Abstract
Description
本PCT出願は、2012年6月12日に第61/658,569号として出願された同時係属米国仮出願および2013年4月8日に第61/809,604号として出願された同時係属米国仮出願の優先権の利益を主張するものであり、これらの各出願は引用することによりその全内容が本明細書の一部とされる。
本発明は、治療上および診断上の有用性を有する抗体に関する。より詳しくは、本発明は、ミスフォールド型のヒトPrPタンパク質によってユニークに提示されるエピトープに選択的に結合する抗体に関する。抗体、それらの結合フラグメントおよびそれらに基づく免疫複合体は、癌、ならびにクロイツフェルト−ヤコブ病(CJD)などの伝達性海綿状脳症を含むPrPのミスフォールディングおよび凝集に関連する疾患の治療および検出において治療上および診断上有用である。
先進国の人口の約3分の1が癌による死を運命付けられている。現行の癌の治療は、化学療法および放射線療法を含め、正常細胞よりも優先的に癌細胞を殺すこと、腫瘍成長の緩徐化のまさにぎりぎりのところまで顕著な副作用を受け入れるいわゆる「治療域」に基づいている。正常細胞を温存する特異的な治療が差し迫って必要である。
本発明は、ミスフォールド型のPrPに選択的に結合する抗体を提供する。より詳しくは、ここで、そのミスフォールド状態においてのみPrPにより提示されるエピトープに選択的に結合する抗体を提供する。本抗体は、その野生型、すわなち、本来のフォールドコンフォメーションのPrPとの結合親和性はほとんど示さないか、全く示さない。このエピトープは、アミノ酸配列MDEYSNQNN(配列番号14)で定義され、リジッドループとして知られるPrPの領域に存在する。このエピトープに対して生成した抗体はミスフォールドPrPに対して結合優先性を示すことが見出されている。これらの抗体、ならびにそれらに基づくそれらの結合フラグメントおよび免疫複合体は、様々な診断および治療適用に有用性を見出す。

本明細書で使用する場合、用語「PrP」は、発現しプロセシングを受けたPRP遺伝子産物を含んでなる成熟ヒトタンパク質を意味し、この成熟タンパク質は、UniProtKB/Swiss−Prot P04156の残基1−230と呼称される。本目的で、用語「PrP」は、ミスフォールド状態において本抗体に対する結合親和性を保持する、このタンパク質の天然変異体をさらに含む。

本抗体、結合フラグメントまたは複合体の治療処方物は、保存のため、所望の純度を有する抗体または複合体と任意選択の薬学的に許容可能な担体、賦形剤または安定剤(Remington’s Pharmaceutical Sciences, 第16版, Osol, A. Ed. 本抗体、結合フラグメントまたは免疫複合体は、生理学上耐用性のある、例えば、薬学的に許容可能な希釈剤、担体、または賦形剤とともに、単位投与形で、また、特定の医学的状態の治療もしくは診断に適合した総合的処置計画の一部として、または診断目的で被験体のイメージングを行うために投与することができる。
本抗体、または結合フラグメントもしくは複合体は、それを必要とする被験体に、有用な他の薬剤と組み合わせて投与することができる。1以上のさらなる治療薬と「組み合わせて」投与するとは、同時(共)投与および任意の順序での連続投与を含む。他の治療計画を本発明の抗癌剤、例えば、抗体または複合体の投与と組み合わせてもよい。例えば、このような抗癌剤で治療される患者は、外部照射療法などの放射線療法もまた受けてよい。代わりに、または加えて、化学療法薬を患者に投与してもよい。このような化学療法薬の調製および投与計画は、製造者の説明書に従うか、または熟練の医師によって経験的に決定されるように使用することができる。このような化学療法の調製および投与計画はまた、Chemotherapy Service Ed., M. C. Perry, Williams & Wilkins, Baltimore, Md. (1992)にも記載されている。化学療法薬は、例えば抗体などの抗腫瘍薬剤の投与(administration or the anti-tumor agent)に先行または後続してよく、あるいはそれと同時に投与されてもよい。本抗体は、複合体に関して上記された毒素のうちいずれと組み合わせてもよく、または任意の他の好適な薬物としては特に、イリノテカン(CPT−11)、シスプラチン、シクロホスファミド、メルファラン、ダカルバジン、ドキソルビシン、ダウノルビシン、およびトポテカン、ならびにチロシンキナーゼ阻害剤が含まれる。
本発明の別の実施態様では、本明細書に記載の障害の診断または治療に有用な材料を含有する製品が提供される。この製品は、本抗体、またはその結合フラグメントもしくは免疫複合体を、容器内に、かつ好適には標識を担持して含んでなる。好適な容器としては、例えば、ボトル、バイアル、シリンジ、および試験管が含まれる。これらの容器は、ガラスまたはプラスチックなどの様々な材料から形成されてよい。容器は病態を検出または治療するために有効な組成物を保持し、無菌アクセスポートを持ち得る(例えば、容器は、皮下注射針により貫通可能なストッパーを備えた静脈内溶液バッグまたはバイアルであり得る)。容器上の、または容器に付随するラベルは、組成物が癌病態を治療するためまたはCJDなどの伝達性海綿状脳症を治療するために使用されることを示す。本製品は、リン酸緩衝生理食塩水、リンゲル液およびデキストロース溶液などの薬学的に許容可能なバッファーを含んでなる(compromising)第2の容器をさらに含んでなって(compromise)よい。本製品はさらに、他のバッファー、希釈剤、フィルター、針、シリンジ、および本発明に従った使用説明を記した添付文書を含め、商業上および使用上の観点から望ましい他の物品を含んでもよい。PrP調製標品など、本方法において有用な対照薬剤または標品も本キットに含むことができる。
標的エピトープに選択的に結合する抗体およびそのフラグメントは、本発明の態様に従って、癌または他の細胞をスクリーニングしてミスフォールドPrPを提示するものを検出するために使用される。好ましい実施態様では、スクリーニングは、PrP抗体療法の候補である被験体から採取した癌細胞のサンプルに適用される。ミスフォールド型のPrPを提示する癌細胞に関して陽性判定の被験体は、次に、本抗体、またはそのフラグメントもしくは免疫複合体による療法の予定を組むことができる。本明細書に記載の抗体または他の結合剤と組み合わせた標準的な技術を癌細胞のスクリーニングに使用することができる。望ましくは、本抗体に検出可能な標識を組み込む。この標識はそれ自体検出可能であり得(例えば、放射性同元素標識または蛍光標識)または酵素標識の場合には、検出可能な基質化合物または組成物の化学変化を触媒し得る。検出可能な標識として機能し得る放射性核種としては、例えば、I−131、I−123、I−125、Y−90、Re−188、Re−186、At−211、Cu−67、Bi−212、およびPd−109が含まれる。
配列MDEYSNQNN(配列番号14)を含んでなるペプチドを、標準的な方法を用いて合成した後、担体タンパク質と結合させた。調製した免疫原は、KLH−Cys−MDEYSNQNNおよびOVA−Cys−MDEYSNQNNの両方を含んでいた。
Maxisorp 96ウェルプレートを、2〜8℃にて一晩、PBS中、100ng/ウェルのBSA−ペプチドでコーティングした。PBST/カゼインでブロッキングした後、一次抗体を加え、室温で1時間インキュベートした。ウサギ抗体は、ヤギ抗ウサギIgG−HRPとTMB基質を用いて検出した。0.25M硫酸で反応を停止させた後、450nmで吸光度を測定した。
組換えPrP(Alicon)をLDSサンプルバッファー(Life Technologies)およびサンプル還元剤(Life Technologies)と混合し、80℃で20分間加熱した。15分間冷却した後、Maxisorp 96ウェルプレートを100ng/ウェルの変性PrPでコーティングし、2〜8℃にて一晩インキュベートした。PBST/BSAでブロッキングした後、一次抗体を加え、室温で1時間インキュベートした。後の行程は、抗ペプチドELISAに関して記載した通りとした。
Maxisorp 96ウェルプレートを2〜8℃にて一晩、PBS中、100ng/ウェルのヤギ抗His−6抗体(QED)でコーティングした。PBST/BSAでブロッキングした後、His−PrP(Alicon)を加え、室温で1時間インキュベートした。一次抗体の添加および後の行程は、抗ペプチドELISAに関して記載した通りとした。
接着腫瘍細胞株および初代細胞を、非酵素系細胞解離バッファー(Invitrogen)を用いてフラスコから解離された。末梢血単核細胞は、採取当日に標準的なフィコール遠心分離法を用い、新鮮なクエン酸血から調製した。他の初代細胞は10%DMSO中で凍結させ、試験当日に解凍した。移植腫瘍をマウスから外科的に摘出した。腫瘍をはさみで刻んだ後、37℃にて30分間、振盪しながらコラゲナーゼ/ヒアルロニダーゼ(Worthington Biochemical)で処理した。この混合物を40μmスクリーンに通すことにより、個々の腫瘍細胞を回収した。
Fc受容体を有する細胞を10%正常ヒト血清で処理して受容体をブロッキングした。細胞を2〜8℃で30分間、一次抗体とともにインキュベートした。洗浄した後、細胞を2〜8℃で30分間、ヤギ抗ウサギAF488とともにインキュベートした。最終の洗浄後、細胞を1μg/mLのヨウ化プロピジウム中でインキュベートした。Becton Dickinson FACSCaliburまたはBecton Dickinson FACS Canto IIのいずれかとFCS Expressソフトウエア(DeNovo Systems)を用いて細胞を分析した。
ペプチドまたは変性PrPに対する抗体の結合は、上記のようにELISAにより行った。腫瘍細胞に対する抗体の結合は、上記のようにFACSにより行った。抗体の力価を測定して結合曲線を得た。EC50値は、GraphPadソフトウエアを用いて計算した。
接着腫瘍細胞株を、非酵素系細胞解離バッファー(Invitrogen)を用いてフラスコから解離させた。初代細胞を10%DMSO中で凍結させ、試験当日に解凍した。細胞を37℃で30分間、種々の濃度のプロテイナーゼKで処理した。その後、細胞を洗浄し、上記のようにFACSにより抗体結合を測定した。
細胞を6ウェルプレートに播種し、一晩接着させた。翌日、培地を除去し、種々の濃度のパクリタキセルを含有する新鮮培地に置換した。細胞を37℃/5%CO2で一晩インキュベートした。翌日、非酵素系細胞解離バッファーを用いて細胞を解離させ、洗浄し、抗体結合を上記のようにFACSにより評価した。
合成
Ab 1c−120とウレアーゼを、リンカーとしてSIAB(スクシンイミジル−(4−ヨードアセチル)アミノベンゾエート)を用いてコンジュゲートすることにより、免疫複合体を形成した。SIABは、N−ヒドロキシスクシンイミド(NHS)エステルおよびヨードアセチル反応性基を介したアニム(anime)−スルフヒドリルコンジュゲーション用の、中程度の長さの架橋剤である。このSIABは、約10.6オングストローム長のスペーサーアームを生じる。SIABは、Thermo Scientificから市販されており、コンジュゲーションにおけるその使用は、例えば、Hermanson, Bioconjugate Techniques, 1996, San Diego, Academic Press pp 542, 553, 568に記載されている。
タンパク質濃度はBCAにより0.5mg/ml;
ウレアーゼ酵素活性は1030U/ml;
ウレアーゼ比活性は2060U/mg;
平均コンジュゲーション比は2IgG/ウレアーゼ;
総生成物:6.0ml溶液中、3mg
バッファー組成物:20nMアルギニン、0.2mM EDTA、1%スクロース、pH7.0。
ウレアーゼまたはウレアーゼ複合体の酵素活性を、グルタミン酸デヒドロゲナーゼ(GLDH)と組み合わせた酵素反応で行った。酸化したNADHの量を、340nmでの吸光度の変化を測定することにより決定した(Kaltwasser, H. and Schlegel, H. G., Anal. Biochem., 16, 132, 1966)。用いた試薬は、0.10Mリン酸カリウムバッファーpH7.6;リン酸バッファー中に調製した1.80M尿素;バッファー中0.025Mアデノシン−5’−ジホスフェート(ADP)(10.7mg/ml);リン酸バッファー中0.008M NADH(5mg/ml);リン酸バッファー中0.025M α−ケトグルタル酸塩(3.7mg/ml);アンモニウムイオン不含グルタミン酸デヒドロゲナーゼ(GLDH)溶液;アッセイ前に新たに調製した50U/mlリン酸バッファーであった。ウレアーゼ溶液は、0.1〜0.5U/mlの濃度となるようにリン酸バッファーに溶かすことによって調製した。この溶液はアッセイ前に新たに調製した。
試薬を2時間、腫瘍細胞とともにインキュベートした。細胞を2回洗浄した後、20mM尿素とともに30分間インキュベートした。細胞生存率は、WST−1を添加し、16〜20時間後に吸光度を測定することにより評価した。
ES−2細胞を細胞培養で増殖させた。試験0日目に、5×106細胞を皮下移植した。腫瘍が平均100mm3に達したところで、週に3回、iv投与を開始した。腫瘍成長は、カリパスで腫瘍寸法を測定することによりモニタリングした。腫瘍体積は、式L×W2/2に従って計算した。マウスは腫瘍が800mm3に達した際に終了とするか、または重度に潰瘍形成させた。
ProMis(商標)アルゴリズム(WO2010/040209に記載)を用いて、ヒトPrPのDSEを同定した。DSE3はリジッドループエピトープと呼ばれ、β−シート2とα−ヘリックス1の間に位置する。
Claims (25)
- ミスフォールドヒトプリオンタンパク質(PrP)エピトープMDEYSNQNNと結合する単離された抗体であって、重鎖と軽鎖を含んでなり、各鎖は定常領域と可変領域を有し、各可変領域はフレームワーク領域と相補性決定領域(CDR)を含んでなり、前記CDRは下記に示されるアミノ酸配列を含んでなる抗体:
重鎖としては:
CDR1 TYAMG(配列番号1)
CDR2 VITKSGNTYYASWAKG(配列番号2)
CDR3 YGIGVSYYDI(配列番号3)
軽鎖としては:
CDR1 QSSQSLYNKNWLS(配列番号4)
CDR2 KASTLES(配列番号5)
CDR3 QGEFSCSSADCTA(配列番号6)。 - 前記軽鎖可変領域が配列番号7を含んでなる、請求項1に記載の抗体。
- 前記重鎖可変領域が配列番号8を含んでなる、請求項1または請求項2に記載の抗体。
- 前記軽鎖が配列番号9を含んでなる、請求項1に記載の抗体。
- 前記重鎖が配列番号10含んでなる、請求項1または請求項4に記載の抗体。
- エピトープMDEYSNQNN、または(1)少なくとも5個の連続する残基を含んでなり、かつ、(2)請求項1に記載の抗体に対して結合親和性を保持するその一部に選択的に結合する、請求項1〜5のいずれか一項に記載の抗体のフラグメント。
- 請求項1〜5のいずれか一項に記載の抗体または請求項6に記載のフラグメントと、それとコンジュゲートされた毒素とを含んでなる、免疫複合体。
- ウレアーゼと、それとコンジュゲートされた請求項1〜5のいずれか一項に記載の抗体または請求項6に記載のフラグメントとを含んでなる、免疫複合体。
- 請求項1〜5のいずれか一項に記載の抗体または請求項6に記載のフラグメントと、それとコンジュゲートされた検出可能な標識とを含んでなる、免疫複合体。
- 請求項1〜8のいずれか一項に記載の抗体、フラグメントまたは免疫複合体と薬学的に許容可能な担体とを含んでなる、医薬組成物。
- ミスフォールドヒトPrPにより媒介される障害の治療のための、請求項1〜8のいずれか一項に記載の抗体、フラグメントまたは免疫複合体の使用。
- 癌またはCJDの治療のための、請求項11に記載の使用。
- 癌の治療のための、請求項12に記載の使用。
- 卵巣癌の治療のための、請求項13に記載の使用。
- ミスフォールドPrPの検出のための、請求項1〜6および9のいずれか一項に記載の抗体、フラグメントまたは免疫複合体の使用。
- 請求項1〜9のいずれか一項に記載の抗体、フラグメントまたは免疫複合体と、ミスフォールドPrPを検出するためのその使用に関する説明書とを含んでなる、診断キット。
- 生体サンプルにおいてミスフォールドPrPを検出するための方法であって、前記サンプルを請求項1〜9のいずれか一項に記載の抗体、フラグメントまたは免疫複合体とともにインキュベートすること、および抗体結合複合体の形成を検出することを含んでなる、方法。
- ミスフォールドPrP表現型を有する罹患細胞を呈する被験体を治療するための方法であって、前記被験体に薬学的に許容可能な担体と請求項1〜8のいずれか一項に記載の抗体、フラグメントまたは複合体とを含んでなる医薬組成物を、前記罹患細胞の成長または増殖を阻害するために有効な量で投与することを含んでなる、方法。
- 前記罹患細胞が癌細胞である、請求項18に記載の方法。
- 前記癌細胞が白血病細胞である、請求項19に記載の方法。
- 前記癌細胞が卵巣癌細胞である、請求項19に記載の方法。
- 前記被験体がパクリタキセルでも治療される、請求項21に記載の方法。
- PrP凝集が関係する疾患を呈する被験体を治療するための方法であって、前記被験体に薬学的に許容可能な担体と請求項1〜9のいずれか一項に記載の抗体、フラグメントまたは複合体とを含んでなる医薬組成物を、PrP凝集の進行を阻害するために有効な量で投与することを含んでなる、方法。
- 前記疾患が伝達性海綿状脳症である、請求項23に記載の方法。
- 前記疾患がCJDである、請求項24に記載の方法。
