Abstract
Description
本発明はまた、生理的に許容される媒体中に加水分解コメ抽出物及びブドウ種子抽出物の組み合わせを含有する組成物を皮膚に適用することを含む、汚染から皮膚を保護して皮膚再生及び修復を改善するための化粧的処理(treatment)方法に関する。
a. 植物性酵素での酵素的加水分解によって加水分解コメ抽出物を得て6kDa未満の分子量の低分子量ペプチドを取得し、
b. 空隙率が低下する複数のフィルターを用いて加水分解コメ抽出物を精製し、
c. pH4〜7の水中のブドウ種子ケーキからブドウ種子抽出物を取得し、
d. 空隙率が低下する複数のフィルターを用いてブドウ種子抽出物を精製し、そして
e. ステップb及びdで取得した抽出物を、5〜9の範囲の加水分解コメ抽出物及び5〜1の範囲のブドウ種子抽出物の比率、好ましくは9:1の比率で組み合わせて最終生成物の70%とし、
f. 次いでステップeで取得した生成物中にグリセロールを添加して最終生成物の30%とし、
g. 場合によって0.5%の安息香酸ナトリウムを添加しても良い。次いで最終溶液を無菌環境下でろ過し、低温殺菌によって殺菌する。

−糖アミン(sugar amine)、グルコサミン、D-グルコサミン、N-アセチル-グルコサミン、N-アセチル-D-グルコサミン、マンノサミン、N-アセチルマンノサミン、ガラクトサミン、N-アセチルガラクトサミン、
−ポリフェノール、イソフラボン、フラボノイド、例えばブドウ(grap)抽出物、松抽出物、オリーブ抽出物、
−動物性油、例えばスクアレン又はスクアラン、
−植物性油、例えばアーモンド油、ヤシ油、ヒマシ油、ホホバ油、オリーブ油、ナタネ油、ピーナッツ油、ヒマワリ油、コムギ胚芽油、トウモロコシ胚芽油、大豆油、綿実油、アルファルファ油、ケシ油、パンプキン種子油、月見草油、雑穀(millet)油、オオムギ油、ライムギ油、サフラワー油、パッションフルーツ油(passion oil)、ヘーゼルナッツ油、パーム油、アプリコットカーネル油、アボカド油、カレンデュラ油、エトキシル化植物油、又はシアバター。
上記の化合物は、植物のペプチド加水分解物のような天然のものであっても良く、またペプチド化合物のような合成のものであっても良い。
A. 加水分解コメ抽出物(Oryza sativa L種)の調製
第1のステップにおいて、タンパク質量が80%に高められた42gのコメ粉末(コメデンプン製造澱粉生成後に得られたもの)を1kgの水中に撹拌しながら懸濁する。次いで、0.04重量%のAlcalase(登録商標)及び0.04重量%のブロメラインのプロテアーゼを55℃で2時間添加し、pHをpH7.0〜7.5の範囲に調整する。
次いで酵素を80℃で2時間不活化させる。次いでpHを4.3〜4.7の範囲の値に調整する。
加水分解したコメ抽出物を次いでMillipore Heliconろ過カートリッジ(カットオフ値6kDa)上で限外ろ過に供する。濃縮液(retentate)中に含まれる高分子量の化合物を除去し、ろ液を回収する。このろ液は、いわゆる加水分解コメ抽出物の原料の形態(raw form)を示す。特にHPLCによる分析後、抽出物が6kDa未満の分子量を有するペプチドを約80%含有することが観察される。
−タンパク質及び低分子量ペプチド:77%
−炭水化物:13%
−アミノ酸:6.3%
−ポリフェノール:1.6%
−灰分(ash):2.1%
を含有する約700gの抽出物が得られる。
抽出物を次いで0.22μmのろ過で滅菌し、最後に65℃で一晩、オーブン中で殺菌する。
この段階で、加水分解コメ抽出物は、32g/lの乾燥抽出物を含有するわずかに黄色の溶液の形態であり、Lowry法で測定したペプチド化合物の濃度は17g/lである。
10% w/wのブドウ種子ケーキ粉末(grape seed cake flour)を、5g/kgの活性炭(Norit SX plus(登録商標))の存在下で水に懸濁し、50℃で1時間放置する。
次いで、ろ過収率を改善するために、10g/kgの珪藻土(ろ過助剤)を添加する。
後の使用のために保存する場合、ブドウ種子抽出物を次いで30%グリセロール及び0.5%安息香酸ナトリウムの添加によって希釈する。次いで得られた抽出物を、95%の糖、4.5%のタンパク質及び0.5%のポリフェノールから構成される乾燥物質0.5%に構成する。
抽出物を、ステップAで得られた加水分解コメ抽出物9部(重量)と、ステップBで得られたブドウ種子抽出物1部(重量)の比率で混合する。次いで、混合物を、15g/kg濃度のタンパク質(低分子量ペプチド)、0.5%濃度の安息香酸ナトリウム、及び30%グリセロールに標準化するために、水及びグリセロールで希釈する。
得られた最終生成物は、pH 4.5、20℃での密度1.07g/cm3、20℃での屈折率1.370、そしてタンパク質含量15g/kg(Lowryアッセイで測定)を有する。保存剤は0.5%濃度の安息香酸ナトリウムである。
NHEK細胞にUVBを照射し、照射後18時間インキュベートして細胞がDNAの光傷害を修復できるようにした。細胞内DNAは約260nmのUV光を効率的に吸収し、隣接するピリミジン間の二量体性光傷害(dimeric photolesion)、シクロブタンピリミジン二量体(CPD)の形成に到る。
NHEKを100J/m2のUVB照射前に実施例1のコメ/ブドウ種子抽出物で24時間前処理した。対応する濃度の活性剤による処理を照射後に延長した。照射18時間後に細胞を回収し、DNAを単離し、ElisaによってCPDを検出した。

NHEKをT75プレートp-2で60-70%のコンフルエンスになるまで増殖させる。
細胞をPBSで洗浄し、トリプシン処理する。
細胞を計数し、p50あたり1×105個でまく。
以下の最終希釈:培地中1、2、1、0.5、0.1及び0.01% v/vの実施例1のコメ/ブドウ種子抽出物(Neovixylともいう)で処理する。
処理(24時間)後、プレートをすすぎ、5ml PBSを添加する。
UV照射チャンバーでサンプルを0.01J/cm2で照射する。
PBSを対応する活性成分を含有する培地と置き換え、インキュベーターに戻して20時間置く。PBS中で細胞をかき取り、遠心分離して、ペレットを凍結する。
煮沸耐性チューブ(boiling secured tube)で10〜15分間DNAを変性させ、次いでチューブを氷浴中に移して少なくとも15〜30分間急速冷却した。
100μlのコーティング混合物をElisaウェルにトリプリケートでアプライした。
プレートを37℃で一晩乾燥させた(プレートを覆わない)。
プレートをElisa 1×洗浄バッファーで2×10分間洗浄した。
150μlのNeptuneブロッキングバッファーを用い、室温で1時間ブロックした。
1×洗浄バッファーで抗CPD抗体(TDM-2)を最終濃度1:12000に希釈した。
一次抗体100μlをアプライし、室温で60分間プレートをインキュベートした。
それぞれ180μlの洗浄バッファーで3×10分間洗浄した。
洗浄後、100μlの希釈二次抗体を添加し、(1:10000の最終希釈物を得るために1×洗浄バッファー中で)室温で1時間インキュベートした。
180μlの洗浄バッファーで3×5分間洗浄した。
PBSで1×3分間洗浄した。
ウェルに100μlの基質をアプライした(基質−9:1を達成するために45mlのパート1及び5mlのパート2)。
直後、及び10分毎に1時間まで、320/405をElisaリーダーで読み取った。
表1及び2において、実施例1のコメ/ブドウ種子抽出物をNeovixylと記載する。
図1及び2において、実施例1のコメ/ブドウ種子抽出物をNeovixyl(NEOVIXYLTM)と記載する。
創傷治癒試験方法
細胞の移動は、細胞外マトリクス、可溶性因子、並びに細胞間相互作用によって調節される。これらの因子の影響を、創傷治癒方法を使用して研究することができる。オルガノタイプスキンを、ハンドヘルドスカルパーを使用して、真皮区画を乱すことなく約2mmの幅に剥いで傷つけた。創傷部位にPBSで希釈した実施例1のコメ/ブドウ種子抽出物のアリコートを添加し、48時間インキュベートした。対応する濃度の実施例1のコメ/ブドウ種子抽出物を下部のインキュベーション培地に同様に添加した。オルガノタイプスキンを緩衝ホルマリン中で固定し、McClain研究所で標準的な組織学的手順によって処理した。オルガノタイプスキンの創傷床の画像を、NikonデジタルカメラDS-R1を備えた40倍及び100倍のOlympus顕微鏡を使用して取得した。創傷底部から移動する角化細胞の先端までの長さを、Nikon Elements BR-3-10プログラムを使用して測定した。両側の創傷の端(edge)からの距離を合わせ、データを長さ(μm)及び未処理の創傷と比較した増加%で表した。
表3において、実施例1のコメ/ブドウ種子抽出物をNeovixylと記載する。
プロトコル
組織培養
正常ヒト表皮角化細胞(NHEK)を60mmのペトリ皿上でおよそ50%のコンフルエンシーに達するまで継代培養(sub-cultivate)した。実施例1のコメ/ブドウ種子抽出物を以下の濃度:0.1%、0.5%及び1%でNHEK媒体中に調製した。次いでNHEKを新しい培地、又は実施例1のコメ/ブドウ種子抽出物を含む培地のいずれかで処理して一晩インキュベートした。
オゾン暴露の前に、細胞培養培地を除去し、2mlのダルベッコリン酸緩衝生理食塩水に置き換えた。プレートを、450アングルに設定した回転ステージを備えたオゾンチャンバー中に置いた。次いで細胞を0.4ppmのオゾンに20分間暴露し、続いてそれぞれの培地を用いて再度2時間インキュベートし、トリプシンで処理して回収した。最後に、細胞を計数し、1×104個の細胞をコメットアッセイに用いた。
NHEKをトリプシン処理し、PBSで洗浄し、1×105細胞/mLでPBS中に懸濁した。細胞を次いで37℃で融解アガロース(Trevigen, cat#4250-050-02)中に1:10の比率で分散させた。細胞/アガロース混合物75μlをコメットスライド(Trevigen, cat#4250-050-03)の各スポット上に均一にピぺッティングし、次いで4℃で10分間インキュベートした。スライドを氷上の冷溶解液(Trevigen, cat#4250-050-01)に3時間浸漬した。溶解液からスライドを取り出し、アルカリ溶液(300mM NaOH, 1mM EDTA, pH>13)中に室温で30分間置いた。次いでスライドを氷で冷やした電気泳動装置内に電極から等距離になるように置いた。冷やしたアルカリ性電気泳動液(300mM NaOH, 1mM EDTA, pH>13)を装置中に注ぎ、スライドを覆う程度にした。電気泳動は23Vで30分間実施した。電気泳動の後、スライドをH2Oですすぎ、70%EtOHに5分間浸漬した。スライドをEtOH溶液から取り出し、タオルの上に置いて一晩風乾した。SYBRグリーン(Trevigen, cat#4250-050-05)をTEバッファー(10 mM Tris-HCl, 1mM EDTA, pH 7.5)で1:10000に希釈した。希釈したSYBRグリーン50μlを各スポット上にピペッティングした。スライドを4℃で5分間インキュベートした。次いで、スライドから過剰量のSYBRグリーンを除いた後にスライドを再び乾燥させた。20倍の対物レンズとFITCフィルターとを備えたOlympus BX51顕微鏡でスライドを観察した。画像はNikon Elementsソフトウェアを使用して取得した。Tri Tek社のComet Scoreソフトウェアを使用してテールモーメントを決定した。
図4は、オゾン暴露後のDNAの損傷を示す。オゾン及び実施例1のコメ/ブドウ種子抽出物で処理したNHEKのコメットスコアを示す。実施例1のコメ/ブドウ種子抽出物をNeovixyl(NEOVIXYLTM)と記載する。
実施例1のコメ/ブドウ種子抽出物は、オゾンによって誘発されるDNA断片化の増大に対抗することができた。実施例1のコメ/ブドウ種子抽出物による処理は、オゾンの影響から細胞を保護する。
Claims (8)
- 化粧用組成物を皮膚に局所的に適用することを含む、大気汚染から皮膚表皮細胞を保護するための化粧的処理方法であって、該組成物が、5〜9の範囲の加水分解コメ抽出物及び5〜1の範囲のブドウ種子抽出物の比率の加水分解コメ抽出物及びブドウ種子抽出物の組み合わせからなる0.01〜5%の活性成分、及び生理的に許容される媒体を含有する、上記方法。
- 上記比率が加水分解コメ抽出物及びブドウ種子抽出物で9:1(w/w)に調節されている、請求項1記載の化粧的処理方法。
- 上記化粧用組成物が、加水分解コメ抽出物及びブドウ種子抽出物の組み合わせを0.5〜2%含有する、請求項1記載の化粧的処理方法。
- 上記方法が:
a. 植物性酵素での酵素的加水分解によって加水分解コメ抽出物を得て6kDa未満の分子量の低分子量ペプチドを取得し、
b. 空隙率が低下する複数のフィルター(decreasing porosity filters)を用いて加水分解コメ抽出物を精製し、
c. pH4〜7の水中のブドウ種子ケーキからブドウ種子抽出物を取得し、
d. 空隙率が低下する複数のフィルターを用いてブドウ種子抽出物を精製し、そして
e. ステップb及びdで取得した抽出物を9:1の比率で組み合わせて最終生成物の70%とし、
f. ステップeで取得した生成物中にグリセロールを添加して最終生成物の30%とする
を含む、請求項1記載の化粧的処理方法。 - 加水分解コメ抽出物及びブドウ種子抽出物の組み合わせが、10〜20g/kgの範囲のタンパク質含量、より好ましくは15g/kgのタンパク質含量を含む、請求項1記載の化粧的処理方法。
- 加水分解コメ抽出物が6kDa未満の分子量を有するペプチドから構成される、請求項1記載の化粧的処理方法。
- 皮膚再生及び修復を改善するために使用される、請求項1記載の化粧的処理方法。
- 汚染の有害な影響による皮膚老化及び光老化の徴候を防止及び/又は処理するために使用される、請求項1記載の化粧的処理方法。
