材料及び方法
動物及び心臓移植モデル
心臓のアロ移植を、以前に説明されたように(5)、全体としてMHC不一致の、LEW.1W(ドナーとしてのRT1.Au)及びLEW.1A(レシピエントとしてのRT1.Aa)のメスラット間で行った。実験は、動物実験についての地方倫理委員会により承認された。
ヒトIgG1の定常ドメインに融合したマウスCD40の細胞外部分をコードするAd(AdCD40Ig)及びリコンビナント非コードアデノウイルス(Ad)Addl324と、移植組織内送達の手法とは、以前に記載されている(5)。簡潔に、アデノウイルス粒子(2.1010個の感染性粒子)を、心室壁内の3点にゆっくり注入した。
4aaのラギングを有する16mer オーバーラップペプチドライブラリを、以前に公開されたように(17〜19)、MHC−I RT1.Au(アルファ1、2、及び3ドメイン)、MHC−II RT1.Bu(全ドメイン)、及びMHC−II RT1.Du(アルファ2及びベータ1ドメイン)の多型配列全体をカバーするように設計し、GL Biochem Ltd(Shangai, China)により調製した。凍結乾燥させたペプチドを、0.4% 無菌DMSO/滅菌水中に溶解させ、−80℃で保存した。対照ペプチドとして、in vitroにおいて、種々の同種非活性化ペプチド#7、26、及び39を使用し、in vivoにおいて、ペプチド#31を使用した。
T細胞を、以前に記載されたように(6)精製した。簡潔に、トータル脾細胞を、抗−γδ T細胞(V65)、抗−CD45RA細胞(OX33)、抗−CD161 NK細胞(3.2.3)、及び抗−CD11b/c単球(OX42)のカクテルにより、磁性ビーズ(Dynal)を使用して枯渇させた。濃縮されたT細胞を、抗−CD45RC−ビオチン(OX22)及びストレプトアビジン−PE−Cy7、抗−CD8α−PE(OX8)、抗−TCRαβ−Alexa 647(R73)、ならびに抗−CD25−FITC(OX39) mAbsによりラベルした。CD8+CD45RClow T細胞及びCD4+CD25− T細胞を、FACSAria(BD Biosciences, Mountain View, CA)によるTCRαβ+細胞のゲーティング後に分取した。分取した集団の純度は、99%超であった。
MLR共培養アッセイ法のために、LEW.1AナイーブラットからのpDC(1.25×104個)、同系CD8+CD40Ig Treg(5×104個)、及び120μg/mL 同種ペプチドを、丸底96ウェルプレート中の完全RPMI−1640培地 200μL中に、トリプリケートで、37℃、5% CO2で6日間蒔いた。pDCを、0.5μM CpG ODN 1826で成熟させた。
細胞外染色のために、細胞を、下記mAb:抗−TCRαβ(R73、Alexa Fluor 647−コンジュート)、抗−CD8α(OX8、PE−Cy7−コンジュート、ebiosciences)、抗−CD4(W3.25、PE−Cy7−コンジュート)、抗−CD45RC(OX22、FITC−コンジュート)、抗−CD28(JJ319、ビオチンラベル)、抗−CD71(OX26、ビオチンラベル)、抗−マウスVb11(KT11、ビオチンラベル、AbD Serotec)、抗−CD25(OX39、ビオチンラベル)、及び抗−MHC−II(OX6、ビオチンラベル)で染色した。
IFNγ、IL−10を、共培養上清中において、BD BiosciencesからのELISAであるOptEIAを使用して測定し、IL−12及びTGFβをそれぞれ、Invitrogen及びR&D SystemからのELISAを使用して測定した。
簡潔に、重鎖RT1−Aa及びβ2ミクログロブリン(β2m)を、pET24中にクローニングし、Escherichia coli BL21-DE3中で産生した。リコンビナントタンパク質を、封入体として産生し、8M 尿素中に溶解させ、ヒトHLA−A2/ペプチド複合体について以前に記載されたように(61)、in vitroでリフォールディングさせた。RT1−Aa、β2m、及びペプチドDu51を、0.4M L−アルギニン、0.1M Tris pH8、2mM EDTA、5mM 還元型グルタチオン、及び0.5mM 酸化型グルタチオン中において、4℃で5日間リフォールディングさせた。ついで、この溶液を濃縮し、アミコンメンブラン10Kd(Millipore, Bedford, MA)上でバッファー交換した。フォールディングされたMHC/ペプチド複合体を、BirA酵素(Avidity, Denvers CO)により、30℃で5時間ビオチン化し、Hiprep 26/10脱塩カラム(GE Healthcare)で脱塩した。ついで、MHC/ペプチド複合体を、アニオン交換Q-Sepharoseクロマトグラフィーにより精製した。ビオチン化を、4:1のモル比でのストレプトアビジン(Sigma Aldrich)との四量体化により試験した。
RT1.Aa/Du51の四量体化を、4:1のモル比での、ストレプトアビジン−PE(Jackson ImmunoResearch)又はストレプトアビジン−APC(BD Biosciences)を、15分間隔で加えた4等分アリコートにおいて加えることにより室温で行った。同様に、対照四量体RT1.Aa/MTF−E(ILFPSSERLISNR)を、ストレプトアビジン−BV421(Biolegend)にコンジュゲートさせ、Du51−特異的細胞の中でも、1.6+/−0.7% 非特異的染色を表わした。これら3つの試薬を混合し、蒔いた細胞に10μg/mLで4℃において1時間加えた。さらに、細胞を、CD8及びCD45RCで染色した。蛍光を、FACSCanto IIサイトメーター(BD Biosciences, Mountain View, CA)で分析した。まず、細胞を、その形態によりゲーティングし、死んだ細胞を、DAPI陰性細胞を選択することにより除外した。
16mer ペプチドを、注入前に0.4% DMSO/PBS中に溶解させた。1回目のプロトコールについて、単回投与のペプチド(500μg/注入)を、移植前後の種々の時点である、−6、−3、0、+3、及び+7日目に、移植したLEW.1Aレシピエント内にi.v注入した。2回目のプロトコールについて、ミニ浸透圧ポンプ(ALZET, Cupertino, CA, USA)を、レシピエントの腹腔内(i.p)に埋め込み、20.83又は41.66μg/時間のいずれかで、16mer ペプチドを連続的に14日間送達させた。1回目のポンプを、移植前−7日に埋め込み、+7日目に2回目のポンプに置き換えた。このポンプは、28日間連続で動物当たりに14又は28mg ペプチドの送達が可能である。枯渇性抗−CD8α mAb(OX8、IgG1、3mg/kg)又は抗−MHCクラスIa及びIb mAb(OX18、3mg/kg)を、−7日目から拒絶までに週2回、i.p注入した。移植臓器を、触診により毎日モニターした。移植臓器拒絶を、触知できる心拍の完全な停止として定義した。
細胞送達を、移植前日に致死量以下で照射(4.5Gy 全身照射)したLEW.1Aレシピエントへの、精製し、分取したトータル又はDu51四量体− CD8+CD40Ig Tregのi.v.注入により行った。トータル脾細胞(1,5.108個)を、同じ日に4.5Gy 全身照射を受けたナイーブなLEW−1Aレシピエントへの心臓移植前日に、養子性にi.v.送達した。レシピエントに、Du51処置ラットからの脾細胞(1回目のspl送達と定義)、ナイーブなラットからの脾細胞(ナイーブなspl送達)、又は細胞無し(未処置照射)を受けさせた。
移植片の上1/3を、パラホルムアルデヒド中で固定し、パラフィン中に包埋した。5μm 冠状切片を、ヘマトキシリン−エオシン−サフランで染色した。組織を、群に対して盲検の病理医(K.R.)により分析した。慢性拒絶を、以前に記載されたように(63)評価した。
アロ抗体を、他の箇所で記載されたように(28)、細胞蛍光測定法により分析した。簡潔に、コラゲナーゼDによる消化及び赤血球溶解後に、同種の脾細胞を、希釈(1/8)した熱不活性化血清と共にインキュベートし、ついで、FITC−コンジュゲート ヤギ抗−ラットIgG抗体(H+L鎖特異的)(Jackson Laboratories)、マウス抗−ラットIgG1 MAb(MCA 194、Serotec)、IgG2a(MCA 278、Serotec)、又はIgG2b(MCA 195、Serotec)と共にインキュベートした。抗体が結合したことを、FITC−結合F(ab)’2 ヤギ抗−マウスIgG(Jackson Laboratories)を使用して証明した。細胞を、FACS Canto IIサイトフルオロメーター(BD Biosciences, Mountain View, CA)を使用して分析した。結果を、各血清についての平均チャネル蛍光として表わした。
ペプチド活性化試験のために、ノンパラメトリックなウィルコクソンの符号順位検定を、1.0の仮定値に対するカラムメジアンと比較して行った。TCR Vβ11発現、活性化された細胞の表現型、サイトカイン発現、増殖アッセイ法、及び四量体染色についての統計学的有意性を、両側マン・ホイットニーt検定により評価した。移植片生存を、カプランマイヤー・ログランク検定により分析した。2要因の分散分析検定及びボンフェローニポストテストを、ドナー特異的抗体分析及び脾細胞表現型特徴決定に使用した。分析を、GraphPad Prism 5.04ソフトウェア(GraphPad, San Diego, CA, USA)により行った。多様性分析のために、クルスカル−ウォリス及びダンの多重比較ポストテストを、GraphPad Prism 6.0cを使用して行った。0.05未満のP値を、有意であるとみなした。
in vitroにおけるCD8+CD40Ig Treg活性化
CD8+CD40Ig Tregによる同種MHC/ペプチド複合体のTCR認識、及び、その機能のその後の活性化を特定するために、抗原性刺激に対する曝露に従って、フローサイトメトリーによる分析を可能にする活性化の特異的マーカーを選択する必要があった。したがって、ポリクローナル抗−CD3及び抗−CD28抗体による刺激に基づいて、CD8+CD40Ig Tregにより、種々の時点で発現された分子をスクリーニングした。新鮮に単離されたCD8+CD40Ig Tregにおける分子の発現は、Q RT−PCRにより以前に評価されており(5)、これらの分子の中でも、CD25及びIFNγが、他の細胞集団から、CD8+CD40Ig Tregを区別するマーカーであったことが証明された。CD71、CD25、及びIFNγの発現を、0、1、2、3、及び6日目において、フローサイトメトリーにより分析した。
ラットのMHCミスマッチ心臓移植臓器モデルにおいて、ドナー(RT1u)とレシピエント(RT1a)とは、全てのMHC分子についてミスマッチである。したがって、ドナーとレシピエントとのMHCI及びIIアミノ酸(aa)配列をアライメントし、ドナーMHCI及びII分子の多型ドメインとマッチした82個のオーバーラップした16merペプチドを設計した(17〜19)。まず、ペプチドを、6〜8個のペプチド(30μg/mL 各ペプチド)のプールに分類し、未成熟又は成熟した同系のレシピエントpDCとCD40Ig処置した長期移植臓器保有レシピエントから分取したCD8+CD40Ig Tregを3又は6日間共培養したin vitroアッセイ法で試験した。未成熟pDCに関して、CD8+CD40Ig Tregの有意な活性化は、ペプチドの同種プールのいずれについても、3日目又は6日目において観察されなかった。成熟pDC及び同種ペプチドプールによる刺激後、3日目に、CD8+CD40Ig Tregの小集団におけるCD25発現のわずかなアップレギュレーションが観察され、6日目に、同種刺激により、CD25発現の有意なアップレギュレーションが観察された。これらの結果から、一部の同種ペプチドは、CD8+CD40Ig Tregにより効率的に認識され、この認識により、CD25発現の増大がもたらされることが示唆された。また、pDCは、当該アッセイ法おいて成熟している必要があることも証明された。
抗原特異的CD8+CD40Ig Tregにより認識される天然のドミナントなドナーペプチド配列を決定するために、変性ペプチドのライブラリを使用した。この変性ペプチドは、1つのaaラギングを有する9mer ペプチド〜ドミナントな16mer Du51由来の2つ以上のaaラギングを有する15mer ペプチド(#51−1〜#51−18でラベル)の範囲とした(図1B及び表1)。このライブラリ設計は、以前の結果に基づいており、レポートで報告されており(20〜23)、上記されたin vitroアッセイ法と同様に試験した。興味深いことに、ランダムペプチドライブラリにより、ラットMHCクラスI RT1−Aa分子が、C末端にアルギニン(R)を有する9〜15mer ペプチドに強力な優位性を示したことが確立されている(23)。
Du51活性化CD8+CD40Ig Tregの表現型を、刺激後6日で分析した。CD8+CD40Ig Tregが、高レベルのIFNγの分泌を介して働き、次に、DC及び移植片ECによりIDO発現を誘導し、この反応が、in vivoでの寛容誘導に必須であったことが、以前に証明されている(5)。これらの結果に基づいて、pDCの存在下におけるペプチドによるCD8+ Tregの刺激が、大方活性化されたCD8+CD40Ig Tregにより、IFNγの有意に増大した発現をもたらすことが観察された。同じ培養上清中において、pDC起源であろうIL−12産生の低下が観察されたが、CD8+CD40Ig Treg及びpDCの両方により産生することができるIL−10及びTGFβの発現の改変は観察されなかった。また、CD71、CD28、及びMHCクラスIIのアップレギュレーションも観察されたが、ペプチド刺激の6日後に、Foxp3発現の改変は観察されなかった。
移植における重要な課題は、依然として、抗原特異的Tregの特定である。より活性なサプレッシブ集団を表わすためである。ここまで、非常に少ない天然エピトープしか特定されていない。加えて、今日、ラット及び移植環境において、抗原特異的CD8+ Treg集団を、直接的に可視化及び検出するのに利用できるツールは存在しない。そのためにも、フィコエリスリン(PE)及びアロフィコシアニン(APC)でラベルした、MHCクラスI四量体であるRT1.Aa/Du51を生成し、脾臓及び移植片中において特定集団を染色した。
Du51刺激CD8+CD40Ig Tregのサプレッシブ能が、以前に証明されている。この実験において、新鮮に分取したRT1Aa/Du51四量体特異的CD8+CD40Ig Tregのサプレッシブ能を研究しようとした(図2A)。ナイーブなCD8+CD45RClow Treg、トータルCD8+CD40Ig Treg、RT1Aa/Du51四量体陰性(四量体−)、及びRT1Aa/Du51四量体陽性(四量体+)のCD8+CD40Igを分取し、同種(直接的経路)又はアロ抗原ロード同系pDC(間接的経路)及びナイーブなCFSEラベル同系CD4+CD25− T細胞と共に、6日間インキュベートした。このアッセイ法において、RT1Aa/Du51四量体特異的CD8+CD40Ig Tregを、四量体結合により活性化させた(データを示さず、及び(27))。以前に説明されたように、トータルCD8+CD40Ig Tregは、アロ抗原定提示の直接的及び間接的経路の両方により誘導されるエフェクターCD4+CD25− T細胞の増殖を、ナイーブなCD8+CD45RClow Tregより効率的にサプレッションした(図2A)。興味深いことに、非自己認識の直接的経路により媒介される四量体+ vs 四量体− CD8+CD40Ig Tregのサプレッシブ活性間に、有意差が観察された。四量体+ CD8+CD40Ig Tregが、最も活性なサプレッサー細胞部分集合であった(図2A)。有意ではなかったが、統計学的な傾向を有して(p=0.0571)、同じ差異が、間接的なアロ抗原提示経路に関して得られた(図2A)。さらに、CD4+CD25− T細胞増殖のサプレッションが、直接的よりむしろ、間接的なアロ抗原提示経路により誘導された場合、Du51特異的Tregにより効果的に達成された。興味深いことに、非特異的四量体− CD8+CD40Ig Tregは、トータルCD8+CD40Ig Tregより低いサプレッシブ能を有する傾向にある。このことは、トータルCD40Ig Tregプールの全体的なサプレッシブ能における、Du51特異的CD8+ Tregの重要な寄与を強調している。これらの結果から、Du51抗原特異的CD8+ Tregが、トータルCD8+CD40Ig Tregプールの最も効率的なTregの部分集団であることが示唆された。
CD8+CD45RClow Tregのin vivoでの生成及び移植臓器生存において特定された免疫ドミナントなペプチドの活性を更に決定するために、動物を他の処置をすることなく、2種類のプロトコールのペプチド投与を使用して別々に処置した。第1のプロトコールにおいて、動物に、500μg ペプチドの静脈内(i.v)注入を、5回受けさせた(図3A)。対照ペプチド又はDu51のいずれかの注入は、移植臓器生存の有意な延長を誘導するのに十分でなかったことが観察された(それぞれ、11及び9.5日、n=4)(図3A)。第2のプロトコールにおいて、このような小型のペプチドがレシピエントの体内から急速に排出されることから、処置の有効性を改善するために、1時間当たりに20.8μg ペプチドの一定の腹腔内送達のためのミニ浸透圧ポンプを、移植前−7日から開始して28日間試験した。興味深いことに、このプロトコールは、対照ペプチドと比較して、Du51を使用する25%の明確でない移植臓器生存を伴って、移植臓器生存の有意な延長を可能にした(対照ペプチド及び未処置と比較して、p<0.01)(図3A)。ペプチド注入により誘導された寛容が、CD+ T細胞及びMHCクラスI提示に依存することを証明するために、以前説明されたように(5)、レシピエントを、ペプチド注入及び枯渇性抗−CD8 mAb(OX8)又は阻害性抗−MHCクラスI mAb(OX18)により同時処置した(図3A)。両抗体の投与により、移植臓器生存が完全に消滅した。このことは、CD8+ T細胞によるMHCクラスI/抗原の認識が、ペプチド注入により得られる寛容の確立に必要であったことを示している。
1mg/日 in vivoでのペプチドDu51処置により生じた長期移植臓器生存に関与する誘導されたレギュラトリー細胞のドミナントなサプレッシブ活性を評価するために、ナイーブな移植され、致死量以下で照射されたレシピエントへの、長期生存レシピエントの脾細胞を使用する養子細胞送達実験を、先に行ったのと同様に(5)行った(図4A)。二次的でナイーブな移植照射レシピエントへの150.106個の脾細胞の1回目の養子性送達は、50%のレシピエントにおける移植臓器生存の十分な延長をもたらした。このことは、ドミナントなレギュラトリー細胞の誘導が、新たに移植され照射されたレシピエントにおいて、移植臓器拒絶を防止可能であったことを証明している。1回目に養子性に送達された長期脾細胞レシピエントの移植片の解剖病理学的状態を調査した。血管病変及び障害が完全に存在しない(すなわち、慢性拒絶のサインがない)ことが観察された。最後に、長期間生存している1回目の養子性に送達されたレシピエントにおいて、高いアロ抗体産生を表わす未処置ラットと比較し、慢性拒絶の非存在に関連する(28)ナイーブなラットと比較可能な、総IgG、IgG1、及びIgG2bアロ抗体産生の総体的な阻害が観察された(図4B)。
Tregがペプチドを認識する方法及びこの認識の役割についての現在の知識は、非常に限られており、ほとんどが、CD4+ TregのTCR遺伝子伝達を使用するトランスジェニックマウスモデル(8、11)、又は、自己免疫疾患及びガンに関与するマウスQa−1制限CD8+ Tregのいずれかに基づいている。このQa−1ペプチドレパートリーは、ここ数年で説明されてきた(29)。しかしながら、これらの研究からは、抗原特異的Tregは、長期移植の結果に重要な影響を及ぼし、寛容の確立に寄与することが示唆されている(30)。現在の研究では、ガン(ヘムオキシゲナーゼ−1由来ペプチド)、自己免疫(Vβ由来ペプチド)の間に、又は、妊娠(マイナー抗原由来ペプチド)の間にも、CD8+ Tregにより認識されるペプチドが特徴決定されているが、移植においてはまだない(2)。移植は、移植片の存在がアロ抗原の連続的な供給源であり、Tregにより認識される抗原が特定される環境である。このことは、レギュラトリー集団の機能及び維持、このため、移植臓器の生存に最も確かに必須である(31)。レギュレーションの文脈におけるアロ抗原の認識は、特に、pDCにより支持される提示の間接的な経路により主に生じることが、本発明者らにより示されている(6、30、32)。

Claims (15)
- アミノ酸配列:REEYARFDSDVGEYR(配列番号:1)又は下記ペプチド:
を除く、その機能保存変異体を含むMHCクラスII分子由来の、15〜40個のアミノ酸の範囲の長さを有する、
単離されたペプチド。 - 前記ペプチドが、アミノ酸配列:REEYARFDSDVGEYR(配列番号:1)又はその機能保存変異体を含むか、又は、からなる、15又は16個のアミノ酸長のペプチドである、請求項1記載のペプチド。
- ペプチドが、配列番号:1に対して、1、2、3、又は4個のアミノ酸が異なってもよい、請求項2記載のペプチド。
- ペプチドが、
からなる群より選択されるアミノ酸配列からなる、請求項1〜3のいずれか一項記載のペプチド。 - アミノ酸配列:REEYARFDSDVGEYR(配列番号:1)もしくはその機能保存変異体を含むか、又は、からなる、15〜40個のアミノ酸の範囲の長さを有するペプチドをコードする核酸配列、又は、このような核酸配列を含む発現ベクター、又は、このような発現ベクターを含むホスト細胞。
- アミノ酸配列:REEYARFDSDVGEYR(配列番号:1)もしくはその機能保存変異体を含むか、又は、からなる、15〜25個のアミノ酸の範囲の長さを有するペプチドを含む、
