材料及び方法
材料
Egg Bio Chemica(ドイツ国ダルムシュタットのAppliChem GmbH)製レシチン;コレステロール(ドイツ国タウフキルヘンのSigma Aldrich);1,2−ジパルミトイル−sn−グリセロ−3−ホスホチオエタノール(チオール修飾レシチン;米国アラバマ州のAvanti(商標) polar lipids);以下に記載されるようにS.アシドカルダリウスから単離したテトラエーテル脂質(TEL);ダルベッコリン酸緩衝生理食塩水(英国のlife technologies(商標)によるgibco(商標));ガラスビーズ(0.75mm〜1.0mm;カールスルーエのCarl Roth GmbH + Co. KG);NAP(商標)−5カラム(英国バッキンガムシャーのGE Healthcare);Triton(商標)X−100(ドイツ国タウフキルヘンのSigma Aldrich);クロロホルム(ドイツ国タウフキルヘンのSigma Aldrich);メタノール(ドイツ国タウフキルヘンのSigma Aldrich);Antra MUPS(オメプラゾール、ヴェーデルのAstra Zeneca GmbH);シリカゲル60(0.063mm〜0.200mm、ドイツ国ゲルンスハイムのMerck);放射性ヨウ素(米国ボストンのPerkin Elmer(商標))。
当該技術分野において知られている通りに細胞の生育及び脂質抽出を行った。S.アシドカルダリウスを培地から分離し、Christ製のDelta 1−20 KDを使用して凍結乾燥した。当該技術分野において知られている通りにクロロホルム/メタノール(2:1)を用いるソックスレー抽出によって脂質を単離した。抽出した溶媒をロータリーエバポレーション(Rotavapor−R、スイス国フラヴィルのBuchi Labortechnik AG)によって除去した。その後、脂質混合物をクロロホルム、メタノール及び塩酸(8:3:1)の混合物に溶解した。脂質頭部基を開裂するために該混合物を3日間加熱した。最後に、脂質を水相からクロロホルム/メタノール(2:1)で抽出した。グリセリルカルジチルテトラエーテル脂質(GCTE)は第1の溶離液(カラムの予備洗浄用)として水/メタノール(1:1)でシリカゲルカラムクロマトグラフィーにより分離した後、望ましくない脂質及びメタノール/クロロホルム(1:1)を除去するために水/メタノール/クロロホルム(1:2.5:1)によって分離し、GCTE画分を得た。
1.CPPペネトラチンの合成
Applied Biosystems 433Aペプチド合成機でフルオレニルメトキシカルボニル/t−ブチル(Fmoc/tBu)化学を使用し、固相合成によって、ペネトラチンを産生した。カップリング条件の残りは記載の通り行う。Reprosil(商標)Gold 120C−18カラム(4μm、150mm×20mm)を備えるLaPrep P110(VWR International)HPLCシステムで精製を行った。0.1%のTFAを含む水及びアセトニトリルを、20ml/分の流速で溶離液として使用した。分離条件は15分で60%〜90%のアセトニトリルの直線濃度勾配であった。溶離液として、水中の0.1%トリフルオロ酢酸(TFA)(溶離液A)及びアセトニトリル中の0.1%TFA(溶離液B)を使用した。合成したペプチドのアイデンティティーは、HPLC−MS(質量分析)分析(Exactive、Thermo Fisher Scientific)によって検証した。
DCM/DMSOの2:1混合物中のチオール修飾リン脂質の1mMの溶液5mlに対して、200μlのDMSOに予め溶解した20mgのペネトラチン−Cysを添加した。反応混合物を室温で一晩撹拌した。溶媒を蒸発させ、粗生成物を20mlのACN/H2Oの4:1混合物に溶解した。Reprosil(商標)Gold 120C−18カラム(4μm、150mm×20mm)を備えるLaPrep P110(VWR International)HPLCシステムで精製を行った。0.1%のTFAを含む水及びアセトニトリルを20ml/分の流速で溶離液として使用した。Chromolithe(商標) Performance RP−C18eカラム(100mm×4.6mm)を備えるAgilent 1100 HPLCシステムで分析的分析を行った。0.1%のTFAを含む水及びアセトニトリルを2ml/分の流速で溶離液として使用した。Waters(商標)Hypersil Gold aqカラム(200mm×2.1mm)を備えるAgilent 1200 HPLCシステム、続いてThermo Scientific Exactive質量分析計でHPLC−MS分析を行った。0.05%のTFAを含む水及びアセトニトリルを200μl/分の流速で溶離液として使用した。
131I放射能標識化のため、PBS中にモデル物質であるバンコマイシンの1mmol原液を用意した。必要量の放射性ヨウ素−131を25μlの原液に添加し、当該技術分野において知られているクロラミンT法を使用して標識化を行った。当該技術分野において知られている半分取(semi-preparative)HPLCによって反応混合物を精製した。その後、5分以内;流量2ml/分;UV吸光度λ=214;γ−検出で、アセトニトリル中の0.1%TFA(溶離液B)に対する水中の0.1%TFA(溶離液A)の直線勾配を適用して、Chromolithe(商標)Performance RP−C18(100mm−3mm)カラムを使用する放射性HPLC(Agilent 1100シリーズ)によって放射能標識の純度を特定した。
SpeedMixer(商標)(DAC150FVZ ドイツ国ハムのHauschild Engineering GmbH & Co. KG)を使用するDAC法によりリポソームを作製した。最初に、脂質をクロロホルム/メタノール9:1に溶解して100mmolの原液を得る一方で、クロロホルム/メタノール1:1(1mmol原液)にCPPを溶解した。必要量のCPP原液(0.1モル%〜1.0モル%)を脂質混合物(85モル%のEPC、10モル%のコレステロール及び5モル%のTEL)に添加し、窒素流(nitrogen stream)によって有機溶媒を蒸発させた。その後、得られた脂質フィルムを真空チャンバーで1時間乾燥した。スピード混合プロセスを開始する前に、20mgの0.075mm〜1.00mmのガラスビーズを添加した。スピード混合、及び総容積250μl(表1を参照されたい)までの3回の実行で種々の量の組み込み物質(例えば、バンコマイシン−PBSに溶解した1mmol原液−;1回目)又はPBS(2回目/3回目)の添加によって、リポソームを作製した。比較のため、標準リポソーム(85モル%のEPC;15モル%のコレステロール)を同じように作製した。
5分以内;流量2ml/分;UV吸光度λ=214nmで、アセトニトリル中の0.1%TFA(溶離液B)に対する水中の0.1%TFA(溶離液A)の直線勾配を適用して、C18カラム(Chromolithe(商標)Performance RP−18e、100mm−3mm)を使用する逆相(rp)HPLC(Agilent 1100シリーズ)によってバンコマイシンの封入効率を特定した。スピード混合プロセスの後、リポソームを100μlずつ2つの部分に分割した。50μlの10%Triton(商標)X−100の添加によってリポソームを破壊し、rpHPLCによってバンコマイシンのAUCを特定することにより、部分1(Part 1)を使用して100%の値を計算した。部分2をSephadex G−25ゲル濾過クロマトグラフィー(NAP(商標)−5カラム)によって精製した後、部分1と同じように処理した。封入効率E(%)を、部分2の希釈の補正後に以下の方程式によって特定した。

1.粒子径、多分散性指数(PDI)及びゼータ電位
全てのリポソーム製剤の粒子径、PDI及びゼータ電位を、Malvern(商標)(英国ウースタシャーのMalvern Instruments Ltd.)製Zetasizer Nano ZSを使用して、室温で特定した。サイズ及びPDIをPBSで0.076mg/mlの濃度に希釈した後に測定し、一方、ゼータ電位を50mmolリン酸バッファーによって0.95mg/mlの濃度に希釈した後に特定した。測定は自動モード及び3回の測定の平均を使用して行った。サイズをnmで、ゼータ電位をmVで明示するが、PDIは無次元値である。
当該技術分野で知られているように、CPP−TEL−リポソームのサイズ及びラメラ構造を特定するため、試料を2/2 QuantifoilグリッドでFEI Vitrobotを使用して凍結した。その後、各サンプルを3秒間グロー放電させ、8秒間〜10秒間4℃及び100%湿度でブロットした。グリッドを200kV及び液体窒素温度にて操作したクライオス(Krios)顕微鏡で観察した。CPP−リポソーム試料の写真を倍率64000倍で撮影した。
1.種々のモル比で凍結乾燥保護剤としてショ糖を使用する凍結乾燥
全てのリポソーム製剤をChrist製のDelta 1−20 KDで凍結乾燥した。主な乾燥を−20℃で2日間行い、続いて少なくとも6時間に亘って0℃で二次乾燥を行った。以前より、ショ糖が良好な凍結乾燥保護剤であることがわかっていたため、100nm〜500nmの範囲で凍結乾燥保護剤としてショ糖を使用した。簡潔には、3回の実行の間にPBSバッファーに代えて所望の濃度のPBD中のショ糖を添加する点で異なるが、上に記載されるようにリポソームを作製した。リポソーム懸濁液をエッペンドルフ管1本当たり50μlに分割して凍結乾燥した。凍結乾燥リポソームを50μlのPBSによって再水和し、サイズ及びPDIを上に記載されるMalvern(商標)製Zetasizer Nano ZSを使用して特定した。
90秒で120℃まで加熱することにより凍結乾燥されたリポソーム100mgを使用して、水分計(ドイツ国バーリンゲンのKern&Sohn)によってCPP−TEL−リポソームの残留水分を特定した。
1.種々の製剤の131I標識化バンコマイシンの血液レベル
体重約220g〜250gの雄性ウィスターラットを用いて、地方自治体(local authorities)に従い、動物研究を行った。種々の製剤の血液レベルを特定するため、バンコマイシンを131Iによって放射能標識化し、幾つかのリポソーム製剤に組み込んだ。経口投与から1時間後のバンコマイシンの血液レベルを、標準と比較して、Berthold LB 951 Gカウンターを使用して、放射能を直接計数することにより測定した。簡潔には、5群(n=6)のウィスターラットを形成した。プロトンポンプ阻害剤であるオメプラゾールによる前処理がリポソーム中へのプロトンの拡散を減少させ、結果的に胃のpHを引き上げることによって封入された薬剤の変性を減少させることが最近の研究により示されたため、実験の前日に強制投与によって懸濁したAntra MUPS(商標)(オメプラゾール)(ラット1匹当たり10mg)でラットを前処理した。実験前12時間に亘り給餌せず、自由飲水でラットを維持した。経口適用を強制投与によって実施した。1時間後にラットを屠殺し、血液試料を採取して計量し、血液試料中の放射能を、標準と比較して、Berthold LB 951 Gカウンターを使用して測定した。血液関連活性は総注入用量(ID)と関連し、組織1グラム当たりの総注入用量の割合(%ID)として表した。その後、総血液レベル(%ID)を計算した。
放射性血液試料の測定によって得られた値を検証するため、CPP−TEL−バンコマイシン−リポソーム製剤のバンコマイシン免疫アッセイ(米国タリータウンのSiemens、ADVIA Centaur(商標)VANC ReadyPack(商標))を行った。放射能標識化による研究と同様、500μlのCPP−TEL−バンコマイシン−リポソームを強制投与によって250gの雄性ウィスターラット(n=6)に投与した。実験の前日、強制投与によって懸濁したAntra MUPS(商標)(オメプラゾール)(ラット1匹当たり10mg)でラットを前処理した。経口投与の1時間後、ラットを屠殺し、血液試料を得て、血漿を分離し、バンコマイシンの量を上に記載される免疫アッセイによって特定した。
TELの単離
単離処理は、質量分析及び薄層クロマトグラフィーによって特定される脂肪親和性鎖中のペンチル環の数がほんのわずかに変化する(3個〜5個)精製形態のテトラエーテル脂質をもたらした。環の数は古細菌の培養中の温度によって影響を受ける。400gの湿質量の細胞(wet cell mass)当たり、平均1gのテトラエーテル脂質を得ることができた。
CPP−リン脂質−複合体の合成及び特性評価
質量分析によって特定されるように、高純度でCPP(ペネトラチン)−リン脂質−複合体を得ることができた(図5)。生成物の全収率は分取HPLCによる精製の後、約59%であった。
バンコマイシンの131I放射能標識化
バンコマイシンの131I放射能標識化は、放射性HPLCによって特定されるように高純度(95%超)の所望の生成物をもたらした(図6)。クロラミンT法を使用する標識化効率は、適用された放射能の約60%であった。
封入効率
CPP−TEL−バンコマイシン−リポソームは、標準リポソームの特定された値(38.73±1.23%)に匹敵する35.38±1.65%の封入効率を示した。
粒子の特性評価
1.粒子径、多分散性指数(PDI)及びゼータ電位
応用DAC法は、サイズ、PDI及び組み込み効率(表2を参照されたい)において高い均一性を有するCPP−TEL−バンコマイシン−リポソームをもたらした。標準リポソームと比較して、サイズ及びPDIはわずかな増加を示したのに対し、CPP−TEL−バンコマイシン−リポソームは標準リポソームより高い正のゼータ電位を特徴とする。
クライオ電子顕微鏡写真(図7)は、85モル%レシチン;10モル%コレステロール;5モル%TEL及び0.1モル%ペネトラチンを含むCPP−TEL−バンコマイシン−リポソームのサイズ及びラメラ構造を示す。撮影されたリポソームの大半は、単層から最大3層までのラメラ層をそれぞれ示す。
長期保存安定性
1.種々のモル比で凍結乾燥保護剤としてショ糖を使用する凍結乾燥
凍結乾燥保護剤として種々のモル比でショ糖を含むCPP−TEL−リポソームを凍結乾燥することは、凍結乾燥プロセスの前に測定したデータと比較して、或る特定のモル比について匹敵するサイズ及びPDIをもたらした(図8A、図8B)。このリポソーム製剤に関し、ショ糖の下限は300mmolであるが、最良の結果は500mMの濃度のショ糖を使用して得られた。凍結乾燥保護剤の濃度の更なる増加(500mmol超のショ糖)は、リポソームのサイズ及びPDIに関してより良好な結果を提供しない。
リポソーム懸濁液の非常に不十分な安定性のため、凍結乾燥リポソームの長期安定性を保証するためには、非常に低い残留水分を示す凍結乾燥物を得ることが望ましいであろう。凍結乾燥保護剤として500mmolのショ糖を含むCPP−TELリポソーム組成物の残留水分は、2.88%±0.79%であった。
実証実験:動物研究
1.種々の製剤の131I標識化バンコマイシンの血液レベル
実証実験の研究は、CPP−TEL−リポソームについて、標準リポソームと比較してバンコマイシン血液レベルの3倍の増加を示した。1種の追加成分(CPP又はTELのいずれか)のみの添加は、バンコマイシン血液レベルのわずかな増加を示したに過ぎない。これは、巨大分子の経口送達手段を提供するため、1つのリポソーム組成物において両方の成分の必要性を強調する(図9)。
免疫アッセイによるバンコマイシン血液レベルの特定は、放射能標識化研究に匹敵する結果(7.83%対8.68%)を示した(図10)。
本発明では、新規クラスのCPP−リン脂質−複合体を確立した。1つが直鎖CPP(モデルCPPペネトラチン、d/l−形態)で、もう1つが環状CPP(環状R9誘導体)の異なる2つのCPP変異体を使用した(図11)。質量分析によって検出される腸液中でのより高い酵素安定性のため、環状CPPを使用した(図13)。さらに、直鎖CPPペネトラチンは、D−アミノ酸で構成される場合、より安定であることがわかった。
全てのCPP(水中1mg/ml)を人工腸液で1:1(容積/容積)に希釈し、当該技術分野で知られているように、常に振蕩しながら37℃でインキュベートした。0分後、15分後、30分後及び60分後、無傷のCPPの回復を検出するため試料をHPLC/MSで分析し、初期の溶液との関係で比較した。
雄性ウィスターラット(200g〜250g)において、モデル物質であるバンコマイシンを用いて様々なリポソーム製剤を試験した。バンコマイシンの放射能標識化血液レベルの測定は、1モル%の直鎖CPP−リン脂質複合体を含むリポソーム製剤を使用することでバンコマイシンの経口アベイラビリティの著しい増加を示した。それにもかかわらず、わずか0.1モル%しか環状CPP−リン脂質−複合体を含まない別の製剤は、複合体を必要としても微量であるという利益を伴って、バンコマイシン血液レベルのより高い増加を示した。得られた結果は、バンコマイシンの免疫アッセイによって確認され、同様の血液レベルを測定することができた(図14)。
体重約200g〜250gの雄性ウィスターラットを使用し、地方自治体に従って動物研究を行った。実証実験の研究では、先に記載される通り、バンコマイシンを131Iで放射能標識化し、様々なリポソーム製剤に組み込んだ。経口投与から1時間後、放射能標識化した試料を直接計数することによりバンコマイシンの血液レベルを測定した。簡潔には、雄性ウィスターラット群(n≧3)を形成した。実験前12時間に亘りラットを給餌せず、自由飲水で維持した。リポソーム及び遊離ペプチドの経口適用を強制投与によって実施した。血液を除去して計量し、Berthold LB 951 Gカウンターを使用し、標準と比較して放射能を測定した。血液関連活性は総注入用量(ID)に関連し、組織1グラム当たりの総注入用量の割合(%ID/g)として表した。統計学的データをPrism(商標)ソフトウェア(米国カリフォルニア州サンディエゴのGraphPad Software)を使用して処理し、平均±平均値の標準偏差(S.D.)として提示した。Prism(商標)ソフトウェアを使用する一元配置分散分析検定によって種々の動物実験群を比較し、*p<0.05、**p<0.01及び***p<0.001で有意とした(図14)。
或る1つの更なる研究では、ペプチド薬物であるリラグルチドのバイオアベイラビリティを検討した。このため、薬物を静脈内注射し、血液レベルを1モル%の環状CPP−リポソームを含む経口リポソーム製剤と比較した。このペプチド薬物に関し、本発明者らのリポソーム製剤の使用によるバイオアベイラビリティは、リラグルチドの静脈内適用と比較して50%超である(図15)。
体重約200g〜250gの雄性ウィスターラットを使用し、地方自治体に従って動物研究を行った。バイオアベイラビリティの特定のため、抗糖尿病薬であるリラグルチドを131Iで放射能標識化し、1モル%の環状CPP−リン脂質−複合体を含むリポソーム製剤に組み込み、先に記載される通り、リラグルチド静脈内注射と比較した。幾つかの時間点において、放射能標識化した試料を直接計数することによりリラグルチドの血液レベルを測定した。簡潔には、雄性ウィスターラット群(n≧3)を形成した。実験前12時間に亘りラットを給餌せず、自由飲水で維持した。リポソームの経口適用を強制投与によって実施した。血液を除去して計量し、Berthold LB 951 Gカウンターを使用し、標準と比較して放射能を測定した。血液関連活性は総注入用量(ID)に関連し、組織1グラム当たりの総注入用量の割合(%ID/g)として表した。統計学的データをPrism(商標)ソフトウェア(米国カリフォルニア州サンディエゴのGraphPad Software)を使用して処理し、平均±平均値の標準偏差(S.D.)として提示した(図15)。
或る1つの更なるパイロット試験では、アダリムマブ(ヒュミラ(商標))の経口アベイラビリティを検討した。このため、抗体を131Iで放射能標識化し、1モル%の環状CPP−複合体を含むリポソームに組み込み、経口投与して、放射能標識化した遊離ペプチドと比較した。精製前及びその後の放射能標識化抗体の直接測定によって検出されるように、リポソーム中への抗体の組み込み効率は約60%であった。ここでも、アダリムマブの経口アベイラビリティの強い増加を検出することができた(図16)。したがって、この新規のリポソーム製剤もまた、抗体等の100kDa超の分子量を有する巨大分子薬物に適していると主張することができた。

別の研究では、頭部基修飾リン脂質へのCPPのカップリングに使用されるリンカーの影響を検討し、ペプチド薬物の組み込み効率は、リンカーのPEG単位の量に依存することがわかった(図17)。図18では、様々なリンカーを使用する幾つかの量(0モル%〜3モル%)のCPP−リン脂質複合体をリポソームに組み込み、その後、リポソームのサイズ及びPDIを特定した。PEG−リンカーが最も良好な結果を示し、8個〜50個の個別のPEG単位からなるPEG−リンカーが好ましいことがわかった。
5分以内(流量2ml/分;UV吸光度λ=214nm)で、アセトニトリル中の0.1%TFA(溶離液B)に対して水中の0.1%TFA(溶離液A)の直線勾配を適用し、C18カラム(Chromolithe(商標)Performance RP−18e、100mm−3mm)を使用する、逆相HPLC(Agilent 1100シリーズ)によってペプチド薬物の封入効率を特定した。スピード混合プロセスの後、リポソームを100μlずつ2つの部分に分割した。50μlの1%Triton(商標)X−100の添加によってリポソームを破壊し、HPLCによってペプチド薬物の曲線下面積(AUC)を特定することにより、部分1を使用して100%の値を計算して得た。部分2をSephadex G−25ゲル濾過クロマトグラフィー(NAP(商標)−5カラム)によって精製し、部分1のように定量した。部分2の精製中におけるNAP(商標)−5カラム上での脂質の潜在的喪失を特定するため、スピード混合プロセスの後、及びNAP(商標)−5カラムを使用する精製の後、リポソーム懸濁液中のコレステロール濃度を直接測定した。両測定に対してリポソームをメタノールに1:10(容積/容積)で溶解した。RP−18カラム上、15分以内(流速2ml/分;UV吸光度λ=208nm)でアセトニトリル/メタノール(80:20容積/容積)の無勾配(isocratic gradient)を適用するHPLCによってコレステロールを定量した。封入効率の計算にNAP(商標)−5カラム上での脂質喪失を含めるため、精製工程前及びその後のコレステロール濃度を比較し、補正係数Cを決定した。以下の方程式を使用して封入効率E(%)を計算した。

サイズ排除クロマトグラフィーによるリポソーム懸濁液の精製後、CPP修飾リポソームのゼータ電位は、CPPの正に帯電したアミノ酸により、未修飾のリポソームと比較して強い増加を示し(図19)、リポソーム中への複合体の組み込みの成功を確認した。
リポソームのゼータ電位を、Malvern(商標)(英国ウースタシャーのMalvern Instruments Ltd.)製のZetasizer Nano ZSの自動モードを使用して、室温で特定した。リポソーム製剤の粒子径、PDI及びゼータ電位を、いずれもMalvern(商標)(英国ウースタシャーのMalvern Instruments Ltd.)製のZetasizer Nano ZSを使用して室温で特定した。自動モードを使用して、pH7.4の10mMリン酸バッファーで0.076mg/mlの脂質濃度に希釈した後、サイズ及びPDIを測定した。pH7.4の50mMリン酸バッファーで0.95mg/mlの脂質濃度に希釈した後、ゼータ電位を特定した。Malvern(商標)(英国ウースタシャーのMalvern Instruments Ltd.)製のZetasizer Nano ZSの自動モードのデフォルト設定は次の通りであった:測定回数=3;実行時間=10秒;実行回数=10;平衡化時間=60秒;溶媒の屈折率1.330;ポリスチレンキュベットの屈折率1.590;粘度=0.8872mPa s;温度=25℃;誘電率=78.5F/m;後方散乱モード(173°);自動電圧選択;スモルコフスキー(Smoluchowski)方程式、リン酸バッファーpH7.4。
抗体マツズマブの放射能標識化により標準リポソームと比較した環状CPP−リン脂質−複合体を含むリポソームの結合能を試験した。新規な複合体を含むリポソームは、標準リポソームよりも有意に高い結合能を示した(図20)。
Caco−2細胞を6ウェルプレートに蒔いた。2週間の分化の後、増殖培地を除去した後に1mlのリポソーム試料を添加した。細胞を37℃で15分間、30分間及び60分間インキュベーションした後、リン酸バッファーで洗浄した。その後、細胞を1mlの1molグリシンバッファーpH2.2と共に5分間インキュベートした。0.3molのNaOHを添加し、0.25%SDSによって細胞溶解を行った。グリシンバッファー及び細胞溶解画分を収集し、Berthold LB 951 Gカウンターを使用し、標準と比較して放射能の量を測定した。
或る1つの更なる研究では、ペプチド薬物であるバンコマイシンのバイオアベイラビリティ(絶対)を検討した。このため、薬物を静脈内注射し、血液レベルを1モル%の環状CPP−リポソームを含む経口リポソーム製剤と比較した。このペプチド薬物に関し、本発明者らのリポソーム製剤の使用によるバイオアベイラビリティは、バンコマイシンの静脈内適用と比較して最大50%である(図21)。
体重約200g〜250gの雄性ウィスターラットを使用し、地方自治体に従って動物研究を行った。バイオアベイラビリティ(絶対)の特定のため、糖ペプチド抗生物質であるバンコマイシンを131Iで放射能標識化し、1モル%の環状CPP−リン脂質−複合体を含むリポソーム製剤に組み込み、先に記載される通り、バンコマイシン静脈内注射と比較した。幾つかの時間点において、放射能標識化した試料を直接計数することによりバンコマイシンの血液レベルを測定した。簡潔には、雄性ウィスターラット群(n≧3)を形成した。実験前12時間に亘りラットを給餌せず、自由飲水で維持した。リポソームの経口適用を強制投与によって実施した。血液を除去して計量し、Berthold LB 951 Gカウンターを使用し、標準と比較して放射能を測定した。血液関連活性は総注入用量(ID)に関連し、組織1グラム当たりの総注入用量の割合(%ID/g)として表した。
別の研究では、2つの抗体、すなわちアダリムマブ及びセツキシマブの組み込み効率を先に記載される通り特定した(図22)。両抗体もまた、ペプチド薬物に匹敵する高い組み込み効率(約50%〜70%)を示すことがわかった。
1モル%の環状CPP−複合体を含むリポソームに組み込まれたセツキシマブの腫瘍濃縮を特定するため、腫瘍を持つマウスに各リポソームを投与した。これらのマウスを得るため、細胞株A431をBalb/cマウス(EGFR過剰発現)に注射した。抗体セツキシマブを放射能標識化し、リポソームに組み込んだ後、マウスに経口適用した。リポソーム製剤を使用することで、遊離ペプチドと比較した腫瘍蓄積の有意な増加がわかった(図23)。

或る1つの更なる研究は、粒子の特徴を特定するため、別のCPP(MAP)を検討した。複合体を合成し、0モル%〜1モル%の範囲でリポソームへと組み込んだ。粒子径は標準リポソームに類似し、ゼータ電位は増加を示し、リポソームへの複合体の組み込みが成功したことを実証した(図24)。

本発明では、多様なCPP−TEL−リポソームの使用により、例えば、バンコマイシン、リラグルチド、インスリン等の様々なペプチド薬物、及びアダリムマブ又はセツキシマブ等の様々な抗体に対する有望で新規な経口送達システムを確立することができた。これらの結果(非常に増強された粘膜取り込み)は、一般に巨大分子薬物(ペプチド、タンパク質及び抗体)に対するプラットフォーム技術としてこの新規技術が役立つ可能性があることを実証する。CPP−TEL−リポソームはいずれも、サイズ、PDI及び組み込み効率(50%超)において高い均一性を示した。人工腸液中での安定性アッセイは、環状CPP−リン脂質−複合体が直鎖のものより安定し、直鎖CPPに関してD−アミノ酸の組み込みが好ましいことを示した。PEG単位の量が異なる幾つかのリンカーに関して、8個〜50個のPEG単位を有するPEG−リンカーが使用される場合、封入効率がより高いことがわかった。リポソームの特徴を他のCPPに伝達可能であることを実証するため、他のCPPに類似する特徴を示したCPP MAPをリポソームに組み込んだ。リポソーム製剤はいずれも、全ての試験した濃度で細胞毒性を示さなかった。さらに、凍結乾燥保護剤としてショ糖を使用する凍結乾燥によってCPP−TEL−リポソームの長期保存をもたらすことができた。雄性ウィスターラット(n=6)を使用する実証実験の研究では、標準リポソームと比較して、131Iで放射能標識化したバンコマイシンの3倍高い血中濃度を検出することができた。また、これらの結果を確認するため、血中濃度を免疫アッセイによって特定したところ、これもまたCPP−TEL−リポソーム製剤を使用するバンコマイシン血液レベルに匹敵する濃縮(7.83%ID対8.68%ID)を示した。リラグルチド;インスリン;アダリムマブ及びセツキシマブ等の更なる物質を放射能標識化して、経口適用後の特定の時間点において放射能を特定することにより、これらの物質に対し、これらの結果(リポソーム製剤による非常に増強された粘膜の取り込み)を確認することができた。
Claims (15)
- リポソームを含むリポソーム組成物であって、前記リポソームが、
(a)テトラエーテル脂質(TEL)と、
(b)細胞浸透性ペプチド(CPP)と、
を含み、
前記CPPが、前記リポソームの脂質二重層の一部である化合物に付着される、リポソーム組成物。 - 前記TELがスルホロブス属の一種に由来する、請求項1に記載のリポソーム組成物。
- 前記TELがスルホロブス・アシドカルダリウスに由来する、請求項2に記載のリポソーム組成物。
- 前記TELがグリセリルカルジチルテトラエーテル(GCTE)、ジグリセリルテトラエーテル(DGTE)、及びそれらの組み合わせからなる群より選択される、請求項1〜3のいずれか一項に記載のリポソーム組成物。
- 前記リポソームが、前記脂質の総量に対して1モル%〜10モル%の量で前記TELを含む、請求項1〜4のいずれか一項に記載のリポソーム組成物。
- 前記CPPが、直鎖又は環化ペネトラチン(配列番号1)、TAT(転写トランス活性化因子)−ペプチド(配列番号2)、MAP(モデル両親媒性ペプチド)(配列番号3)、R9(配列番号4)、pVEC(配列番号5)、トランスポータン(配列番号6)及びMPG(配列番号7)、並びにそれらの組み合わせ及びそれらの二量体からなる群より選択される、請求項1〜5のいずれか一項に記載のリポソーム組成物。
- 前記リポソームが前記脂質の総量に対して0.1モル%〜1モル%の量で前記CPPを含む、請求項1〜6のいずれか一項に記載のリポソーム組成物。
- 前記CPPが付着される前記化合物が、コレステロール及びその誘導体、リン脂質、リゾリン脂質、並びにテトラエーテル脂質からなる群より選択される、請求項1〜7のいずれか一項に記載のリポソーム組成物。
- 前記CPPが共有結合的に付着される、請求項1〜8のいずれか一項に記載のリポソーム組成物。
- 前記CPPがリンカーを介して付着される、請求項1〜9のいずれか一項に記載のリポソーム組成物。
- 前記リンカーが二官能性PEG−リンカーからなる群より選択される、請求項10に記載のリポソーム組成物。
- 少なくとも1種の治療剤及び/又は少なくとも1種の診断剤を更に含む、請求項1〜11のいずれか一項に記載のリポソーム組成物。
- 医薬として用いられる、請求項1〜12のいずれか一項に記載のリポソーム組成物。
- 経口投与用である、請求項13に記載のリポソーム組成物。
- 少なくとも1種の治療剤及び/又は少なくとも1種の診断剤を経口送達するための、請求項1〜12のいずれか一項に記載のリポソーム組成物の使用。
