Abstract
Description
(1)標的化リガンド(分子結合要素)をマイクロバブルシェルに結合させるためのビオチン−(ストレプト)アビジン結合化学の伝統的使用。ストレプトアビジンとアビジンは免疫原性のタンパク質であり、ヒトでの使用は避けた方が良い;

が含まれる。

貯蔵法1:形成された(洗浄済み、または未洗浄の)(分子結合要素が結合した、または結合していない)微粒子の凍結。これには瞬間凍結と容器のヘッドスペース中の所望のガスとの/無しでの−80℃でのそれらの微粒子の貯蔵が含まれる。
貯蔵法2:形成された(洗浄済み、または未洗浄の)(分子結合要素が結合した、または結合していない)微粒子の凍結乾燥。形成された微粒子に対して凍結乾燥前にマトリックス剤(例えば、炭水化物)を添加しても添加しなくてもよい。それらの微粒子を再形成するため、適切な水ベースの溶液を添加し、その後に所望のガスの存在下で剪断混合(超音波処理または好ましくは手動/機械振盪)してよい。
貯蔵法3:分子結合要素が結合した、または結合していない微粒子のための有機溶媒中の脂質混合物の凍結乾燥(そうして最初の凍結乾燥物を作製する)。それらの成分を有機溶媒(例えば、シクロヘキサン:クロロホルム)に溶解および混合する。その後、凍結乾燥前にマトリックス剤(例えば、炭水化物)を添加しても添加しなくてもよい。それらの微粒子を形成するため、その凍結乾燥物を適切な水ベースの溶液に溶解し、その後にその溶液を所望のガスの存在下で剪断混合前にその混合物中の脂質の鎖融点(Tc)より上(例えば、60〜65℃)まで昇温しても昇温しなくてもよい。
かった分子結合要素を除去することが好ましい。例えば、分子結合要素を含有する複合体化された成分(ii)を使用前に精製してよい。また、成分(ii)上の残留マレイミド部分(すなわち、活性マレイミド機能を含有する部分)は、例えば、加水分解または過剰なチオールとの反応により不活性化され得る。とりわけ、分子当たり一つのチオール基を有する適切な分子、例えば、L−システインを使用することが好ましくあり得る(未反応チオールを除去するための精製が後で必要とされ得る)。精製のためのあらゆる適切な方法、例えば、高速液体クロマトグラフィー(HPLC)を用いることができる。複合体化した成分の検証はあらゆる適切な方法、例えば、マススペクトロメトリー、核磁気共鳴、およびアミノ酸分析(分子結合要素および/または反応物質の性質に依存する)によるものであり得る。
物質、ポリエチレングリコール、カルボキシメチルセルロースナトリウム、ポリアクリレート、ワックス、ポリエチレン・ポリオキシプロピレンブロック重合体、ポリエチレングリコールおよび羊毛脂が含まれるが、これらに限定されない。
抗体
マウスEselに対するラットIgG2a,κであるMES−1モノクローナル抗体(mAb)10およびそのF(ab’)2断片(MES−1 F(ab’)2)はD Brown博士(UCB Celltech、英国)によって提供された。AF488−MES−1(7分子のAlexa Fluor(登録商標)488蛍光色素で標識されたMES−1)と還元型MES−1 F(ab’)2(トリス(2−カルボキシエチル)ホスフィンヒドロクロリド(TCEP)還元によりF(ab’)2当たり2つのチオール(SH)基)を以下に記載されるように調製した。マウスPECAM−1に対するラットIgG2a,κであるMEC13.3 mAb(BD Biosciences)。マウスPECAM−1に対するアロフィコシアニン標識mAb(BD Pharmingen)。ラットIgG2a,κアイソタイプ陰性対照mAb(BD Biosciences)。ラットIgG2aに対するビオチン化ウサギmAb(二次抗体)(Vector Laboratories)。
4モル過剰のTCEP(シグマアルドリッチ)を使用してMES−1 F(ab’)2を還元した。交換緩衝液(50mMの2−(N−モルホリノ)エタンスルホン酸(シグマアルドリッチ)、2mMのエチレンジアミンテトラ酢酸(シグマアルドリッチ)、pH6)中のMES−1 F(ab’)2(83.3μΜ、8.3mg/ml)とTCEP(333.3μΜ、0.096mg/ml)を一定速度で撹拌しながら37℃で1時間保温した。その反応体積は1〜1.6mlの範囲であった。氷上においてその反応を停止し、製造業者(Perbio Science)の指示に従って4℃で5ml−Zeba脱塩スピンカラム(サイズ排除限界:1,000Da)を使用するスピンカラムゲル濾過クロマトグラフィーにより還元したF(ab’)2を直ぐに精製した。そのスピンカラムは事前に冷交換緩衝液で平衡化された。次の改変と共に製造業者の指示に従ってエルマン試薬(
Perbio Science)を使用するエルマン試験を用いてそのF(ab’)2の還元の程度(F(ab’)2当たりのSHの数)を分光光度的に決定した。2.5μlのエルマン試薬(10mM、4mg/ml)、7.5μlの交換緩衝液および90μlの交換緩衝液中の精製した還元型F(ab’)2からなるエルマン反応を、光の曝露を最小限にするためにアルミホイルで覆って室温で(rt)保温した。エルマン反応の開始から24分の時点で412nmでの吸光度(A412)を測定し、交換緩衝液(pH6)中の既知の濃度のSH含有化合物であるL−システイン塩酸(Perbio Science)を使用するエルマン反応の検量線を参照してその還元型F(ab’)2試料中のSHの濃度を決定した。その還元型F(ab’)2の代わりに交換緩衝液を添加した二つ一組のエルマン「ブランク」反応を検査試料からのA412のベースライン減算のために使用した。還元型F(ab’)2の濃度を(交換緩衝液を使用してゼロを合わせた分光光度計を用いて)、エルマン試薬がA280に干渉するのでエルマン試薬が存在しない中でA280から決定した。F(ab’)2還元の程度をF(ab’)2当たりのSH=[SH(μM)]/[F(ab’)2(μΜ)]として計算した。その還元型F(ab’)2はF(ab’)2の分子当たり2つのSH基を含有した。その精製された還元型F(ab’)2をその後の微粒子への複合体化のために交換緩衝液中に約8mg/ml(80μΜ)の濃度で保持した。
Alexa Fluor(登録商標)488カルボン酸、2,3,5,6−テトラフルオロフェニルエステル、5−異性体(Molecular Probes)を使用してMES−1を次のように標識した。50μΜのMES−1 mAb(リン酸緩衝生理食塩水(PBS)pH7.5中に194μΜまたは29.1mg/mlの濃度で25.7μl)、750μΜのAF488(水の中に11.3mMまたは10mg/mlの濃度で6.6μl)、2.6μl(添加されたMES−1 mAbの10分の1の体積)の1MのNaHCO3、pH8.5および65.1μlの蒸留水からなる100μlの標識反応混合物を手でやさしく30分撹拌して室温で1時間保温した。対照は色素またはMES−1 mAbを含まなかった。次に6kDaのサイズ排除限界を有するゲル濾過クロマトグラフィースピンカラム(SSCパッキング緩衝液を含むバイオスピンP−6カラム、BioRad)を次の特定の条件で製造業者の指示に従って使用してAF488標識MES−1をその反応混合物から精製し、PBS(pH7.5)中に懸濁した。3回の洗浄サイクルを用いてまずバイオスピンカラムをPBSで緩衝液交換した。全ての遠心分離を20℃で実行した。分光光度計を使用してAF488標識MES−1の濃度と標識の程度を決定した。試料をPBS中に100μlの体積まで希釈して二つ一組に揃え、吸光度を280nm(A280)および495nm(A495)で測定した。標識されたmAbの濃度を次のように計算した。[mAb(mg/ml)]=[A280−(A495×0.11)]/1.4×希釈係数であり、0.11はA280へのAF488の寄与に対する補正率である。[mAb(μM)]=[mAb(mg/ml)]/150000×106を用いてmg/ml単位のmAbの濃度がμΜに換算された。その式で150000はmAbのDa単位の分子量であり、106はMをμΜに変換するための増倍率である。AF488の濃度を次のように計算した。[AF488(μM)]=A495/71000×希釈係数×106であり、その式で71000は494nmでのAF488色素のおおよそのモル吸光係数(cm−1M−1単位)であり、106はMをμΜに変換するための増倍率である。最後に、標識の程度を次のように計算した。標識の程度=[AF488(μM)]/[mAb(μΜ)]。光の曝露を最小限にするために全てのステージでアルミホイルを使用した。このプロトコルの下で未反応AF488がカラム中に保持され、「MES−1mAb無し」対照反応によって確認された。15対1(AF488:MES−1)の標識反応モル比がそれぞれ7分子のAF488で標識されたMES−1を46%の標識効率、約89%の収率で産生した。精製されたAF488標識−MES−1(6.748mg/ml、
44.984μΜ)を分割し、アルミホイルで覆い、そして使用するまで−20℃で貯蔵した。AF488標識MES−1のEselに対する感度と特異度の保存はリポ多糖(LPS)で前処置された野生型(WT)マウスとEselノックアウト(KO)マウスの凍結心臓切片に対する免疫組織化学を用いて確認された。
未変性の(複合体化されていない)マレイミド官能化脂質の殻で覆われたオクタフルオロプロパン(C3F8)微粒子は、1,2−ジステアロイル−sn−グリセロ−3−ホスフォコリン(DSPC;Avanti Polar Lipids、アラバマ州)、1,2−ジステアロイル−sn−グリセロ−3−ホスフォエタノールアミン−N−(マレイミド(ポリエチレングリコール)−2000)(DSPE−PEG2000−Mal;Avanti Polar Lipids)、モノ−ステアレートポリ(エチレン)グリコール(ステアリン酸PEG40;シグマアルドリッチ)、および蛍光色素1,1’−ジオクタデシル−3,3,3’,3’−テトラメチルインドカルボシアニンパーコレート(DiI;Molecular Probes)を75:9:14:2のモル比で含む水性懸濁液のC3F8の存在下での超音波処理により調製された。Esel標的化粒子を作成するためにF(ab’)2当たり2つのSH基を含有するTCEP還元MES−1 F(ab’)2をマレイミド・チオール複合体化によりこれらの未変性粒子のシェル外表面にグラフトした。その結合反応比率は粒子当たり4.338×106個のF(ab’)2分子(109粒子当たり7.2nmolのF(ab’)2)であった。(注意:水性懸濁液中のDSPE−PEG2000−Mal分子の総数/その水性懸濁液から作製された未洗浄粒子の総数=4.338×106である。その水性懸濁液中の成分の10%以下が粒子シェルに組み込まれ、且つ、そのシェル上の成分のモル比が最初の水性懸濁液の成分のモル比に近いままである場合56、集団平均サイズを有する粒子は4.338×105以下のマレイミド分子を含有し、推定されるF(ab’)2:マレイミドの結合反応モル比は10対1以上である。)その結合反応は一定速度でやさしく撹拌しながらC3F8雰囲気下、中性に近いpHで4℃において30分間実施された。あらゆる未反応SHの反応を停止するために過剰なN−エチルマレイミド(NEM)を添加して反応を停止した。組込まれなかった成分と粒子断片を除去するために粒子複合体化の前後にC3F8雰囲気下において4℃で複数のサイクルの遠心分離浮揚を用いて粒子を脱気した冷通常生理食塩水により洗浄した。新しく調製された粒子を直ぐに20〜50μlのアリコットに分割し、キャップをし、パラフィルム(American National Can)で密封し、その後に液体窒素中で瞬間凍結し、そして使用するまで−80℃で貯蔵した。後に融解されたEsel標的化粒子の濃度は異なる時期に調製された5バッチの間で1〜3×109粒子/mlの範囲であった。
した。プローブの先端を溶液の中に約2mmの位置に配置し、約60℃の初期温度、約120Wの音響出力で高強度超音波ホーン(20〜21kHz)により超音波処理を30〜60秒間実施した。より多くのC3F8ガスをそのマイクロバブル分散液に注入し、その容器にキャップをして直ぐに氷冷水の中に入れて(3分)超音波処理過程中に生じた熱を消散させた。作製した微粒子は、Beckman Coulter Allegra X−15R遠心分離機(Beckman Coulter)を使用して、1,000g、4℃で15〜25分間の遠心分離浮揚により洗浄(精製)された。遠心分離後に粒子は試料バイアル瓶の上部に浮遊する。浮上分離液を取出し、同量の脱気した冷通常生理食塩水(pH7.4)と交換した。その洗浄ステップを7回繰り返して組込まれなかったシェル成分と粒子断片を除去した。Esel標的化微粒子を作製するため、これらの洗浄済み未変性微粒子を還元型MES−1 F(ab’)2に混合しながら添加した(各還元型F(ab’)2分子は上記のように調製され、2つのSH基を含有する)。上出来のEsel標的化粒子を作製するために使用された結合反応比率は粒子当たり4.338×106個のF(ab’)2分子であった。その結合反応混合物中の粒子とF(ab’)2の濃度はそれぞれ5〜8×109/mlと35〜60uM(3.5〜6mg/ml)の範囲であった。その反応混合物はその還元型F(ab’)2由来の約3分の2の体積の交換緩衝液(pH6)と洗浄済み粒子由来の約3分の1の体積の通常生理食塩水(pH7.4)を含む。その結合反応を4℃で30分間保温し、垂直に傾いた回転ホイール上で穏やかに連続的に混合した。20モル過剰の乾燥ジメチルスルホキシド(DMSO、シグマアルドリッチ)に溶解した80mMのNEM(シグマアルドリッチ)をF(ab’)2に添加することにより粒子複合体化を停止した。その反応混合物を回転機上において4℃で30〜60分間保温した。典型的には、その反応混合物中のNEMとDMSOの濃度はそれぞれ約1mMと1.7%(体積/体積)以下であった。その後、それらの粒子を上記のように160g、4℃で5分間の遠心分離浮揚により冷通常生理食塩水で4回洗浄した。これにより組込まれなかったF(ab’)2、未反応NEM、DMSOおよび粒子断片を除去した。粒子の喪失を最小にするために全ての洗浄と保温は(それらの粒子からのC3F8ガスの拡散に適した濃度勾配を減少させるために)C3F8雰囲気下おいて、且つ、(ガス拡散速度を低下させるために)4℃で粒子濃度を高く保って(1×109粒子/ml超)実施された。それらの粒子DiI蛍光色素を保存するためにDiI化合物、凍結乾燥物または粒子を光から保護した。
例えば冷通常生理食塩水中に1:200に希釈して粒子を血球計算板(Reichert明線金属化血球計算板、Hausserscientific、ペンシルバニア州)に配置して粒子の完全性(球状の形態、凝集の不在)を顕微鏡観察で検討した。粒子濃度と粒度分布を30μm径開口部計数チューブが装着されたCoulter Multisizer IIe(Coulter Electronics)での製造業者の指示に従うエレクトロセンシングにより決定した。その構成により0.72〜18μmのサイズ検出範囲と0.09μmの解像度が可能になった。粒子荷電分析について、それらの粒子を1mlの1mMのKCl(pH7.4)に約107粒子/mlの濃度で分散した。その正味の電荷をZetasizer Nano ZS(Malvern Instruments)での製造業者の指示に従う光分散によりゼータ電位として測定した。
100μlの2.5×107粒子/mlの濃度のEsel標的化粒子または非標的化(未変性)粒子を200μlの7nMの濃度の組換えホモ二量体マウスEselタンパク質(R&D Systems)で被覆された逆向きのポリスチレン製ペトリディッシュ(ディッシュE)に、または500μlの67nMの濃度の過剰なMES−1 F(ab’)2で事前にブロックされたEsel被覆ディッシュ(ディッシュB)に、またはディッシュの被覆にEselの代わりにリン酸緩衝生理食塩水pH7.5(PBS)が使用された
非被覆ディッシュ(ディッシュP)に配置した。1分後に結合しなかった粒子をやさしく洗い流した。その後、液浸対物レンズを装着した正立型光学顕微鏡での即時の検査のためにそれらのディッシュに脱気した冷PBSを再充填した。各ディッシュに結合した粒子の数を計数し、10箇所のランダムな光学場(OF)から平均を求めて結合粒子密度を決定した。
野生型(WT)マウス:成熟オスC57B16/Jax(Charles River、英国)。Eselノックアウト(KO)マウス:K Norman博士とP Hellewell教授(シェフィールド大学、英国)から寄贈されたマウスからその地域で繁殖させたC57B16バックグラウンドの成熟オスEselホモ接合性KO57。全ての動物作業は1986年の動物(科学的処置)法を受けて内務省により許可されたプロジェクトライセンスとパーソナルライセンスの下で実施された。地域の倫理審査委員会から倫理的な許可をさらに得た。
WTマウスとEsel KOマウスを通常生理食塩水中に200μlの体積にまでした50μgの大腸菌0111:B4株由来のLPS(シグマアルドリッチ)で腹腔内(ip)注射により前処置して全身性炎症を誘導した。ip注射によるLPSの全身性投与により全身性炎症が引き起こされ、その全身性炎症には心臓と腎臓を含む複数の器官におけるEsel発現の誘導が含まれる58、59。この動物モデルは、(i)その動物モデルが外科的外傷または血液凝固などの交絡変数を導入し得る外科的処置を必要としない点、(ii)それらの粒子が標的とする分子が複数の器官に存在し(たった1つではない)、ユニークにも標的化粒子特異度の評価を複数の器官において同時に可能にし、且つ、他の組織による「粒子窃盗」が存在する中で目的の組織において標的分子を検出する撮像技術の能力を評価する点、および(iii)心筋における標的分子発現が基本的に全体的で均一であり、ユニークなことに標的とされた粒子シグナルの減衰の検討を可能にする点で粒子標的化において使用される他のモデル(例えば、虚血再灌流傷害、心臓移植拒絶、血栓形成、慢性虚血/腫瘍血管形成の動物モデル)と異なる。さらに、活性化内皮細胞上で単独で発現しているEselとLPS前処置マウスの心筋における基本的に全体的で均一なE
selの発現の組合せがそのLPSマウスモデルとEselの組合せを、分子発現の音響学的定量を試験するための理想的なインビボモデルにする。
免疫組織化学を新しく採取したWT(LPS前処置有り/無し)マウスとEsel KO(LPSで前処置済み)マウスの心臓のアセトン固定凍結切片に対して実施した。100μlの1:1000ウサギ血清(シグマアルドリッチ)を用いて室温(rt)で1時間(h)非特異的結合部位をブロックした後に100μlの0.01mg/mlの濃度の一次抗体:MES−1(Esel用)、MEC13.3(PECAM−1用、内皮マーカー)またはアイソタイプ陰性対照と切片をrtで1時間保温した。その後、各切片を100μlの0.005mg/mlの濃度のビオチン化二次抗体とrtで60分間保温した。0.3%のΗ2O2メタノールをrtで20〜30分間用いて内在性ペルオキシダーゼをブロックした後にホースラディッシュペルオキシダーゼベースの検出系であるVectastain ABCキット(Vector Laboratories)を発色基質としての3,3’−ジアミノベンジジン溶液(SIGMA FAST(商標)DABタブレット、シグマアルドリッチ)と共に使用した。ハリス改変ヘマトキシリン溶液(シグマアルドリッチ)と1%のNaHCO3を使用して切片を対比染色し、その後に70〜100%のエタノールにより脱水し、乾燥し、そしてHistomount(VWR)を載せ、そして光学顕微鏡観察により検査した。LPS前処置の時間と即時の組織採取のための動物の殺処理との間の期間をLPS時間として表した。
WTマウスを上記のようにLPSで前処置した。LPS前処置の時間と即時の組織採取のための動物の殺処理との間の期間をLPS時間として表した。新しく採取された組織をRNAlater(登録商標)溶液(Ambion)中に保持してリボ核酸(RNA)をインサイチュで保存した。製造業者の指示に従ってTRIzol試薬(Invitrogen)を使用して総RNAを後に抽出した。その後、Qiagen Omniscript(登録商標)逆転写キット(Qiagen)を製造業者の指示に従って使用してファーストストランド相補性デオキシリボ核酸(cDNA)合成を実施した。これに続いてEselとヒポキサンチンホスフォリボシルトランスフェラーゼ−I(HPRT−I)についてのSYBR(登録商標)グリーン検出法を用いるリアルタイムqPCRをiCycler(商標)(iCycler iQリアルタイムPCR検出システム、Bio−Rad)中の96ウェルプレート上で製造業者の指示に従って実施した。全てのPCR反応を同じプレートの三つ一組のウェルで実施した。プライマー配列は:Eselフォワードプライマー5’−CTCATTGCTCTACTTGTTGATG−3’、Eselリバースプライマー5’−GCATTTGTGTTCCTGATTG−3’、HPRT−Iフォワードプライマー5’−ATTAGCGATGATGAACCAG−3’、HPRT−Iリバースプライマー5’−AGTCTTTCAGTCCTGTCCAT−3’であった。データ分析のためにiCycler(商標)iQオプティカルシステム・ソフトウェア・バージョン3.0a(Bio−Rad)を使用して増幅プロットから閾値サイクル(Ct)を決定した。EselプライマーペアとHPRT−IプライマーペアのPCR効率が5%以下しか異ならなかった(それぞれ93±4%と92±3%(平均値±SD);n=4ずつ)ので比較Ct方法を使用して次の式を使用してHPRT−Iのメッセンジャーと比較したEselメッセンジャー(mRNA)の量を推定した:

外科手術の2時間前にWTマウスとEsel KOマウスにおいて50ngの組換えIL−1β(R&D Systems)の陰嚢内注射することにより精巣挙筋の炎症を引き起こした。24Gの0.7×19mm静脈内(iv)カテーテル(空所、約50μl)(BD Medical)で尾静脈をカニューレ処置した。通常生理食塩水中に1mg/mlキシラジン(Rompum、バイエル)と10mg/mlケタミン塩酸(Ketalar、Parke−Davis)を含む200〜300μlの混合物の腹腔内(ip)注射を用いて長期間の麻酔を達成した。注文製作した温度管理(37℃)IVMステージ上に動物を配置した。解剖顕微鏡下で陰嚢切開により右または左の精巣をやさしく露出させた。精巣挙筋に沿って長軸方向の切開を行い、次にその精巣挙筋を広げて半透明の顕微鏡観察ステージにピンで留めた。露出させた筋肉は温度管理されたタイロード塩緩衝液溶液(9.6gタイロード塩(シグマアルドリッチ)+1g炭酸水素ナトリウム、滅菌蒸留水で1Lにした)の連続灌流により維持された。明視野顕微鏡観察および蛍光顕微鏡観察用に20倍および40倍の液浸対物レンズ(Water Achroplan、カールツァイス)、電荷結合素子(CCD)カメラ(コントローラー付きカラー冷却3CCDカメラ、浜松ホトニクス)、シリコン電子倍増ターゲット(SIT)管カメラ(C−2400−08、浜松ホトニクス)、モニター(Triton、ソニー)、S−VHSレコーダー(モデルAG6730 SVHS 625、パナソニック)およびパーソナルコンピューターが装備された正立顕微鏡(Axioskop、カールツァイス)を使用して観察と記録を行った。粒子結合に対する剪断速度をWT動物とEsel KO動物の間で比較するために光学ドップラー流速計(微小循環研究所、テキサスA&M大学、テキサス)を使用して
WTマウスとKOマウスの20〜40μmの直径の細静脈における微小血管中心線赤血球流速(Vrbc)を粒子の注射前に測定した。動物当たり5つのランダムな血管部分を測定した。
麻酔した(キシラジン/ケタミン混合物、ip)WTマウスとEsel KOマウスの精巣挙筋における生体内顕微鏡観察評価のため、尾静脈カテーテルを介した急速ivボーラスとして100μlの通常生理食塩水中に1.5×107個のEsel標的化粒子を投与し、続いて直ぐに100μlの通常生理食塩水洗浄液を投与した。その筋肉の露出の2時間前に全ての動物を50ngの組換えIL−1β(R&D Systems)の陰嚢内投与で前処置して精巣挙筋炎症を誘導した。明視野顕微鏡観察および蛍光顕微鏡観察用に20倍および40倍の液浸対物レンズ、CCDカメラおよびSIT管カメラが装備された正立顕微鏡を使用して観察を行った。粒子結合に対する剪断速度を決定するために光学ドップラー流速計を使用して動物当たり5つのランダムな血管部分に由来する20〜40μmの直径の細静脈においてVrbcを粒子の注射前に測定した。S−VHSレコーダーおよびパーソナルコンピューターを使用して全ての記録を行った。実験の最後に全ての動物を殺処理した。設定の詳細については上記を参照のこと。多数の異なる血管を包含する幾つかのOFについて、血流と粒子を明視野顕微鏡観察および蛍光顕微鏡観察で評価した。モニター上の一つのOFにおける自由循環粒子の数を5分、7分、10分±15分の時点で蛍光顕微鏡観察により10秒にわたって計数した。一つのOF場における結合した粒子の蓄積(3秒を超えて動かないものと定義される)を粒子の注射から15分後までについて評価した。動物当たり2〜6本の細静脈で細静脈当たり1〜5か所の400μm長の部分から、20〜40μmの直径の細静脈に結合した粒子の数を(自由循環粒子が存在しなかった/最小であった)粒子の注射から約15分後の時点で計数した。数匹の動物では、断続的な明視野顕微鏡観察および蛍光顕微鏡観察の組合せを用いて同じOF中にある結合した粒子を最大で90分間追跡し、組織間質への移動または細胞内移行の存在/不在について調査した。分析に関して、結合した粒子の密度を血管表面積(VSA)当たりの粒子数で表し、VSA(mm2)=πD(mm)×L(mm)であり、DとLはそれぞれ血管部分の直径と長さであった。各細静脈の結合粒子密度はその細静脈の部分の平均とし、各動物の結合粒子密度はその動物の細静脈の平均とした。次のようにVrbcから剪断速度を推定した。剪断速度(s−1)=8×Vb(cm/s)/D(cm)であり、その式で、Vb(cm/s)=Vrbc(cm/s)/αであり、Vbは平均バルク速度であり、αはVrbcをVbに変換する計数であり、静脈におけるポアズイユの流れに1.6が採用される60。各動物の剪断速度は試料とされた5箇所のランダムな細静脈部分の平均とした。
50ngのIL−1βの陰嚢内投与により2.5時間前に前処置されたWTマウスとEsel KOマウスに、上記のIVMについて説明された方法を用いて1.5×107個のEsel標的化粒子を投与した。15分後に通常生理食塩水中に50μgのAF488−MES−1(Esel用)と25μgのマウスPECAM−1(内皮マーカー)に対するアロフィコシアニン標識mAbを含む150μlのカクテルの急速ivボーラスをiv投与し、続いて100μlの通常生理食塩水洗浄液を投与した。さらに15〜20分後にキシラジン/ケタミン混合物のivボーラスを用いて動物に終末麻酔を施し、続いてPBSの灌流により循環中の結合しなかった粒子とmAbを除去した。その直後に精巣挙筋を採取し、共焦点顕微鏡観察(zスタック作製)用に4%パラホルムアルデヒドPBS中に固定した。40倍の液浸対物レンズを有する正立共焦点レーザー走査顕微鏡(LSM 5
PASCAL、カールツァイス)を使用して3種類の異なる蛍光であるDiI(粒子)、Alexa Fluor(登録商標)488(Esel)およびアロフィコシアニン(PECAM−1)を各深度でz軸の次の深度に移動する前に連続して走査した。Zeis

WTマウスとEsel KOマウスを全てLPSで前処置し、上記のように尾静脈をカニューレ処置し、キシラジン/ケタミン混合物で麻酔した。その後、胸部、腹部および骨盤部の毛刈りを行い、動物を仰向けに配置した。ECG電極パッド(Ambu(登録商標)ブルーセンサーP、Ambu)を足に取り付け、「小動物ECGフィルター」が装備されたUS機械(Acuson Sequoia(登録商標)512USシステム、シーメンス、カリフォルニア州)に接続した。一層の温かいゲル(超音波および電気伝達用ゲル、Henleys Medical)が皮膚とUS変換機(15L8−s直線配列変換機、26mmのフットプリント、シーメンス)の間に連結された。使用したUSの設定は14MHz(P14MHz、空間分解能:約0.2mm)のコントラストパルスシーケンシング(CPS)モード、スキャナーにより推定される0.22〜0.26の低メカニカルインデックス(MI)を生じる9dBの送信力、55dBのダイナミックレンジ、0%のタイムゲイン、8のCPSゲイン、15dBの基礎2Dゲイン、カラーマップM:3(粒子シグナルがヒートオブジェクトスケール(「CPSコントラストのみ」画像)で提示され、組織シグナルがグレースケール(「B−モード」画像)で提示される)であり、TEQは使用されなかった。粒子の注射前にベースラインの心臓の胸骨傍短軸(PSA)断面を乳頭筋レベルで3秒のデジタルクリップとして記録し、「領域拡大選択」(RES;解像度が上昇した拡大画像を生じる)を行い、且つ、行わずに心臓の胸骨傍長軸(PLA)断面と心尖部四腔(A4C)断面を3秒のデジタルクリップとして記録した。その後、自由直立クランプを使用して所定の位置に固定された前記変換機を用いて撮像をPSA断面に保持した。その後、ストップウォッチをスタートし、1×108個のEsel標的化粒子(通常生理食塩水で調節された100μlの体積中)を10秒の時点で尾静脈カテーテルを介して急速ivボーラスとして1〜2秒にわたって注入し、続いて20秒の時点で100μlの通常生理食塩水洗浄液を1〜2秒にわたって注入した。ストップウォッチで0〜1分23秒まで中断することなく連続的US超音波照射を行い、その後に中断し、その後にデジタル画像の取得のたびに3秒間だけ超音波照射を再開した。数回の連続的心周期を含む心臓の3秒のデジタルクリップ(RES活性化)をストップウォッチ上で10秒と13秒の時点で記録し、その後に10秒の間隔で20秒から1分20秒まで記録し、その後に1分の間隔で2分20秒から10分20秒まで記録し、その後に2分の間隔で12分
20秒〜30分20秒まで記録し、その後に5分の間隔で60分20秒まで記録した(左心室(LV)内腔(中央血液プール)内の粒子コントラスト強調が見られなくなった場合にはそれらより早く画像取得を停止した)。PSA断面の心臓および周囲の組織を含む胸部の拡大されていない(非RES)画像を約5分の間隔で記録した。心臓の他の断面(PLA断面およびA4C断面)を最後に取得した。数匹の動物では0.22のMIで撮像する7MHz(P7MHz、空間分解能:約0.4mm)CPS(ゲインと他の設定は14MHzでの撮像と同じに維持された)もベースライン時と14MHz撮像検査の最後に取得された。14MHzのCPS撮像から7MHzのCPS撮像に切り換えるときにUS周波数を低下させる前に送信出力を最初に−9dBから−19dBまで低下させて不注意な粒子破壊を引き起こすMIの上昇(約0.7まで)を回避した。数匹の動物では、胸部、腹部および骨盤部の他の組織の撮像を前後方向撮影法でベースライン時と心臓撮像検査の最後に実施して心臓外組織における粒子保持を調査した。これを行うためにプローブを横方向に配置し、3秒毎の画像取得の間に首の真下から骨盤部まで尾方向にゆっくりと動かした。14MHzのCPSと7MHzのCPSでの非RES画像を取得した。以前の粒子投与からの持ち越し効果(例えば、Esel結合部位のブロッキング)を回避するために全ての動物は1用量だけの粒子を受容した。撮像の最後に動物を殺処理し、凍結切片免疫組織化学とqRT−PCRのために上記のように組織を直ぐに採取した。
YABCO(著作権)ソフトウェア(LLCシャーロッツビル、バージニア州)を使用するビデオ濃度測定法を用いてオフラインの粒子シグナル強度を定量した。PSA断面の心臓の拡張末期画像フレーム(「CPSコントラストのみ」画像)を選択して整列し、(例えば、人為的な大きな動きのために)整列させることができない画像フレームを除外した。図4aに示されるように心筋の中部前壁(M)とLV内腔中の隣接する領域(C)に目的の領域(ROI)を設置した。これらの領域では全ての動物において一貫してUS減衰の影響が最低または最小であったので、それらの領域を選択した。ビデオシグナル強度(VI)を次の式を使用する対数展開により「線形化」した:
上のTICに従って得られた、微粒子投与から20分10秒後の時点での心筋におけるベースラインを差し引いたバブルシグナル強度(R20)を分析に使用した。TICの定量的分析に基づく代替的な新しい方法(TICベースの方法)もEsel発現のレベルを定量するために使用した。そのTICベースの方法は、インビボでの保持微粒子半減期と循環微粒子半減期を含む他の変数が同時に決定されることも可能にした。US分子イメージングのためのLPS時間はLPS前処置の時間と標的化微粒子の投与との間の期間とした。各動物の平均心拍数(HR)は心臓撮像中の様々な時点で記録された全てのHRから計算された。
ピアソンの相関、線形回帰分析または非線形回帰分析を示されるように実施した。示される場合にはスチューデントのt検定またはテューキーのポストホック分析付きANOVAを有意性検定のためにp<0.05を有意として使用した。
Esel発現
凍結切片免疫組織化学により、EselはLPSで前処置された全てのWTマウスの心臓(n=35、LPS時間=4〜9時間)において発現することが示された。空間分布は基本的に心筋を通して均一であったが後毛細管細静脈と毛細管に限定された。Eselは陰性対照であるLPSで処置されなかったWTマウス(n=5)、およびLPSで前処置されたEsel KOマウス(n=10、LPS時間=4〜7時間)では検出されなかった(図1)。
RT−qPCRにより、LPS前処置から約3時間後に心臓におけるEsel mRNAの濃度は時間と共に指数関数的に低下し、約9時間(n=42、LPS時間=3.2〜15.7時間)までに非常に低いレベルに達することが示された(図7a)。この傾向は、Eppihimerら58によってiv投与放射性標識mAbを使用して、同じマウス(n=19)の系統、性別、および年齢、ならびに同じLPS投与の用量および経路を用いて決定された細胞表面Eselタンパク質濃度(注射用量の放射活性に対する%/g組織(%ID/g組織)として表される)の傾向と類似していた(図7b)。
設計されたEsel標的化粒子の図式が図2aに示されている。これらの粒子は球状の形態を示し、粒子間凝集または架橋は最小限であった(図2b)。粒子径分布は独立した機会に調製された5バッチの間で再現性を有し、粒子直径=2.2(平均値)±0.2(SEM)μmであり、それらの粒子の98.6%または100%の直径がそれぞれ6μmまたは10μmより下であった(図2c)。それらの粒子(洗浄済み)は中性に近い電荷を有し、ゼータ電位はpH7.4で約5mVであった。
Esel標的化粒子は被覆ディッシュ上のEselに結合した(ディッシュE):2060(平均値)±1070(SEM)粒子/mm、n=5(図2d)。Eselに対する標的化粒子の特異度は過剰な抗Esel抗体で事前にブロックされたEselへのそれらの標的化粒子の最小の結合により実証された(ディッシュB):ディッシュE上の標的化粒子の8(平均値)±4(SEM)%、n=5。Eselで被覆されていないディッシュ(ディッシュP)に対して標的化粒子または非標的化(未変性)粒子を使用する、またはディッシュEおよびBに対して非標的化粒子を使用する追加の陰性対照により同様の低レベルの粒子結合が示された:ディッシュPに対する標的化粒子(9±3%、n=5)、ディッシュEに対する非標的化粒子(10±9%、n=2)、ディッシュBに対する非標的
化粒子(7±7%、n=2)およびディッシュPに対する非標的化粒子(7±7%、n=2)。テューキーのポストホック分析付きANOVAによりディッシュE上の標的化粒子と陰性対照との間に結合した粒子の相対密度について有意差(p<0.0001)が示された。陰性対照間では有意差は観察されなかった。
Esel標的化粒子を5匹のWTマウス(体重:25(平均値)±2(SD)g、範囲:23〜27g))と5匹のEsel KOマウス(24±2g、範囲:22〜27g)にそれぞれIL−1β前処置から3時間06分〜4時間03分および3時間17分〜4時間30分の時点で投与した。尾静脈を介したivボーラス投与から約7〜17秒後にそれらの粒子が循環し、精巣挙筋に到達することが観察された。自由循環粒子の数は時間と共に減少し、血液プールからのそれらの排出がKOよりもWTで早く起こった(図3a)。粒子投与後10分を越えると血液プール中の自由循環粒子の数は全ての動物において最小か、または検出不可能であった。それらの粒子はWT動物の精巣挙筋細静脈壁に結合および蓄積した(370(平均値)±46(SEM)粒子/mm2 VSA、n=5動物)。これはKO動物では最小であった(11±3、n=5)、p<0.0001(スチューデントのt検定)(図3c〜d)。精巣挙筋血管壁上の粒子蓄積は粒子ボーラス投与後直ぐにプラトーに達した(図3b)。蓄積した粒子は血液プールからの自由循環粒子の排出を越えて持続した。ごく少数の粒子においてローリングが観察された。結合した粒子の完全な脱離はまれにしか見られなかった。粒子による血管内閉塞は観察されなかった。最大で90分間断続的に観察すると、結合した粒子の組織間質への移動または細胞内移行は検出されなかった。共焦点顕微鏡観察により、結合した粒子はWT精巣挙筋細静脈(n=3動物、体重:21.8〜24g)の内皮細胞表面上で発現したEselと共局在することが示された。KO動物(n=2、25.7〜28.4g)ではEsel発現は検出できなかった(図3e)。
(1)動物
15匹のWTマウス(年齢:5.7(平均値)±0.2(SD)週、範囲:5.1〜6.1週;体重19.5(平均値)±1.5(SD)g、範囲:17〜22g)と8匹のEsel KOマウス(年齢:7.9±3.2週、範囲:5〜13.6週;体重22.3±3.7g、範囲:17〜28g)を撮像した。LPS注射と標的化粒子の投与との間の期間(LPS時間)はWT群について3時間26分〜5時間59分であり、KO群について4時間27分〜5時間39分であった。
粒子投与の前後の両方に存在する非線形CPSの人為的シグナル(オレンジ色)をWT動物とEsel KO動物の両方で見ることができた。これらの人為的シグナルは小さく、心筋の外側に存在した(図4a(フレーム0:10)、図4a2(フレーム20:20))。iv粒子ボーラス投与の後に粒子シグナルがまず4回の心臓の拍動(約1秒)の間に右心室において検出され、シグナル強度が急速に上昇した。これに続いて粒子が肺循環
から戻ってきて左心室に粒子シグナルが現れた。LV内腔と心筋における粒子シグナルの強度はそれぞれ粒子投与後から6〜7回の心臓の拍動(約1.5〜2秒)と9〜12回の心臓の拍動(約2〜3秒)の間にピークを迎えた。その後、粒子シグナル強度は時間と共に心室と心筋において低下した(図4a)。高い粒子濃度に起因する著しいUS減衰が粒子ボーラス投与後の早い時期にUS経路の遠位に位置する心臓の領域(例えば、PSA断面の心筋と隣接するLV内腔の5〜10時の位置、図4a)内のシグナルの大幅な喪失と共に起こった。血液プール(LV内腔)内の循環粒子濃度が時間と共に低下すると、その減衰は少なくなるが(フレーム0:30と6:20を比較されたい、図4a)、消失することはなかった(フレーム20:20、図4a1)。それは、上に被さる骨、肺の中の空気および保持されている粒子によって引き起こされる減衰が最も可能性のある原因である。重要なことに、心筋におけるEsel発現はリアルタイムで可視化され、血液プール(LV内腔)内の自由循環粒子が時間と共に減少および消失するとWT心筋内での粒子シグナルの残留が示された。KO心筋では、粒子シグナルの残留は低く/最少であり、心筋における低い/最少の程度の非特異的粒子保持と一致した(図4a〜b)。US経路の近位に位置する上に被さる骨、肺の中の空気および保持されている粒子によって引き起こされるUS減衰が撮像走査平面に応じて心筋のある特定の部分に影響を及ぼした。これによりWT動物におけるEselの標的結合粒子シグナルの擬似的な喪失が引き起こされた(例えば、14MHzのCPS撮像によるPSA断面のLVの中部後壁、中部下壁、中部中隔後壁、中部中隔前壁(それぞれ、反時計回りに5〜10時の位置)における保持された粒子シグナルの人為的喪失が存在した)。しかしながら、上に被さる物体の相対位置を変えるために走査平面を(例えば、PSA断面からPLA断面またはA4C断面へ)変化させることにより、またはUSの透過深度を増加させるためにUS周波数を(例えば、14MHzから7MHzへ)減少させることにより、保持されている粒子シグナルを良く回復してこれらの減衰効果を克服することができた(図4b)。こうしてWT心筋におけるEselの全体的発現がUS撮像で実証され、免疫組織化学データと一致した。
ベースライン像(粒子投与前)を参照したWT動物とEsel KO動物との間の胸部、腹部、および骨盤部の走査像の比較により、非RES組織における標的化粒子の非特異的保持が低い/最少であることが示された。WTの腎皮質でEsel発現が検出されたが、KO動物では検出されなかった(図5aおよび5b)。免疫組織化学と共焦点顕微鏡観察によりEselは主に腎糸球体の内皮細胞で発現しており、後者が腎臓に集中していることが確認された。予想通り、標的化粒子はWT動物とKO動物の両方で、粒子排出に係る主要なRES組織である脾臓と肝臓によって取り込まれた(図5aおよび5b)。
Esel標的化粒子に起因する死または顕著な有害事象は観察されなかった。局所的な心臓壁運動異常として現れる局所的な心筋血流の喪失を引き起こす、心筋中の顕著な粒子介在性血管内閉塞は検出されなかった。
標的化粒子のivボーラス投与後のTICから異なる特徴を有する三相が認められた(図4a)。
(1)動物
12匹のWTマウス(年齢:5.7(平均値)±0.3(SD)週、範囲:5.1〜6.1週;体重19.7(平均値)±1.4(SD)g、範囲:18〜22g)と8匹のEsel KOマウス(年齢:7.9±3.2週、範囲:5〜13.6週;体重22.3±3.7g、範囲:17〜28g)を使用した。LPS時間はWT群について3時間53分〜5時間59分の範囲であり、KO群について4時間27分〜5時間39分の範囲であった。3匹のWT動物が(i)微粒子の誤投与(1匹の動物)、(ii)Esel発現レベルに関する不確実性(2匹の動物)、および(iii)これらの3匹の動物のうちの1匹がおそらくは減衰に関する重大な人為的シグナルのために適切に定量できなかったTICを有していることから定量分析から除外された。
WT群とEsel KO群の両方で血液プール(LV内腔)内の自由循環粒子シグナルは108個の粒子のivボーラス投与から約20分後まで存在しなかった/最少であった。したがって、バブル投与から20分10秒後の時点までに残留しているあらゆる自由循環粒子のシグナルへの寄与は中央血液プールにおけるそれらの粒子の低い濃度(LV内腔におけるシグナル強度)のために無視できるものであったので、この時点での心筋におけるバックグラウンドを差し引いたI(R20)を使用してその保持された粒子だけの強度を表した。例えば、LV内腔内の0.1AUは心筋内のわずかに0.005〜0.024AUに寄与し、それは相対的心筋血液量(血液である心筋の%)の約5〜24%であった65〜67。R20はKO動物(0.06±0.03、−0.01〜0.24、n=8)よりもWT動物(0.46±0.16(SEM)、範囲:0.01〜1.61、n=12)において有意に(p<0.05)高かった。R20はWTマウス(r=−0.78、p<0.05、n=12)においてLPS時間と強力に相関したがKOマウス(r=−0.
05、p=0.9、n=8)では相関しなかった(図8c)。R20はEsel mRNAの濃度とも強力に相関した(r=0.81、p<0.005、n=12)(図8d)。
心筋における最大保持粒子シグナル強度ArはEsel KO動物(0.4±0.1、0〜1、n=8)よりもWT動物(2.3±0.4(SEM)、範囲:0.8〜4.2、n=12)において有意に(p<0.005)高かった。KO動物における低いAr値は低い程度の非特異的粒子保持を示唆した。ArはWTマウス(r=−0.87、p<0.0005、n=12)ではLPS時間と強力に相関したが、KOマウス(r=−0.08、p=0.86、n=8)では相関しなかった(図8a)。図7aの曲線を使用してLPS時間をEsel mRNAの濃度に変換することにより、ArがEsel mRNAの濃度と強力に相関する(r=0.84、p<0.001、n=12)ことが図8bにより示され、その範囲の後者は細胞表面Eselタンパク質の濃度とほぼ線形的に関係した(図7c)。
自由循環粒子のインビボ半減期はWT(n=12)動物とKO(n=8)動物について約2(平均値)±1(SD)分であり、1〜5分の範囲であった。これはほとんどの市販の非標的化マイクロバブル(範囲:約1〜3分)と同等であり、作製された微粒子が商業レベルの品質以上であることを示した。心筋における保持粒子の排出は心筋内の保持粒子の最大濃度の上昇と共に低下した。その関係は非線形的であり、指数関数またはシグモイド関数にその関係を経験的に適合させることができた。これにより、最大保持粒子濃度が低いほど保持粒子の半減期が短くなった。これらの群の動物の心筋における音響学的に有効な保持粒子のインビボ半減期はKO動物(n=8)における4±4分、1〜14分の範囲と対照的にWT動物(n=12)では約7(平均値)±5(SD)分で3〜18分の範囲であった。
標的面への結合した微粒子の蓄積は基本的に粒子ボーラス投与後直ぐに完了した。保持された標的化微粒子と自由循環標的化微粒子の排出がインビボで一次カイネティクスに従うことが新しいTICベースの方法を用いてわかった。

Claims (76)
- 分子イメージングに適切な微粒子組成物であって、中央領域とシェルを有するコア微粒子構造を備える微粒子を含み、且つ、前記コア微粒子構造が
(i)ホスファチジルコリン脂質、
(ii)少なくとも1つのマレイミド部分を含むホスファチジルエタノールアミン脂質、および
(iii)アルコキシル化脂肪酸
を含む前記組成物。 - 前記コア微粒子構造のシェルに共有結合している少なくとも1つの分子結合要素をさらに含む、請求項1に記載の微粒子組成物。
- 前記少なくとも1つの分子結合要素が前記少なくとも1つのマレイミド部分を介して前記コア微粒子構造に共有結合している、請求項2に記載の微粒子組成物。
- 前記コア微粒子構造がミセル状である、請求項1から3のいずれか一項に記載の微粒子組成物。
- (i)〜(iii)のモル比が次の範囲:
(i)70〜80、
(ii)5〜15、および
(iii)10〜20
を満たす、請求項1から4のいずれか一項に記載の微粒子組成物。 - 前記コア微粒子構造が標識部分をさらに含む、請求項1から5のいずれか一項に記載の微粒子組成物。
- 前記組成物における前記標識部分のモル比が0.2〜50である、請求項6に記載の微粒子組成物。
- 前記組成物における前記標識部分のモル比が0.5〜5である、請求項7に記載の微粒子組成物。
- 前記標識部分が蛍光色素である、請求項6から8のいずれか一項に記載の微粒子組成物。
- 前記標識部分が1,1’−ジオクタデシル−3,3,3’,3’−テトラメチルインドカルボシアニンパーコレート(DiI)である、請求項9に記載の微粒子組成物。
- 前記コア微粒子構造がホスファチジルエタノールアミン脂質をさらに含む、請求項1から10のいずれか一項に記載の微粒子組成物。
- 前記ホスファチジルエタノールアミン脂質が炭素数10〜20の飽和型ホスファチジルエタノールアミン脂質である、請求項11に記載の微粒子組成物。
- 前記ホスファチジルエタノールアミン脂質がジステアロイルホスファチジルエタノールアミン(DSPE)である、請求項12に記載の微粒子組成物。
- 前記組成物における前記ホスファチジルエタノールアミン脂質のモル比が0.5〜5で
ある、請求項11から13のいずれか一項に記載の微粒子組成物。 - 前記コア微粒子構造の中央領域が液体媒質を含有する、請求項1から14のいずれか一項に記載の微粒子組成物。
- 前記液体媒質が生理的に許容可能なガスを含む、請求項15に記載の微粒子組成物。
- 前記生理的に許容可能なガスが空気、窒素、二酸化炭素、キセノン、クリプトン、六フッ化硫黄、クロロトリフルオロメタン、ジクロロジフルオロメタン、ブロモトリフルオロメタン、ブロモクロロジフルオロメタン、テトラフルオロメタン、ジブロモジフルオロメタン、ジクロロテトラフルオロエタン、クロロペンタフルオロエタン、ヘキサフルオロエタン、ヘキサフルオロプロピレン、オクタフルオロプロパン、ヘキサフルオロブタジエン、オクタフルオロ−2−ブテン、オクタフルオロシクロブタン、デカフルオロブタン、パーフルオロシクロペンタン、ドデカフルオロペンタン、およびテトラデカフルオロヘキサンから選択される、請求項16に記載の微粒子組成物。
- 前記生理的に許容可能なガスがオクタフルオロプロパンを含む、請求項16または17に記載の微粒子組成物。
- 前記少なくとも1つの分子結合要素がタンパク質、ペプチド、または低分子有機部分を含む、請求項2から18のいずれか一項に記載の微粒子組成物。
- 前記少なくとも1つの分子結合要素が抗体である、請求項19に記載の微粒子組成物。
- 成分(ii)に対する前記少なくとも1つの分子結合要素の結合反応モル比が1対1以上である、請求項2から20のいずれか一項に記載の微粒子組成物。
- 成分(ii)に対する前記少なくとも1つの分子結合要素の結合反応モル比が5対1以上である、請求項21に記載の微粒子組成物。
- 前記微粒子が微粒子当たり少なくとも約1×105個の分子結合要素を有する、請求項2から22のいずれか一項に記載の微粒子組成物。
- 前記ホスファチジルコリン脂質が炭素数10〜20の飽和型ホスファチジルコリン脂質である、請求項1から23のいずれか一項に記載の微粒子組成物。
- 前記ホスファチジルコリン脂質が1,2−ジステアロイル−sn−グリセロ−3−ホスフォコリンである、請求項24に記載の微粒子組成物。
- 少なくとも1つのマレイミド部分を含む前記ホスファチジルエタノールアミン脂質が炭素数10〜20の飽和型ホスファチジルエタノールアミン脂質である、請求項1から25のいずれか一項に記載の微粒子組成物。
- 少なくとも1つのマレイミド部分を含む前記ホスファチジルエタノールアミン脂質が少なくとも500の分子量を有するポリエチレングリコール鎖を含む、請求項1から26のいずれか一項に記載の微粒子組成物。
- 前記少なくとも1つのマレイミド部分が前記ポリエチレングリコール鎖に結合している、請求項27に記載の微粒子組成物。
- 少なくとも1つのマレイミド部分を含む前記ホスファチジルエタノールアミン脂質が1,2−ジステアロイル−sn−グリセロ−3−ホスフォエタノールアミン−N−[マレイミド(ポリエチレングリコール)−2000]である、請求項28に記載の微粒子組成物。
- 前記アルコキシル化脂肪酸がポリエチレングリコール脂肪酸エステルである、請求項1から29のいずれか一項に記載の微粒子組成物。
- 前記ポリエチレングリコール脂肪酸エステルが炭素数10〜20の飽和型ポリエチレングリコール脂肪酸エステルである、請求項30に記載の微粒子組成物。
- 前記ポリエチレングリコール脂肪酸エステルがステアリン酸PEG40である、請求項31に記載の微粒子組成物。
- 前記微粒子の平均直径が0.5〜5μmの範囲にある、請求項1から32のいずれか一項に記載の微粒子組成物。
- 分子イメージング微粒子組成物の製造に適切な中間微粒子組成物であって、
(i)ホスファチジルコリン脂質、
(ii)少なくとも1つのマレイミド部分を含むホスファチジルエタノールアミン脂質、および
(iii)アルコキシル化脂肪酸
を含む前記中間組成物。 - 少なくとも1つの分子結合要素をさらに含む請求項34に記載の中間微粒子組成物。
- 前記少なくとも1つの分子結合要素が前記少なくとも1つのマレイミド部分に共有結合している、請求項35に記載の中間微粒子組成物。
- (i)〜(iii)のモル比が次の範囲:
(i)70〜80、
(ii)5〜15、および
(iii)10〜20
を満たす、請求項34から36のいずれか一項に記載の中間微粒子組成物。 - 標識部分をさらに含む請求項34から37のいずれか一項に記載の中間微粒子組成物。
- 前記組成物における前記標識部分のモル比が0.2〜50である、請求項38に記載の中間微粒子組成物。
- 前記組成物における前記標識部分のモル比が0.5〜5である、請求項39に記載の中間微粒子組成物。
- 前記標識部分が蛍光色素である、請求項38から40のいずれか一項に記載の中間微粒子組成物。
- 前記標識部分が1,1’−ジオクタデシル−3,3,3’,3’−テトラメチルインドカルボシアニンパーコレート(DiI)である、請求項41に記載の中間微粒子組成物。
- ホスファチジルエタノールアミン脂質をさらに含む請求項34から42のいずれか一項
に記載の中間微粒子組成物。 - 前記ホスファチジルエタノールアミン脂質が炭素数10〜20の飽和型ホスファチジルエタノールアミン脂質である、請求項43に記載の中間微粒子組成物。
- 前記ホスファチジルエタノールアミン脂質がジステアロイルホスファチジルエタノールアミン(DSPE)である、請求項44に記載の中間微粒子組成物。
- 前記組成物における前記ホスファチジルエタノールアミン脂質のモル比が0.5〜5である、請求項43から45のいずれか一項に記載の中間微粒子組成物。
- 前記少なくとも1つの分子結合要素がタンパク質、ペプチド、または低分子有機部分を含む、請求項35から46のいずれか一項に記載の中間微粒子組成物。
- 前記少なくとも1つの分子結合要素が抗体である、請求項47に記載の中間微粒子組成物。
- 成分(ii)に対する前記少なくとも1つの分子結合要素の結合反応モル比が1対1以上である、請求項35から48のいずれか一項に記載の中間微粒子組成物。
- 成分(ii)に対する前記少なくとも1つの分子結合要素の結合反応モル比が5対1以上である、請求項49に記載の中間微粒子組成物。
- 前記ホスファチジルコリン脂質が炭素数10〜20の飽和型ホスファチジルコリン脂質である、請求項34から50のいずれか一項に記載の中間微粒子組成物。
- 前記ホスファチジルコリン脂質が1,2−ジステアロイル−sn−グリセロ−3−ホスフォコリンである、請求項51に記載の中間微粒子組成物。
- 少なくとも1つのマレイミド部分を含む前記ホスファチジルエタノールアミン脂質が炭素数10〜20の飽和型ホスファチジルエタノールアミン脂質である、請求項34から52のいずれか一項に記載の中間微粒子組成物。
- 少なくとも1つのマレイミド部分を含む前記ホスファチジルエタノールアミン脂質が少なくとも500の分子量を有するポリエチレングリコール鎖を含む、請求項34から53のいずれか一項に記載の中間微粒子組成物。
- 前記少なくとも1つのマレイミド部分が前記ポリエチレングリコール鎖に結合している、請求項54に記載の中間微粒子組成物。
- 少なくとも1つのマレイミド部分を含む前記ホスファチジルエタノールアミン脂質が1,2−ジステアロイル−sn−グリセロ−3−ホスフォエタノールアミン−N−[マレイミド(ポリエチレングリコール)−2000]である、請求項55に記載の中間微粒子組成物。
- 前記アルコキシル化脂肪酸がポリエチレングリコール脂肪酸エステルである、請求項34から56のいずれか一項に記載の中間微粒子組成物。
- 前記ポリエチレングリコール脂肪酸エステルが炭素数10〜20の飽和型ポリエチレングリコール脂肪酸エステルである、請求項57に記載の中間微粒子組成物。
- 前記ポリエチレングリコール脂肪酸エステルがステアリン酸PEG40である、請求項58に記載の中間微粒子組成物。
- 前記組成物が凍結乾燥物である、請求項34から59のいずれか一項に記載の中間微粒子組成物。
- 前記組成物が水性分散液である、請求項34から59のいずれか一項に記載の中間微粒子組成物。
- 請求項34から61のいずれか一項に記載の中間組成物、および生理的に許容可能なガスを含む液体媒質の供給源を含むキット。
- 前記生理的に許容可能なガスが空気、窒素、二酸化炭素、キセノン、クリプトン、六フッ化硫黄、クロロトリフルオロメタン、ジクロロジフルオロメタン、ブロモトリフルオロメタン、ブロモクロロジフルオロメタン、テトラフルオロメタン、ジブロモジフルオロメタン、ジクロロテトラフルオロエタン、クロロペンタフルオロエタン、ヘキサフルオロエタン、ヘキサフルオロプロピレン、オクタフルオロプロパン、ヘキサフルオロブタジエン、オクタフルオロ−2−ブテン、オクタフルオロシクロブタン、デカフルオロブタン、パーフルオロシクロペンタン、ドデカフルオロペンタン、およびテトラデカフルオロヘキサンから選択される、請求項62に記載のキット。
- 前記生理的に許容可能なガスがオクタフルオロプロパンを含む、請求項63に記載のキット。
- 請求項1から33のいずれか一項に記載の微粒子組成物の調製方法であって、
(i)成分(i)から(iii)を含む中央領域とシェルを有するコア微粒子構造を形成すること
を含む前記方法。 - 請求項65に記載の微粒子組成物の調製方法であって、
(ii)前記コア微粒子構造のシェルに少なくとも1つの分子結合要素を共有結合すること
をさらに含む前記方法。 - 生理的に許容可能なガスの存在下でステップ(i)が実行される、請求項65または66に記載の微粒子組成物の調製方法。
- ステップ(i)における成分(ii)に対する前記少なくとも1つの分子結合要素の結合反応モル比が1対1以上である、請求項65から67のいずれか一項に記載の微粒子組成物の調製方法。
- ステップ(i)における成分(ii)に対する前記少なくとも1つの分子結合要素の結合反応モル比が5対1以上である、請求項68に記載の微粒子組成物の調製方法。
- 請求項1から33のいずれか一項に記載の微粒子組成物、および薬学的に許容可能な担体または賦形剤を含む医薬組成物。
- 分子イメージングにおける造影剤として使用される請求項70に記載の医薬組成物。
- 分子イメージングにおける造影剤として使用される請求項1から33のいずれか一項に記載の微粒子組成物。
- 請求項1から33のいずれか一項に記載の微粒子組成物、または請求項70に記載の医薬組成物を被験者に投与すること、および
少なくとも1つの撮像技術を用いて前記被験者を撮像すること
を含む撮像方法。 - 前記撮像技術が超音波撮像法(US)を含む、請求項73に記載の撮像方法。
- 前記撮像技術が磁気共鳴撮像法(MRI)を含む、請求項73に記載の撮像方法。
- 分子イメージングに適切な微粒子組成物であって、中央領域とシェルを有するコア微粒子構造を備える微粒子を含み、且つ、前記コア微粒子構造が
(i)ホスファチジルコリン脂質、
(ii)ホスファチジルエタノールアミン脂質、および
(iii)アルコキシル化脂肪酸
を含む、前記組成物。
