中間安定化siRNAは、dTsdT突出がない点で完全安定化siRNAと異なる。いかなる2’−F修飾も示さなかった。
RPMI 1640培地を用いて、C4−2細胞を、この細胞株について記載される標準的な手順に従って培養した。
リポフェクタミン2000を用いて、C4−2細胞を様々な濃度のsiRNAでトランスフェクトした。siRNAの濃度は、トランスフェクション実験において、50nM、10nM、1nMまたは0.1nMであった。インキュベーション時間は24時間であった。それ以外は標準的な手順を用いた。トランスフェクション効率を、ハウスキーパーsiRNAを測定することにより決定した;トランスフェクションの有効性は、全ての場合において少なくとも93%であった。
標準的な手順を用いてIC50値を決定した。より具体的には、示される様々なsiRNAを用いて記載されるようにトランスフェクトされたC4−2細胞を用いて、Notch 1のmRNAの発現が50%まで減少するsiRNA濃度を決定した。
ナノ担体(パーフルオロカーボンナノ担体)の調製のために、リン脂質の混合物を用いてパーフルオロオクチルブロマイド(パーフルブロン)を乳化した。1グラムの混合物は、蒸留水および75mMのリン酸二水素ナトリウム(NaH2PO4)バッファー中に、ホスファチジルコリン(980mg)、スフィンゴミエリン(10mg)、コレステロール(5mg)、リゾホスファチジルコリン(5mg)を含む。1000μlのパーフルオロカーボンナノ担体を得るために、475μlのパーフルオロオクチルブロマイド、36mgのリン脂質、200μlの75mM NaH2PO4(pH7.4)および325μlの蒸留水を用いた。
Notch siRNA−負荷ナノ担体を、上述のナノ担体に基づいて調製した。負荷する目的のために、所望のsiRNA種をナノ担体に加えて、そのようにして得られた混合物を、超音波により15秒間500Wで用いてホモジナイズに供した。
皮下接種されたPANC−1ヒト膵臓腫瘍由来の腫瘍を有する、60匹の雌無胸腺ヌードFoxn1nuマウスを、112匹のプールから選択した。この動物モデルは、Lieber M et al.(M.Lieber,et al.,“Establishment of a continuous tumor−cell line (panc−1) from a human carcinoma of the exocrine pancreas”,Int J.Cancer 15(5),741(1975))により最初に説明され、Madden ME and Sarras MP(M.E.Madden and M.P.Sarras,Jr.,“Morphological and biochemical characterization of a human pancreatic ductal cell line (PANC−1)”,Pancreas 3(5),512(1988))により生化学的および形態学的に特徴付けられた、PANC−1細胞株を用いて膵癌および膵臓腫瘍についてそれぞれ実証された動物モデルである。そのような細胞株、および、そのような細胞株を用いて実証された動物モデルは、抗癌剤の評価に用いられている。Schultz RM et al.(R.M.Schultz,et al.,“Evaluation of new anticancer agents against the MIA PaCa−2 and PANC−1 human pancreatic carcinoma xenografts”,Oncol.Res.5(6−7),223(1993))。
66匹の雌無胸腺ヌードFoxn1nuマウスを、PANC−1ヒト膵臓腫瘍細胞で同所性接種した。生着率(take−rate)を、3匹のマウスにおいて、接種後20日および27日にそれぞれ評価した。接種後32日に、残りの60匹のマウスを、体重により、10匹の6つのグループにランダム化した(0日目)。
C4−2細胞を培養し、実施例1に記載の表1に示される様々なsiRNAでトランスフェクトし、ここで、siRNA濃度は50nMであった。結果を図1に示す。図1は、様々なsiRNAを用いたC4−2細胞のトランスフェクション時のNotch 1の相対的発現を示すダイアグラムである。発現は、コントロールを用いたC4−2細胞におけるNotch 1の発現に対して標準化される。コントロールは、タンパク質をコードするいかなる公知のmRNAも標的化しないsiRNAであった。
表1および表2のsiRNA分子XD00404およびXD−00409を、実施例1にも記載のように、中間安定化および完全安定化に供した。このようにして修飾されたsiRNA分子は、以下のとおりである。

実施例1に記載のPANC−1ヒト膵臓腫瘍を用いた動物試験を行なった。用いたsiRNA種は、完全安定化XD−00409であった。
−ゲムシタビン単剤療法、およびNotch/ゲムシタビンの組み合わせを用いた処置の後に、軽度の腫瘍阻止が明らかであったが(腫瘍重量および腫瘍増殖の割合の測定により示される)、Notch/ゲムシタビン併用療法のみが、腫瘍重量に関してビヒクルと有意に異なった。腫瘍阻止における相乗の応答(腫瘍増殖の割合により測定される)は、併用療法Notch/ゲムシタビンにより示された。
−ゲムシタビン単剤療法およびNotch/ゲムシタビン併用療法により処置されたマウスは、腫瘍壊死の発生が最も低かった。
実施例1に記載のPANC−1ヒト膵臓腫瘍を用いた動物試験を行なった。用いたsiRNA種は、完全安定化XD−00409であった。
