Abstract
Description
i)対象の腫瘍細胞にアデノウイルスベクターを感染させる工程、
ii)対象の樹状細胞にアデノウイルスベクターを感染させる工程、
iii)工程i)の腫瘍細胞および工程ii)の樹状細胞からMHC−I分子を単離し、両グループからMHC−I関連ポリペプチドを同定する工程、
iv)感染しなかった腫瘍細胞からMHC−I分子を単離し、MHC−I関連ポリペプチドを同定する工程、
v)工程iii)およびiv)の感染した腫瘍および感染しなかった腫瘍、ならびに工程iii)の樹状細胞によって提示されたポリペプチドを同定する工程、
を含む方法に関する。
腫瘍免疫学および免疫ペプチドーム
樹状細胞(DC)は、骨髄由来の専門的抗原提示細胞である。DCは、腫瘍抗原エピトープをCD8+およびCD4+T細胞に提示するために最適な抗原提示細胞である3。外因性抗原は、CD8+T細胞への「交差提示(クロスプレゼンテーション)」のためにMHCクラスIに積載(ロード)されうる4。交差提示は、その結果がDCの活性化状態によって決定される現象である5。癌細胞では、腫瘍抗原交差提示をもたらすDC成熟の程度は、不利な腫瘍微小環境および局所リンパ節での腫瘍由来免疫抑制のために、通常、非常に低い。これらの障害は、腫瘍を破壊するウイルスがいずれも、DC活性化を駆動し、腫瘍免疫抑制を妨害して隠れた免疫原性抗原を露出させるのに必要な「危険信号」を提供するため、腫瘍崩壊性ウイルス治療によって克服することができる6−8。
ペプチドで被覆されたアデノウイルスは、任意のタイプおよび種のアデノウイルス科のもの(例えば、ヒトアデノウイルスに限定されない)であってよい。本発明の一実施形態では、アデノウイルスは、腫瘍に対する抗ウイルス免疫応答を転換しながら複製し癌細胞を死滅させることができる(図1)。患者由来の腫瘍特異的免疫活性化ペプチドで被覆された本発明の癌破壊性ウイルスは、抗ウイルス免疫から抗腫瘍免疫への転換を増強する。
本発明によれば、アデノウイルスのカプシドは、対象においてペプチド特異的免疫応答を刺激することができる合成ポリペプチドまたはペプチドでコーティングされる。アデノウイルスベクターをコーティングするために使用されるポリペプチドは、前記アデノウイルスベクターによって遺伝的にコードされていない。本明細書において、用語「ポリペプチド」および「ペプチド」は、任意の長さのアミノ酸のポリマーを指すために交換可能に使用される。
本発明は、障害を治療するための治療方法および使用だけでなく、前記方法および治療用途で使用するための医薬組成物も提供する。そのような医薬組成物は、被覆アデノウイルスを単独で、または治療上有効な薬剤(一または複数)および/または薬学的に許容されるビヒクル(一または複数)などの他の薬剤と組み合わせて含む。

本発明は、対象から少なくとも腫瘍特異的およびMHC−I特異的なポリペプチドを同定するための方法を明らかにする。この方法は、具体的にはインビトロで腫瘍溶解物に曝露されたDCおよび腫瘍のMHC−I免疫ペプチドームに関する、定性的および定量的研究を利用する。方法論は、図5に簡潔に要約すると、インビトロで腫瘍崩壊物でパルスされたDC(ウイルス感染腫瘍細胞)および腫瘍細胞の両方からのMHC−I分子の単離、および質量分析に基づく技術によるMHC関連ポリペプチドの配列決定を含む。免疫学的に関連するペプチドは、腫瘍溶解物でパルスされた樹状細胞および腫瘍の両方によって提示される。例えば、OVA発現モデルマウスの使用は系の検証を容易にすることができ、実際によく知られた免疫原性OVA由来ペプチド(例えばSIINFEKL)はマウス実験からもたらされ、ポジティブコントロールとして役立ちうる。
すべての腫瘍崩壊性アデノウイルス(OAd)を生成し、先に記載した標準的なプロトコルを用いて増殖させた(8)。簡潔に述べると、10のT175フラスコに70〜80%コンフルエントA549細胞を30の感染多重度(MOI)で感染させることによりウイルスを増幅した。感染の3日後、細胞を回収し、4回の凍結(−80℃)および解凍(37℃)サイクルにより溶解した。次いで、CsCl勾配の2回の超遠心分離(22,000および27,000rpm)により、細胞破片および不純物からアデノウイルス粒子を分離した。回収したバンドを、連続攪拌しながらA195緩衝液に対して4℃で一晩透析することにより精製した。具体的には、10,000kDaの分子量カットオフを有する透析カセット(Pierce、Life Technologies)を使用した。精製されたウイルスをカセットから回収し、等分し、−80℃で保存した。
方法1a:
C57BL/6マウスからマウスCD11c+分類骨髄樹状細胞を採取し、1週間培養した23。次いで、細胞を以下の溶液に曝した。
A)コントロールとしてのPBS、
B)B16−OVA細胞からの腫瘍崩壊物(腫瘍崩壊物は、完全に溶解するまで腫瘍崩壊性アデノウイルスAd5D24に感染させたB16−OVA細胞に由来する)
C)細胞の凍結および融解により得られたB16−OVA細胞溶解物。
様々な時点で、ペプチドを負荷したMHC−Iを、軽度の酸溶出を用いて生DCから単離した25。分析の時点で、ペプチドを水溶液に溶解し、LTQ−Orbitrap Elite質量分析計(Thermo Fisher Scientific)でナノLC−MS/MSにより分析した。データベース検索は、51536配列および24497860残基を含む国際タンパク質インデックスマウスデータベースバージョン3.23(http://www.ebi.ac.uk/IPI/IPIhelp.html)に対して実施した。関連ペプチドは、(DC自己ペプチドを排除するために)パルスを与えていないDCを除いた溶解物でパルスしたDC群と、(ウイルス特異的ペプチドを排除するために)感染していないB16−OVAを除いたウイルス感染B16−OVA群の両方に共通して存在するペプチドによる群とした。
本発明者らはまず、方法1aの免疫ペプチドームの複雑さをインシリコで減少させた。 MHC−Iクラスペプチドの予測(http://www.syfpeithi.de/home.htm)。タンパク質の機能的注釈(http://david.abcc.ncifcrf.gov)および(http://www.ingenuity.com)を用いた。
OVA由来のペプチドは非常によく知られているので、概念実証として、本発明者らは最初にOVA特異的に被覆ウイルスを生成した(図6)。より具体的には、本発明者らは、SIINFEKL被覆アデノウイルス(SIINFEKL(配列番号1)は、最も免疫原性の高いOVA由来ペプチドである);SIINFEDL被覆ウイルス(SIINFEDL(配列番号7)はSIINFEKLペプチドのアンタゴニストである);FILKSINE被覆ウイルス(FILKSINE(配列番号3)はSIINFEKLのスクランブルペプチドである)を生成した。
ペプチド被覆腫瘍崩壊性アデノウイルスを生成するために、異なる戦略が考慮された(図7)。
I.静電相互作用。陰性ウイルスカプシドと複合体を形成した陽性荷電ペプチド26。
II.共有結合。カプシドのタンパク質のシステインとのジスルフィド結合27,28。
III.共有結合。アミド結合。カプシドのリジンのアミン基とのスクシンイミジルエステル反応28。
連結の方法は対応する参考文献に記載されている。
PeptiCRAd複合体形成
この研究に記載されているすべてのPeptiCRAd複合体は、以下のプロトコールに従って、腫瘍崩壊性ウイルス(「腫瘍崩壊性アデノウイルス調製物」の項で記載)とポリKエピトープとを1:500の比で混合することによって調製した(図8Aおよび12参照)。i)用いたウイルス調製物1マイクロリットルにつき、存在するタンパク質のマイクログラムの対応する数を計算し、ii)次に、ウイルスタンパク質1マイクログラムにつき500μgのペプチドを添加し、iii)ボルテックス後、混合物を室温(RT)で15分間インキュベートし、iv)溶液をボルテックスし、アッセイまたは動物注射に使用した。 新鮮な試薬を用いて各実験の前に新しいPeptiCRAdを調製した。インキュベーション前に必要とするウイルスおよびペプチドをすべて、pH7.4に調整した滅菌Milli−Q水で希釈した。次いで、PeptiCRAdをアッセイに必要とされる緩衝液で希釈した。
方法2aのこのペプチド被覆ウイルスの感染性を、異なる細胞株(ヒトおよびマウス)においてルシフェラーゼアッセイおよびqPCRによってインビトロで評価した30。感染性を評価するために、CARの発現レベルが異なる異種腫瘍細胞株のパネルに、異なる濃度のルシフェラーゼ発現被覆ウイルス(Ad5D24−Luc)(1、10、100、1000VP/細胞)を感染させ、被覆されていないウイルスを常にコントロールとして用いた。異なる時点でルシフェラーゼ発現を定量した。同時に、全DNAを回収し、qPCRによりウイルスDNA複製を定量した。インビトロでの腫瘍崩壊活性をTCID50およびMTSアッセイにより試験した31。
ICCによる感染性アッセイ
腫瘍細胞を、1ウェルあたり2.0×105個の細胞で3または5回反復して24ウェルプレートに播種した。翌日、細胞を100μlのウイルス希釈液で感染させた。次いで、プレートを37℃で1,000rcfで90分間遠心分離し、続いて48時間インキュベートした。インキュベーション期間の後、培地を除去し、250μlの氷冷メタノールで15分間インキュベートすることによって細胞を固定した。メタノールを除去した後、細胞を1%ウシ血清アルブミン(BSA)を補充した300μlのPBSで3回洗浄した。次いで、細胞を1:2,000に希釈した250μlのマウスモノクローナル抗ヘキソン抗体(Novus Biologicals、Littleton、CO、USA)を用いて暗所で室温で1時間かけて染色した。次いで、細胞を洗浄し、PBS/1%BSAで1:500に希釈した250μlのビオチン−ストレプトアビジン結合ヤギ抗マウス抗体で、暗所で室温で1時間かけて染色した。その後、細胞を1:200に希釈した250μlのextravidin−ペルオキシダーゼ(Sigma-Aldrich、St. Louis、MO、USA)と共にRTで30分間インキュベートした。細胞を広範囲に洗浄し、DAB染色溶液(Sigma-Aldrich、St.Louis、MO、USA)を製造者の指示に従って調製した。次に、合計250μlのDAB染色溶液を各ウェルにアプライし、細胞を暗点の出現について顕微鏡下でモニターした。最適なシグナル対ノイズ比に達したら、PBS/1%BSA(1ウェル当たり500μl)の添加により反応を停止させた。各複製物(すなわち、ウェル)について、AMG EVOS XL顕微鏡(AMGグループ、Life Technologies)を用いて、非重複フィールドの5つの画像を取得した。以下の式を用いて感染力価を決定した:

感染性の比較のために、データは、各フィールドにおける平均スポット数として表した。
負に荷電したアデノウイルスカプシドを腫瘍特異的ペプチドで静電的に被覆した。この複合体は、ペプチドの量に比例するZ電位の変動を有していた。このZ電位の変化は、正に荷電したペプチドが電荷の反転を決定するウイルスカプシドに結合していることを示した(図8Aの点線)。カプシドの負の電荷すべてが飽和した後、Z電位はプラトーになるようであった(図12の円を付した線)。インビトロおよびインビボの有効性に進むために、500nMを超えるポリペプチド濃度で均一な単分散複合体を形成することができる。
生存率アッセイ
腫瘍細胞を、5%FBSを含む成長培地に、96ウェルプレート上に1.0×104細胞/ウェルで播種した。翌日、培地を除去し、2%FBSを含む成長培地で希釈した50μlのウイルスを用いて、細胞を37℃で2時間かけて感染させた。その後、5%FBSを含む100μlの成長培地を添加し、細胞を再び37℃でインキュベートした。成長培地は1日おきに交換した。最も感染力の強い条件(100vp /細胞)が広範囲の細胞変性効果(>90%)を示した場合、製造業者のプロトコール(CellTiter 96 AQueous One Solution細胞増殖アッセイ;Promega、Nacka、Sweden)に従ってMTSアッセイによって細胞生存率を測定した。分光光度データは、Varioskan Flash Multimode読み取り機(Thermo Scientific、Carlsbad、CA、USA)を用いて取得した。
サンプルサイズは以下の式を使用して決定した。

ここで、Cはα値とβ値に基づく定数であり、sは推定された変数であり、dは観測される効果である(34)。すべての動物実験について、少なくとも80%のパワー(1−β)と0.05の有意性(α)が考慮された。データ収集を停止するための条件は、i)1または複数の群におおける60%を超えるマウスの死、およびii)腫瘍の全クリアランスとした。実験終了前に死亡したマウスをすべて増殖曲線から除外して、分析の統計的完全性を維持した。
ヒト肺癌細胞株A549、ヒト結腸直腸腺癌細胞株CACO−2、ヒト悪性メラノーマ細胞株SK−MEL−2、ヒトメラノーマ細胞株HS294Tおよびマウスメラノーマ細胞株B16−F10は、アメリカンタイプカルチャーコレクション(ATCC;Manassas、VA、USA)より購入した。ニワトリOVAを発現するマウスメラノーマ細胞株である細胞株B16−OVA(35)は、Richard Vile教授(Mayo Clinic、Rochester、MN、USA)により快く提供された。
ペプチドの正味電荷は、ペプチド特性計算機バージョン3.1オンラインツール(http://www.biosyn.com/PeptidePropertyCalculator/PeptidePropertyCalculator.aspx)によって算出した。
方法3a:
本発明者らは、被覆ウイルス対被覆されていない一般的な腫瘍崩壊性ウイルスの有効性、免疫原性、毒性、生体内分布をインビボで試験した。有効性および免疫原性を、B16−OVA腫瘍を持つC57BL/6マウスにおいて試験した。SIINFEKL被覆ウイルスは、他の被覆ウイルス(アンタゴニスト、スクランブルおよび非被覆)と比較して、より顕著な腫瘍制御(有効性)に変換される、より強固な抗OVA応答を示した。同時に、放射能標識細胞(DCおよびT細胞)の適応的移入を介して、これらの細胞の腫瘍微小環境への輸送も評価した。最後に、改変されたアデノウイルスベクターの毒性および生体内分布も研究した。
i)OVA−ペプチド(SIINFEKL(配列番号1))、ii)B16ペプチドTRP2(SVYDFFVWL(配列番号5))、iii)hgp100ペプチド(KVPRNQDWL(配列番号6))またはiv)方法1で同定された新規ペプチドで被覆されたウイルスの抗腫瘍活性および免疫原性を比較した。
図7ストラテジーIに記載するように、本発明者らは、OVA特異的PeptiCRAd(SIINFEKL被覆腫瘍崩壊アデノウイルス)を生成した。簡潔には、合成SIINFEKLペプチドを合成し、ポリリジンリンカー(ポリK−SIINFEKL)に結合させて、ペプチドに正味の正電荷を与え、注射の30分前に正味の負電荷を有するネイキッドなウイルスと複合体を形成させた。次いで、この複合体を皮下B16−OVA腫瘍を有するマウスに腫瘍内投与した。腫瘍増殖をモニターし、実験の最後にマウスを安楽死させ、腫瘍を収集し、OVA特異的T細胞をフローサイトメトリーによって定量した(図10)。
方法4:
第二世代のPeptiCRAdは、より強力で多価の免疫応答を引き出すために、腫瘍崩壊性ウイルスを1つより多くのペプチドでコーティングすることによって作製した。これらの新しいウイルスは方法2のように特徴付けられ、有効性は方法3に記載するように評価した。続いて、本発明者らは、サイトカイン武装腫瘍崩壊性アデノウイルスをいくつかのタイプのポリペプチドでコーティングした。ポリペプチドは、同じ抗原の異なるMHC−I特異的ペプチド、または異なる抗原由来のMHC−Iペプチド、またはMHC−IおよびMHC−II拘束ペプチドの組み合わせのいずれであってよい。
ゼータ電位および動的光散乱(DLS)分析
被覆された腫瘍崩壊性ウイルス試料は、タイトル「PeptiCRAd複合体形成」の項に記載したように調製した。次いで、各サンプルをボルテックスし、pH7.4に調整した滅菌Milli−Q水で最終容量700μlに希釈した後、サンプルをポリスチレン使い捨てキュベットに移して複合体のサイズを決定した。 次いで、試料をキュベットから回収し、ゼータ電位測定のためにDTS1070使い捨て毛細管(Malvern、Worcestershire、UK)に移した。 すべての測定は、Zetasizer Nano ZS(Malvern)を用いて25℃で行った。
ポリK−SIINFEKLまたはSIINFEKLとOAdとの相互作用をSPRを用いて評価した。測定は、マルチパラメトリックSPR Navi(商標)220A装置(Bionavis Ltd、Tampere、Finland)を用いて行った。この装置は、統合された流体システムを備えた温度制御二重流体チャネルと、緩衝液および試料の取り扱いのためのオートサンプラーとを含む。泳動緩衝液として、pHを7.4に調整したMilli−Q水を使用した。さらに、30μl/分の一定流量を実験を通して使用し、温度を+20℃に設定した。表面プラズモン励起には波長670nmのレーザー光を用いた。
新鮮な脾臓をナイーブC57BL/6マウスから採取し、70μm細胞ストレーナー(Fisher Scientific、Waltham、MA、USA)に通した。5mlのACK溶解緩衝液(Life Technologies)と共に試料を室温で5分間インキュベートすることにより、赤血球を溶解させた。その後、脾細胞を洗浄し、アッセイ用に調製した(試験した各条件について10%RPMI−1640培地800μl中2×106細胞)。SIINFEKL、polyK−SIINFEKL、SIINFEKL−polyKまたはSIINFEKL−AHX−polyKペプチド希釈物(0.19μg/μl)の合計200μlを脾細胞に添加した。OVA−PeptiCRAdを試験するために、100vp/細胞の感染条件を使用した(200μlの10%RPMI−1640中37.5μgのpolyK−SIINFEKLと混合した合計7.9×109vp)。PeptiCRAd複合体を方法2の記載に従って調製した。次いで、脾細胞を37℃で2時間インキュベートした。その後、細胞を十分に洗浄し、SIINFEKLに結合したAPC抗マウスH−2KbまたはAPCマウスIgG1(κアイソタイプCtrl)(BioLegend、San Diego、CA、USA)のいずれかで染色した。氷上で30分間インキュベートした後、サンプルを洗浄し、フローサイトメトリーで分析した。
処置したマウスの腫瘍、脾臓およびリンパ節を収集し、70μm細胞ストレーナーに通し、10%RPMI−1640培地中で一晩培養した。必要に応じて、サンプルをRPMI−1640(10%FBSおよび10%DMSOを含む)中で凍結し、−80℃で保存した。単一細胞懸濁液を蛍光色素結合モノクローナル抗体で染色し、BD LSR II(BD Biosciences)またはGallios(Beckman Coulter)フローサイトメーターおよびFlowJoソフトウェア(Tree Star、Ashland、OR、USA)を用いて分析した。滅菌PBSを染色緩衝液として使用した。エピトープ特異的T細胞をMHCクラスIペンタマー(ProImmune、Oxford、UK)を用いて研究した。使用した他の抗体には以下のものを含めた:ネズミおよびヒトFcブロックCD16/32(BD Pharmingen)、FITC抗マウスCD8およびFITC抗ヒトCD8(ProImmune)、PE/Cy7抗マウスCD3ε、PE/Cy7抗マウスCD19、FITC抗マウスCD11c、PerCp/Cy5.5抗マウスCD86、SIINFEKLに結合したAPC抗マウスH−2Kb、およびAPCマウスIgG1(κアイソタイプCtrl)(BioLegend)。すべての染色手順は、製造業者の推奨に従って実施した。
統計的有意性は、GraphPad Prism 6(GraphPad Software、Inc.、La Jolla、CA、USA)を用いて測定した。各実験からのデータを分析するために使用される統計的方法の詳細な説明は各図面の簡単な説明に記す。
動物実験は、フィンランドおよびヨーロッパの法律および法案の下で行われた。動物許可証(ESAVI/5924/04.10.03/2012)は、フィンランド当局(ヘルシンキ大学実験動物委員会および南フィンランド州政府)によって改訂され、承認された。完全免疫応答性C57BL/6マウスをScanbur(Karlslunde、Denmark)から入手し、免疫不全トリプルノックアウトNOD.Cg−Prkdcscid−IL2rgtm1Wjl/SzJマウスをJackson Laboratories(Bar Harbor、ME、USA)から入手した。すべてのマウスを4〜6週齢で購入し、試験前に2週間隔離した。マウスは隔離され調節された気流でケージに保持され、研究期間全体にわたって無制限に食べ物を利用可能とした。マウスの健康状態は頻繁に監視され、動物は痛みまたは苦痛の最初の兆候で屠殺した。すべての手順は、無菌条件下でのバイオセーフティーレベル2のキャビネット内で行った。
アデノウイルスカプシドの負電荷は、正に荷電した免疫原性ペプチドを複合させて、PeptiCRAdを形成するために使用することができる
アデノウイルスカプシドは非常に高い正味の負電荷を有する(36)。ゆえに、本発明者らは、正に荷電したMHC−I拘束ペプチドが、免疫学的に関連するペプチド(すなわち、腫瘍特異的MHC−I拘束ペプチド)でウイルスを覆う静電相互作用によってカプシドに結合すると仮定した。本発明者らの仮説を検証するために、B16−OVA腫瘍モデルを用いた(37)。この細胞株はニワトリ卵白アルブミン(OVA)を発現し、MHC−I上にモデルエピトープとして用いられるOVA由来ペプチドSIINFEKLを提示する。
効果的な細胞傷害性Tリンパ球媒介性免疫応答を誘導するためには、ペプチドはAPC上のMHC−Iを介してナイーブCD8+Tリンパ球に交差提示されなければならない。したがって、本発明者らは、ポリK鎖の存在および位置が交差提示の効率に影響を及ぼすかどうかを調べた。この目的のために、本発明者らは、天然SIINFEKLまたは2つの異なるリジン伸長バージョン:ポリK−SIINFEKL(N末端伸長)およびSIINFEKL−polyK(C末端伸長)のいずれかを用いて、エクスビボで培養した脾細胞(C57BL/6マウス由来)をパルスした。ネガティブコントロールとして、本発明者らは、プロテオソームのタンパク質分解活性を阻害することができるリジンのよく知られた類縁体である、アミノカプロン(AHX)残基を含む伸長SIINFEKLを含めた。次に、SIINFEKLを搭載したMHC−Iを特異的に認識する抗体を用いてSIINFEKLの交差提示を評価した(38)。
OAdは腫瘍細胞に選択的に感染し、OAd複製サイクルを介してそれらを溶解することができる。したがって、本発明者らは、改変されたペプチドでウイルスをコーティングすることがそれらの生物学的特性に影響を与えるかどうかを調べた。本発明者らは、低レベルのコクサッキーおよびアデノウイルスレセプター(CAR)を発現するヒト結腸直腸腺癌細胞株(CACO−2)および高いレベルのCARを発現する2つのヒトメラノーマ細胞株(SK−MEL−2およびA2058)を選択して試験した。OVA−PeptiCRAdと非改変ウイルスAd5D24とを比較するインビトロ生存率アッセイを最初に実施し(図14A)、結果は腫瘍崩壊活性に関して有意差を示さなかった。予想通り、最も感染性の高い条件(100vp/細胞)は、すべての細胞株において最も低い生存率と相関していた。さらに、本発明者らは、ペプチドpolyK−SIINFEKLが細胞に対して毒性作用を及ぼさないことを示した。
PeptiCRAdの抗腫瘍効果およびそれが促進する抗腫瘍免疫を徹底的に研究するために、初めにニワトリOVA(B16−OVA)を過剰発現するメラノーマのネズミ科モデルを使用した(35)。具体的には、B16−OVAをマウスの側腹部に移植し、その後、定着した腫瘍を処置した。実験は、E1A(Ad5D24)にD24欠損を有するOAd(37)を用いて行い、CpGリッチアデノウイルス(Ad5D24−CpG)(39)を繰り返し用いてさらに免疫を増強した(図15)。試験群にはOVA−PeptiCRAd、非複合Ad5D24−CpGおよびSIINFEKL(Ad5D24−CpG+SIINFEKL)、OAd(Ad5D24−CpG)またはペプチド(SIINFEKL)単独または生理食塩水(mock)で処置したマウスを含めた。
腫瘍崩壊性ワクチンを使用する主な利点の1つは、誘発された免疫応答が、原発腫瘍だけでなく播種性転移を標的とすることを容易にすることである。この理由のために、本発明者らは、メラノーマのネズミ科モデルにおける、未処置対側腫瘍に対するPeptiCRAdの抗腫瘍効果を調べた。同様の一連の実験において、本発明者らはまた、単一のものではなく、2つの腫瘍抗原(多価PeptiCRAdを介して)を標的とすることが全体の有効性を増加させるかどうかを検討した。したがって、本発明者らは、腫瘍崩壊性ウイルスAd5D24−CpGをコートするために2つの腫瘍特異的MHC−I拘束エピトープ:SVYDFFVWL(TRP−2180−188;ネズミ科MHC−I分子H−2Kbに限定)およびKVPRNQDWL(ヒトgp10025−33またはhgp100;ネズミMHC−I分子H−2Db(40)に限定)を選択した。これらの実験では、両方の腫瘍抗原を発現する非常に侵攻性の高いメラノーマB16−F10を用いた(41)。先のSIINFEKLについてのように、ペプチドをN末端でポリK鎖で修飾してウイルスカプシドへの吸着を促進した。
最後に、臨床現場への転換の実行可能性に関する情報を提供することができるモデルにおけるPeptiCRAdの有効性評価を試みた。したがって、本発明者らはより高性能のヒト化マウスモデルを選択した。この目的のために、まずトリプルノックアウトマウス(NOD.Cg−Prkdcscid−IL2rgtm1Wjl/SzJ、またはNSG)にヒトメラノーマ細胞株SK−MEL−2を移植した。腫瘍が触知可能な大きさに達したとき、健康なドナー由来の部分的に適合したヒト末梢血単核球(PBMC)を同じマウスに移植した。1日後、マウスをPeptiCRAd、被覆されていないウイルスまたは生理食塩水で処置した。この実験では、メラノーマ関連抗原A1(MAGE−A196−104;SLFRAVITK)由来のペプチドを選択し、それを改変してウイルスカプシド(polyK−SLFRAVITK)との相互作用を可能にした。この実験では、本発明者らはヒトの免疫系を十分に研究していたので、ヒトGM−CSFを発現するOAdを選択した。このOAdは本発明者らが以前に癌患者を含む免疫応答系で活性を増強することを示したものである(8)。
任意のMHC−I特異的ポリペプチドが、DCおよび対象の感染した疾患細胞に代表されるMHC−I拘束ポリペプチドを比較することによって同定される。両方の細胞群によって提示される1つ又は複数のポリペプチドは、アデノウイルスベクターをコーティングするために選択される。
コーティングされたベクターは、患者の疾患を治療するために使用される。

Claims (26)
- 必要とする対象においてペプチド特異的な免疫応答を刺激する方法であって、ウイルスカプシド上に付着したポリペプチドを含むアデノウイルスベクターの対象への投与を含み、当該ポリペプチドが前記アデノウイルスベクターによって遺伝的にコードされていない方法。
- 対象においてペプチド特異的な免疫応答を刺激する際に使用するための、ウイルスカプシド上に付着したポリペプチドを含むアデノウイルスベクターであって、当該ポリペプチドが前記アデノウイルスベクターによって遺伝的にコードされていないアデノウイルスベクター。
- 前記ペプチド特異的免疫応答が、抗腫瘍、抗癌、抗感染および抗ウイルスの免疫応答からなる群から選択される、請求項1に記載の方法または請求項2に記載のアデノウイルスベクター。
- 必要とする対象における癌の治療方法であって、対象におけるペプチド特異的免疫応答を刺激することが可能であり、ウイルスカプシド上に付着しているポリペプチドを含むアデノウイルスベクターの、対象への投与を含み、当該ポリペプチドが前記アデノウイルスベクターによって遺伝的にコードされていない方法。
- 対象における癌の治療の際に使用するための、対象におけるペプチド特異的免疫応答を刺激することが可能であり、ウイルスカプシド上に付着しているポリペプチドを含むアデノウイルスベクターであって、当該ポリペプチドが前記アデノウイルスベクターによって遺伝的にコードされていないアデノウイルスベクター。

- 前記対象がヒトまたは動物である、請求項1から6のいずれか一項に記載の方法またはアデノウイルスベクター。
- 前記アデノウイルスベクターの投与が、腫瘍内、動脈内、静脈内、胸膜内、膀胱内、腔内または腹腔内注射、または経口投与によって行われる、請求項1から7のいずれか一項に記載の方法またはアデノウイルスベクター。
- ウイルスカプシドがポリペプチドと結合しており、ポリペプチドと結合したアデノウイルスベクターが対象におけるペプチド特異的免疫応答を刺激することができる、アデノウイルスベクター。
- 前記ポリペプチドがウイルスカプシド上に共有結合または非共有結合的に接着している、請求項1から9のいずれか一項に記載の方法またはアデノウイルスベクター。
- アデノウイルスのカプシドを改変する方法であって、ポリリジン改変ポリペプチドを、共有結合的ないしは非共有結合的にアデノウイルスカプシドに連結することを含み、当該改変されたアデノウイルスベクターが対象においてペプチド特異的な免疫応答を刺激することが可能である方法。
- 対象においてペプチド特異的な免疫応答を刺激することが可能であるポリリジン改変ポリペプチドの使用であって、共有結合的ないしは非共有結合的に前記ポリペプチドをカプシドに連結することによってアデノウイルスのカプシドを被覆するための使用。
- 前記ポリペプチドが、静電的、ジスルフィドまたはアミド結合連結によってカプシドに結合されているか、または同時送達されて単一のナノ粒子のカプシドに付着されている、請求項1から12のいずれか一項に記載の方法、使用またはアデノウイルスベクター。
- 前記ウイルスカプシド上に付着したポリペプチドは、すべて同じポリペプチドであるか、または2種以上の異なるポリペプチドから選択される異なるペプチドである、請求項1から13のいずれか一項に記載の方法、使用またはアデノウイルスベクター。
- 前記ウイルスカプシド上に付着したポリペプチドが、クラスI(MHC−I)特異的ポリペプチドの主要組織適合性複合体、クラスII(MHC−II)特異的ポリペプチドの主要組織適合性複合体、疾患特異的ポリペプチド、腫瘍特異的ポリペプチドおよびDC特異的ポリペプチドからなる群から選択される、請求項1から14のいずれか一項に記載の方法、使用またはアデノウイルスベクター。
- 前記ウイルスカプシド上に付着したポリペプチドが、同時にMHC−I特異的および疾患特異的なポリペプチドの両方、MHC−I特異的および腫瘍特異的なポリペプチドの両方、同時にMHC−I特異的およびDC特異的および疾患特異的なポリペプチド、または同時にMHC−I特異的およびDC特異的および腫瘍特異的なポリペプチドである、請求項1から15のいずれか一項に記載の方法、使用またはアデノウイルスベクター。
- 前記アデノウイルスベクターの骨格の血清型が、血清型3または5から選択される、請求項1から16のいずれか一項に記載の方法、使用またはアデノウイルスベクター。
- 前記アデノウイルスベクターが腫瘍崩壊性アデノウイルスベクターである、請求項1から17のいずれか一項に記載の方法、使用またはアデノウイルスベクター。
- 前記アデノウイルスベクターが24bp欠失またはE1遺伝子欠失を含むか、または前記ベクターがヘルパー依存性ベクターである、請求項1から18のいずれか一項に記載の方法、使用またはアデノウイルスベクター。
- 前記アデノウイルスベクターが一または複数の導入遺伝子を含む、請求項1から19のいずれか一項に記載の方法、使用またはアデノウイルスベクター。
- 前記アデノウイルスベクターがカプシド修飾を含む、請求項1から20のいずれか一項に記載の方法、使用またはアデノウイルスベクター。
- 前記アデノウイルスベクターが、Ad5核酸骨格と、Ad3ファイバーノブ、Ad35ファイバーノブ、Ad5/3キメラファイバーノブおよびAd5/35キメラファイバーノブからなる群から選択されるファイバーノブとを含むAd5/3またはAd5/35である、請求項1から21のいずれか一項に記載の方法、使用またはアデノウイルスベクター。
- 請求項9、10および13から22のいずれか一項に記載のアデノウイルスベクターを含む薬学的組成物。
- 対象から腫瘍特異的およびMHC−I特異的なポリペプチドを同定するための方法であって、
i)対象の腫瘍細胞にアデノウイルスベクターを感染させる工程、
ii)対象の樹状細胞にアデノウイルスベクターを感染させる工程、
iii)工程i)の腫瘍細胞および工程ii)の樹状細胞からMHC−I分子を単離し、両グループからMHC−I関連ポリペプチドを同定する工程、
iv)感染しなかった腫瘍細胞からMHC−I分子を単離し、MHC−I関連ポリペプチドを同定する工程、
v)工程iii)およびiv)の感染した腫瘍および感染していない腫瘍、ならびに工程iii)の樹状細胞によって提示されたポリペプチドを同定する工程、
を含む方法。 - MHC−I分子を非感染の樹状細胞から単離し、MHC−I関連ポリペプチドを同定すること、および、
工程iii)およびiv)の感染した腫瘍および感染していない腫瘍によって提示されるポリペプチド、および感染していない樹状細胞ではなく工程iii)の感染した樹状細胞によって提示されるポリペプチドを同定すること
をさらに含む、請求項24に記載の方法。 - アデノウイルスカプシドをコーティングするための1または複数の腫瘍特異的およびMHC−I特異的ポリペプチドを選択するための、請求項24または請求項25に記載の方法。
