本明細書に開示する一態様は、抗ZIKV抗体またはその抗原結合部分を提供する。総称して、新規抗体は、「抗ZIKV抗体」と本明細書で呼ばれる。「抗体」という用語は、明細書全体で使用される一方、抗体断片(すなわち、抗ZIKV抗体の抗原結合部分)もまた本開示に含まれ、明細書全体に記載される実施形態(方法及び組成物)に含まれ得ることに留意すべきである。ある特定の実施形態では、抗ZIKV抗体の抗原結合部分は、Fab、Fab’、F(ab’)2、Fv、ジスルフィド連結Fv、scFv、単一ドメイン抗体、またはダイアボディである。
A.ウサギモノクローナル抗ZIKV抗体の生成
モノクローナル抗体は、参照により本明細書に組み込まれるKohler and Milstein(1975)Nature 256:495によって記載されるものなどのハイブリドーマ法を用いて調製することができる。ハイブリドーマ方法を用いて、マウス、ハムスター、ウサギまたは他の適切な宿主動物を免疫付与し、ZIKVに特異的に結合する抗体のリンパ球による産生を誘発する。リンパ球はまた、インビトロにおいても免疫付与することができる。免疫付与後、リンパ球は、単離され、かつ、例えば、ポリエチレングリコールを用い、好適な骨髄腫細胞株と融合され、ハイブリドーマ細胞を形成し、これらは次に融合されないリンパ球と骨髄腫細胞から選別することができる。免疫沈降、免疫ブロット法、またはインビトロ結合アッセイ(例えばラジオイムノアッセイ(RIA);酵素結合免疫吸着アッセイ(ELISA))により決定されるように、ZIKVに対して特異的に向けられたモノクローナル抗体を産生するハイブリドーマは、次に、標準方法(参照により本明細書に組み込まれるGoding,Monoclonal Antibodies:Principles and Practice,Academic Press,1986)を使用するインビトロ培養において、または動物内の腹水腫瘍としてインビボにおいてのいずれかで増殖することができる。このモノクローナル抗体は次に、先のポリクローナル抗体について説明されたように、培養培地または腹水液から精製することができる。

重鎖、軽鎖、またはその断片をコードするDNAを単離するため、総RNAは、RNeasy(登録商標)キット(Qiagen,Hilden,Germany)を用いて、ハイブリドーマ細胞から単離される。一本鎖及び二本鎖cDNAの合成は、OneTaq(登録商標)One-Step RT-PCR キット(New England BioLabs,Ipswich,MA)を用いて行うことができる。PCR産物は、アガロース電気泳動によって分離され、断片を切り取って、QIAquick(登録商標)ゲル抽出キット(Qiagen,Hilden,Germany)によって精製することができる。断片は、ゴールデンゲートクローニング技術によって、BspQI(New England BioLabs,Ipswich,MA)との発現ベクターに直接クローニングしてもよい。各反応物から少なくとも4つのコロニーを、例えば、SequeMid(登録商標)DNA精製キット(Aline Biosciences,Woburn,MA)によって、ミニプレップスケールプラスミド精製のためにスケールアップする。各プラスミドにサンガーシーケンシングを施す。分析後、独自の組換え重鎖を独自の組換え軽鎖と対合させる。これらのプラスミド対を哺乳動物細胞にトランスフェクトする。8~12日後、各対合からの馴化培地を、ZIKVまたはZIKV Eタンパク質に結合するためのFLOW(商標)またはOctet(商標)によってスクリーニングする。
ヒト化抗体の遺伝子操作の方法も用いることができ、当該技術分野において周知である。ヒト化抗体は、非ヒト、例えば、限定されないが、マウス、ラット、ウサギ、非ヒト霊長類または他の哺乳動物である供給源由来の1つ以上のアミノ酸残基を有し得る。これらの非ヒトアミノ酸残基は、しばしば「インポート」残基と称される残基によって置き換えられ、インポート残基は、典型的には、既知のヒト配列の「インポート」可変、定常または他のドメインから取られる。
抗体は、宿主細胞からの発現によって産生してもよく、重鎖及び軽鎖をコードする発現ベクター(複数可)は、標準的技術によって、宿主細胞にトランスフェクトされる。「トランスフェクション」という用語の種々の形態は、原核または真核宿主細胞内への外因性DNAの導入のために一般に使用されている多種多様の技術、例えば、エレクトロポレーション、リン酸カルシウム沈殿、DEAE-デキストラントランスフェクションなどを包含することを意図する。原核または真核宿主細胞のいずれかにおいて抗体を発現することは可能であるが、真核細胞における抗体の発現が好ましく、哺乳動物宿主細胞における抗体の発現が最も好ましい。なぜなら、このような真核細胞(特に、哺乳動物細胞)が、適切に折り畳まれ、免疫学的に活性な抗体を構築し、分泌する可能性が原核細胞より高いからである。
本開示はまた、抗体またはその抗原結合部分、及び薬学的に許容される担体、希釈剤または賦形剤を含む、医薬組成物を提供する。抗体を含む医薬組成物は、これらに限定されないが、ZIKV感染によって引き起こされる容態またはその1つ以上の症状の防止、治療、管理、または寛解において使用される。特定の実施形態では、医薬組成物は、本発明による1つ以上の抗体またはその抗原結合部分を含む。別の実施形態では、医薬組成物は、1つ以上の本発明による抗体またはその抗原結合部分、及び有害なZIKVの感染を治療するための、抗体以外の1つ以上の予防または治療剤を含む。
本明細書に記載される単離された抗体及び抗原結合部分は、インビボ及びインビトロの両方でZIKVを中和することができるのが好ましい。したがって、そのような抗体及び抗原結合部分を使用して、例えば、ZIKVに暴露された細胞培養において、ZIKVに感染したヒト対象または他の哺乳動物対象において、ZIKVの感染を阻害することができる。
本発明による抗ZIKV抗体またはその抗原結合部分はまた、ZIKV感染の診断または検出などの非薬学的目的、または分析目的に使用することもできる。
本開示はまた、感染性ZIKVの中和、ZIKV感染の少なくとも1つの症状の予防、治療、または寛解、及び妊婦の胎児にZIKV感染を伝播させる尤度の低下、及び/または男性生殖器官への伝播の予防に使用されるキットを提供する。加えて、本開示はまた、ZIKV感染の診断にインビトロで使用されるキットを提供する。
動物及び免疫付与
合計2匹の(2)New Zealand White(NZW)雌ウサギ(TAK 466及びTAK 472)(Covance Research Products(Denver,PA)から全て入手した)は、それらの高い抗ZIKV ELISA抗体力価により選択した。動物の登録及び治療群の割り当ての詳細を表1に列挙する。
リンパ球単離
ウサギTAK466及びTAK472由来の脾臓をAbcamに送達し、Abcamのプロトコルに従って、最後の免疫付与の4日後に脾細胞を単離した。簡潔に言えば、脾臓を培養皿に移して、鉗子及びハサミを用いて過剰な脂肪をトリミングした。その後、脾臓に穿刺し、滅菌針及び注射器を用いて、培養培地(1:100希釈)(ペニシリン/ストレプトマイシン/ファンギゾンを有するRPMI培地、フィッシャーカタログ番号BW17745E)でフラッシュし、次いで、それを注射器の端で粉々に砕いた。その後、懸濁液を数回ピペットし、セルストレーナーを通過させて、濾液をファルコンチューブに収集した。細胞破片を同じ方法で3~5回処理し、ストレーナーを通過させて、全ての濾液を同じファルコンチューブに収集した。次いで、細胞懸濁液を1600rpmで15分間スピンし、上清を廃棄した。次に、赤血球溶血緩衝液を細胞ペレットに加え、4分間インキュベートした。培養培地をさらに加え、細胞を1600rpmでスピンした。最終的に、細胞ペレットを培地中に再懸濁し、細胞計数を行った。細胞を凍結用培地(90%FBS及び10%DMSO)にアリコートし、-80℃で一晩凍結した後、液体窒素保存に移した。脾臓及び単離された脾細胞の詳細を表3に記載する。
モノクローナル融合ごとに、2億個の脾細胞を1億個の融合パートナー細胞(240E-W2細胞、Epitomics/Abcamによって開発され、特許化された)と融合し、20枚の96ウェルプレートに播種した。TAK466及びTAK472脾細胞を用いて、4回の融合を翌日行い、得られたハイブリドーマを、合計で80枚の96ウェルプレートに播種した。
融合後、マルチクローン上清を、ZIKV-VLP(Native Antigen,Inc.)及びZIKV Eタンパク質(Native Antigen,Inc.)との反応性について標準ELISAによって、Abcamでスクリーニングした。ELISAスクリーニングごとに、抗原を50ng/ウェルでコーティングした。TAK466及びTAK472ウサギ出血(表1に示すように、PIZVで3回免疫付与した後にプールした過免疫血清)を、表5の対応するプレートについて、1:100希釈で陽性対照として使用した。細胞で調節されていない新鮮な培地を、80枚全てのプレートについて陰性対照として使用した。
ZIKV Eタンパク質及びZIKV-VLPへのハイブリドーマ結合の確認:
Abcamから受け取った全てのクローンを再試験し、間接ELISAによって、ZIKV Eタンパク質及びZIKV-VLPへの結合を確認した。
クローン102-1、242-3、289-3、306-2、11-3、78-2、270-12、181-4及び260-3に遺伝子シーケンシングを施した。アミノ酸及び核酸配列の両方を決定した。表6は、6つのクローンの抗ZIKV抗体の重鎖及び軽鎖、可変領域及びCDRのアミノ酸を記載する。抗ZIKV抗体の重鎖及び軽鎖及び可変領域に対応する核酸配列を表7に記載する。
・表7の各核酸配列の下線付きセグメントは、シグナルペプチドをコードするリーダー配列である。
デングとの交差反応性のためのハイブリドーマ上清のスクリーニング:
ハイブリドーマ上清に存在する抗体がZIKVに特異的であるかどうか決定するため、全てのマルチクローン及び/またはサブクローンを、不活化デングウイルス1(DENV1)[West Pacific 74(Microbix)];デングウイルス2(DENV2)[16681(Microbix)];デングウイルス3(DENV3)[CH53489(Microbix)];及びデングウイルス4(DENV4)[TVP-360(Microbix)]に対して、間接ELISAによってスクリーニングした。生産プロセスの一部として、製造者によって、DENV-1、3及び4は、ガンマ線照射で不活化し、DENV2は、ホルマリンで不活化した。ZIKV-VLP(天然抗原)及び組換えZIKV Eタンパク質(天然抗原)の両方は、陽性対照抗原として使用した。
TCID50に基づくマイクロ中和力価(MNT)アッセイを使用して、96ウェルプレート中のハイブリドーマ上清のウイルス中和抗体力価を検査した。TCID50は、50%組織培養感染量を表す。簡潔に言えば、連続希釈したハイブリドーマ上清を100 TCID50/ウェルのPRVABC59ジカ生ウイルスと混合した後、ベロ細胞の単層に加え、エンドポイント力価の細胞変性効果(CPE)の存在または不在について、プレートを感染5日後に観察した。
ZIKV-VLP及びZIKV Eタンパク質への結合について、上清を間接ELISAによって試験した。全てのクローンは、ZIKV-VLPへの良好な結合を示した(表10)。
ハイブリドーマ上清による精製ZIKV-VLP(天然抗原)の解離速度定数(koff)は、Octet 96デバイス(Pall ForteBio)を用いて、バイオレイヤー干渉法(BLI)によってモニタリングした。簡潔に言えば、ランニング緩衝液((0.1%のウシ血清アルブミン(BSA)、PBS 0.05% Tween 20(PBS-T))及びウサギIgGで5倍に希釈したハイブリドーマ上清を、プロテインAバイオセンサー(Pall ForteBio)によって捕捉した。バイオセンサーを、3.3μg/mLの会合用のZIKV-VLP溶液(10分)に移した後、解離用のランニング緩衝液に移した。各mAbのkoffは、Octet分析ソフトウェア(バージョン9.0 ForteBio)を用いて、1:1結合モデルによって計算した。
BLI技術によって、抗体ライブラリーを特徴付けて、抗体ライブラリーを、特異的抗原上の異なるエピトープに結合するビンに分類することができる。この実験では、競合アッセイを適用して、mAbをエピトープビンに分類した(図3)。簡潔に言えば、ZIKV-VLPをアミノプロピルシラン(APS)バイオセンサーにコーティングして、PBS中の1%BSAで遮断した。ビニング実行ごとに、表12から選択された一次抗体を結合させて、ZIKV-VLP結合部位を10分間飽和させた(水平矢印)後、二次抗体に10分間交差結合させた(垂直矢印)。各一次抗体に対する二次抗体の反応シグナル読み出しは、SAS JMP 13.1.0(SAS,Cay,NC,USA)を用いて、階層的クラスタリング(Ward Method)によってクラスタリングした。
選択されたトップハイブリドーマクローンは、ジカレポーターウイルス粒子(RVP)アッセイによって、それらの中和活性について試験した。このアッセイでは、ジカRVPを生ジカウイルスの代わりに使用して、中和抗体力価を決定した。ジカRVPは、カプシド、エンベロープ、プレ膜及び膜タンパク質CprMEを含む、野生型ビリオンの抗原決定基を保持する(Integral Molecular,Philadelphia,PA)。ジカRVPは、許容細胞が感染する際に、ウミシイタケルシフェラーゼレポーター遺伝子を発現する。抗体の最大半量有効濃度EC50力価は、ウミシイタケルシフェラーゼとセレンテラジン基質の相互作用によってグロータイプの発光シグナルを生成する生物発光反応を用いて決定した。発光シグナルは、発光対応プレートリーダーを用いて測定した。血清の存在下における発光シグナルの低下は、中和を示す。
ProteinSimple Wes(San Jose,CA)は、目的の抗体を用いて、プロセスサンプルから特定のタンパク質を検出することができるキャピラリーに基づく技術である。ハイブリドーマ上清のZIKV-VLPへの結合特異性を決定するため、選択された上清に、Wesシステムの製造者の指示を用いたWes分析を施した。
組換え抗体クローニング
選択されたサブクローン由来のハイブリドーマ細胞を収集し、溶解し、商業的に入手可能なポリ(A)+RNA単離キットを用いて、ポリ(A)+mRNAを単離した。その後、cDNAを、RNA産物からRT-PCRによって合成した。重鎖及び完全長カッパ軽鎖のウサギIgG可変領域は、遺伝子特異的プライマーを用いて、個々にPCR増幅した。PCR産物のゲル精製後、完全長カッパ軽鎖断片を哺乳動物軽鎖発現ベクターにクローニングした。重鎖では、可変断片を、標準的な分子生物学法を用いて、重鎖ベクターのウサギ重鎖定常領域にインフレーム融合した。重鎖及び軽鎖DNA哺乳動物発現プラスミドの両方を、サンガーシーケンシング及び抗体発現のために調製した。
組換えウサギモノクローナル抗体(RabMAb(登録商標))を発現するため、脂質仲介トランスフェクション試薬を用いて、軽鎖及び重鎖哺乳動物発現プラスミドを、指数関数的に増殖する293-6E細胞に同時トランスフェクトした。無血清培養上清は、遠心分離によって、トランスフェクションの5日後に収穫した。
収穫した培養培地を遠心分離して、細胞破片を除去し、分泌モノクローナル抗体を含有する透明な上清を、MabSelect SuReタンパク質Aカラムクロマトグラフィーを通じて精製した。溶出抗体をPBS緩衝液中で透析し、滅菌濾過し、pHを7.4に調整した。精製抗体を連続希釈し、ZIKV-VLP及び組換えZIKV Eタンパク質に対して、ELISAで試験した。また、濃度決定は、ヤギ抗ウサギIgGを用いて、OD405nmでのELISAによって行った。組換え抗体の純度及び完全性は、非還元及び還元条件下で、SDS-PAGEによって検証した(図12)。200μl及び約1mg/mL濃度のアリコートを調製した。組換えウサギモノクローナル抗体生成は、全てのクローンについておよそ3ヵ月で達成された。RabMAb(登録商標)開発の組換え段階には、クローニング、発現、並びにスケールアップ及び精製前の検証を含んだ。
ウェスタンブロット分析(Wesシステムを用いて)は、精製mAbに対して、ZIKV-VLP及びジカPRVABC59生ウイルスを用いて行った(図13)。289-3を除く全てのmAbは、ZIKV-VLP及び生ZIKVの両方のZIKV Eタンパク質に結合した。したがって、ELISAデータ(表16)に基づいて、289-3 mAbは、立体配座的にZIKV-VLPにのみ結合し、非常に強いMNT力価が得られることで、289-3 mAbは、ウェスタンブロット試薬または条件に感受性があり得るEタンパク質の2つ以上のドメインを伴う複合体エピトープに結合することが考えられる。この仮説を真に確認するため、本文書において以下に述べるように、このmAb上でエピトープマッピングを行った。
クローンのエピトープマッピングは、それらのアラニンスキャニング変異誘発ライブラリーを用いて、Integral Molecular,Inc.で実施した。抗体結合を担う重要なアミノ酸残基は、アラニンスキャニング変異誘発ライブラリーを用いて決定した。

(d)前記シグナルを検出すること、
(e)工程(b)で調製された前記複数の溶液の残りで工程(a)~(d)を繰り返すこと;及び
(f)前記二次抗体のシグナルによる希釈曲線を、工程(b)で調製された前記複数の溶液の各々の濃度に対してプロットすることであって、前記希釈曲線のIC 50 は、前記試験抗体の効力を示す、前記プロットすること、
を含む、前記方法。

試験PIZVの効力/濃度の決定方法であって、
(a)プレートのウェルを、試験PIZV上の第1のエピトープに結合して、前記試験PIZVを捕捉する第1の抗ZIKV抗体で被覆すること;
(b)前記被覆ウェルを前記試験PIZVでインキュベートすること;
(c)前記被覆ウェルを、前記試験PIVZ上の第2のエピトープに結合する第2の抗ZIKV抗体の複数の溶液のうちの1つでさらにインキュベートすることであって、前記第2の抗ZIKV抗体の前記複数の溶液は、前記第2の抗ZIKV抗体の濃縮溶液の連続希釈によって調製される、前記インキュベートすること、
(d)前記被覆ウェルを、酵素またはレポーターにコンジュゲートした二次抗体でさらにインキュベートすることであって、前記二次抗体の前記第2の抗ZIKV抗体への結合は、シグナルを生成する、前記インキュベートすること、
(e)前記シグナルを検出すること、
(f)工程(c)で調製された前記第2の抗ZIKV抗体の前記複数の溶液の残りで工程(a)~(e)を繰り返すこと、及び
(g)前記二次抗ZIKV抗体のシグナルによる希釈曲線を、工程(c)で調製された前記第2の抗ZIKV抗体の前記複数の溶液の各々の濃度に対してプロットすることであって、前記希釈曲線のIC 50 は、前記試験PIZVの効力を示し、
前記第1の抗ZIKV抗体及び前記第2の抗ZIKV抗体のうちの少なくとも1つは、項目1~9、19~21、25~27、31、33~44、57~68、73~76及び78のいずれか1項に記載の単離された抗体であり、前記第1のエピトープは、前記第2のエピトープと重複しない、前記プロットすること
を含む、前記方法。
Claims (18)
- ZIKVに結合する単離された抗体またはその抗原結合部分であって、配列番号5のアミノ酸配列を含むCDR3ドメイン、配列番号4のアミノ酸配列を含むCDR2ドメイン、及び配列番号3のアミノ酸配列を含むCDR1ドメインを含む重鎖可変領域、並びに配列番号10のアミノ酸配列を含むCDR3ドメイン、配列番号9のアミノ酸配列を含むCDR2ドメイン、及び配列番号8のアミノ酸配列を含むCDR1ドメインを含む軽鎖可変領域を含む、前記単離された抗体またはその抗原結合部分。
- ZIKVに結合する単離された抗体またはその抗原結合部分であって、配列番号15のアミノ酸配列を含むCDR3ドメイン、配列番号14のアミノ酸配列を含むCDR2ドメイン、及び配列番号13のアミノ酸配列を含むCDR1ドメインを含む重鎖可変領域、並びに配列番号20のアミノ酸配列を含むCDR3ドメイン、配列番号19のアミノ酸配列を含むCDR2ドメイン、及び配列番号18のアミノ酸配列を含むCDR1ドメインを含む軽鎖可変領域を含む、前記単離された抗体またはその抗原結合部分。
- ZIKVに結合する単離された抗体またはその抗原結合部分であって、配列番号35のアミノ酸配列を含むCDR3ドメイン、配列番号34のアミノ酸配列を含むCDR2ドメイン、及び配列番号33のアミノ酸配列を含むCDR1ドメインを含む重鎖可変領域、並びに配列番号40のアミノ酸配列を含むCDR3ドメイン、配列番号39のアミノ酸配列を含むCDR2ドメイン、及び配列番号38のアミノ酸配列を含むCDR1ドメインを含む軽鎖可変領域を含む、前記単離された抗体またはその抗原結合部分。
- ZIKVに結合する単離された抗体またはその抗原結合部分であって、配列番号45のアミノ酸配列を含むCDR3ドメイン、配列番号44のアミノ酸配列を含むCDR2ドメイン、及び配列番号43のアミノ酸配列を含むCDR1ドメインを含む重鎖可変領域、並びに配列番号50のアミノ酸配列を含むCDR3ドメイン、配列番号49のアミノ酸配列を含むCDR2ドメイン、及び配列番号48のアミノ酸配列を含むCDR1ドメインを含む軽鎖可変領域を含む、前記単離された抗体またはその抗原結合部分。
- ZIKVに結合する単離された抗体またはその抗原結合部分であって、配列番号89のアミノ酸配列を含むCDR3ドメイン、配列番号88のアミノ酸配列を含むCDR2ドメイン、及び配列番号87のアミノ酸配列を含むCDR1ドメインを含む重鎖可変領域、並びに配列番号94のアミノ酸配列を含むCDR3ドメイン、配列番号93のアミノ酸配列を含むCDR2ドメイン、及び配列番号92のアミノ酸配列を含むCDR1ドメインを含む軽鎖可変領域を含む、前記単離された抗体またはその抗原結合部分。
- 請求項1に記載の単離された抗体またはその抗原結合部分であって、配列番号2に記載のアミノ酸配列を含む重鎖可変領域、及び配列番号7に記載のアミノ酸配列を含む軽鎖可変領域を含む、前記単離された抗体またはその抗原結合部分。
- 請求項6に記載の単離された抗体またはその抗原結合部分であって、配列番号1に記載のアミノ酸配列を含む重鎖、及び配列番号6に記載のアミノ酸配列を含む軽鎖を含む、前記単離された抗体またはその抗原結合部分。
- 請求項2に記載の単離された抗体またはその抗原結合部分であって、配列番号12のアミノ酸配列を含む重鎖可変領域、及び配列番号17のアミノ酸配列を含む軽鎖可変領域を含む、前記単離された抗体またはその抗原結合部分。
- 請求項8に記載の単離された抗体またはその抗原結合部分であって、配列番号11に記載のアミノ酸配列を含む重鎖、及び配列番号16に記載のアミノ酸配列を含む軽鎖を含む、前記単離された抗体またはその抗原結合部分。
- 請求項3に記載の単離された抗体またはその抗原結合部分であって、配列番号32のアミノ酸配列を含む重鎖可変領域、及び配列番号37のアミノ酸配列を含む軽鎖可変領域を含む、前記単離された抗体またはその抗原結合部分。
- 請求項10に記載の単離された抗体またはその抗原結合部分であって、配列番号31に記載のアミノ酸配列を含む重鎖、及び配列番号36に記載のアミノ酸配列を含む軽鎖を含む、前記単離された抗体またはその抗原結合部分。
