Description
本発明は、そのいくつかの実施形態において、MaSp(大瓶状腺スピドロイン((ampullate spidroin))タンパク質に由来するタンパク質に基づく複合材料、及びその調製を対象とする。
クモドラグラインシルクは、クモの巣のフレーム及び半径だけでなくクモが降りる又は危険を逃れるときのライフラインを構築するために、円網を作るクモによって使用されるシルクとして当技術分野で知られている。これらの作業を実行できるように、ドラグライン繊維は、高い弾性及び強度の組み合わせにより著しく高い靭性を示し、天然又は人工に関わらず最も強靭な繊維として位置づけられている。例えば、ドラグラインは、その直径において高張力鋼の6倍強く、これまでに作られた最強の合成繊維の1つであるケブラーよりも3倍強靭である。
天然のクモシルクに似た機械的性質を有する繊維を生成するための改良された組成物及び方法に対する必要性が満たされていない。

材料及び方法
プラスミド:Geneart(Regensburg,Germany)から入手したPCR−ScriptAmpSK(+)プラスミド中のDNA配列。pFastBacHTaをInvitrogenから入手した。
クモドラグラインシルクタンパク質の単一リピート単位をコードする配列の合成:ADF−4(Genbank登録U47856)の反復領域を構成する15個のリピートの平均コンセンサス配列を表す35アミノ酸長の配列を設計した。平均的なコンセンサス配列のペプチド配列は、105個のDNA塩基対の配列:5'−TCTGGTCCTGGAGGTTATGGCCCAGGAAGCCAAGGACCATCTGGTCCAGGAGGATATGGTCCAGGCGGACCTGGCTCTAGTGCAGCAGCTGCCGCAGCAGCTGCA−3'(配列番号15)によってコードされるSGPGGYGPGSQGPSGPGGYGPGGPGSSAAAAAAAA(配列番号14)である。上記合成DNAは、PCR−ScriptAmpSK(+)プラスミドの中で得られる。該配列を、クモシルクタンパク質及び繊維の合成に使用されるスポドプテラ・フルギペルダ細胞のコドン使用に従って発現のために最適化した。

エポキシ樹脂を含む複合材料
二成分系を複合材料のマトリックスとして試験した。目的は、系全体の良好な機械的性質を可能にするために、マトリックスの性質を改善することである。カスタムメイドのPTFE金型を材料調製のために使用した。
例示的な手順において、パートA−樹脂ベース及びB−硬化剤を製造業者の推奨に従って秤量した。クモシルク(「SS」、本明細書全体を通して「SSS」とも呼ばれる)繊維を、0、1%及び2%w/wの異なるローディング比率で秤量し、金型に注いだ。硬化プロファイルを、室温で24時間、その後、80℃で3時間であった。被検査物を金型から取り出し、試料の引張強度、弾性率、歪み及び破壊までの仕事量のエネルギー(work to failure energy)について、引張試験機による機械的性質について試験した。各試験セットは、統計データを確立するために、5〜6種の被検査物からなっていた。
熱可塑性及び熱硬化性ポリウレタン(PU)を含む複合材料
いくつかのポリウレタンを試験した。引裂抵抗及び引張強度の改善を評価した。熱硬化PUは主に二成分系であり、熱可塑性PUは、シート状又はペレットで供給され、SS繊維が添加された有機溶媒に溶解した。溶液を金型にキャストし、蒸発させた。
供給されたシートを、溶解速度について試験するために、いくつかの有機溶媒に溶解した。溶解はまた、最終被検査物から生成された最終の機械的性質に影響を与えた。調製方法は、全ての試料について均一であった。
U−2047の二成分ポリウレタン化合物−ベース(パートA−ポリオール)及び硬化剤(パートB−HDI)。ポリマーG−Formulating Successによって提供される。2成分の調製は製造業者の指示に従う。繊維補強の場合、繊維を、パートBと混合する前にパートAと混合した。混合物を、脱気するために、1000RPMで4分間遠心分離し、次いで、さらに脱気するために、100mBarで7分間吸引した。硬化プロファイルは、室温で24時間、その後、80℃で3時間、後硬化した。実験を以下のバッチ:対照、1%、2%及び3%SSバッチを用いて行った。性質の改善が、測定した全4つのパラメータで観察され、LLOYD引張試験機で試験した。
配合及び射出成形によって生成されるナイロンを含む複合材料
ナイロン6、ナイロン6,6及びナイロン12の使用は業界で普及している。クモシルクでナイロンを補強することにより、機械的性質の有意な改善が示された。最初の試験を、受け取ったままのペレットで行った。これは、大きなペレットが完全に溶融せず、SSとの十分な混合が達成されなかった時に、混合相で問題を引き起こした。さらなる試験を、ナイロン12の微粉末形態で実施した。例示的な手順において、配合をDSM Xploreツインスクリューマイクロ配合機(DSM,Heerlen,Netherlands)で行った。調製手順は次の通りである:直径約100μmのフィラメントを、マイクロ配合機を用いて取得し、機械的性質について試験した。5つの被検査物を試験した。
電界紡糸によるクモシルクでの被覆
電気紡糸は、有機溶媒に溶解されたポリマー及び繊維の電界加速によって、非常に細いワイヤを生成することができる方法である。この方法は、30キロボルトの電圧で負に帯電した、集電装置に向かう内容物の加速によって、溶媒の急速蒸発によって機能する。高電位差は、集電装置に向かうポリマー及び溶媒の液滴内容物を引き寄せる電界を形成し、溶媒は急速に蒸発し、ポリマーは集電装置上で固化し、システムの設定パラメータに従ってナノからマイクロの繊維の形成を可能にする。

式中、Fは測定された力であり、Aは測定断面であり、見掛け密度はρmat/ρbulkであり、ρbulkはデンプンタップ粉体(starch tapped powder)密度であり、ρmatは繊維マット密度である。
電界紡糸方法:電界紡糸方法は、繊維の平均直径が0.8μmであるTPU 12%(w/w)安定方法、及び繊維の平均直径が3.0μmであるTPU 16%(w/w)安定方法をもたらした。補強溶液は粘度が高すぎることが判明したので、約10.5%に希釈した。
BUTVAR、EVOH、PHEMA、及びUV硬化型アクリル酸製剤を含む複合材料
合成繊維の機械的性質を特徴付けるために、合成繊維により補強されたポリマーマトリックス試料を調製した。研究のために選択したポリマーマトリックスは、
・pHEMA(ポリヒドロキシエチルメタクリレート)
・ポリビニルブチラール樹脂(BUTVAR B−98)
・Vero Clear(Stratasysから供給されるUV硬化インクジェット材料)
であった。
合成繊維の架橋
さらなる例示的な手順は、織物の最終的な性質がスレッド、ケーブルなどに製織できるようにするために、クモシルクに基づいて、より長い連続繊維を作成することを目的とした。方法は、異なるpHレベルの無機緩衝剤ベースの溶液中で架橋剤を用いる湿式化学に基づく。繊維を分散用緩衝液に添加し、次いで、架橋剤を添加する。pHレベル、架橋剤の濃度、温度、インキュベーション時間などの方法のパラメータは、達成される架橋の程度に影響を与える。様々な注入速度での注入方法などの方法のパラメータは、開発される繊維生成のための連続方法を作るために、架橋剤が繊維と接触するのを可能にする。湿式紡糸、乾式紡糸、ゲル紡糸のような方法をベースとする紡糸口金である他の方法も、繊維生産のための生成方法として使用することができる。以下の架橋剤は水系反応に利用され、いくつかは以下に詳述する有機溶媒を用いて行われる。
材料:EDC(1−エチル−3−[3−ジメチルアミノプロピル]カルボジイミド塩酸塩);NHS(N−ヒドロキシスクシンイミド);結合緩衝液:リン酸緩衝生理食塩水(PBS)、100mMのナトリウム;リン酸、150mMのNaCl;pH7.2;及び活性化緩衝液:0.1MのMES、0.5MのNaCl、pH6.0。
ESEM中のSSの湿潤研究
材料及び方法:いくつかの試料を調製した:凍結乾燥SS繊維をエタノールに分散させ、スライドガラス上に置き、風乾した。凍結乾燥SS繊維を、カーボンテープ上に広げた。凍結乾燥SS繊維を石英シート上に広げた。対照実験として、セルロース繊維をエタノールに分散させ、スライドガラス上に置いた。
クモシルク上での3D細胞成長の評価
例示的な手順において、クモシルク上での3D細胞成長の評価を、いくつかの態様:コーティングしていない培養皿及びコラーゲンコーティングした培養皿上での形態、接着、増殖及び細胞の2D成長比較で実施した。
96ウェルプレート:細胞を、200,000個のL929細胞/mlの密度まで新鮮な培地で希釈した。200μlの細胞懸濁液を、指定のウェルに加え、5%CO2、加湿した37℃のインキュベーターに入れた(最終播種濃度:40,000細胞/ウェル)。
細胞を、SS境界線上に播種し、横方向だけでなく3次元構造で成長させ、生存可能な細胞クラスターを形成させた。培養物の成長及び形態を、倒立顕微鏡を用いて毎日検査する。細胞を、異なる倍率及び異なる焦点面で顕微鏡を用いて観察する。培養皿の底から開始し、ダイアルをゆっくり回し、より高い平面に焦点を合わせる。SS上に播種した細胞によって形成されたクラスターがいくつかの焦点面上に見られ、各焦点レベルで異なる細胞の輪郭を明らかにする。
コーティングプレートのクモシルク(SS)上への細胞の播種及びコーティングされていないプレート上へのSSと混合した細胞の播種
HEK293細胞をSSでコーティングしたプレート上に播種し、未処理プレートにSSを細胞培地と混合して播種した。
SSコーティングプレート上に播種した細胞:HEK293のクラスターが播種後3〜4日目に観察された。SSコーティング上に播種した高密度と低密度の両方の細胞は、播種後5日以内に多層3次元構造で組織化することが示された(図47A)。コーティングしていないプレート上に播種した対照細胞は単層として成長した(図47B)。細胞死は5%未満であった。ほとんどの領域で、細胞の3以上の層が観察された。
Claims (30)
- タンパク質の混合物及び少なくとも1つのポリマーを含む複合材料であって、前記タンパク質の混合物が異なる分子量のタンパク質の「m」型を含み、前記混合物中の各タンパク質は、独立して、大瓶状腺スピドロイン(MaSp)タンパク質、又はその機能的相同体、バリアント、誘導体若しくは断片の「n」リピートを含み、m及びnは、独立して2〜70の整数である、複合材料。
- 前記タンパク質の混合物が、
(a)各リピートが2kDa〜3.5kDaの範囲の分子量を有する;
(b)「m」に対する「n」の比が1.5:1〜1:1.5の範囲にある;
からなる群から選択される1以上の性質によって特徴付けられる、請求項1に記載の複合材料。 - 前記タンパク質の混合物が繊維の形態にある、請求項1〜2のいずれか一項に記載の複合材料。
- 少なくとも1つのポリマーが、非MaSpタンパク質由来のポリマーである、請求項1〜3のいずれか一項に記載の複合材料。
- 前記タンパク質のそれぞれが、独立して、配列番号1:
(X1)zX2GPGGYGPX3X4X5GPX6GX7GGX8GPGGPGX9X10
(式中、X1が、独立して、各場合においてA又はGであり、(X1)Zの少なくとも50%がAであり、Zが5〜30の整数であり、X2がS又はGであり、X3がG又はEであり、X4がG、S又はNであり、X5がQ又はYであり、X6がG又はSであり、X7がP又はRであり、X8がY又はQであり、X9がG又はSであり、かつX10がS又はGである)
に記載のアミノ酸配列を含む、請求項1〜4のいずれか一項に記載の複合材料。 - 前記反復領域が、配列番号3(AAAAAAAASGPGGYGPGSQGPSGPGGYGPGGPGSS)に記載のアミノ酸配列を含む、請求項1〜5のいずれか一項に記載の複合材料。
- タンパク質の前記混合物を含まないポリマーの性質と比較して少なくとも1つの改良された機械的性質によって特徴付けられ、前記性質が、ヤング率、引張強度、破断歪み、降伏点、靭性、破壊までの仕事量、衝撃強度、引裂強度、曲げ弾性率、曲げ歪み及び特定の割合の伸長時の応力からなる群から選択される、請求項1〜6のいずれか一項に記載の複合材料。
- 前記ポリマーが、合成ポリマー、熱可塑性ポリマー、熱硬化性エポキシ、ポリエステル、ポリアミド、ポリオール、ポリウレタン、ポリエチレン、ナイロン、ポリアクリレート、ポリカーボネート、ポリ乳酸(PLA)又はそのコポリマー、シリコン、液晶ポリマー、無水マレイン酸グラフト化ポリプロピレン、ポリカプロラクトン(PCL)、ゴム、セルロース、又はそれらの任意の組み合わせである、請求項1〜7のいずれか一項に記載の複合材料。
- 前記タンパク質の総濃度が、前記複合材料の総重量の約0.1重量%〜約10重量%の範囲にある、請求項1〜8のいずれか一項に記載の複合材料。
- 少なくとも0.5ミクロンの厚さの層を有する、請求項1に記載の複合材料。
- 前記繊維が、少なくとも30%の多孔度によって特徴付けられる、請求項3に記載の複合材料。
- 請求項1〜10のいずれか一項に記載の複合材料を含む物品。
- 生体適合性材料又は埋め込み用材料を含む、請求項12に記載の物品。
- 請求項1〜10のいずれか一項に記載の複合材料及び薬学的に許容される担体を含む、医薬組成物。
- 前記複合材料を形成するために、前記タンパク質の混合物を前記ポリマーに接着させる工程を含む、請求項1〜10のいずれか一項に記載の複合材料を作製する方法。
- 溶融ポリマーを取得するために前記ポリマーを溶融し、前記タンパク質の混合物を前記溶融ポリマーに変換する工程を含む、請求項15に記載の方法。
- 前記ポリマーを溶解し、溶解ポリマーを取得し、前記タンパク質の混合物を前記溶融ポリマーに変換する工程を含む、請求項15及び16のいずれか一項に記載の方法。
- 前記ポリマーを押し出す工程を含む、請求項15〜17のいずれか一項に記載の方法。
- 連続した電界紡糸により前記タンパク質の1以上の層を形成することを含む、請求項15〜18のいずれか一項に記載の方法。
- (a)異なる分子量のタンパク質の「m」型を含み、前記タンパク質の「m」型中の各タンパク質が、独立して、大瓶状腺スピドロイン(MaSp)タンパク質、又はその機能的相同体、バリアント、誘導体若しくは断片の反復領域の「n」リピートを含み、m及びnは独立して2〜70の整数である繊維、並びに(b)生体適合性材料、細胞培地、細胞、又はそれらの任意の組み合わせを含む、組成物。
- 前記生体適合性材料が、組織足場、医療用装置、創傷ケア包帯、又は縫合糸である、請求項20に記載の組成物。
- 細胞遊走を増強するか、細胞増殖を増強するか、細胞死を阻害するか、細胞の接着を増強するか、又はそれらの任意の組み合わせの方法であって、異なる分子量のタンパク質の「m」型を含み、前記タンパク質の「m」型中の各タンパク質が、独立して、大瓶状腺スピドロイン(MaSp)タンパク質、又はその機能的相同体、バリアント、誘導体若しくは断片の反復領域の「n」リピートを含み、m及びnは独立して2〜70の整数である繊維を含む組成物と、前記細胞とを接触させることを含む、方法。
- 前記組成物が表面上に接着している、請求項22に記載の方法。
- 前記組成物が細胞培地と混合される、請求項22に記載の方法。
- 表面上の所定数の細胞層をアセンブルするための方法であって、異なる分子量のタンパク質の「m」型を含み、前記タンパク質の「m」型中の各タンパク質が、独立して、大瓶状腺スピドロイン(MaSp)タンパク質、又はその機能的相同体、バリアント、誘導体若しくは断片の反復領域の「n」リピートを含み、m及びnは独立して2〜70の整数である所定量の繊維を前記表面に適用することを含み、前記所定量が所定数の細胞層と相関する、方法。
- 前記所定数の細胞層が、1〜4枚の細胞層である、請求項23に記載の方法。
- 1層が1×103〜8×105の前記繊維/cm2を必要とする、請求項24に記載の方法。
- 2層が4×105〜18×105の前記繊維/cm2を必要とする、請求項24に記載の方法。
- 3〜4層が18×105の前記繊維/cm2を必要とする、請求項24に記載の方法。
- 前記方法が、インビトロ法又はエクスビボ法である、請求項25に記載の方法。
