Abstract
Description
(1)神経前駆細胞を神経栄養因子を含有する培養液中で培養する工程、
(2)(1)の工程で得られた細胞を解離させる工程、および
(3)(2)の工程で得られた細胞をコーティング処理されていない培養容器を用いて神経栄養因子を含有する培養液中で接着培養する工程。
本発明の他の態様は、前記神経栄養因子がGDNF、BDNFおよびNT-3から成るグループより選択される因子である、前記方法を提供することである。
本発明の他の態様は、次の工程をさらに含む、前記方法を提供することである。
(4)(3)の工程で得られた細胞を解離させる工程、ならびに
(5)(4)の工程で得られた細胞をコーティング処理されていない培養容器を用いてGDNF、BDNFおよびNT-3から成るグループより選択される因子を含有しない培養液中で接着培養する工程。
本発明の他の態様は、さらに、前記工程(5)で得られた細胞を解離させ、コーティング処理されていない培養容器を用いてGDNF、BDNFおよびNT-3から成るグループより選択される因子を含有しない培養液中で接着培養する工程を、少なくとも2回繰り返す、前記方法を提供することである。
本発明の他の態様は、次の工程をさらに含む、前記方法を提供することである;
(6)(5)の工程で得られた細胞を解離させる工程、ならびに
(7)(6)の工程で得られた細胞をゼラチンコートした培養容器を用いてGDNF、BDNFおよびNT-3から成るグループより選択される因子を含有しない培養液中で接着培養する工程。
本発明の他の態様は、前記工程(1)が66日以上の期間培養する工程である、前記方法を提供することである。
本発明の他の態様は、前記工程(3)が30日以上の期間培養する工程である、前記方法を提供することである。
本発明の他の態様は、前記工程(5)が20日以上の期間培養する工程である、前記方法を提供することである。
本発明の他の態様は、前記神経前駆細胞が、多能性幹細胞をBMP阻害剤およびTGFβ阻害剤を含有する培養液中で培養する工程を含む方法により製造される細胞である、前記方法を提供することである。
本発明の他の態様は、前記BMP阻害剤がDorsomorphinであり、前記TGFβ阻害剤がSB431542である、前記方法を提供することである。
本発明の他の態様は、前記多能性幹細胞から神経前駆細胞を製造する工程が、胚様体を形成後、接着培養させる工程を含む、前記方法を提供することである。
本発明の他の態様は、前記神経前駆細胞がヒト神経前駆細胞である、前記方法を提供することである。
本発明の他の態様は、前記方法で製造されたアストロサイトを提供することである。
本発明の他の態様は、ニューロンとアストロサイトが混在する細胞群よりアストロサイトを選択的に培養する方法であって、次の工程を含む方法を提供することである;
(I)細胞群を解離させる工程、および
(II)(I)の工程で得られた細胞をコーティング処理されていない培養容器を用いて培養する工程。
本発明の他の態様は、前記工程(II)の培養する工程が、神経栄養因子を含有する培養液中で接着培養する工程である、前記方法を提供することである。
本発明の他の態様は、次の工程をさらに含む、前記方法を提供することである;
(III)前記工程(II)で得られた細胞を解離させる工程、ならびに
(IV)前記工程(III)で得られた細胞をコーティング処理されていない培養容器を用いてGDNF、BDNFおよびNT-3から成るグループより選択される因子を含有しない培養液中で接着培養する工程。
本発明の他の態様は、さらに、前記工程(IV)で得られた細胞を解離させ、コーティング処理されていない培養容器を用いてGDNF、BDNFおよびNT-3から成るグループより選択される因子を含有しない培養液中で接着培養する工程を、少なくとも2回繰り返す、前記方法を提供することである。
本発明の他の態様は、前記工程(II)が30日以上の期間培養する工程である、前記方法を提供することである。
本発明の他の態様は、前記工程(IV)が20日以上の期間培養する工程である、前記方法を提供することである。
本発明の他の態様は、前記ニューロンとアストロサイトが混在する細胞群が、次の工程
により多能性幹細胞から製造された細胞群である、前記方法を提供することである;

本発明の神経前駆細胞を培養する工程において用いる培養液は、動物細胞の培養に用いられる培地を基礎培地として調製することができる。基礎培地としては、例えばIMDM培地、Medium 199培地、Eagle's Minimum Essential Medium (EMEM)培地、αMEM培地、Dulbecco's modified Eagle's Medium (DMEM)培地、Ham's F12培地、RPMI 1640培地、Fischer's培地、Neurobasal Medium(ライフテクノロジーズ)およびこれらの混合培地などが包含される。好ましくは、Neurobasal Mediumである。培地には、血清が含有されていてもよいし、あるいは無血清でもよい。必要に応じて、培地は、例えば、アルブミン、トランスフェリン、Knockout Serum Replacement(KSR)(ES細胞培養時のFBSの血清代替物)、N2サプリメント(Invitrogen)、B27サプリメント(Invitrogen)、脂肪酸、インスリン、コラーゲン前駆体、微量元素、2-メルカプトエタノール、3'-チオールグリセロールなどの1つ以上の血清代替物を含んでもよいし、脂質、アミノ酸、L-グルタミン、Glutamax(Invitrogen)、非必須アミノ酸、ビタミン、増殖因子、低分子化合物、抗生物質、抗酸化剤、ピルビン酸、緩衝剤、無機塩類などの1つ以上の物質も含有し得る。好ましい培地は、B27サプリメントおよびGlutamaxを含有するNeurobasal Mediumである。
Factor (NGF)、Brain-derived Neurotrophic Factor (BDNF)、Neurotrophin 3 (NT-3)、Neurotrophin 4/5 (NT-4/5)、Neurotrophin 6 (NT-6)、basic FGF、acidic FGF、FGF-5、Epidermal Growth Factor (EGF)、Hepatocyte Growth Factor (HGF)、Insulin、Insulin Like Growth Factor 1 (IGF 1)、Insulin Like Growth Factor 2 (IGF 2)、Glia cell line-derived Neurotrophic Factor (GDNF)、TGF-b2、TGF-b3、Interleukin 6 (IL-6)、Ciliary Neurotrophic Factor (CNTF)およびLIFなどが挙げられる。本発明において好ましい神経栄養因子は、GDNF、BDNFおよびNT-3から成るグループより選択される因子である。
細胞を解離させる工程においては、互いに接着して集団を形成している細胞を個々の細胞に解離(分離)させる。
細胞を解離させる方法としては、例えば、力学的に解離する方法、プロテアーゼ活性と
コラゲナーゼ活性を有する解離溶液(例えば、Accutase(商標)およびAccumax(商標)など)またはコラゲナーゼ活性のみを有する解離溶液を用いた解離方法が挙げられる。好ましくは、プロテアーゼ活性とコラゲナーゼ活性を有する解離溶液(特に好ましくは、Accutase(商標))を用いてヒト多能性幹細胞を解離する方法が用いられる。
コーティング処理されていない培養容器とは、当業者が汎用的に用いている細胞培養用ディッシュ、プレートまたはフラスコを意味し、その形状は特に限定されない容器であり、培養に際して、別途コーティング剤による処理工程を経ていない容器である。好ましくは、ポリスチレン製の培養容器である。コーティング剤としては、例えば、マトリゲル(BD)、コラーゲン、ゼラチン、ラミニン、ヘパラン硫酸プロテオグリカン、またはエンタクチンが挙げられ、本工程では、少なくともこれらのコーティング剤にて処理を行わない培養容器を用いることが好ましい。
本発明では、アストロサイトを製造するにあたり、さらに得られた細胞を解離させ、コーティング処理されていない培養容器を用いてGDNF、BDNFおよびNT-3から成るグループより選択される因子を含有しない培養液中で接着培養しても良い。細胞の解離は、上記と同様の方法を用いて行うことができ、プロテアーゼ活性とコラゲナーゼ活性を有する解離溶液を用いて行うことが望ましい。
F12培地、RPMI 1640培地、Fischer's培地、Neurobasal Medium(ライフテクノロジーズ)およびこれらの混合培地などが包含される。好ましくは、Neurobasal Mediumである。培地には、血清が含有されていてもよいし、あるいは無血清でもよい。必要に応じて、培地は、例えば、アルブミン、トランスフェリン、Knockout Serum Replacement(KSR)(ES細胞培養時のFBSの血清代替物)、N2サプリメント(Invitrogen)、B27サプリメント(Invitrogen)、脂肪酸、インスリン、コラーゲン前駆体、微量元素、2-メルカプトエタノール、3'-チオールグリセロールなどの1つ以上の血清代替物を含んでもよいし、脂質、アミノ酸、L-グルタミン、Glutamax(Invitrogen)、非必須アミノ酸、ビタミン、増殖因子、低分子化合物、抗生物質、抗酸化剤、ピルビン酸、緩衝剤、無機塩類などの1つ以上の物質も含有し得る。好ましいGDNF、BDNFおよびNT-3から成るグループより選択される因子を含有しない培養液は、N2サプリメントおよびGlutamaxを含有するDMEM/F12または血清およびGlutamaxを含有するDMEM/F12である。
上述した神経前駆細胞を神経栄養因子を含有する培養液中で培養する工程後には、アストロサイトに加えニューロンも同時に製造され得る。しかしながら、アストロサイトはニューロンなどと比較して、コーティング処理されていない培養容器に接着しやすいので、本発明の方法を用いると、アストロサイトとニューロンが混在する細胞群から高効率でアストロサイトを選択的に得ることができる。従って、本発明は、アストロサイトとニューロンが混在する細胞群よりアストロサイトを選択的に培養する方法を提供する。詳細には、上述した細胞を分離する工程およびコーティング処理されていない培養容器を用いて培養する工程を含む方法である。
本発明の方法で製造されたアストロサイトは、ゼラチンコートをした培養容器を用いて、上述したGDNF、BDNFおよびNT-3から成るグループより選択される因子を含有しない培養液中で培養することができる。
本発明において、神経前駆細胞は、生体内から単離された神経前駆細胞でもよく、in vitroで他の細胞種より誘導された細胞でもよいが、好ましくは多能性幹細胞から分化誘導される神経前駆細胞である。
al. Neuron. 28:31-40, 2000)、(3)マトリゲル上に薬剤を添加して培養する方法(Chambers SM, et al. Nat Biotechnol. 27:275-80, 2009)などが挙げられる。多能性幹細胞から神経前駆細胞への好ましい分化誘導方法は、多能性幹細胞をBMP阻害剤およびTGFβ阻害剤を含有する培養液中で培養する工程を含む方法を用いることができる。

本発明で使用可能な多能性幹細胞は、生体に存在する全ての細胞に分化可能である多能性を有し、かつ、増殖能をも併せもつ幹細胞であり、それには、例えば胚性幹(ES)細胞、核移植により得られるクローン胚由来の胚性幹(ntES)細胞、精子幹細胞(「GS細胞」)、胚性生殖細胞(「EG細胞」)、人工多能性幹(iPS)細胞、培養線維芽細胞や骨髄幹細胞由来の多能性細胞(Muse細胞)などが含まれる。本発明では、胚の破壊を行わずに得られるという意味では、iPS細胞またはMuse細胞を用いることが好ましい。
ES細胞は、ヒトやマウスなどの哺乳動物の初期胚(例えば胚盤胞)の内部細胞塊から樹立された、多能性と自己複製による増殖能を有する幹細胞である。

精子幹細胞は、精巣由来の多能性幹細胞であり、精子形成のための起源となる細胞である。この細胞は、ES細胞と同様に、種々の系列の細胞に分化誘導可能であり、例えばマウス胚盤胞に移植するとキメラマウスを作出できるなどの性質をもつ(M. Kanatsu-Shinohara et al. (2003) Biol. Reprod., 69:612-616; K. Shinohara et al. (2004), Cell, 119:1001-1012)。神経膠細胞系由来神経栄養因子(glial cell line-derived neurotrophic factor (GDNF))を含む培養液で自己複製可能であるし、またES細胞と同様の培養条件下で継代を繰り返すことによって、精子幹細胞を得ることができる(竹林正則ら(2008),実験医学,26巻,5号(増刊),41〜46頁,羊土社(東京、日本))。
胚性生殖細胞は、胎生期の始原生殖細胞から樹立される、ES細胞と同様な多能性をもつ細胞であり、LIF、bFGF、幹細胞因子(stem cell factor)などの物質の存在下で始原生殖細胞を培養することによって樹立しうる(Y. Matsui et al. (1992), Cell, 70:841-847; J.L. Resnick et al. (1992), Nature, 359:550-551)。
人工多能性幹(iPS)細胞は、特定の初期化因子を、DNA又はタンパク質の形態で体細胞に導入することによって作製することができる、ES細胞とほぼ同等の特性、例えば分化多能性と自己複製による増殖能、を有する体細胞由来の人工の幹細胞である(K. Takahashi and S. Yamanaka (2006) Cell, 126:663-676; K. Takahashi et al. (2007), Cell, 131:861-872; J. Yu et al. (2007), Science, 318:1917-1920; Nakagawa, M.ら,Nat. Biotechnol. 26:101-106 (2008);国際公開WO 2007/069666)。初期化因子は、ES細胞に特異的に発現している遺伝子、その遺伝子産物もしくはnon-coding RNAまたはES細胞の未分化維持に重要な役割を果たす遺伝子、その遺伝子産物もしくはnon-coding RNA、あるいは低分子化合物によって構成されてもよい。初期化因子に含まれる遺伝子として、例えば、Oct3/4、Sox2、Sox1、Sox3、Sox15、Sox17、Klf4、Klf2、c-Myc、N-Myc、L-Myc、Nanog、Lin28

、酵母人工染色体(YAC)、細菌人工染色体(BAC、PAC)などが含まれる。プラスミドとしては、哺乳動物細胞用プラスミドを使用しうる(Science, 322:949-953, 2008)。ベクターには、核初期化物質が発現可能なように、プロモーター、エンハンサー、リボゾーム結合配列、ターミネーター、ポリアデニル化サイトなどの制御配列を含むことができるし、さらに、必要に応じて、薬剤耐性遺伝子(例えばカナマイシン耐性遺伝子、アンピシリン耐性遺伝子、ピューロマイシン耐性遺伝子など)、チミジンキナーゼ遺伝子、ジフテリアトキシン遺伝子などの選択マーカー配列、緑色蛍光タンパク質(GFP)、βグルクロニダーゼ(GUS)、FLAGなどのレポーター遺伝子配列などを含むことができる。また、上記ベクターには、体細胞への導入後、初期化因子をコードする遺伝子もしくはプロモーターとそれに結合する初期化因子をコードする遺伝子を共に切除するために、それらの前後にLoxP配列を有してもよい。
剤耐性遺伝子をマーカー遺伝子として導入した場合は、対応する薬剤を含む培養液(選択培養液)で培養を行うことにより樹立したiPS細胞を選択することができる。また、マーカー遺伝子が蛍光タンパク質遺伝子の場合は蛍光顕微鏡で観察することによって、発光酵素遺伝子の場合は発光基質を加えることによって、また発色酵素遺伝子の場合は発色基質を加えることによって、iPS細胞を選択することができる。
nt ES細胞は、核移植技術によって作製されたクローン胚由来のES細胞であり、受精卵由来のES細胞とほぼ同じ特性を有している(T. Wakayama et al. (2001), Science, 292:740-743; S. Wakayama et al. (2005), Biol. Reprod., 72:932-936; J. Byrne et al. (2007), Nature, 450:497-502)。すなわち、未受精卵の核を体細胞の核と置換することによって得られたクローン胚由来の胚盤胞の内部細胞塊から樹立されたES細胞がnt ES(nuclear transfer ES)細胞である。nt ES細胞の作製のためには、核移植技術(J.B. Cibelli et al. (1998), Nature Biotechnol., 16:642-646)とES細胞作製技術との組み合わせが利用される(若山清香ら(2008),実験医学,26巻,5号(増刊), 47〜52頁)。核移植においては、哺乳動物の除核した未受精卵に、体細胞の核を注入し、数時間培養することで初期化することができる。
Muse細胞は、WO2011/007900に記載された方法にて製造された多能性幹細胞であり、詳細には、線維芽細胞または骨髄間質細胞を長時間トリプシン処理、好ましくは8時間または16時間トリプシン処理した後、浮遊培養することで得られる多能性を有した細胞であり、SSEA-3およびCD105が陽性である。
本発明において、上述した分化誘導法によって作製されたアストロサイトは、例えば、GFAPなど任意のマーカーによって染色することで同定し、当業者に周知の方法で純化され得る。

本発明は、神経前駆細胞からアストロサイトを分化誘導するためのキットを提供する。本キットには、上述した分化誘導に用いる増殖因子、化合物、培養液、解離溶液および培養容器のコーティング剤を含んでもよい。本キットには、さらに分化誘導の手順を記載した書面や説明書を含んでもよい。
2名の健常者より同意を得て3 mmの皮膚生検で得られた外植片からヒト皮膚線維芽細胞(HDF)を樹立した。樹立したHDFへOkitaらの方法に従ってエピソーマルベクターを用いて、ヒトcDNA(SOX2、KLF4、OCT4、L-MYC、LIN28)およびp53のshRNAを導入した(Okita et al.,
Nat Methods. 2011, 8, 409-412)。導入の数日後、HDFを回収し、SNLフィーダー細胞層上に再播種した。翌日、培地を4 ng/ml bFGF (Wako Chemicals, Osaka, Japan)を補充した霊長類胚性幹細胞用培地(Reprocell, Kanagawa, Japan)で置換した。一日おきに培地を交換した。cDNAの導入から30日後、各iPS細胞コロニーを1つ選択した(N116213およびN117322)。さらに、Okitaらが樹立した細胞を受領し(409B2)、計3種のiPS細胞を本実施例に用いた。
1.神経前駆細胞の誘導
上記の方法で得られたiPS細胞と共培養しているフィーダー細胞をCTK溶液(ReproCell)を用いて選択的に剥離、さらに残ったiPS細胞をAccutase(Innovative Cell Technologies)により解離した。解離したiPS細胞を、2μM Dorsomorphin(Sigma-Aldrich)および10μM SB431542(Cayman Chemical)を添加したDFK5%培地(5%KSR(Invitrogen)、L-glutamine(Sigma-Aldrich)および0.1M 2-mercaptoethanol(Invitrogen)を添加したDMEM/Ham’s F12(Gibco))に懸濁、さらに2% Pluronic F-127(Sigma-Aldrich)ethanol溶液でコートしたU-bottom 96-wellプレートに播種し胚様体(EB)を形成させ、浮遊培養により8日間培養した。続いて、得られたEBを マトリゲル(BD)でコートした6-well プレートへ移し、1x N2 supplement(Invitrogen)、2μM Dorsomorphinおよび10μM SB431542を添加したDFK5%培地にて接着培養により16日間培養し(通算24日間)、神経前駆細胞を得た。
上記の方法で得られた神経前駆細胞をAccutase(Innovative Cell Technologies)により解離させ、マトリゲルコートした12-wellプレートを用いて、1x B27 without Vitamin A(Invitrogen)、1x Glutamax(Invitrogen)、10ng/ml BDNF、10ng/ml GDNFおよび10ng/ml NT-3を添加したNeurobasal medium(Invitrogen)にて接着培養により66日間培養した(通算90日間)。続いて、得られた細胞をAccutaseを用いて解離させ、何もコートしていない6-cmディッシュへ移し、1x B27 without Vitamin A、1x Glutamax、10ng/ml BDNF、10ng/ml GDNFおよび10ng/ml NT-3を添加したNeurobasal mediumにて接着培養により30日間培養した(通算120日間)。この時、接着しなかった細胞はアノイキスにより死滅した。この接着した細胞をAccutaseにより解離させ、何もコートしていない6-cmディッシュへ移し、1x N2 supplementを添加したDMEM/F12, Glutamax(Invitrogen)にて、接着培養により30日間培養した(通算150日間)。さらに、2度得られた細胞を解離させ同様の条件にて30日間培養し、アストロサイトを得た(通算200日間)。続いて、グルタミン取り込み試験に用いる場合は、得られた細胞を解離させ、0.1%ゼラチンコートした6-cmディッシュへ移し、1x N2 supplementを添加したDMEM/F12, Glutamaxにて培養した。以上の工程を図1に示す。
1.Microarray分析
上述の方法で得られたアストロサイト(通算90日目および200日目)およびプライマリーアストロサイト(Lonza)からRNeasy mini kit(Qiagen)を用いて全RNAを取り出し、Ovation Pico WTA System/ Encore Biotin Module kit(NuGENE)を用いて断片化およびビオチン付加cDNAへ変換した。このcDNAサンプルをGeneChip Human Gene 1.0 ST Array(Affymetrix)を用いてハイブリダイズさせ、G2565BA Microarray Scanner System(Agilent)によりスキャンした。スキャンしたデータはGeneSpring GX7.3.1 software(Agilent)により分析した。このように分析されたクラスター解析の結果を図2に示す。この結果より、上述のアストロサイトの誘導方法において通算90日目よりも200日目の方がよりプライマリーアストロサイトに近いことが確認された。
上述の方法で得られたアストロサイトは、4%パラホルムアルデヒド(pH 7.4)で30分間室温静置し固定した。0.2% Triton X-100を含むPBSにて洗浄し、10% donkey serumを含むPBSでブロッキングした。GFAP抗体(Dako)(1:2000希釈)を用いて4℃で一晩反応させた後、蛍光タグ2次抗体およびDAPIで染色した。染色像を図3に示す。またこの時のDAPI染色に対するGFAP陽性細胞数の割合を図4に示す。以上の結果より、得られたアストロサイトはおよそ9割GFAP陽性の細胞であることが確認された。
上述の方法で得られたアストロサイトを0.1%ゼラチンコートした48-wellプレートの1ウェルあたり4×104個の移し、3日間培養した。培養後、最終濃度が250 μM になるようにL-glutamate(Nacalai)を添加し、細胞外のグルタミン酸濃度をGlutamate Assay Kit colorimetric assay II(Yamasa Corporation)を用いて測定し、細胞内へのグルタミン酸の取り込み量を経時的に測定・算出した。この結果を図5に示す。得られたアストロサイトはグルタミン取り込み能を有していることが確認された。
Claims (25)
- 神経前駆細胞からアストロサイトを製造する方法であって、次の(1)から(3)の工程を含む方法;
(1)神経前駆細胞を神経栄養因子を含有する培養液中で培養する工程、
(2)(1)の工程で得られた細胞を解離させる工程、および
(3)(2)の工程で得られた細胞をコーティング処理されていない培養容器を用いて神経栄養因子を含有する培養液中で接着培養する工程。 - 前記神経栄養因子がGDNF、BDNFおよびNT-3から成るグループより選択される因子である、請求項1に記載された方法。
- 次の(4)および(5)の工程をさらに含む、請求項1または2に記載された方法;
(4)(3)の工程で得られた細胞を解離させる工程、ならびに
(5)(4)の工程で得られた細胞をコーティング処理されていない培養容器を用いてGDNF、BDNFおよびNT-3から成るグループより選択される因子を含有しない培養液中で接着培養する工程。 - さらに、前記工程(5)で得られた細胞を解離させ、コーティング処理されていない培養容器を用いてGDNF、BDNFおよびNT-3から成るグループより選択される因子を含有しない培養液中で接着培養する工程を、少なくとも2回繰り返す、請求項3に記載された方法。
- 次の(6)および(7)の工程をさらに含む、請求項3または4のいずれか1項に記載された方法;
(6)(5)の工程で得られた細胞を解離させる工程、ならびに
(7)(6)の工程で得られた細胞をゼラチンコートした培養容器を用いてGDNF、BDNFおよびNT-3から成るグループより選択される因子を含有しない培養液中で接着培養する工程。 - 前記工程(1)が66日以上の期間培養する工程である、請求項1から5のいずれか1項に記載された方法。
- 前記工程(3)が30日以上の期間培養する工程である、請求項1から6のいずれか1項に記載された方法。
- 前記工程(5)が20日以上の期間培養する工程である、請求項3から7のいずれか1項に記載された方法。
- 前記神経前駆細胞が、多能性幹細胞をBMP阻害剤およびTGFβ阻害剤を含有する培養液中で培養する工程を含む方法により製造される細胞である、請求項1から8のいずれか1項に記載された方法。
- 前記BMP阻害剤がDorsomorphinであり、前記TGFβ阻害剤がSB431542である、請求項9に記載された方法。
- 前記多能性幹細胞から神経前駆細胞を製造する工程が、胚様体を形成後、接着培養させる工程を含む、請求項9または10に記載された方法。
- 前記神経前駆細胞がヒト神経前駆細胞である、請求項1から11のいずれか1項に記載された方法。
- 請求項1から12のいずれか1項に記載の方法で製造されたアストロサイト。
- ニューロンとアストロサイトが混在する細胞群よりアストロサイトを選択的に培養する方法であって、次の(I)および(II)の工程を含む方法;
(I)細胞群を解離させる工程、および
(II)(I)の工程で得られた細胞をコーティング処理されていない培養容器を用いて培養する工程。 - 前記工程(II)の培養する工程が、神経栄養因子を含有する培養液中で接着培養する工程である、請求項14に記載の方法。
- 次の(III)および(IV)の工程をさらに含む、請求項14または15に
記載の方法;
(III)前記工程(II)で得られた細胞を解離させる工程、ならびに
(IV)前記工程(III)で得られた細胞をコーティング処理されていない培養容器を用いてGDNF、BDNFおよびNT-3から成るグループより選択される因子を含有しない培養液中で接着培養する工程。 - さらに、前記工程(IV)で得られた細胞を解離させ、コーティング処理されていない培養容器を用いてGDNF、BDNFおよびNT-3から成るグループより選択される因子を含有しない培養液中で接着培養する工程を、少なくとも2回繰り返す、請求項16に記載された方法。
- 前記工程(II)が30日以上の期間培養する工程である、請求項14から17のいずれか1項に記載された方法。
- 前記工程(IV)が20日以上の期間培養する工程である、請求項16から18のいずれか1項に記載された方法。
- 前記ニューロンとアストロサイトが混在する細胞群が、次の(i)および(ii)の工程により多能性幹細胞から製造された細胞群である、請求項14から19のいずれか1項に記載された方法;
(i)多能性幹細胞をBMP阻害剤およびTGFβ阻害剤を含有する培養液中で培養する工程、および
(ii)(i)の工程で得られた細胞を神経栄養因子を含有する培養液中で培養する工程。 - 前記工程(i)が、多能性幹細胞をBMP阻害剤およびTGFβ阻害剤を含有する培養液中で胚様体を形成後、接着培養させる工程である、請求項20に記載された方法。
- 前記BMP阻害剤がDorsomorphinであり、前記TGFβ阻害剤がSB431542である、請求項20または21に記載された方法。
- 前記神経栄養因子がGDNF、BDNFおよびNT-3から成るグループより選択される因子である、請求項14から22のいずれか1項に記載された方法。
- 前記工程(ii)が66日以上の期間培養する工程である、請求項20から23のいずれか1項に記載された方法。
- 前記ニューロンとアストロサイトが混在する細胞群がヒト細胞群である、請求項14から24のいずれか1項に記載された方法。
