Abstract
【解決手段】標的細胞を12.8kDaから約15kDaの分子量を有するポロキサマーおよびレトロウイルスベクターと接触させる段階を含む標的細胞に形質導入するための方法、ポロキサマーをポリカチオン性重合体またはポリカチオン性ペプチドと組み合わせて標的細胞をレトロウイルスベクターで形質導入するための使用、および、ポロキサマーとポリカチオン性物質とを含むキット。

Description
(1)標的細胞を12.8kDaから約15kDaの分子量を有するポロキサマーおよびレトロウイルスベクターと接触させることを含む、標的細胞に形質導入するための方法;
(2)標的細胞がリンパ球、腫瘍細胞、リンパ球系細胞、神経細胞、上皮細胞、内皮細胞、初代細胞、幹細胞からなる群から選択される細胞である、(1)の方法;
(3)リンパ球が初代リンパ球であり、および/または腫瘍細胞が造血腫瘍細胞、ニューロン腫瘍細胞または上皮腫瘍細胞である、(2)の方法;
(4)レトロウイルスベクターがレンチウイルスベクターである、(1)〜(3)のいずれかの方法;
(5)レンチウイルスベクターが、VSV−Gで、および/またはVSV−Gに融合された抗体断片で偽型化される、(4)の方法;
(6)ポロキサマーが、式HO−[CH2CH2O]x−[CH2C2H4O]z−[CH2CH2O]y、ここで、平均でx+y=265.45およびz=50.34、を有するか;またはポロキサマーが、式HO−[CH2CH2O]x−[CH2C2H4O]z−[CH2CH2O]y、ここで、平均でx+y=236.36およびz=44.83、を有する、(1)〜(5)のいずれかの方法;
(7)該標的細胞がさらにポリカチオン性重合体またはポリカチオン性ペプチドの群から選択される1つ以上のポリカチオン性物質と接触される、(1)〜(6)のいずれかに記載の方法;
(8)該ポリカチオン性重合体が、ポリ(エチレングリコール)−ポリ(L−リジン)ブロック共重合対(PEG−PLL)および1,5−ジメチル−1,5−ジアザ−ウンデカ−メチル−ポリメソブロミド(ポリブレン)からなる群から選択される;および/または該ポリカチオン性ペプチドが、硫酸プロタミンおよび平均分子量1から300kDaを有するポリ−l−リジン(PLL)からなる群から選択される、(7)の方法;

細胞株および化学物質
ヒト胚腎臓細胞HEK293Tは、10%(vol/vol)ウシ胎仔血清(FCS)および2mMグルタミンが補われたDMEMで増殖した。未分化大細胞リンパ腫細胞株KARPAS−299、SUDHL−1、SR−786、およびSUP−M2は10%FCSと2mMグルタミンが補われたRPMI1640で、膵癌細胞株AsPC−1は20%FCS、2mMグルタミンおよび1mMピルビン酸ナトリウムとのPRMI1640中で、およびPANC−1は10%のFCSおよび4mMグルタミンが補足されたDMEM中で、培養された。すべての化学的アジュバントの候補は、Sigma−aldrichから購入されて、100mg/mlの原液を得るために水に溶解された。
6ウェルプレートにおいて、1ウェルあたり2×105の細胞が三重に播種され、1μg/mlから1000μg/mlの範囲内で規定されたアジュバント濃度で処理された。24時間のインキュベーション(5%CO2、37℃)の後に、培地を新鮮な培地に交換し、細胞をさらに48時間インキュベートした。製造業者の指示に従い、細胞増殖はWST−1比色分析細胞増殖アッセイ(Roche)で決定された。まもなく、細胞は10μlのWST−1基質での2時間の100μl細胞培養(5%CO2、37℃)に続いて、トリプシン処理された。アッセイは96ウェルマイクロタイタープレートで実施され、吸光度はTECAN−Infiniteマイクロプレートリーダー(TECAN)を使用して、490nmで測定された。
レンチウイルス導入ベクター、pGreenPuro(pGP)およびpSIH1−H1−copGFP(pSIH1)(System Biosciences)は内部CMVプロモーターによって駆動されたcopGFPの発現を許容する。複製欠失レンチウイルス粒子(GP)は、製造業者の指示に従い、トランスフェクション試薬としてリポフェクタミン2000(Life Technologies)を使用して、16μg、8μgおよび4μgのパッケージプラスミドpMDLg/pRRE、pRSV.RevおよびpMD2.G(D. Trono, Ecole polytechnique federale de Lausanneからの好意)および8μgのpGPベクターでの、10cmペトリ皿中のHEK293T細胞の一時的な同時トランスフェクションで作製された。次に、レンチウイルス粒子(SIH1)は、業者の指示(System Biosciences)に従い、pPackH1−プラスミドパッケージングミックスを使用してHEK293T細胞内で作製された。
HEK293T細胞(1ウェルあたり2×105細胞)はアジュバントの有無にかかわらず、レンチウイルス(GP)を含む1mlの培地でカバーされた。膵臓癌AsPC−1およびPANC−1細胞(1ウェルあたり104細胞)は、アジュバントの有無にかかわらず、レンチウイルス(SIH1)を含む250μl上清でカバーされた。特定の場合、プレートは60分間、1,000gで遠心分離された。24時間のインキュベーション(5%CO2、37℃)の後に、上清は新鮮な培地に交換され、さらに48時間インキュベートされた。感染後48時間で組み合わせ顕微分析および照合を実施した(HBO 50/AC and AxioCam MRC, Carl Zeiss AG)。
ヘルシンキ宣言で示された原則および地方の倫理委員会での許容および要求に従って
2人の健康なドナーのPBMCが回収された。PBMCは、フィコール勾配遠心分離を通して単離され、ヒト血清、50U/ml IL−2(Chiron Vaccines)および50ng/ml OKT3(LGC Standards)が補足されたRPMI培地で3日間以上培養された。活性化の後に、1ウェルあたり5×105の細胞がアジュバント有りまたは無しのレンチウイルス(GP)を含む500μlの補足培地で再懸濁された。プレートは、90分間、800gで遠心分離され、一晩培養され、その後洗浄および新鮮な補足培地で再懸濁され、さらに24時間インキュベートされた。
レンチウイルス形質導入後に、細胞はPBSで洗浄され、1μg/mlのヨウ化プロピジウム(Invitrogen)とPBS中に再懸濁された。氷上で10分間インキュベーションした後、30000の事象が、FACSDiva(BD Biosciences)で前方/側方錯乱光特性、530nmでの緑色蛍光放射、および610nmでの赤色蛍光放射を分析された。アポトーシス測定に関して、細胞はPBSで洗浄され、70%(vol/vol)のエタノールで再懸濁された後、氷上で45分間インキュベーションされた。固定の後に、細胞は遠心分離され、ペレットは40μg/mlのヨウ化プロビジウムおよび100μg/mlのRNAse(Qiagen)と共にPBSで再懸濁された。細胞はFACSDivaで赤色蛍光放射のみ分析され、30000の事象が、アポトーシスを起こした細胞のSub−G1割合(<2n)を測定するために分析された。
すべての実験が少なくとも二重で行われた。対応するグラフでは、他に言及がない場合、平均値±標準誤差(s.e.m.)で表される。
レンチウイルス遺伝子導入のための潜在的アジュバント物質から、我々はポリカチオン様のクラスから5種類およびポロキサマー様化学物質のクラスから7種類の候補を選択した(表1)。HEK293T細胞は規定された細胞濃度で共インキュベーションされ、その後培地を変更して、細胞は細胞増殖の測定の前にさらに48時間インキュベートされた。異なる物質での特定のHEK293T増殖阻害を試験するため、我々は、WST1基質の取り込みおよび物質との共インキュベーションなしの対照細胞集団との相関を測定した(図1)。この手順は、LVおよびアジュバント(ポリブレン)が洗浄される前に、標的細胞との24時間のLVインキュベーションを含む一般的な形質導入プロトコルに応じて、準備された。
HEK293T細胞は、LVと最大許容濃度(1000μg/ml)の特定のアジュバントの混合物でカバーされた(24時間)。CopGFP−コーディング遺伝子を運ぶLVは、2.5から0.0025の減少しているMOIで適用された。培地変更の後、細胞は、さらに48時間培養され、緑色蛍光放射がフローサイトメトリーで測定された。図3aでは、4つの代表的実験がドットブロットで表される。アジュバント無しの形質導入は、全体のMOI範囲にわたって、低導入効率を示した。ポリブレンは、推奨濃度10μg/mlで、導入効率を31.5%から48.4%(MOI0.25)まで増加させる能力を有した。
前臨床的な研究において、多くの確立した腫瘍細胞株が、一般的なウイルス性および非ウイルス性のツールで低い導入率のみを提供する。浮遊培養で増殖する未分化大細胞リンパ腫(ALCL)細胞株は、腫瘍細胞株のこの感染させにくいサブセットに属する8。LV感染中にスピノキュレーション段階を含む改良プロトコルで、我々は、シンペロニックF108がKARPAS−299、SR−786、SUDHL−1、およびSUP−M2リンパ腫細胞のレンチウイルス感染を容易にするかどうか試験した(図4b)。とりわけ、同じ平均MOI率(1.5)で、Karpas−299細胞はポリブレンの存在下で20.8%感染され、SR−786は既に58.8%の感染率を示した(図4a)。KARPAS−299細胞は、LVとのプレミックスにおいて、ポリブレン存在下での58.4%から、シンペロニックF108存在下での85.2%まで、GFP−陽性細胞の増加を示した(図4b)。下側のMOI範囲で、ポリブレン補助形質導入率は21.9%から33.7%またはそれぞれ41.0%まで、併用処理(ポリブレンおよびシンペロニックF108添加)で高められる。SR−786細胞の形質導入率は、KARPAS−299セルと同様に、64.9%(ポリブレン)から84.1%(シンペロニックF108)までMOI1.5で増加した。他のリンパ腫細胞株(SUDHL−1およびSUP−M2)はシンペロニックF108によるLV送達の増進において類似作用を示した。(図7a)。
前の実験では、非イオン性ポロキサマー、シンペロニックF108はレンチウイルス形質導入の補助において、当分野でアジュバントとして知られるポリブレンよりも良い効率を示した。ポロキサマーは直接細胞膜と相互作用すると説明されるので、我々は、標的細胞の透過状態がシンペロニックF108によって変えられるかどうか検討した。リンパ腫細胞膜の透過能力をモニターするための方法として、我々は、ポリブレン、シンペロニックF108または両方の物質とのインキュベーションの後に、細胞への取り込みのためにヨウ化プロピジウム(PI)を投与した。一般に、PIは非常に透過性の、または、そうでなければ、漏出性細胞中にのみ蓄積する(図4c)。SUDHL−1細胞を除いて、すべてのリンパ腫細胞株が、ポリブレン処理と比較して、シンペロニックF108の存在下で著しく高いPI透過率を示した(図4dおよび図7b)。アポトーシスを起こした細胞(<2n)のバックグラウンドをモニターするために、我々は並列断片化DNA(parallel fragmented DNA)を測定した(図4c、d)。これらの知見は、シンペロニックF108の、細胞膜を通してレトロウイルス粒子の成功できる形質導入を許容する好ましい無毒の膜効果を示す。
細胞株実験では、シンペロニックF108は特にポリカチオン、ポリブレンと組み合わせてレンチウイルス感染で高性能を示した。初代細胞がそれらのウイルス性遺伝子導入への敏感さにおいて実質的に異なるように、我々は、2人の健康なドナーの血液から単離した初代リンパ球の活力およびレンチウイルス形質導入率を評価した。両方の試料は、同じスピノキュレーションプロトコルで処理された細胞株と比較して、高いMOIレベルであっても(2.5から25)、より低い、ドナー依存の形質導入率を示した(図5)。シンペロニックF108単独およびポリブレンとの組み合わせは、第2因子(22.2%から41.7%、ドナー#1、図5a)によって、または第4因子(5.2%kara19.2%,ドナー#2、図5b)によってさえ、初代リンパ球の形質導入の増加に成功した。シンペロニックF108単独だけでなく組み合わせ処理も、それらの標的細胞における顕著な細胞死を引き起こさなかった。リンパ腫細胞株および初代リンパ球で得られた結果は、シンペロニックF108のレンチウイルス粒子の形質導入アジュバントとしての有望な役割を明確に示す。
HEK293T細胞は、特定のアジュバント、5000μg/mlの最大適用濃度までのシンペロニックF108と実施例3で使用されたLVの混合物でカバーされた(24時間)。CopGFP−コーディング導入遺伝子を運ぶ、実施例3で使用されるレンチウイルス粒子(GP)は、MOI0.25、および0.025で適用された。培地交換の後、細胞はさらに48時間培養され、緑色蛍光放射がフローサイトメトリーで測定された。シンペロニックF108は、試験した全ての濃度で目立った毒性なしで、よく許容された(1000μg/ml、2500μg/mlおよび5000μg/ml)。シンペロニックF108は、ポリブレン補助LV感染よりも良い感染を示した(図3c)。ポリブレン誘導形質導入と比較して、シンペロニックF108単独またはポリブレン(10μg/ml)と組み合わせは、感染細胞の平均量(2つの独立した実験)を増加させた。とりわけ、MOI0.25で、感染細胞の量が49.5%(ポリブレン10μg/ml)から、シンペロニックF−108が(1000μg/ml)の濃度で使用されたときに、66.2%、2500μg/ml濃度で68.2%、5000μg/ml濃度で64.3%まで、増加した。シンペロニックF108がポリブレン(10μg/ml)と組み合わせて使用されたとき、平均感染はさらに69.7%(1000μg/mlのシンペロニックF108)、71.2%(2500μg/mlのシンペロニックF108)、および64.8%(5000μg/mlのシンペロニックF108)に増加した。アジュバント無しで、0.25のMOIを使用するとき、23%の細胞の平均形質導入が観察された。3つの独立した生物学的反復からの二標本両側スチューデントt−検定での統計分析(表2、下記)は、ポリブレン(10μg/ml)およびシンペロニックF108(1000μg/ml)の両方がHEK293細胞の感染を有意な(p<0.0)増加で高く仲介したこと、およびシンペロニックF108(1000μg/ml)は、ポリブレン(10μg/ml)と比較して有意に(p<0.05)HEK293細胞感染を増加したことを示した。
LV感染の間にスピノキュレーション段階を含む改良プロトコルで、シンペロニックF108は、KARPAS−299およびSR−786リンパ腫細胞のレンチウイルス感染を容易にすることについて、最大適用濃度(5000μg/ml)まで試験された(図4e)。CopGFP−コーディング遺伝子を運ぶ実施例3で使用されたレンチウイルス粒子(GP)は、MOI2.5および0.25で適用された。シンペロニックF108は、試験した全ての濃度で目立った毒性なしで、よく許容され(1000μg/ml、2500μg/mlおよび5000μg/ml)、単独およびポリブレンとの組み合わせで、ポリブレン補助LV感染のみと比較して、より良い感染を示した。とりわけ、MOI2.5で、59.5%のKarpas−299細胞がポリブレン(10μg/ml)単独の存在下で感染し、他方で、シンペロニックF−108が1000μg/mlの濃度で存在すると83.3%、2500μg/ml濃度で81.7%、5000μg/ml濃度で77.5%感染した。KARPAS−299細胞が、同じMOI(2.5)で、シンペロニックF108およびポリブレン(10μg/ml)の組み合わせを使用したとき、感染細胞の量はさらに86%(1000μg/mlシンペロニックF108、10μg/mlポリブレン)、87.2%(2500μg/mlシンペロニックF108、10μg/mlポリブレン)、および78.9%(5000μg/mlシンペロニックF108、10μg/mlポリブレン)まで増加した(図4e)。
LV感染の間のスピノキュレーション段階効果はKARPAS−299およびSR−786リンパ腫細胞のレンチウイルス感染を容易にするアジュバントの使用と組み合わせて試験された(図4f)。CopGFP−コーディング導入遺伝子を運ぶ実施例3で使用されたレンチウイルス粒子(GP)は、アジュバントの存在下、スピノキュレーション(室温、90分間、800gの遠心分離)プロトコルを適用および適用せずに、2つの異なったMOI(2.5および0.25)で適用された。試験されたすべてのMOIで、スピノキュレーションプロトコルは細胞の、より良い感染をもたらした。とりわけ、MOI2.5でのスピノキュレーションプロトコルで、59.5%のKarpas−299細胞がポリブレン(10μg/ml)単独の存在下で感染し、他方で、シンペロニックF−108が1000μg/mlの濃度で存在すると83.2%、ポリブレンおよびシンペロニックを両方使用すると86%感染した。MOI2.5で、62.2%のSR−786細胞がポリブレン(10μg/ml)単独の存在下で遺伝子導入され、シンペロニックF−108(1000μg/ml)で82.3%、両方のアジュバントを使用して88%が導入された。

Claims (15)
- 標的細胞を12.8kDaから約15kDaの分子量を有するポロキサマーおよびレトロウイルスベクターと接触させることを含む、標的細胞に形質導入するための方法。
- 標的細胞がリンパ球、腫瘍細胞、リンパ球系細胞、神経細胞、上皮細胞、内皮細胞、初代細胞、幹細胞からなる群から選択される細胞である、請求項1に記載の方法。
- リンパ球が初代リンパ球であり、および/または腫瘍細胞が造血腫瘍細胞、ニューロン腫瘍細胞または上皮腫瘍細胞である、請求項2に記載の方法。
- レトロウイルスベクターがレンチウイルスベクターである、請求項1〜3のいずれか1項に記載の方法。
- レンチウイルスベクターが、VSV−Gで、および/またはVSV−Gに融合された抗体断片で偽型化される、請求項4に記載の方法。
- ポロキサマーが、式HO−[CH2CH2O]x−[CH2C2H4O]z−[CH2CH2O]y、ここで、平均でx+y=265.45およびz=50.34、を有するか;またはポロキサマーが、式HO−[CH2CH2O]x−[CH2C2H4O]z−[CH2CH2O]y、ここで、平均でx+y=236.36およびz=44.83、を有する、請求項1〜5のいずれか1項に記載の方法。
- 該標的細胞がさらにポリカチオン性重合体またはポリカチオン性ペプチドの群から選択される1つ以上のポリカチオン性物質と接触される、請求項1〜6のいずれか1項に記載の方法。

- ポリカチオン性物質が、1,5−ジメチル−1,5−ジアザ−ウンデカ−メチル−ポリメソブロミドおよび/または硫酸プロタミンである、請求項8に記載の方法。
- 該ポロキサマーが、約50から5000μg/mlの濃度で提供される、請求項1〜9のいずれか1項に記載の方法。
- 該ポロキサマーが、約500から1000μg/mlの濃度で提供される、請求項10に記載の方法。
- 該標的細胞と該ポロキサマーを接触させる前、させると同時またはさせた後に、該標的細胞と該レトロウイルスベクターをスピノキュレーションさせる(spinoculating)さらなる段階を含む、請求項1〜11のいずれか1項に記載の方法。
- 請求項1〜12のいずれか1項に規定されるポロキサマーの、任意に請求項7〜9のいずれか1項に規定されるポリカチオン性物質と組み合わせての、標的細胞をレトロウイルスベクターで形質導入するための使用。
- 請求項1〜12のいずれか1項に規定されるポロキサマー、請求項7〜9のいずれか1項に規定されるポリカチオン性重合体および/またはポリカチオン性ペプチド、および任意に使用説明書を含むキット。
- 請求項1〜12のいずれか1項に規定されるレトロウイルスベクターをさらに含む、請求項14に記載のキット。
