Abstract
Description
抗体−薬物結合体(ADC)は、組織または生体において標的部位へ薬物を送達する効果的な手段を提供することができる。抗体による腫瘍などの標的の認識は、標的ではない組織の毒性のある化学療法剤への曝露を最小にし、「遊離」薬物(すなわち、抗体などの担体に結合していない)の毒性に関連する副作用を限定する。ADCは、いくつかの技術によって調製することができる。ヒトまたは他の被験体への投与の前に、遊離薬物および他の不純物を取り除くため、結合体は精製される。
一部において、本発明は、ベンゾジアゼピンADCおよびベンゾジアゼピン薬物関連不純物を含む混合物(本明細書においてADC混合物ともいう)からのベンゾジアゼピン薬物関連不純物の除去は、ベンゾジアゼピン薬物の特質のために非効率的であるという知見、ならびに混合物へのシクロデキストリンの添加が、ベンゾジアゼピン薬物関連不純物の効率的な排除を可能にするという発見に基づく。本発明者らは、とりわけ、ベンゾジアゼピンADCおよびベンゾジアゼピン薬物関連不純物を含む混合物に、接線流ろ過を行う前にシクロデキストリンを添加することによって、ベンゾジアゼピン薬物関連不純物の効率的な排除が可能になるということを見出した。

ベンゾジアゼピン抗体−薬物結合体
ベンゾジアゼピン抗体−薬物結合体は、ベンゾジアゼピン2量体に、典型的にはリンカーを介して結合体化した抗体をいうが、リンカーは必ずしも必要ではない。ベンゾジアゼピン薬物リンカーは、リンカーに付いたベンゾジアゼピン2量体をいう。ベンゾジアゼピン薬物リンカーのリンカー構成成分は、典型的には、抗体に付くための反応性基を有する。ベンゾジアゼピン化合物は、ジアゼピン環へ縮合したベンゼン環をその中心に有する。ジアゼピン環へ縮合したベンゼン環の例示的な環構造は以下の通りである。

本発明は、特に、ベンゾジアゼピンADCおよびベンゾジアゼピン関連不純物を含む混合物からベンゾジアゼピン薬物関連不純物を除去する方法を対象とする。ベンゾジアゼピン薬物関連不純物は、抗体とベンゾジアゼピン2量体またはベンゾジアゼピン薬物リンカーとの結合体化反応から生じる任意の薬物関連不純物である。ベンゾジアゼピン薬物関連不純物には、例えば、ベンゾジアゼピン2量体遊離薬物、ベンゾジアゼピン薬物リンカー、クエンチされたベンゾジアゼピン薬物リンカー、またはベンゾジアゼピン薬物リンカー分解生成物が含まれ得る。ベンゾジアゼピン薬物関連不純物は、抗体と結合体化したベンゾジアゼピン薬物でも抗体と結合体化した薬物リンカーでもない。
典型的には、結合体化反応および任意選択のクエンチの後、ベンゾジアゼピンADCおよびベンゾジアゼピン薬物関連不純物を含む混合物は、接線流ろ過(TFF)に供される。本発明者らは、混合物から薬物関連不純物を効果的に除去するためには、シクロデキストリンを使用し、ろ過の間を通じてシクロデキストリンの最小レベルを維持することが好ましいということを見出した。ベンゾジアゼピン関連不純物の排除を最適化するために、シクロデキストリンは、ADC混合物の構成成分(例えばADC)の溶解性を実質的に維持するレベルに維持される。シクロデキストリンは、ろ過プロセスの開始の前(例えば、結合体化および任意選択のクエンチの後であるが接線流ろ過を始める前)に、またはろ過プロセスの開始(もしくは始まり)時に、混合物に好ましくは添加される。いくつかの局面において、シクロデキストリンは、ろ過プロセスの開始後であるが不純物の実質的な除去の前に、ADC混合物へ添加される。可溶化剤(例えばシクロデキストリン)は、混合物が接線流ろ過に供される前に、混合物に好ましくは添加される。シクロデキストリンのレベルは、ろ過の間を通じて最小レベルに好ましくは維持される。
シクロデキストリンは、デンプンから製造される非還元環状グルコースオリゴ糖である。α−1,4グリコシド結合によって連結した6、7または8個のグルコース単位を有する3種の一般的なシクロデキストリンがある(それぞれ、α−、β−およびγ−シクロデキストリン)。シクロデキストリンは、ゲスト分子をそれらの内部空洞内に捕捉して、それによって包摂複合体を形成することによって、分子容器として作用することができる。α−シクロデキストリンはより小さな空洞を有するのに対し、β−およびγ−シクロデキストリンはより大きな空洞を有する。
本発明は、ベンゾジアゼピンADCおよびベンゾジアゼピン薬物関連不純物を含む混合物(結合体化反応混合物およびクエンチされた結合体化反応混合物を含む)、TFF被保持物製剤、ならびにベンゾジアゼピンADCおよびシクロデキストリンを含む医薬製剤を提供する。TFF被保持物製剤とは、TFFを使用して精製されたが最終的な製剤化がなされていないADC混合物をいう。
本発明は、ベンゾジアゼピンADCおよびシクロデキストリンを含む医薬製剤を提供する。本発明者らは、約2%w/vまたはそれ超のシクロデキストリンを含有するシクロデキストリン含有製剤は、製剤中に0.5%w/vまたはそれ未満のシクロデキストリンを含有する製剤と比較して、凝集の速度および程度の低減を奏し、シクロデキストリンを含有しない製剤と比較して、酸性種の増大の低減を奏するということを発見した。本発明者らは、約6%w/vまたはそれより多いシクロデキストリンを含有する製剤は、製剤中に2%w/vまたはそれ未満のシクロデキストリンを含有する製剤と比較して、ベンゾジアゼピン薬物リンカーの化学的分解の低減を奏するということも発見した。したがって、一部において、本発明は、約6%w/vのシクロデキストリンを含むベンゾジアゼピンADCは、より少量のシクロデキストリンを含有する製剤またはシクロデキストリンを全く含有しない製剤と比較して、改善された安定性を奏するという発見に基づいている。改善された安定性は、例えば、以下の1つまたは複数によって実証され得る:(i)凝集の速度および程度の低減、(ii)酸性種の増大の低減、および(iii)薬物の化学的分解の低減。ある特定の局面において、本発明は、治療に使用するための安定化された液体または凍結乾燥ベンゾジアゼピンADC製剤を提供する。特に、治療のためのベンゾジアゼピンADCが、長期間にわたって安定で、様々な投与経路を通じて投与され得るように安定化された製剤を提供する。そのような製剤は、例えば、ADCの抗体構成成分によって認識される抗原を発現する標的細胞に対して細胞傷害性または細胞増殖抑制効果を発揮することによる疾患または障害の処置(例えば、がんの処置)において使用する予定のベンゾジアゼピンADCにとりわけ有用である。
PBD薬物リンカーは、重鎖の239位を介してh2H12ec抗体またはh1F6ecと結合させた(抗体の名称の後の記号「ec」は、組み込み操作されたシステインを239位に有する抗体を指す)。簡潔に言うと、239位(EUインデックスによるナンバリング)に導入されたシステインを有するh2H12およびh1F6を、還元し、部分的に再酸化し(すなわち、鎖間ジスルフィドについて再酸化し)、WO2011/130613に記載の方法を使用してPBD薬物リンカーと結合してADCを形成した。PBD薬物リンカーは、導入されたシステイン残基を介して、部分的に再酸化された抗体と結合された(平均して、抗体当たり2つの薬物リンカー)。h2H12−1は、化合物1と結合体化したヒト化2H12ec抗体を指し、h1F6−1は、化合物1と結合体化したヒト化1F6ec抗体を指す。h2H12−2は、化合物2と結合体化したヒト化2H12ec抗体を指し、h1F6−2は、化合物2と結合体化したヒト化1F6ec抗体を指す。h2H12−3は、化合物3と結合体化したヒト化2H12ec抗体を指し、h1F6−3は、化合物3と結合体化したヒト化1F6ec抗体を指す。実施例4〜7は、239位に導入されたシステインを介して化合物1と結合体化したh2H12ecまたはh1F6ec抗体のいずれかを使用して行われた。
透析、バッファー交換カラムまたは遠心ろ過を使用して出発物質を試験製剤バッファーへバッファー交換することによって、製剤を調製した。製剤は、図の凡例に示した通りである。存在すると示されている場合のアルギニンの濃度は、0.5Mである。最終ADC濃度は、UV分光分析によって決定して1.3mg/mLから3.1mg/mLの範囲であった。製剤化したADCは、予め殺菌した試験管に充填し、以下のICH条件にて保存した:2〜8℃、25℃/65%RH、および/または40℃/75%RH。%HMWは、サイズ排除クロマトグラフィーを使用してモニターした。図1の製剤はh2H12−1を含有し、保存は25℃であり、図2の製剤はh1F6−1を含有し、保存は25℃であり、図3の製剤はh2H12−3を含有し、保存は40℃であり、図4の製剤はh1F6−3を含有し、保存は40℃であり、図5の製剤はh2H12−2を含有し、保存は40℃であり、図6の製剤はh1F6−2を含有し、保存は40℃であり、図13の製剤はh1F6−1を含有し、保存は25℃である。

透析、バッファー交換カラムまたは遠心ろ過を使用して出発物質を試験製剤バッファーへバッファー交換することによって、製剤を調製した。概して、製剤としては、対照としての初期製剤(主にアルギニン)、バッファー、シクロデキストリン3%およびシクロデキストリン6%が含まれていた。最終ADC濃度は、UV分光分析によって決定して1.3mg/mLから3.1mg/mLの範囲であった。製剤は、予め殺菌した試験管に充填し、以下のICH条件にて保存した:2〜8℃、25℃/65%RH、および/または40℃/75%RH。薬物リンカーの安定性は、還元PLRP/MSを使用した分解物の%、またはペプシン消化マップを使用した、インタクトな薬物リンカーの%によって測定された。結果は、左欄に挙げた製剤について、以下の通りである。表2は、シクロデキストリンを6%含有する製剤が、40℃にて1週間後に、シクロデキストリンを2%含有する製剤より少ない薬物リンカー分解を示したということを実証している。表3は、シクロデキストリンを6%含有するh2H12−1製剤が、25℃にて1週間後に少ない薬物リンカー分解を示したということを実証しており、表4は、hpbシクロデキストリンを6%含有するh1F6−1製剤が、40℃にて1週間のインキュベーション後に、インタクトな薬物リンカーを高いレベルで維持したということを実証している。
この実験は、h2H12−1を含むクエンチされた結合体化反応混合物(QCR)のダイアフィルトレーション中の凝集を防ぐための、ダイアフィルトレーションバッファーの構成成分としてのヒドロキシプロピル−β−シクロデキストリン(hpb−CD)3w/v%の有用性を試験したものである。この実験の第一の目的は、ダイアフィルトレーションに用いられる膜がhpb−CDを透過させ得るか否かを決定することにあった。膜がhpb−CDを透過させにくい場合は、バッチ中のその濃度は、ダイアフィルトレーションバッファー中の濃度を超えて増加することになるのに対し、透過させやすい場合は、バッチ中のhpb−CD濃度は、ダイアフィルトレーションバッファー中の濃度に達して、そのレベルを維持することになる。
この実験においては、pH8.0のトリス50mM/EDTA5mM中に、h2H12−1、プロピレングリコール50w/w%、クエンチされた薬物リンカー(化合物4)およびNACを含有するQCRを利用した。この混合物を、pH8.0のトリス50mM/EDTA5mM中のhpb−CD50w/v%溶液の25体積%を添加することによって、hpb−CD10w/v%にした。その後、この混合物を、実施例4と同様の条件であるが適切な規模にしたダイアフィルトレーションに付した。サンプルを各ダイアフィルトレーション容量の後に取り出し、分析まで凍結した。クエンチされた薬物リンカーの濃度を、各サンプル中で決定した。クエンチされた薬物リンカーの濃度は、ln(C/C0)とダイアフィルトレーション容量#との間の直線関係:C/C0=exp(−SN)によって示される、定容ダイアフィルトレーションを通じた排除についての確立されたモデルに従うようにして、ダイアフィルトレーション全体を通じて下降した。CおよびC0は測定される被検体の濃度であり、Nはダイアフィルトレーション容量数であり、Sは、ふるい係数であって、膜の透過側の被検体濃度を膜の被保持物側の被検体濃度で割ったものとして定義される。開始濃度は16.3μMであり、最終濃度は0.14μMであった。
実施例4および5に記載の実験は、シクロデキストリンを使用したダイアフィルトレーションプロセスによって、クエンチされたPBD薬物リンカーを混合物から除去し得ることを示した。この実験の目的は、この方法が、クエンチされたPBD薬物リンカーの出発濃度がより高いQCRから、クエンチされた薬物リンカーをうまく清浄化できるかどうかを決定することにあった。
TFFプロセスを、クエンチされた結合体化反応混合物に対して、分子量30kDをカットオフする0.1m2のUltracel Pellicon3膜(Millipore)を使用して行った。初めは流速によって、その後は被保持物バルブによって、TMPを20psiに維持した。TFFプロセスは、20個のダイアフィルトレーション容量で行い、2個のダイアフィルトレーション容量毎にサンプルを採取した。クエンチされたPBD薬物リンカーの濃度を、各サンプル中で決定した。
クエンチされた薬物リンカー(化合物4)を、定容ダイアフィルトレーションを使用して、QCRから精製した。クエンチされた結合体化反応混合物を、接線流ろ過装置内へ導入した。クエンチされた結合体化反応混合物は、トリス、NaCl、およびプロピレングリコール50%を含む。接線流ろ過バッファーも、トリス、NaCl、およびプロピレングリコール50%を含む。一連の限外ろ過/ダイアフィルトレーションの後、1.1μMのベンゾジアゼピン薬物関連不純物が混合物中に残留し、その際、4個のダイアフィルトレーション容量の後に排除が失速した(データ示さず)。
シクロデキストリンを6%含有するADC製剤が細胞傷害行性を保持しているかどうかを決定するため、細胞傷害性研究を行った。結果は、ADC製剤が細胞傷害行性を保持していることを実証していた(データ示さず)。
7.57のSlogP値を有するクエンチされていないPBD薬物リンカー(化合物1)を、定容ダイアフィルトレーションを使用して、クエンチされていない結合体化反応混合物から精製した。シクロデキストリンは、3%に維持した。驚くべきことに、クエンチされていないPBDリンカーの排除(データ示さず)は、前の実施例に示したクエンチされたPBD薬物リンカーの排除ほど効率的ではなかった。この実験は、PBD薬物リンカーの疎水性を低くすること(化合物4は5.76のSlogP値を有する)によって排除が改善されることを実証した。
Claims (64)
- ベンゾジアゼピンADCおよびベンゾジアゼピン薬物関連不純物を含む混合物からベンゾジアゼピン薬物関連不純物を除去する方法であって、ベンゾジアゼピンADCおよびベンゾジアゼピン薬物関連不純物を含む混合物を、該混合物中で少なくとも約1%w/vのシクロデキストリン濃度を維持しながら接線流ろ過に供する工程を含む、方法。
- 前記シクロデキストリンが、接線流ろ過の開始時に前記混合物中に存在する、請求項1に記載の方法。
- 前記シクロデキストリンが、ベンゾジアゼピン薬物関連不純物の実質的な除去の前に前記混合物に添加され、シクロデキストリンが、該混合物中で少なくとも約1%w/vの濃度でその後維持される、請求項1に記載の方法。
- 少なくとも約2%のシクロデキストリン濃度が前記混合物中で維持される、請求項1から3のいずれか一項に記載の方法。
- 少なくとも約3%の濃度が前記混合物中で維持される、請求項1から3のいずれか一項に記載の方法。
- 前記接線流ろ過が、定容ダイアフィルトレーションである、前記請求項のいずれか一項に記載の方法。
- 接線流ろ過装置が、ポンプ;入口、ろ液出口、被保持物出口を有するろ過ホルダー;約50Kdまたはそれより小さい孔サイズを有する限外ろ過膜であって、該ろ液出口を通って該ろ過ホルダーから出る前に全てのろ液が該入口に入り該限外ろ過膜を通り抜けるように、該ろ過ホルダーを上流区画と下流区画とに分ける限外ろ過膜;結合体化反応混合物を保持するためのサンプル貯留槽;および該サンプル貯留槽と流体連通しているバッファー貯留槽を含み、該バッファー貯留槽中のバッファーが、少なくとも約1%w/vのシクロデキストリン、少なくとも約2%w/vのシクロデキストリン、または少なくとも約3%w/vのシクロデキストリンを含む、請求項1から3のいずれか一項に記載の方法。
- 前記バッファーが、ろ液流と同じ速度でろ過容量を置き換え、前記接線流ろ過装置中の容量を一定に維持する、請求項7に記載の方法。
- 前記ろ過が不連続ダイアフィルトレーションである、請求項1から5のいずれか一項に記載の方法。
- (i)ベンゾジアゼピンADCを含む結合体化反応混合物を形成するのに十分な条件下で、抗体をベンゾジアゼピン薬物リンカーと接触させる工程、および(ii)該反応混合物をクエンチング剤と接触させて、クエンチされた結合化反応混合物を形成する工程をさらに含み、接線流ろ過に供される前記混合物が、クエンチされた結合体化反応混合物である、前記請求項のいずれか一項に記載の方法。
- 前記ベンゾジアゼピン薬物関連不純物が、クエンチされたベンゾジアゼピン薬物リンカーである、請求項1から10のいずれか一項に記載の方法。
- 前記クエンチされたベンゾジアゼピン薬物リンカーが、7.50を超えないまたは6.5を超えないSlogP値を有する、請求項11に記載の方法。
- 前記ベンゾジアゼピン薬物関連不純物が、約1μMもしくはそれ未満、0.5μMもしくはそれ未満、0.1μMもしくはそれ未満、または0.05μMもしくはそれ未満のレベルまで低減される、前記請求項のいずれかに記載の方法。
- 前記シクロデキストリンが、化学修飾されたベータシクロデキストリンである、前記請求項のいずれか一項に記載の方法。
- 前記シクロデキストリンが、ヒドロキシプロピルベータシクロデキストリンまたはスルホブチルエーテルベータシクロデキストリンである、請求項14に記載の方法。
- 前記シクロデキストリンが、ヒドロキシプロピルベータシクロデキストリンである、請求項15に記載の方法。
- 前記ベンゾジアゼピンADCが、ピロロベンゾジアゼピン2量体と結合体化したモノクローナル抗体である、前記請求項のいずれか一項に記載の方法。
- 前記ベンゾジアゼピンADCが、インドリノベンゾジアゼピン2量体またはオキサゾリジノベンゾジアゼピン2量体と結合体化したモノクローナル抗体である、請求項1から16のいずれか一項に記載の方法。
- 前記請求項のいずれか一項に記載の方法を使用して精製された混合物。
- 製剤中にベンゾジアゼピン薬物関連不純物を0.1μMまたはそれ未満含む、ベンゾジアゼピンADC製剤。
- ベンゾジアゼピンADC、および
約3%w/vから約30%w/vの濃度のシクロデキストリン
を含む、医薬製剤。 - 少なくとも1種のバッファー剤をさらに含み、水溶液であり、該少なくとも1種のバッファー剤の濃度が生理学的に適したpHを維持するのに効果的である、請求項24に記載の医薬製剤。
- 前記シクロデキストリンが、約5%w/vまたは約6%w/vから、約30%w/vまでの濃度で存在する、請求項24または25に記載の医薬製剤。
- 前記シクロデキストリンが、約6%w/vから約10%w/vの濃度で存在する、請求項26に記載の医薬製剤。
- 凍結乾燥用保護剤をさらに含む、請求項24から27のいずれか一項に記載の医薬製剤。
- 前記少なくとも1種の凍結乾燥用保護剤が糖である、請求項28に記載の医薬製剤。

- 請求項25から59のいずれか一項に記載の水性製剤へ再構成することができる、安定化された凍結乾燥抗体−薬物結合体製剤。
- 抗体に付いたベンゾジアゼピン薬物リンカーの化学的分解および断片化を防止する方法であって、少なくとも約6%w/vのガンマシクロデキストリンまたは化学修飾されたベータシクロデキストリンを用いて、該抗体に付いたベンゾジアゼピン薬物リンカーを製剤化する工程を含む、方法。
- 前記製剤中に存在するベンゾジアゼピン薬物リンカーの化学的分解の低減によって測定したとき、2%w/vまたはそれ未満のシクロデキストリンを有するベンゾジアゼピン薬物リンカーと比較して、改善された安定性を有する、請求項23から59のいずれか一項に記載の製剤。
