RAGEに結合するアプタマーを、SELEX法により選択した。SELEX法は、国際公開2006/080262号公報の実施例1と同様にして実施した。5’末端にフォワードプライマー(AGCTCAGAATGGATCCAAACGCTCA)(配列番号12)と同じ配列、3’末端にリバースプライマー(GATCCGGCCGAATTCTCATGTCGAA)(配列番号13)に相補的な配列を含み、その間に40塩基のランダム配列を含む90塩基のテンプレートDNAを合成し、このテンプレートDNAをフォワードプライマーおよびリバースプライマーを用いて増幅して、ランダムDNAアプタマープールを得た。得られたランダムDNAアプタマーを、RAGEのVドメイン(v−RAGE)を付着させたカラムに通し、結合活性の無いアプタマーは除去して、RAGEに結合するアプタマーを抽出した。v−RAGEは、RAGEのcDNAをテンプレートとして大腸菌用ベクターにサブクローニングした後、発現させ、精製した。使用したv−RAGEはヒトRAGEのaa23-121に相当し、アミノ酸配列は以下のとおりである。

3.1.方法
ヒトRPTEC細胞に100μg/mlのAGEまたは50μg/mlのウシ血清アルブミン(BSA)を投与し、RAGEアプタマー(RAGE−apt)(#4)(0.1μM)またはコントロールアプタマー(Ctr−apt)(0.1μM)とともに4時間(ROSまたはmRNAの測定の場合)または24時間(タンパク質の測定の場合)インキュベートした。ROSの産生はCM−H(2)DCFDAにより測定した。mRNAおよびタンパク質の発現は、定量的リアルタイムPCRおよびウェスタンブロッティングにより測定した。
CM−H(2)DCFDAを用いて細胞内のROSを測定した結果、AGEの投与により誘導されるROSの産生は、RAGEアプタマーの投与により抑制された(図4)。
AGEの投与により誘導される線維化促進因子または炎症性サイトカインであるTGF−β、CTGF、およびMCP−1のmRNA発現は、RAGEアプタマーの投与により、コントロールと同じレベルまで抑制された(図5)。また、AGEの投与により誘導されるRAGEのmRNA発現も、RAGEアプタマーにより抑制された(図5)。
AGEの投与により誘導される線維化促進因子のTGF−βおよびCTGFの発現がRAGEアプタマーにより抑制されることが、タンパク質レベルでも確認された(図6)。
4.1.方法
8週齢雄ウィスターラットにストレプトゾトシン(Stz)(60mg/kg)(クエン酸緩衝液中)を1回腹腔内投与することで糖尿病を誘発した。コントロールのラットにはクエン酸緩衝液のみを投与した。前記ラットを2群に分け、RAGEアプタマー(RAGE−apt)(#1)またはコントロールアプタマー(Ctr−apt)を、浸透圧ミニポンプを用いた持続注入により投与した(0.873μg/日)(コントロールラット(Con)+Ctr−apt(n=4);コントロールラット(Con)+RAGE−apt(n=4);糖尿病ラット(Stz)+Ctr−apt(n=6);糖尿病ラット(Stz)+RAGE−apt(n=6))。2週間のアプタマーの持続注入後、尿、血液、および腎臓を採取した。採取した尿中のアルブミン量および8OHdG量をELISAにより測定した。アルブミン尿は、糖尿病性腎症の早期マーカーであり、8OHdGはDNAの酸化障害マーカーである。また、腎炎のマーカーであるMCP−1の発現を定量的リアルタイムPCRにより評価した。さらに、採取した腎臓から糸球体切片を作成し、この実験系においてはabcam社製の抗RAGEポリクローナル抗体を用いた免疫染色により糸球体におけるRAGEの発現を検討した。
糖尿病を誘発したラットでは、コントロールのラットと比較して尿中アルブミンと尿中8OHdGの値が高かったが、RAGEアプタマーを2週間投与することにより、コントロールアプタマーを投与したラットと比較して尿中アルブミンと尿中8OHdGの値が有意に減少した(図7)。
糖尿病を誘発したラットでは、コントロールのラットと比較して糸球体におけるRAGEの発現が上昇したが、RAGEアプタマーの投与によりコントロールのレベルまでRAGEの発現が減少した(図8)。
糖尿病を誘発したラットでは、コントロールのラットと比較してRAGEおよびMCP−1のmRNA発現が上昇したが、RAGEアプタマーの投与によりこれらの発現上昇は有意に抑制された(図9)。
5.1.方法
ヒトMesangiumu細胞に50μg/mlのAGEまたはウシ血清アルブミン(BSA)を投与し、RAGEアプタマー(RAGE−apt)(#1)(0.1μM)またはコントロールアプタマー(Ctr−apt)(0.1μM)とともに4時間(ROSの測定の場合)または24時間(mRNAの測定の場合)インキュベートした。ROSの産生はCa−H(2)DFFDAにより測定した。mRNAの発現は、定量的リアルタイムPCRにより測定した。
Ca−H(2)DFFDAを用いて細胞内のROSを測定した結果、AGEの投与により誘導されるROSの産生は、RAGEアプタマーの投与により抑制された(図10)。
AGEの投与により誘導されるRAGEおよび炎症性サイトカインまたは細胞接着性因子であるMCP−1、ICAM−1、VCAM−1のmRNA発現は、RAGEアプタマーの投与により、コントロールと同じレベルまで抑制された(図11)。
6.1.方法
片腎摘した8週齢雄のC57Bl/6Jマウスに、アルドステロンの前駆体である酢酸デオキシコルチコステロン(DOCA)50mgを皮下投与し、アルドステロン誘導性高血圧モデルマウスを作成した。図12に示す投与計画に従って、浸透圧ポンプを用いて、スピロノラクトン(20mg/kg/日、ゾンデにて1日1回経口投与)、RAGEアプタマー(#4)またはコントロールアプタマーを投与した。21日後に屠殺し、尿、血液、および腎臓を採取した。
AGEの1つであるCMLの組織沈着を調べるために、4μmの腎組織切片を用いて免疫染色を行った。Abcamより販売されている抗CML抗体を使用し、発色にはDABのキットを使用した。また、抗RAGEポリクローナル抗体(Abcam)を用いた免役染色によりRAGEの発現を調べた。染色結果をTIFにて取り込み、ImageJ を用いて染色された面積を計算し、群間での比較を行った。結果を図13に示す。DOCA投与により糸球体内でCML沈着が著明に亢進したが、RAGEアプタマーによって有意に抑制された。また、DOCA投与によって糸球体内のRAGE発現は著明に亢進したが、RAGEアプタマーによって有意に抑制された。
酸化ストレスの1つであるONOO−のマーカーであるニトロタイロシンを染色することにより、ニトロタイロシンとCMLの二重染色を行った。抗ニトロタイロシン抗体(abcam)と前述の抗CML抗体を1次抗体として、4μmの腎組織切片をインキュベートし、Alexa fluor の488と561を二次抗体として、二重染色を行った。結果を図14に示す。DOCA投与によりポドサイト(糸球体上皮細胞)でニトロタイロシンの増加を認め、それはCMLと共局在していた。RAGEアプタマーはニトロタイロシンの発現を有意に抑制した。
4μmの腎組織切片を1%BSA/PBSでブロッキングした後に、1:100で希釈した抗ポドシン抗体(京都大学大学院淺沼克彦先生より譲渡)と終夜インキュベーションした。その後、Alexa-fluor488で発色し、ImageJを使用し、蛍光発色量および強度を測定した。また、採取した尿から尿中アルブミン排泄量をELISAにより測定した。結果を図15に示す。DOCA投与によりポドシンが著明に減少したが、RAGEアプタマーによって有意に抑制された。また、RAGEアプタマー投与により尿中アルブミン排泄が減少した。
酸化ストレスのマーカーである8OHdGを用いて、腎組織における酸化ストレスを測定した。トリプルバッファー(Triple buffer)およびプロテアーゼ阻害剤を用いてバッファーを作成し、腎組織をホモジナイズした。BCAにてタンパク質濃度を一定とした後に、8OHdGを1次抗体としてウェスタンブロッティングを行った。結果を図16に示す。DOCA/食塩群で著明に増加した8OHdgは、RAGEアプタマー投与にて有意に抑制された。
6.5.と同様の方法でウェスタンブロッティングを行い、腎組織におけるRAGE発現を検出した。結果を図17に示す。また、コスモバイオ社から販売されているCML測定ELISAキットを使用して、CMLの血漿中濃度を測定した。結果を図18に示す。DOCAによるAGE−RAGE系の亢進は、腎臓組織において著明であり、RAGEアプタマーによって有意に抑制された。
高血圧性腎障害の代表的な組織障害所見の1つである細胞外基質の増加を調べるために、4μmの腎組織切片をPAS染色し、糸球体、特にメサンギウム領域の拡大を、ImageJを用いる面積測定により調べた。結果を図19に示す。細胞外基質はDOCA/食塩群で増加し、RAGEアプタマー投与により有意に抑制された。
4μmの腎組織切片を1%BSA/PBSにて2時間ブロッキングし、ミネラルコルチコイド受容体(MR)(Abcam)と終夜インキュベーションした。その後、EnVisionTM G|2 Doublestain System(Dako)にて発色を行い、ImageJを使用し染色面積を測定した。また、M.O.M. Immunodetection Kitで2時間ブロッキングを行った後に、Rac1(NewEast Biosciences)に対する抗体を使用し、終夜インキュベーションを行った。その後、EnVisionTM G|2 Doublestain System(Dako)にて発色し、ImageJを使用し染色面積を測定した。MRの発現は、ウェスタンブロッティングによっても確認した。結果を図20に示す。DOCAによって誘導されるMRの過剰発現は、RAGEアプタマーによってAGE−RAGE系を抑制することにより、有意に抑制された。また、DOCAによって誘導されるRac1の過剰発現は、RAGEアプタマーにより有意に抑制された。
RAGEアプタマーは、RAGEとそのリガンドであるAGEまたはHMGB1との結合を阻害し、また、RAGEとAGEとの結合を阻害することにより、細胞内の酸化ストレスの産生、およびRAGEの下流因子である炎症性サイトカインの発現を抑制した。RAGEアプタマーは、RAGEを介したシグナル伝達を阻害することができ、RAGE関連疾患の治療または予防に有用である。実際に、RAGEアプタマーは、糖尿病または高血圧のモデル動物において腎障害の発症を抑制した。
