I.バイアルリングを形成しない脱気組成物の調製
組換えタンパク質を含む特定の組成物のバイアルにリングが形成され得ることが分かっている。「リング」とは、本明細書では、バイアル又は他の容器の空気−液体−ガラスの界面における粒子の堆積のことであり、バイアル又は他の容器に見て明らかな円形又は楕円形の見かけ上の欠陥をもたらす。一実施形態では、組換えタンパク質を含む組成物は、界面活性剤を含まない。一実施形態では、組換えタンパク質を含む組成物は、その製造プロセス中に界面活性剤が添加されない。特定の場合には、組換えタンパク質を含む組成物は、少なくとも約24時間、少なくとも約36時間、又は少なくとも約48時間、少なくとも約3日間、少なくとも約4日間、少なくとも約1週間、少なくとも約1か月間、少なくとも約6か月間、少なくとも約1年間、少なくとも約2年間、少なくとも約3年間陽圧下で保存されている。
限定されるものではないが、真空の強度、真空が適用される期間、及び容器及び溶液の特性(例えば、限定されるものではないが)、溶液の量、液体の高さ、及び空気の高さ/ヘッドスペース)を含む様々な因子が、組換えタンパク質を含む組成物を脱気する真空の能力に影響を与える。任意の状況で適用され得る真空の程度及び真空の期間は、様々であり得るが、試験バッチの実施及び脱気が任意のゴールを達成するのに十分であるか否かの観察によって定性的に決定することができる。別法では、特定の実施形態のみにおいて、真空の程度及び真空の期間は、以下の式に従った気泡指数の計算を用いて決定することができる:

脱気ステップの様々な利点が以下に説明され得るが;全ての利点を、それぞれの状況で提示することができるわけではない。一態様では、脱気ステップにより、組換えタンパク質を含む組成物を陽圧下で保存することができる。別の実施形態では、組換えタンパク質を含む組成物は、界面活性剤を含まない。別の実施形態では、界面活性剤が、組換えタンパク質を含む組成物に添加されない。
このプロセスは、組換えタンパク質を含む複数の組成物に適用することができる。一実施形態では、組換えタンパク質は抗体である。別の実施形態では、組換えタンパク質は、RSVに対する抗体である。
本脱気方法は、組換えタンパク質を含む組成物の多数の異なる生産プロセスに組み入れることができる。特定の生産プロセスは、本明細書に概略が説明されるが、本脱気方法は、本明細書に概略が説明される利点を達成するために他の製造プロセスに含めることができる。一実施形態では、脱気方法が使用されない場合、充填ラインの気泡が存在し、かつ/又は脱気方法が使用されない場合、リングが、バイアル又は他の容器に存在する。

脱気に対する真空、量、及び時間の影響を決定するために試験を行った。65Lのタンクを用いて、原薬を濾過して清浄なタンクに入れ、20psigに加圧し、2〜8℃で1週間保存して溶存酸素を増加させた。脱気のために、真空を適用して目標の真空を達成し、弁を閉じ、指定の期間脱気した。
実施例1のプロトコルに続いて、1mg/mlのMEDI−524を含む組成物の脱気に対する真空の影響を評価した。脱気圧力は、99mbar、268mbar、及び505mbarと異なるようにした。脱気の時間は、以下の表に示されているように1〜4日間とした。

実施例1のプロトコルに続いて、1mg/mlのMEDI−524を含む組成物の脱気に対する時間の影響も、40Lの固定量の溶液及び99mbarの固定圧力で評価した。脱気の時間は、24時間及び46時間(約1日間又は2日間をシミュレーションする)と異なる条件にした。
実施例1のプロトコルに続いて、組成物の脱気に対するタンパク質濃度の影響も、40Lの固定溶液量及び99mbarの固定圧力で2日(以下に示されるように実際の時間は僅かに異なる)の期間で評価した。1mg/mlの濃度(MEDI−524)と100mg/ml(Synagis(登録商標))の両方を評価した。気泡は、試験した条件のいずれでも観察されなかった。結果は、表6に示されている。
気泡形成の発生を予測するために、以下のパラメーターを考慮して、気泡指数の式に適用した:

Synagis(登録商標)バイアル、言い換えれば、例えば、50mg及び100mgのバイアルでリングを観察した。これは、指示矢印の左側のバイアルのガラスに実質的に水平に示されている白色リングとして図4に示されているように、ガラス壁の側面に固着した空気−液体界面に位置する非常にかすかな白色リングとして出現する。
・溶存ガス及び気泡は、Synagis(登録商標)の原薬プロセスにつきものである;
・気泡が存在して界面活性剤が存在しないと、バイアルにリング形成が生じる;
・薬品充填最終プロセスは、リング形成に影響を与えず、タンクから手動で直接充填された原薬にリングが形成される;
・リングの存在は、製品の品質に影響を与えないが、視覚的に厄介な見かけ上の欠陥であり得る;
・トリプシン消化によるリングの分離は、リング中のタンパク質がSynagis(登録商標)であることを示す;
・リング形成は、小規模で再現可能である。
Synagis(登録商標)バイアルの壁に形成されるリングの組成を決定するために評価を行った。FTIRを使用して、フィルター表面に収集したリング分離物を確認した。上記のように調製したサンプルを、FTIR顕微鏡を用いて分析した。収集物は、IRシグネチャが、タンパク質(アミド−Iバンド;1600〜1700cm−1及びアミド−IIバンド;1510〜1580cm−1)及びポリジメチルシロキサン(1260cm−1)に一致することを示した。アミドIIバンドの第2の誘導体は、典型的には天然IgG分子で観察される天然βシート構造に一致する1638cm−1における主要なバンドを示した。これらの結果は、リングタンパク質が天然βシート構造から構成されていることを示唆している。熱又はせん断応力IgG1は、第2の誘導体の主ピークを1628cm−1にシフトさせ、これは、凝集(可溶性又は不溶性)と共に示される分子間相互作用を示唆している。FTIRは、リングがタンパク質及びシリコーン油を含むことを示している。
界面活性剤PS−80(0.02%)を含む又は含まない原薬を加圧して、溶存ガス及び気泡を増加させた。バイアルを、M&O Perryフィルターを用いてステンレス鋼回転ピストンポンプで充填した。約160mLのパリビズマブDSを、2つの250mLのPETG容器のそれぞれに濾過注入した。1つの容器を、0.02%ポリソルベート80(PS−80)でスパイクした。両方のPETG容器を、特注のステンレス鋼タンクに入れて、20psigの空気で加圧した。タンクを、オービタルシェーカーで約43時間、2〜8℃で保存して溶存ガス/気泡を増加させた。各PETG容器からM&O Perryフィルターを用いてB&S回転ピストンポンプで、約112の3ccのバイアルに充填した。バイアルを、それぞれ56のバイアルの2つのセットに分けて、リングを検出するために黒色及び白色の背景で、充填の2日後及び7日後に目視検査した。対照バイアルは全てリングを有していたが、PS−80スパイクバイアルは1つもリングを有していなかった。気泡は、対照及びPS−80スパイク原薬の両方の充填ラインで観察された。これは、PS−80が、タンパク質の空気−液体界面への吸着(気泡)を防止するという仮説を立証している。

Claims (20)
- 組換えタンパク質を含む組成物を含むバイアルを製造する方法であって:
a.組換えタンパク質を含む組成物を含む容器を用意するステップであって、任意選択により、前記容器が、陽圧下で保存されている、ステップ;
b.前記容器に真空を適用するステップ;
c.前記真空により前記組成物を脱気するステップ;及び
d.組換えタンパク質を含む前記脱気組成物でバイアルを充填するステップを含む、方法。 - 前記組換えタンパク質が、Synagis(登録商標)である、請求項1に記載の方法。
- 前記組成物が:
a.配列番号1のアミノ酸配列を有する重鎖及び配列番号6のアミノ酸配列を有する軽鎖;
b.配列番号1又は配列番号2の重鎖可変領域、及び配列番号6の軽鎖の軽鎖可変領域;又は
c.アミノ酸配列TSGMSVG(配列番号:3)を有するH1相補性決定領域(CDR)、アミノ酸配列DIWWDDKKDYNPSLKS(配列番号:4)を有するH2 CDR、アミノ酸配列SMITNWYFDV(配列番号5)を有するH3 CDR、アミノ酸配列KCQLSVGYMH(配列番号7)を有するL1 CDR、アミノ酸配列DTSKLAS(配列番号8)を有するL2 CDR、及びアミノ酸配列FQGSGYPFT(配列番号9)を有するL3 CDRを含む、請求項1に記載の方法。 - 組換えタンパク質を含む組成物を含む前記容器が陽圧下で保存されている、請求項1に記載の方法。
