進行性核上麻痺(PSP)は、脳の特定の容量の漸進的な悪化及び死を伴う変性疾患である。
大脳皮質基底核変性症(CBD)は、神経細胞の損失及び大脳皮質及び基底核を含む脳の複数の領野の萎縮(収縮)を特徴とする進行性神経障害である。症状としては、協調運動不全(poor coordination)、無動、硬直、平衡障害(disequilibrium)、四肢ジストニア、認知及び視覚空間障害、失行、ためらいがちな及びたどたどしい発話、ミオクローヌス、並びに嚥下障害が挙げられる。これらの症状は、パーキンソン病で見られる症状と非常によく似ている。現在、この疾患に対する利用可能な治療は存在せず、正確な原因は依然として不明である(Langの文献(2005)、進行性核上麻痺及び大脳皮質基底核変性症の治療(Treatment of progressive supranuclear palsy and corticobasal degeneration)、Movement Disorders 20: S83-S91)。
上で論じられているように、神経細胞の損失は、多くのCNS障害及び損傷、特に、神経変性疾患の症状の主な原因である。他方、神経及び軸索の成長を促進する多くの分子が存在するという事実にもかかわらず、成体哺乳動物のCNSは、損傷後に再生しない。対照的に、成体哺乳動物の末梢神経系(PNS)は、実際に、ある程度再生する。理論に束縛されることを望むものではないが、CNSにおける再生の欠如は、再生を積極的に妨害又は阻害する分子の存在によって生じると考えられる。PNSでは、ニューロンが再生することができる範囲内で、これらの阻害因子が除去されるか又は不活性であると考えられ、そのため、再生する軸索に遭遇することがない。したがって、十分に立証されている損傷後の成体哺乳動物のCNSの再生不能は、阻害分子の優位に起因すると考えられる。

下記の実施例において、CNS細胞の増幅及び機能に対するBRD7アンタゴニストの効果が評価されている。下に示されているように、BRD7アンタゴニストは、脳のサイズを拡大し、発生途中の脳及び脊髄の神経始原細胞を増加させる。BRD7アンタゴニストは、多能性遺伝子の発現も誘導する。
下記の実施例において、CNS細胞の増幅及び機能に対するIkarosアンタゴニストの効果が評価されている。下に示されているように、Ikarosアンタゴニストは、脳のサイズを拡大し、発生途中の脳及び脊髄の神経始原細胞を増加させる。Ikarosアンタゴニストは、多能性遺伝子の発現も誘導する。
下記の実施例において、CNS細胞の増幅及び機能に対するCRBN活性化因子の効果が評価されている。下に示されているように、CRBN活性化因子は、脳のサイズを拡大し、発生途中の脳及び脊髄の神経始原細胞を増加させる。CRBN活性化因子は、多能性遺伝子の発現も誘導する。
(6.1 実施例1−ゼブラフィッシュ胚における全身性の遺伝子過剰発現)
ゼブラフィッシュは、脊椎動物発生の解析のための重要な脊椎動物モデル及びヒト疾患のモデル系である。Barbazukらの文献、ゼブラフィッシュゲノムとヒトゲノムのシンテニー関係(The Syntenic Relationship of the Zebrafish and Human Genomes)、Genome Res., 10: 1351-1358(2000)を参照されたい。ヒトゲノムとゼブラフィッシュゲノムの間に高いシンテニー関係が存在し、ゲノムシンテニーの70〜80%がヒトとゼブラフィッシュの間で保存されている。Lieschke及びCurrieの文献、ヒト疾患の動物モデル:ゼブラフィッシュが泳いで姿を現す(Animal Models of Human Disease: Zebrafish Swim into View)、Nature Reviews Genetics., 8, 353-367(2007)を参照されたい。したがって、ゼブラフィッシュモデルは、発生生物学者の間で好んで選ばれるモデルとして長年にわたって使用されている。Fleischらの文献、CNSニューロンの再生及び機能的統合の研究:ゼブラフィッシュ遺伝学及び他の魚種からのレッスン(Investigating Regeneration and Functional Integration of CNS Neurons: Lessons from Zebrafish Genetics and Other Fish Species)、Biochim. 全身性に発現されたときに胚の非特異的な背方化を示すlhx2遺伝子を検討するために、及び脳の体積に対して及ぼされる効果を具体的に調べる正確な実験のために、胚の将来頭部になる領域へのインビボRNAリポフェクションを実施した。リポフェクションの方法は、Andoらによって開発及び公表された技法に完全に従うものであった。Ando及びOkamotoの文献、ゼブラフィッシュにおける遺伝子発現の空間的時間的制御のための効率的なトランスフェクション戦略(Efficient Transfection Strategy for the Spatiotemporal Control of Gene Expression in Zebrafish)、Biotechnol., 8(3): 295-303(2006)を参照されたい。
ノックダウンされる遺伝子のcDNA配列中の翻訳開始コドンの付近に対応する25-merのアンチセンスモルフォリノオリゴヌクレオチド(AMO、Gene Tools, LLC)の水溶液(700ng/μlの注射条件はRNAに対して用いられる条件と同じであった)を1細胞期の胚に注射した。CRBN遺伝子ノックダウンに用いられたAMOの配列は、
このプロセスは、以下の手順を除き、Andoらの技法に完全に従って実施された。Andoらの文献、Lhx2はゼブラフィッシュの前脳の成長におけるSix3の活性を媒介する(Lhx2 Mediates the Activity of Six3 in Zebrafish Forebrain Growth)、Dev. Biol., 287(2): 456-468(2005)を参照されたい。頭部特異的遺伝子発現は、RNAのケージ解除の代わりに、上の実施例2に記載のインビボリポフェクション法によって実施した。日本特許第2002-315576号、及びAndoらの文献、ゼブラフィッシュ胚におけるケージ化RNA/DNAを用いた光媒介遺伝子活性化(Photo-mediated Gene Activation Using Caged RNA/DNA in Zebrafish Embryos)、Nat. Genet., 28, 317-325(2001)を参照されたい。基本原理は、次の通りである:機能的関連を有することが示唆される2つの遺伝子のうちの1つ(A)が上の実施例3に記載の方法によってノックダウンされている胚を受精後6時間(原腸期)まで培養し、この胚の将来前脳になる領域で、もう1つの遺伝子(B)の発現を上の実施例2に記載のインビボリポフェクション法によって誘導する。受精後24時間以内に、Aをノックダウンした効果がレスキューされるのに対し、Bをノックダウンした効果がAとBの反対の組合せによってレスキューされない場合、Aは機能的にBの上流にあると決定される。
ゼブラフィッシュの初期ニューロンの抗体染色を以下の技法によって実施した。受精後24〜28時間の胚を、4%パラホルムアルデヒド/リン酸バッファー(pH 8.0)中で、4℃で12時間固定した。リン酸バッファーで4回洗浄した後(この場合、各洗浄は15分間行なう)、ブロッキングを、0.5%Triton X-100/リン酸バッファーに溶解した5%新生ヤギ血清中で、常温で1時間実施した。一次抗体反応を、同じ溶液に1:1000で希釈したモノクローナル抗アセチル化チューブリン抗体を用いて、4℃で12時間実施した。次いで、この胚をリン酸バッファーで同様に洗浄した後、一次抗体反応で適用された条件と同じ条件下でAlexa Fluoro 488(Molecular Probes)とコンジュゲートされた抗マウス抗体を用いて、二次抗体反応を実施した。その後、この胚をリン酸バッファーで洗浄し、30%/50%/70%グリセロール(リン酸バッファーに溶解したもの)で透明化し、その後、488nmで励起しながら、蛍光顕微鏡で観察した。
このプロセスは、基本的に、Thisseらの技法に従って実施した。ホールマウントゼブラフィッシュ胚に対する高分解能インサイチュハイブリダイゼーション(High-resolution In Situ Hybridization to Whole-mount Zebrafish Embryos)、Nat. Protcol., 3(1)59-69,(2008)を参照されたい。相違点は、5mg/mlのトルラ酵母RNAをプローブハイブリダイゼーション溶液中のブロッカーとして用いること、及び0.5%のブロッキング試薬(Roche)を抗体反応溶液中のブロッカーとして用いることであった。プローブとして用いられた遺伝子(six3.2遺伝子、emx1遺伝子、pax2.1遺伝子、foxg1遺伝子、及びotx2遺伝子)のcDNAは、原提供者からの寄贈品であった。lhx2遺伝子のプローブとしては、Andoによって最初にクローニングされたものを用いた。CRBN遺伝子及び関連遺伝子のプローブについては、プライマーをゼブラフィッシュのESTデータベースに基づいて設計し、プローブをcDNAライブラリーからクローニングした。
適当な濃度のローダミンデキストラン(分子量10,000)及び600ng/μlの最終濃度のCRBN RNAをヌクレアーゼ非含有水に溶解させ、その後、上の実施例1に記載の方法の条件下で、1細胞期のゼブラフィッシュ胚に注射した。得られた胚は、ドナーとしての役割を果たした。ドナーの受精から3〜4時間後、10〜50個の細胞を、蛍光顕微鏡観察下で、ガラスマイクロキャピラリーによって吸引した。これらの細胞を同じ段階の宿主胚の動物極に移植した。いかなる修飾も加えずに宿主胚を2日間培養した後、CRBNを発現するドナー由来細胞の分化の局在を蛍光顕微鏡下で観察した。また、陰性対照として、胚発生に影響がないと考えられる緑色蛍光タンパク質(GFP)を発現させた細胞を同じ条件下で移植し、分化の局在を実験胚と対照胚で比較した。
このプロセスは、基本的に、Groismanらの方法に従って実施した。Groismanらの文献、Cell, 113:357-367(2003);Groismanらの文献、Genes Dev., 20:1429-1434(2006)を参照されたい。まず、CRBN及び結合タンパク質(CRBN複合体と呼ぶ)を、M2 FLAGアガロースビーズ(SIGMA)を用いて、FLAGエピトープタグと融合したCRBNを発現する哺乳動物細胞の細胞ライセート溶液から精製した。その後、このように精製されたCRBN複合体を、Uba1(E1)、UbcH5b(E2)、及び組換えGST融合ユビキチン(Ub)タンパク質を含む水溶液と混合し、ATPを添加した後、混合物を30〜37℃で2時間放置しておいた。その後、反応をSDSによって終結させ、結合タンパク質の自己ユビキチン化及びユビキチン化をポリアクリルアミドゲル電気泳動及び免疫ブロッティングによって可視化し、それにより、ユビキチンリガーゼ活性を検出及び測定した。
このプロセスは、基本的に、Ohtakeらの方法に従って実施した。Ohtakeらの文献、Nature, 446:562-566(2007)を参照されたい。プロテアソーム阻害剤であるMG132を、FLAGエピトープタグと融合したCRBNを発現する哺乳動物細胞に適用し、細胞を静置しておいた。その後、これらの細胞を破壊し、得られた細胞ライセート溶液から、FLAGを精製し、CRBNを抽出した。その後、免疫ブロッティングを、上記と同様の、しかし、より厳しい条件下で実施し、それにより、自己ユビキチン化を検出及び測定した。
2日齢のゼブラフィッシュを用いた(グリア細胞染色及びセロトニン産生細胞染色のために、それぞれ、受精後56時間及び49時間のゼブラフィッシュを用いた)。ゼブラフィッシュを4%パラホルムアルデヒド(PFA)で固定し、その後、リン酸バッファー(PBS)で洗浄し、その後、表皮の部分消化のために、10μg/mlのプロテアーゼKで処理した。消化反応の後、ゼブラフィッシュをPBST(PBS+0.5%Triton X-100)で20分間洗浄し、その後、PFAで再び固定した。固定されたゼブラフィッシュを、PBSTで、室温で1時間洗浄し、その後、PBST+5%ヤギ血清を用いてブロッキングした。得られた標本をモノクローナル抗グリア抗体(zrf-1/zrf-2)又はウサギ抗セロトニン抗体と4℃で一晩(12〜18時間)反応させた。その後、標本をPBST+5%ヤギ血清で、室温で1時間洗浄し、抗体の代わりにヤギ抗マウスIgG抗体(グリア細胞染色用のCy-2コンジュゲート)又はヤギ抗ウサギIgG抗体(セロトニン産生細胞染色用のCy-5コンジュゲート)を4℃で一晩(12〜18時間)用いることにより、二次抗体反応を実施した。その後、標本を、PBSTで、室温で1時間洗浄した。洗浄後、PBSTを30%、50%、及び70%グリセロール/PBSに交換し、ゼブラフィッシュ全体をスライドガラス上で標本化して、プレパラートを得た。蛍光をCy-2及びCy-5の励起波長下で観察し、グリア細胞又はセロトニン産生細胞の分布を記録した。グリア細胞は、ゼブラフィッシュの側面から観察したが、セロトニン産生細胞は、正中線に沿った細胞の分布を観察するために、背側から撮影した。
CRBNをコードするRNA(700ng/μl)と2%ローダミンデキストランの混合溶液を、受精直後の1細胞期のゼブラフィッシュ胚に注射した。この胚を受精後4時間まで培養し、10〜20個の細胞を吸引キャピラリーによって回収し、これを受精後30時間の胚の間脳脳室に注射し、それにより、細胞を移植した。移植を受けたゼブラフィッシュを受精後3日までゼブラフィッシュ生理食塩水(E3リンガー)中で飼育し、その後、4%パラホルムアルデヒドで固定した。常法に従って、一次抗体反応及び二次抗体反応を、それぞれ、モノクローナル抗アセチル化チューブリン抗体及びAlexa Fluor(登録商標)(488nmで励起される)とコンジュゲートされた抗マウスIgG抗体を用いて実施した。その後、蛍光標識された神経細胞軸索を観察しながら、ローダミンデキストラン(543nmで励起される)で標識された移植細胞の分布を調べた。
正常胚(図1、左上のパネル)と比較して、lhx2遺伝子ノックダウン胚(図1、左下のパネル)で、脳の縮小が観察された。一方、CRBN遺伝子過剰発現胚(図1、右上のパネル)と同様に、lhx2遺伝子がノックダウンされている胚、及びCRBN遺伝子が過剰発現されている胚で、脳の拡大が観察された(図1、右下のパネル)。この観察から、CRBNがLhx2の下流で機能し、中枢神経幹細胞の増殖及びこれらの細胞の神経細胞への分化を直接誘導するものと考えられる。
大脳でのCRBNによる神経幹細胞への分化の誘導を確認するために、CRBN過剰発現実験をゼブラフィッシュ胚の前脳及び中脳で実施した。結果として、両方の脳は、その形状を保持しながら、体積が約1.5倍拡大した(図2、右)。その上、脳のニューロンネットワークは形態学的に正常であった(図2、右)。
まず、CRBNが過剰発現されるように、CRBNをコードするmRNA及び蛍光物質(ローダミン)をドナー胚に一緒に注射した。得られた胞胚を別の個体に移植し、脳室に分布したドナー由来細胞の分化を蛍光顕微鏡下で観察した。結果として、移植されたドナー由来細胞は、ゼブラフィッシュの終脳組織である嗅球へと顕著に分化した(図3、矢尻)。これは、CRBN発現細胞が脳室に分布すると、それらが神経幹細胞へと分化し、吻側移動経路(RMS)と呼ばれる細胞移動経路に沿って移動し、哺乳動物で大脳皮質が発生する背側終脳の脳組織に分化したことを示している。
CRBN(FH-CRBN複合体)を、Uba1(ユビキチン活性化酵素)、UbcH5b(ユビキチン転移酵素)、及び組換えGST融合ユビキチンタンパク質(Ub/E1/E2)を含む水溶液に添加したとき、CRBNが存在しなければ検出されないタンパク質が検出された(図4、左、中央、及び右の画像のレーン2)。タンパク質のユビキチン化のためには、ユビキチンの他に、3種類の酵素、すなわち、ユビキチン活性化酵素、ユビキチン転移酵素、及びユビキチンリガーゼが、通常必要とされる。ユビキチンリガーゼが存在しなかったので、ユビキチン化タンパク質は、左、中央、及び右の画像のレーン1で検出されなかった。一方、左、中央、及び右の画像のレーン2では、おそらくは、ユビキチン化タンパク質を産生するユビキチンリガーゼとしてのCRBNの機能のために、ユビキチン化タンパク質が電気泳動によって検出された。
MG132を添加したとき、CRBN含有タンパク質を表すいくつかのバンドが検出された(図5、右のレーン)。対照的に、MG132を添加しなかったとき、CRBN含有タンパク質を表すバンドはほとんど検出されなかった(図5、左のレーン)。
CRBN過剰発現個体(図6及び7、右の写真を参照)では、グリア細胞(図6参照)及びセロトニン産生細胞(図7参照)が増加する一方で、正常な空間的分布が維持された。これは、CRBNが増殖するように及び細胞を分化させるように作用する一方で、脳の空間パターンを正常に認識することを意味している。
CRBNを発現しない細胞を移植したとき、これらの細胞は脳組織に分化しなかったが(図8A〜8D)、CRBNを発現する細胞を移植したとき、これらの細胞は脳組織に分化した(図8E〜8H)。適当な濃度のローダミンデキストラン(分子量10,000)及び600ng/μlの最終濃度のCRBN RNAが溶解しているヌクレアーゼ非含有水を、上記の方法の条件下で、1細胞期のゼブラフィッシュ胚に注射し、得られた胚はドナーとしての役割を果たした。ドナーの受精から3〜4時間後、10〜50個の細胞を、蛍光顕微鏡観察下で、ガラスマイクロキャピラリーによって吸引した。これらの細胞を同じ段階の宿主胚の動物極にさらに移植した。いかなる修飾も加えずに宿主胚を2日間培養した後、CRBNを発現するドナー由来細胞の分化の局在を蛍光顕微鏡下で観察した。また、陰性対照として、胚発生に影響を及ぼさないと考えられる緑色蛍光タンパク質(GFP)を発現させた細胞を同じ条件下で移植し、分化の局在を実験胚と対照胚で比較した。
実施例19は、BRD7のノックダウンがゼブラフィッシュ胚の脳を拡大させることを示している。脳の成長に対するBRD7のアンタゴニストの効果を調べるために、BRD7をゼブラフィッシュ胚でノックダウンするか又は過剰発現させた。BRD7のノックダウンは、アンチセンスモルフォリノオリゴヌクレオチド(AMO)のマイクロインジェクションによって実施した。BRD7の過剰発現は、合成されたキャップ化mRNAのマイクロインジェクションによって実施した。窒素ガス圧マイクロインジェクターIM 300(Narishige)を用いて、AMO又はキャップ化mRNAを1細胞期胚に導入した。マイクロインジェクションの条件は次の通りである:ガス圧は約15ピコシーメンスであり;放出期間は、注射1回当たり約30〜約50ミリ秒であり;AMO及びキャップ化mRNAの濃度は、それぞれ、ヌクレアーゼ非含有水中、400ng/μl及び250ng/μlであった。キャップ化mRNAは、mMESSAGE mMachine(登録商標)インビトロ転写キット(Ambion)を用いて、pCS2+プラスミドにクローニングされたcDNAからインビトロで合成された。
ゼブラフィッシュ胚におけるPou5f1(Oct3/4)、Klf4、c-Myc、及びHuC(Elav13)の発現をインサイチュハイブリダイゼーションによって解析した。胚を、4%パラホルムアルデヒドで、室温で1時間固定し、リン酸緩衝生理食塩水(PBS)ですすいだ。固定された胚を100%メタノールで再固定し、-80℃で12時間以上置いた。この胚をPBSですすぐことにより脱固定し、皮膚をプロナーゼ(2mg/ml)で消化し、その後、再固定及び脱固定し、HYB*バッファー(50%ホルムアミド、5×SSC、10mMクエン酸、0.1%Tween 20)中で15分間、及びHYB+(5mg/mlトルラ(酵母)RNA、50μg/mlヘパリンとしてHYB*に添加されたもの)中で1時間、65℃でプレハイブリダイズさせた。1ng/μlのDIG(ジゴキシゲニン)標識プローブを添加した後、ハイブリダイゼーションを65℃で一晩実施した。ハイブリダイゼーション後、胚を、2×SSC中の50%ホルムアミドにより30分間(2回)、2×SSCにより15分間、0.2×SSCにより30分間(2回)、65℃で洗浄した。その後、ブロッキング処理を、PBSTw(PBS+0.1%Tween 20)中の0.5%精製カゼインにより、室温で1時間行った。その後、免疫染色を、PBSTw中の0.5%精製カゼイン中の抗DIG Fab-AP断片の1/4000希釈液中で、室温で4時間実施した。
BRD7ノックダウンゼブラフィッシュ胚は、CRBN過剰発現ゼブラフィッシュ胚と同様の表現型を示す。例えば、未処理ゼブラフィッシュ胚、CRBN過剰発現ゼブラフィッシュ胚、及びBRD7ノックダウンゼブラフィッシュ胚におけるHuCの発現を定量的PCRによって比較した。定量的PCRは次のように実施した: ReverTra Ace qPCR RT Kit(TOYOBO, Japan)によって(鋳型として)合成された1.7μgのランダムヘキサン及びオリゴ-dTプライムドcDNAをTHUNDERBIRD(商標) SYBR(登録商標) qPCR Mix(Toyobo, Japan)による逆転写に用いた。0.25mMの各々のフォワードプライマー及びリバースプライマーを反応に用い、その場合、フォワードプライマーの配列は
サリドマイドとBRD7の機能的関係を解析するために、サリドマイドで処理されたBRD7ノックダウンゼブラフィッシュの脳の発達を調べ、サリドマイドで処理された正常ゼブラフィッシュの脳の発達と比較した。図12に示されているように、BRD7ノックダウンゼブラフィッシュは、脳の発達中のサリドマイド処理に対して抵抗性である。図12A及び12Bは、400μMのサリドマイドで処理された正常ゼブラフィッシュを示している。図12C及び12Dは、400μMのサリドマイドで処理されたBRD7ノックダウンゼブラフィッシュを示している。図12A'及び12C'は、400μMのサリドマイドで処理された正常ゼブラフィッシュの脳領域を示している。図12B'及び12D'は、400μMのサリドマイドで処理されたBRD7ノックダウンゼブラフィッシュの脳領域を示している。示されているように、正常な魚は、400μMのサリドマイドの存在下で脳の発達の遅滞を示した(図12A、12B、12A'、及び12C')。対照的に、BRD7ノックダウン幼生の脳は、サリドマイド溶液中でほぼ正常に発達した(図12C、12D、12B'、及び12D')。サリドマイド処理あり又はなしでの、正常ゼブラフィッシュ及びBRD7ノックダウンゼブラフィッシュの体長に対する脳の厚さのパーセンテージも測定した。図12Eに示されているように、サリドマイド処理なしでは、BRD7ノックダウン幼生の脳のサイズは、通常のE3培地中の正常な幼生の脳のサイズの120.7%であり、BRD7ノックダウン幼生の脳のサイズは、400μMのサリドマイド中の正常な幼生の脳のサイズの140.0%である(n=12)。サリドマイド中でインキュベートされたBRD7ノックダウン幼生の脳のサイズは、サリドマイドの非存在下でインキュベートされたBRD7ノックダウン幼生の脳のサイズの98.8%である。
11hpfの未処理ゼブラフィッシュ胚、CRBN過剰発現ゼブラフィッシュ胚、CRBNノックダウンゼブラフィッシュ胚、BRD7過剰発現ゼブラフィッシュ胚、BRD7ノックダウンゼブラフィッシュ胚、及びサリドマイド処理ゼブラフィッシュ胚におけるPou5f1、Nanog、Zic3、及びSox2の発現を定量的PCRによって解析した。図13に示されているように、Pou5f1の発現は、11hpfのCRBN過剰発現及びBRD7ノックダウン胚で有意に増加した。Zic3(Pou5f1標的遺伝子)及び別の多能性遺伝子Nanogの発現もCRBN過剰発現及びBRD7ノックダウン胚で増加した。対照的に、Pou5f1、Zic3、及びNanogの発現は、同じ段階のCRBNノックダウン胚、BRD7過剰発現胚、及びサリドマイド処理胚で減少した。
Ascl1、Brn2(Pou3f2)、及びMyt1lという3つの遺伝子のプールの発現は、胚性線維芽細胞及び出生後線維芽細胞を機能的ニューロンへと直接的に再プログラミングするのに十分である。例えば、Vierbuchen及びWernigの文献、Nat Biotechnol. 29:892-907(2011)を参照されたい。そこで、Ascl1、Pou3f3a、Pou3f2b、及びMyt1laの発現を、定量的PCRにより、未処理ゼブラフィッシュ胚及びBRD7ノックダウンゼブラフィッシュ胚で解析した。図14に示されているように、BRD7のノックダウンは、28hpfでAscl1、Pou3f3a、及びMyt1laの発現を増加させ、BRD7のノックダウンは、Ascl1、Myt1l、及びBrn2(Pou3f2b)の発現を増加させた。
実施例25は、Ikarosのノックダウンがゼブラフィッシュ胚の脳を拡大させることを示した。脳の成長に対するIkarosのアンタゴニストの効果を調べるために、Ikarosをゼブラフィッシュ胚でノックダウンするか又は過剰発現させた。Ikarosのノックダウンは、アンチセンスモルフォリノオリゴヌクレオチド(AMO)のマイクロインジェクションによって実施した。Ikarosの過剰発現は、合成されたキャップ化mRNAの注射によって実施した。AMO又はキャップ化mRNAを、窒素ガス圧マイクロインジェクターIM 300(Narishige)を用いることによって、1細胞期胚に導入した。マイクロインジェクションの条件は次の通りである:ガス圧は約15ピコシーメンスであり;放出期間は、注射1回当たり約30〜約50ミリ秒であり;AMO及びキャップ化mRNAの濃度は、それぞれ、ヌクレアーゼ非含有水中、400ng/μl及び250ng/μlであった。キャップ化mRNAは、mMESSAGE mMachine(登録商標)インビトロ転写キット(Ambion)を用いて、pCS2+プラスミドにクローニングされたcDNAからインビトロで合成された。
ゼブラフィッシュ胚の脳の発達に対する2つのIkaros変異体の過剰発現の効果を解析した。2つの新しいIkaros変異体ikzf1-006及びikzf1-007を単離した。

CRBN過剰発現ゼブラフィッシュ胚及びIkarosノックダウンゼブラフィッシュ胚におけるAscl1a及びPou3f2bの発現をインサイチュハイブリダイゼーションによって解析した。胚を、4%パラホルムアルデヒドで、室温で1時間固定し、リン酸緩衝生理食塩水(PBS)ですすいだ。固定された胚を100%メタノールで再固定し、-80℃で12時間以上置いた。この胚をPBSですすぐことにより脱固定し、皮膚をプロナーゼ(2mg/ml)で消化し、その後、再固定及び脱固定し、HYB*バッファー(50%ホルムアミド、5×SSC、10mMクエン酸、0.1%Tween 20)中で15分間、及びHYB+(5mg/mlトルラ(酵母)RNA、50μg/mlヘパリンとしてHYB*に添加されたもの)中で1時間、65℃でプレハイブリダイズさせた。1ng/μlのDIG(ジゴキシゲニン)標識プローブを添加した後、ハイブリダイゼーションを65℃で一晩実施した。ハイブリダイゼーション後、胚を、2×SSC中の50%ホルムアミドにより30分間(2回)、2×SSCにより15分間、0.2×SSCにより30分間(2回)、65℃で洗浄した。その後、ブロッキング処理を、PBSTw(PBS+0.1%Tween 20)中の0.5%精製カゼインにより、室温で1時間行った。その後、免疫染色を、PBSTw中の0.5%精製カゼイン中の抗DIG Fab-AP断片の1/4000希釈液中で、室温で4時間実施した。
CRBNとIkarosの遺伝的関係性を調べるために、Ikaros発現を、インサイチュハイブリダイゼーションにより、24hpfのCRBN過剰発現胚で解析した。図23A及び23Bは、24hpfの未処理ゼブラフィッシュ胚及びCRBN過剰発現ゼブラフィッシュ胚におけるインサイチュハイブリダイゼーションによるIkaros発現を示している。図23Cは、24hpfの未処理ゼブラフィッシュ胚及びCRBN過剰発現ゼブラフィッシュ胚における定量的PCRによるIkaros、Tp53、及びTp63発現を示している。示されているように、Ikarosの発現は、主に、未処理胚の中枢神経系及び中間細胞塊ICMで観察された(図23Aの紫色のシグナル)。対照的に、Ikarosの発現は、CRBN過剰発現胚で有意に減少した(図23B参照)。定量的PCRにより、同じ段階のCRBN過剰発現ゼブラフィッシュ胚におけるIkaros発現の激しい減少が確認された(図23C参照)。

Claims (96)
- 患者の中枢神経系(CNS)細胞の疾患、障害、もしくは疾病、又はこれらの症状を治療又は予防する方法であって、該患者に、(i)ブロモドメイン含有7(BRD7)アンタゴニスト、(ii)Ikarosファミリージンクフィンガータンパク質1(Ikaros)アンタゴニスト、又は(iii)セレブロン(CRBN)活性化因子の有効量を投与し;それにより、該患者のCNS細胞欠損性疾患、障害、もしくは疾病、又はこれらの症状を治療することを含む、前記方法。
- 前記量が、前記患者のCNS細胞又はそのCNS始原細胞/前駆細胞の集団を増幅させ、それにより、前記CNS細胞欠損性疾患、障害、もしくは疾病、又はこれらの症状を治療又は予防するのに有効である、請求項1記載の方法。

- 前記CNS組織を前記BRD7アンタゴニスト、Ikarosアンタゴニスト、又はCRBN活性化因子と接触させることをさらに含む、請求項39記載の方法。
- 対象のCNS組織内のCNS細胞又はそのCNS始原細胞/前駆細胞の増殖を改善する方法であって、CNS細胞又はそのCNS始原細胞/前駆細胞の集団をBRD7アンタゴニスト、Ikarosアンタゴニスト、又はCRBN活性化因子の有効量と接触させること、及び該CNS組織を該細胞の集団と接触させることを含み、その結果、該細胞の増殖が向上する、前記方法。
- 対象のCNS組織内のCNS細胞塊を増加させる方法であって、CNS細胞又はそのCNS始原細胞/前駆細胞の集団をBRD7アンタゴニスト、Ikarosアンタゴニスト、又はCRBN活性化因子の有効量と接触させること(ここで、該CNS始原細胞/前駆細胞を任意にCNS細胞へとさらに分化させる);及び該CNS組織を該CNS細胞の集団と接触させることを含み、その結果、CNS細胞塊が増加する、前記方法。
- 前記CNS細胞又はそのCNS始原細胞/前駆細胞が、幹細胞、神経幹細胞、神経始原細胞、神経前駆細胞、神経細胞、星状細胞、グリア細胞、放射状グリア、及び希突起神経膠細胞からなる群から選択される、請求項32〜42のいずれか一項記載の方法。
- 前記対象がヒトである、請求項33及び37〜42のいずれか一項記載の方法。
- 前記対象がそれを必要としている患者である、請求項33及び37〜42のいずれか一項記載の方法。
- 前記BRD7アンタゴニストが、BRD7タンパク質の阻害因子、BRD7遺伝子の核酸配列の少なくとも一部を含む核酸、及びBRD7が下方調節されている幹細胞、神経始原細胞、又は神経前駆細胞からなる群から選択される、請求項1〜45のいずれか一項記載の方法。
- 前記BRD7アンタゴニストが、BRD7タンパク質の阻害因子である、請求項46記載の方法。
- 前記BRD7タンパク質の阻害因子が、BRD7産生の阻害因子、BRD7作用の阻害因子、及びBRD7の阻害因子のコード領域を含む核酸からなる群から選択される、請求項47記載の方法。
- 前記BRD7アンタゴニストがBRD7遺伝子の核酸配列の少なくとも一部を含む核酸である、請求項46記載の方法。

- 前記CRBN活性化因子が神経細胞への分化を誘導する、請求項72記載の方法。
- (a)CNS始原細胞/前駆細胞のCNS細胞への分化を誘導し、(b)CNS細胞又はCNS始原細胞/前駆細胞の増殖を改善し、又は(c)CNS細胞又はCNS始原細胞/前駆細胞の生存を改善する方法であって、該細胞を(i)BRD7アンタゴニスト、(ii)Ikarosアンタゴニスト、又は(iii)CRBN活性化因子と接触させることを含む、前記方法。
- CNS細胞又はCNS始原細胞/前駆細胞が、幹細胞、神経幹細胞、神経始原細胞、神経前駆細胞、神経細胞、星状細胞、グリア細胞、放射状グリア、及び希突起神経膠細胞からなる群から選択される、請求項84記載の方法。
- (a)患者の中枢神経系(CNS)細胞の疾患、障害、もしくは疾病、又はこれらの症状を治療又は予防する方法;
(b)患者の損傷したCNS組織を治療する方法;
(c)対象のCNS組織にCNS細胞又はそのCNS始原細胞/前駆細胞を生着させる方法;
(d)対象のCNS組織内のCNS細胞又はそのCNS始原細胞/前駆細胞の増殖を改善する方法;或いは
(e)対象のCNS組織内のCNS細胞塊を増加させる方法
における使用のための、(i)BRD7アンタゴニスト、(ii)Ikarosアンタゴニスト、及び/又は(iii)CRBN活性化因子を含む組成物。 - 請求項86記載の使用のための組成物であって、前記方法が:
(a)CNS細胞又はそのCNS始原細胞/前駆細胞の集団をドナー(好ましくは、該ドナーは前記患者である)から採取すること;
(b)該CNS細胞又はそのCNS始原細胞/前駆細胞の集団を培養すること;
(c)該CNS細胞又はそのCNS始原細胞/前駆細胞の集団を該組成物と接触させ、それにより、該CNS細胞又はそのCNS始原細胞/前駆細胞の集団を増幅させること;及び
(d)増幅させた集団のCNS細胞又はそのCNS始原細胞/前駆細胞を該患者に移植し、それにより、該患者の該CNS細胞欠損性疾患、障害、もしくは疾病、又はこれらの症状を治療すること
を含む、前記組成物。 - 前記方法が、前記組成物を前記患者に投与することをさらに含む、請求項87記載の使用のための組成物。
- 前記方法が、
(i)前記患者におけるCNS細胞塊を前記組成物の投与の前後で比較すること(ここで、該組成物の投与前と比較したときの該組成物の投与後のCNS細胞塊の増加は、該CNS細胞欠損性疾患、障害、もしくは疾病、又はこれらの症状を予防もしくは治療する際の該組成物の効力を示す);或いは
(ii)該患者における山中因子、Nanog、NeuroD、Zic3、Elav13、及び/又はBAM因子の発現レベルを該組成物の投与の前後で比較すること(ここで、該組成物の投与前と比較したときの該組成物の投与後の該山中因子、Nanog、NeuroD、Zic3、Elav13、及び/又はBAM因子の発現レベルの増加は、該CNS細胞欠損性疾患、障害、もしくは疾病、又はこれらの症状を予防もしくは治療する際の該組成物の効力を示す)
によって、該組成物の効力を評価することをさらに含む、請求項86〜88のいずれか一項記載の使用のための組成物。 - 前記方法が、前記組成物の投与に対する臨床応答を予測し、該投与に対する臨床応答をモニタリングし、又は該投与に対する患者コンプライアンスをモニタリングするために、CNS細胞欠損性疾患、障害、もしくは疾病、又はこれらの症状を有する患者の集団を、該患者における(i)CNS細胞塊、又は(ii)山中因子、Nanog、NeuroD、Zic3、Elav13、及び/もしくはBAM因子の発現レベルに基づいて選択することをさらに含む、請求項86〜89のいずれか一項記載の使用のための組成物。
- 前記山中因子が、Oct 3/4、Sox2、c-Myc、もしくはKlf4であり、及び/又は前記BAM因子が、Brn2(Pou3f2)、Ascl1、もしくはMyt11である、請求項89又は90記載の使用のための組成物。
- 前記CNS細胞欠損性疾患、障害、又は疾病が、大脳皮質の疾患、大脳皮質の外科的損傷、又は神経変性疾患である、請求項86〜91のいずれか一項記載の使用のための組成物。
- 前記CNS細胞の疾患、障害、又は疾病が、パーキンソン病;アルツハイマー病;クロイツフェルト・ヤコブ病;大脳皮質基底核変性症;筋萎縮性側索硬化症;多発性硬化症;進行性運動衰弱;免疫性神経障害、CNS外傷;パーキンソニズムを伴うアルツハイマー病;運動緩慢;無動;細かい運動制御及び手指の器用さを損なう運動障害;発声不全;単調な発話;硬直;ジストニア;パーキンソン病と関連する炎症;顔、顎、舌、姿勢の振戦;パーキンソン病様歩行;すり足歩行;小刻み歩行;加速歩行;気分、認知、感覚、睡眠の障害;認知症;鬱病;薬物誘発性パーキンソニズム;血管性パーキンソニズム;多系統萎縮症;進行性核上麻痺;一次性タウ病変を有する障害;皮質基底核神経節変性症;認知症を伴うパーキンソニズム;多動性障害;舞踏病;ハンチントン病;ジストニア;ウィルソン病;トゥレット症候群;本態性振戦;ミオクローヌス;及び遅発性運動障害からなる群から選択される、請求項86〜92のいずれか一項記載の使用のための組成物。
- 前記BRD7アンタゴニストが、BRD7タンパク質の阻害因子、BRD7遺伝子の核酸配列の少なくとも一部を含む核酸、及びBRD7が下方調節されている幹細胞、神経始原細胞、又は神経前駆細胞からなる群から選択され、好ましくは、
該BRD7タンパク質の阻害因子が、BRD7産生の阻害因子、BRD7作用の阻害因子、及びBRD7の阻害因子のコード領域を含む核酸からなる群から選択され;並びに/又は
該核酸が、BRD7遺伝子に特異的なアンチセンス分子、RNAi分子、もしくはBRD7遺伝子に特異的なモルフォリノオリゴヌクレオチドである、
請求項84もしくは請求項85記載の方法、又は請求項86〜93のいずれか一項記載の使用のための組成物。 - 前記Ikarosのアンタゴニストが、Ikarosタンパク質の阻害因子、Ikaros遺伝子の核酸配列の少なくとも一部を含む核酸、及びIkarosが下方調節されている幹細胞、神経始原細胞、又は神経前駆細胞からなる群から選択され、好ましくは、
該Ikarosタンパク質の阻害因子が、Ikaros産生の阻害因子、Ikaros作用の阻害因子、及びIkarosの阻害因子のコード領域を含む核酸からなる群から選択され;並びに/又は
