Abstract
Description
全てのキット試薬をそれらの使用前に室温にする。
バクテリア細胞を含有する生体試料を室温で1−6時間、マイナス20℃の温度で保存されていた場合は好ましくは1.5から3時間、例えば2から2.5時間、+4℃で保存されていた場合、約60分、例えば30分間放置する。
ゆっくりかつ穏やかに攪拌しながら、ビーズを含有する懸濁液を置く。
・ 液体形態の試料の場合、約105−107細胞/mlの濃縮倍率が達成されるまで、0.1%滅菌ペプトン生理食塩水で1:10の段階希釈を行う。
・ 無水試料の場合、滅菌バッグ中で試料を1:10で水を加えて元に戻し、そして、ストマッキングして調製物を均質化する。次に、液体形態の試料の場合などのその後の1:10の希釈が行われる。
・ 2分間室温で攪拌およびインキュベートしながら0.5mlの適した希釈液に、2.5μlのチアゾールオレンジTOおよび1.5μlのヨウ化プロピジウムPIを加え;かつ
・ ビーズを含有する50μlの懸濁液を加えて、試料を細胞蛍光測定解析に供する。
−段階希釈により、105から107細胞/mlの濃度を有するラクトバチルス・ラムノサスGG(ATCC53103)の間欠滅菌されたバクテリア細胞を含有する試料を調製すること;
−試料に第一試薬チアゾールオレンジおよび第二試薬ヨウ化プロピジウムを加えて、溶液を得ること;
−溶液にアジ化ナトリウム中の蛍光微粒子の懸濁液を加えて、試験試料を得ること;
−試験試料をフローサイトフルオロメトリーによる生細胞および死細胞を含む総細胞の計数並びに死細胞だけの計数に供すること;
−間欠滅菌された試料中に存在する死細胞を計数すること。
−1x106から1x1010UFC/gの濃度を有するラクトバチルス・ラムノサスGG(ATCC53103)のバクテリア細胞培養物を調製すること;
−培養物を、間欠滅菌プロセスに供して、間欠滅菌されたバクテリア細胞の培養物を得ること;
−上述の計数方法を適用すること。

Claims (8)
- ラクトバチルス・ラムノサス(Lactobacillus rhamnosus)GG(ATCC53103)の間欠滅菌されたバクテリア細胞の試料中のインタクトな細胞膜を有する死細胞の数を計数するための方法であって:
−段階希釈により、105から107細胞/mlの濃度を有するラクトバチルス・ラムノサスGG(ATCC53103)の間欠滅菌されたバクテリア細胞を含有する試料を調製すること;
−試料に第一試薬チアゾールオレンジおよび第二試薬ヨウ化プロピジウムを加えて、溶液を得ること;
−溶液にアジ化ナトリウム中の蛍光微粒子の懸濁液を加えて、試験試料を得ること;
−試験試料をフローサイトフルオロメトリーによる生細胞および死細胞を含む総細胞の計数並びに死細胞だけの計数に供すること;
−間欠滅菌された試料中に存在する死細胞を計数すること、
を含む、方法。 - 0.5mlの、105から107細胞/mlの濃度を有するラクトバチルス・ラムノサスGG(ATCC53103)の間欠滅菌されたバクテリア細胞を含有する試料に、2.5マイクロリットルの第一試薬および1.5マイクロリットルの第二試薬が加えられ、溶液を形成する、請求項1に記載の方法。
- アジ化ナトリウム中の蛍光微粒子の懸濁液がポリスチレン微粒子を含み、好ましくはビーズ懸濁液が、アジ化ナトリウム0.1%溶液中の蛍光ポリスチレン微粒子の懸濁液である、請求項1または2に記載の方法。
- 10から100μl、好ましくは40から60μlが含まれる、既知の量のビーズが加えられ、細胞数の決定を可能にする、請求項3に記載の方法。
- インタクトな細胞壁を有するラクトバチルス・ラムノサスGG(ATCC53103)の間欠滅菌されたバクテリア細胞を製造するための方法であって、:
−1x106から1x1010UFC/gの濃度を有するラクトバチルス・ラムノサスGG(ATCC53103)のバクテリア細胞培養物を調製すること;
−培養物を、間欠滅菌プロセスに供して、間欠滅菌されたバクテリア細胞の培養物を得ること;
−請求項1−4のいずれか一項に記載の計数方法を適用すること、
を含む、方法。 - 請求項5に記載の方法により得られる、ラクトバチルス・ラムノサスGG(ATCC53103)の、間欠滅菌され、インタクトかつ免疫学的に活性なバクテリア細胞の培養物。
- ラクトバチルス・ラムノサスGG(ATCC53103)の間欠滅菌されたバクテリア細胞の試料中のインタクトな細胞膜を有するラクトバチルス・ラムノサスGG(ATCC53103)の、間欠滅菌された、死んだバクテリア細胞の数を計数するためのフローサイトフルオロメトリーの使用。
- インタクトな細胞膜を有するラクトバチルス・ラムノサスGG(ATCC53103)の、間欠滅菌され、死んだバクテリア細胞が、請求項1−4のいずれか一項に記載の方法により計数される、請求項7に記載の使用。
