Abstract

本明細書に記載されている抗体分子は、動物モデルにおいて評価することができる。例えば、動物モデルを使用して、デングウイルス感染、複製および/または伝染の低下における本明細書に記載されている抗体分子の有効性を検査することができる。本明細書に記載されている抗体分子の評価に使用することができる例示的な動物モデルとして、AG129マウスモデル(例えば、Tharakaramanら、Proc Natl Acad Sci U S A.2013年;110巻(17号):E1555〜64頁;Johnsonら、J Virol.1999年;73巻(1号):783〜6頁に記載);非マウス適応マウスモデル(例えば、TanらPLoS Negl Trop Dis.2010年;4巻(4号):e672頁に記載されている非マウス適応DENV−2 D2Y98Pマウスモデル);ヒト化マウスモデル(例えば、Sridharanら、J Virol.2013年;87巻(21号):11648〜58頁に記載);非ヒト霊長類モデル(例えば、GoncalvezらProc Natl Acad Sci U S A.2007年;104巻(22号):9422〜7頁に記載);および蚊モデル(例えば、VuらPLoS Negl Trop Dis.2010年;4巻(7号):e757頁に記載)が挙げられるがこれらに限定されない。
一部の態様において、本開示は、薬学的に許容される担体と共に製剤化された、本明細書に記載されている抗デング抗体分子を含む組成物、例えば、薬学的に許容される組成物を提供する。本明細書において、「薬学的に許容される担体」は、生理的に適合性のありとあらゆる溶媒、分散媒、等張性および吸収遅延剤その他を含む。担体は、静脈内、筋肉内、皮下、非経口的、直腸、脊髄または上皮投与(例えば、注射または注入による)に適することができる。ある特定の実施形態において、医薬組成物における抗体分子の約5%未満、例えば、約4%、3%、2%または1%未満が、凝集体として存在する。他の実施形態において、医薬組成物における抗体分子の少なくとも約95%、例えば、少なくとも約96%、97%、98%、98.5%、99%、99.5%、99.8%またはそれ超が、単量体として存在する。一部の実施形態において、抗体凝集体または単量体のレベルは、クロマトグラフィー、例えば、高速分子ふるいクロマトグラフィー(HP−SEC)によって決定される。
本開示は、本明細書に記載されている抗デング抗体分子の重および軽鎖可変領域ならびにCDRをコードするヌクレオチド配列を含む核酸も特色とする。例えば、本開示は、本明細書に開示されている抗体分子、例えば、表1の抗体または抗体の部分、例えば、表2の可変領域のうち1種または複数から選択される抗デング抗体分子のそれぞれ重および軽鎖可変領域をコードする第1および第2の核酸を特色とする。本核酸は、本明細書における表におけるアミノ酸配列、またはそれと実質的に同一の配列(例えば、それと少なくとも約85%、90%、95%、99%もしくはそれを超えて同一の、または本明細書における表に示す配列から3、6、15、30もしくは45ヌクレオチド以下が異なる配列)のうちいずれか1種をコードするヌクレオチド配列を含むことができる。
本明細書に記載されている抗体分子をコードするヌクレオチド配列を含むベクターがさらに本明細書に提供される。一部の実施形態において、本ベクターは、本明細書に記載されている抗体分子をコードするヌクレオチドを含む。一部の実施形態において、本ベクターは、本明細書に記載されているヌクレオチド配列を含む。ベクターとして、ウイルス、プラスミド、コスミド、ラムダファージまたは酵母人工染色体(YAC)が挙げられるがこれらに限定されない。

本明細書に開示されている抗体分子は、in vitroおよびin vivo診断ならびに治療および予防有用性を有する。一部の実施形態において、抗体分子は、デングウイルスを中和する。例えば、このような分子は、培養中の、in vitroもしくはex vivoの細胞に、または対象、例えば、ヒト対象に例えばin vivoで投与して、デングウイルスを中和することができる。したがって、一部の態様において、本開示は、対象におけるデングウイルス感染を処置する方法であって、デングウイルス感染が処置されるように、本明細書に記載されている抗体分子を対象に投与するステップを含む方法を提供する。例えば、このような抗体分子は、培養中の細胞に、例えばin vitroもしくはex vivoで、または対象に、例えばin vivoで投与して、デングウイルス感染を処置、防止および/または診断することができる。
デングウイルスは、ウイルス表面にE(エンベロープ)タンパク質を呈する。Eタンパク質は、宿主細胞へのウイルスの付着に寄与する。Eタンパク質は、DIドメイン(9本鎖ベータ−バレル)、DIIドメイン(宿主細胞との融合に関係付けられる疎水性ドメイン)およびDIIIドメイン(細胞外ドメイン)を含む。理論に制約されることを望まないが、一部の実施形態において、本明細書に記載されている抗体分子は、そのEタンパク質DIII(EDIII)ドメインに結合することにより、例えば、ウイルスが宿主細胞と融合するのを防止する、ウイルスが宿主細胞に結合するのを防止する、Eタンパク質の構造を破壊する、またはウイルスを不安定化することにより、デングウイルスを中和することができる。
本抗デング抗体分子は、他の治療法と組み合わせて使用することができる。例えば、併用療法は、1種または複数の追加的な治療剤、例えば、抗ウイルス剤(他の抗デング抗体を含む)、ワクチン(デングウイルスワクチンを含む)または免疫応答を増強する薬剤と同時製剤化および/または同時投与した抗デング抗体分子を含むことができる。他の実施形態において、本抗体分子は、静脈内水分補給、解熱剤(アセトアミノフェン等)または輸血等、他の治療的処置様式と組み合わせて投与される。斯かる併用療法は、投与される治療剤のより低い投薬量を有利に利用し、これにより、様々な単独療法に伴う可能性のある毒性または合併症を回避することができる。
一部の態様において、本開示は、in vitro(例えば、血液試料等、生物学的試料における)またはin vivo(例えば、対象におけるin vivoイメージング)でデングウイルスEタンパク質の存在を検出するための診断方法を提供する。本方法は、(i)試料を本明細書に記載されている抗体分子に接触させるステップ、または対象に抗体分子を投与するステップと、(任意選択で)(ii)参照試料、例えば、対照試料(例えば、血漿または血液等、対照生物学的試料)または対照対象を本明細書に記載されている抗体分子に接触させるステップと、(iii)抗体分子および試料もしくは対象または対照試料もしくは対象の間の複合体の形成を検出するステップであって、対照試料または対象と比べた試料または対象における複合体の形成の変化、例えば、統計的に有意な変化が、試料におけるデングウイルスの存在を示すステップとを含む。抗体分子は、検出可能物質で直接的にまたは間接的に標識して、結合または非結合抗体の検出を容易にすることができる。適した検出可能物質は、上述のおよびより詳細に後述する様々な酵素、補欠分子族、蛍光材料、発光材料および放射性材料を含む。
抗デング抗体の構造にガイドされる設計
エピトープおよび鋳型同定
様々な中和エピトープが、デングウイルスEタンパク質二量体に存在する。これらのエピトープは、例えば、EDI、EDII、EDIII、融合ループ、EDI/IIヒンジおよびEDIII「A」β鎖における領域を含む。EDIおよびEDIIは、ヒトにおいて免疫優性であり、弱く中和性だが高度に交差反応性の抗体を誘導することができる。EDIIIは、強力な中和抗体を誘導することができる。EDIIに位置する融合ループに対する抗体は、多くの場合、交差血清型反応性を提示するが、弱い中和活性である。EDI/IIヒンジ領域に対する抗体は、強力な中和を提示することができるが、典型的には、このエピトープ領域の低い保存のために血清型特異的である。EDIII A鎖に対する抗体は、多くの場合、一部には、抗体へのこの領域のより優れた到達性のために高い効力を提示するが、多くの場合、限定的な交差血清型反応性を有する。
タンパク質工学のための従来の計算的アプローチは、典型的には、エネルギー論に基づく方法に頼る。これらの方法の結果は、一般に、構造またはモデルにおける正確な原子位置に対し高度に感応性であり、したがって、これらの方法は、典型的には、的確なモデリングおよび有益変異予測のために、タンパク質−タンパク質複合体の高分解能かつ高品質の結晶構造または同様のデータを必要とする。その上、タンパク質工学に対する従来のエネルギー論に基づくアプローチは、典型的には、抗体特異的特性および知識を取り込まない。
4E11−EDIII構造的モデルを作製し、親和性増強変異/位置を予測した。例えば、特異的部位における個々の変異を予測し、合理的集中的コンビナトリアルライブラリーの作製のための位置を選択した。図23に示す通り、得られたヒト化抗体D88は、抗体4E11と比べて、DENV−4に対する10,000倍の親和性増と、同時発生的なDENV1〜3に対する親和性改善を実証する。
抗デング抗体4E11(この抗体は、Thullierら、J Biotechnol.1999年4月15日;69巻(2〜3号):183〜90頁に記載)およびA11(4E5A)の間の比較は、Tharakaramanら、Proc Natl Acad Sci U S A.2013年;110巻(17号):E1555〜64頁に示されている。4E5Aは、A55E(VH)、R31K(VL)、N57E(VL)、E59Q(VL)およびS60W(VL)に、4E11と比べて5個の点変異を有する(Tharakaramanら、2013年、Proc Natl Acad Sci U S A.2013年;110巻(17号):E1555〜64頁)。
Del26は、DV−4中和活性を改善する。
B11は、A11(4E5A)と比べて、フレームワーク1(FR1)におけるS26の重鎖欠失を有する抗デング抗体である。4G2は、対照抗デング抗体である。4種のデングウイルス血清型由来のEDIIIに対する各抗体の中和活性を、フォーカス減少中和検査(FRNT)により検査した。
ヒト化抗デング抗体。
図4は、A11およびB11に関係するいくつかのヒト化抗体について記載する。様々なヒト化フレームワークを検査した。最右4列は、デングウイルス血清型DV−1、DV−2、DV−4およびDV−4のEDIIIに対する各抗体の親和性を示す。重鎖FW3におけるA98V変異(抗体B48+A98Vを抗体B48と比較)は、他の血清型への結合を保持または改善しつつ、DV−4への結合を約5倍改善した。
ヒト化抗デング抗体の逆変異。
特にDV−4におけるEDIIIに対する抗体親和性を改善するために、選択されたヒト化抗体の重鎖N末端に様々な逆変異を作製した。図5は、N末端の完全マウス復帰であるD48が、完全にヒト化されたN末端を有する抗体と比べて、親和性に約2倍の改善を有することを示す。他の事例において、逆変異は、EDIII−DV−4に対し同様の、僅かにより低い、または僅かにより高い親和性をもたらした。
親和性増強変異の組合せによる抗体親和性の改善。
DV−4に対するヒト化抗体の親和性をさらに改善するために、様々な親和性増強変異を単独でまたは組み合わせて検査した。図6において、次の変異を検査した:全てVHにおけるT33V、del26、G27A、G27Y、F28W、F28G、F28AおよびF28Y。Del26およびT33Vは共に(抗体D88における)、T33V変異単独(抗体C88における)と比較して、EDIII−DV−4に対する親和性を約4倍改善することが見出された。抗体D88に存在する二重del26およびT33V変異は、また、del26変異単独を有する抗体よりも親和性を改善する。このような結合の相加的または相乗的改善は、予想外であった。
ヒト化抗デング抗体における追加的な結合試験。
数種類のヒト化抗デング抗体を、デングウイルス血清型DV−1、DV−2、DV−3およびDV−4由来のEDIIIに結合するその能力に関して検査した。図8は、競合ELISAアッセイによって決定される抗体の親和性を示し、図9は、SPRによって決定される抗体の親和性を示す。全ヒト化抗体は、強い親和性を有するが(例えば、全て24.35nM未満またはそれに等しく、多くはナノモル濃度に満たない(sub−nanomolar)範囲内である)、一部の抗体は、特に強い結合を有する。例えば、C88およびD88は、C98の場合よりも約20倍優れた親和性でDV−1に結合する。図9の全3種の抗体は、DV−2およびDV−3に対しほぼ等しい親和性を有する。図9における抗体のうち、D88は、DV−4に対し最も強い親和性を有する。
ヒト化抗デング抗体は、血清型DV−2、DV−3およびDV−4の多数の株由来のEDIIIに結合する。
D88(図11、最右列)およびC88(右から2番目の列)を、デングウイルスの様々な株から単離されたEDIIIに結合する能力に関して検査した。株の血清型および地理的位置を最左2列に示す。一般に、抗体は、結合の十分な幅を呈し、広範囲のデングウイルス株に対し有効である可能性があることを示す。
ヒト化抗デング抗体は、フォーカス減少中和検査においてデングウイルスを中和する。
ヒト化抗体は、フォーカス減少中和検査においてDV−1、DV−2、DV−3およびDV−4を中和する能力を有する。このアッセイは、上の実施例2に記載されている。図12、図13および図14は、代表的DV−3およびDV−4株によるこれらの実験の結果を示す。ヒト化抗体は、このアッセイにおけるデングウイルスの中和において有効である。例えば、A48、C98およびD88は、マウス抗デング抗体A11よりも低いDV−4に対するEC50を達成する(図12)。図13は、抗体D88、D188およびD128が、DV−3のこの株に対する426〜506ng/mlの範囲内のEC50を有することを示す。加えて、D88、D188およびD128は、マウス抗デング抗体A11よりも低いDV−4に対するEC50を有した(図14)。ヒト化抗体は、DV−1およびDV−2を中和する能力も有する(データ図示せず)。ヒト化抗体が、マウス抗デング抗体よりも高い活性を達成することは予想外であった。少なくとも一部には、本明細書に示す結合データおよび公知デング分離菌の配列解析に基づき、本明細書に記載されているヒト化抗デング抗体分子は、デングウイルスの多様性の代表的な本明細書に収載されている株によるあらゆる血清型および遺伝子型にわたり幅広い中和能力を有する可能性がある。
ヒト化抗体の熱安定性を熱シフト解析により検査した。
試薬Sypro Orangeを使用した熱シフト解析により、選択されたヒト化抗体を安定性に関してアッセイした。実験の結果を図15A、図15Bおよび図15Cに示す。一般に、ヒト化抗体は、多くの場合、約64〜68の間のTmを有する優れた安定性を示した。
デングウイルスのためのマウスモデル。
本明細書に記載されている抗体分子は、動物モデルにおける有効性に関して検査することができる。斯かるモデルの1種、AG129マウスモデルは、Tharakaramanら、2013年、Proc Natl Acad Sci U S A.2013年;110巻(17号):E1555〜64頁に記載されている。I型およびII型インターフェロン受容体の両方を欠くAG129マウス株は、ウイルス血症および他の疾患徴候を含む、デングの臨床症例において観察される疾患顕在化の一部を再現する一般的に使用される動物モデルである。感染したAG129マウスは、このウイルスに感染したヒト患者においてごく稀にしか観察されない、脳におけるDENV複製に伴う神経学的機能障害を経験することができる。これは多くの場合、ウイルス負荷のほぼ18日後に、感染したAG129マウスに死亡をもたらす(Stein, D.A.ら、J Antimicrob Chemother、2008年、62巻(3号):555〜65頁;Johnson AJ、Roehrig JT. J Virol.1999年1月;73巻(1号):783〜6頁)。ヒト感染と類似性を欠くにもかかわらず、このモデルは、抗ウイルス処置の評価において有用であり、原理研究の証明において使用することができる。簡潔に説明すると、AG129(IFN−α/βおよびIFN−γ受容体を欠損)マウスに、デングウイルスを負荷し、候補治療抗体分子を投与する。典型的には、ウイルス血症(血液試料におけるウイルス力価)は、実験のエンドポイントである。ウイルス血症は、例えば、定量的RT−PCRにより測定することができる。この試験において、ウイルス血症、体重変化および生存を、DENV−2のニューギニアC(NGC)株感染後の疾患徴候として使用した。本実施例に示す結果は、DENV−2マウスモデルにおける抗体D88の強いin vivo阻害活性を実証する。
in vivoにおけるADEからの保護
本明細書に記載されている抗体分子は、動物モデルにおける有効性に関して検査することができる。斯かるモデルの1種、AG129マウスにおける抗体増強性重症デングウイルス感染は、Balsitisら、2010年(Lethal antibody enhancement of dengue disease in mice is prevented by Fc modification、PLoS Pathogens、2010年2月12日;6巻(2号):e1000790頁)に記載されている。簡潔に説明すると、デングウイルス(例えば、D2S10)負荷1日前に、AG129マウスにデングウイルス増強抗体(例えば、DV1抗血清または4G2モノクローナル抗体)を投与する。負荷1日前に(予防)または負荷1もしくは2日後に(治療)、候補抗体分子を投与する。典型的には、死亡率は、実験のエンドポイントである;血清におけるウイルス血症および炎症性サイトカイン(例えば、TNF−α)レベルをエンドポイントとすることもできる。
昆虫および哺乳動物細胞において繁殖したデングウイルス血清型の中和。
上の実施例2に記載されている通り、フォーカス減少中和検査(FRNT)により、4種のデングウイルス血清型に対するヒト化抗体の中和活性を試験した。4種のデングウイルス血清型(DENV−1、DENV−2、DENV−3およびDENV−4)を昆虫細胞または哺乳動物細胞のいずれかにおいて繁殖させ、宿主Vero(サル)細胞の感染に使用した。図16〜図18は、代表的DENV−1〜4株によるこれらの実験の結果を示す。データは、相対感染力として表現する。例えば、EC50またはFRNT50値は、50%ウイルス中和の達成に必要とされる抗体の濃度を表す。図16〜図18に示す通り、被験抗体は、昆虫および哺乳動物細胞において繁殖したDENV−1、DENV−2、DENV−3およびDENV−4株の中和において有効である。
抗体の阻害活性を評価するためのデングウイルスの蚊モデル。
本明細書に記載されている抗体分子は、蚊モデルにおいて有効性を検査することができる。デングウイルスは、蚊に伝染されるRNAウイルスである。ある特定のデングウイルスは、in vivo適応度利点を生じることができ、これは、より高確率のヒトから蚊への伝染をもたらすことができる(Vuら、PLoS Negl Trop Dis.2010年;4巻(7号):e757頁)。デングウイルスに対する抗体の阻害活性を評価するための蚊モデルを確立するために、ウイルスを含有する血液を、様々な希釈の抗体と混合し、37℃で30分間インキュベートする。抗体をスパイクした血液を蚊の給餌器に加え、蚊に約1時間給餌する。蚊を寒冷麻酔し、充血した蚊を選択する。血液給餌7日後に、蚊の腹部を採取する。ウイルス負荷は、qRT−PCRによって測定することができる。腹部感染を有する蚊の比率は、PCRにより検査した腹部の総数で割った感染した腹部の数として計算することができる。
抗デング抗体のHP−SEC評価。
高速分子ふるいクロマトグラフィー(HP−SEC)を行って、ネイティブ非ストレス条件下における抗デング抗体の凝集傾向を評価した。この方法は、単量体からの抗体二量体および凝集体の識別を可能にする。二量体および凝集体は、免疫原性のリスク増加をもたらし得る。

- 抗ウイルス剤が、アルファ−グルコシダーゼI阻害剤、アデノシンヌクレオシド阻害剤、NS3および/またはその補助因子NS2Bの阻害剤、RNA依存性RNAポリメラーゼ(RdRp)阻害剤、宿主ピリミジン生合成の阻害剤、ウイルスNS4Bタンパク質の阻害剤またはイミノ糖のうち1種または複数から選択される、請求項50に記載の方法。
- 前記アルファ−グルコシダーゼI阻害剤が、セルゴシビルであり、前記アデノシンヌクレオシド阻害剤が、NITD008であり、前記NS3および/またはその補助因子NS2Bの阻害剤が、[5−アミノ−1−(フェニル)スルホニル−ピラゾール−3−イル]化合物であり、前記RNA依存性RNAポリメラーゼ(RdRp)阻害剤が、NITD107であり、前記宿主ピリミジン生合成の阻害剤が、NITD−982もしくはブレキナルから選択される宿主ジヒドロオロト酸デヒドロゲナーゼ(DHODH)の阻害剤であり、前記ウイルスNS4Bタンパク質の阻害剤が、NITD−618であり、または前記イミノ糖が、UV−4である、請求項54に記載の方法。
- ワクチンを前記対象に投与するステップをさらに含む、請求項50に記載の方法。
- 投与が、非経口的である、請求項50に記載の方法。
- 投与が、静脈内である、請求項50に記載の方法。

- 対象および蚊の間のデングウイルスの伝染を低下させる方法であって、デングウイルスの感染前または後に対象に、単離された請求項1〜39のいずれかに記載の抗体分子を、前記デングウイルスの伝染を低下させるのに有効な量で投与するステップを含む方法。
- 生物学的試料におけるデングウイルスを検出する方法であって、(i)前記試料または対象(および任意選択で、参照試料または対象)を、請求項1〜39のいずれかに記載の抗体分子に、前記抗体分子とポリペプチドとの相互作用が起こるのを可能にする条件下で接触させるステップと、(ii)前記抗体分子と、前記試料または前記対象(および任意選択で、前記参照試料または対象)との間の複合体の形成を検出するステップとを含む方法。
