材料および方法
菌株、プラスミドおよび増殖条件。
本明細書で使用した細菌株およびプラスミドを以下に挙げる。
NCBIのBasic Local Alignment Search Tool(BLAST)を用いてアシネトバクター(Acinetobacter)特異的OTaseのタンパク質配列を解析し、タンパク質のアノテーションにConserved Domain Databaseを用いてタンパク質ドメインを同定した(Marchler−Bauerら,2004、Marchler−Bauerら,2009、Marchler−Bauerら,2011)。
既に記載されている方法論(Hardingら,2013)に従ってウエスタンブロット解析を実施した。使用した一次抗体は抗rsPilAM2またはペンタ−His抗体(Qiagen社)であった。使用した二次抗体はヤギ抗ウサギIgG(H+L)、アルカリホスファターゼ抗体(Molecular Probes社)およびヤギ抗マウスIgG(H+L)、アルカリホスファターゼ抗体(Molecular Probes社)であった。BCIP/NBT Liquid Substrate System(Sigma社)で膜を発色させた。
既に記載されているものに以下の修正を加えて線毛剪断調製物を調製した。簡潔に述べれば、寒天表面から菌叢を取り出し、1×プロテアーゼ阻害剤(Roche社)を添加した氷冷DPBS 5mLに再懸濁させた。細菌懸濁液をA600nmでの吸光度70に正規化した。表面に露出したタンパク質を剪断するため、細菌懸濁液を高速で1分間ボルテックス処理した。細菌を10,000×g、4℃で10分間ペレット化した。上清を収集し、再び10,000×g、4℃で10分間遠心分離した。上清を収集し、20,000×g、4℃で5分間遠心分離することによりさらに清澄化した。剪断された表面タンパク質を最終濃度30%の硫酸アンモニウムで沈殿させた。沈殿したタンパク質を20,000×g、4℃で10分間遠心分離することによりペレット化した。上清を捨て、ペレットを1×Laemmli緩衝液100μLに再懸濁させた。調製物を10分間煮沸し、SDS−PAGEで泳動し、クーマシー染色を実施し、バンドを切り出し、Shevchenkoら(2006)に従って質量分析用に調製した。簡潔に述べれば、バンドを水で洗浄し、アセトニトリル(ACN)で繰り返し脱水した。ジスルフィド結合を37℃で60分間、50mM NH4HCO3に溶かした10mM DTTで還元した後、室温の暗所で60分間、50mM NH4HCO3に溶かした50mMヨードアセトアミドでシステインチオール基をアルキル化した。次いで、ゲル片を50mM NH4HCO3で洗浄し、100%ACNで脱水し、乾燥させた。ピリンを37℃で16時間、50mM NH4HCO3に溶かした0.02mg/mLトリプシン(Promega社)で消化した。100%ACNおよび水でペプチドを溶離し、凍結乾燥させた。トリプシンペプチドを0.1%トリフルオロ酢酸に再懸濁させ、C18 ZipTip(Millipore社、米国)で脱塩した。60%ACNを用いてペプチドを溶離し、これをspeedvacで乾燥させ、0.1%ギ酸に再懸濁させた。既に記載されている通りに(Wangら,2007)、nanoACQUITY(Waters社)超高速液体クロマトグラフィーシステムと接続したQ−TOF Premier(Waters社、マンチェスター、イギリス)を用いて解析を実施した。MassLynx、v.4.1(Waters社)を用いてデータを解析した。M2菌株からOmpA−Hisを精製し、上記のようにESI−QTOF MS/MS解析用に調製した。
既に記載されているTRI試薬法(YiおよびHackett,2000)により、A.バイリイ(A.baylyi)および青枯病菌(R.solanacearum)のLPSを一晩培養物から抽出した。既に記載されている通りに(TsaiおよびFrasch,1982)、等量のLPSを12.5%SDS−PAGEゲルに負荷してLPSを分離した後、銀染色を実施した。
大腸菌(E.coli)CLM24細胞を3種類のプラスミド、すなわち、C.ジェジュニ(C.jejuni)のグリコシル化遺伝子座をコードするプラスミド、単一のO−OTase遺伝子をコードするプラスミドおよびdsbA1−HisをコードするプラスミドpAMF22で形質転換した。プラスミドを選択するため、必要に応じてアンピシリン(100μg/ml)、トリメトプリム(50μg/ml)およびクロラムフェニコール(10μg/ml)を添加した。細胞を37℃でOD600が0.4〜0.6になるまで増殖させ、次いで0.1mM IPTGおよび/または0.2%アラビノースで誘導した。アラビノース誘導を必要とする培養物には、4時間後に2回目の誘導を実施した。定常期に全細胞溶解物を入手し、ウエスタンブロット解析を用いてDsbA1の修飾を判定した。
若干修正を加えたLithgowら(2014)に従って、糖ペプチド濃縮および定量解析に供するペプチドライセートを調製した。簡潔に述べれば、乾燥させた膜濃縮タンパク質試料2mgを6M尿素、2Mチオウレア、40mM NH4HCO3で可溶化し、10mMジチオスレイトール(DTT)で還元した。還元し可溶化したペプチドを光の不在下、25mMヨードアセトアミド(IAA)で1時間アルキル化した。得られたアルキル化タンパク質混合物を25℃で4時間、Lys−C(1/100 w/w)で消化し、40mM NH4HCO3で1:5に希釈した後、25℃で一晩、トリプシン(1/50 w/w)で消化した。1%トリフルオロ酢酸(TFA)を加えて消化を停止させた。C18 empore(Sigma−Aldrich社、セントルイス、ミズーリ州)STop And Go Extraction(STAGE)チップ(Rappsilberら,2007)を用いて一級アミドおよび塩を除去し、ペプチド消化物を精製した。
若干修正を加えた(Scottら,2011)に従ってZIC−HILIC濃縮を実施した。1mm2に切り取ったC8 Empore(商標)ディスク(Sigma社)を含むp10チップに充填した10μm ZIC−HILICレジン(Sequant社、ウメオ、スウェーデン)からなるマイクロカラムをベッド長が0.5cmになるまで充填した。使用前、カラムを超純水、次いで95%アセトニトリル(ACN)で洗浄し、次いで80%ACNおよび5%ギ酸(FA)で平衡化した。試料を80%ACN/5%FAに再懸濁させ、16,100×g、4℃で5分間遠心分離することにより不溶性物質を除去した。試料の濃度を3μg/μLに調整し、ペプチド物質150μgをカラムに負荷し、10負荷体積の80%ACN、5%FAで洗浄した。未結合画分を収集してプールし、真空遠心分離により乾燥させた。ZIC−HILIC結合ペプチドを3負荷体積の超純水で溶離させ、真空遠心分離を用いて濃縮した。新たに調製した試薬を用いて、別個にバイオロジカルレプリケートをZIC−HILIC濃縮に供した。
精製した糖ペプチド/ペプチドを緩衝液A(0.5%酢酸)に再懸濁させ、糖ペプチドの定性分析にはLTQ−Orbitrap Velos(Thermo Scientific社、サンノゼ、カリフォルニア州)と接続したAgilent 1290 Series HPLC(Agilent Technologies社、ミシサガ、オンタリオ州)、定量試験にはQ−exactiveと接続したEASY−nLC1000システムで逆相クロマトグラフィーを用いて分離した。A.バイリイ(A.baylyi)ADP1糖ペプチドの定性分析には、社内で充填した20cm、内径75μm、外径360μmのReproSil−Pur C18 AQ1.9μm(Dr.Maisch社、アンマーブーフ−エントリンゲン、ドイツ)カラムを用いた。定量試験には、社内で充填した45cm、内径50μm、外径360μmのReproSil−Pur C18 AQ1.9μmカラムを用いた。両システムとも、試料をトラップカラム、すなわち、社内で充填した2cm、内径100μm、外径360μmでAqua 5μm C18を含むカラム(Phenomenex社、トーランス、カリフォルニア州)に5μL/分で負荷した後、グラジエント分離を実施し、質量分析用に注入した。0%の緩衝液B(80%ACN、0.5%酢酸)から32%のBまで140分、次いで32%のBから40%のBまで5分、次いで100%のBまで2.5分かけて上昇させ、100%のBで2.5分間保持し、次いで、さらに20分かけて0%のBまで低下させる180分間のグラジエントをかけた。ZIC−HILIC糖ペプチド濃縮で得た未結合画分を糖ペプチドの定性分析と同じ機器設定を用いた分析に供した。両機器とも、定性分析には、既に記載されている通りに(Scottら,2011)、Xcalibur v2.2(Thermo Scientific社)を用いて、MS、CID MS−MSおよびHCD MS−MSの間で自動的に切り替わるデータ依存モードでキャピラリー温度275℃にて稼働させ、定量試験には、MS(分解能70k、AGC標的1×106)とHCD MS−MSイベント(分解能17.5k、AGC標的1×106、最大注入時間60ミリ秒、20%ステップのNCE28)との間で切り替わる上位10のデータに依存する方法を用いて稼働させた。
定性的グリコシル化分析の未処理ファイルを既に記載されている通りに(Scottら,2011,2012)処理した。簡潔に述べれば、Proteome Discoverer v.1.2(Thermo Scientific社)を用い、得られた糖ペプチドに関するデータをA.バイリイ(A.baylyi)ADP1のデータベース(UNIPROT、http://www.uniprot.org/、681 2014−06−10から入手、Taxon識別番号:3263個のタンパク質配列を含む62977)にMASCOT v2.4を用いて検索した。以下のパラメータ:ペプチド質量精度20ppm;フラグメント質量精度0.02Da;酵素特異性無し、固定修飾(カルバミドメチル)、可変修飾(メチオニン酸化および脱アミドN、Q)を用いてMascot検索を実施した。以前の研究でHCDスペクトル内に四極子様断片化が示されていることから(Olsenら,2007)、MALDI−QUAD−TOFの機器設定を選択した。MASCOT検索からペプチドが同定されなかったスキャンイベントをMicrosoft Excel(Microsoft社、レイモンド、ワシントン州)にエクスポートした。エクスポートしたマッチしないスキャンの中に糖ペプチドの可能性のある物質を同定するため、「mgfグラフ」と称するGPMAW8.2のMS−MSモジュールを用いて、204.08m/zのHexNAcのオキソニウムが含まれるHCDスキャンイベントを洗い出した。オキソニウム204.08m/zイオンが含まれる全スキャンイベントを手動で調べ、糖ペプチドの可能性のある物質を同定した。糖ペプチドの帰属を容易にするため、204.08オキソニウムが含まれるHCDスキャンイベントを手動で調べ、脱グリコシル化ペプチドイオンの可能性のある物質を同定した。Xcalibur v2.2のスペクトルリスト機能を用いて、これらのHCDスキャンのうち、脱グリコシル化ペプチドの質量を下回る質量に対応するMS特性(m/z、電荷および強度)を抽出した。検出されたMS特性に関して得られた数値をmgfファイルに書き込み、ペプチドの質量を脱グリコシル化ペプチドの質量に設定した。次いで、上記のMASCOT設定を用いて、得られたmgfファイルを検索した。デコイオプションを起動して全スペクトルを検索したところ、このデータベースにマッチするものは検出されず、偽発見率(FDR)は0%であった。
アシネトバクター(Acinetobacter)がコードする2種類のOTaseホモログは異なるpfamドメインを含む
これまでに、A.バイリイ(A.baylyi)ADP1がWzy_Cスーパーファミリーのドメインを含むタンパク質を2種類コードすることが報告されている(Schulzら,2013)。本発明者らはバイオインフォマティクス解析により、主要IVa型ピリンサブユニットpilAのすぐ下流に進化的に関係のあるWzy_Cスーパーファミリーのドメインを含むタンパク質をコードすると予想される2つのオープンリーディングフレーム(ORF)を有するアシネトバクター(Acinetobacter)菌種を数種類同定した(図1、パネルA)。
図2Aは、同系のpilA変異株であるM2菌株の全細胞溶解物のウエスタンブロット解析を示しており、その補完されたpilA変異菌株から、PilAが見かけの分子量の異なる2つの分子形態で存在するという本発明者らのそれまでの知見が裏付けられた。存在量が多く分子量が大きい方の形態のPilAは翻訳後修飾されたPilA種であると考えられ、分子量が小さい方の形態のPilAは未修飾種であった。TfpOがPilAの翻訳後修飾に及ぼす影響を明らかにするため、同系のtfpO変異株を構築し、PilAの発現を調べた。tfpOを欠く菌株のPilAは分子量が小さい方の形態でのみ存在していた(図2A)。PilAの電気泳動移動度の増大と翻訳後修飾の喪失とで一致がみられる。さらに、補完されたtfpO変異菌株のPilAは主として分子量の大きい方の形態で存在し、PilAの翻訳後修飾にはTfpOが必要であることが裏付けられた。
PilAがTfpOによってグリコシル化されることを裏付けるため、M2菌株、超線毛型M2ΔpilT変異株および超線毛型M2ΔtfpO::kanΔpilT::strep変異株の表面剪断調製物からPilAを精製した。pilT遺伝子は予測収縮ATPアーゼをコードし、したがって、pilTを欠く変異株は超線毛型の表現型を呈し、表面に露出したPilAの量が多くなる。剪断調製物中のタンパク質をSDS−PAGEによって分離し、クーマシー染色を実施し、切出し、質量分析に供した。図2Bは、M2およびM2ΔpilTの両菌株のPilAのMS/MS解析により、PilAにHexNAc残基2つ、ヘキソースおよびN−アセチルデオキシヘキソースからなる四糖が存在することが確認されたことを示している。MS/MS解析から、この四糖がカルボキシ末端のトリプシン119NSGTDTPVELLPQSFVAS136ペプチド上に存在することがわかった。M2ΔtfpO::kanΔpilT::strep変異株のPilAには修飾がみられず、PilAグリコシル化にはTfpOが必要であることが裏付けられた(データ不掲載)。
緑膿菌(P.aeruginosa)1244では、ピリンタンパク質PilAがTfpO依存性にグリコシル化される(Castric,1995)。のちにグリコシル化部位がカルボキシ末端セリン148の位置にあることが明らかにされた(Comerら,2002)。A.ノソコミアリス(A.nosocomialis)M2を含めたアシネトバクター(Acinetobacter)菌種のPilAタンパク質のアミノ酸配列を緑膿菌(P.aeruginosa)1244 PilAと比較した。配列の相同性は低いが、M2菌株のPilA配列にもC末端セリンが含まれており、これはMSによって同定された糖ペプチドに含まれていたものである(図2B)。
Huらは最近、主要多糖抗原(MPA)遺伝子座を含むアシネトバクター(Acinetobacter)菌種の血清型を分子レベルで判定するスキームを開発した。保存されたfkpA遺伝子とlldP遺伝子の間に存在するMPA遺伝子座は、研究対象に含めシーケンシングを実施した全アシネトバクター(Acinetobacter)菌株に確認されたほか、A.ノソコミアリス(A.nosocomialis)M2菌株にもみられた(Huら,2013,Carruthersら,2013)。A.ノソコミアリス(A.nosocomialis)M2菌株のMPA遺伝子座には、予測グリコシルトランスフェラーゼ3種類(wafY、wafZおよびwagBと呼ばれる)および予測開始グリコシルトランスフェラーゼ1種類(weeHまたはpglCと呼ばれる)が含まれている(図4A)。MPA遺伝子座がPilAM2の翻訳後修飾に必要であるかどうかを明らかにするため、予測グリコシルトランスフェラーゼをそれぞれ欠く同系変異株を個別に構築した。図4BはweeHを欠く菌株の全細胞溶解物のウエスタンブロット解析を示しており、この解析から、PilAが分子量の小さい方の形態で存在することが明らかになり、PilAのグリコシル化にはWeeHが必要であることがわかった。ほかの3種類のグリコシルトランスフェラーゼの欠失では、中間の電気泳動移動度を示すPilAタンパク質が得られた。wafY::kan変異株のPilAはWT PilAの移動度に最も近い位置に移動し、これにwafZ::kan変異株のPilA、wagB::kan変異株のPilAがこの順に次いだ(図4B)。wafZ::kan変異株およびwagB::kan変異株のバックグラウンドから部分的に修飾された形態および未修飾の形態のPilAがともに同定されたのは興味深い。いずれの変異菌株も補完に成功し、完全に修飾されたPilAの産生にはwafY遺伝子、wafZ遺伝子、wagB遺伝子およびweeH遺伝子の産物がいずれも必要であることがわかった。
図2Aでは、Wzy_Cドメインを含む第二のORFであるpglLM2はピリングリコシル化に必要ではないことが示された。pglLM2は、A.バウマンニ(A.baumannii)ATCC17978で既に特徴が明らかにされているPglLと同じように汎用O−OTaseであり、ピリン以外の標的タンパク質をグリコシル化するのではないかと考えられる。本発明者らは最近、タンパク質OmpAをコードするA1S_1193−Hisが様々な菌株PglLによって認識されることから、アシネトバクター(Acinetobacter)菌株特異的グリカンを単離および同定するベイトアクセプタータンパク質としての役割を果たす可能性があることを明らかにした(Scottら,2014)。本発明者らは、菌株M2、M2ΔtfpO::kan、M2ΔpglL::kanおよびM2ΔweeH::kanの各菌株でカルボキシ末端Hisタグを含むOmpA−Hisを発現させた。
上記の通り、A.バイリイ(A.baylyi)ADP1のゲノムでは、Wzy_Cスーパーファミリーのドメインを含むOTase様タンパク質が2種類コードされている。そのうちの1つであるpglLComP(ACIAD3337)はcomPに隣接している。Schulzら(2013)は、ComPの修飾にpglLComP(ACIAD3337)が必要であることを明らかにしており、これとは別に本発明者らもこれを確認している(図6A)。さらに、同系のpglLADP1(ACIAD0103)変異菌株のComP−His発現を調べたウエスタンブロット解析では、ComPの翻訳後修飾にPglLADP1は必要ではないことが明らかになった(図6A)。
髄膜炎菌(N.meningitidis)のDsbA1およびA.バウマンニ(A.baumannii)ATCC17978のOmpAは、それぞれの菌株で汎用O−OTaseによっても修飾され、以前はグリコシル化を研究するためのモデルとして用いられた(Vikら,2009;Iwashkiwら,2012;Gebhartら,2012;Lithgowら,2014)ものであり、PglLADP1がこれをグリコシル化することができるかどうかを検討した。この2種類のタンパク質を野生型、ΔpglLComPおよびΔpglLADP1のA.バイリイ(A.baylyi)菌株で別個に発現させた。DsbA1−His(図6C)およびOmpA−His(図6D)は、野生型およびΔpglLComPバックグラウンドよりもΔpglLADP1バックグラウンドの方が電気泳動移動度が大きかった。以上の実験は、PglLADP1が汎用O−OTaseとしての役割を果たすことを裏付けるものである。
A.バイリイ(A.baylyi)のADP1グリコシル化での両推定O−OTaseの役割を明らかにするため、A.バイリイ(A.baylyi)ADP1のグリコプロテオームをADP1ΔpglLComP変異株またはAPD1ΔpglLADP1変異株のいずれかと比較した。糖ペプチド濃縮のためのZIC−HILIC(Iwashkiwら,2012;Nothaftら,2012;Scottら,2014;Lithgowら,2014)および複数のMS/MS断片化法(Scottら,2011)を用いて、A.バイリイ(A.baylyi)ADP1に8種類のタンパク質基質に由来する21種類の固有の糖ペプチドが360個同定された。ほかのアシネトバクター(Acinetobacter)菌種に観察されている多様性(Scottら,2014)と同じく、286−217−HexNAc3(1112.41Da、図8Aおよび8D)、286−217−245−HexNAc2(1154.41Da、図8Bおよび8F)、286−217−HexNAc−245−HexNAc(1154.41Da、図8Bおよび8G)および286−217−2452−HexNAc(1196.41Da、図8Cおよび8E)からなる4種類の五糖グリコフォームのうちの1種類で修飾された糖ペプチドを有する固有のグリカンがA.バイリイ(A.baylyi)ADP1で得られた。A.バイリイ(A.baylyi)ADP1ΔpglLComPの膜タンパク質の糖ペプチド解析によって同じ糖ペプチドが同定され、グリコプロテオームはこの遺伝子の欠失による影響を受けないことが示唆された。これに対し、野生型A.バイリイ(A.baylyi)ADP1に観察された21種類の糖ペプチドのうちA.バイリイ(A.baylyi)ADP1ΔpglLADP1の抽出物中に検出されたものはなく、A.バイリイ(A.baylyi)ADP1ではpglLADP1が一般的なタンパク質グリコシル化を担っていることが示唆された。
図19は、ジメチル標識を用いたA.バイリイ(A.baylyi)ADP1 WT、A.バイリイ(A.baylyi)ADP1ΔpglLADP1およびA.バイリイ(A.baylyi)ADP1ΔpglLComPのグリコシル化の定量解析を示している。
アシネトバクター(Acinetobacter)染色体上の主要IVa型ピリン遺伝子と下流のO−OTase遺伝子との間に強い遺伝的連鎖があることから、これらのO−OTaseがそのコグネイトピリンタンパク質に対して特異的でるかどうかを明らかにしようとした。A.ノソコミアリス(A.nosocomialis)M2菌株のPilAM2を発現するプラスミドを異なるアシネトバクター(Acinetobacter)種で発現させた後、ウエスタンブロット解析を実施し、ピリンタンパク質の発現および電気泳動移動度を調べた。分子量が大きい方の形態のPilAM2および分子量が小さい方の形態のPilAM2の両方が存在することからわかるように、PilAM2は、ともにtfpOホモログおよび末端セリン残基を含むピリンをコードするA.バウマンニ(A.baumannii)ATCC19606および臨床分離株のA.バウマンニ(A.baumannii)27413によって修飾された(図9A)。A.バウマンニ(A.baumannii)ATCC19606のグリカンは、A.バウマンニ(A.baumannii)ATCC17978で同定された五糖と同じものであることがわかった。A.バウマンニ(A.baumannii)ATCC19606で発現したPilAM2は電気泳動の移動速度が最も遅く、大きいグリカンがPilAM2と結合していることが示された(Scottら,2014)。tfpOホモログを欠くA.バウマンニ(A.baumannii)ATCC17978およびA.バイリイ(A.baylyi)ADP1はともに、PilAM2をグリコシル化することができなかった。
材料および方法:
菌株およびプラスミド
本研究に使用した細菌株およびプラスミドを表4に記載する。DNeasy blood and tissue kit(Qiagen社)を用いてA.バイリイ(A.baylyi)のゲノムDNAを単離した。遺伝子aciad3337(pglLComP)およびその上流の非コード領域の増幅には、プライマーigrF(ACTGGTCGACTAGTAGTACTATATGGCTTTAAA;配列番号89)およびigrR(ACTGCTGCAGTTAATATTCTATTGAACAAAATTTTAAC;配列番号90)を用いた。得られたPCR産物をSalIおよびPstIで消化しpEXT20の同じ部位に挿入してpMN8を作製した。ComPをpBAV−ComP−hisからpEXT20のBamHIおよびSalI部位にサブクローン化することによってプラスミドpMN1を構築した。pglLComPおよびその上流領域をpMN8のSalIおよびPstI部位からpMN1にサブクローン化することによってプラスミドpMN2を構築した。
Dowerらによって記載されているプロトコル(Dowerら,1988)に従って、エレクトロコンピテントな大腸菌(E.coli)CLM24、CLM37またはSDB1を調製した。グリカン合成遺伝子座、アクセプタータンパク質およびOTaseをコードするプラスミドで細胞を形質転換させた。プラスミドの選択には、アンピシリン(100μg/ml)、テトラサイクリン(20μg/ml)、トリメトプリム(50μg/ml)、クロラムフェニコール(12.5μg/ml)、カナマイシン(20μg/ml)およびスペクチノマイシン(80μg/ml)を必要に応じて添加した。細胞をLBブロス中、37℃でOD6000.4〜0.6まで増殖させ、必要に応じて0.05mMもしくは0.1mM IPTGまたは0.2%アラビノースで誘導し、37℃で一晩放置した。アラビノース誘導が必要な培養物には4時間後に2回目のアラビノース投与を実施した。定常期に全細胞溶解物を入手し、そのうち0.1ODを12.5%SDS−PAGEゲルに負荷し、次いでニトロセルロース膜に転写した。ウエスタンブロット解析を用いてタンパク質修飾を判定した。使用した抗体を表5にまとめる。Odyssey Infrared Imaging System(LiCor Biosciences社、米国)を用いてニトロセルロース膜を可視化した。
テリフィックブロス中37℃で一晩増殖させた細胞をリン酸緩衝生理食塩水(PBS)緩衝液で洗浄し、同緩衝液に再懸濁させた。フレンチプレスを20kpsi(1.4×105kPa)で用いて細胞の破壊を2回実施した後、プロテアーゼインヒビターカクテル(Roche社)を加えることにより細胞を溶解させた。ライセートを10000×gで30分間遠心分離して細胞残屑を除去し、次いで上清を100000×gで60分間超遠心分離して全膜をペレット化した。PBS 10mlに0.1%Triton×100または1%n−ドデシル−β−D−マルトシド(DDM)を含有する緩衝液にペレットを再懸濁させ、一晩回転させることにより膜タンパク質を可溶化した。この懸濁液に等体積のPBSを加えて界面活性剤の濃度を低下させ、100000Gで60分間超遠心分離した。可溶化膜タンパク質に対応する上清をニッケルアフィニティータンパク質精製用のカラムに負荷した。
ニッケルアフィニティークロマトグラフィーにより可溶化全膜からヘキサ−ヒスチジンタグ化タンパク質を精製した。簡潔に述べれば、20mMイミダゾールを含有する緩衝液で予め平衡化させたニッケル−ニトリロ三酢酸(Ni−NTA)アガロースカラム(Qiagen社)に全膜を負荷した。未結合タンパク質を除去するため、カラムを4回、それぞれ20mMイミダゾールを含有する緩衝液および30mMイミダゾールを含有する緩衝液で洗浄した。250mMイミダゾールを含有する溶離緩衝液でカラムに結合したHisタグ化タンパク質を6つの画分に溶離させた。
透析を一晩実施した後、0.25%DDMを含有するPBS 250mlからなる透析緩衝液で2時間の透析を2回実施することによりイミダゾールを除去した。DCキット(biorad社)を用いてタンパク質を定量化した後、試料を約6μg/mlに希釈し、マウス1匹当たり0.6μg注射した。マウス10匹からなる2つのグループに非グリコシル化ComPまたはCPSコンジュゲートComPのいずれかを注射した。免疫感作前および免疫感作から7日後と21日後にマウスから血清を採取した。第14日に追加免疫を実施した。
肺炎球菌(S.pneumoniae)血清型14(Statens Serum Institut社、デンマーク)をBHIブロス中、37℃、5%CO2好気条件下で一晩増殖させた。細胞を1×PBSで洗浄し、ODを0.6に調節した。次いで、細胞を60℃で2時間インキュベートすることによって熱不活化した後、Corning高結合96ウェルプレートに固定化した(50μL/ウェル)。プレートを4℃で一晩インキュベートした。翌日、ウェルを1×PBS(100μL/ウェル)で3回洗浄した後、5%脱脂乳で2時間ブロックした。ウェルをPBSで3回洗浄した。次いで、PBSTに溶かした2.5%脱脂乳で1:500に希釈したマウス血清(100μL/ウェル)をプレートに加え、室温で1時間インキュベートした。陽性対照には、血清型14のCPSに対する市販のウサギポリクローナル抗体を用いた(Statens Serum institute社)。陰性対照ウェルは一次抗体の代わりに脱脂乳で処理した。一次抗体とインキュベートした後、ウェルをPBSで3回洗浄した。次いで、二次HRPコンジュゲート抗体(100μL/ウェル)と1時間インキュベートした。使用した抗体は、PBSTに溶かした2.5%脱脂乳で希釈した抗マウスIgM(1:10000)、抗マウスIgG(1:10000)および抗ウサギ(1:5000)HRPコンジュゲート抗体であった。インキュベーション後、ウェルをPBSで3回洗浄し、各ウェルに発色基質TMB(Cell Signaling Technology社)100μLを加え、プレートを室温で5分間インキュベートした後、BioTek(商標)プレートリーダーを用いて650nmでの吸光度を測定した。
本実施例では、大腸菌(E.coli)を用いPglLComPを介してComPとコンジュゲートさせたCPS14を注射することにより、肺炎球菌(S.pneumoniae)血清型14の莢膜に対するIgG免疫応答が生じることを示す証拠が提供される。

Claims (26)
- PglLComPO−オリゴサッカリルトランスフェラーゼを用いるComPのグリコシル化を含む、糖タンパク質の合成方法。
- 前記ComPを任意選択で第二のタンパク質、アジュバントまたはキャリアと融合させる、請求項1に記載の方法。
- グラム陰性菌で実施する、請求項1または2に記載の方法。
- 前記細菌がアシネトバクター(Acinetobacter)または大腸菌(E.coli)である、請求項3に記載の方法。
- 前記グリコシル化が、O−グリコシル化、N−グリカン、O抗原または莢膜多糖体に由来する糖を用いるものである、請求項1〜4のいずれか1項に記載の方法。
- 前記莢膜多糖体がストレプトコッカス(Streptococcus)の莢膜である、請求項5に記載の方法。
- 前記ストレプトコッカス(Streptococcus)がストレプトコッカス・ニューモニエ(Streptococcus pneumoniae)である、請求項6に記載の方法。
- 前記N−グリカンが、カンピロバクター(Campylobacter)に由来するものである、請求項5に記載の方法。
- 前記N−グリカンがカンピロバクター・ジェジュニ(Campylobacter jejuni)のN−七糖である、請求項8に記載の方法。
- 前記アシネトバクター(Acinetobacter)が、A.バイリイ(A.baylyi)、A.バウマンニ(A.baumannii)、A.ノソコミアリス(A.nosocomialis)またはA.カルコアセティカス(A.calcoaceticus)である、請求項4に記載の方法。
- ComPのグリコシル化のためのO−オリゴサッカリルトランスフェラーゼ。
- 前記グリコシル化をグラム陰性菌で組換えにより生じさせる、請求項11に記載のO−オリゴサッカリルトランスフェラーゼ。
- 前記細菌がアシネトバクター(Acinetobacter)または大腸菌(E.coli)である、請求項12に記載のO−オリゴサッカリルトランスフェラーゼ。
- 前記グリコシル化が、O−グリコシル化、N−グリカン、O抗原または莢膜多糖体に由来する糖を用いるものである、請求項11〜13のいずれか1項に記載のO−オリゴサッカリルトランスフェラーゼ。
- 前記莢膜多糖体が、ストレプトコッカス(Streptococcus)由来のものである、請求項14に記載のO−オリゴサッカリルトランスフェラーゼ。
- 前記ストレプトコッカス(Streptococcus)がストレプトコッカス・ニューモニエ(Streptococcus pneumoniae)である、請求項15に記載のO−オリゴサッカリルトランスフェラーゼ。
- 前記N−グリカンが、カンピロバクター(Campylobacter)に由来するものである、請求項14に記載のO−オリゴサッカリルトランスフェラーゼ。
- 前記N−グリカンがカンピロバクター・ジェジュニ(Campylobacter jejuni)のN−七糖である、請求項17に記載のO−オリゴサッカリルトランスフェラーゼ。
- PglL ComPである、請求項13〜18のいずれか1項に記載のO−オリゴサッカリルトランスフェラーゼ。
- 請求項1〜10のいずれか1項に記載の方法に従って合成した糖タンパク質を含む、ワクチン。
- ストレプトコッカス(Streptococcus)ワクチンである、請求項20に記載のワクチン。
- 前記ストレプトコッカス(Streptococcus)がストレプトコッカス・ニューモニエ(Streptococcus pneumoniae)である、請求項21に記載のワクチン。
- ComPまたはグリコシル化部位を有するComPのフラグメントを含む融合タンパク質である、糖タンパク質。
- タンパク質のグリコシル化へのPglLComPの使用。
