Abstract
Description
試料をウェル内の試薬と反応させる反応ステップであって、2次元アレイに配置された複数のウェルを有するマイクロプレートが使用される反応ステップと、
ウェルの底壁が回転軸に対して外方向に配置されるようにマイクロプレートを回転させる遠心分離ステップを含み、遠心分離ステップにおいて、凝集された試料物質が、ゲル物質またはビーズマトリックスなどの分離物質によって、凝集されていない試料物質から分離される遠心分離ステップと、
マイクロプレートの上側の少なくとも1つの画像およびマイクロプレートの下側の少なくとも1つの画像を撮影する撮像ステップと、
上記試料ウェル内の試料を、凝集反応に関して陽性または陰性であると決定する決定ステップであって、マイクロプレートの上側と下側の画像内の上記ウェルの色強度および/またはグレーレベルが比較される決定ステップと
を含む。

Claims (14)
- 凝集反応の結果を決定するための方法であって、
試料をウェル(2)内の試薬と反応させる反応ステップであって、2次元アレイに配置された複数のウェル(2)を有するマイクロプレート(1)が使用される反応ステップと、
前記ウェル(2)の底壁(4)が回転軸(24)に対して外方向に配置されるように前記マイクロプレート(1)を回転させる遠心分離ステップにおいて、凝集された試料物質が、ゲル物質またはビーズマトリックスなどの分離物質によって、凝集されていない試料物質から分離される遠心分離ステップと、
前記マイクロプレート(1)の上側の少なくとも1つの画像および前記マイクロプレート(1)の下側の少なくとも1つの画像を撮影する撮像ステップと、
前記試料ウェル(2)内の前記試料を、凝集反応に関して陽性または陰性であると決定する決定ステップであって、前記マイクロプレートの前記上側と前記下側の前記画像内の前記ウェルの色強度および/またはグレーレベルが比較される決定ステップと
を含む方法。 - 前記遠心分離ステップにおいて、前記マイクロプレート(1)が水平軸の周りで回転される請求項1に記載の方法。
- 前記遠心分離ステップの前に培養ステップをさらに含む請求項1または2に記載の方法。
- 前記分離物質が、前記試薬を含む懸濁液中に混合される請求項1〜3のいずれか一項に記載の方法。
- 前記試薬が、前記分離物質の上に提供される請求項1〜4のいずれか一項に記載の方法。
- 前記試料が、血液細胞の試料を含む請求項1〜5のいずれか一項に記載の方法。
- 前記試薬が、所定の抗体および/または抗原を含む請求項1〜6のいずれか一項に記載の方法。
- 前記マイクロプレートが、前記方法の最初に、空のウェルを含み、前記反応ステップの前に、前記ウェルの少なくとも幾つかが、前記分離物質、前記試薬、および前記試料物質を充填される請求項1〜7のいずれか一項に記載の方法。
- 2次元アレイに配置される複数のウェル(2)を有する、凝集反応の生成物を決定するためのマイクロプレートであって、
前記ウェルの少なくとも1つが分離区域(7)を備え、前記分離区域(7)が、ゲルまたはビーズマトリックスなどの分離物質を含み、
前記分離区域(7)が、下に向かって先細りにされた少なくとも1つの円錐部分(9)を備え、それにより、前記分離物質を通過する試料物質が、前記それぞれのウェル(2)の中央に集中されるマイクロプレート。 - 前記ウェル(2)が、前記ウェルの底端部に、前記分離物質を通過した前記試料物質を収集するための収集区域(10)を備え、前記収集区域(10)が、好ましくは中空円筒体の形状を有する請求項9に記載のマイクロプレート。
- 前記分離区域(7)が、中空円筒体を含む請求項9または10に記載のマイクロプレート。
- 前記ウェル(2)が、前記ウェル(2)の上端部に充填区域(5)を備え、前記充填区域(5)の断面領域が、前記分離区域(7)の断面領域よりも大きい請求項9〜11のいずれか一項に記載のマイクロプレート。
- 前記マイクロプレート(2)が、少なくとも96個のウェルを備え、および/または前記反応ウェル(2)の前記内部高さが、5mm〜20mmの間である請求項9〜12のいずれか一項に記載のマイクロプレート。
- 遠心分離機(14)と、前記反応ウェル(2)の上側を検出するためのカメラ(44)と、前記反応ウェルの下側を検出するためのカメラ(45)と、請求項1〜8のいずれか一項に記載の方法を実施するための制御ユニットとを備える試験装置であって、好ましくは、請求項9〜13のいずれか一項に記載のマイクロプレートが使用される試験装置。
