本出願は、神経発生及びグリア発生を促進する物質、及びそのような物質についてのアッセイの分野である。
間葉間質細胞は、間葉幹細胞として知られている多能性細胞集団を含む([1]に概説される)。大人の哺乳類における間葉幹細胞(MSC)の主要源は骨髄であり;骨髄から得られる多能性細胞は、間葉幹細胞(MSC)、骨髄付着間質細胞(MASC)、骨髄付着幹細胞及び骨髄間質細胞(BMSC)として多様に知られている。間葉幹細胞は、神経組織の修復のための有望な細胞療法として研究されてきた([2]に概説される)。神経系へのMSC又はMSC誘導体の移植は、脳卒中、パーキンソン病、脊髄損傷、多発性硬化症、及び新生児低酸素性虚血性脳損傷を包含する神経変性疾患の多くのモデルにおいて有益であることが示されてきた[3〜9]。
培養下で異なった神経細胞の増殖及び分化に影響を及ぼす因子の効果を定量化する能力を最適化する条件下で、神経細胞集団と、MSC及び/又はそれらの誘導体(例えば、SB623細胞)との共培養のためのインビトロシステムが本明細書に提供される。
1.神経発生を促進する物質についての試験方法であって、
(a)固体支持体上で間葉幹細胞(MSC)を培養し;
(b)MSCにより生成される細胞外マトリックスが支持体上に残るよう、支持体からMSCを除き;
(c)工程(b)の支持体上で胚性皮質細胞を培養し;
(d)工程(c)の培養物に物質を添加し;そして
(e)ニューロンの増殖を測定することを含んでなり、
ここで、ニューロンの増殖が、前記物質が神経発生を促進することを示唆する、方法。

種々の異なったタイプの神経細胞の増殖及び分化を支持するであろうインビトロ培養条件を確立することは困難であることが分かっている。本発明者は、神経前駆体細胞、ニューロン、星状細胞及びオリゴデンドロサイトがすべて増殖し、そして分化することができるインビトロ培養システムを考案した。従って、本明細書に開示される培養システムは最初に、神経細胞の増殖及び分化に対する試験物質の効果の定量的評価を可能にする。このシステムは、試験物質の存在下で細胞外マトリックス上での神経細胞(例えば、胚性齧歯類皮質細胞)の培養、続いて、1又は2以上のマーカー分子の発現についての神経細胞培養物の分析を包含する。それらのアッセイで使用される細胞外マトリックスは、(a)間葉幹細胞、又は(b)Notch細胞内ドメインをコードするが、しかし完全長Notchタンパク質はコードしない核酸によりトランスフェクトされた間葉幹細胞(例えば、SB623細胞)により生成される。

MSC及びSB623細胞の調製:
MSC及びSB623細胞の調製は記載されている[29]。手短に言えば、ヒト成人骨髄吸引物(Lonza, Walkersville, MD)を、10%ウシ胎児血清(FBS)(Hyclone, Logan, UT)、2mMのL−グルタミン及びペニシリン/ストレプトマイシン(両者とも、Invitrogen, Carlsbad, CAから)により補充されたαMEM(Mediatech, Herndon, VA)において増殖させた。第2継代上で、一部の細胞を低温保存し(MSC調製)、そして一部の細胞をSB623細胞の調製のためにプレートした。SB623細胞調製に関しては、MSCを、ヒトNotch1細胞内ドメイン(NICD)をコードするpCI−neo発現プラスミドによりトランスフェクトした。培養の1日後、トランスフェクトされた細胞を、7日間、G418(Invitrogen)による選択下に置き、その後、選択は解除され、そして培養物を、2度の継代により成長せしめ、そして増殖した。次に、SB623細胞を回収し、そしてCryostor CS5 (BioLife Solutions, Bothell, WA)を用いて低温保存した。3種の異なったドナーからの細胞を、本明細書に記載される研究に使用した。MSC及びSB623細胞を、解凍し、そして使用の前にαMEMで1度洗浄した。共培養実験に関しては、次に、細胞を、2%B27及び0.5mMのGlutaMAX(すべては、Invitrogenから)により補充されたNeurobasal培地から成る神経増殖培地に再懸濁した。ECMコーティングの生成又はならし培地(CM)の生成に関しては、細胞を、10%FBS及びペニシリン/ストレプトマイシンにより補充されたαMEMにプレートした。
ECMにより被覆されたウェルの調製のために、SB623細胞を、96−ウェルプレートに又はガラスカバースリップ(Fisher Scientific, Pittsburgh, PA)上に、3×104個の細胞/cm2でプレートし、それらを12−ウェルプレート(すべてのプレートは、Corning Inc, Corning, NYから購入された)に配置し、そして5日間、増殖した。続いて、培地を、血清を含まない培地に変え、そして細胞を、さらに2日間、培養した。次に、細胞を、いくらかの改良を伴って、前述のプロトコール[29]を用いて、ECMから除いた。手短に言えば、細胞を、室温で40分間、水中、0.2%Triton X-100(Sigma-Aldrich, St. Louis, MO)により処理し;次に細胞溶解物を注意して吸引し、そして水中、濃縮されたNH4OH(Sigma−Aldrich)の1:100(v/v)溶液を、5〜7分間、ゆっくりと添加し、次に除いた。洗浄のために、ウェルをPBSで完全に満たし、そして少なくとも3時間インキュベートした。ウェルは、すぐに使用されるか、又は4℃で保存された。
MSC又はSB623細胞を、3×104個の細胞/cm2でプレートし、そして10%FBS及びペニシリン/ストレプトマイシンにより補充されたαMEMにおいて3〜4日間、コンフルエントになるまで増殖した。次に、培地を、Neurobasal培地(Invitrogen)により置換し、そして培養物を1〜2時間、インキュベートした。この培地を捨て、そして細胞増殖のために典型的に使用される容量の半分を用いて、新鮮なNeurobasal培地により置換した。細胞を24時間インキュベートし、その後、培地を回収し、そして粒状物を、遠心分離により除去した。培地を分注し、そして−70℃で保存した。MSC−CM調製物を、使用の前に2%B27及び0.5mMのGlutaMAXにより補充した。
ラット胚性(E18)大脳皮質対を、BrainBits(Springfield, IL)から購入し;そして細胞懸濁液を記載のようにして調製した[29]。手短に言えば、皮質を、0.25%トリプシン/EDTAと共に37℃で5〜7分間インキュベートし、そしてトリプシンを除いた。組織を、10%FBSを含むαMEM、次にPBSにより洗浄した。次に、0.25mg/mlのDNase(MP Biomedicals, Solon, OH)を添加し、そして管の内容物を、30秒間、渦動により混合した。得られる細胞懸濁液を、粉末化し、PBSにより希釈し、ペレット化し、そして次に、神経増殖培地(上述の)に懸濁した。

様々な神経マーカーをコードするmRNAの発現の定量化のために、細胞の増殖及び培養を、96ウェルプレートにおいて実施した。示される時間の培養の後、培養培地を、10μlのピペット先端を備えたNunc Immuno Washerを用いて、注意して且つ完全に吸引し;そして細胞を、Cell-to-Signal(商標)(Applied Biosystems/Ambion, Austin, TX)又はSideStep(商標) (Agilent Technologies, Santa Clara, CA)の何れかの溶解緩衝液20μl(ウェル当たり)により3分間、溶解した。次に、溶解物を注意して上下にピペッティングし、そしてサンプル(四重反復下で)をペアで組み合わせて(従って、生物学的複製物を製造し)、保存プレートに移し、そして−70℃で凍結した。
神経細胞を、通常使用されるよりも10倍低いB27(0.2%)を含む神経増殖培地において、1.5×104個の細胞/cm2で、単独で、又は1.5×103個の細胞/cm2でMSC又はSB623細胞の何れかと一緒に、ECMで覆われたガラスカバースリップ上にプレートし、そして18〜24時間、増殖させた。次に、上記のようにして、培養物を、MAP2発現のために固定し、そして染色し、そしてDAPI含有培地をマウントした。約12〜15の倍率で、同じ露出時間を用いて、写真撮影し、そして条件当たり合計20のニューロンを分析した。最長の神経突起の長さを、各細胞について測定し、そして細胞当たりの神経突起の数を計数した。
間葉細胞の存在下又は不在下で増殖されたECMに基づく神経培養物の組成を、ネスチン、神経幹細胞/初期前駆体のマーカー及びMAP2、ニューロンマーカーについて、免疫細胞化学を用いて、最初に分析した。様々な時点で、培養物を固定し、そしてラット−特異的抗−ネスチン抗体及び抗−MAP2抗体と共にインキュベートした。すべての培養物をまた、核−特異的染料DAPIにより対比染色した。次に固定され、そして染色された培養物を、蛍光顕微鏡により検査した。1日目、単一陽性Nes+及びMAP2+細胞を、MAP2染色及びネスチン染色が共局在した、いくらかのMAP2+Nes+二重陽性細胞と共に、試験されたすべての培養物に存在した。小画分の二重陰性細胞もまた存在した。3日目までに、二重陽性細胞が検出されない一方で、単一陽性細胞はそれらの突起を拡張した。5日目に、MAP2+ニューロンは、神経突起を拡張し続ける一方で、Nes+細胞は数的に有意に増加した。この時点で、Nes+細胞はコロンーを形成した。より多くのコロニー数及びより大きなコロニーサイズが、間葉細胞の存在下で観察された。二重陽性細胞は検出されなかった。
星状細胞の成長を、グリア線維性酸性タンパク質(GFAP)の発現についての免疫組織化学分析によりアッセイした。グリア線維性酸性タンパク質(GFAP、星状細胞マーカー)及びネスチンについてのECMに基づく培養物の二重染色は、すべての培養において3日目の前、GFAP反応性の不在を示した。約5日目、GFAP−発現細胞が、単一又は二重陽性細胞としてのNes+コロニー内の共培養に観察されるようになった。GFAP−発現細胞は、間葉細胞を含まない培養において観察されなかった。別のコロニーは、可変比率のGFAP+Nes+細胞及びより十分に分化されたGFAP+Nes-細胞を有した。GFAP+細胞は、間葉細胞を欠いている培養においてはこの時点で検出されなかった。9日目、すべての3種の表現型(GFAP+Nes+、GFAP+Nes+及びGFAP+Nes-)が、すべての培養物に存在し、そしてGFAP+Nes-細胞が卓越した。
オリゴデンドロサイト発生を、初期オリゴデンドロサイトマーカー2’,3’−環状ヌクレオチド3’−ホスホジエステラーゼ(CNPase)の発現の免疫組織化学分析により評価した。CNPase+細胞は、共培養の9日目では、検出できなかった。12日目、間葉細胞の不在下でのECM上で増殖される培養においては、CNPase反応性は、非常に少数の、通常分裂する細胞においてのみ検出され得た。同じ時点で、間葉細胞を含む共培養においては、GNPase+細胞はクラスターで出現し、そしてCNPase発現は核周囲領域に局在した。SB623細胞による共培養においては、CNPase染色は、細胞質全体に、より強く、より広範囲に拡張した。CNPaseの発現は、ネスチン発現とは共局在しなかった。
神経細胞の増殖を、1.5×103個の細胞/cm2のMSCの存在下又は不在下で、1.5×104個の神経細胞/cm2を含む培養物において、2種の方法を用いてアッセイした。最初の方法によれば、DAPI染色された核が、上記のような免疫化学分析のために調製されたスライド上で計数された。200X倍率での5種の顕微鏡視野を、2つの実験から、計数し、そして条件ごとに平均した。非常に凝縮されるか、又は断片化された核は、死亡細胞を示すものとして考えられた。MSC核を、それらの独得のサイズに基づいて、計数から除外した。
この実験においては、ECM上で培養される神経細胞における種々のニューロン(ダブルコルチン、MAR2)、神経前駆体(ネスチン)及びグリアマーカー(GFAP及びCNPase)の発現の経時変化を、200個のMSC/ウェルの存在下及び不在下で分析した。サンプルを、様々な時点で集め、そして凍結した。続いて、全てのサンプルを融解し、そしてqRT−PCRにより並行してアッセイした。プライマー、プローブ及び増幅条件は上記の通りであった。結果を、図2に示す。神経マーカーダブルコルチン(DCX)及びMAP2のレベルは、最初は高く、そして培養及び共培養において時間と共に上昇する。中間の培養時で、共培養は、DCX及びMAP2 RNAレベルに対してはほとんど効果を有していないように見る一方で;後の時間では、共培養の活性効果が顕著になる。ネスチン、すなわち神経前駆体細胞マーカーの発現は、共培養の開始で、ほとんど検出できなかったが、しかし7日間の培養にわたって着実に高められ、そしてMSCの存在により増強された。GFAP mRNA、すなわち星状細胞マーカーの発現は、MSCとの共培養の不在下で検出されない一方で;共培養においては、それは、最初に、4日目で検出され、そして6日目と7日目との間で発現の大きな増加が存在した。CNPase mRNA、すなわちオリゴデンドロサイトマーカーの発現は、MSCとの共培養において、最初に4日目で検出され、そして6日目と7日目との間で急増が示された。
定量アッセイに関しては、ラット皮質細胞(5000個の細胞/ウェル)を、単独で培養するか、又はウェル当たり500〜32個の細胞の減少する数のMSCと共に共培養した。対照として、MSCはまた、500個の細胞/ウェルで単独で培養された。ラットネスチン、MAP2、GFAP及びCNPase mRNAについてのqRT−PCR Taqmanアッセイを用いて、遺伝子発現を定量化した。ヒト特異的グリセルアルデヒド3−リン酸デヒドロゲナーゼ(huGAP)のqRT−PCR Taqmanアッセイを用いて、MSC数を見積もった。図3及び4は、共培養サンプルにおけるrNes、rMAP2、rCNPase又はrGFAP遺伝子発現の総レベルが存在するMSCの数に直接的に依存することを示しており、ここで前記MSC数は、ヒト特異的GAP(huGAP)mRNAについてのアッセイにより定量化される。それらの効果は、MSC単独では、ラット特異的プローブ及び条件を用いてシグナルを付与しないので、ヒト配列の増幅により引き起こされなかった。
この実施例においては、神経細胞分化に対する生存MSCの効果を、それらのならし培地(CM)の効果に比較し;そして支持体としてのECMの使用の効果を、ポリ−D−リシンの使用と比較した。
上記実験においては、プレートのECMコーティングは、間葉細胞のコンフルエント層により生成され、ここで前記細胞の濃度は、共培養に使用される間葉細胞の最高濃度よりも約40倍、高かった。共培養における神経幹/初期子孫細胞増殖及び分化の強い刺激が共培養における間葉ECMの存在により引き起こされるかどうかを明確にするために、皮質細胞を、減少する数のMSC又はSB623細胞と共に混合し、そして共培養を、ポリ−D−リシン(PDL)により被覆されたプレートにおいて、非接着性条件下で実施した。対照として、皮質細胞を、単独で、FGF2及びEGFの存在下又は不在下でプレートし;そして間葉細胞を、共培養に使用される最高濃度で、単独でプレートした。
共培養における神経細胞の分化に対する接着条件の効果を評価した。このために、神経細胞を、SB623細胞由来のECM被覆されたプレート、オルニチン/フィブロネクチン−被覆されたプレート、及びUltra Low Attachment (ULA)プレート (Corning, Lowell, MA)上で、MSCと共に共培養した。オルニチン/フィブロネクチン−被覆されたプレートは、MSCの接着を支持した。ULAプレート上で、神経細胞を、5倍高い濃度のMSC(ECM又はオルニチン/フィブロネクチン上での共培養と比較して)、又は各20ng/mlの線維芽細胞増殖因子−2(FGF2)及び上皮増殖因子(EGF)の何れかと共に培養した。
前述の実施例に記載されるように、豊富なネスチン−発現細胞に対するECMの肯定的効果を考慮して、ネスチン発現に対するヘパリン硫酸プロテオグリカン成分の効果を調べた。
定量的RT−PCRを用いて、下記表4に示されるように、一定の増殖因子及びサイトカインをコードするmRNAのMSCによる発現を測定した。
ECM上で培養される神経細胞に、組換え因子を単独で、又は5%MSCならし培地(MSC−CM)と共に添加することにより、MSCにより生成される多くの増殖因子及びサイトカインを、神経発生及びグリア発生を刺激するそれらの能力について試験した。
線維芽細胞増殖因子−2(FGF2)は、MSCにより分泌され(上記表4)、そして皮質細胞へのFGF2の添加は、ネスチン、MAR2及びCNPaseの発現を刺激した(上記表5)。2種の追加の実験を行い、ネスチン発現の刺激におけるFGF2つの役割を確かめた。最初に、FGF2に対するブロッキング抗体を、神経細胞及びMSCの共培養物に添加した。第二に、MSCならし培地からFGF2を枯渇し、そしてそれを、神経細胞培養物に添加した。
間葉幹細胞の効果を、ECM被覆プレート上での神経細胞培養におけるネスチン及びGFAPの発現に対する間葉幹細胞からのならし培地の効果と比較した。図12に示されるように、類似するレベルのネスチンmRNAが、ウェル当たり200のMSCにより、及び10%MSCならし培地により誘導された。しかしながら、ならし培地によるGFAP発現の誘導は、細胞自体により誘導されたその発現の誘導よりも低かった。この結果は、星状細胞成長を担当できる成分が、MSC自体におけるよりもMSCならし培地においてあまり豊富ではなかった(そして/又はあまり活動的ではなかった)ことを示唆した。
ECM上で、Nes+細胞の増殖は、免疫染色及びqRT−PCRを用いて示される場合、生存間葉細胞又はそれらのならし培地により、用量依存性的に有意に拡大された一方で、PDL上で、それらの因子に対する応答は低められた(図1及び5、並びに表2)。これは、Nes+幹/初期前駆体細胞の増殖が分泌された間葉細胞−由来の因子により刺激され、そして間葉細胞ECM上での増殖により相乗的に増強されたことを示唆する。この相乗効果のための最も可能性が高い機構は、マトリックスプロテオグリカンによる神経細胞への間葉細胞由来の増殖因子の効果的蓄積、保存及び提供である。神経Nes+細胞の増殖が離れて作用するMSC−由来の可溶性因子により刺激され得る結論は、最近の報告と一致しており、この報告は、マウスMSCと共に共培養されたが、しかし半透膜によりそれらから分離されたマウスニューロスフェアは、高い百分率のKi−67−陽性細胞を有したことを示している[38]。実際、MSCは、インビボでの神経前駆体の維持に関与していることが示されている多くの増殖因子を分泌することが知られており[16、30]、そしてBMP4、FGF2、EGF、VEGF及びPDGF−AAを包含する、それらのいくつかの分泌が本明細書で使用されるMSC及びSB623細胞バッチにおいて確認されている[39]。

Claims (23)
- スクリーニングデバイスを製造するための方法であって、
(a)固体支持体上で間葉細胞を培養し;そして
(b)間葉細胞により生成される細胞外マトリックスが支持体上に残るよう、支持体から間葉細胞を除くことを含み、
前記スクリーニングデバイスは、神経発生を促進する、グリア発生を促進する、神経前駆体細胞の増殖を促進する、又は神経前駆体細胞の分化を促進する物質についての定量的アッセイに用いられる、前記方法。 - 前記間葉細胞が、
(a)間葉幹細胞、及び
(b)Notch細胞内ドメインをコードする核酸によりトランスフェクトされている間葉幹細胞の子孫、
から成る群から選択される、請求項1に記載の方法。 - 前記間葉細胞がヒトから得られる、請求項1又は2に記載の方法。
- 前記固体支持体が、プラスチック、ニトロセルロース及びガラスから成る群から選択される、請求項1〜3の何れか一項に記載の方法。
- 胚性皮質細胞を細胞外マトリックス上で培養することを更に含む、請求項1〜4の何れか一項に記載の方法。
- 前記胚性皮質細胞がマウス又はラットから得られる、請求項5に記載の方法。
- 試験物質をスクリーニングデバイスに接触させることを更に含む、請求項5又は6に記載の方法。
- 前記試験物質が化合物又はポリペプチドである、請求項7に記載の方法。
- 前記試験物質が、試験細胞、試験細胞培養物、又は細胞培養物からの試験ならし培地である、請求項7に記載の方法。
- 前記試験細胞が、
(a)間葉幹細胞、及び
(b)Notch細胞内ドメインをコードする核酸によりトランスフェクトされている間葉幹細胞の子孫、
から成る群から選択される、請求項9に記載の方法。 - 前記皮質細胞の増殖及び/又は分化が測定される、請求項7〜10の何れか一項に記載の方法。
- 前記皮質細胞からニューロンへの分化が測定される、請求項11に記載の方法。
- 前記皮質細胞から星状細胞への分化が測定される、請求項11に記載の方法。
- 前記皮質細胞からオリゴデンドロサイトの分化が測定される、請求項11に記載の方法。
- 物質が、神経発生を促進する、グリア発生を促進する、神経前駆体細胞の増殖を促進する、又は神経前駆体細胞の分化を促進するか否かについて決定するための該物質の試験に使用するための組成物であって、
前記組成物は、固体支持体及びその上に接着される生物学的層を含み、前記生物学的層が、
(a)間葉幹細胞(MSC)、又は
(b)核酸によりトランスフェクトされているMSCであって、ここで、前記核酸がNotch細胞内ドメインをコードするが、しかし完全長Notchタンパク質はコードしない、MSC
により接着される細胞外マトリックスであり、
前記組成物は、神経発生を促進する、グリア発生を促進する、神経前駆体細胞の増殖を促進する、又は神経前駆体細胞の分化を促進する物質についての定量的アッセイに用いられる、前記組成物。 - 前記MSCがヒトから得られる、請求項15に記載の組成物。
- 前記固体支持体が、プラスチック、ニトロセルロース及びガラスから成る群から選択される、請求項15又は16に記載の組成物。
- 胚性皮質細胞をさらに含む、請求項15〜17の何れか一項に記載の組成物。
- 前記胚性皮質細胞がマウス又はラットから得られる、請求項18に記載の組成物。
- 試験物質をさらに含む、請求項15〜19の何れか一項に記載の組成物。
- 前記試験物質が化合物又はポリペプチドである、請求項20に記載の組成物。
- 前記試験物質が、細胞、細胞培養物、又は細胞培養物からのならし培地である、請求項20に記載の組成物。
- 前記細胞が、
(a)間葉幹細胞、及び
(b)Notch細胞内ドメインをコードする核酸によりトランスフェクトされている間葉幹細胞の子孫、
から成る群から選択される、請求項22に記載の組成物。
