Abstract
Description
毒素を本発明の抗体と共に使用して、免疫毒素を得ることができる。例示的な毒素としては、リシン、アブリン、ジフテリア毒素およびこれらのサブユニット、ならびにボツリヌス毒素A〜Fが挙げられる。これらの毒素は商業的供給源(例えば、Sigma Chemical Company,St.Louis,Mo.)から容易に入手することができる。ジフテリア毒素は、コリネバクテリウム・ジフテリアエ(Corynebacterium diphtheriae)から単離される。リシンは、リキヌス・コムニス(Ricinus communis)(トウゴマの実)からのレクチンRCA60である。この用語はまた、この毒素変異体も参照する。例えば、米国特許第5,079,163号明細書および同第4,689,401号明細書を参照されたい。リキヌス・コムニス(Ricinus communis)凝集素(RCA)は、それぞれ約65および120kDの分子量により、RCA60およびRCA120と呼ばれる2つの形態で生ずる(Nicholson & Blaustein,J.Biochem.Biophys.Acta 266:543(1972))。A鎖はタンパク質合成を不活性化し、細胞を殺滅する役割を担う。B鎖は細胞表面のガラクトース残基にリシンを結合させ、サイトゾル中へのA鎖の輸送を助ける(Olsnes,et al.,Nature 249:621−631(1974)および米国特許第3,060,165号明細書)。
PE38などの目的のPEに関して、その保存的に改変された変異体またはその細胞毒フラグメントは、アミノ酸のレベルで、少なくとも80%の配列類似性、好ましくは少なくとも85%の配列類似性、より好ましくは少なくとも90%の配列類似性、最も好ましくは少なくとも95%の配列類似性を有する。
一実施形態では、目的のポリペプチドはCD22と特異的に結合する抗体を含む。「CD22」は、Igスーパーファミリーに属する系列制限B細胞抗原をいう。これはB細胞リンパ腫および白血病の60〜70%に発現し、B細胞の成長初期段階の細胞表面や幹細胞には存在しない。例えば、Vaickus et al.,Crit.Rev.Oncol/Hematol.77:267−297(1991)を参照されたい。他の実施形態では、目的のポリペプチドは、CD22と結合する抗体フラグメント(例えば、FabまたはscFv)である。

本発明のPE免疫複合体は、細菌細胞などの細胞内で好適には発現し、その後、封入体から単離される。次いで、封入体から単離されたPE免疫複合体は、本明細書に記載の通り下流の工程でさらに精製/単離される。
ある実施形態では、活性免疫複合体、すなわち、所望の標的に結合して、細胞標的物質(例えば、抗体、抗体断片または他のタンパク質)と結合した化合物(免疫毒素)を送達することができる免疫複合体を調製する方法が本明細書に提供される。
概説
CAT−8015(モキセツモマブ・パスドトックス(Moxetumomab pasudotox))は、大腸菌(E.coli)封入体に産生される組換え免疫毒素である。活性モキセツモマブ・パスドトックス(Moxetumomab pasudotox)の生成には、好適には、不活性前駆体からのリフォールディングと、リフォールディングされた生成物の4カラムプロセスによる精製を使用する。図1は、再生および精製プロセスの概略を示す。
1.封入体可溶化
好適な細胞から産生されたVH−PE38(VH)およびVL封入体を室温で12〜24時間解凍する。VHおよびVL IB可溶化開始濃度は、リフォールド1リットル当たり0.3gのVHおよび0.07gのVLである。封入体は、1:1モル比でVHとVLを混合し、トリス/EDTA緩衝液(50mMトリス、20mM EDTA、pH7.4)を添加することにより、最終VH濃度10g/Lに調節する。濃度調節封入体溶液1kgずつに、6kgの封入体可溶化緩衝液(50mMエタノールアミン、8M尿素、0.5Mアルギニン、2mM EDTA、10mM DTT、pH9.3±0.1)を添加することにより、封入体を可溶化する。可溶化は、定速で撹拌しながら、室温で90±15分間実施する。
可溶化した封入体を一連の深層フィルターを通した濾過(国際公開第2012/059212号パンフレットを参照)により清澄化する。清澄化した濾過物は、5kDa分子量カットオフ(MWCO)限外濾過膜を用いて、最終リフォールド重量の1/10までタンジェント流濾過により濃縮する。
モキセツモマブ・パスドトックス(Moxetumomab pasudotox)は、予冷した(2〜8℃)リフォールディング緩衝液(50mMエタノールアミン、1Mアルギニン、2mM EDTA、1.0mM酸化グルタチオン、pH9.4)への清澄化および濃縮封入体濾過物の10倍(w/w)希釈によりリフォールドする。添加(L/時)は、清澄化および濃縮封入体濾過物が、4時間にわたって予冷リフォールド緩衝液に添加される(好適には、毎時リフォールド緩衝液1L当たり26mLの可溶化封入体)ように設定する。リフォールド反応は、連続的に混合しながら、2〜8℃で48〜72時間進行させた後、室温まで温めてから、濃縮およびダイアフィルターを行う。
a.Fractogel TMAE(M)クロマトグラフィ
濃縮およびダイアフィルターリフォールド溶液を0.2μmフィルターにより滅菌濾過し、10倍カラム容積(CV)のTMAE平衡化緩衝液(20mMリン酸緩衝液、pH7.4)で平衡化したFractogel TMAEカラム(EMD Biosciencesまたは同等物)にロードする。クロマトグラフィ工程は、他に注記のない限り、200cm/時の線流速で実施する。ロードした後、まずカラムを4CVのTMAE平衡化緩衝液(20mMリン酸塩、pH7.4)で洗浄した後、洗浄緩衝液1(20mMリン酸塩、0.1%Triton X−100)を用いた6CV洗浄および洗浄緩衝液2(20mMリン酸塩、100mM NaCl、pH7.4)を用いた8CV洗浄を実施する。生成物を3CVの溶出緩衝液(20mMリン酸塩、200mM塩化ナトリウム、pH7.4でカラムから溶出する。溶出後、カラムを3CVのストリッピング緩衝液(50mMエタノールアミン、0.5Mアルギニン、2mM EDTA、8M尿素、10mM DTT、pH9.3)でストリッピングする。ストリップ工程中に、流速を低下させてもよい。続いて、カラムを3CVの注射用蒸留水(WFI)で洗浄した後、3CVの再生溶液(2M NaCl)で再生させる。カラムを少なくとも3CVの殺菌溶液(1N水酸化ナトリウム)で殺菌した後、3CVの短期保存液(0.1N水酸化ナトリウム)または3CVの長期保存液(20mMリン酸塩、20%(w/v)エタノール、pH7.4)を用いて保存する。
ヒドロキシアパタイトクロマトグラフィ工程は、フロースルークロマトグラフィ工程として操作する。クロマトグラフィ工程は、他に注記のない限り、250cm/時の線流速で実施する。5CVの前平衡化緩衝液(400mMリン酸塩、200mM塩化ナトリウム、pH7.4)および5CVの平衡化緩衝液(20mMリン酸カリウム、200mM塩化ナトリウム、pH7.4)で平衡化したセラミックヒドロキシアパタイトカラム(Bio−Rad Laboratoriesまたは同等物)に、捕捉ステップ生成物をロードする。生成物をフロースルー画分中に回収する。ロードした後、カラムを3CVの平衡化緩衝液で洗浄する。カラムを3CVの前平衡化緩衝液で再生し、3CVの殺菌緩衝液(1N水酸化ナトリウム)で殺菌した後、室温で3CVの保存緩衝液(10mMリン酸塩、0.1N水酸化ナトリウム)中に保存する。
ヒドロキシアパタイト生成物をロード調製緩衝液(20mMリン酸塩、1.2M硫酸ナトリウム、pH7.4)で1:1比(w/w)に希釈し、5CVの平衡化緩衝液(20mMリン酸塩、0.6M硫酸ナトリウム、pH7.4)で平衡化したPhenyl 650Mカラム(Tosohまたは同等物)にロードする。ロードした後、カラムを1CVの平衡化緩衝液で洗浄する。生成物を0〜100%溶出緩衝液(20mMリン酸ナトリウム、pH7.4)の20CVの直線的勾配で溶出させる。カラムを2CVの注射用蒸留水で洗浄した後、2CVの8M尿素で再生させる。カラムを3CVの殺菌緩衝液(0.5N水酸化ナトリウム)で殺菌した後、3CVの保存緩衝液(20mMリン酸塩、20%(w/w)エタノール、pH7.4)を用いて室温で保存する。
ダイアフィルターしたPhenyl 650M生成物をQ Sepharose HPカラム(GE Healthcareまたは同等物)にロードした後、5CVの前平衡化緩衝液(10mMトリス、1M塩化ナトリウム、pH8.0で前平衡した後、5CV平衡化緩衝液(10mMトリス、pH8.0)で平衡する。カラムを1CV平衡化緩衝液で洗浄した後、3CVの65%(v/v)平衡化緩衝液、35%(v/v)溶出緩衝液(10mMトリス、0.5M塩化ナトリウム、pH8.0)で洗浄する。65%(v/v)平衡化緩衝液、35%(v/v)溶出緩衝液から45%(v/v)平衡化緩衝液、55%(v/v)溶出緩衝液への10CV直線的勾配を用いて、生成物を溶出する。カラムを2CVの前平衡化緩衝液でストリッピングした後、3CVの殺菌緩衝液(1N水酸化ナトリウム)で殺菌する。3CVの短期保存液(0.1N水酸化ナトリウム)または3CV長期保存液(20mMリン酸塩、20%(w/v)エタノール、pH7.4)のいずれかで、カラムを保存する。
ダイアフィルターしたQ Sepharose HP生成物を、製剤化緩衝液を用いて、最終濃度0.95〜1.05mg/mLまで希釈した後、製剤化スパイク溶液(10%(w/v)ポリソルベート−80)を用いて、0.02%(w/v)ポリソルベート−80まで調節することにより、薬剤物質を製造した。薬剤物質を0.2μmで濾過して滅菌HDPEボトルに入れ、≦−70℃で保存する。
A.材料および方法
1.フィルターおよび膜
C0HCおよびX0HC Millistak+HC PODフィルター(各々0.55m2)は、Millipore製であった。Pellicon 2 BioMax−5(Vスクリーン、2m2)およびPellicon 2 BioMax−10(Aスクリーン、0.5および2.5m2)タンジェント流濾過膜は、Millipore製であった。Durapore Millipak20、SHC Opticap XL150およびSHC Opticap XL300フィルターは、Millipore製であった。
Fractogel TMAE(M)はEMD製であった。ヒドロキシアパタイト、タイプ1、40μmは、BioRad製であった。Phenyl 650Mは、Tosoh製であった。Q Sepharose HPは、GE Healthcare製であった。Fractogel TMAE精製は、BPG 140×500カラム(GE Healthcare)で実施した。ヒドロキシアパタイトおよびPhenyl 650M精製は、BPG 100×500カラムで実施した。Q Sepharose HP精製は、Millipore QuikScale 70×550カラム(Millipore)で実施した。全ての精製は、AKTA Pilotクロマトグラフィシステムで実施した。
3.39kgのVH封入体(IB)スラリーおよび0.33kgのVL IBスラリーを3.77kgのTE緩衝液で、最終濃度10g/Lまで希釈した。44.9kgのIB可溶化緩衝液を用い、90分間室温でIBを可溶化させた。可溶化IB溶液を、X0HC深層フィルター(0.55m2)と連続して接続したC0HC深層フィルター(0.55m2)で清澄化した。可溶化および清澄化IB溶液を、Pellicon 2 BioMax−5(Vスクリーン、2m2)膜を用いたタンジェント流濾過により、最終限外濾過物(UF)1リテンテート重量25.5kgまで濃縮した。25kgのUF1リテンテートを2〜8℃に温度調節し、4時間にわたり、定速で混合しながら(好適には、毎時リフォールド緩衝液1L当たり26mLの可溶化封入体)、225kgの予冷リフォールド緩衝液(pH9.4)に添加した。リフォールドは、リフォールド溶液の温度を室温まで上昇させることにより、66時間後に停止させた。リフォールド溶液は、Pellicon 2 BioMax−10(Aスクリーン、2.5m2)膜を用いたタンジェント流濾過により、24.9kgまで濃縮した後、10倍体積のTMAE平衡化緩衝液を用いてダイアフィルターした。
モキセツモマブ・パスドトックス(Moxetumomab pasudotox)の精製を前述のように実施した。9.9Lおよび9.3Lの濃縮およびダイアフィルターしたリフォールド溶液をパックしたFractogel TMAE(M)カラムにロードした(ベッド高さ:18cm、体積:2.77L)。2つの精製サイクルを実施した(連続した溶出の間にストリッピング緩衝液を用いた)。Fractogel TMAE(M)溶出液画分を合わせて単一のTMAE生成物プールとし(7.1kg)、ヒドロキシアパタイトカラムにロードした(ベッド高さ:21.8cm、体積:1.71L)。ヒドロキシアパタイトフロースループールを、7.91kgロード調製緩衝液で1:1希釈してから、Phenyl 650Mカラムにロードした(ベッド高さ:17.5cm、体積:1.37L)。Phenyl 650M生成物プール(11.6kg)を、Pellicon 2 BioMax−10膜(Aスクリーン、0.5m2)を用いたタンジェント流濾過により、8.1kgに濃縮した後、10倍体積のQ Sepharose HP平衡化緩衝液を用いてダイアフィルターした。濃縮およびダイアフィルターしたPhenyl 650M生成物をQ Sepharose HPカラムにロードした(ベッド高さ:18.2cm、体積:0.70L)。Q Sepharose HP生成物プール(2.8kg)をPellicon 2 BioMax−10(Aスクリーン、0.5m2)膜を用いたタンジェント流濾過により、2g/Lに濃縮した後、7倍体積の製剤化緩衝液を用いてダイアフィルターした。濃縮およびダイアフィルターしたQ Sepharose HP生成物を、製剤化緩衝液を用いて最終タンパク質濃度1.02g/Lまで希釈した(量:4.7kg)後、製剤化スパイク溶液で、0.02%(w/v)ポリソルベート−80に調節した。製剤化したモキセツモマブ・パスドトックス(Moxetumomab pasudotox)をDurapore Millipak20フィルターで滅菌濾過して、PETGボトルに導入し、≦−70℃で保存した。
1.再生
可溶化、清澄化およびUF1工程の収率をRP−HPLCにより評価し、表IおよびIIに示す。深層濾過の前後に、清澄化効率を濁度測定値により評価した(表III)。
モキセツモマブ・パスドトックス(Moxetumomab pasudotox)は、濃縮およびダイアフィルターしたリフォールド溶液から200Lリフォールド規模で、Fractogel TMAE(M)、ヒドロキシアパタイト、Phenyl 650MおよびQ Sepharose HPクロマトグラフィにより精製した。各精製工程のクロマトグラムを図2〜5に示す。

3.40kgのVH IBスラリーおよび0.33kgのVL IBスラリーを3.77kgのTE緩衝液で、最終濃度10g/Lまで希釈した。45.0kgのIB可溶化緩衝液を用い、IBを90分間室温で可溶化させた。可溶化IB溶液を、X0HC深層フィルター(0.55m2)と連続して接続したC0HC深層フィルター(0.55m2)で清澄化した。可溶化および清澄化IB溶液を、Pellicon 2 BioMax−5(Vスクリーン、2m2)膜を用いたタンジェント流濾過により、25.5kgの最終UF1リテンテート重量まで濃縮した。25kgのUF1リテンテートを2〜8℃に温度調節し、4時間にわたり、定速で混合しながら(好適には、毎時リフォールド緩衝液1L当たり25mLの可溶化封入体)、225kgの予冷リフォールド緩衝液(pH9.4)に添加した。リフォールドは、リフォールド溶液の温度を室温まで上昇させることにより、66時間後に停止させた。リフォールド溶液は、Pellicon 2 BioMax−10(Aスクリーン、2.5m2)膜を用いたタンジェント流濾過により、22.6kgまで濃縮した後、10倍体積のTMAE平衡化緩衝液を用いてダイアフィルターした。
モキセツモマブ・パスドトックス(Moxetumomab pasudotox)の精製を前述のように実施した。9リットルおよび8.7リットルの濃縮およびダイアフィルターしたリフォールド溶液をパックしたFractogel TMAE(M)カラムにロードした(ベッド高さ:18cm、体積:2.77L)。2つの精製サイクルを実施し、連続した溶出の間に、開示するストリッピング緩衝液を用いてストリッピングを行った。Fractogel TMAE(M)溶出液画分を合わせて単一のTMAE生成物プール(6.2kg)とし、ヒドロキシアパタイトカラムにロードした(ベッド高さ:21.8cm、量:1.71L)。ヒドロキシアパタイトフロースループールを、6.7kgロード調製緩衝液で1:1希釈してから、Phenyl 650Mカラムにロードした(ベッド高さ:17.5cm、体積:1.37L)。Phenyl 650M生成物プール(10.1kg)を、Pellicon 2 BioMax−10膜(Aスクリーン、0.5m2)を用いたタンジェント流濾過により、8.0kgに濃縮した後、10倍体積のQ Sepharose HP平衡化緩衝液を用いてダイアフィルターした。濃縮およびダイアフィルターしたPhenyl 650M生成物をQ Sepharose HPカラムにロードした(ベッド高さ:18.2cm、量:1.4L)。Q Sepharose HP生成物プール(5.1kg)をPellicon 2 BioMax−10(Aスクリーン、0.5m2)膜を用いたタンジェント流濾過により、2g/Lに濃縮した後、7倍体積の製剤化緩衝液を用いてダイアフィルターした。濃縮およびダイアフィルターしたQ Sepharose HP生成物を、製剤化緩衝液を用いて最終タンパク質濃度1.05g/Lまで希釈した(量:5.4kg)後、製剤化スパイク溶液を用いて、0.02%(w/v)ポリソルベート−80に調節した。製剤化したモキセツモマブ・パスドトックス(Moxetumomab pasudotox)をDurapore Millipak20フィルターで滅菌濾過して、PETGボトルに導入し、≦−70℃で保存した。
1.再生
可溶化、清澄化およびUF1工程の収率をRP−HPLCにより評価し、表VIIIおよびIXに示す。深層濾過の前後に、清澄化効率を濁度測定値により評価した(表3.2.2.1〜3)。
モキセツモマブ・パスドトックス(Moxetumomab pasudotox)は、濃縮およびダイアフィルターしたリフォールド溶液から200Lリフォールド規模で、Fractogel TMAE(M)、ヒドロキシアパタイト、Phenyl 650MおよびQ Sepharose HPクロマトグラフィにより精製した。各精製工程のクロマトグラムを図7〜10に示す。
実施例2および3に記載の2つの精製について、同等の薬剤物質収率が達成された。これらのデータは、本明細書に記載の再生および精製方法の生産性、ならびにこれらの方法が、臨床試験に好適な高品質薬剤物質を生成する能力を証明している。
経済的に実現可能な精製方法の操作には、複生産サイクルにクロマトグラフィ樹脂を用いることが必要である。これは、効率的なカラム洗浄方法と共に、生成物ならびに生成物およびプロセス関連の混入物が、次の精製サイクルの開始前に、カラムから許容可能なレベル未満まで除去されるという立証を必要とする。
1.クロマトグラフィ媒体および計器類
Fractogel TMAE(M)はEMD製であった。精製は、Tricorn5/200カラムで実施した。全ての精製は、AKTA Explorerクロマトグラフィシステムで実施した。
Fractogel TMAE(M)精製を前述のように実施した。Fractogel TMAE(M)カラムに、濃縮し、ダイアフィルターしたリフォールド溶液を20gタンパク質/L樹脂までロードした。キャリーオーバー分析のために、前述と同様に、しかしタンパク質をロードせずに、また、記載されていれば、Triton X−100洗浄緩衝液なしで、Fractogel TMAEカラムをランした。
図11は、カラム洗浄および殺菌溶液として、2M NaCl(ストリップ1)、8M尿素(ストリップ2)および1N水酸化ナトリウムを用いた、Fractogel TMAE捕捉工程クロマトグラムを示す。Fractogel TMAE(M)精製順序を表XVに記載する。
陰イオン交換捕捉に代わる手段として、Cibacron Blue Dyeクロマトグラフィを用いて、やや酸性の条件下でリフォールド後UF/DF2プールからモキセツモマブ・パスドトックス(Moxetumomab pasudotox)を捕捉した。モキセツモマブ・パスドトックス(Moxetumomab pasudotox)とCibacron Blue色素リガンドとの相互作用は、天然において多モードであり、しかもかなり強度が高い。Cibacron Blue樹脂へのモキセツモマブ・パスドトックス(Moxetumomab pasudotox)の選択的結合は、イオン成分や、疎水性成分(プロピレングリコールなど)で調節することができる。本明細書に記載するようなCibacron Blue Dyeによるモキセツモマブ・パスドトックス(Moxetumomab pasudotox)の捕捉。
1.クロマトグラフィ媒体および計器類
Capto Blue Sepharoseは、GE Healthcare製であった。Capto Blue Sepharose精製は、XK16/20カラム(GE Healthcare)で実施した。精製は全て、AKTA Explorerクロマトグラフィシステムで実施した。
Capto Blue Sepharose精製は、前述のように実施した。濃縮し、ダイアフィルターし、調節したリフォールド溶液をCapto Blueカラム(カラム容積:39mL)上に、10gモキセツモマブ・パスドトックス(Moxetumomab pasudotox)/L樹脂のロード負荷までロードした。
ロード中、非結合不純物に起因し得るフロースルー吸収シグナルが認められたが、モキセツモマブ・パスドトックス(Moxetumomab pasudotox)生成物のほぼ全部が、Capto Blue Sepharose樹脂によって捕捉された。図17は、代表的なCapto Blue捕捉工程クロマトグラムを示す。ごくわずかのモキセツモマブ・パスドトックス(Moxetumomab pasudotox)しかロードフロースルー画分に検出されないため、図18のSDS−PAGEゲルにおいてモキセツモマブ・パスドトックス(Moxetumomab pasudotox)の選択的結合を認めることができる。Capto Blue Sepharoseは、高分子量種の優れた除去を示した。図18からわかるように、UF/DF2プールは、高レベルの高分子量種を含有しており、これは、フロースルー画分ならびに8M尿素ストリップピーク中でも認められたが、溶出プールには検出されなかった。その結果、溶出プールは、UF/DF2プールと比較して、非常に清浄であった。
Claims (54)
- 活性免疫複合体を調製する方法であって、前記免疫複合体は、ジスルフィド結合により連結された2つのポリペプチド鎖から構成されており、フェドバッチリフォールディングプロセスで前記免疫複合体をリフォールディングする工程と、リフォールディングされた前記免疫複合体を1つまたは複数のクロマトグラフィカラムで精製する工程とを含む、方法。
- 活性免疫複合体を調製する方法であって、前記免疫複合体は、ジスルフィド結合により連結された2つのポリペプチド鎖から構成されており、前記免疫複合体をリフォールディングする工程と、リフォールディングされた前記免疫複合体を、陰イオン交換カラムを用いて精製する工程とを含み、カラムサイクル同士の間に、前記カラムを、アルギニン、尿素およびジチオトレイトール(DTT)を含むストリッピング緩衝液でストリッピングする、方法。
- 前記免疫複合体のリフォールディングが、pH9.5以下のリフォールド緩衝液を含む、請求項2または3に記載の方法。
- 前記免疫複合体が、抗体またはその抗原結合フラグメントを含む、請求項1〜3のいずれか一項に記載の方法。
- 前記抗体または抗原結合フラグメントが、Fab、Fab’、F(ab’)2、Fd、単鎖FvもしくはscFv、ジスルフィド結合Fv、V−NARドメイン、IgNar、細胞内抗体、IgGΔCH2、ミニボディ、F(ab’)3、四重特異性抗体、三重特異性抗体、二重特異性抗体、単一ドメイン抗体、DVD−Ig、Fcab、mAb2、(scFv)2、またはscFv−Fcを含む、請求項4に記載の方法。
- 前記抗体または抗原結合フラグメントが、細胞表面受容体に結合する、請求項4または5に記載の方法。
- 前記細胞表面受容体が、CD22である、請求項6に記載の方法。
- 前記免疫複合体が、毒素を含む、請求項1〜7のいずれか一項に記載の方法。
- 前記毒素が、シュードモナス(Pseudomonas)外毒素、リシン、アブリン、ジフテリア毒素およびそのサブユニット、ならびにボツリヌス毒素A〜Fおよびこれらの変異体、および誘導体からなる群から選択される、請求項8に記載の方法。
- 前記毒素が、シュードモナス(Pseudomonas)外毒素、またはその変異体である、請求項8または9に記載の方法。
- 前記シュードモナス(Pseudomonas)外毒素、またはその変異体が、配列番号16〜22からなる群から選択されるアミノ酸配列を有する、請求項10に記載の方法。

- 前記フェドバッチプロセスにおいて、毎時リフォールド緩衝液1L当たり約52mLの可溶化封入体〜毎時リフォールド緩衝液1L当たり約13mLの可溶化封入体の添加速度を使用する、請求項1に記載の方法。
- 前記フェドバッチプロセスにおいて、毎時リフォールド緩衝液1L当たり約35mLの可溶化封入体〜毎時リフォールド緩衝液1L当たり約17mLの可溶化封入体の添加速度を使用する、請求項19に記載の方法。
- 前記フェドバッチプロセスにおいて、毎時リフォールド緩衝液1L当たり約30mLの可溶化封入体〜毎時リフォールド緩衝液1L当たり約18mLの可溶化封入体の添加速度を使用する、請求項20に記載の方法。
- 前記フェドバッチプロセスにおいて、毎時リフォールド緩衝液1L当たり約26mLの可溶化封入体の添加速度を使用する、請求項21に記載の方法。
- ストリッピング緩衝液が、約0.25〜約0.75Mアルギニン、約7〜9M尿素および約9〜11mM DTTを含む、請求項2に記載の方法。

- 活性免疫複合体を調製する方法であって、前記免疫複合体は、ジスルフィド結合により連結された2つのポリペプチド鎖から構成されており、pHが9.5以下のリフォールド緩衝液を用いて、フェドバッチリフォールディングプロセスで前記免疫複合体をリフォールディングする工程と、リフォールディングされた前記免疫複合体を、イオン交換カラムを用いて精製する工程とを含み、カラムサイクル同士の間に、前記カラムを、アルギニン、尿素およびジチオトレイトール(DTT)を含むストリッピング緩衝液でストリッピングする、方法。
- 前記調製方法から回収される免疫複合体の量が、フェドバッチリフォールディングプロセスおよび/または前記ストリッピング緩衝液を用いてストリッピングされたイオン交換カラムでの精製を含まない方法を使用して回収された免疫複合体の量と比較して、少なくとも三百%(300%)多い、請求項32に記載の方法。
- 前記ポリペプチドまたは免疫複合体が、抗体またはその抗原結合フラグメントを含む、請求項33に記載の方法。
