ウイルスによる腫瘍モデル及びEP腫瘍モデルは様々な組織におけるがんの発生進化的側面を研究するためにますます用いられているが、様々な遺伝子送達方法に関して問題がある。推定上のがんドライバー遺伝子のインビボの自然発症モデル化のためには本発明の方法が最も適すると推論し、原理証明のためにHrasG12V(よく使用される活性化がん遺伝子)を選択した。推定上の単一コピーのヘテロ接合のマウスにおける増殖ダイナミックスの組織学的分析から、HrasG12V細胞は、EGFP+集団と比較した場合に、急速に過剰増殖したということが示された(図13A)。さらに、本発明者は、ホモ接合性へmTmGマウスを繁殖させることによって、細胞のヘテロ接合集団及びホモ接合集団を識別でき得ると推論した。具体的には、理論的には4つの可能性が組換えまたは挿入後に生じ、各々は遺伝的マーカーの異なる組み合わせを有するだろう(図2B)。生後2日のEP後に、HrasG12Vを用いてEPを行ったホモ接合のmTmGのすべては、急速にグリオーマを発症し、3〜4か月以内に最終的に死亡した(図2a;n=4)(CAGプロモーターによって駆動される同じがん遺伝子を使用して、本発明者はHrasG12VのPB−EPが100%浸透するグリオーマをもたらすことを以前に示している)。ホモ接合のmTmGマウスにおいて、10ng/μlのプラスミドを使用する場合でさえ、MADR反応は高度に効率的であった(図13B)。興味深いことには、このホモ接合のmTmGグリオーマにおいて、青色のみの細胞(Rosa26HrasG12V×2)は、青色及び緑色の両方を発現する細胞(Rosa26HrasG12V×1)よりも、腫瘍横断面のうちの大きなパッチを占めた(図2c、7b)。以前に、HrasG12Vのコピー数は、表現型の差(増殖及びアポトーシスの率等)を付与することが示されている。本発明者は、PB−EPを使用して、より明るいEGFPタグ付加HrasG12V細胞が、薄暗いEGFP+細胞よりも、リン酸化Rb1(pRb1)を多く発現することも観察した(図13C)。同様に、HrasG12Vを用いてEPを行ったmTmGマウスにおける大部分の推定上のホモ接合のRosa26HrasG12V×2細胞はpRb1を発現するが、ヘミ接合のRosa26HrasG12V×1は発現しないように思われる(図7b〜7c)。このデータは、GEMMを使用して以前に観察されたように、がん遺伝子のコピー数が、生じた腫瘍集団のプロファイルを有意に変更する可能性を示す。
最近、CRISPR/Cas9は、EPを使用する遺伝子の突然変異についてインビボで高度に効率的なことが実証されている。しかしながら、これらの研究の欠点は、修飾された細胞の系譜を追跡する決定的な手法である。この問題に取り組むために、同時に細胞を標識し突然変異させる、SM_BFP2−P2a−SpCas9含有ドナープラスミドを作成し、インビボでの突然変異細胞の正確な追跡を可能にした(図15A)。Nf1、Trp53及びPtenのタンデムmiR−E及びCas9媒介性ノックアウトによる本発明の前述の所見を考慮し、これらの同じPCRを行った(PCR−ed)sgRNAを使用して、Nf1及びTrp53を標的化した。EPを行った細胞において成功した標的化は、シーケンシングによって腫瘍集団のゲノムDNA中で確認された(図15B)。大まかに5か月で、EPを行った動物での最終的な死亡が観察された。病理学的分析により、主に腫瘍における壊死の存在に起因して、多形性グリオブラストーマの診断が導かれた。免疫組織化学的に、腫瘍では、血管を例外として、TdTomatoに標識された集団がほとんど無いことが観察された(図15C〜C1)。小さなEGFP集団は、標的化部位が存在することが予想される付近で観察された(図15C、C2;矢頭)。しかしながら、腫瘍は、MADR MAX SM_BFP−V5で標識された細胞によって区切られ、半球を超えて異常増殖した(図15C、C3)。大部分のこの体積はOlig2+集団により満たされていが、シグナルが欠如した領域が観察された(図15;矢頭;15E)。Olig2は、前神経性グリオーマサブタイプのマーカーであるので(本発明者及び他の者は間充織発達に先行することを観察している)、これが腫瘍発達の部位であるかどうかを、間充織マーカー(CD44)についての染色によって評価した。とりわけ、CD44は、腫瘍の全体にわたって見出されたがこのOlig2減退領域でエンリッチされていた(図15E〜F、データ不掲載)。TdTomatoの血管が顕著であること、及びSM_BFP−V5が腫瘍細胞系譜を識別する能力に起因して、V5タグ付加血管周囲細胞の顕著な集団は他の集団から際立っていた(図16A;矢印)。腫瘍細胞と間質とを遺伝的マーカーにより遺伝的に識別する本発明の能力と一緒に、これらの細胞の所在位置及び形態を考慮して、グリオーマ細胞の分化転換に関する現在の論争を再考した。具体的には、複数のグループは、グリオーマ細胞が内皮細胞へと分化転換し得ることを示した。これは、その代りに周皮細胞への分化転換であるという所見によって争われた。他のグループは、かなりの調査にもかかわらずいずれについても証拠が欠如していることを見出している。Pecam1(内皮細胞の真のマーカー)のTdTomatoとの共局在化の試験において、本発明者は、実質的にすべてのPecam1+細胞が共標識されていると示し、血管は腫瘍由来でないことを示唆した(図16B)。しかしながら、非常に稀なPecam1+/TdTomato−シグナルが示され、小さなサイズ(<5μm)であること及び正常な内皮細胞のサイズのプロファイルがほんのわずかのであることを考慮すると、その大部分は残骸であると思われる(図16C〜D)。しかしながら、管に結び付いた細胞は見出されず、これらの推定上の細胞数は体系的調査に適していなかった(図16C〜D)。反対に、十分に検証された周皮細胞マーカーであるPdgfrb及びDes(すなわちデスミン)の両方を使用して、本発明者は、前述のOlig2−/Cd44+部位で及びその周辺で、不連続な所在位置を観察することができ(図15D中で矢頭によって描写された領域に大まかに隣接する切片において)、この不連続な所在位置はTdTomatoの非存在下においてどちらの周皮細胞マーカーについても陽性を示した(図15G〜H)。TdTomatoの抗体増幅及びこれらの領域におけるTdTomatoシグナルの人為的に過度な露出にもかかわらず、いくつかの周皮細胞においてTdTomato及びDesの共局在化は観察されなかった(図16E)。しかしながら、腫瘍の大部分の領域(特にOlig2エンリッチ領域)において、これらの周皮細胞マーカーは、TdTomato膜と厳密に共局在することが見出され、この分化転換が、前神経性サブタイプが優勢な領域中に広がっていないことを示唆した(図16F)。したがって、本発明の新規の自然発症モデル(それはインビボで複数の集団の明確な遺伝学的標識を可能にする)を使用して、本発明者は、GBMにおける腫瘍細胞の周皮細胞への局所性分化転換についての証拠を提供する。これは、グリオーマ細胞の周皮細胞への分化転換がグリオーマの不均一な発達(前神経性サブタイプから間充織サブタイプのものが含まれる)と相関し得ることを示唆する。
トランスポゾン(PB及びSleeping Beauty等)はEPと組み合わせてますます使用されて、近年これらの技法を利用する複数の報告により安定的な体細胞遺伝子導入体が産生される。トランスポゾンは長期的な発生的研究及びさらにインビボの腫瘍生成を可能にするので、非常に魅力的なツールである。本発明の新しい方法はトランスポゾン系の2つの大きな問題:ランダムなゲノム挿入及びコピー数変動を克服する。ノッチシグナリングは、遺伝子用量感受性のある分子経路の1つの重要な例である。加えて、他の複数の細胞運命決定要素は、それらの発現レベルに基づいて劇的に差次的な表現型をもたらすことが示されている。例えば、グリア前駆細胞におけるNfiaの高発現はアストロサイトへの分化を促進する一方で、低発現はオリゴデンドロサイトになる細胞において観察され、別の例において、より高いFezf2発現は、ノッチシグナリングエフェクターのアップレギュレートによって脳室帯/脳室下帯におけるNSC休止を誘導する。本発明の方法は迅速にインビボでかかる経路を査定するために使用することができる。本明細書において示される1コピーvs2コピーの比較パラダイムを使用して、かかる核内因子は、天然の切片内の対照細胞により1匹のマウスにおいて調査することができる。GEMM操作と比較した場合に、本発明のインビボのMADRは拡張可能であり、比較的低価格である。さらに、出生後のEP手順が迅速であり(動物の1腹仔あたり約35分)、侵襲手術を要求しないという事実を考慮すると、本方法論は容易に多くの研究室によって採択され得る。
本ストラテジーは、デュアルリコンビナーゼ部位を保有する任意の既製のGEMM(例えばAi14マウス、R26−CAG−LF−mTFP1マウス、Ribotagマウス、及びIKMCマウス)と共に用いることができ、定義された供与量でのインビボのGOFタンパク質及びLOFタンパク質機能の決定的な査定を可能にする(図16)。目的の遺伝子座の周囲にloxP及びFRTの部位を既に保有する何千もの遺伝子導入マウスがある。Rosa26mTmGと同様に、Ai14は別の広く用いられるレポーターマウス系統であり、それは典型的にはリコンビナーゼ発現遺伝子導入マウスと交配繁殖される。図16中で図示されるように適切にリコンビナーゼ認識部位を配向させることによって、ドナープラスミドをAi14マウスにおける使用のために作成することができる。RibotagはCre組換えを使用して、Cre発現に定義されたmRNAの親和性免疫沈降のために、タグ付加していないRpl22エクソン4をHAタグ付加バリアントとスワップする。MADRにより、代替タグは、タグについての連続的な免疫沈降を使用する同時のオルソゴナルなmRNAの精製を可能にするように、追加のエレメントを挿入することができる。加えて、スプライスアクセプターにより開始するORFを作成し、このコンストラクトを使用して、外来のシス調節環境下で導入遺伝子を置換する効果を調査することができる(図16)。同時に、蛍光レポーターを備えたドナープラスミドは単純にエレクトロポレーションして(Flp及びCreと共に)、loxP部位及びFRT部位が重要な遺伝子座に隣接する様々な遺伝子導入マウスにおいて、Cre発現マウスについての必要性なしに、焦点からの系譜追跡を可能にすることができる。

クローニング
Flp−Creコンストラクトは、Y.Voziyanovによって惜しみなく提供され、dRMCEのコンテキスト中で検証された。本発明のドナープラスミドは、標準的な制限消化技法と組み合わせたIn−Fusion technique(Clontech)を使用して、PGKneotpAlox2から導かれた。FRT部位は、2つのオリゴをアニールし、PGKneotpAlox2の中へ挿入物をインフュージョンすることによって作成した。他のドナープラスミドの下流の生成は、既存のORFを除去しIn−Fusionまたはライゲーションを使用して、新しいカセットを追加することによって行った。PB−CAGプラスミドは以前に記載された。
Gt(ROSA)26Sortm4(ACTB−tdTomato,−EGFP)Luo/Jマウスは、Jackson Laboratoryから購入した。mTmGを野生型CD1マウス(Charles River)と交配させて、ヘテロ接合のマウスを生成した。すべてのマウスは、Cedars−Sinai研究機関内動物実験委員会に従って使用した。出生後の側脳室EPは、以前に記載されるように実行した。生後1日〜3日の仔マウスを約5分間氷上に置いた。すべてのDNA混合物は、トリス−EDTAバッファー中で希釈された、0.5〜1μg/μlのFlp−Cre発現ベクター、ドナープラスミド、hypBase、またはCAGレポータープラスミドを、特別に示さない限り含有した。ファストグリーン色素を混合物へ添加し(10%v/v)、それを側脳室の中へ注入した。Platinum Tweezertrodesは、ECM 830 System(Harvard Apparatus)からの120Vの5パルス(50ミリ秒;950ミリ秒で分離される)を送達した。SignaGelを適用してコンダクタンスを増加させた。マウスを加熱ランプ下で暖め、ケージへ戻した。
麻酔後に、マウス脳を単離し、4℃で一晩、4%のPFA中で固定した。脳を4%の低融点アガロース中に包埋し、ビブラトーム(Leica)で70μmの切片にした。
免疫組織化学は以前に記載されるように標準的方法論を用いて遂行した。すべての二次抗体(Jackson ImmunoResearch)は1:500で使用した。一次抗体についての詳細は表3に見出すことができる。
3匹のヘテロ接合の生後0日のmTmG仔マウス脳を分離して、本研究において使用するマウス神経幹細胞株を確立した。以前に記載されるように細胞株を維持した。ビタミンA不含有B−27(Life Technologies 12587−010)、GlutaMAX(Life Technologies 35050)、抗生物質−抗真菌剤(Life Technologies 15240)、hEGFP(Sigma E9644)、ヘパリン(Sigma H3393)、及びbFGF(Millipore GF003)を補足したNeurobasal(登録商標)−A Medium(Life Technologies 10888−022)を含有する培地中で、細胞を増殖させた。神経幹細胞ヌクレオフェクションは、製造者の推奨(Lonza AG)に従ってNucleofector 2bデバイス及びマウス神経幹細胞キットを使用して遂行した。ヌクレオフェクション混合物は、10ng/μlの等しい濃度のプラスミドを含有していた。
すべての固定画像は、Nikon A1Rの倒立レーザー共焦点顕微鏡で収集した。mNSCのライブ画像はEVOSのデジタル蛍光倒立顕微鏡で得た。全脳画像のために、Nikon Elementsの自動化スティッチング機能を使用した。次いでND2ファイルをImageJの中へインポートしてZ投影画像を作成し、後続してそれをAdobe Photoshop CS6で編集した。Adobe Illustrator CS6を最終的な図の制作のために使用した。
以前に記載されるように細胞を収集した。細胞をPBS中で短時間リンスし、Accutase(Millipore)を使用する酵素による解離によって取り出し、250gで3分間ペレット化し、培地中で再懸濁した。FACSは、Cedars−SinaiフローサイトメトリーにおいてBeckman Coulter MoFloで行った。
細胞ペレットをLaemmliバッファー中で再懸濁し、95℃で5分間煮沸した。タンパク質濃度をThermoScientific NanoDrop 2000で測定した。SDS−PAGE分離及びニトロセルロース膜上への転写後に、0.1% PBS−Tween中の5% 脱脂乳中で希釈した、表3でリストされた抗体を使用して、タンパク質を検出した。すべての二次抗体(Li−cor IRDye(登録商標))は1:15000で使用した。赤外線検出はLi−Cor Odyssey(登録商標)CLX Imaging Systemによって達成した。
ドキシサイクリン(Clontech 631311)を100ng/mlの最終濃度で培養培地へ添加した。ピューロマイシン(Clontech 631305)を1μg/mlで使用した。
本発明者は、FlExベースの導入遺伝子発現(特にEGFPカセット(FlEx−EGFP)のCre媒介性の逆位及び活性化)を以前に使用した。Nf1、Pten、及びTrp53を標的化する本発明のmulti−miR−Eを試験するために、多重miR−Es(FlEx−multi−miR−E)を保有するCAG駆動性FlExベースのコンストラクトを製作した。出生後のmNSC株をCD1仔マウス脳の分離によって確立し、EGFPまたはFlEx−multi−miR−E及びCreリコンビナーゼベクターをトランスフェクションした。蛍光細胞を選別し、mRNAの抽出、ならびにNf1、Pten及びTrp53についてのqPCRプローブを使用するSYBRベースのFluidigm BioMarkダイナミックアレイを行った。
各々の条件について、2匹の仔マウスにpCAG−TagBFP2−nls、pDonor−smFP−HA、及びFlp−2A−Creをエレクトロポレーションした。脳をEP後2日に得て、各々の脳からの2つの隣接しない切片をHAタグ抗体及びEGFPにより染色した。各々の切片について、約25のBFP+細胞をランダムに選択し、その中で、BFP+細胞中の、HA+細胞及びEGFP+細胞をカウントした。各々の群について4つの切片にわたって、割合を平均化した。
短いリバースプライマー及びultramerフォワードプライマー(IDT DNA)をPCR反応及び後続する精製において組み合わせて、濃縮したsgRNAを製作した(Ran et al.,2013)。100ngの各々の断片をEPのためにプラスミドDNAと組み合わせた。
腫瘍細胞の純粋な集団をFACSによって得て、ゲノムDNAを単離した(Qiagen DNeasy)。gRNA標的部位に隣接するプライマーを使用して、NF1、Trp53、及びPtenについてのInDel突然変異を含有すると予想される領域をPCR増幅した。PCR増幅断片をThermo Fisher Zero Blunt TOPOキットを使用してtopoクローニングし、One Shot MAX Efficiency DH5−T1R細胞の中に形質転換した。
