Abstract
Description
圧縮成形することで得られる厚み5mm未満の口腔内崩壊性シート状製剤。
[態様2]
口腔内崩壊時間が20秒以下で、かつシート強度が0.04N以上である、態様1に記載の口腔内崩壊性シート状製剤。
[態様3]
賦形剤及び崩壊剤を含む、態様1又は2に記載の口腔内崩壊性シート状製剤。
[態様4]
崩壊剤としてクロスポビドン、クロスカルメロースナトリウム、低置換度ヒドロキシプロピルセルロース及びカルボキシメチルスターチから成る群から選択される1成分以上を含む、態様3に記載の口腔内崩壊性シート状製剤。
[態様5]
賦形剤としてD−マンニトール、ラクトース、トレハロース、セルロース、キシリトール及びリン酸カルシウムから成る群から選択される1成分以上を含む、態様3又は4に記載の口腔内崩壊性シート状製剤。
[態様6]
更に、結合剤を含む、態様3〜5のいずれか一項に記載の口腔内崩壊性シート状製剤。
[態様7]
更に、流動化剤を含む、態様3〜6のいずれか一項に記載の口腔内崩壊性シート状製剤。
[態様8]
態様1〜7のいずれか一項に記載の口腔内崩壊性シート状製剤の製造方法であって、該製剤に含まれる各種成分を混合した後、圧縮成形することを含む、前記製造方法。
・口腔内で素早く崩壊する。
・水なしで服用出来る。
・嚥下しやすい。
・服薬量を簡単に調整できる。
・患者のコンプライアンスが向上する。
・製造工程が単純である。
・製造場所(薬局などを含む)を問わず製造可能である。
・使用できるコーティング粒子の選択肢が広がる。
・液状で不安定な成分も使用することができる。
・口蓋に張り付かない。
・乾燥工程が不要である。
実施例および比較例で得た各錠剤について、以下の方法によってシート厚み、シート強度及び口腔内崩壊時間を測定した。これらの測定結果を表1に示す。
シート厚み:デジタルノギス(シンワ測定株式会社)を用いて、シート厚み(mm)を測定した。
シート強度:テクスチャーアナライザー(英弘精機株式会社)を用いて、シート強度(N)を測定した。強度の測定には三点曲げ冶具を使用し、支点間距離を16mmとして評価を実施した。
口腔内崩壊時間については、150mgのシート状錠剤を口腔内に含み、力を加えないように舌と上顎で挟んだ状態を保って、錠剤が完全に崩壊するまでの時間を測定した。成人男1名が2回ずつ実施し、それらの平均値を測定結果とした。
マンニトール(D−マンニトール、メルク株式会社)495g、ステアリン酸マグネシウム(太平化学産業株式会社)5gを混合し、配合物とした。この配合物を乾式造粒機(TF−LABO、フロイント産業株式会社)にてロール圧縮し、シート状製剤(厚み:1.26mm)を得た。
※口腔内崩壊時間は重量150mgを服用したときのものである
Claims (8)
- 圧縮成形することで得られる厚み5mm未満の口腔内崩壊性シート状製剤。
- 口腔内崩壊時間が20秒以下で、かつシート強度が0.04N以上である、態様1に記載の口腔内崩壊性シート状製剤。
- 賦形剤及び崩壊剤を含む、請求項1又は2に記載の口腔内崩壊性シート状製剤。
- 崩壊剤としてクロスポビドン、クロスカルメロースナトリウム、低置換度ヒドロキシプロピルセルロース及びカルボキシメチルスターチから成る群から選択される1成分以上を含む、請求項3に記載の口腔内崩壊性シート状製剤。
- 賦形剤としてD−マンニトール、ラクトース、トレハロース、セルロース、キシリトール及びリン酸カルシウムから成る群から選択される1成分以上を含む、請求項3又は4に記載の口腔内崩壊性シート状製剤。
- 更に、結合剤を含む、請求項3〜5のいずれか一項に記載の口腔内崩壊性シート状製剤。
- 更に、流動化剤を含む、請求項3〜6のいずれか一項に記載の口腔内崩壊性シート状製剤。
- 請求項1〜7のいずれか一項に記載の口腔内崩壊性シート状製剤の製造方法であって、該製剤に含まれる各種成分を混合した後、圧縮成形することを含む、前記製造方法。
