Description
−ネスチン+:発生中の多くの細胞および特に神経堤の細胞によって発現される中間径フィラメントタンパク質(この発現は、一過性であり、成人期になっても続くものではない)、
−OCT4+(八量体結合転写因子4):未分化胚性幹細胞の自己複製に関与する多分化能マーカー、
−SOX2+(性決定領域Y−ボックス2):未分化胚性幹細胞の自己複製の維持に不可欠な転写因子
を発現している細胞として定義される。
−その相乗作用的抽出物は、皮膚細胞において、皮膚幹細胞(SKP)の「幹」の特徴に関連する生化学的マーカー、特にネスチン+、OCT4+、SOX2+の量を増加させ;
−その相乗作用的抽出物は、皮膚の弾性、皮膚の柔軟性および/または皮膚の硬度を高め、
−その相乗作用的抽出物は、皮膚の再生を増加させ;
−その相乗作用的抽出物は、皮膚細胞外マトリックスのタンパク質、例えば、コラーゲン、特にコラーゲンI、IIIまたはV;プロコラーゲン、特にプロコラーゲンI、IIIまたはV;およびトロポエラスチンの合成を増加させ;
−その結果、その相乗作用的抽出物は、皮膚老化に関連する皮膚の徴候に対抗することが可能である。
−線維芽細胞によって発現されるSOX2+のメッセンジャーRNAの量を少なくとも8倍増加させること、および/または
−線維芽細胞によって発現されるネスチン+のメッセンジャーRNAの量を少なくとも2倍増加させること、および/または
−線維芽細胞によって発現されるOCT4+のメッセンジャーRNAの量を少なくとも6倍増加させること
が可能であり得る。
−ある量のPalmaria palmataを、10/1〜50/1という水とPalmaria palmata(乾燥重量)との重量比で水に溶解する工程;
−Palmaria palmataの水溶液を40〜80℃の温度においてカルボヒドラーゼおよびエンドプロテアーゼで加水分解する工程;
−ジャスミン花頭を、先に得られたPalmaria palmataの水性抽出物中で浸軟する工程(ここで、その藻類の乾燥重量と花頭の乾燥重量との重量比は、40/60〜95/5である);
−浸軟され得られた生成物を濾過し、次いで、2〜24時間にわたって、40〜90℃の温度で加熱することにより、カルボヒドラーゼおよびエンドプロテアーゼ酵素を不活性化する工程。
a)乾燥され、フレークの形態に細かく粉砕された、ある量のPalmaria palmataを、10/1〜50/1、好ましくは、20/1〜40/1という水とPalmaria palmataとの重量比で水に溶解する工程、
b)Palmaria palmataの水溶液をカルボヒドラーゼおよびエンドプロテアーゼで加水分解する工程であって、好ましくは、キシラナーゼおよびブロメラインを用いて、3〜6、好ましくは、4〜5.5、さらにより好ましくは、4〜4.5のpH、40〜80℃、好ましくは、50〜60℃、さらにより好ましくは、55℃の温度で、少なくとも1時間、好ましくは、2時間にわたって行われる工程;
c)必要に応じて濾過助剤の添加および遠心分離を行った後、Palmaria palmataの水性抽出物を得る工程;
d)乾燥されたジャスミン花頭を、少なくとも2時間かつ長くとも4時間にわたって、外界温度において、工程c)で得られたPalmaria palmataの水性抽出物中で浸軟する工程(ここで、その藻類の乾燥重量と花頭の乾燥重量との重量比は、40/60〜95/5であり、好ましくは、その藻類の乾燥重量と花頭の乾燥重量との重量比は、90/10に等しい);
e)工程d)においてそのようにして得られた浸軟された生成物を濾過することにより、Palmaria palmataおよびジャスミン花頭の抽出物を回収する工程であって、少なくとも2時間かつ最長24時間、好ましくは、12時間または一晩にわたって、40〜90℃、好ましくは、80℃の温度において、加熱することにより、カルボヒドラーゼおよびエンドプロテアーゼ酵素を不活性化する工程;および
f)それを必要に応じて濾過によって精製することにより、Palmaria palmataとジャスミン花頭との相乗作用的抽出物を得る工程。

−ビタミンB3、より詳細には、ニコチンアミド、ニコチン酸トコフェロール、
−ビタミンB5、ビタミンB6、ビタミンB12、パンテノール、
−ビタミンC、特に、アスコルビン酸、アスコルビルグルコシド、テトラパルミチン酸アスコルビル、リン酸アスコルビルマグネシウムおよびリン酸アスコルビルナトリウム、

−植物ペプチド抽出物、例えば、ダイズ、スペルトコムギ、ブドウ、ナタネ、アマ、イネ、トウモロコシまたはエンドウマメの抽出物、
−酵母抽出物、Artemia salinaの抽出物、
−デヒドロ酢酸(DHA)、
−合成または天然起源のフィトステロール、
−サリチル酸およびその誘導体、アルファ−およびベータ−ヒドロキシ酸、シラノール、
−アミノ糖、グルコサミン、D−グルコサミン、N−アセチル−グルコサミン、N−アセチル−D−グルコサミン、マンノサミン、N−アセチルマンノサミン、ガラクトサミン、N−アセチルガラクトサミン、
−ポリフェノール、イソフラボン、フラボノイドの抽出物、例えば、ブドウの抽出物、マツの抽出物、オリーブの抽出物、
−脂質(例えば、セラミドまたはリン脂質)、動物起源の油(例えば、スクアレンまたはスクアラン);植物油(例えば、甘扁桃油、コプラ油、ひまし油、ホホバ油、オリーブ油、菜種油、落花生油、ヒマワリ油、麦芽油、トウモロコシ胚芽油、ダイズ油、綿実油、アルファルファ油、ケシ油、カボチャ油、月見草油、キビ油、オオムギ油、ライムギ油、ベニバナ油、トケイソウ油、ヘーゼルナッツ油、パーム油、杏仁油、アボカド油、キンセンカ油);エトキシ化植物油、シアバター、
−UVスクリーンおよび日焼け止め。
20gのJasminum officinaleの乾燥した花頭を1kgの水に入れ、撹拌しながら外界温度で2時間浸軟する。次いで、固体残渣を廃棄し、抽出物を清澄化するために、多孔度を低下させた(20〜50μmから0.3〜0.5μmまで)フィルターを用いる連続的な濾過を行う。
50gのフレークの形態のPalmaria palmataを1kgの水に溶解する。反応混合物に添加した2gのキシラナーゼおよび1gのブロメラインを用いて、加水分解を行う。次いで、反応混合物を55℃で2時間加熱する。濾過工程により、固体残渣を廃棄することおよび炭水化物が豊富な濾液だけを維持することができる。このために、10g/kgのCelatom(登録商標)(遠心分離助剤)を添加し、溶液を4000rev/分で10分間遠心した。遠心分離の後、残留固体を廃棄し、次いで、上清を、7〜20μmまで多孔度を低下させたフィルターによる濾過によって清澄化し、80℃で一晩維持する。翌日、抽出物を、0.3〜0.5μmまで多孔度を低下させたフィルターで再度濾過する。得られた抽出物は、30±2g/kgの乾物を特徴とする。次いで、抽出物を水で希釈して、20±2g/kgの乾物を得る。次いで、pHを4〜4.5に調整する。
結果は、図1に示されているように、無処理コントロールと比べて、1%のPalmaria palmataの参照抽出物で処理された線維芽細胞において、SOX2、ネスチンおよびOCT4のmRNA発現のそれぞれ29%、26%および34%という統計学的に有意な増加を示す。
実施例1に従って得られた相乗作用的抽出物で前処理された線維芽細胞では、無処理コントロール細胞、ジャスミンの参照抽出物および藻類の参照抽出物と比べて、SOX2、ネスチンおよびOCT4のより高いmRNAレベルが観察される。実施例1に従って得られた相乗作用的抽出物は、皮膚細胞の「幹」の特徴に特徴的なマーカーの相乗作用的な増加をもたらす。
結果は、図2に示されているように、SKP細胞を含まない無処理コントロールと比べて、SKP細胞を含む無処理の真皮等価物において、コラーゲンIおよびコラーゲンIIIの新しく合成されたタンパク質の発現の18%および23%という有意な増加を示した。
SKP皮膚幹細胞を含む真皮等価物における、実施例1に従って得られた1%の相乗作用的抽出物の適用は、プロコラーゲンIおよびコラーゲンIIIの発現を刺激する。
顕微鏡法による観察(倍率*40)は、実施例1に従って得られた相乗作用的抽出物を適用した条件において、無処理と比べて、SOX2、OCT4およびネスチン標識の強度の増加を示した。図7は、SOX2およびネスチン標識に対して得られた写真を再現している。
SKP様細胞における、実施例1に従って得られた1%の相乗作用的抽出物の適用は、SOX2、OCT4およびネスチン幹細胞マーカーの発現を刺激する。
Claims (14)
- 藻類Palmaria palmataとJasminum属の植物の花頭との相乗作用的抽出物であって、前記抽出物は、i)藻類Palmaria palmataの水性抽出物を調製する工程、続いて、ii)Jasminum属の植物の花頭を前記水性抽出物中で浸軟する工程を含む方法によって入手可能であり、該藻類の乾燥重量と該花頭の乾燥重量との重量比は、40/60〜95/5であり、該藻類と該花頭の両方が、該相乗作用的抽出物を調製するための原材料として使用される、相乗作用的抽出物。
- 前記藻類の乾燥重量と前記花頭の乾燥重量との重量比が、8/1〜10/1であることを特徴とする、請求項1において特許請求された抽出物。
- 前記Jasminum属の植物が、Jasminum grandiflorum、Jasminum officinale、Jasminum odoratissimum、Jasminum sambac、Jasminum auriculatum、Jasminum flexile、好ましくは、Jasminum officinale種から選択されることを特徴とする、前述の請求項のうちの1項に記載の抽出物。
- 前記花頭が、乾燥した花頭全体であることを特徴とする、前述の請求項のうちの1項に記載の抽出物。
- 工程i)において、前記Palmaria palmataの水性抽出物が、カルボヒドラーゼおよびエンドプロテアーゼを用いる酵素加水分解、次いで、濾過および/または遠心分離によって得られることを特徴とする、前述の請求項のうちの1項に記載の抽出物。
- 前記カルボヒドラーゼが、キシラナーゼ、好ましくは、前記藻類の乾燥重量の2〜6wt%、好ましくは、4%の濃度で使用されるキシラナーゼであること、および前記エンドプロテアーゼが、該藻類の乾燥重量の1〜3重量%、好ましくは、2重量%の濃度で使用されるブロメラインであることを特徴とする、請求項5において特許請求された抽出物。
- 26.8〜30.8g/kgの乾物含有量、1.3〜2.3g/kgのタンパク質含有量および25.3〜29.3g/kgの糖類含有量を有することを特徴とする、前述の請求項のうちの1項に記載の抽出物。
- 工程ii)の終わりに、水、グリセロール、エタノール、プロパンジオール、ブチレン グリコール、ジプロピレングリコール、エトキシル化もしくはプロポキシル化ジグリコール、環状ポリオールまたはこれらの溶媒の任意の混合物から選択される1種以上の生理的に許容され得る溶媒に希釈されることを特徴とする、請求項7において特許請求された抽出物。
- 請求項1〜8のうちの1項において特許請求されたPalmaria palmataとJasminum属の植物の花頭との相乗作用的抽出物を得る方法であって、該方法は、以下の工程:
a)ある量の乾燥され細かく粉砕されたPalmaria palmata藻類を、10/1〜50/1、好ましくは、20/1〜40/1という水とPalmaria palmataとの重量比で水に溶解する工程、
b)Palmaria palmataの該水溶液を、3〜6、好ましくは、4〜5.5、さらにより好ましくは、4〜4.5のpHにおいて、40〜80℃、好ましくは、50〜60℃、さらにより好ましくは、55℃の温度において、少なくとも1時間、好ましくは、2時間にわたってカルボヒドラーゼおよび/またはエンドプロテアーゼで加水分解する工程;
c)必要に応じて濾過助剤の添加および遠心分離を行った後、Palmaria palmataの水性抽出物を得る工程;
d)Jasminum属の植物の乾燥した花頭を、少なくとも2時間かつ長くとも4時間にわたって、外界温度において、工程c)において得られたPalmaria palmataの水性抽出物中で浸軟する工程であって;該藻類の乾燥重量と該花頭の乾燥重量との重量比は、40/60〜95/5である、工程、
e)工程d)におけるように得られた浸軟生成物を濾過して、Palmaria palmataおよびJasminum属の植物の花頭の抽出物を回収し、それを少なくとも2時間かつ最大24時間、好ましくは、12時間または一晩にわたって、40〜90℃、好ましくは、80℃の温度で加熱して、該カルボヒドラーゼおよびエンドプロテアーゼ酵素を不活性化する工程;および
f)それを必要に応じて濾過によって精製して、Palmaria palmataとJasminum属の植物の花頭との相乗作用的抽出物を得る工程
を含むことを特徴とする、方法。 - 請求項1〜8のうちの1項において特許請求されたまたは請求項9に記載の方法によって得られる、Palmaria palmataとJasminum属の植物の花頭との相乗作用的抽出物を生理的に許容され得る溶媒中に含む、化粧品組成物。
- Palmaria palmataとJasminum属の植物の花頭との前記相乗作用的抽出物を、前記組成物の総重量の0.0001%〜20%乾燥重量の濃度、好ましくは、該組成物の総重量の0.05%〜5%乾燥重量の濃度で含むことを特徴とする、請求項10において特許請求された組成物。
- ビタミンA、レチノイン酸、レチノール、ビタミンB3、ビタミンB5、ビタミンB6、ビタミンB12、ビタミンC、ビタミンE、ビタミンF、ビタミンH、ビタミンK、ビタミンPP、コエンザイムQ10、メタロプロテイナーゼ阻害剤、アミノ酸、カルニチン、カルノシン、タウリン、天然または合成ペプチド、植物ペプチド抽出物、酵母抽出物、Artemia salinaの抽出物、合成または天然起源のフィトステロール、サリチル酸、オリゴ糖、多糖、アミノ糖、ポリフェノール、フラボノイド、脂質、リン脂質、動物起源の油、植物油、エトキシ化植物油、UVスクリーンおよび日焼け止めから選択される少なくとも1つのさらなる活性な作用物質をさらに含むことを特徴とする、請求項10または11のうちの1項において特許請求された組成物。
- 成体の皮膚幹細胞(SKP)の「幹」の特徴の維持を支持することによって、皮膚老化の徴候に対抗するための、請求項1〜8のうちの1項において特許請求されたPalmaria palmataとJasminum属の植物の花頭との相乗作用的抽出物または請求項10〜12のうちの1項において特許請求された組成物の美容的使用。
- 皮膚の弾性、柔軟性および/または硬度を改善するための、請求項13において特許請求された使用。
