Abstract
Description
慢性閉塞性肺疾患(COPD)、喘息、気管支拡張症、嚢胞性線維症、および間質性肺疾患を含めた多くの肺疾患が、慢性炎症および酸化的ストレスを伴なう。COPDは米国で第4位の主要な死因であり、毎年の死亡全体の約4.5%を占める。有病推定値は13,500,000人プラス“未診断”で最大15,000,000人に及ぶ;200万人は肺気腫を伴なう。それは炎症、水腫および分泌物により引き起こされる多様性疾患であり、結果として肺の全領域に形態変化が生じる。肺機能は年齢と共に低下し、進行性の不可逆的臓器不全として、特に肺気腫および慢性気管支炎において顕性になる。これらの障害は世界的に廃疾および死亡の主な負荷となっており、現在はそれらの自然経過を著しく変化させる全肺移植片の不足のため療法はない。
脳は、高レベルの酸素を必要とする結果として、ROS仲介による損傷に対して特に感受性が高い。血液−脳関門の背後で、正常なミトコンドリア呼吸からの漏出および活性化されたミクログリアからの呼吸によるRSバーストにより発生する酸化的ストレスが、本明細書中で考察するアルツハイマー病、パーキンソン病、外傷性脳傷害、多発性硬化症、老人性認知症、筋萎縮性側索硬化症その他を含めた中枢神経系の難病における神経死に関与することが長い間疑われてきた。研究により、特に高齢対象の脳において悪質かつ自律永続性の自己中毒サイクルになる可能性があるものに、DNA、脂質、タンパク質、カルシウムバランス、および神経伝達物質活性の酸化的損傷の証拠が見出された。RS活性のマーカーがすべての主要なCNS疾患に見出された。神経変性疾患に対する最も信頼性のあるリスク因子は老化であり、これは高齢化に際して脳がRS傷害をより受けやすくなる可能性があること、および/またはそれらの作用が長期間かけて増強される可能性があることを示唆する。“すべてではないとしても大部分の細胞死モデルはフリーラジカル種および酸化的ストレスを伴なう。したがって、フリーラジカルが酸化的損傷を仲介するのを停止または遅延させることを目標とした療法戦略を考案することにより、神経変性疾患における細胞死を妨げることができる可能性がある”(12)。

大部分の主要な慢性疾患においてアンチオキシダントの有効性は広く研究されており、それには下記のものが含まれる:カロテノイド、フラボノイド、ビタミン(アスコルベートおよびトコフェロールを含む)、無機物(亜鉛、セレン)、果実および野菜ならびに抽出物、ユビキノン(補酵素Q−10)、グルタチオン(グルタチオンエステル、グルタチオンペルオキシダーゼミメティック、グルタチオン生合成誘導物質)、リポ酸、メラトニン、チオール化合物(N−アシステリン(N-acystelyn)、N−イソブチリルシステイン、合成新規チオール類、およびN−アセチル−L−システイン)、ニトロンスピントラップ剤(nitrone spin trap)、スーパオキシドジスムターゼ(SOD)およびカタラーゼ、SODミメティック、ならびにレドックスセンサー阻害剤。若干の研究は有効性を示したが、外的要因を適切に制御するのに苦闘し、しばしば類似の研究を再現するのに失敗した。SODおよびカタラーゼは生物学的に有効な内因性アンチオキシダント酵素であるが、これらの酵素は細胞によって容易には取り込まれないので、それらを外から導入するのは問題が多い。生物学的利用能は外因性アンチオキシダント戦略全般における重要な複雑な要因である;たとえ炎症部位に適切に配置したとしても、大部分のアンチオキシダントはフリーラジカルとの1回の相互作用で消費され、その結果、連続投与の手段なしではそれらのスカベンジング効力は制限される。
・セリアナノ粒子は、パーキンソン病のMPTPマウスモデルにおいて線条体ドーパミンを保存し、黒質のドーパミン作動性ニューロンを保護し、用量応答性は釣鐘形である。
1.卓越した療法特性を備えた有効な生物学的利用能のある酸化セリウムナノ粒子プラットホームを作製する。セリアナノ粒子(直径5〜10nm)は生物活性をもち、ペルオキシナイトライトおよびスーパーオキシドのレベルを低下させ、その結果、炎症の傷害性細胞作用を低減する。新規コーティングおよびキャリヤーまたは多官能性炭化水素を用いて修飾したナノ粒子を合成することができ、レドックス活性を増強するための内部修飾を開発し、粒子を特定の組織ヘターゲティングさせ、隔離環境への粒子透過を増大させることができる(肺の上皮層を通した吸収、血液脳関門の透過増大)。選択的取込みのために肺への吸入により投与できまたは静脈内に投与できる組成物および表面特性、パッケージングならびに送達ビヒクルの工学作製は、特異的にフリーラジカル発生部位における蓄積を最小化し、生物学的利用能を増大させ、有効性を最適化するであろう。

Claims (20)
- 酸化セリウムナノ粒子および配位子シェルを含む多層カプセル化した酸化セリウムナノ粒子(“CeNP”)であって、場合により炭化水素付加、電子シャトリング系、脂質シェル、ターゲティング分子付着、またはその組合わせを含むことができるCeNP。
- 配位子シェルがCeNPに対する最も内側の連結である、請求項1に記載のCeNP。
- 配位子シェルが、炭素2〜40個の長さを有する炭化水素を含む、請求項1に記載のCeNP。
- 配位子シェルがキレート化カルボン酸を含む、請求項3に記載のCeNP。
- キレート化カルボン酸が、ブチル、t−ブチル、ヘキシル、デシル、ヘキシルデシルカルボン酸、またはカルボン酸とエーテル、エステル、エポキシド、ペルオキシド、チオールもしくはアセタールとの対向官能基を備えた炭化水素、またはその組合わせのうち少なくとも1つを含む、請求項4に記載のCeNP。
- 配位子シェルが、ステアリン酸、オレイン酸、ポリアクリレート、クエン酸、またはその組合わせを含む、請求項1に記載のCeNP。
- 炭化水素付加がn−末端アミン炭化水素を含む、請求項1に記載のCeNP。
- 炭化水素付加がリンカーである、請求項7に記載のCeNP。
- アミン炭化水素が、ブチル、t−ブチル、ヘキシル、デシル、ヘキシルデシルアミン、またはω−末端のエーテル、エステル、エポキシド、ペルオキシド、チオールもしくはアセタールなどの二重官能基を備えたアミンを含む、請求項7に記載のCeNP。
- 電子シャトリング系が、大型のコンジュゲートした系、または固定ベンジル環の系、または炭化水素鎖上の交互二重結合を含む、請求項1に記載のCeNP。
- 脂質シェルが長鎖大型脂質を含む、請求項1に記載のCeNP。
- 脂質シェルが、種々のヘッド基およびテイル選択肢を備えたリン脂質、スフィンゴ脂質またはステロール類を含む、請求項1に記載のCeNP。
- ターゲティング分子付着が、カルボン酸、チオール基またはアミンを使ってカップリングする小分子を含む、請求項1に記載のCeNP。
- ターゲティング分子付着が、L−DOPA、ドーパミン、セロトニン、アセチルコリン、6OHDA、その誘導体、またはペプチドを含む、請求項1に記載のCeNP。
- 反応性酸素種に対するCeNP作用を制御および指向する方法であって、
少なくとも1つのアンジッピング形成を備えたCeNPを作製し;
CeNPを反応性酸素種またはフリーラジカルの存在に曝露し、それにより、反応性酸素種またはフリーラジカルが形成される部位または多量に存在する部位に対するCeNPのアンチオキシダント活性が得られるようにする
ことを含む方法。。 - CeNPが、処理されたセリウム表面に連結した脂質カプセルを含む、請求項15に記載の方法。
- CeNPが、セリウムの修飾された表面における脂質コートの形成を容易にする短い連結炭化水素を含む、請求項16に記載の方法。
- 配位子シェルおよび脂質シェルの両方またはいずれか一方に埋め込まれた化学結合が、反応性酸素種またはフリーラジカルによる攻撃に対して感受性である、請求項17に記載の方法。
- CeNPと体内の血液および組織との相互作用を制限するための方法であって、
多層カプセル化した酸化セリウムナノ粒子(“CeNP”)をその必要がある対象に投与し、その際、CeNPの固有の抗酸化活性を制限するようにCeNPを形成する
ことを含む方法。 - CeNPを、炭素鎖または他の嵩高い付加物、たとえばt−ブチル、シクロアルカン類、樹状構造体、ポリプロピレン官能基で不動態化する、請求項19に記載の方法。
