Description
[0001] 本出願は、MODELING THE BRAIN DYNAMICS IN NORMAL AND DISEASED STATESという表題の2013年1月31日に出願された米国仮特許出願公開第61/759,363号に対する優先権を主張し、その内容全体が、全ての目的で参照により本明細書に援用される。

1)細胞型特異性を用いたげっ歯類におけるオプトジェネティックfMRIにより、VNS作用機序を同定することと、
2)別個の発作型を発生させ、各型に関してVNSの有効性を評価して、発作VNSのいずれの発作型が有効であるかを同定し、各発作型に関する特有のEEGの形跡を同定することと、
3)EEGおよび発作と同期されたMRIにより患者におけるEEGの形跡を同定し、VNSと同期されたfMRIを行って、VNS作用機序を確認することと
を提供する。
[0092] てんかんは、高い有病率の支障を来す神経障害であり、世界で26人のうち1人が罹患しており、そのうちのおよそ3分の1が、既存の治療法に対して不耐性または不応性である。さらに、その不均一な病態生理学、ならびに発作オンセットおよび伝播に関連付けられる直接的な神経経路を現在は観察することができないことが、より有効な治療の開発に特に難題を課している。本発明のオプトジェネティック機能的磁気共鳴画像法(ofMRI)を利用して、発作は、細胞型、位置、時間的発火パターンおよびオンセット時間を含むそれらの起源の正確な知識により初めて発生する一方で、関連する全脳下流伝播ネットワークが同定される。シータ周波数を用いた背側海馬でのCAMKIIa発現ニューロンの特異的な刺激は、主に海馬体に制限される短期持続期間の血行動態応答をもたらす。より高いガンマ周波数での刺激は、中隔を含む脳のより広範囲の野で刺激よりも長く続く、より長い持続期間の血行動態応答を生じる。シータ刺激のビデオ脳波(EEG)モニタリングが、誘起されたEEG応答を示す一方で、ガンマ刺激は、より長い持続期間の血行動態応答に相当するエレクトログラフ発作を誘導する。しかしながら、行動発作は、背側海馬を刺激することからは誘導されない。対比して、腹側海馬でのCaMKIIa発現ニューロンのガンマ周波数刺激は、行動発作を付随するエレクトログラフ発作をもたらす。相当するofMRIにより同定されるネットワークは、前面皮質野の関与を示す。重要なことに、これらの観察は、ofMRIの、多重シナプスにわたる全脳動態ネットワーク機能を明らかにする能力を示している。臨床上のてんかんは、より複雑なプロセスを表し得る一方で、正確な起源の繰り返し可能な発作伝播経路の直接的な可視化は、発作に関与するネットワークを系統的に精査して、投薬、局所手術および神経刺激に関する標的とされる治療戦略の設計を可能にする前例のない機会を提供する。
オプトジェネティックfMRIデータ獲得および解析
[00103] 麻酔をかけた(およそ1.0〜1.5%イソフルラン)動物を473nmレーザーに結合して、7T小動物専用MRIスキャナーを使用してグラジエントリコールエコー(GRE)BOLD画像配列によりスキャンしながら刺激した。獲得したデータを、750ms毎に再構築させて、動きを補正して、平均化のために共通の選択画像に整列させた。続いて、平均化したデータセットを、刺激に同期的な周波数構成要素の有意性をコンピュータ計算する周波数ドメイン解析により解析した。
[00104] ofMRI研究後に、各げっ歯類被検体に頭蓋骨EEG電極を移植した。次に、動物を順化させて、3台のカメラを有するダイヤモンド形状の箱で試験して、行動を記録した。全ての実験において、被検体は、ベースライン獲得の5分後に、データ獲得の開始から5、10および15分で20または40秒間刺激した。EEGシグナルは、1kHzのサンプリングレートで記録した。6、10、20、40Hz刺激を有する5つの異なるタイプの施行を実施した。

[00107] pAAV−CaMKIIα−ChR2(H134R)−EYFPプラスミドは、MlulおよびEcoRI制限部位を使用して、pAAV−CaMKIIα−ChR2(H134R)−EYFPをAAV主鎖中にクローニングすることにより構築した。マップは、www.optogenetics.org.にてオンラインで入手可能である。組換えAAVベクターは、AAV5コートタンパク質を用いて血清型決定して、ノースカロライナ大学ウイルスベクターコアによりパッケージングした。力価は、2×10個の粒子/mLであった。
[00108] AAV5−CamKIIa−ChR2−EYFPウイルスベクターは、Deisseroth研究室から入手して、ノースカロライナ大学ベクターコアでパッケージングして、−80℃で保管した。AAV5は、それが順行性標識を生成して、注射の部位で細胞体を有するニューロンを選択的に標的とするため選択した。CamKIIαプロモーターを使用して、CamKIIα発現興奮性ニューロンにおいてのみChR2を生産した一方で、EYFPを使用して、検証のためChR2発現を共局在化させた。スプラーグドーリーラット(11週齢を超える、250〜350g)を被検体として使用して、動物畜産および実験操作は、国立衛生研究所およびUCLA動物実験委員会(IACUC)からのガイドラインに厳密に従った。ラットは、誘導チャンバー中で5%イソフルランにより麻酔をかけて、手術中にノーズコーンを用いて2〜3%で維持した。動物を定位フレームおよび加熱パッド上へ固定した後、人工涙液を目に垂らして、手術中の乾燥を防いだ。疼痛緩和のため、ブプレノルフィン(0.05mg/kg)を皮下注射した。頭部を剪毛して、70%エタノールおよびベタジンで消毒して、感染の可能性を最低限に抑えた。正中切開後、定位フレームにマウントした歯科用ドリルを使用して、小さな開頭術を行った。2つのタイプの手術を行った:I)背側CA1(+1.2mm ML右半球、−4.3mm AP、−3.6mm DVで注射)におけるウイルス注射(ラット1匹当たり注射1回、2μl/部位)およびカニューレ(3.4mm突出)移植、II)腹側CA1(+5.2mm ML右半球、−5.8mm AP、−4.6mm DVで注射)におけるウイルス注射(ラット1匹当たり注射1回、2μl/部位)およびカニューレ(4.4mm突出)移植。MRIおよび免疫組織化学により、ウイルス発現および刺激位置の精度を検証した。誤ったプローブ位置を有する動物は排除した。濃縮ウイルスを、10μlシリンジおよび34ゲージ金属針(World Precision Instruments Inc.)を使用して、マイクロシリンジポンプ制御器により駆動される流速150nl/分で送達した。シリンジ針を適所に10分間残した後、ゆっくりと引き抜いた。ベースから所望のDV座標へ伸長する光ファイバーを伴う特注設計した光ファイバーカニューレを、頭蓋骨上に取り付けて、凝固するまで(10分)メタボンド(Parkell Inc.)の一層を使用して固定した。5−0ナイロン皮膚縫合を行うことで、切開を縫合して閉じて、麻酔から回復するまで(通常、約15分)動物を加熱パッド上に保持した。術後の不快感を最低限に抑えるために、ブプレノフィンを一日二度、術後48時間、皮下注射した。
[00109] fMRIスキャニングは、ULCAにあるAhmanson−Lovelace Brain Mapping Centerにおいて、7T Broker Biospec小動物専用MRIシステムで実施した。動物に初めに、5%イソフルランを有する誘導チャンバー中で麻酔をかけて、挿管した後、特注のMRI適合性クレードル上へ配置させて、耳および歯を固定した。外径39mm内径25mmの特注設計した送信/受信単一ループ表面コイルを、頭蓋骨上の関心対象の領域にわたって中央に置いた。コア径62.5μmの光ファイバーを、青色レーザー供給源(473nm)へ接続して、光ファイバーカニューレと連結させた。fMRIスキャニング中、動物は、換気装置(Harvard Apparatus、モデル683小動物換気装置)を使用して浅麻酔下で人工的に換気(45〜55ストローク/分)して、O2(35%)、N2O(63.5%)およびイソフルラン(1.5%)の混合物で気化器を較正しながら、磁石のアイソセンターへ配置させた。呼気CO2を3〜4%に保って、加熱気流を使用して、体温を36〜38℃で維持した。T2荷重高分解能解剖学的画像をfMRIスキャニング前に獲得して、脳損傷およびプローブ配置に関してチェックした。グラジエントリコールエコー(GER)BOLD法を使用して、種々の光学刺激パラメーターを受けてfMRI画像を獲得した。1回60秒サイクルの刺激は、30%デューティサイクルによる所望の周波数での光学刺激20秒、および光学刺激なし40秒を含んでおり、実験1回当たり6回のサイクルを可能にした。刺激は、6、10、20、40および60Hzの周波数で実施した。光ファイバー出力をおよそ2.5mWに設定し、これは、対照実験では任意の加熱関連人工産物を誘導しなかった。
[00110] グラジエント−エコー(GRE)−BOLD配列を獲得に使用した。パルス配列は、面内視野(FOV)35×35mm2、空間分解能0.5×0.5×0.5mm3、および反復時間毎(750ms)に画像をアップデートすることができるスライドウィンドウ再構築を有するように設計した。GRE−BOLD fMRIは、二次元であり、マルチスライスであり、4つのインターリーブスパイラル読み取りを伴うグラジエントエコー配列であるように設計した;反復時間(TR)750msおよびエコー時間(TE)12msは、11.5mmのスライス方向容量を網羅する23個のスライスを生じる。この特異的な設計により、オプトジェネティック制御中に脳の大容量マッピングが可能となった。特注設計したGPUベースの平行再構築および動き補正の完了時に、周波数ドメイン解析が実施され、コヒーレンス値が算出された。特注のアルゴリズムおよびソフトウェア(MRVista)を使用して、コヒーレンス値閾値0.35を受けて、活性化マップを獲得した。デジタル標準ラット脳アトラスを使用して、移植したカニューレの位置を確認して、特異的脳領域へ活性化ボクセルを局在化させた。
[00111] ofMRI GRE BOLDスパイラルkスペースサンプルをまず、2次元(2D)グリッディング再構築法により再構築させた。背側および腹側CA1実験の両方に関して、最終的なfMRI画像は、6匹の動物における8回の独立した実験(それぞれが6回のスキャンを含有する)から平均化される。精密な平均化に関して、画像は全て、誤整列が存在しないように登録した。第1の工程として、他の画像が登録するのに必要とされる移行および回転が最小限に保持されるように、正中位置を有する画像を選択した。次に、選択した実験データの3D時間枠の1つを抽出して、登録用に実験間画像変換行列計算用の鋳型として使用した。特注開発されたMatlabツールをまず使用して、登録されるべき各実験の選択3D画像を、選択した鋳型画像へ変換することができる6自由度行列を決定した。続いて、別の特注のGPUベースの平行動き補正ツールを使用して、実験間登録に関して実際の変換を行い、続いて各実験の時系列および6つの反復スキャン内で動きに関して補正した。全ての画像を整列させた後に、それらを平均化して、最終的なfMRI4D画像をもたらした。次に、周波数ドメイン解析によりデータセットを解析した。
[00112] ofMRI研究の完了後、動物に、EEG電極移植に関するさらなる手術を施した。動物は、誘導チャンバー中で5%イソフルランにより麻酔をかけて、手術中にノーズコーンを用いて2〜3%で維持した。動物を定位フレームおよび加熱パッド上へ固定して、人工涙液を垂らして、手術中の乾燥を防ぎ、ブプレノルフィン(0.05mg/kg)を皮下注射した。ラットの頭部を剪毛した。70%エタノールおよびベタジンを消毒薬として裸の頭皮へ適用した。滅菌メスを用いて正中切開を行い、頭蓋骨上の再生した膜を滅菌綿棒で除去した。2つのステンレス製のねじ(0−80、直径1.5mm、Plastics One Inc.)をおよそ2.0cmの絶縁線(30ゲージ、R30Y0100、Wire Wrapping Wire, O.K. Industries)に取り付けて、前頭大脳皮質上で頭蓋骨へ付けた。一方の電極を、参照用にブレグマの前方およそ3.0mmおよび右に2.0mmに配置させ、他方の電極を、海馬EGG記録用に、I)背側CA1またはII)腹側CA1上の大脳皮質の縁に、各動物における光ファイバー移植位置に対して尾側およそ1.5mmで配置させた。電極を頭蓋骨に取り付けた後、それらを凝固するまでメタボンド(Parkell Inc.)の一層を使用して固定して、およそ10分を要した。5−0ナイロン皮膚縫合を行うことで、切開を閉じた。およそ15分で麻酔から回復するまで、動物を加熱パッド上に保持した。疼痛を最低限に抑えるために、ブプレノフィンを12時間毎に、術後48時間、皮下注射した。動物を一週間、不快感または感染に関して厳密に観察して、それらの水の中にはトリメトプリム−スルファメトキサゾール抗生物質(48mg/100ml)を供給した。
[00113] 各端に、および上部上にカメラ取り付けたダイヤモンド形状の箱に、ラットを順化させた。この形状により、箱内のカマー(comer)をより鈍角にさせ、動物を正面から可視化するのを助長する。注意散漫となるものはケージに配置させなかった。動物には、試行の間、食物および水を与えた。EEG記録は、ベースラインの5分間、続いて各試行が20分間続くように、5、10および15分での20または40秒のオプトジェネティック光刺激(間での4:20の刺激なし)を実施した。チャネル1つ当たり毎秒1000ポイントでサンプリングする市販のデジタルEEGシステムを使用して、EEGを記録した。刺激周波数の種々のパラメーターは、無作為の試行順序で試験した。5つの試行(6、10、20、40および偽)を含有する試験を、各動物について三度繰り返した。偽実験は、レーザーの力を、脳中で2桁分、光を減衰させるレベルに低減させて、したがって任意の影響を無視し得るものにしたが、動物および研究者らに目に見える鮮やかな青色閃光を生み出すことが依然として可能であった。EEG記録中に、3つのビデオカメラは全て、動物の行動を記録した。ofMRIは、O2(およそ35%)、N2O(およそ63.5%)およびイソフルラン(1.5%)の吸入で麻酔をかけたラットで記録されたため、ビデオ−EEGもまた、この混合物を吸入している3匹の動物における実験で解析した。

てんかんに関する神経刺激の変換
1.課題、革新および影響評価:
[00117] 深部脳刺激(DBS)は、パーキンソン病、振戦、うつ病、疼痛、てんかんおよび多くの他のものを含む脳障害に関して潜在的に有用な新たな治療法である。しかしながら、DBSメカニズムの初歩的な理解が有効性を制限する。DBSは、局所組織を阻害または励起させて、ネットワーク同期性を崩壊させて、通路の線維を活性化させて、不確かな遠隔作用を発生させることができる。最良の標的は必ずしも既知ではないが、使用される標的は、刺激に対するそれらの応答では不均一である。臨床試験における刺激パラメーターは、大部分が推量である。新たな技術であるオプトジェネティック機能的磁気共鳴画像法(ofMRI)は、これらの制限を克服する立場にある。ofMRIは、神経学的状態に関与する特異的脳ネットワークを表示することができ、脳の全3D空間におけるニューロン活動を可視化することができる。オプトジェネティクスは、光の制御可能かつ反復可能なパルスにより、特異的ニューロン集団を励起または阻害させることができる。ofMRIを使用して、脳刺激のメカニズムを精査することが可能であり、得られた洞察を、電気脳刺激に対する合理的アプローチを設計するのに用いる。例としててんかんを使用して、海馬から伝播する発作の制御可能なモデルを開発して、伝播ネットワークを通じてこれらの発作を追跡または阻害し、オプトジェネティックマッピングにより発見される小領域およびパラメーターへ電気刺激を仕立ててもよい。
[00118] てんかん用のDBSは、治療的DBSの見込みおよび制限の実例として解することができる。投薬抵抗性てんかんに関する脳刺激の2つの近年の多施設無作為対照試験(1つは本出願人により導かれる)は、統計学的に有意な有益性を示したが、ほんの約40%の発作の中央値の低減を有したに過ぎなかった(図13)。視床前核(ANT)は、ほとんどの発作が由来する刺激の前面および近心側頭領域に影響を与えるその能力のため、試行の1つにおいて刺激の標的として使用された。有益性の度合いは、刺激のパラメーターに非常に依存した。例えば、1人の患者は、5Vの刺激が6分サイクルで活性化されるたびに、発作が誘発されたが、続いて4Vの刺激による発作では、実質的な低減が見られた。刺激の最良のパラメーターが既知である場合に、どれくらいより多くの有益性が実現されたのであろうか?最良の電圧(または電流)、刺激周波数、パルス幅、両極性対参照刺激、連続的、間欠的または応答性刺激とは何であろうか?最も重要なことには、最良の標的とは何であろうか?ターゲッティングは、電極を配置させるための全般的な領域だけではなく、通路の線維の活性化を伴って、または伴わずに、特異的細胞集団を刺激するための努力を包含する。例えば視床下核の刺激は、皮質−視床下線維の逆行性活性化により発作を阻害するとされてきた。ANTSの電気刺激は、局所ニューロンだけでなく、乳頭体視床束(mammillothalamic tract)、セロトニン作動性、コリン作動性およびグルタミン酸作動性の通路の線維もまた活性化する。

[00124] オプトジェネティクスは、単一構成要素の微生物光活性化膜コンダクタンス制御因子が、細胞型特異的プロモーターを使用して特異的に標的とされる細胞型および回路要素へ導入されて、in vivoでミリ秒スケールの標的活動調節を可能にする革命的な神経調節技術である。チャネルロドプシン(ChR2)は、470nmの青色光への曝露後にNa+イオンが細胞に入るのを可能にする一価陽イオンチャネルであるのに対して、ハロロドプシン(NpHR)は、580nmの黄色光による照射時に活性化するクロライドポンプである。これらの2つのタンパク質の最適活性化波長が100nm以上離れているため、それらは、活動電位発火(firing)を開始するために、または無傷組織において神経活動を抑止するために独立して制御することができ、一緒にニューロン同期性を調節し得る。両方のタンパク質が、ミリ秒のスケールで迅速な時間的キネティクスを有し、ChR2を使用して、in vivoで信頼性ある一連の高周波数活動電位を駆動させて、NpHRを使用して一連の高周波数スパイク内で単一活動電位を抑止することが可能である。これまでは、神経科学における最大の課題の1つは、脳の種々の回路要素を選択的に制御して、その機能を理解することの難しさであった。オプトジェネティクスは、遺伝的に標的とされる回路要素のin vivo制御を可能にすることにより、各回路要素の系統的寄与の研究を可能にする。

[00166] てんかんならびに多くの他の神経学的疾患のための薬物開発における重要な課題は、化合物の将来の臨床的有効性および副作用の精密な前臨床予測性である。神経系の複雑さにより、in vitroでの情報、ex vivoでのデータまたは単純な動物行動モデルにおいて代理を使用して、候補化合物の影響を推論することが極めて困難となっている。疾患メカニズムの正確な先験的知識を用いずに、薬物の有効性を正確に試験するのに使用することができる疾患モデルを作製することは困難である。本発明者らの提唱は、オプトジェネティック機能的磁気共鳴画像法(ofMRI)技術を利用して、薬物投与の影響のin vivoでの完全脳定量化とともに精密な疾患モデル作製を通じてこのプロセスを基本的に変革することを目的とする。ofMRIは、オプトジェネティクスと呼ばれる、細胞型特異的な時間的に正確な刺激方法を、fMRI読み出しと組み合わせる新規技術である。PIにより出版された概念研究の最初の証拠(Nature 2010)は、その遺伝的同一性、細胞体位置、軸索突出標的および時間的シグナル伝達パターンにより特定される細胞型に起因する脳全体にわたる(brain-wide)神経活動を正確に可視化するofMRI技術の能力を実証した。
[00170] 3つの市販の抗てんかん薬(AED):フェニトイン(PHT)、ロラゼパムおよびレベチラセタム(LEV)が使用され得る。それらは、それらの別個の前臨床抗けいれん効果および別個の薬物作用機序に関して選択した。さらに、それらは、最大電気ショック(MES)、全身性ペンチレンテトラゾール(PTZ)および扁桃体キンダリングのような共通てんかんモデルにおける有効性の種々のプロフィールを示す。PHTは、Na+チャネルの不応期に影響を及ぼし、ロラゼパムは、GABA作動性効率に影響を及ぼし、LEVは、新規シナプス前A2タンパク質に影響を及ぼす。ロラゼパムは、3つの従来モデル全てにおいて有効性を示すのに対して、LEVは、扁桃体キンダリングにおいてのみ有効性を示した。臨床上、PHTおよびロラゼパムは、部分的および全身性強直−間代発作における有効性を示す一方で、LEVは、部分的な強直−間代および非けいれん性全身性発作に関して有効性を示す。種々の薬物は、種々の動物モデルに関する有効性における明らかな差を示すが、臨床的有効性は、動物モデル研究により必ずしも予測されるわけではない。さらに、MESおよびPTZモデルのみが使用される場合、LEVは、病院に到達しない可能性が高い。したがって、病因へ予測性を提供し得る標的てんかん患者群を表す適正な動物モデルを利用することが、非常に重要である。このため、本発明者らは、動物モデルが提供する発作の精密な理解、ならびに薬物が動物モデルにおいて影響を与える包括的読み出しを有する必要がある。
[00179] 本発明者らは、同じ動物に対して繰り返される実験を実施して、各薬物の少なくとも5倍の半減期を実験用の種々の薬物間で通過することが可能である(例えば、1週を上回る)。本発明者らはまた、発作閾値を評価して、経時的に著しい「キンドリング」効果を除外する。同じ動物に対して実験を繰り返す能力は、薬物実験用のモデルの将来の魅力を大いに高める。ofMRI研究を行って、信頼性を考慮する本発明者らの経験、およびin vivoで正確な再現性に起因して同じ動物において連続実験を実施する能力、非侵襲的画像能力から、本発明者らは、1群当たりたった5匹の動物を必要とする。2つのモデルを用いる場合、このことは、総計10匹の動物につながり、消耗を前提としない。10匹の動物を使用して、まず、発作伝播のofMRIが、図28、図29に見られるようにマップに精密に示す。次に、同じ動物において、薬物を投与する。図28、図29に示されるように脳領域全てを分割して、また薬物注射前および後の間の差の統計学的有意性に注目して、本発明者らは、薬物がどのようにして、各脳領域に影響を及ぼすかを解析する。さらに、2つのモデルに関する薬物有効性の差は、全体的な活動容量がどれくらい変化したか、ならびにどの脳領域の活動が変化したかに関する具体的な特徴により評価する。概要すると、注射前および後の2つのモデルおよび3つの薬物は、2つのモデルに関して注射前に関する2つのデータセット、2つの発作モデルにおける3つの薬物の注射後に関する6つのデータセットを付与する。8つの事例が、総計10匹の動物における局所データの統計学的比較を通じて比較される。本発明者らはまた、同じ動物において3度、同じ実験条件の繰り返し実験を行い、統計学的パワーを増す。このことは、3回の繰り返しで総計5匹を付与し、たった10匹の動物を用いた場合のシナリオ1回(6つの事例)当たり15回の試行をもたらす。本発明者らは、薬物治療の有意性対分散の他の重要な原因を評価するために、局所活性化の連続的な可変性マーキング容量に対して、反復測定統計学によりANOVAを使用してデータを解析する。本発明者らはまた、薬物投与の前および後に活性化される領域対活性化されない領域のノンパラメトリック尺度に対するカイ二乗検定(または5よりも小さなビンサイズに関する比例差検定)を実施する。

Claims (21)
- 正常状態および罹患状態での脳動態をモデリングする方法であって、
細胞型特異性を用いたオプトジェネティック機能的磁気共鳴画像法(ofMRI)により、神経刺激の作用機序を同定する工程と、
別個の発作型を発生させ、いずれの発作型か同定するために各発作型に関して神経刺激の有効性を評価して、各発作型に関する特有の脳波(EEG)の形跡を同定する工程と、
EEGおよび発作と同期されたofMRIにより、患者におけるEEGの形跡を同定する工程と
を含む方法。 - 前記神経刺激が、迷走神経刺激(VNS)、深部脳刺激(DBS)および応答性神経刺激(RNS)のうちの1つである、請求項1に記載の方法。
- 神経刺激の作用機序を確認するために、神経刺激と同期されたofMRIを行う工程
をさらに含む、請求項1に記載の方法。 - 前記神経刺激が、迷走神経刺激(VNS)、深部脳刺激(DBS)および応答性神経刺激(RNS)のうちの1つである、請求項3に記載の方法。
- 正常状態および罹患状態での脳動態をモデリングする方法であって、
脳における罹患状態モデルを創出する工程と、
オプトジェネティック刺激および機能的磁気共鳴画像法(fMRI)画像に基づいて前記モデルを分類する工程と
を含む方法。 - 光応答性刺激中の分類に基づいて脳波(EEG)測定を解釈するために、さらなる測定を行う工程をさらに含む、請求項5に記載の方法。
- 前記さらなる測定が、EEGを使用して行われる、請求項6に記載の方法。
- 前記罹患状態モデルが、発作のモデルである、請求項5に記載の方法。
- 前記罹患状態モデルが、発作の動物モデルである、請求項5に記載の方法。
- 前記発作が、背側CA1刺激、中間CA1刺激、腹側CA1刺激または海馬台刺激の少なくとも1つから発生する、請求項9に記載の方法。
- 神経刺激および薬物設計に関して前記発作の前記動物モデルを使用する工程をさらに含む、請求項9に記載の方法。
- 正常状態および罹患状態での脳動態をモデリングする方法であって、
背内側、内側、腹内側および/または外側CA1領域において、錐体ニューロンをオプトジェネティック的に刺激する工程と、
オプトジェネティック機能的磁気共鳴画像法(ofMRI)を用いて、連関した脳ネットワークを通じて発作をモニタリングする工程と、
ofMRI活動の位置での局所電場電位および単一ユニット記録を測定する工程と
を含む方法。 - 前記オプトジェネティック的に刺激する工程が、阻害性介在ニューロンまたは通路の線維の直接的な刺激を起こすことなく遂行される、請求項12に記載の方法。
- 前記オプトジェネティック的に刺激する工程が、短期持続活動を発生するように20Hz未満の周波数刺激を利用して遂行される、請求項12に記載の方法。
- 前記オプトジェネティック的に刺激する工程が、短期持続活動を発生するように6〜10Hzの周波数刺激を利用して遂行される、請求項14に記載の方法。
- 前記オプトジェネティック的に刺激する工程が、20Hzを超える周波数刺激を用いて遂行される、請求項12に記載の方法。
- 前記オプトジェネティック的に刺激する工程が、長期持続活動を発生するように20〜60Hzの周波数刺激を利用して遂行される、請求項16に記載の方法。
- 前記オプトジェネティック的に刺激する工程が、40Hzの光刺激を用いて遂行される、請求項17に記載の方法。
- 錐体ニューロンを刺激または阻害するために、AAV5−CamKIIa−ChR2−EYFPまたはAAV5−CamKIIa−NpHR−mCherryの少なくとも1つを注射する工程と、
前記ニューロンをオプトジェネティック的に誘発するために、青色および黄色光の少なくとも1つを適用する工程と
をさらに含む、請求項12に記載の方法。 - 青色光が、毎秒1、10、60または100サイクルで送達される、請求項19に記載の方法。
- 青色光が、前記脳をスキャンしながら、毎秒1、10、60または100サイクルで、毎分20秒間で、総計6分間送達される、請求項20に記載の方法。
