Bは、NK92CIRB処置群におけるPC−3腫瘍の増殖が第1回注射後に緩徐化されたことを示している。最終NK92CIRB細胞の注射後、未処置群と比較して、約17日間の有意な腫瘍増殖遅延が第1回〜第4回NK92CIRB細胞の注射期間中に記録された(**P<0.01)。これとは対照的に、NK92IL2処置群の腫瘍は未処置腫瘍群と比較して7日間の腫瘍遅延しか生成しなかった(*P<0.05)。
インターロイキンIL4、IL7、IL9、IL15およびIL21は、IL2と同一ファミリーに属し、同一の共通IL2Rgを使用する。それらは、それらの固有のα受容体に加えてIL2Rβを使用するIL2およびIL15を除いて、全部がそれらの固有のプライベート受容体を有する(図11)。本発明者らは、PC−3細胞に対して、キメラNK92CIRBを発現するNK92細胞の細胞傷害性に添加されたサイトカイン(IL2、IL4、IL7およびIL21)が及ぼす影響について試験した。本発明者らは、細胞傷害性を劇的に増加させることができたのがIL21だけであることを見いだした(図12は、IL21およびIL2の影響だけを示している)。これに鼓舞されて、本発明者らは、IL2およびIL21が同一IL2Rgを使用する場合、1つのキメラサイトカイン受容体において両方のシグナル伝達を結び付けることが可能であるかどうかを調査した。それを解決するために、IL21Rの全細胞質ドメインをクローニングし、次にキメラCIRB内のIL2RβのC末端に頭−尾結合で加えた。これは新規なIL2−IL2Rβ−IL21Rキメラ(図13に例証した、いわゆるCIRB21)を生じさせ、これを次にNK92細胞内に導入するとNK92CIRB21が産生した。

Claims (26)
- 介在性リンカーとともに、インターロイキン2受容体β(IL2Rβ)のN末端に融合したインターロイキン2(IL2)を含む融合タンパク質。
- 前記IL2は配列番号34を含む、および/または前記IL2Rβは、配列番号35のアミノ酸27〜551を含む、請求項1に記載の融合タンパク質。
- IL2とIL2Rβの前記N末端との間の前記介在性リンカーは、IL2Rαの細胞外ドメインを含む、請求項1に記載の融合タンパク質。
- IL2Rαの前記細胞質ドメインは、配列番号28を含む、請求項3に記載の融合タンパク質。
- 任意選択的にそれらの間の介在性リンカーとともに、IL2Rβの前記C末端でIL21Rの細胞質ドメインをさらに含む、請求項1に記載の融合タンパク質。
- IL21Rの前記細胞質ドメインは、配列番号36のアミノ酸254〜538を含む、請求項5に記載の融合タンパク質。

- 好ましくは前記NK細胞は、さらにCD16ならびに任意選択的にNKP44、NKP46およびNKP30を発現する、請求項1〜8に記載の融合タンパク質を発現する単離ナチュラルキラー(NK)細胞。
- 被験者、好ましくは癌を有するヒト被験者を治療する方法であって、それを必要とする被験者に請求項1〜8に記載の融合タンパク質を発現する治療有効量のナチュラルキラー(NK)細胞を投与する工程を含む方法。
- 前記被験者は、固形腫瘍を有する、請求項12に記載の方法。
- 1種以上の抗腫瘍モノクローナル抗体もしくはチェックポイント阻害剤を投与する工程をさらに含む、請求項12に記載の方法。
- 前記NK細胞は、静脈内投与される、請求項12に記載の方法。
- 前記NK細胞は、投与される前に500〜1000cGyのγ線照射を受ける、請求項12に記載の方法。
- GVHDもしくは自己免疫疾患を有する被験者を治療する方法であって、それを必要とする被験者に請求項7または8に記載の融合タンパク質を発現する治療有効量の制御性T細胞(T−reg)を投与する工程を含む方法。
- 前記T−reg細胞は、静脈内投与される、請求項17に記載の方法。
- 前記NK細胞は、投与される前に500〜1000cGyのγ線照射を受ける、請求項17に記載の方法。
- 被験者、好ましくは癌を有するヒト被験者を治療する際に使用するための、請求項1〜8に記載の融合タンパク質を発現するナチュラルキラー(NK)細胞。
- 前記被験者は、固形腫瘍を有する、請求項20に記載の使用のためのNK細胞。
- 1種以上の抗腫瘍モノクローナル抗体もしくはチェックポイント阻害剤を投与する工程を含む方法において使用するための、請求項20に記載の使用のためのNK細胞。
- 前記NK細胞は、静脈内投与するために処方される、請求項20に記載の使用のためのNK細胞。
- 前記NK細胞は、投与される前に500〜1000cGyのγ線照射を受ける、請求項20に記載の使用のためのNK細胞。
