はじめに
補体系とは、異種細胞ならびにアポトーシスおよび酸化損傷の産物のような潜在的に危険な物質を認識し、タグづけし、除去するため、それ自体で、および他の免疫成分と協働する脊椎動物中の40〜50個の血液タンパク質のセットに与えられた名称である(1)。補体系を回避するのに細菌が用いる戦略は、新規抗生物質およびワクチンの潜在的標的であり、補体系の治療的調節は広範囲の臨床状況において積極的に研究されている(2)。
タンパク質の用意
プール血漿から精製された、ヒト補体タンパク質は、Complement Technologies Inc.(テキサス州タイラー)から入手し、供給業者の指示に従って保存し、希釈する前に新しく解凍し、さらなる精製なく使用した。組換えCFHモジュール切断変異体(「断片」、図1参照)のライブラリーはピキア・パストリスから調製され、以前記載された(35)ように特徴分析され、−80℃で保存された。
すべての表面プラズモン共鳴実験は、25℃、Biacore T200装置(GE Healthcare)上で、50μMのエチレンジアミン四酢酸(EDTA)を補充した、10mMのHEPES、150mMのNaCl、0.05%v/vの界面活性剤P20、pH7.4(HBS−P+)を用いて行った。Ni2+(51RU)およびその後(そのN末端hexaHis−SUMOタグを介して)sPspCN(144)をBiacoreNTAセンサーチップ上に固定した。(血漿または組換えP・パストリスからの)全長CFH、またはCFH断片の溶液のアリコートは示される濃度で180秒間、100μl/分でチップ上を通し、次に1500秒の解離フェーズを行った。CFHによる不可逆的結合のため、センサーチップは、1MのNaCl中の350mMのEDTAの30秒注入2回、その後の50mMのNaOH、次に0.5(w/v)%のドデシル硫酸ナトリウム(SDS)の30秒注入により、測定の間で再生した。データはBiacore評価ソフトウェアを用いて分析し、曲線をあてはめ、1:1結合モデルに基づき動的パラメータを推定した。
実験は、C3b(272RU);C3c(166RU);およびC3d(69RU)を、標準的なアミン結合を用い、Biacore C1センサーチップの3つの異なるフローセル上に固定した(第4のフローセルは背景差分に用いた)以外、上記のとおり行った。(2倍モル過剰のPspCN有りまたは無しでプレインキュベートした)因子Hを指示濃度でチップ上に注入(90秒、30μl/分)した後、400秒の解離フェーズを行った。センサーチップは1MのNaClの2回×30秒注入により再生した。データはBiacore評価ソフトウェアを用いて分析し、平衡親和性パラメータは1:1結合親和性モデルを用いて計算した。
CFHまたはCFH:PspCNの崩壊促進活性は、以前記載された(参考文献)SPRに基づくアッセイにおいて測定した。簡潔には、C3bの535RUを標準的なアミン結合によってBiacoreCM5センサーチップ上に固定した。その後、C3コンバターゼ(C3b.Bb)を、CFBおよびCFD(それぞれ500nM+50nM)の120秒、10μl/分での注入により、チップ上で組み立てた。C3b.Bb(〜145RU)の形成、その後の初期解離フェーズ(200秒)を(SPRにより)観察し、内因性C3コンバターゼ崩壊(Bbの離脱)の速度のモニタリングを可能にした。この後、CFHまたはCFH:PspCN複合体を指示濃度で注入(120秒、10μl/分)し、次にさらなる解離フェーズを行った。チップはサイクル間で、0.2μMのCFHの30秒注入、その後の1MのNaClの2回の30秒注入により再生した。
ヒツジおよびウサギ全血はTCS Biosciences(英国、バッキンガム)から購入し、CFH欠乏血清はComplement Technologyから購入した。溶血アッセイは96ウェルプレートにおいて、より従来的なベローナル緩衝液(37)の代わりにHEPESを用いた以外、Pangburn(36)により記載された方法に基づいて行った。5mlの全血からの赤血球を20mMのHEPES;145mMのNaCl;10mMのEDTA;0.1%(w/v)のゼラチン(ブタ皮膚、Fluka)、pH7.3(緩衝液H1)で3回、20mMのHEPES;145mMのNaCl;100μMのEDTA;0.1%(w/v)のゼラチン、pH7.3(緩衝液H2)でさらに3回洗浄した。CFH欠乏血清はCFH構成物で2μMまで再構成し、最終指示濃度で使用した。赤血球は、水で完全に溶解される場合、1(ウサギ)または1.2(ヒツジ)の最終A412をもたらす濃度で用いた。CFHで再構成された血清は次に氷上で、最終指示濃度まで、赤血球および緩衝液H2と組み合わせ、45μlの最終体積を得た。溶解は50mMのMg2+、50mMのエチレングリコール四酢酸、pH7.3の溶液5μlの添加により開始した。反応物は37℃で30分間インキュベートし、すぐに200μlの緩衝液H1の添加によりクエンチした。未溶解細胞をその後1500g、10分間、4℃での遠心分離によりペレット化し、次に100μlの浮遊物を除去し、A412を記録した。
タンパク質試料(〜0.5μM)を、20mMのリン酸カリウム、pH7.5中の1mlのHiTrap Heparinクロマトグラフィーカラム(GE Healthcare)に適用し、1MのNaClの直線勾配で20のカラム体積にわたり溶出した。
NMRスペクトルは5mmのTCI CryoProbe(Bruker)を備えるAvance II 800MHz分光計(Bruker)上で得た。NMRデータはTopSpin3.1(Bruker)を用いて処理し、CCPNMR分析ソフトウェアスイートを用いて分析した[Vranken WF,Boucher W,Stevens TJ,Fogh RH,Pajon A,Llinas M,Ulrich EL,Markley JL,Ionides J,Laue ED.The CCPN data model for NMR spectroscopy:development of a software pipeline.Proteins.2005 Jun 1;59(4):687−96]。15N標識FH8〜9および15N標識PspCNの試料を用い、NaCl濃度を0〜150mM、pHを4〜6.5、および温度を15℃〜70℃と変動させることにより条件を最適化した。その後、20mMのリン酸カリウム、pH6.2中の40μMの15N標識FH8〜9の試料を用い、1H、15N HSQCスペクトルを310Kで記録し、次にPspCNを100μMの最終濃度に付加し、スペクトルを記録した。別の実験において、20mMのリン酸カリウム、pH6.0中の30μMの15N標識PspCNの試料を用い、1H、15N HSQCスペクトルを298Kで記録し、次にFH8〜9を40μMの最終濃度に付加し、スペクトルを再度記録した。
血漿精製CFH(20μg)または血漿精製CFHおよびPspCN(20μgのCFH、対PspCN1:1.15のモル比)のいずれかの新しく解凍した試料を、ビス(スルホスクシニミジル)スベラート(BS3)で、タンパク質対BS3の1:4の質量比、および反応混合物中の最終[CFH]=0.56μg/μlを用いて架橋した。架橋生成物にドデシル硫酸ナトリウム−ポリアクリルアミドゲル電気泳動(SDS−PAGE)[(NuPAGE4〜12%、Bis−Trisゲル、MOPS泳動用緩衝液(Invitrogen)]を行った。ゲルバンドをInstantBlue(Expedeon)で染色し、5つの異なる架橋生成物に対応するバンド(図5A参照)を選択し、ポリアクリルアミドゲルから切断した。これらに記載されたようなゲル内トリプシン消化を行った[Chen ZA,Jawhari A,Fischer L,Buchen C,Tahir S,Kamenski T,Rasmussen M,Lariviere L,Bukowski−Wills JC,Nilges M,Cramer P,Rappsilber J.Architecture of the RNA polymerase II−TFIIF complex revealed by cross−linking and mass spectrometry.EMBO J.2010 Feb 17;29(4):717−26]。各試料について、架橋ペプチドはSCX−StageTipを用いて画分し[Ishihama Y,Rappsilber J,Mann M.Modular stop and go extraction tips with stacked disks for parallel and multidimensional Peptide fractionation in proteomics.J Proteome Res.2006 Apr;5(4):988−94];非架橋ペプチドは主に低塩(60mMの酢酸アンモニウム)で溶出すべきであるが、架橋ペプチドは高塩画分(500mMの酢酸アンモニウム)で濃度増加すべきである。高塩画分は、Q Exactive装置(Thermo Fisher Scientific)を用いる液体クロマトグラフィー−MS/MSを用いて分析した。ペプチドはC18材料(ReproSil−Pur C18−AQ 3μm;Dr Maisch GmbH、ドイツ)を充填した分析カラム上でPicoTipエミッタ(New Objective、マサチューセッツ州ウーバン)に分離した。直線勾配を1.6%(v/v)の水性アセトニトリルから32%(v/v)の水性アセトニトリルまで109分かけて適用した後、76%(v/v)のアセトニトリルまで急速に増加させた。質量分析データ取得はデータ依存設定で行った:各MS1スキャン後、10個のもっとも強いイオンを高エネルギーCトラップ解離により分離および断片化した。MS1およびMS2スペクトルの両方をオービトラップ分析計において、MS1スキャンは70000、MS2スキャンは35000の解像度で記録した。「動的排除」を60秒に設定し、「繰り返し回数」を1とした。
PspCNのN末端領域はCFHに不可逆的に結合する
S・ニューモニエ(D39株)からのPspCのN末端領域(図1)は以前示され(Dave S,et al.Indian J Med Res.2004 May;119 Suppl:66−73、およびHammerschmidt S,et al.The host immune regulator factor H interacts via two contact sites with the PspC protein of Streptococcus pneumoniae and mediates adhesion to host epithelial cells.J Immunol.2007 May 1;178(9):5848−58)、CFH結合部位を含有するが、その正確な境界は十分に定義されないままであった。我々は大腸菌において、PspC残基37〜140(sPspCN)および37〜292(sPspCNR1)(すなわちR1ドメイン、図1を含む)に対応する2つのN末端hexaHis−SUMOタグをつけたタンパク質を産生した。SPRを用い、我々はSUMO融合パートナー(データ記載せず)もR1ドメインの存在(データ記載せず)もCFHまたはCFH断片への結合には顕著に影響を及ぼさなかったことを見出した。SUMO自体はCFHに結合しなかった(データ記載せず)。我々は一連の濃度を用意し、SPRを用い、ニトリロ三酢酸(NTA)で予め固定し、Ni2+を充填したカルボキシメチル化デキストランでコーティングしたチップ上に固定したsPspCNについての、血漿精製CFHおよび(P・パストリスからの)組換えCFH(35)の親和性を測定した(図2、表1)。両方の例において、結合は測定するには遅すぎるオフレートおよびサブpM KDで事実上不可逆的であった。結合CFHは高塩洗浄により除去することができず;NTAチップをEDTA、NaOHおよびSDS溶液の連続処理で除去することによってのみ、sPspCNコーティング表面は測定間で再生することができた。それにもかかわらず、再生sPspCNコーティング表面は、2つの反応単位内で再生可能であった。
PspCは、CCP8〜11、13〜15および19〜20を含むCFHのさまざまな部位に結合すると以前報告された(Dave S,et al.2004 Mayand Hammerschmidt S,et al..2007;Duthy TG,et al.Infect Immun.2002 Oct;70(10):5604−11)。我々は、我々のライブラリー(15)からの関連組換えCFH断片を有するPspCN(図1)を用い、この問題について再考した。CFH6〜8(すなわち、CFH CCP6〜8に対応する組換え構成物)は、NTAチップ上のNi2+に、全長CFHまたはその他の試験断片より強く結合したことがわかった(図示せず)が、これはそれ以上調査されなかった。
架橋剤の非存在下、血漿精製CFH試料のSDS−PAGE後、予想どおり単一のバンドを検出した。同じCFH試料(0.56μg/μL)を3.9mMBS3と60分のインキュベーション後、ともに単量体CFHの候補である2つのSDS−PAGEバンドが現れた(図5A)。よりゆっくり移動するバンドはより多くのヘッド−テイル(例えばCCP20−CCP4およびCCP19−CCP20)架橋(図5B)を含有し、CFHの折り畳み構造の捕捉を反映した。もとの試料におけるこうした構造の相対存在量は、比較的希少である場合でも時間とともに蓄積するので、不明である。
市販の血漿精製ヒトC3b、C3cおよびC3dは、標準的なアミン結合を用い、同じC1 SPRチップ(Biacore)の異なるチャネルに固定した。その後、PspCNがC3b、C3cまたはC3dに結合しないことを示した(データ記載せず)ので、C3bおよびその各種分解生成物について、CFH、またはCFH:PspCN複合体の親和性を同じ実験で測定した(図6)。固定化C3bについて顕著に一致した親和性は、0.5〜0.6mMの文献KD値とよく一致する、組換えおよび血漿精製CFHについて得られた(表1)。興味深いことには、CFH:PspCN複合体はC3bに、KD=0.15〜0.2mMで、一貫して3倍良好に結合した。実質的に同じ結果をPspCNR1について得た(データ記載せず)。
PspCNがCFHに非常に密接に結合し、その構造を修飾する能力を発達させたとして、PspCN結合はC3b増幅の阻害剤としてCFHの機能的特性に対して何の効果を有するのかという疑問が出てくる。我々は、SPRチップ上でコンバターゼを組み立て、これがCFH(またはCFH:PspCN)付加前後でその成分に解離するのを観察することにより、崩壊促進活性に対するその効果を試験した(図7)。10nMのCFHは(予想どおり)崩壊に対して明らかな促進効果を有することがわかるが、これは2.5nMのCFHおよび2.5nMのPspCNの混合物により約2倍超となる。
我々はP・パストリスにおいて変異体CFH(D1119G)を産生した(図8A)。CCP19内のこのaHUS関連変異は、精製全長CFHの状況では、以前は完全には特徴分析されていなかった。他方でC末端の断片、CFH19〜20の状況では、D1119GはC3dおよびC3b結合のほぼ全損失を引き起こすことが報告された(39)。また、CFH19〜20:C3d複合体内で、D1119は分子間界面における主要な位置を占める(39)。興味深いことには、我々は、CFH(D1119D)がSPRチップ表面上でC3bに、CFH(wt)とほとんど同じ親和性で結合することを見出した(表2)。予想どおり、CFH(D1119G)はiC3bまたはC3dについて顕著な親和性を有さなかった。
CFHは補体系の作動における中心的な事象であるC3b増幅の制御に重要である。CFHは宿主細胞上および異種細胞上のC3b間で区別することが不可欠である。保護を必要とする表面でのCFHの選択的な濃度増加のみの問題である可能性は低く;これは微生物によるCFHの非常に容易なハイジャックを可能にし、補体系を感染に対する防御の最前線として実質的に無用にする。よってCFHはその補体調節部位のいくつかを「隠し」、それらを特定の分子マーカーの存在下でのみ明らかにするように進化し得た。我々は、こうした仮説戦略が、なぜCFHがリガンドに直接結合するとは考えられないこれほど多数のCCPモジュールを有するのかを説明することができると推量した。それらはCFHに異なる構造をとらせ、表面特異的な分子署名に応えて、異なるリガンド結合部位を露出することができた。ここで示される我々の結果は、PspCNおよびCFHの間で形成された複合体の研究に基づき、こうしたモデルを強く支持する。

PNH様赤血球の溶血に対する保護
PNH様ヒト赤血球はEzzell et al.(1991)の修正版に従って用意した。簡潔には、ヒト全血はドナーから静脈切開により入手し、アルセバー液中に4℃で保存した。細胞はRBC洗浄緩衝液(20mMのHEPES;145mMのNaCl;10mMのEDTA;0.1%w/vのゼラチン、pH7.3)で3回洗浄し、PBS中で最終洗浄後に再懸濁させた。各洗浄ステップ後、細胞ペレットの上部10%を吸引し、白血球含有バフィーコートを除去した。1mlアリコートの充填細胞を、HClを用いてpH8.0まで滴定した臭化2−アミノエチルイソチオウロニウム(AET、Sigma−Aldrich)溶液の8%w/v溶液に添加した。細胞懸濁液を9分間37℃で、一定に撹拌しながらインキュベートし、この時間後EDTAを含有しないRBC洗浄緩衝液で3回細胞を洗浄した。
PspC(52〜140)、PspC(62〜140)、PspC(72〜140)、PspC(37〜130)、PspC(37〜120)およびPspC(37〜110)のコード配列を含有する構成物を、PspCN(37〜140)配列を生成するのに用いた方法に従って生成した。次に、CFH結合を、PspCN(37〜140)タンパク質のCFH結合親和性を決定するように開発された同じSPRアッセイを用いて評価した。
PspCNのNおよびC末端を、システインおよび柔軟なペプチドリンカーセグメントを含有するように修飾した。N末端でCys修飾したPspCN(Cys−PspCN)の例では、含まれる追加の残基は:CGSGSGSGSGG−PspCN(配列番号7)であり、C末端でCys修飾したPspCN(PspCN−Cys)の例では、含まれる追加の残基は:PspCN−GSGSGSGSGGC*(配列番号8)であった。PspCNのこれらの修飾バージョンをクローニングし、PspCNのために開発されたものと同じ方法を用いて大腸菌においてSUMO融合タンパク質として発現させた。これらのタンパク質の精製も、システイン残基のチオール基を保存するための精製の全ステップ中にトリス(2−カルボキシエチル)ホスフィン(TCEP)を含んだ以外、PspCNに用いたものと同様であった。
His−SUMO−Cys−PspCNおよびHis−SUMO−PspCN−CysをSPR実験におけるリガンドとして、ならびに市販の血漿精製CFH(Complement Technology、テキサス州タイラー)を用い、sPspCNのCFH結合能力を評価するのに用いたものと同じ方法を用いてSPRを行った。
Cys−PspCNを20倍モル過剰のメトキシ−PEG−マレイミド(Sigma−Aldrich)と混合し、2.5時間室温で、1mMのTCEPを補充したPBS中で撹拌しながらインキュベートした。推定PEG化Cys−PspCNのアリコートに還元条件下でSDS−PAGEを行った。うまくPEG化したタンパク質は未処理のタンパク質に対するバンドシフトを示したものであると判断された。
正常ヒト血清中のCFHレベルを定量化するELISA型アッセイを標準的な平底96ウェルプレートにおいて行った。ウェルはsPspCN(100μl;8μg/ml)で一晩、4℃でコーティングし、その後5%w/vの乾燥無脂肪乳粉末を補充したリン酸緩衝生理食塩水(PBS)を用いてブロックした。ウェルは0.05%v/vのTween20を補充したPBSで3回洗浄した。ウェルは次に1時間室温で、一次抗体(OX24−Biotin(Lifespan Biosciences LS−C62878))の1:800希釈物とインキュベートした。ウェルは次に0.05%v/vのTween20を補充したPBSで3回洗浄した。ウェルは次に1時間室温で、ストレプトアビジン−ペルオキシダーゼ超感受性ポリマー(Sigma−Aldrich S−2438)の1:250希釈物とインキュベートした。ウェルは次に0.05%v/vのTween20を補充したPBSで3回洗浄した。ウェルを次に20分間、O−フェニレンジアミン二塩酸塩(OPD)基質とインキュベートした後、等体積の2.5MのH2SO4を添加した。ウェルの吸光度を490nmで読み取った。
PspCNは酸性化血清による「PNH様」赤血球の溶血を阻害することができる。図9AおよびBにおける溶血アッセイの結果は、CFHの添加により溶血を効果的に阻害することができることを示す。また、反応物へのPspCNの添加は溶血をさらにより効果的に阻害した(IC50値は28nMであると計算された)。この観察のもっとも可能性のある説明は、C3bおよびC3dへのCFHの結合のアッセイの結果と組み合わせた場合、PspCNの添加が血清中の内因性CFHを活性化するということである。
図10A〜Fにおいて示されるセンサーグラムは、C末端からのPspCN(残基37〜140)の切断が少なくとも1000倍の結合親和性の低下をもたらすが、N末端から少なくとも24個の残基を除去し、依然として非常に高いCFH結合親和性を保持することが可能であることを示す。他方で、PspC(71〜140)タンパク質は少なくとも1000倍の結合低下を示す。62〜140構成物もCFHを活性化構造で安定させる能力を保持することはまだ確認されていない。
図11において示される曲線の形状は、野生型PspCNの例と同様に、Cys−PspCN(図10A)およびPspCN−Cys(図10B)の両方がCFHに事実上不可逆的に結合する能力を有し、検出可能なオフレートを示さないことを示す。Biacore評価ソフトウェアを用いて行ったCys−PspCNおよびPspCN−Cysの相互作用の動的分析は、(相互作用はSPRを用いて正確なKDを計算するには強すぎたが)相互作用のKDが約10−13Mであることを示した。この計算されたKDは未修飾PspCNについて計算されたものと同じ桁数である。
メトキシ−PEG−マレイミドは、図12においてSDS−PAGE下での移動度シフトにより見ることができるように、穏やかな条件下でCys−PspCNに効果的に結合する。15kDaおよび20kDaマーカーの間を移動する下方バンドは未修飾PspCNを表す。PEG化反応後、依然として顕著な量のPspCNがPEG化されずに残る。25kDaマーカーのすぐ上を移動する上方バンドは、他方でうまくPEG化されたPspCNを表す。PspCNジスルフィド結合二量体、または複数のPEG部分を含むPspCN種の形成の証拠はほとんどない。全体的にこのPspCNのPEG化方法は、約50%収率のPEG化PspCNをもたらし;収率パーセントをより正確に決定するにはより多くの定量的研究が必要となるだろう。
既知の濃度で、血漿精製CFH(Complement Technology)を用いて標準曲線(図13)を構築し、これは少なくとも二桁にわたる良好な直線検出をもたらした。標準曲線と比較した場合、正常ヒト血清中のCFHレベルを決定することができ、現在許容される基準範囲内であることがわかった。
N末端のCys残基および残基GSGSGSGSGG(Cys−PspCN)からなるリンカーを含有するように修飾したPspCN(37〜140)を、Ultralink Iodoacetylクロマトグラフィー樹脂(Thermo Scientific)にそのN末端のCys残基を介して結合した。結合は製造業者の指示に基づいて行った。Ultralink Iodoacetyl樹脂(10ml)は、50mlの結合緩衝液(50mMのTris、5mMのEDTA、pH8.5)で、重力供給式クロマトグラフィーカラムを用いて洗浄した。2.5mMのTCEPを補充した結合緩衝液中のCys−PspCN(2mg/mlで20ml)の溶液を、重力供給式クロマトグラフィーカラムにおいてUltralink Iodoacetyl樹脂と15分間室温で混合した。カラムを次に直立させ、さらに30分間室温でインキュベートした。次に流体相を排出し、カラムを30mlの結合緩衝液で洗浄することにより、未結合Cys−PspCNを除去した。樹脂上の未反応ヨードアセチル基をブロックするため、樹脂を次に結合緩衝液中の50mMのL−システイン.HCl20mlと15分間室温で混合した。カラムを次に直立させ、さらに30分間室温でインキュベートした。樹脂は次に1MのNaCl60ml、その後0.05%のNaN3を補充したリン酸緩衝生理食塩水(PBS)60mlで洗浄した。この方法を用い、各1mlの樹脂に約1〜1.5mgのCys−PspCNを結合した。
アルセバー液中のヒト血漿(正常混合プール)をTCS Biosciencesから購入した。PspCN樹脂(5ml)を100mlのヒト血漿と4℃で1時間インキュベートした。インキュベーション後、混合物を空の重力供給式クロマトグラフィーカラムに適用し、血漿を排出した。樹脂を次に3×50mlのHBS(20mMのHEPES;150mMのNaCl;pH7.5)で洗浄し、その後2×50mlの4MのNaClでのさらなる洗浄を行った。高度に精製したFHをカラムから0.1Mのグリシン緩衝液、pH2.5を用いて溶出した。
図18を参照すると、システインタグをつけたPspCNを、製造業者の指示に従い、示されるような結合緩衝液中の異なる濃度のCys−PspCNを用いてマレイミド活性化ポリスチレン96ウェルプレート(Pierce)上に固定した。ウェルをその後新鮮な正常ヒト血清と室温でインキュベートし、補体沈着を引き起こした。表面へのC3bの沈着は従来のELISAプロトコルに類似した方法で抗C3抗体を用いて検出した。沈着したC3bは結合緩衝液中のPspCNの濃度の関数として測定した。N末端およびC末端でシステインタグをつけたPspCNの両方について、C3bの沈着は100μg/mlのPspCNをコーティングした表面上で約50%減少した。
図19を参照すると、Cys−PspCNを、製造業者の指示に従い、50μg/mlの結合緩衝液中のある濃度のPspCNを用いてマレイミド活性化96ウェルプレート(Pierce)上に固定した。プレートをその後精製ヒト因子Hとインキュベートした後、抗FH自己抗体について陽性であることが知られる患者または正常な対照に由来する精製IgGとインキュベートした。その後、ビオチンに結合した抗ヒトIgG特異的抗体、次にストレプトアビジンペルオキシダーゼを標準的なELISAに類似した方法で用い、抗FH自己抗体を検出した。その後のインキュベーション間でウェルはリン酸緩衝生理食塩水で3回洗浄した。
ここではTIGR4 PspCと称される、ストレプトコッカス・ニューモニエのTIGR4株のPspCタンパク質の断片は、CFHに強く結合することが示された。TIGR4 PspC(37〜179)はCFHに10−16MのKDで結合し、TIGR4 PspC(68〜148)はCFHに10−15MのKDで結合する。
図20を参照すると、S・ニューモニエのTIGR4株からのPspCNは、D39株からのPspCNを試験するのに用いたものと同一の方法で、Biacore NTAチップに結合した。残基37〜179または残基68〜148からなるTIGR4株からのPspCNの断片への結合を示すセンサーグラムを取得した。各例において、TIGR4由来のPspCN配列へのFHの結合は、各例で計算された10−16M台のKDでの、D39株について観察されたものと同様に強い結合をもたらした。

- 対象または補体活性の低減が有益である場合における異常な補体調節活性に関連する疾患(症状)または他の内科的合併症の治療または予防に有用な請求項1〜12のいずれか1項に記載のタンパク質。
- 前記疾患(症状)または他の内科的合併症が:
−発作性夜間ヘモグロビン尿症(PNH);
−非典型溶血性尿毒症症候群(aHUS);
−密沈積症(DDD);
−加齢性黄斑変性症(AMD);
−全身性紅斑性狼瘡(SLE);
−敗血症;および
−アルツハイマー病
の1つ以上である、請求項32に記載のタンパク質。 - 請求項1〜12のいずれか1項に記載のタンパク質またはこれをコードする核酸を含む医薬製剤。
- 医薬的に許容可能な担体をさらに含む、請求項34に記載の医薬組成物。
- 適切な賦形剤溶液を希釈剤として用い、凍結乾燥物として製剤される、請求項34または35に記載の医薬組成物。

- 請求項1〜12のいずれか1項に記載のタンパク質の前記医薬的に効果的な量が、タンパク質約1μg/対象の体重1kg〜タンパク質約500mg/対象の体重1kgであり得る、請求項45に記載の方法。
- 請求項1〜12のいずれか1項に記載のタンパク質の対象における特定の部位または組織への標的化を含む、請求項41〜45のいずれか1項に記載の方法。
