抗体及び抗体断片
本発明者等は、ヒトCK8タンパク質(ref.: Uni-ProtKB P05787)の353位〜360位のアミノ酸配列を有するペプチドに特異的に結合する抗体又は抗体断片を開発した。ヒトCK8タンパク質の353位〜360位のアミノ酸配列は、配列番号29に提示される。「特異的に」とは、抗体又は抗体断片が、上記配列を有するヒトCK8ペプチドのみに結合することを意味する。抗体又は抗体断片は、上記配列を含む任意のタンパク質/ペプチド断片に結合することができることが理解されよう。

本発明はまた、8〜16個のアミノ酸を有するポリペプチドからなり、かつ配列番号29の配列を含むヒトCK8抗原ペプチドに関する。特に、本発明は、配列番号29に従う配列からなるヒトCK8抗原ペプチドに関する。
本発明はまた、ヒトCK8タンパク質(配列番号29)の353位〜360位のアミノ酸に相当する配列を有するヒトCK8に由来するペプチドをコードするポリヌクレオチドに関する。好ましくは、本発明のポリヌクレオチドは、DNA型、特に二重鎖DNA型を有する。「ポリヌクレオチド」という用語はまた、修飾ポリヌクレオチドも指す。
本発明はまた、本発明の抗体又は抗体断片を含む薬学的組成物又はキットに関する。抗体又は抗体断片は、上述の通りであり得る。特に、抗体又は抗体断片は、Fv、Fab、F(ab')2、scFv、抗体、好ましくはモノクローナル抗体である。
CK8に特異的なマウスモノクローナル抗体は、標準的なハイブリドーマ技法を使用して生産した(Zola等、Aust J. Exp Biol Med Sci. 1981年; 59:303〜6頁)。2つの異なるCK8関連ペプチドを合成して、マウス免疫化に使用した。ペプチド配列は、下記の通りである:

樹立ヒト結腸がん(CC)Isreco-1、Isreco-2、Isreco-3、HCT116+/+、HCT116-/-又はHT29(CLB)を、10%熱失活ウシ胎児血清(FBS)(Sigma社、St Quentin Fallavier、フランス)、4nMのL-グルタミン(Sigma社、St Quentin Fallavier、フランス)及び50U/mL、50μg/mLのペニシリン-ストレプトマイシン(Sigma社、St Quentin Fallavier、フランス)を補充したダルベッコ変法イーグル培地(Sigma社、St Quentin Fallavier、フランス)中で成長させた。
細胞表面でのCK8細胞発現を、フローサイトメトリーにより分析した。簡潔に述べると、96ウェル当たり2.105個の細胞を、10μg/mLの非結合型抗CK8マウスMabの希釈物とともにインキュベートした後、1/10で希釈した。結合されていない抗体を、1%ウシ血清アルブミン(Sigma社、St Quentin Fallavier、フランス)を補充したPBS(Invitrogen社、Villebon sur Yvette、フランス)で洗い流した。続いて、細胞を遠心分離して(400gで5分)、結合した抗体を、フルオレセインイソチオシアネート(FITC)結合型ヤギ(Fab')2ポリクローナル抗マウス(MP Biomedical社、Illkirch、フランス)を用いて、4℃で30分間検出した。検出試薬を洗い流して、細胞を遠心分離して(400gで5分)、PBS 300μL中に懸濁させた。結合された検出抗体を、FACS CAN(BD Biosciences社、Rungis、フランス)で定量化する(FL1チャネル、1回の獲得につき2000回の事象)。
96ウェル当たり2.105個のIsreco-1細胞を、種々の濃度の試験したペプチド1若しくはペプチド2を伴って、又は伴わずに、ビオチン化抗体抗CK8(12.5μg/mL)とともにインキュベートして、4℃で30分間インキュベートした。結合されていない抗体を、1%ウシ血清アルブミン(Sigma社、St Quentin Fallavier、フランス)を補充したPBS(Invitrogen社、Villebon sur Yvette、フランス)で洗い流した。続いて、細胞を遠心分離して(400gで5分)、結合した抗体を、フィコエリトリン結合型ストレプトアビジン(Interchim社、Montlucon、フランス)を用いて、4℃で30分間検出した。検出試薬を洗い流して、細胞を遠心分離して(400gで5分)、PBS 300μL中に懸濁させた。結合された検出抗体を、FACS CAN(BD Biosciences社、Rungis、フランス)で定量化した(FL2チャネル、1回の獲得につき2000回の事象)。実験中、各々のアイソタイプ対照を含ませて、任意の非特異的な結合事象を排除した。実験の結果を図2に示す。ペプチド1の存在は、M20 Mab細胞染色に対していかなる著しい影響も与えずに、mD-D6又はmD-A10 Mabによる細胞標識を阻害する。
種々のアイソフォームを、推定分子量(kDa)及びそれらの相当するアミノ酸配列とともに、Table 6(表6)に示す
競合実験に関して、抗体M20(1μg/mL)又はmD-A10(1μg/mL)若しくはmD-D6(1μg/mL)等のマウスモノクローナル抗体を、TBS-T溶液中で1/100の分子比(ペプチド分子100個につき抗体分子1個)でペプチド1又はペプチド2とともに、室温で2時間、プレインキュベートした。次に、抗体-ペプチド混合物を、HRPに連結された抗マウス二次抗体による顕色のため、2.5%乳汁を有するTBS-Tの溶液中で1時間、膜と接触させた。結果を図3に示す。ペプチド2の存在は、mD-D6 Mabによる全てのCK8アイソフォームの認識を減少させて、抗体M20による、及びmD-A10 MabによるCK8アイソフォームの認識を妨げない。逆に、ペプチド1の存在は、mD-A10及びmD-D6 Mabによる全てCK8アイソフォームの認識を強力に減少させるのに対して、それは、抗体M20によるCK8アイソフォームの認識を妨げない。
リアルタイム測定機器xCELLigence(ACEA Biosciences(商標))は、微小電極上での細胞固定に比例するインピーダンス値に基づく。細胞の浸潤能を評価するのに使用するシステムは、RTCA DPシステムである。このシステムは、16個のウェルを含有するCIMプレートを使用する。ウェルは、下部チャンバー及び上部チャンバーにより形成される。上部チャンバーは、微小電極を上回る8μM直径の孔を通じて、下部チャンバーと連絡する。Isreco-1浸潤能を評価する場合、下部チャンバーは、10%ウシ胎児血清(FBS)を補充した培養培地を含有していた。FBSを伴わない培養培地の存在下のIsreco-1細胞(20000個の細胞)は、抗CK8抗体(50μg/mLのM20、mD-A10又はmD-D6)を伴って、又は伴わずに、上部チャンバーに入れる。この上部チャンバーは、その底部でマトリゲル(matrigel(商標)、BD biosciences社)の層により予め覆われていた。浸潤プロセス中、細胞は、下部チャンバーと上部チャンバーとの間で確立されているFBS勾配により引き付けられ、マトリゲルを分解し、それによって、孔を通って遊走することが可能となり、下部チャンバーの微小電極に接触する。細胞接触により発生するこのインピーダンス測定は、70時間中に15分毎に測定され、RTCA DPシステムに連結されるコンピューターにより記録される。細胞の浸潤能の結果を図4に提示する。Isreco-1浸潤能の阻害のパーセントは、3回の別個の実験に相当する標準偏差とともに表される。Isreco-1細胞のFBSにより誘発される浸潤能に対する著しい阻害は、抗体M20又はmD-A10の存在下で観察され、mD-D6を用いた場合にはいかなる影響もなかった。
CellTiter-Glo(登録商標)Luminescent Cell Viabilityアッセイ(Promega社、Charbonnieres les Bains、フランス)を使用して、代謝的に活性な細胞の指標である存在するATPの定量化に基づいて、培養液中の生存細胞の数を決定した。検出は、ルシフェラーゼ反応を使用することに基づき、生存細胞由来のATPの量を測定する。膜完全性の損失の数分後に、細胞は、ATPを合成する能力を失い、内因性ATPアーゼは、任意の残存するATPを破壊し、したがって、ATPのレベルは、急激に下がる。細胞培養物(5.104個の細胞/mL)を、単独で、又は10μg/mlで試験した抗体(B-Z1、B-E4、M20、1E8、D-D6、D-A10)の存在下で72時間インキュベートした後、1/10で希釈した(図5)。アイソタイプ対照Mab(IgG1-B-Z1、IgG2a-B-E4)は、Diaclone社(Besancon、フランス)で購入した。CellTiter-Glo(登録商標)試薬を、試薬50μLの培養培地200μLに対する比で培養液中の細胞へ直接添加した。アッセイプレートを室温で10分間インキュベートして、標準的なマルチウェル蛍光光度計Mithras LB940(Berthold社、Thoiry、フランス)を使用して、生物発光シグナルを記録した。結果を図5に示す。Mab 1E8抗CK8のみが、Isreco-1細胞増殖を阻害する。
30mg/kgのMabにより処理したか、又は処理しないマウスのIsreco-1異種移植片腫瘍から組織を単離した。抗体M20、mD-A10又はmD-D6の、カスパーゼ3活性化を誘発する効力を評価した。染色のため、組織試料を10%緩衝ホルマリン中で固定して、パラフィン中に包埋させた。ホルマリン固定したパラフィン包埋組織の4μm厚の組織切片を、従来の手順に従って調製した。免疫組織化学は、DABmapキットを使用した自動免疫染色機(immunostainer)(Ventana Discovery XT、Roche社、Meylan、フランス)で、製造業者の指示書に従って実施した。緩衝液細胞コンディショニング(CC1)を用いた回復(retrieval)手順後に、切片を、ウサギモノクローナル抗切断カスパーゼ3抗体(1:200で希釈、クローン5A1E、Cell signaling社)とともにインキュベートした。染色は、発色性基質として3,3'-ジアミノベンジジンを有するDAB溶液で可視化させた。最終的に、切片をGillのヘマトキシリンで対比染色させた。1mm2当たりのカスパーゼ3活性化陽性細胞の数の定量化は、顕微鏡可視化後に、カメラに連結させたHistolabソフトウェアで行った。各腫瘍に関して、カスパーゼ3活性化陽性細胞の数の計数は、表面積2mm2上で実現させた。腫瘍の壊死領域及び線維性被膜は、分析から除外した。活性化カスパーゼ3の定量化の結果を図6Aに提示する。カスパーゼ3活性化陽性細胞の数は、1群当たり3匹に相当する標準偏差ともに表す。図6Bでは、免疫組織化学分析の例は、未処理の動物から、及びmD-A10 Mab、M20及びmD-D6 Mabで処理した動物から得られる厚い組織切片に関して示される。図6Aに示されるように、カスパーゼ3活性化陽性細胞の数は、未処理の動物と比較して、mD-A10 Mabで処理した動物由来の腫瘍において、著しく高い。M20で、又はmD-D6 Mabで処理した動物においては、著しい影響は観察されなかった。この結果により、mD-A10が、in vivoでIsreco-1細胞のカスパーゼ3活性化に関連したアポトーシスを誘発することが可能であるのに対して、M20又はmD-D6 Mabは、カスパーゼ切断を誘発しなかったことが明確に分かった。これらの結果により、M20及びmD-A10は、非常に特徴的なメカニズムにより、それらの抗腫瘍成長効果を示すことが実証される。mD-A10 Mabは、カスパーゼ3誘発アポトーシス誘発に関連したアゴニスト性Mabとして定義される一方で、M20 Mabは、この経路に非依存的である。
Isreco-1細胞(10×106)を、12匹のSCID CB17マウスに皮下注射した。15日後、腫瘍は、100〜150mm3の体積に達した。この時点で、マウスを3匹の4つの群へ分割した。各群に、腫瘍の反対部分で実施される腹腔内注射による特異的な処理を施したか、又は施さなかった。4週の間、1週間に一度(15日目、22日目、29日目及び36日目)、マウスに、抗体M20、mD-A10 Mab又はmD-D6を用いて30mg/kgの注射を施したか、又は施さなかった(図7)。各マウスに関して、実験全体にわたって、1週間に二度、腫瘍体積を測定した。各マウス群の腫瘍体積の平均値は、1群当たり3匹のマウスに相当する標準偏差とともにグラフ上に表す。図7に示すように、抗体M20又はmD-A10は、マウスにおいて皮下移植したIsreco-1結腸直腸がん細胞の成長を阻害したのに対して、mD-D6は、腫瘍発達を制御しなかった。
結腸直腸がん細胞Isreco-1から入手した腫瘍Isreco-1断片を、18匹のSCID CB17マウスに皮下移植した。15日後、腫瘍は、100〜150mm3の体積に達した。この時点で、マウスを3匹の6つの群へ分割した。各群に、腫瘍の反対部分で実施される腹腔内注射による特異的な処理を施したか、又は施さなかった。4週の間、1週間に一度(15日目、22日目、29日目及び36日目)、マウスに、1mg/kg、3mg/kg、10mg/kg、30mg/kg又は60mg/kgのmD-A10 Mabの注射を施したか、又は施さなかった。各マウスに関して、実験全体にわたって、1週間に二度、腫瘍体積を測定した。各マウス群の平均腫瘍体積は、1群当たり3匹のマウスに相当する標準偏差とともにグラフ上に表す。結果を図8に提示する。マウスにおいて皮下移植したIsreco-1結腸直腸がん細胞の成長の阻害は、1mg/kgの最低Mab濃度でさえ観察された。
HCT116細胞(10×106)を、12匹のSCID CB17マウスに皮下注射した。6日後、腫瘍は、100〜150mm3の体積に達した。この時点で、マウスを3匹の4つの群へ分割した。各群に、腫瘍の反対部分で実施される腹腔内注射による特異的な処理を施したか、又は施さなかった。3週の間、1週間に一度(6日目、13日目及び20日目)、マウスに、30mg/kgの抗体抗CK8(M20、mD-A10又はmD-D6)の注射を施したか、又は施さなかった。各マウスに関して、実験全体にわたって、1週間に二度、腫瘍体積を測定した。マウスの各群の平均体積は、1群当たり3匹のマウスに等しい標準偏差とともにグラフに示す。図7に示すように、mD-A10は、抗体M20と比較して、マウスにおいて皮下移植したIsreco-1結腸直腸がん細胞の成長を最高レベルで阻害したのに対して、mD-D6は、腫瘍発達を制御しなかった。
HCT116細胞(10×106)を、18匹のSCID CB17マウスに皮下注射した。10日後、腫瘍は、100〜150mm3の体積に達した。この時点で、マウスを3匹の6つの群へ分割した。各群に、腫瘍の反対部分に、腹腔内注射による特異的な処理を施したか、又は施さなかった。3週の間、1週間に一度(10日目、17日目及び24日目)、マウスに、1mg/kg、3mg/kg、10mg/kg、30mg/kg又は60mg/kgのmD-A10 Mabで処理したか、又は処理しなかった。各マウスに関して、実験全体にわたって、1週間に二度、腫瘍体積を測定した。マウスの各群の平均体積は、1群当たり3匹のマウスに等しい標準偏差とともにグラフに示す。結果を図10に示す。
従来の手順を使用して(例えば、マウス抗体の重鎖及び軽鎖をコードする遺伝子に特異的に結合することが可能であるオリゴヌクレオチドプローブを使用することにより)、抗体をコードするDNAを単離及び配列決定することができる。ハイブリドーマ細胞は、かかるDNAの好ましい供給源として役立つ。
ハイブリドーマ由来の可変領域に相当するcDNAを、2つのアプローチを使用して入手した。第1のアプローチは、N末端配列決定した後に生成した縮重N末端アミノ酸関連プライマー組のPCRにおける利用で構成された。第2のアプローチは、IMGT(登録商標)プライマーデータベース及びこれまでに記載された(Essono等、J Immunol Methods. 2003年; 203: 279:25〜66頁、Wang等、Mol Immunol. 1991; 28:1387〜97頁)特異的なプライマーにより生成した縮重プライマー組のPCRにおける利用で構成された。N末端可変領域の配列は、Edman分解により決定した。総RNA抽出は、供給業者Sigmaにより記載されるプロトコールに従って、Tri試薬キットを使用して実施した。増幅VL及びVH断片を、ジデオキシ終結方法により配列分析用にTOPO-TAクローニングベクター(Invitrogen社)にクローニングした(Sanger等、Nature. 1977年; 265:687〜95年)。続いて、抗体変異構築物を、PCRにより増幅させて、発現ベクターにクローニングした。
- mD-A10 Mabの可変マウス軽鎖のヌクレオチド配列(配列番号30)及びそれ由来のアミノ酸配列(配列番号31)。
- mD-A10 Mabの可変マウス重鎖のヌクレオチド配列(配列番号32)及びそれ由来のアミノ酸配列(配列番号33)。
- chD-A10 Mabの定常ヒト重鎖のヌクレオチド配列(配列番号48)及びそれ由来のアミノ酸配列(配列番号47)。
- chD-A10 Mabの定常ヒト軽鎖のヌクレオチド配列(配列番号50)及びそれ由来のアミノ酸配列(配列番号49)。
ヒト化H及びL鎖は、PCR法によるCDRグラフトを使用して生成させた。マウスモノクローナル抗体のCDRがヒト抗体上へグラフトされるヒト化抗体を生成するために、マウスモノクローナル抗体の可変領域とヒト抗体の可変領域との間に高い相同性を得ることが好ましい。したがって、マウス抗ヒトCK8モノクローナル抗体のH鎖及びL鎖のV領域は、ソフトウェアIMGT/DomainGapAlignを使用して、全ての公知のヒト抗体のV領域と比較する。マウス抗体が、従来技術によりヒト化される場合、CDRを支持するマウス抗体のV領域FRの幾つかのアミノ酸配列は、望ましい場合には、ヒトV領域のFR上へグラフトされてもよい。
C末端システインのSH基でウシ血清アルブミン(BSA)と連結されたか、又は連結されていないCK8ペプチドを、96ウェルプレートのウェルの底部で、室温で一晩固定化した(それぞれ、0.5μ/ ml又は1μg/ml)。未結合のペプチドは、Tween(Sigma社、St Quentin Fallavier、フランス)を補充した精製水で洗い流した。次に、5%BSAを含有する生理リン酸緩衝液(PBS)で、室温で(RT)2時間、ウェルを飽和させる。続いて、Tweenを補充した精製水で、プレートを洗い流す。1%ウシ血清アルブミン(Sigma社、St Quentin Fallavier、フランス)を補充したPBS(Invitrogen社、Villebon sur Yvette、フランス)中で、RTで2時間のインキュベーション後に、コーティングされたペプチドへのMab結合を、種々のMab濃度で試験する。未結合のMabは、Tweenを補充した精製水で洗い流す。続いて、結合Mabは、ホースラディッシュペルオキシダーゼ(HPR)に連結されたヤギ抗マウスIgG二次抗体(Biorad、フランス)により顕色させる。吸光度を450nmで読み取り、630nmで補正する。種々の実験に関する特定の要件を以下で詳述する。
C末端システインの付加により修飾されるヒトCK8のペプチド1、2、3又は4を、C末端システインのSH基でBSAに連結させて、96ウェルプレートの底部で固定化した。483個のアミノ酸(aa)を含む完全ヒトCK8配列(ref Uni-ProtKB P05787)を図11に示す。ペプチド1、2、3及び4の配列の位置は、ヒトCK8配列上で灰色及び黒色の矢印で示す:

ヒトCK8の非結合型ペプチド1、5〜28を、96ウェルプレートの底部で固定化した(2μg/ml)。種々のMab濃度(10μg/mLから、続いて1/10で希釈)を、室温で2時間、単独で、又は試験する各ペプチドとともにインキュベートした。光学密度値(OD)を450nmで読み取り、630nmで補正した。結果を図13A(mD-A10に関連)又は図13B(M20に関連)に示す。比較分析を以下のTable 8(表8)で概要する。これらの結果は、試験するペプチドパネルに対するM20抗体との比較での、種々のmD-A10反応性プロファイルを明らかに実証した。
種々のCK8ペプチドを用いたmD-A10 Mabの競合アッセイを設定する場合、ペプチド-抗体混合物を、30分間インキュベートした後、ウェルの底部に固定化した、検出しようとするペプチドの存在下に置いた。mD-A10 Mabは、BSA結合型ペプチド1(配列番号1)の認識に関して、又はBSA結合型ペプチド17(配列番号17)の認識に関しては、ペプチド1、5〜28(配列番号1、配列番号5〜配列番号28)のいずれかの存在下に置いた。

- 配列番号29のアミノ酸配列を有するペプチドに特異的に結合する抗体、又はその抗体断片であって、前記抗体が、
- 下記の3つのCDR、それぞれCDR-H1、CDR-H2及びCDR-H3を含む重鎖であって、
CDR-H1が、配列番号37の配列、又は配列番号37の配列との最適なアラインメント後に少なくとも80%、好ましくは85%、90%、95%及び98%同一性を有する配列を含み、
CDR-H2が、配列番号38の配列、又は配列番号38の配列との最適なアラインメント後に少なくとも80%、好ましくは85%、90%、95%及び98%同一性を有する配列を含み、
CDR-H3が、配列番号39の配列、又は配列番号39の配列との最適なアラインメント後に少なくとも80%、好ましくは85%、90%、95%及び98%同一性を有する配列を含む
重鎖、並びに
- 下記の3つのCDR、それぞれCDR-L1、CDR-L2及びCDR-L3を含む軽鎖であって、

- 配列番号30、又は配列番号30の配列との最適なアラインメント後に少なくとも80%、好ましくは85%、90%、95%及び98%同一性を有する配列のマウス軽鎖可変領域のヌクレオチド配列、及び配列番号32、又は配列番号32の配列との最適なアラインメント後に少なくとも80%、好ましくは85%、90%、95%及び98%同一性を有する配列のマウス重鎖可変領域のヌクレオチド配列を有するポリヌクレオチドによりコードされる可変領域を含む、請求項1に記載の抗体。
- 配列番号29のアミノ酸配列を有するペプチドに特異的に結合するヒト化抗体、又はその抗体断片であって、前記抗体が、

- 浸潤がんの治療における使用のための、及び/又は結腸直腸がん、好ましくは浸潤結腸直腸がんの治療における使用のための、請求項8に記載の抗体又は抗体断片。
- 8〜16個のアミノ酸を有するポリペプチドからなり、かつ配列番号29の配列を含むヒトCK8抗原ペプチド、特に配列番号29の配列からなるヒトCK8抗原ペプチド。
- 治療薬としての使用のための、請求項10に記載のヒトCK8抗原ペプチド。
- 請求項1から6に記載の抗体又は抗体断片、及び最終的に、適切な薬学的担体を含む薬学的組成物又は診断用のキット。
- 前記抗体又は抗体断片が、Fv、Fab、F(ab')2、scFV、抗体、好ましくはモノクローナル抗体である、請求項12に記載の薬学的組成物又はキット。
- がん、自己免疫疾患、炎症性状態、ウイルス感染若しくはウイルス疾患の治療若しくは防止における、及び/又は腫瘍細胞のアポトーシスを誘発するための医薬としての使用のための、請求項12又は13に記載の薬学的組成物。
- 細胞が、その表面上で、CK8タンパク質、特に配列番号29に従う配列のペプチドを発現する腫瘍の治療における使用のための、請求項14に記載の薬学的組成物。
- 別の抗体及び/又は別の抗がん薬を更に含む、請求項12から15のいずれか一項に記載の薬学的組成物又はキット。
- がんを検出するために使用するための、請求項12に記載のキット。
