(a)配列番号:4〜6に記載のアミノ酸配列、配列番号:4〜6に記載のアミノ酸配列に対して80%以上の相同性を有するアミノ酸配列、配列番号:4〜6に記載のアミノ酸配列の少なくともいずれかにおいて、1若しくは数個のアミノ酸が置換、欠失、付加及び/又は挿入されたアミノ酸配列、配列番号:10〜12に記載のアミノ酸配列、配列番号:10〜12に記載のアミノ酸配列に対して80%以上の相同性を有するアミノ酸配列、並びに配列番号:10〜12に記載のアミノ酸配列の少なくともいずれかにおいて、1若しくは数個のアミノ酸が置換、欠失、付加及び/又は挿入されたアミノ酸配列からなる群から選択される少なくとも1のアミノ酸配列を、CDRとして含む可変領域を保持する

後述の実施例において示す通り、本発明者らは、ヒト由来のEva1タンパク質に対して高い親和性を示す、3種のマウスモノクローナル抗体(B2E5−48抗体、C3抗体、A5D11−10抗体)を得ることに成功した。さらに、かかる抗体が高いADCC活性及び/又はCDC活性を有することも見出した。また、メラノーマ細胞を投与したマウスに、前記抗体を投与することによって、当該細胞の肺への転移等が抑制されることも明らかにした。さらに、これら抗体の軽鎖可変領域及び重鎖可変領域の各々の相補性決定領域(CDR)1〜3の配列も決定した。

本発明の抗体は、後述の実施例において示す通り、ヒト由来のEva1タンパク質に対して高い親和性を示し、またADCC活性及び/又はCDC活性等を発揮することから、Eva1タンパク質に関連する疾患の治療又は予防に利用することができる。したがって、本発明は、本発明の抗体を有効成分とする医薬組成物(例えば、本発明の抗体を有効成分とする抗がん剤)、並びに本発明の抗体の治療上又は予防上の有効量を、ヒトを含む哺乳類に投与する工程を含んでなる、Eva1タンパク質に関連する疾患(例えば、がん)の治療又は予防の方法をも提供するものである。
以上、本発明の抗体の好適な実施形態について説明したが、本発明の抗体は上記実施形態に限定されるものではない。本発明の抗体の標的であるEva1タンパク質は、がん、特にがん幹細胞において高発現しているため、例えば、光感受性物質、毒性ペプチド、化学療法剤又は放射性化学物質等の細胞傷害性物質を結合した本発明の抗体は、いわゆるミサイル療法において有用である。
また、本発明の抗体は、上述の治療方法、予防方法、及びこれらの方法に用いられる薬剤の態様に制限されることなく、診断方法、及び該方法に用いられる薬剤としても利用し得る。
以下に記載の方法にて、ヒトEva1タンパク質に対して高い親和性を示すマウス由来のモノクローナル抗体(後述の、B2E5−48抗体、C3抗体、A5D11−10抗体)を作製した。さらにそれら抗体の可変領域のアミノ酸配列を決定すると共に、相補性決定領域(CDR1〜3)を同定した。また、B2E5−48抗体及びC3抗体に関しては、これら抗体を基にし、ヒト由来の定常領域を保持するキメラ抗体を作製した。
親和性の高い抗Eva1抗体を得るため、その抗原を調製すべく、先ず、ヒトEva1タンパク質の細胞外ドメイン 27〜150位のアミノ酸からなる領域に、免疫グロブリンのFc部位を融合させたタンパク質(以下、「Fc融合Eva1」とも称する)を、HEK293細胞において発現させた。
前記28のハイブリドーマから、より抗原親和性の高い抗体を産生するハイブリドーマを選別するために、表面プラズモン共鳴解析(surface plasmon resonance assay)装置 Biacoreを行い、各抗体(以下「候補抗体」とも称する)について、K D 値等を算出した。具体的には、先ず、デキストランコートされたセンサーチップ(CM5,GE health care社製)にマウスIgGをキャプチャーする抗体を固定化した。次いで、当該抗体を介して各候補抗体をセンサーチップ上にキャプチャーさせた後に、抗原であるEva1タンパク質を流した。なお、抗原を流す際には、10mM−Tris−HCl(pH7.5),150mM−NaCl,3mM−EDTA(pH8.0)溶液を用い、また抗原を候補抗体から解離させるための溶液(キャプチャーさせた候補抗体蘇生溶液)として、10mM グリシン溶液(pH1.7)を用いた。そして、このような溶液を用い、各候補抗体と抗原との結合反応、解離反応をBiacoreにて検出し、付属のソフトウェアを用いて解析することにより、解離定数(K D )、結合速度定数(k a )、解離速度定数(k d )を算出した。その結果、前記28ハイブリドーマから産生される抗体において、高親和性を示し(K D の値が小さく)、かつ解離の遅い(k d の値が小さい)抗体上位4種(C3抗体、B1E4−32抗体、B2E5−48抗体、A5D11−10抗体)を選抜した。なお、これら抗体において、解析の結果、C3抗体とB1E4−32抗体とは、起源を同一とする抗体であることが明らかになったため、以後の解析においては、C3抗体、B2E5−48抗体及びA5D11−10抗体を対象とした。
ヒトEva1とマウスEva1との相同性は極めて高いにも関わらず、上述の通り、B2E5−48抗体は、ヒトEva1タンパク質のみを特異的に認識し、マウスEva1タンパク質に対しては結合性が認められなかった。したがって、この結果から、B2E5−48抗体はヒトEva1タンパク質とマウスEva1タンパク質との間で保存されていない12アミノ酸のいずれかを認識している可能性が示唆される。
A5D11−10抗体のFabとヒトEva1タンパク質との複合体に関し、その結晶構造を2.0Åの分解能で決定した。その構造から、A5D11−10抗体はヒトEva1タンパク質の30番目のチロシン、31番目のスレオニン、33番目のアルギニン、46番目のリシン、48番目のスレオニン、49番目のフェニルアラニン、51番目のセリン、102番目のグルタミン酸、103番目のアルギニン、104番目のチロシンを認識していることが明らかになった。次に、前記構造解析の結果から結合に重要と考えられたヒトEva1タンパク質上の30番目のチロシン、46番目のリシン、102番目のグルタミン酸、103番目のアルギニン、104番目のチロシンに関し、各アミノ酸をアラニンに置換した変異体を作製し、それらを用いた表面プラズモン共鳴法を利用した結合実験を行い、A5D11−10抗体のエピトープの確認と、該抗体が認識する各アミノ酸の結合における重要性とを解析した。その結果、Y30A変異体に対し、A5D11−10抗体の結合性は喪失した。前記構造解析の結果、当該30番目のチロシンは、A5D11−10抗体の重鎖CDR3の103番目のグリシンと104番目のグリシンの主鎖と疎水結合を形成していることが明らかになっている。また側鎖部分は、自身の50番目のセリンと水素結合を形成し、構造維持に寄与していると考えられる。そのため、アラニンに置換することにより、チロシン側鎖の環構造部分で形成されていた疎水結合が完全に断ち切られたことと、安定化に寄与していた水素結合を失ったことにより、A5D11−10抗体のヒトEva1タンパク質に対する結合性も完全に失われたと考えられる。また、K46A変異体及びY104A変異体においては、K D 値に関し、ヒトEva1タンパク質(野生型)と比較してが10−2ほど向上し、親和性の低下が確認された。前記構造解析の結果、当該46番目のリシンの側鎖は、軽鎖CDR3のアスパラギン酸の側鎖及び主鎖の両方と、水素結合を結合し、軽鎖CDR1の32番目のチロシンと分子間相互作用を形成している。また前記104番目のチロシンに関しては、側鎖部分で重鎖CDR1の32番目のアルギニンの側鎖と主鎖の両方と水素結合を形成、重鎖CDR3の102番目アスパラギン酸、103番目グリシンと分子間相互作用を形成している。そのため、かかるアラニン置換により、いくつかの結合が断ち切られ、その結果としてK D 値が10−2大きくなったと考えられる。さらに、結晶構造では、多くの結合を抗体側と結んでいる102番目のグルタミン酸と103番目のアルギニンであるが、それぞれのアラニン置換体に対し、K D 値は25倍程度の上昇が認められた。前記構造解析の結果、102番目のグルタミン酸は、A5D11−10抗体の重鎖CDR2の57番目のセリンの側鎖及び主鎖のそれぞれと水素結合を、さらには55番目のグリシン、56番目のグリシンの主鎖とも水素結合を結んでいる。一方、103番目のアルギニンは、A5D11−10抗体の重鎖CDR以外の側鎖、59番目のアスパラギン酸により水素結合を形成することで固定され、さらに53番目のトリプトファンとの間に分子間相互作用による結合が生じ、総合的に強い結合が生じていることが、構造解析から明らかになっている。そのため、前記K D 値の上昇が25倍程度であり、完全に結合を失わなかったのは、103番目のアルギニンをアラニンに置換した変異体では弱いながらも重鎖CDR2の53番目のトリプトファンとの間に分子間力が保持されていた事に基づくと予想される。また、多数の水素結合を形成しているヒトEva1タンパク質上の102番目のグルタミン酸をアラニンに置換した変異体においては、全ての水素結合が失われると予想されたにもかかわらず、25倍のK D 値の上昇であったのは、この側鎖の抗原・抗体における結合への寄与度が低いことを意味する。
上述の通り、C3抗体は、ヒトEva1タンパク質上の認識部位において、完全又は一部、A5D11−10抗体と競合していることが考えられた。そこで、上記A5D11−10抗体のエピトープ解析同様に、アラニン変異体を用いた表面プラズモン共鳴法を利用した結合実験を行った。その結果、C3抗体は、ヒトEva1タンパク質において、46番目のリシン、96番目のセリン及び103番目のアルギニンを認識していることが明らかになった。C3抗体が認識するヒトEva1タンパク質上の46番目のリシン、96番目のセリン及び103番目は、A5D11−10抗体の認識アミノ酸でもあり、これら2つの抗体がヒトEva1タンパク質との結合において競合したことと一致する。しかしながら、ヒトEva1タンパク質におけるY104A変異は、A5D11−10抗体の結合性を著しく低下させたが、C3抗体の結合性には影響を与えなかった。一方、S96A変異は、A5D11−10抗体の結合性に影響を与えなかったが、C3抗体との結合に影響を与えたことから、これら抗体において、認識部位は部分的に重なるが、認識機構は異なるとことが考えられる。
<抗Eva1抗体をコードする遺伝子のクローニングと配列決定>
B2E5−48及びC3に関し、各ハイブリドーマから常法に従ってRNAを抽出した。そして、それらを鋳型とし、SMART又はRACE cDNA増幅キット(Clontech社製)を用いて5’RACE PCRを行い、VHDHJH領域の抗体重鎖可変領域(VH鎖)及びVLJL領域の軽鎖可変領域(VL鎖)のcDNAを増幅した。

C3抗体、B2E5−48抗体及びA5D11−10抗体を各々産生するハイブリドーマを、ヌードマウスに移植し、当該マウスから腹水を採取した。次いで、腹水を4M 硫安で処理し、得られた沈殿を透析、可溶化した後、プロテインGカラムにてこれら抗体(以下、下記キメラ抗体と区別するため、C3マウス抗体、B2E5−48マウス抗体、A5D11−10マウス抗体とも称する)を精製した。
上記マウス由来のC3抗体及びB2E5−48抗体の定常領域を、ヒト由来の抗体のそれに置き換えたキメラ抗体を以下の通りに作製した。
上記の通りに調製した、ヒトEva1タンパク質に対して高い親和性を示すマウス抗体、及びキメラ抗体について、以下に記載の方法にて、ADCC活性、CDC活性、インビボにおける抗腫瘍活性を解析した。
上記の通りに調製した、C3マウス抗体、B2E5−48マウス抗体、C3キメラ抗体及びB2E5−48キメラ抗体に関し、ADCC活性(抗体依存性細胞障害活性)を解析した。すなわち、これら抗体のFc部位にFc−γ受容体を介してエフェクター細胞が結合することによって、当該細胞が活性化し、それに伴って当該抗体が認識する標的細胞を溶解し得るを、以下の方法にて解析した。
溶解率(%)=(ADCC活性による殺傷で細胞内から放出されたカルセインの蛍光値−細胞内から自律的に放出されるカルセインの蛍光値/TritonX(最終濃度1%)による殺傷で細胞内から放出されたカルセインの蛍光値−細胞内から自律的に放出されるカルセインの蛍光値)x100。得られた結果を図4〜7に示す。
上記の通りに調製した、C3マウス抗体、B2E5−48マウス抗体、C3キメラ抗体及びB2E5−48キメラ抗体について、補体の存在下、当該抗体が認識する標的細胞を溶解する活性(CDC活性:補体依存性細胞障害活性)を、以下の方法にて解析した。
溶解率(%)=(補体活性による殺傷で細胞内から放出されたカルセインの蛍光値−細胞内から自律的に放出されるカルセインの蛍光値/TritonX(最終濃度1%)による殺傷で細胞内から放出されたカルセインの蛍光値−細胞内から自律的に放出されるカルセインの蛍光値)x100。得られた結果を図9〜14に示す。
上記の通りに調製した抗Eva1抗体のインビボでの抗腫瘍活性を評価すべく、以下の通り、当該抗体をがん転移モデルマウスに投与して解析を行った。
(1)B16−Eva1を、5〜8週齢のC57BL/6マウスに1×105個 尾静脈内投与した。当該メラノーマ接種1日後から、3日間おきに、B2E5−48マウス抗体を100μgずつ尾静脈内に計4回投与した。そして、メラノーマを接種してから13日後に肺を採取し、形成されたメラノーマのコロニー数を計測した(n=5)。得られた結果を、図15及び16に示す。
(2)B16−GFPを、5〜8週齢のC57BL/6マウスに各々1×105個 尾静脈内投与した。当該メラノーマ接種1日後から、3日間おきに、B2E5−48マウス抗体又はB2E5−48キメラ抗体を100μgずつ尾静脈内に計4回投与した。そして、メラノーマを接種してから13日後に肺を採取し、形成されたメラノーマのコロニー数を計測した(n=5)。得られた結果を、図17〜20に示す。
(3)B16−GFPを、5〜8週齢のC57BL/6マウスに各々1×105個 尾静脈内投与した。当該メラノーマ接種13日後から、3日間おきに、B2E5−48マウス抗体を100μgずつ尾静脈内に計3回投与した。そして、メラノーマ接種してから22日後に肺を採取し、形成されたメラノーマのコロニー数を計測した(n=5)。また、形成されたコロニー数に応じてマウスを群に分け、それらの匹数をも計測した。得られた結果を、図21〜22に示す。
(4)B16−Eva1を、5〜8週齢のC57BL/6マウスに1×105個 尾静脈内投与した。当該メラノーマ接種1日後から、4日間おきに、C3マウス抗体を100μgずつ尾静脈内に計4回投与した。そして、メラノーマを接種してから14日後に肺を採取し、形成されたメラノーマのコロニー数を計測した(n=5)。
得られた結果を、図23及び24に示す。
(5)B16−GFPを、5〜8週齢のC57BL/6マウスに1×105個 尾静脈内投与した。当該メラノーマ接種1日後から、4日間おきに、C3マウス抗体を100μgずつ尾静脈内に計4回投与した。そして、メラノーマを接種してから14日後に肺を採取し、形成されたメラノーマのコロニー数を計測した(n=5)。
得られた結果を、図23及び24に示す。

Claims (6)
- ヒト由来のEva1タンパク質に結合し、下記(a)〜(c)のいずれかに記載の特徴を有する抗体
(a)配列番号:4〜6に記載のアミノ酸配列、配列番号:4〜6に記載のアミノ酸配列に対して80%以上の相同性を有するアミノ酸配列、配列番号:4〜6に記載のアミノ酸配列の少なくともいずれかにおいて、1若しくは数個のアミノ酸が置換、欠失、付加及び/又は挿入されたアミノ酸配列、配列番号:10〜12に記載のアミノ酸配列、配列番号:10〜12に記載のアミノ酸配列に対して80%以上の相同性を有するアミノ酸配列、並びに配列番号:10〜12に記載のアミノ酸配列の少なくともいずれかにおいて、1若しくは数個のアミノ酸が置換、欠失、付加及び/又は挿入されたアミノ酸配列からなる群から選択される少なくとも1のアミノ酸配列を、CDRとして含む可変領域を保持する
(b)配列番号:16〜18に記載のアミノ酸配列、配列番号:16〜18に記載のアミノ酸配列に対して80%以上の相同性を有するアミノ酸配列、配列番号:16〜18に記載のアミノ酸配列の少なくともいずれかにおいて、1若しくは数個のアミノ酸が置換、欠失、付加及び/又は挿入されたアミノ酸配列、配列番号:22〜24に記載のアミノ酸配列、配列番号:22〜24に記載のアミノ酸配列に対して80%以上の相同性を有するアミノ酸配列、並びに配列番号:22〜24に記載のアミノ酸配列の少なくともいずれかにおいて、1若しくは数個のアミノ酸が置換、欠失、付加及び/又は挿入されたアミノ酸配列からなる群から選択される少なくとも1のアミノ酸配列を、CDRとして含む可変領域を保持する
(c)配列番号:27〜29に記載のアミノ酸配列、配列番号:27〜29に記載のアミノ酸配列に対して80%以上の相同性を有するアミノ酸配列、配列番号:27〜29に記載のアミノ酸配列の少なくともいずれかにおいて、1若しくは数個のアミノ酸が置換、欠失、付加及び/又は挿入されたアミノ酸配列、配列番号:32〜34に記載のアミノ酸配列、配列番号:32〜34に記載のアミノ酸配列に対して80%以上の相同性を有するアミノ酸配列、並びに配列番号:32〜34に記載のアミノ酸配列の少なくともいずれかにおいて、1若しくは数個のアミノ酸が置換、欠失、付加及び/又は挿入されたアミノ酸配列からなる群から選択される少なくとも1のアミノ酸配列を、CDRとして含む可変領域を保持する。 - ヒト由来のEva1タンパク質に結合し、下記(a)〜(c)のいずれかに記載の特徴を有する抗体
(a)配列番号:3に記載のアミノ酸配列、配列番号:3に記載のアミノ酸配列に対して80%以上の相同性を有するアミノ酸配列、又は配列番号:3に記載のアミノ酸配列の少なくともいずれかにおいて、1若しくは数個のアミノ酸が置換、欠失、付加及び/又は挿入されたアミノ酸配列を含む軽鎖可変領域と、
配列番号:9に記載のアミノ酸配列、配列番号:9に記載のアミノ酸配列に対して80%以上の相同性を有するアミノ酸配列、又は配列番号:9に記載のアミノ酸配列の少なくともいずれかにおいて、1若しくは数個のアミノ酸が置換、欠失、付加及び/又は挿入されたアミノ酸配列を含む重鎖可変領域とを保持する
(b)配列番号:15に記載のアミノ酸配列、配列番号:15に記載のアミノ酸配列に対して80%以上の相同性を有するアミノ酸配列、又は配列番号:15に記載のアミノ酸配列の少なくともいずれかにおいて、1若しくは数個のアミノ酸が置換、欠失、付加及び/又は挿入されたアミノ酸配列を含む軽鎖可変領域と、
配列番号:21に記載のアミノ酸配列、配列番号:21に記載のアミノ酸配列に対して80%以上の相同性を有するアミノ酸配列、又は配列番号:21に記載のアミノ酸配列の少なくともいずれかにおいて、1若しくは数個のアミノ酸が置換、欠失、付加及び/又は挿入されたアミノ酸配列を含む重鎖可変領域とを保持する
(c)配列番号:26に記載のアミノ酸配列、配列番号:26に記載のアミノ酸配列に対して80%以上の相同性を有するアミノ酸配列、又は配列番号:26に記載のアミノ酸配列の少なくともいずれかにおいて、1若しくは数個のアミノ酸が置換、欠失、付加及び/又は挿入されたアミノ酸配列を含む軽鎖可変領域と、
配列番号:31に記載のアミノ酸配列、配列番号:31に記載のアミノ酸配列に対して80%以上の相同性を有するアミノ酸配列、又は配列番号:31に記載のアミノ酸配列の少なくともいずれかにおいて、1若しくは数個のアミノ酸が置換、欠失、付加及び/又は挿入されたアミノ酸配列を含む重鎖可変領域とを保持する。 - ヒト由来の定常領域を有する、請求項1又は2に記載の抗体。
- ADCC活性及びCDC活性から選択される少なくとも1の細胞障害活性を有する、請求項1〜3のいずれか一項に記載の抗体。
- 請求項1〜4のいずれか一項に記載の抗体を有効成分として含む医薬組成物。
- 抗がん剤である、請求項5に記載の医薬組成物。
