Abstract
Description
1)前記したような熱水抽出液、その濃縮液又はその凍結乾燥物をそのままふりかけとして、又はティーパックやカプセルの中に充填して使用する方法。
2)前記したような熱水抽出液又は減圧濃縮液に糖類、酸類、塩類及び香料類等を調合して使用する方法。
3)前記したような熱水抽出液、その減圧濃縮液又はその凍結乾燥物をベイク品、発酵品、練り製品、乳製品、油脂製品、調味料、菓子等の食品、又はコーヒー、ココア、茶、果実ジュース、野菜ジュース、発酵飲料、清涼飲料等の飲料の製造工程で添加して使用する方法。
実施例1
直井白芝の子実体の乾燥粉砕物300gに精製水3000mlを加え、緩やかに撹拌しながら、還流下に95℃で2時間熱水抽出した後、12000rpmで10分間遠心分離して熱水抽出液を得た。得られた熱水抽出液を40℃で減圧濃縮し、更に凍結乾燥して、水可溶性成分を得た(収率15.1%)。
直井白芝の種菌を、ブドウ糖、麦芽エキス及び寒天等を用いて調製したpH6.8の液体培地に接種し、ジャーファーヌンターを用い、28℃で14時間、通気培養した後、濾過して、培養菌糸体を得た(収率3.8g/L)。この培養菌糸体に5倍量の精製水を加え、緩やかに撹拌しながら、還流下に95℃で2時間熱水抽出した後、濾過して、熱水抽出液を得た。この熱水抽出液を加熱して7倍に濃縮し、冷却した後、等量のエタノールを加え、8000rpmで20分間遠心分離して、エタノール沈殿物を分離し、更に凍結乾燥して、水可溶性成分を得た(収率898mg/L)。
直井赤芝、直井黄芝及び直井白芝の各子実体の乾燥粉砕物を等量混合したもの300gに精製水300mlを加え、緩やかに撹拌しながら、還流下に95℃で2時間熱水抽出した後、12000rpmで10分間遠心分離して熱水抽出液を得た。得られた熱水抽出液を40℃で減圧濃縮し、更に凍結乾燥して、水可溶性成分を得た(収率13.5%)。
供試したSlc:Wistar雄ラット(日本エスエルシー社製)は、4週齢で購入し、7日間の予備飼育の後、一般症状観察及び尿酸検査で異常が認められなかったラットを試験に供した。試験開始時のラットの体重は約100gであった。ラットは温度23±2℃、相対湿度55±5%、12時間暗照光、換気回数20回/時、12時間照明(午前7時点灯、午後7時消灯)のバリアシステムの環境下の飼育条件下で、1群5匹とし、プラスチックケージに5匹ずつ同居させて飼育した。食餌性高尿酸ラットを作製するための飼料は、クレア固型飼料(CE−7)粉末に100%ビール酵母粉末(キリンヤクルトネクストステージ社製、蛋白質49.33%、核酸(RNA)2.33%、脂質4.42%、糖質2.17%、食物繊維4.18%、その他、アミノ酸、ビタミン、ミネラルを含有)を25%又は50%となるよう配合し、蛋白質及び核酸含量の異なった配合飼料とした。前記のように各群5匹ずつ3群(ビール酵母粉末0%、25%又は50%の3群)に分け、6日間飼育し、飼料と水は自由に摂取させた。
試験区分2と同様にして前記のCE−7粉末飼料に50%ビール酵母粉末を配合し飼育するラットに、試験区分1で分離した実施例1、実施例2及び参考例1の水可溶性成分を表3及び表4に記載の割合で6日間、強制的に胃ゾンデにより経口投与し、尿中尿酸値を測定すると共に血中尿酸値を測定した。測定は試験区分2と同様に行ない、結果を表3及び表4に示した。
実施例3
試験区分1の実施例2の場合と同様にして熱水抽出液を得た後、該熱水抽出液1kgに砂糖100g、蜂蜜15g、カラメル5g、アスコルビン酸0.75g、クエン酸0.3g及びレモン系香料0.2gを調合し、健康飲料を製造した。
試験区分1の実施例1の場合と同様にして熱水抽出液を得た後、該熱水抽出液を減圧濃縮し、凍結乾燥した凍結乾燥物10g及びアスコルビン酸0.5gをリンゴ搾汁液2kgに調合してリンゴジュースを製造した。
前記の実施例4の場合と同様にして凍結乾燥物を得た後、若干量の塩化カルシウム及び第三リン酸ナトリウムと共に該凍結乾燥物5gを、採肉し、水さらしして脱水した後、予冷した魚肉2kgにそのすりつぶし工程で添加し、凍結して冷凍すり味を製造した。

- 直井白芝の子実体、直井白芝の培養菌糸体及び/又はこれらの処理物から分離した水可溶性成分から成ることを特徴とする尿酸値上昇抑制剤。
- 水可溶性成分が、直井白芝の子実体、直井白芝の培養菌糸体及び/又はこれらの処理物を熱水で抽出処理し、その抽出液から得たものである請求項1記載の尿酸値上昇抑制剤。
- 水可溶性成分が、抽出液を更に凍結乾燥したものである請求項2記載の尿酸値上昇抑制剤。
- 直井白芝の子実体、直井白芝の培養菌糸体及び/又はこれらの処理物から分離した水可溶性成分を用いて成ることを特徴とする尿酸値上昇抑制作用を有する飲食品。
