Description
歯周再建のための活性以外に各抽出物の抗炎症効果を確認するために、歯肉の炎症及び結合組織の破壊と関連したインターロイキン−1(IL−1)と腫瘍壊死因子(TNF−α)との数値変化に及ぼす影響を調べた。RPMI 1640に10%のFBSが添加された培養培地を用いて、ヒト由来のTHP−1を培養した後、細胞株に、炎症誘発物質であるリポ多糖(Lipopolysaccharide、LPS)と本発明に係る各々の抽出物とを同時処理した後、培養液内TNF−αの分泌量を酵素免疫測定法(ELISA)により測定した。
歯周組織由来の幹細胞は、成体幹細胞として、一般に生体内で組織を維持し、損傷を再生する作用を呈する。したがって、抗炎症効果に最も優れたメタカラコウ抽出物を活用して、歯周組織の再建におけるメタカラコウ抽出物による歯周靱帯幹細胞への影響を確認するために、前記メタカラコウ抽出物が歯周靱帯幹細胞の増殖を促進する効果を調べる試験を行った。
歯周靱帯幹細胞の増殖は、KLF4、NOTCH1、MYC、及びPCNA遺伝子が非常に密接な影響を及ぼす。したがって、本発明に係るメタカラコウ抽出物による前記遺伝子の発現変化を確認するために、遺伝子mRNAの発現を確認した。その結果を図3〜6に示す。
本発明に係るメタカラコウ抽出物が、実際、組織の再生効果を発現することができるか否かを確認するために、スクラッチ損傷分析モデルを用いて、前記メタカラコウ抽出物に対する幹細胞の移動と、損傷した組織への転移能力とを試験した。その結果を図7に示す。
Claims (6)
- アザミ抽出物、ヒマワリ抽出物、タンポポ抽出物、キク抽出物、ゴボウ抽出物、コスモス抽出物、メタカラコウ抽出物、ヨモギ抽出物、チコリ抽出物、及びこれらの混合物からなる群より選択されたいずれか1つのキキョウ目に属する双子葉植物抽出物を含む、歯周組織の再生促進用組成物。
- アザミ抽出物、ヒマワリ抽出物、タンポポ抽出物、キク抽出物、ゴボウ抽出物、コスモス抽出物、ヨモギ抽出物、チコリ抽出物、及びこれらの混合物からなる群より選択されたいずれか1つのキキョウ目に属する双子葉植物抽出物、並びに
メタカラコウ抽出物
を含む、歯周組織の再生促進用組成物。 - 前記キキョウ目に属する双子葉植物抽出物は、水、炭素数1〜10の有機溶媒、亜臨界流体、超臨界流体、及びこれらの混合物からなる群より選択されたいずれか1つの溶媒にて抽出されてなる抽出物である、請求項1に記載の歯周組織の再生促進用組成物。
- 前記キキョウ目に属する双子葉植物抽出物は、水、炭素数1〜10の有機溶媒、亜臨界流体、超臨界流体、及びこれらの混合物からなる群より選択されたいずれか1つの溶媒にて抽出されてなる抽出物である、請求項2に記載の歯周組織の再生促進用組成物。
- 請求項1に記載の歯周組織の再生促進用組成物を含む、食品組成物。
- 請求項1に記載の歯周組織の再生促進用組成物を含む、薬学組成物。
