Abstract
Description
本発明は、対象の細胞媒介性免疫能を測定するための方法に関する。特に、この方法は、免疫細胞、例えば対象から得られた末梢血単核細胞(PBMC)を含む試料に対して細胞媒介性イムノアッセイ法を実施すること、および少なくとも3種のウイルス抗原に対する免疫応答を検出することを含む。
免疫能は、適応(T細胞およびB細胞)免疫応答および先天(補体およびToll様受容体)免疫応答を含む正常に機能している免疫応答が個体に存在することに関する。免疫能、特に細胞媒介性免疫能を測るように設計されたアッセイ法を用いて、抗原に対するドナーのT細胞応答をモニターすることができる。個体の免疫能、特に、細胞媒介性免疫応答を開始する一般能力を測るまたはモニターすることが望ましい。例えば、免疫能の評価は、移植、癌、自己免疫疾患、HIVを含むいくつかの疾患状態およびワクチン試験に関して、臨床医にとって有用である。

本発明者らは、細胞媒介性イムノアッセイ法(CMIアッセイ法)を用いて対象の免疫能を測定する方法を特定した。免疫能、特に細胞媒介性免疫能を測定するように設計されたCMIアッセイ法を用いて、抗原に対するドナーのT細胞応答をモニターすることができる。本発明者らは、多種多様な個体における細胞媒介性免疫応答を検出するために、ウイルス抗原の適切なプールを選択できることを確認した。ウイルス抗原のプールは、免疫適格性の個体において、個体の背景に関わらず、応答をもたらす。したがって、このアッセイ法を用いて、イムノコンピテントな個体、およびまた、効果的な免疫応答を開始できない個体を特定することができる。
細胞媒介性免疫(CMI)応答は、個体の免疫状態を明らかにするために使用される。典型的には、臨床免疫学の分野において、CMI応答という用語は、皮膚試験、リンパ球増殖アッセイ法、および特異的抗原の存在下で免疫細胞、例えば末梢血単核細胞(PBMC)によって産生されるサイトカインの検出を包含する。本発明の方法は、インビトロでの細胞媒介性免疫応答を検出することを含んでよい。具体的には、インビトロでの、ペプチドに対するサイトカインに基づくCMI応答が、本発明の方法において検出され得る。
インビトロで検出される免疫応答は、本発明の少なくとも3種のウイルス抗原によって誘発される任意の応答であってよい。免疫応答は、任意のタイプの免疫細胞によって媒介されてよい。特に好ましい態様において、免疫応答は、T細胞、ナチュラルキラー(NK)細胞、および単球より選択される1種または複数種の免疫細胞を含み得る末梢血単核細胞(PBMC)によって媒介される。典型的には、免疫応答は、サイトカイン分泌を評価することによって、好ましくはインターフェロンγ(IFN-γ)分泌を評価することによって、測定される。サイトカイン分泌を測定する方法は、当技術分野において周知である。免疫応答は、好ましくはT細胞応答である。
本発明の方法によるウイルス抗原の断片は、親配列のN末端および/またはC末端の切断によって誘導される、5個もしくはそれより多いアミノ酸を含む配列であってよい。例えば、断片は、約5個もしくはそれより多い、約6個もしくはそれより多い、約7個もしくはそれより多い、約8個もしくはそれより多い、約9個もしくはそれより多い、約10個もしくはそれより多い、約11個もしくはそれより多い、約12個もしくはそれより多い、約13個もしくはそれより多い、約14個もしくはそれより多い、約15個もしくはそれより多い、約16個もしくはそれより多い、約17個もしくはそれより多い、約18個もしくはそれより多い、約19個もしくはそれより多い、約20個もしくはそれより多い、約21個もしくはそれより多い、約22個もしくはそれより多い、約23個もしくはそれより多い、約24個もしくはそれより多い、約25個もしくはそれより多い、約26個もしくはそれより多い、または約27個もしくはそれより多いアミノ酸を含んでよい。断片は、約5〜約27、約6〜約26、約7〜約25、約8〜約24、約9〜約23、約10〜約22、約11〜約21、約12〜約20、約13〜約19、約14〜約18、約12〜約18、約12〜約15、約15〜約18、約13〜約17、約14〜約16、約5〜約10、約10〜約15、約15〜約20、約20〜約25、または約10〜約20アミノ酸長であってよい。
本発明の方法は、1つまたは複数の断片を含むペプチドのプールに対する免疫応答をインビトロで検出することを含んでよい。ペプチドのプールは、単一種のウイルスの、複数種のウイルスの、またはすべて異なるウイルスのウイルス抗原に由来する断片を含んでよい。ペプチドのプールは、同じウイルスおよび異なるウイルスのウイルス抗原に由来する断片を含んでよい。本発明の好ましい局面において、1つまたは複数の断片は、少なくとも3種のウイルス抗原、すなわち、2種のウイルス、好ましくは3種、4種、5種、または6種のウイルスに由来する少なくとも3種のウイルス抗原に由来する。好ましい態様において、1つまたは複数の断片は、少なくとも3種のウイルス抗原、すなわち、ヒトアデノウイルス3、インフルエンザウイルス、エプスタイン・バーウイルス、および呼吸器合胞体ウイルスのうちの1つまたは複数に由来する少なくとも3種のウイルス抗原に由来する。
1つの態様において、プール中の断片は、タンパク質断片ライブラリーを形成する。タンパク質断片ライブラリーは、親タンパク質、すなわち本発明の場合、ヒトアデノウイルス3に由来するヘキソンタンパク質、インフルエンザウイルスに由来する基質タンパク質1、インフルエンザウイルスに由来するヌクレオカプシドタンパク質、エプスタイン・バーウイルスに由来するBALF-4、BMLF-1、BRLF-1、BZLF-1 EBNA-1、EBNA-3A、EBNA-3B、EBNA-3C、EBNA-4、LMP-1、およびLMP-2タンパク質、ならびに呼吸器合胞体ウイルスに由来する融合糖タンパク質G0、に由来する複数の断片であって、親タンパク質の配列の少なくとも10%、例えば、少なくとも20%、少なくとも30%、少なくとも40%、少なくとも50%、少なくとも60%、少なくとも70%、少なくとも75%、少なくとも80%、少なくとも85%、少なくとも90%、少なくとも95%、少なくとも98%、少なくとも99%、または100%を全体として包含する、複数の断片を含む。本発明において、プール中の断片は、好ましくは、それらの断片の由来元であるタンパク質の配列の少なくとも80%を包含するタンパク質断片ライブラリーを形成する。より好ましくは、プール中の断片は、それらの断片の由来元であるタンパク質の配列全体を包含するタンパク質断片ライブラリーを形成する。
本発明の少なくとも3種のウイルス抗原に対する免疫応答のインビトロ検出は、対象から得られた試料を用いて実施する。好ましい態様において、試料は、血液試料である。
本発明の方法は、任意の適切な対象の免疫能を測定するのに使用され得る。通常、対象はヒト対象である。
1つの態様において、本発明の方法は、免疫細胞、例えば末梢血単核細胞を含む試料に対して細胞媒介性イムノアッセイ法を実施することを含み、この方法は、少なくとも3種のウイルス抗原に由来するペプチドのプールに対する免疫応答をインビトロで検出することを含む。ペプチドのプールは、ウイルス抗原のうちの1種もしくは複数種の1つもしくは複数の断片または1種もしくは複数種のウイルス抗原のそれぞれの1つもしくは複数の断片を含んでよい。これらのウイルス抗原は、1つまたは複数のタンパク質断片ライブラリーを構成してよい。
前述したように、本発明の方法は、少なくとも3種のウイルス抗原に対する免疫応答をインビトロで検出することを含む。免疫応答の検出は、対象が免疫適格性であることを示す。免疫応答の検出の欠如(または検出のないこと)は、対象が免疫応答性ではなく、低い免疫能を有していることを示す。例えば、免疫適格性アッセイ法における免疫適格性ドナーとは、陰性対照においてスポットがなく、陽性対照において20個より多いスポットを示し、イムノコンピテンシーアッセイ法(ICA)抗原に応答して特異的IFN-γスポットを生じるドナーである。
本発明は、免疫細胞、例えば対象からの末梢血単核細胞(PBMC)を含む試料に対して細胞媒介性イムノアッセイ法(CMI)を実施することを含み、少なくとも3種のウイルス抗原に由来するペプチドのプールに対する免疫応答をインビトロで検出することを含む、対象の免疫能を測定するための方法を提供する。本発明の方法は、少なくとも3種のウイルス抗原に対するドナーの幅広いT細胞応答をモニターするために、臨床で有用であり得る。これは、移植、癌、自己免疫疾患、HIVを含むいくつかの疾患状態に関する患者の免疫能を評価する際、およびワクチン試験において、臨床医の助けになり得る。
ELISPOTアッセイ法によって、23個の市販のペプチドプールを用いて健常ドナー試料(31〜167)をスクリーニングした。免疫適格性ドナーのカバー率が最も良かったペプチドプールを特定するために、任意の10スポットのカットオフを設定した。次いで、各ペプチドプールに対する応答を、このカットオフを用いて評価した。表1は、応答性ドナーの比率に基づいてペプチドプールを順位付けしたものである。
特別仕様のペプチドプール(ヘキソン、EBV、MP1、NP、FGF0、およびBZLF1)を合成し、凍結乾燥させたプールとして供給した。ジメチルスルホキシド(DMSO)を用いて、これらのペプチドプールを溶解して、プール中のペプチド1つ当たり625μg/mLの濃度にした。
使用濃度(3μg/mL)の各ペプチドプールの安定性を、加速安定性試験を用いて評価した。手短に言えば、次のとおりである:使用濃度のペプチドプールを37℃で8週間保存した。次いで、ELISPOTアッセイ法において、ストレスを加えたペプチドプールを、新たに希釈したペプチドプール(対照)と比較して試験した。各ドナーについて、3つのレプリケートを試験した。これらの研究の結果を図1に示している。
ELISPOTアッセイ法でのペプチドプールのスクリーニングから得られたデータを再評価して、あたかも様々な抗原に由来するペプチドが混合されて1つのプールにされたかのように、ドナーの理論的カバー率を決定した。これは、個々のペプチドプールについてのスポット計数値を足すことによって実施した(例えば、ドナー15、ヘキソンスポット計数値=5、EBVスポット計数値=26、NPスポット計数値=5、理論上のミックス16スポット計数値=36)。スポット10〜350個の範囲内のドナーの比率を各ミックスについて計算した。この解析の結果を表2に示している。
・10種のミックスが、評価したドナーの95%超に応答を生じさせたであろうこと。
・ミックス25および26が、ELISPOTプレートリーダー上で正確に計数可能である上限であるスポット計数値(400+スポット)を理論上与えるであろうこと。これにより、スポット計数値10〜350個の範囲内のドナーの比率が低下した。
・13種のミックスをELISPOTアッセイ法でさらに試験するために選択した(表3)。
溶解させたペプチドプール(プール中のペプチド1つ当たり625μg/mL)を混合して、イムノコンピテンシーアッセイ法(ICA)ミックスとした。調製時に問題は認められなかった。
35名のコーカサス人ドナー、10名のアフリカ系アメリカ人ドナー、10名のラテンアメリカ系ドナー、および9名のアジア人ドナーから採取した血液試料を用いるELISPOTアッセイ法において、13種の選択されたICAミックスを試験した。この試験で得られた結果を図3に示している。
ICAミックスによって誘導される応答の自然変動を明らかにするために、ELISPOTアッセイ法でICAミックスを用いて、6名の免疫適格性血液ドナーを次の時点:0日目、3日目、7日目、14日目、28日目、56日目、および84日目に試験した。

Claims (18)
- 対象からの免疫細胞を含む試料に対して細胞媒介性イムノアッセイ法(CMI)を実施することを含み、少なくとも3種のウイルス抗原に由来するペプチドのプールに対する免疫応答をインビトロで検出することを含む、対象の細胞媒介性免疫能を測定するための方法。
- 免疫細胞が末梢血単核細胞(PBMC)である、請求項1に記載の方法。
- 免疫細胞がT細胞である、請求項1または請求項2に記載の方法。
- プールまたはペプチドが、少なくとも4種、5種、または6種のウイルス抗原に由来する、請求項1に記載の方法。
- ペプチドのプールが、3種、4種、または5種のウイルス抗原に由来する、請求項1に記載の方法。
- ウイルス抗原が、ヒトアデノウイルス3のヘキソンタンパク質、インフルエンザウイルスの基質タンパク質1(H3N2)、エプスタイン・バーウイルスのBZLF-1タンパク質、インフルエンザウイルスのヌクレオカプシドタンパク質、呼吸器合胞体ウイルスの融合糖タンパク質G0より選択される、請求項1〜5のいずれか一項に記載の方法。
- ウイルス抗原が、ヒトアデノウイルス3のヘキソンタンパク質、呼吸器合胞体ウイルスの融合糖タンパク質G0、およびインフルエンザウイルスのヌクレオカプシドタンパク質からなる、請求項1〜6のいずれか一項に記載の方法。
- ウイルス抗原が、ヒトアデノウイルス3のヘキソンタンパク質、インフルエンザウイルスの基質タンパク質1、インフルエンザウイルスのヌクレオカプシドタンパク質、および呼吸器合胞体ウイルスの融合糖タンパク質G0からなる、請求項1〜6のいずれか一項に記載の方法。
- ウイルス抗原が、ヒトアデノウイルス3のヘキソンタンパク質、エプスタイン・バーウイルスのトランスアクチベータータンパク質BZLF1、および呼吸器合胞体ウイルスの融合糖タンパク質G0からなる、請求項1〜6のいずれか一項に記載の方法。
- ウイルス抗原が、ヒトアデノウイルス3のヘキソンタンパク質、エプスタイン・バーウイルスのBALF-4、BMLF-1、BRLF-1、BZLF-1 EBNA-1、EBNA-3A、EBNA-3B、EBNA-3C、EBNA-4、LMP-1、およびLMP-2タンパク質、インフルエンザウイルスの基質タンパク質1、インフルエンザウイルスのヌクレオカプシドタンパク質、ならびに呼吸器合胞体ウイルスの融合糖タンパク質G0からなる、請求項1〜6のいずれか一項に記載の方法。
