Abstract
Description
本出願は、「Multiplex Pyrosequencing Using Non-Interfering Noise-Cancelling Polynucleotide Identification Tags」という題名の、2012年9月24日に出願された米国特許仮出願第61/705,089号の優先権を主張し、該出願は参照により本明細書に組み入れられる。
本出願は、EFS-Webを介してASCII形式において提出されかつその全体が参照により本明細書に組み入れられる配列表を含有する。2013年9月23日に作成された該ASCIIコピーは、15892-0002_SL.txtという名称であり、サイズは15,781バイトである。
本発明は、核酸試料を解析するためおよびシーケンシングするための方法論に関する。より具体的には、本発明は、特異的なポリヌクレオチド識別タグの使用を通して増幅核酸試料をシーケンシングするための方法に関する。
核酸増幅の方法、例えばポリメラーゼ連鎖反応(PCR)の開発により、分子診断試験を含む、様々な用途のためのDNA増幅の使用が可能になっている。しかしながら、分子鑑別診断(MDD)アッセイのためのPCRの使用には課題が付随している。PCRは、特異的なプライマーまたはプライマーセット、温度条件、および酵素を利用する。例えば、PCR反応は容易に汚染され、プライマー結合は、異なるプライマーについて異なる条件を必要とする可能性があり、かつ、プライマーは、その標的配列のみを増幅するために標的配列に特異的であるべきである。これらの限界により、単一の試料から多数の配列を増幅することがさらにいっそう困難になっている。
パイロシーケンシングは、核酸合成中に放出されるピロリン酸の検出に基づく核酸シーケンシング技術である。組み込まれたヌクレオチドの数に比例する可視光が、この酵素反応中に生じる。無機ピロリン酸が、酵素ポリメラーゼによるヌクレオチドの組み込みの結果として放出される。いくつかの態様において、ルシフェラーゼが、フォトダイオード、光電子増倍管、または電荷結合素子(CCD)カメラにより容易に検出される光を生じるために利用される。付加されたヌクレオチドが既知であるため、鋳型核酸の配列が決定され得る。パイロシーケンシングは、DNAおよびRNAの両方で可能である。本質的に、相補鎖を一度に一塩基対合成すること、および、ヌクレオチドの組み込み中に放出される光の測定により各段階でどの塩基が付加されたかを検出することによって、前記方法により一本鎖の核酸のシーケンシングが可能になる。パイロシーケンシングは、臨床試料からの同定を手助けする、細菌、ウイルス、真菌、および他の類似した供給源由来の核酸適用のための迅速で正確な方法の代表例である。しかしながら、パイロシーケンシングは、混合した試料を解析するために、概して多数の並行したシーケンシングランを必要とする、並行シーケンシング法である。このシーケンシング法を、マルチプレックスPCRシーケンシング法のように単一の格納容器中で行うことは、必要な結果を生じるために必要とされる費用および時間を有意に低減することができる。本発明者らは、パイロシーケンシングが真のマルチプレックス様式で行われること、すなわち、多数の並行反応ではなく、単一のマルチプレックス反応において多数の試料をシーケンシングおよび検出することを可能にする方法を、開発した。

d.標的固有の非干渉性、非キャンセル性のポリヌクレオチド識別タグをパイロシーケンシングする段階
を含み、標的固有の非干渉性、非キャンセル性のポリヌクレオチド識別タグの識別が標的ポリヌクレオチドの存在を示す、方法。
