試薬
制限酵素はFermentas and Promegaから、アルカリホスファターゼはAmersham Pharmacia Biotechから、dNTP及びT4DNAリガーゼはFermentasからのものであった。プライマーは、University of Turku,Department of Biotechnologyから、及びEurogentecからのものであった。Qiagenキットを、DNA精製に使用した。
製造者によって「Biomagnetic Techniques in Molecular Biology:Technical handbook」(Dynal A.S,2nd edition,1995)に記載される手順に従って、Dynal poly−dT磁気ビーズによって、5A10MAb産生ハイブリドーマ細胞(2E6細胞、Lilja et al,1991)からmRNAを抽出した。Super Script酵素(Gibco BRL)によって、mRNAからのcDNA合成を実行した。まず、mRNAと結合している磁性粒子を、1×反応緩衝剤、0.1MのDTT、2mMのdNTP、及びリボヌクレアーゼ阻害剤(50μlの全反応物体積中1.5μl)を含有する反応物中、42℃で2分間インキュベートした。その後、1μlのSuper Script(200U)を添加し、42℃で1時間反応を進めた。cDNA合成後、粒子及び反応混合物を磁性粒子回収機によって分離し、液体を除去し、粒子をTE緩衝剤中に懸濁させ、95℃で1分間インキュベートして、mRNAを放出させた。インキュベーション後、粒子に結合したcDNAを回収し、mRNA含有緩衝剤を除去し、粒子をTEで洗浄し、最後に50μlのTE中に懸濁して、保管した。

Diaprost ABによって提供された試薬を、製造者のガイドラインに従って、NovexからのTRIS−Tricineの10〜20%のアクリルアミドゲル上に泳動させた。
これらの結果から、抗体及び抗原試料は高品質かつ高純度のものであることが明らかである(結果示さず)。
(a)以下に記載される条件を使用して、相互作用の親和性を決定するために、2つの実験を実行した。

解離相データを適合し、解離速度定数(koff)を推定した(表1を参照されたい)

会合速度定数を推定するために、解離速度定数(表1)を適合した等式において使用した。
試験した抗体のそれぞれの解離定数(KD)を、表1に示す。
●全ての抗体について、会合プロセスは非常に高速であり、3〜9回の各抗体の実験に基づいて、会合速度定数(kon)は全て105M−1s−1の範囲内である。
●解離プロセスは非常に緩徐であり、ほぼBiacoreの技術的な限界の範囲内である。3〜9回の各抗体の実験に基づいて、解離速度定数(koff)は全て10−6s−1の範囲内である。
●全ての抗体について、解離定数(KD)は10−11Mの範囲内である。
全体の要約
IgGの6つのバリアントに、動的光散乱(DLS)研究を実行して、それらの凝集傾向を研究した。DLS結果は、全ての構築物が妥当な大きさ(球状タンパク質想定して、200kDaまたはわずかに200kDa超)を有し、凝集をほとんどまたは全く有さないことを示す。
動的光散乱を使用して、6つのIgG構築物をオリゴマー状態に関して特性評価すること。
動的光散乱
Malvern Zetasizer APS装置を使用して、20℃、2連の試料で動的光散乱を測定した。各試料を3回測定した。HBS−EP緩衝剤(顧客より)を対照として使用して、緩衝剤が粉塵及び凝集体を妥当に含まないことを確実にした。HBS−EP緩衝剤中の凝集体の信頼できる測定は行うことができず、これは良いことであった(緩衝剤は凝集体を含まないべきであるため)。数分布関数を使用して、全ての試料を確実に測定することができた。最も豊富な種の平均半径を、その種の多分散とともに計算した。その種の平均質量分布もまた計算した(表2を参照されたい)。
動的光散乱は、全ての構築物が妥当な大きさを有し、凝集をほとんどまたは全く有さないことを示す。6つ全ての構築物のサイズ分布は、重複している。驚くべきことに、緩衝剤様粒子(<1kDa)は全てのマウスIgG試料において見出される一方で、より大きな凝集体はヒトIgG試料のみにおいて見出される。
この研究は、111Inで標識した場合の、マウス5A10とヒト5A10とのインビボ体内分布を比較する。
ヒト化対応物の抗体h5A10を、以下に記載されるように産生した。
●抗体:配列番号12に従う重鎖及び配列番号13に従う軽鎖を含む、例示的なヒト化モノクローナル抗体5A10(IgG1/カッパ、HEK293細胞内に一過性発現、実施例2を参照されたい)は、Innovagen AB,Lund(ロット番号90475.33、PBS中濃度1.0mg/ml、pH7.4)によって提供された。非特異的IgG抗体を、アイソタイプ対照として利用した(マウス血清からのIgG抗体、Sigma I−8765)。
CHX−A’’−DTPAと5A10とのコンジュゲーション:PBSまたはNaCl中のマウス及びヒト化5A10mAbの溶液を、0.07Mのホウ酸ナトリウム緩衝剤(Sigma Aldrich)を使用してpH9.2に調節した。その後、タンパク質溶液を、3:1のキレート剤対抗体のモル比で、キレート剤CHX−A’’−DTPA(Macrocyclics,USA)と、40℃で4時間コンジュゲートさせた。反応を終了させ、20mlの0.2Mの酢酸アンモニウム緩衝剤(pH5.5)で平衡化したNAP−5カラム(GE Healthcare)でのサイズ排除クロマトグラフィーによって、CHX−A’’−DTPA−11B6(DTPA−11B6)を遊離キレートから分離した。コンジュゲートされた5A10抗体を1mlの酢酸アンモニウム緩衝剤で溶出させ、200uLのアリコートに分けた試料を−20℃で保管した。
インビボ研究について、hK2を発現する前立腺癌腫細胞株LNCaP(ATCC,Manassas,VA,USA)を使用した。細胞を、Thermo Scientificからの、10%のウシ胎仔血清及びPEST(ペニシリン100IU/ml及び100μg/mlのストレプトマイシン)で補助したRPMI1640培地(Thermo Scientific)中で培養した。細胞を、5%のCO2を有する加湿インキュベーター内で37℃で維持し、トリプシン−EDTA溶液(0.25%のトリプシン、0.02%のEDTA緩衝剤、Thermo Scientific)で剥離した。LNCaP細胞の異種移植時には、Matrigelマトリックス(BD−Biosciences,San−Jose,California,USA)を使用した。
111In−DTPA−h5A10及び111In−DTPA−m5A10のSPECT/CT撮像研究を実行した。撮像中、動物を、2%〜3%のイソフルランガス(Baxter;Deerfield,IL,USA)で麻酔した。皮下LNCaP異種移植片を有するNMRI−nuマウスの尾静脈に、約13〜15MBq、100uLのPBS中50ugのmAbで、111In−DTPA−h5A10(n=4)または111In−DTPA−h5A10(n=4)を静脈内注射した。NSP−106マルチピンホールマウスコリメータを有する前臨床SPECT/CTスキャナ(NanoSPECT/CT Plus,Bioscan;Washington,DC,USA)を使用することによって、動物を1時間撮像した。注射の1、2、3、及び7日後に、撮像を実行した。HiSPECTソフトウェア(SciVis;Goettingen,Germany)を使用して、SPECTデータを再構築した。各全身SPECTの前に、CT撮像を行った。InVivoScope2.0ソフトウェア(inviCRO;Boston,MA,USA)を使用して、SPECT/CT画像を分析し、CT画像を解剖学的基準として使用して、対象領域ROIを引き出した。
111In−DTPA−h5A10及び111In−DTPA−m5A10両方の体内分布研究を実行した。マウスの動物(n=12)群に、111In−DTPA−h5A10(約3〜4MBq、100uLのPBS中50ugのmAb)または111In−DTPA−m5A10(3〜4MBq、100uLのPBS中50ugのmAb)を静脈内注射した。動物を注射の7日後に屠殺し、対象器官を回収し、3インチのNaI(Tl)検出器(1480WIZARD,Wallac Oy,Turku,Finland)を有する自動NaI(Tl)ウェル計数器で分析した。

SPECT/CT撮像
111In−DTPA−h5A10及び111In−DTPA−m5A10注射の最大7日後にスキャンしたLNCaP異種移植片の代表的なSPECT/CT画像を、図1及び2に示す。活性は、24時間以内までにLNCaP腫瘍内に急速に蓄積し、放射線活性は、注射の7日後、高いままである。活性の高い蓄積はまた、肝臓内でも検出された。ROI分析は、対側脚の腫瘍対軟部組織の比率が、注射の7日後、111In−DTPA−m5A10の場合は5.2±0.84、及び111In−DTPA−h5A10の場合は6.4±1.3であることを示した。
異なる器官における活性蓄積をより徹底的に調査するために、より多くの数の動物(1つの抗体当たりn=12)においてエキソビボ体内分布を実行した。
この研究の結果は、以下のことを実証する。
●ヒト化5A10抗体111In−h5A10が、インビボで前立腺腫瘍を効果的に標的とすること、
●ヒト化h5A10抗体が、そのマウス抗体よりも予想外に良好な腫瘍蓄積を呈すること、及び
●ヒト化h5A10抗体が、マウス抗体よりも(より高い腫瘍対器官比によって示される)良好な撮像対比を提供すること。
放射性核種療法(RNT)の場合、放射線源は全体的に分布しており、放射線活性は通常放射性医薬品として全身投与される。放射線活性分布は、経時的に異なる組織内に蓄積する放射性医薬品の量(患者間で異なるものである)に依存する(1)。

- 治療される前記転移性前立腺癌が、リンパ系の転移、骨の転移(脊椎、椎骨、骨盤、肋骨を含む)、骨盤、直腸、膀胱、尿道内の転移である、請求項73に記載の方法。
- 前記患者が、前立腺癌を有し、前立腺癌の診断時点及び/または治療時点で、70歳未満、65歳、60歳、55歳、50歳、45歳、40歳以下である、請求項70〜74のいずれか1項に記載の方法。
- 前記患者が、父親または兄弟などの家族が、以前に前立腺癌と診断されていることを特徴とする、請求項70〜75のいずれか1項に記載の方法。
- 治療される前記前立腺癌が、去勢抵抗性前立腺癌(CRPC)である、請求項70〜76のいずれか1項に記載の方法。
- 前立腺癌細胞を除去するために、前記抗体ポリペプチドの投与後に、放射線誘導手術が、前記患者に実行される、請求項70〜77のいずれか1項に記載の方法。
- 対象の血液中の前立腺腫瘍細胞を検出するためのインビトロ方法であって、
(a)試験される対象からの血液試料を提供することと、
(b)任意で、前記血液試料中に存在する細胞を抽出及び/または精製することと、
(c)請求項1〜46のいずれか1項に記載の抗体ポリペプチドを、前記血液試料中に存在する細胞と接触させることと、
(d)前記抗体ポリペプチドが、遊離PSAに結合するかどうかを判定することと、
を含み、遊離PSAへの前記抗体ポリペプチドの前記結合が、対象の前記血液中の前立腺腫瘍細胞の存在を示す、前記方法。 - 対象の組織中の前立腺腫瘍細胞を検出するためのインビトロ方法であって、
(a)試験される対象からの組織試料を提供することと、
(b)任意で、前記組織試料中に存在する細胞を抽出及び/または精製することと、
(c)請求項1〜46のいずれか1項に記載の抗体ポリペプチドを、前記組織試料中に存在する細胞と接触させることと、
(d)前記抗体ポリペプチドが、遊離PSAに結合するかどうかを判定することと、
を含み、遊離PSAへの前記抗体ポリペプチドの前記結合が、対象の前記組織中の前立腺腫瘍細胞の存在を示す、前記方法。 - 前記組織試料が、組織学的試料である、請求項80に記載の方法。
- ステップ(d)が、ELISAによって実行される、請求項79〜81のいずれか1項に記載の方法。
- 前記試料中の前記前立腺腫瘍細胞の定量化を更に含む、請求項79〜82のいずれか1項に記載の方法。
- 対象における前立腺癌を診断するための、請求項79〜83のいずれか1項に記載の方法。
