本発明は、グルコース刺激によるインスリン分泌の能力を有するインビトロ生成ベータ細胞を含む組成物、胚性幹細胞もしくは人工多能性幹細胞に由来するベータ様細胞またはヒト脂肪由来幹細胞(ADSC)からベータ細胞成熟を誘導する方法、および、1型糖尿病、2型糖尿病、または関連障害の治療のためにインビトロ生成ベータ細胞を使用する方法を提供する。
若齢/新生児のヒトおよびげっ歯類ベータ細胞は、グルコース刺激によるインスリン分泌(GSIS)の機能が乏しい。ベータ細胞成熟の過程で、新生児細胞は、グルコース刺激に応答してインスリンを分泌する能力を発達させる。発生の過程で、ベータ細胞は、グルコースに応答してインスリンを強く分泌する能力を獲得する。従って、ベータ細胞は、機能的ベータ細胞成熟期間のあいだにインスリン分泌を獲得する。完全に機能的なベータ細胞は寿命が長く、インスリン産生および栄養摂取(例えばアミノ酸、遊離脂肪酸、およびグルコースの摂取)に応答した分泌を制御することにより、全身のグルコース恒常性を緊密に制御する。インスリンは、肝臓、骨格筋、および脂肪のような末梢組織において脂質合成およびグルコース取り込みを促進させるので、ベータ細胞機能の喪失が1型および2型の両方の糖尿病につながることは驚きではない。

本発明は、ベータ様細胞においてERRガンマを過剰発現することによってベータ様細胞をリプログラミングし、ベータ様細胞がグルコース刺激によってインスリンを分泌できるようにするための方法を提供する。典型的には、ERRガンマの過剰発現は、ミトコンドリア代謝活性の上昇にも関連する。
本発明の方法において有用な細胞としては、ベータ細胞またはベータ細胞前駆体において典型的に発現されるマーカーを発現する、実質的にあらゆる細胞型が挙げられる。特定の実施形態において、本発明において有用な細胞は、ERRガンマの過剰発現によって、グルコース刺激によるインスリン分泌を獲得するように誘導することができる。
本発明はさらに、本発明のいずれかの天然ポリペプチドのアナログを包含する。アナログは、本発明の天然ポリペプチドと比べて、アミノ酸配列の違い、翻訳後修飾、またはその両方によって異なり得る。本発明のアナログは、一般的に、本発明の天然ERRガンマアミノ酸配列の全部または一部分と少なくとも85%、より好ましくは90%、そして最も好ましくは95%あるいは99%もの同一性を示すであろう。配列比較の長さは少なくとも5、10、15、または20アミノ酸残基であり、好ましくは少なくとも25、50、または75アミノ酸残基であり、より好ましくは100アミノ酸残基超である。ここでもやはり、同一性の程度を決定する例示的アプローチにおいてBLASTプログラムを使用することができ、e-3とe-100の間の蓋然性スコアが、密接に関係した配列を示す。修飾には、ポリペプチドのインビボおよびインビトロにおける化学誘導体化が含まれ、例えばアセチル化、カルボキシル化、リン酸化、またはグリコシル化が含まれる。そのような修飾は、ポリペプチドの合成もしくはプロセシングの際に、または、単離された修飾酵素を用いた処理の後に、起こり得る。アナログは、一次配列の変化によっても本発明の天然ポリペプチドから異なり得る。これらには、自然の、または誘導された遺伝学的バリアント(例えば、放射線照射もしくはエタンメチルスルフェートへの暴露によるランダム変異誘発からもたらされるもの、または、Sambrook, Fritsch and Maniatis, Molecular Cloning: A Laboratory Manual (2d ed.), CSH Press, 1989もしくは上記Ausubel et al.に記述されているように部位特異的変異誘発によるもの)が含まれる。例えばD-アミノ酸または非天然もしくは合成アミノ酸(例えばベータまたはガンマアミノ酸)のような、L-アミノ酸以外の残基を含む環化ペプチド、分子、およびアナログも包含される。
本発明は、1型糖尿病、2型糖尿病、糖尿病前症の治療、および、ベータ細胞数の欠乏(例えば、膵臓細胞の数の減少)またはベータ細胞の生物学的活性のレベルの不十分さ(例えば、グルコース刺激によるインスリン分泌の欠乏、インスリン産生の欠乏)に関連するその他の代謝性疾患もしくは障害の治療を提供する。例えば、本発明は、インスリン産生性膵臓細胞の数または活性の減少により十分なレベルのインスリンを欠く糖尿病患者の治療のための組成物を提供する。細胞数の欠乏に関連する多くの疾患は、ベータ細胞の消失またはベータ細胞死の増加により特徴付けられる。本発明の方法は、その欠乏を補充することができる細胞(例えばインスリン発現細胞)を生成することにより、そのような1型糖尿病、2型糖尿病、および関連疾患、障害を寛解させる。そのような細胞は、ある細胞を目的の細胞型にリプログラミングすることから(例えばベータ様細胞、胚性幹細胞、人工多能性細胞のリプログラミング)、または、ベータ細胞、膵臓組織、もしくは臓器の再生を促進させることにより、生成される。一般的に、本発明は、細胞をリプログラミングするための方法を提供し、この方法には、その細胞(例えば、人工多能性幹細胞もしくは脂肪細胞由来幹細胞から誘導されるもののようなベータ様細胞、内皮細胞、膵臓細胞、およびそれらの前駆細胞または幹細胞)を、ERRガンマをコードするポリヌクレオチドに接触させ、それによってその細胞をリプログラミングすることが関わる。特定の実施形態では、ベータ様細胞におけるERRガンマの発現は、その細胞における少なくとも1つ、2つ、3つ、4つ、5つ、またはそれより多数のポリペプチドの発現レベルを変化させ、および/またはミトコンドリア代謝活性を増加させる。
精製された細胞を含む本発明の組成物は、例えば等張性水溶液、懸濁液、乳濁液、分散系、または粘稠性組成物のような、無菌液体調製物として簡便に提供され得、これらは選ばれたpHに緩衝され得る。液体調製物は通常、ゲル、その他の粘稠性組成物、および固形組成物よりも調製することが容易である。加えて、液体組成物は投与(特に注射によるもの)がいくらか簡便である。一方で、粘稠性組成物は、特定の組織との間でより長い接触時間を提供するように適切な粘度範囲内に製剤することができる。液状または粘稠性の組成物は担体を含み得、これは、例えば水、食塩水、リン酸緩衝食塩水、ポリオール(例えばグリセロール、プロピレングリコール、液状ポリエチレングリコール等)およびそれらの適切な混合物を含有する、溶媒または分散媒であり得る。
本発明のERRガンマ発現ベータ様細胞またはその前駆細胞/子孫を含む組成物は、1型糖尿病、2型糖尿病、糖尿病前症の治療または予防、および、ベータ細胞数の欠乏(例えば、膵臓細胞の数の減少)またはベータ細胞の生物学的活性のレベルの不十分さ(例えば、グルコース刺激によるインスリン分泌の欠乏、インスリン産生の欠乏)に関連するその他の代謝性疾患もしくは障害の治療のために、対象に全身的または直接的に提供され得る。一実施形態では、本発明の細胞は、問題の臓器(例えば膵臓)に直接注射される。あるいは、本発明のベータ様細胞を含む組成物は、例えば循環系(例えば膵臓脈管構造)への投与によって、問題の臓器へ間接的に提供される。細胞の投与の前、最中、または後に、増殖剤および分化剤を供給して、インビトロまたはインビボでインスリン産生能力を有する細胞の産生を増加させることができる。細胞は生理学的に許容されるあらゆる媒体中において投与され得、通常は血管内に投与されるが、細胞が再生および分化のための適切な場所を見つけ得る都合のよい他の部位に導入されてもよい。
ここでは、1型糖尿病、2型糖尿病、糖尿病前症の治療または予防、および、対象におけるベータ細胞数の欠乏(例えば、ベータ細胞の数の減少)もしくはベータ細胞の生物学的活性のレベルの不十分さ(例えば、グルコース刺激によるインスリン分泌の欠乏、インスリン産生の欠乏)に関連するその他の代謝性疾患または障害の治療のための方法が提供される。特定の実施形態では、本発明は、1型糖尿病、2型糖尿病、糖尿病前症の治療または予防、および、ベータ細胞数の欠乏(例えば、膵臓細胞の数の減少)もしくはベータ細胞の生物学的活性のレベルの不十分さ(例えば、グルコース刺激によるインスリン分泌の欠乏、インスリン産生の欠乏)に関連するその他の代謝性疾患または障害の治療のための方法を提供する。糖尿病または代謝性障害を有する患者は、一般に、ベータ細胞の活性または機能の低下により同定され、例えば、血液中の血清糖レベルを監視することにより同定される。
本発明は、1型糖尿病、2型糖尿病、糖尿病前症の治療または予防、および、ベータ細胞数の欠乏(例えば、膵臓細胞の数の減少)もしくはベータ細胞の生物学的活性のレベルの不十分さ(例えば、グルコース刺激によるインスリン分泌の欠乏、インスリン産生の欠乏)に関連するその他の代謝性疾患または障害の治療のためのキットを提供する。一実施形態では、キットは、ERRガンマを発現する細胞(例えばベータ様細胞)の有効量をユニット投与量形態において含有する治療用または予防用組成物を含む。いくつかの実施形態では、キットは、治療用または予防用細胞組成物を含有する無菌容器を含む。そのような容器は、箱、アンプル、ボトル、バイアル、チューブ、袋、パウチ、ブリスター包装、または本技術分野において知られるその他の適切な容器形態であり得る。そのような容器は、プラスチック、ガラス、ラミネート紙、金属ホイル、または医薬を保持することに適したその他の材料で作られ得る。
ベータ細胞は出生後に機能的に成熟することが知られており、それには、グルコースに応答して確実にインスリンを分泌する能力の獲得が含まれる。2週齢新生仔マウスから単離された膵島は、未成熟表現形に合致して、グルコース負荷に応答してインスリンを分泌することができなかった(図1A)。グルコース刺激によるインスリン分泌(GSIS)のために必要な重要経路を同定するために、出生後成熟のあいだ、単離膵島のトランスクリプトームを比較した。ベータ細胞の最終分化は出生後には実質的に完了しているということ、および、成体β細胞は幹細胞分化ではなく自己複製により形成されるという事実と合致して(ENREF_13)(Dor et al., 2004, Adult pancreatic beta-cells are formed by self-duplication rather than stem-cell differentiation. Nature 429, 41-46)、げっ歯類およびヒトにおける膵臓内分泌の発生を制御することが知られる遺伝子の発現は、出生後にはほぼ変化していなかった(図2A、表2、Conrad et al., 2014, Trends in endocrinology and metabolism: TEM, 25(8): 407-414)。
膵臓ベータ細胞の機能的成熟におけるERRガンマの役割を調査するために、ERRガンマlox/loxマウスをラットインスリン2プロモーター(RIP)-Creマウスと交配することにより、ベータ細胞特異的ERRガンマノックアウト(ベータERRガンマKO)マウスを作製した。βERRガンマKOマウスは、メンデルの法則で予測される頻度で出生し、正常な体重と平均寿命とを示した(図4Aおよび4B)。野生型(wild-type)ERRガンマlox/lox膵島と比べてβERRガンマKOでは、RIP-Creリコンビナーゼが、視床下部ERRガンマ発現に有意な影響を与えることなくERRガンマ発現を選択的に80%減少させたが(図4Aおよび4B)、これは最近出された同様の報告と一致している(Tang et al., 2003, Cell metabolism 18, 883-895)。βERRガンマKOマウスの自由摂食血中グルコースレベルの監視により、雌マウスにおいて8週齢までに消える一時的な上昇が見出された(図4Cおよび4D)。しかしながら、8週齢では、ERRγlox/lox(WT)およびRIP-Cre(WT(RIP-Cre))同齢集団と比べてβERRγKOマウスは雄も雌もグルコース不耐容となった(グルコース負荷試験(GTT:グルコース負荷試験)により決定)(図5Aおよび6A)。インスリン感受性において著しい差は見られなかったが、βERRγKOマウスは、グルコース負荷に応答してインスリン分泌を増加させることができなかった(図5C、5D、ならびに図6Cおよび6D)。注目すべきことに、このβERRγKO表現型は代謝的ストレスによって増大された;4週齢から4週間にわたって高脂肪・高スクロース食餌で給餌されたβERRγKOマウスは、インスリン感受性の有意な変化を伴うことなく、より顕著なグルコース不耐性およびインスリン分泌異常を示した(図5B、5E、ならびに図6Bおよび6E)。
ミトコンドリア機能と形態は密接に相関することから(Tang et al., 2013, Cell metabolism 18, 883-895; Narkar et al., Cell metabolism 13, 283-293)、βERRγKOベータ細胞のミトコンドリアにおいて構造的変化が検出可能かどうかを調べた。電子顕微鏡により、インスリン顆粒およびプロインスリン顆粒、ならびに全般的ミトコンドリア数は、ERRガンマ欠失により影響されなかったことが明らかになった(図10Aおよび10B)。しかしながら、クリステ構造の崩壊を伴うミトコンドリア膨張が、ERRガンマ欠損ベータ細胞において見られ、これは、機能的に欠陥があるミトコンドリアの特徴である、ミトコンドリアの長さ、幅、および体積の著しい増加を伴っていた(図10A〜10C)。
移植可能なベータ細胞を多能性幹細胞から生成することは、幹細胞治療学の主要なゴールである。しかしながら、現在のiPSC由来ベータ様細胞は、グルコース負荷に応答してインスリンを分泌する能力を欠く点で胎生細胞に類似している(Hackenbrock et al., 1966, The Journal of Cell Biology 30, 269-297; Anello et al., 2005, Diabetologia 48, 282-289; Hrvatin et al., 2014, Proc. Natl. Acad. Sci. 111(8): 3038-3043; D’Amour et al., 2006, Nature Biotechnology 24, 1392-1401; Kroon et al., 2008, Nature Biotechnology 26, 443-452; Schulz et al., 2012, PloS one 7, e37004; Xie et al., 2012, Cell Stem Cell 12, 224-237; Sneddon et al., 2012, Nature 491, 765-768)。ベータ細胞成熟の際の増強される酸化的代謝におけるERRガンマの提唱された制御的役割に基づいて、ERRガンマの過剰発現が、ヒトiPSC由来ベータ様細胞が代謝の観点から成熟したベータ細胞に成熟することを促進させ得るか否かを調査した。この問題に取り組むために、ヒトインスリンプロモーター駆動GFPレポーターをスクリーニングおよび単離のために利用して、ヒトiPSCからインスリン陽性ベータ様細胞を産生するための分化プロトコールを最適化した(Pagliuca et al., 2014, Development 140, 2472-2483; Hrvatin et al., Proceedings of the National Academy of Sciences of the United States of America; 111(8): 3038-3043.)。最適化されたプロトコールでは、ヒト内皮細胞株HUVECに由来するiPSCから、分化開始後18〜21日で、インスリン陽性細胞が生成された(図13A〜13Cおよび図14)。iPSC分化のあいだの発現プロファイリングにより、ベータ様細胞の生成が確認された。具体的には、多能性マーカーNanogの発現が分化開始に際して失われた一方、分化の開始後18〜21日に、ベータ細胞終末分化マーカーであるインスリンが現れた。胚体内胚葉マーカーSOX17、膵臓前駆体マーカーHNF1β、および内分泌前駆体マーカーSOX9は、第7日目、15日目、および18日目頃にそれぞれ一時的に上昇した(図14)。さらに、PDX1、MAFA、PAX6、NEUROD1、GCK、CHGA、およびVAMP2を含む追加のベータ細胞マーカーの発現は、第21日において強く誘導された(図14)。免疫組織化学解析により、この最適化された21日間分化プロトコールが、ベータ細胞マーカーPDX1、c-ペプチド、およびプロホルモンカルボキシラーゼ1/3(PC1/3)を発現するベータ様細胞を生じたことが確認された(図5A)。この最適化分化プロトコールは、インスリン陽性、グルカゴン陰性β様細胞(本明細書においてiβL細胞と定義される)を再現性をもって産生し、電子顕微鏡によりインスリン顆粒の存在が明らかにされた(図13Hおよび図15B〜15E)。さらに、これらiPSC由来ベータ様細胞は、グルコース負荷に応答してc-ペプチドを分泌することはできなかった一方、KClによる直接的細胞脱分極媒介性インスリン分泌には応答性であった(図15F)。このことは、インスリン陽性iβL細胞におけるNkx6-1およびMafAの発現を確認した(図15H)。これらの結果は、ヒトベータ様細胞の生成を確証した。特定の理論に拘束されるわけではないが、これは、ミトコンドリア機能が、β様細胞におけるGSIS機能欠陥の1つの理由かもしれないことを示していた。
ベータ細胞特異的ERRガンマノックアウトマウス(βERRγKO)は、純粋なC57BL/6J遺伝学的バックグラウンドにあるERRγlox/loxマウスとRIP-Cre(B6N.Cg-Tg(Ins2-cre)25Mgn/J)マウスとを交配することにより産生した。タモキシフェン誘導可能ベータ細胞特異的ERRガンマノックアウトマウスは、ERRγlox/loxマウスとRIP-CreER(ストックTg (Ins2-cre/Esr1)1Dam/J)マウスとを交配することにより産生した。膵臓特異的ERRガンマノックアウトマウス(PERRγKO)は、ERRγlox/loxとPDX1-Cre(B6.FVB-Tg(PDX1-cre)6Tuv/J)とを交配することにより産生した。インスリンプロモーターGFP(MIP-GFP)マウス(Tg(Ins1-EGFP)1Hara)はジャクソンラボラトリーから購入した。ERRγ-LacZノックインマウスは以前に記述されている(Alaynick et al., 2007, Cell metabolism 6, 13-24)。グルコース負荷試験は、タモキシフェン誘導可能β細胞特異的ERRγノックアウトマウスの処置前(12週齢)および処置3週間後(16週齢)に行った。雄マウスにタモキシフェンを7日間毎日注射した(トウモロコシ油中2mg/kg、腹腔内)。
IP-GTTは、一晩絶食させたマウスにおいて行った。2 g/kgのグルコースの腹腔内投与の前ならびに15、30、60、および120分後に、グルコースPILOTを使用して血中グルコース値を評価した。グルコースの腹腔内投与の前ならびに5、15、および30分後に、ラット/マウスインスリンELISAキット(ミリポア)を使用して血清インスリンレベルを評価した。IP-ITTアッセイは、6時間の絶食後のマウスにおいて0.75 U/ kgのインスリン(ヒューマリンR、イーライリリー)の注射により行った。
マウス膵島は、ラットについて以前記述されたようにして単離した(Sutton et al., 1986, Transplantation 42, 689-691)。手短に述べると、HBSSバッファー中に希釈した0.5 mg/mlコラゲナーゼP(ロシュ)を総胆管を通して注入し、灌流された膵臓をウォーターバス中で解剖しインキュベートした(37℃、21分間)。消化された外分泌細胞と無傷の膵島とを、ヒストパック1077(シグマ)を使用して遠心分離(900g、15分間)で分離し、無傷の膵島を手で取り出した。全てのヒト膵島は、認可されたプロトコールのもとIntegrated Islets Distribution Program(IIDP)により提供された。ヒト膵島に関する追加的情報を表5に提供する。
無傷の膵島からのインスリン放出は、バッチインキュベーション法を使用して監視した。一晩培養した単離膵島(10%(v/v)ウシ胎仔血清および1%(v/v)抗菌・抗真菌剤(ギブコ)で補足されたRPMI-1640)を37℃にて30分間、前培養した(129.4 mM NaCl、3.7 mM KCl、2.7 mM CaCl2、1.3 mM KH2PO4、1.3 mM MgSO4、24.8 mM NaHCO3(5% CO2、95% O2、pH7.4で平衡化)、10mM HEPES、および0.2%(v/v)BSA(V画分、シグマ)を含有する(KRBH)クレブスリンゲル炭酸水素バッファー(KRBB)と3 mMグルコース)。それから、インスリン分泌レベルを決定するために膵島を3 mMまたは20 mMのグルコースを伴うKRBHバッファー中にインキュベートした(500μl/10膵島)。30分後、遠心分離により膵島をペレットにして、ELISAによってインスリンレベルを決定した(マウスおよびヒトの膵島について、それぞれ、ラット/マウスインスリンELISAキット(ミリポア)およびヒトインスリンELISAキット(ミリポア))。ヒトiPSC由来細胞については、細胞(24ウェルのウェル当たり1x106細胞)を3mMグルコースKRBHバッファー(500μl/ウェル)中で前培養した。それから、インスリン分泌レベルの指標としてのc-ペプチド分泌レベルを決定するために、細胞を3 mMまたは20 mMのグルコースを伴うKRBHバッファー(200μl/ウェル)中にインキュベートした。30分後、遠心分離により膵島をペレットにして、ヒトc-ペプチドELISAキット(ミリポア)によりc-ペプチドレベルを決定した。
INS-1細胞は、10%(v/v)ウシ胎仔血清、1%(v/v)抗菌・抗真菌剤(ギブコ)、10 mM HEPES、2 mMグルタマックス、1 mMピルビン酸ナトリウム、および50μMβメルカプトエタノールで補足されたRPMI-1640(シグマアルドリッチ)(INS-1用RPMI培地)において、空気中5%のCO2中で37℃にて培養した。INS-1細胞は、Plus試薬(インビトロジェン)を含有するLipofectamine2000でトランスフェクトした。INS-1細胞を、ERRガンマsiRNA(キアゲン)または陰性対照スクランブルsiRNA(キアゲン)で72時間トランスフェクトした。インスリン分泌は、前インキュベーションした細胞(一次膵島についてのインスリン分泌アッセイにおいて記述したように、3 mMグルコースを伴うKRBHにおいて37℃で30分間)において、30分間のグルコース負荷(3 mMまたは20 mMのグルコースを伴うKRBHバッファー)後にラット/マウスインスリンELISAキット(ミリポア)を用いて測定した。
TRIzol試薬(インビトロジェン)およびRNeasyキット(キアゲン)を使用して全RNAを抽出した。逆転写は、SuperScript IIIファーストストランド合成システムキット(インビトロジェン)またはPrimeScript RT試薬キット(タカラ)を用いて行った。リアルタイム定量的RT-PCR(qPCR)はSYBRグリーン(バイオラッド)を用いて行った。PCR分析は、表6に記載したオリゴヌクレオチドプライマーを用いて行った。
マウスインスリノーマ、MIN-6細胞からクロマチンを調製した。手短に述べると、MIN-6細胞を1%ホルムアルデヒドで10分間架橋し、その後125 mMのグリシンを加えた。クロマチンを酵素で剪断し(CHIP ITエクスプレスキット、アクティブモチーフ)、2μgの抗H3、対照マウスIgG、または抗ERRガンマ抗体で免疫沈降した。ChIP-qPCRプライマーは表6に記載されている。
膵臓試料を1 mm2の切片に切り、2%パラホルムアルデヒドおよび2%グルタルアルデヒドを含む0.1 mMリン酸ナトリウムバッファー(pH 7.4)中で4℃において36時間に渡り固定した。それからその組織片を洗浄し、段階的なアセトンを用いて脱水し、エポン・アラルダイト中に包埋した。ウルトラミクロトームを使用して超薄切片を調製した。切片化組織を1%トルイジンブルーホウ砂溶液で染色し、銅グリッド上に載せ、酢酸ウラニルで二重染色した後にJEM 100 CX-II電子顕微鏡で調べた。
H&E染色はパシフィックパソロジー(サンディエゴ)によって行われた。免疫染色は、膵臓の凍結切片および4% PFA固定化細胞に対して、以下の抗体を使用したZEISS共焦点顕微鏡分析によって可視化した:インスリン(1/100、アブカムab7842)、c-ペプチド(1/100、アブカムab14182)、グルカゴン(1/100、アブカムab10988)、ソマトスタチン(1/100、アブカムab103790)、プロホルモンカルボキシラーゼ1/3(1/100、ミリポアAB10553)、Pdx-1(1/100、アブカムab47267)。核染色のためにDAPI含有マウンティング媒体(蛍光用VECTASHIELDマウンティング媒体)を使用した。ERRγノックインマウス(Alaynick et al., 2007, Cell metabolism 6, 13-24)からの全膵臓はパラホルムアルデヒドおよびグルタルアルデヒドで固定し、凍結切片をX-galにより染色した。
Trizol試薬(インビトロジェン)を使用して、Ad-GFPまたはAd-Creで感染された膵島から全RNAを抽出し、その品質をアジレント2100バイオアナライザーにより決定した。イルミナTotalPrep RNA増幅キット(アンビオン)を使用して、500 ngのRNAをcRNAに逆転写しビオチンUTP標識した。cRNAは、アジレントバイオアナライザー2100を用いて定量化し、標準的プロトコール(イルミナ)を用いてイルミナのマウスRefseq-8v2 Expression BeadChipにハイブリダイズさせた。画像データは、イルミナGenomeStudioを用いて非正規化サンプルプローブプロファイル(unnormalized Sample Probe Profiles)に変換した。データはGeneSpring GXソフトウェアにより分析した。手短に述べると、チップ当たりの正規化は75パーセンタイルに対して設定し、遺伝子当たりの正規化は中央値および特定のサンプルに対して設定した。不在として指定された遺伝子はデータセットから消去され、WTより発現差が2倍の遺伝子を選択した。主にDAVIDソフトウェアを使用して、GO、経路解析、およびクラスター解析による組合せ解析を行った(Huang et al., 2009, Nature protocols 4, 44-57; Huang et al., 2009, Nucleic acids research 37, 1-13)。マイクロアレイデータはNCBI遺伝子発現オムニバスに登録され、GEOシリーズアクセス番号GSE56080によりアクセス可能である。
RNA miniキット(キアゲン)を使用して、RNAlaterで処理された細胞ペレットから全RNAを単離し、室温で30分間 DNaseI(キアゲン)で処理した。配列決定用ライブラリーは、製造会社のプロトコールに従ってTruSeq RNAサンプル調製キットv2(イルミナ)を使用して100〜500ngの全RNAから調製した。手短に述べると、mRNAを精製し、断片化し、ファーストストランドおよびセカンドストランドcDNA合成に使用した後、3’末端をアデニル化した。サンプルを固有のアダプターにライゲートし、PCR増幅した。それからライブラリーを、2100バイオアナライザー(アジレント)を用いて確認し、正規化し、配列決定のためにまとめた。
各実験条件について2〜3個の生物学的複製から調製されたRNA-Seqライブラリーを、イルミナHiSeq 2500において、バーコード化マルチプレックスおよび100 bpリード長を使用して配列決定した。画像解析およびベース判定はイルミナCASAVA-1.8.2を用いて行った。これは、サンプル当たり中央値29.9Mの使用可能リードを生じた。RNA-SeqアライナーSTARを使用して、短いリード配列をUCSC mm9参照配列にマッピングした(Dobin et al., 2013, Bioinformatics 29, 15-21)。アライナーにはmm9における既知スプライスジャンクションを供給し、新規のジャンクション発見も許容した。差次的遺伝子発現解析、統計学的検定、およびアノテーションは、Cuffdiff 2を用いて行った(Trapnell et al., 2013, Nature biotechnology 31, 46-53)。転写物の発現は、百万のマッピングされた断片につき1キロベースのエクソンモデルごとの断片(fpkm:fragments per kilobase of exon model per million)の遺伝子レベル相対的存在量として計算し、転写物存在量バイアスについての補正を採用した(Roberts et al., 2011, Bioinformatics 27, 2325-2329)。対象とする遺伝子のRNA-Seq結果はUCSCゲノムブラウザーを使用して視覚的にも探索した。RNA-Seqデータは、NCBI配列リードアーカイブにおいてアクセス番号SRP048600およびSRP048605のもとアクセスすることができる。
Huvecから誘導されたヒト人工多能性幹細胞(hiPSC)および胚性幹細胞(H9ES)はステムセルコア(ソーク研究所)から入手した。マトリゲル(BD)コーティングディッシュ上にて完全mTeSR培地中で細胞を維持した。膵臓分化のためには、スピンフェクション(800g、1時間)によってhPCをヒトインスリンレポーターレンチウイルス(pGreenZeroレンチレポーターヒトインスリン、System Biosciences)で感染させ、それから培地を2日間、100ng/mlヒトアクチビン(シグマ)、25ng/ml組換えヒトWnt3a(シグマ)を含む分化培地(800ml DMEM/F12、13.28g BSA、10mlグルタマックス、560mg NaHCO3、330mgチアミン、100mg還元グルタチオン、3300 mgビタミンC、14μgセレニウム、10ml NEAA、2mlトレースエレメントB、1mlトレースエレメントC、7μl β-ME、2ml DLC、2ml GABA、2ml LiCl、129.7μg PA、インスリン2mgを1000mlまでにする)に変え、それからさらに2日間、100ng/mlヒトアクチビンを含む分化培地に変えた(ステージ1)。続いて培地を、1μMドルソモルフィン(カルバイオケム)、2μMレチノイン酸(シグマ)、および10μM SB431542を伴う分化培地に7日間置き換えた(ステージ2)。それから培地を、10μMフォルスコリン(シグマ)、10μMデキサメタゾン(ステムジェント)、10μM TGFβRIキナーゼ阻害剤II(カルバイオケム)、10mMニコチンアミド(シグマ)を伴う分化培地に10日間置き換えた(ステージ3)。培地は、毎日(ステージ1)、毎日または2日毎に(ステージ2)、および2日毎に(ステージ3、ベータ様細胞)交換した。
酸素消費速度(OCR:Oxygen consumption rate)および細胞外酸性化速度(ECAR:extracellular acidification rate)は、XF96シーホース(Seahorse Biosciences)を使用して96-ウェルプレートにおいて記録した。手短に述べると、70単離膵島/ウェルをXF DMEM培地(pH7.4)および3mMグルコースで1時間前培養し、その後、グルコースを、最終濃度20mMになるまで段階的に追加した。グルコースを追加するあいだ、OCR(3mMグルコースと比較した%変化として報告される)を記録した。
レンチウイルスは、HEK293T細胞株において第2世代または第3世代レンチウイルス系を使用して産生した。アデノウイルスEGFPおよびCreはイリノイ大学から購入し、アデノウイルスERRガンマはウェルゲン社から購入した。
免疫不全NOD-SCIDマウス(NOD.Cg-Prkdcscid Il2rgtm1Wjl/SzJ)はジャクソンラボラトリーから購入し、ソーク研究所のSPF施設にて、オートクレーブ済みケージ中で飼育し維持した。マウスは、高用量のストレプトゾトシン(STZ;180mg/kg)の腹腔内(i.p.)単一注射により糖尿病にした。STZ注射の1週間後、400mg/dLより高いレベルの血中グルコースを有するマウスを、移植分析のためのレシピエントとして使用した。
代謝ケージ分析は、包括的実験動物モニタリングシステム(コロンバスインストゥルメンツ)を用いて行った。データを記録する前に少なくとも24時間適応させ、CO2産生、O2消費、呼吸交換比(RER)、およびx-ピークによる歩行カウントを5日連続して昼と夜に決定した。
結果は、平均±平均の標準誤差(s.e.m.:standard error of the mean)として表現した。統計学的比較はステューデントのt検定を使用して行った。統計学的有意差は*P<0.05として定義した。
上記説明から、本明細書に記述される発明にバリエーションおよび修飾を施して様々な使用および条件に適応させ得ることが明らかであろう。そのような実施形態も下記特許請求の範囲の範囲内である。
Claims (39)
- ベータ様細胞を機能的なベータ細胞へとリプログラミングするための方法であって、ベータ様細胞において組換えエストロゲン関連受容体(ERR)ガンマを発現させ、それによって前記ベータ様細胞を機能的なベータ細胞へとリプログラミングすることを含む、方法。
- グルコース刺激によるインスリン分泌の能力を有する細胞を生成するための方法であって、ベータ様細胞において組換えエストロゲン関連受容体(ERR)ガンマを発現させ、それによってグルコース刺激によるインスリン分泌の能力を有する細胞を生成することを含む、方法。
- 前記ベータ様細胞は、Nkx2.2、NeuroD1、Foxa2、Pax6、HNF4a、Pdx1、MafA、およびNkx6-1のmRNA発現からなる群から選択される1つ以上のβ細胞転写因子を発現する、請求項1または2に記載の方法。
- 前記ベータ様細胞は、インスリン1、インスリン2、グルカゴン、およびソマトスタチンからなる群から選択される1つ以上のβ細胞マーカーを発現する、請求項1または2に記載の方法。
- 前記ベータ様細胞は、胚性幹細胞、人工多能性幹細胞、脂肪細胞由来幹細胞、ヒト臍帯静脈内皮細胞(Huvec)、またはそれらの前駆細胞もしくは幹細胞である、請求項1または2に記載の方法。
- 前記ベータ様細胞は、インビトロまたはインビボでERRガンマを発現するように改変される、請求項1または2に記載の方法。
- 前記ベータ様細胞は、ERRガンマをコードするウイルスベクターにより形質導入される、請求項1または2に記載の方法。
- 前記リプログラミングされた細胞はインスリンを発現する、請求項1または2に記載の方法。
- 前記リプログラミングされた細胞は、グルコース刺激に応答してインスリンを分泌する、請求項1または2に記載の方法。
- 前記ベータ様細胞は、培養中の胚性幹細胞または人工多能性幹細胞を、アクチビンA、ウィングレス型MMTV挿入部位ファミリーメンバー3A(Wnt3a)、インスリン増殖因子(IGF)-2、エクステンディン(Ex)-4、線維芽細胞増殖因子(FGF)-2、ニコチンアミド、および/またはB27の内の1つ以上に接触させることにより得られる、請求項1〜9のいずれかに記載の方法。
- 前記ベータ様細胞は、3〜4日間に渡り毎日50〜100 ng/mlのアクチビンAにより処理される、請求項10に記載の方法。
- 前記ベータ様細胞は、第0日に25 ng/mlのWnt3aで処理される、請求項10に記載の方法。
- 前記ベータ様細胞は、第3日に、50 ng/mlのIGF-2、50ng/mlのEx-4、10 ng/mlのFGF-2、10 mMのニコチンアミド、および1%のB27で処理される、請求項10に記載の方法。
- 得られる前記ベータ細胞は、Pdx1、インスリン、Mafa、Pax6、NeuroD、GCK、CHGA、VAMP2、PC1/3、Glut2、Nkx6.1、GCG、SST、およびU36B4からなる群から選択される1つ以上のmRNAを発現する、請求項13に記載の方法。
- 前記ベータ様細胞は、胚性幹細胞または人工多能性幹細胞において、Oct4、Nanog、Sox17、FoxA2、Pdx1、Nkx6.1、およびNgn3からなる群から選択される1つ以上の転写因子を発現させることにより得られる、請求項1に記載の方法。
- 前記ベータ細胞は、インスリン、Pdx1、Mafa、Pax6、Glut2、NeuroD1、グルコキナーゼ、グルカゴン、ソマトスタチン、クロモグラニンA、およびVAMP2からなる群から選択される1つ以上のポリペプチドを発現する、請求項1〜15のいずれかに記載の方法。
- 必要とする対象において高血糖を寛解させる方法であって、組換えエストロゲン関連受容体(ERR)ガンマを発現するベータ様細胞を前記対象に投与して、それによって前記対象における高血糖を寛解させることを含む、方法。
- 必要とする対象において1型糖尿病または2型糖尿病を治療する方法であって、組換えエストロゲン関連受容体(ERR)ガンマを発現するベータ様細胞を前記対象に投与して、それによって前記対象における1型糖尿病または2型糖尿病を治療することを含む、方法。
- 前記方法は、前記対象において血中グルコースレベルを低下させる、請求項17または18に記載の方法。
- 前記方法は、前記対象において血中グルコースレベルを正常化させる、請求項17または18に記載の方法。
- 前記対象は獣医学的な対象またはヒト対象である、請求項17または18に記載の方法。
- ERRガンマポリペプチドをコードする核酸配列に作用可能に連結されたベータ細胞特異的プロモーターを含む発現ベクター。
- 前記プロモーターが、インスリン、インスリンII、Pdx1、Mafa、Nkx6-1、Pax4、およびNeuroD1プロモーターからなる群から選択される、請求項22に記載の発現ベクター。
- 前記発現ベクターがウイルスベクターである、請求項22に記載の発現ベクター。
- 前記ウイルスベクターは、レンチウイルスベクターまたはアデノ随伴ウイルスベクター(AAV)である、請求項24に記載の発現ベクター。
- 必要とする対象において高血糖を寛解させる方法であって、ERRガンマをコードするAAVベクターにベータ様細胞を接触させ、それによって前記細胞がグルコース刺激によるインスリン分泌の能力を有するようにすること、および、前記対象に前記細胞を投与し、それによって前記対象における高血糖を寛解させることを含む、方法。
- 前記ベータ様細胞は、前記対象に由来するものであるか、または前記対象に由来しないものである、請求項26に記載の方法。
- 請求項22または23に記載の発現ベクターを含む宿主細胞。
- 前記細胞は、胚性幹細胞、新生児幹細胞、成体幹細胞、人工多能性幹細胞、脂肪細胞由来幹細胞、ヒト臍帯静脈内皮細胞(Huvec)、またはそれらの前駆細胞もしくは幹細胞である、請求項28に記載の宿主細胞。
