Abstract
Description
HIV−1 gp120の内側ドメインの構造は、公開されているgp120構造から失われている[4、9および10]。しかしながら、その内側ドメインの層の相互作用は、CD4リガンドが結合する際にgp120において生じる立体配座の変化において重要である。本発明は、HIV−1 gp120および可溶性HIV−1 gp140の内側ドメインにおける層間相互作用の安定化に関する。
gp120の内側ドメインの新しく解明された構造は、CD4結合状態において、内側ドメインが、β−サンドイッチ構造から標的細胞膜に向かって突き出た層1、2および3からなる3つの層に組織化されることを示す(図1)[11−13]。内側ドメインは、SF162株由来のHIV gp120のアミノ酸1〜248および458〜497に対応するアミノ酸で構成される(配列番号2)。層1は、SF162株由来のHIV gp120のアミノ酸42〜76に対応するアミノ酸で構成される。層2は、SF162株由来のHIV gp120のアミノ酸90〜117および192〜223に対応するアミノ酸で構成される。層3は、SF162株由来のHIV gp120のアミノ酸239〜249および459〜470に対応するアミノ酸で構成される。内側ドメインは、ポリペプチドがリガンド非結合(un−liganded)状態からCD4結合状態に移行する際に広範な立体配座変化を起こすgp120の領域として同定された[14,15]。CD4結合時に起こる内側ドメインの立体配座変化は、標的細胞へのHIVウイルスの融合が成功するために必要な下流の2つの事象に寄与する。これらの事象は:
1.最初の弱いCD4接触の安定化、および
2.コレセプター結合のためのgp120上への保存された部位の露出、すなわち、CD4結合エピトープの露出
である。

本発明は、CD4結合エピトープとCD4結合部位エピトープの両方を同時に露出する立体配座で安定化されたHIV gp120または可溶性gp140ポリペプチドを含む単離されたポリペプチドを提供する。「CD4結合エピトープとCD4結合部位エピトープの両方を同時に露出する立体配座で安定化された」とは、HIV gp120または可溶性gp140ポリペプチドの内側ドメインが、CD4リガンドの非存在下でさえもCD4が結合した立体配座を取ることを意味する。したがって、野生型gp120または可溶性gp140ではCD4に結合したときだけ露出される立体配座的にマスクされたエピトープは、本発明の安定化されたgp120または可溶性gp140ポリペプチドでは恒常的に露出される。さらに、本発明の安定化されたgp120または可溶性gp140ポリペプチドは、CD4の非存在下においてCD4が結合した立体配座を取るので、CD4結合エピトープと同時に、CD4結合部位エピトープも露出され得る。
(i)抗gp120 CD4結合部位抗体、すなわち、gp120におけるCD4結合部位に特異的な抗体、
(ii)gp120におけるCD4誘導型の保存された結合部位に特異的な抗体、および
(iii)必要に応じて、さらなる抗gp120抗体または抗可溶性gp140抗体
に特異的に結合するポリペプチドである。
本発明は、本発明のポリペプチドをコードする核酸も提供する。本発明は、本発明のポリペプチドをコードするヌクレオチド配列と配列同一性を有するヌクレオチド配列を含む核酸を提供する。そのような核酸は、同じアミノ酸をコードするために代替のコドンを使用するものを含む。
本発明は、免疫原性組成物も提供する。本発明の免疫原性組成物は、本発明のポリペプチド、その免疫原性フラグメント、本発明の融合タンパク質、本発明の三量体ポリペプチド、本発明のポリヌクレオチドおよび/またはそれらの組み合わせを含み、それらは、本明細書中で抗原と称され得る。そのような免疫原性組成物は、ワクチンとして有用であり得る。これらのワクチンは、予防的(すなわち、感染を予防する)または治療的(すなわち、感染を処置する)であり得るが、代表的には、予防的であり得る。
本発明の単離されたポリペプチド、その免疫原性フラグメント、本発明の融合タンパク質、本発明の三量体ポリペプチド、本発明のポリヌクレオチド、本発明の免疫原性組成物および/またはそれらの組み合わせは、被験体において免疫応答を発生させる方法、ならびに被験体におけるHIVによる感染および/またはAIDSを処置するおよび予防する方法において使用され得る。好ましい実施形態において、被験体は、ヒトである。
用語「〜を含む(comprising)」は、「〜を含む(including)」ならびに「〜からなる」を包含し、例えば、X「を含む」組成物は、もっぱらXからなり得るか、またはさらなる何かを含み得る(例えば、X+Y)。
システイン変異およびジスルフィド架橋のための部位の特定
ジスルフィド架橋が2つのシステイン残基間に形成されるために、側鎖は、互いに対して適切な距離かつ正しい向きで存在しなければならない。本発明者らは、層1、2および3の界面に存在するすべての残基を調査し、ジスルフィド形成のためのシステインによる置換の標的になり得る残基を同定しようと試みた。本発明者らは、gp120の結晶構造[13]を使用し、この選択のために以下の基準を使用した:
a.以前に解明されたgp120構造においてジスルフィド架橋を形成したシステインのCβ(炭素−ベータ)原子間の平均距離は、約4.22Åであり、範囲は3.9Å〜4.7Åである。ゆえに、本発明者らは、この範囲内に入るCβ−Cβ距離を有する相互作用層における残基についてスクリーニングした
b.本発明者らは、システイン残基による置換によって人工的なジスルフィド架橋が導入されたgp120の結晶構造も調査した[6]。この構造では、2対の残基(W90−E268)および(I103−Q413)を、システイン(W90C−E268C)および(I103C−Q413C)に置換した。天然の残基およびシステイン置換された残基のCβ距離は、それぞれ4.1Åおよび4.3Åだった。さらに、システインで置換された残基におけるジスルフィド架橋は、適切に形成され、これらのジスルフィド架橋されたgp120のCD4が結合した構造は、天然のgp120にCD4が結合した構造と識別できなかった(図2)。
上に記載されたように、層の界面において近距離(およそ(approxaimately)4〜5オングストローム)で存在し、システインで置換された場合にジスルフィド結合を形成することができるであろう残基を、入手可能な構造情報[13]を用いて視覚的に選択した。本発明者らはまた、Disulfide−by−Designソフトウェアを使用して、システインによって置換された場合にジスルフィド結合を形成することができるのに十分近いと推定される残基をさらに推測した。
本発明者らは、システイン置換のための残基を決定したら、Quick Change部位特異的変異誘発(Stratagen)に従って、相補的なプライマーをデザインした。野生型SF162 gp120をコードするプラスミドを鋳型として使用し、変異誘発の各回の後に、クローンを選択し、配列決定して、確実に正しい変異が存在し、意図されない変異が存在しないようにした。それらの正しいクローンをラージスケールのプラスミドDNA調製のためにさらに増幅させた。
システイン置換が層1と層2とを架橋するために選択される標的は、層1のバリン59および層2のセリン109である(図3および5)。この架橋は、本明細書中でL1−SS−L2と称される。CD4が結合した新しいgp120構造におけるこれらの残基間のCβ原子間距離は、3.7Åであり、これは、ジスルフィド架橋にとって理想的な距離である。これらの2つの残基を、Stratagen(登録商標)Quick Changeプロトコルを使用する部位特異的変異誘発によってシステインに変異させ、DNA配列決定によって確かめた(表1)。これらの残基をシステインに変異させることに関連して、gp120の発現またはプロセシングに悪影響がないことを、小規模なトランスフェクションおよびウエスタンブロッティングによって確かめた(図6)。
上に記載された類似の基準に基づいて、本発明者らは、ジスルフィド結合によって架橋され得る層2および層3に存在するいくつかの残基を同定した。これらの残基およびCβ原子間の距離を以下に列挙する;V95−W465:4.5Å、V95−R462:5.3Å、V95−L469:5.6ÅおよびH99−R462:5.8Å。残基V95およびW465は、システイン変異および層2と層3とのジスルフィド架橋に対する主要な標的である(図4および5)。これらの選択された残基を変異させ、変異が存在することおよび変異による悪影響が存在しないことを、上に記載されたように確かめた(表1および図6)。
さらなる安定化を達成するために、本発明者らは、表1に示されるようなおよび下記に記載されるような組み合わせ変異誘発を進めた。これらは、
a.V59CとS109Cとの間およびV95CとW465Cとの間のジスルフィド結合を用いる、層1−2および層2−3の二重架橋の組み合わせ
b.V59CとS109Cとの間のジスルフィド結合を使用する、以前に公開されたジスルフィド架橋されたgp120構造[6](W90CとE268C;I103CとQ413C)との、層1−2架橋の組み合わせ
c.V95CとW465C;W90CとE268C;またはI103CとQ413Cにおけるジスルフィド結合と組み合わせた、以前に公開されたジスルフィド架橋されたgp120構造(W90CとE268C;I103CとQ413C)[6]との、層2−3架橋の組み合わせ
を含む。
組換えHIV−1エンベロープ(Env)糖タンパク質であるgp120およびgp140は、サブタイプB CCR5向性株HIV−1 SF162に由来し、HEK293T細胞のトランスフェクションによって生成された。ループ1および2がジスルフィド安定化されたgp120(gp120 L1−SS−L2)およびgp140(gp140 L1−SS−L2)もまた、SF162に由来し、HEK293T細胞において生成された。4つすべての糖タンパク質を、Srivastavaら[52]が記載しているように、Galanthus Nivalis−Agarose(GNA)アフィニティーカラム、陽イオン交換DEAEカラムおよび最後のセラミックヒドロキシアパタイト(CHAP)カラムを含む3工程精製プロセスを用いて精製した。次いで、精製された糖タンパク質を、SDS−PAGEによって(純度レベルについて)解析し、免疫ブロット(抗SF162 gp140ポリクローナルウサギ血清に対する)によって特異的反応性について解析した。精製された糖タンパク質は、>98%の純度で均質だった(gp120については>95%モノマー;gp140については>80%三量体)。糖タンパク質中のエンドトキシンレベルを、Endosafe(登録商標)カートリッジおよびEndosafe(登録商標)−PTSTM分光光度計(Charles River Laboratories International,Inc.,Wilmington,MA)を用いて測定したところ、≦0.05EU/免疫化用量であることが見出された。
各変異体からの精製されたタンパク質の構造の変化および立体配座の固定を、特異的リガンドへの結合の動態を計測するSPRによってアッセイした。野生型gp120およびジスルフィド安定化されたgp120であるgp120 L1−SS−L2の結合親和性を測定するために、本発明者らは、SPRベースのBIAcore3000を使用した。200RUのsCD4またはmAbであるb12もしくは17bを、アミンカップリングによってCM5センサーチップ上に直接固定化した。次いで、様々な濃度の、野生型または(of)ジスルフィド安定化型のいずれかのgp120を80μl/分で注入した。ランニング緩衝液としてHBS−EP緩衝液を使用し、25℃において結合解析を行った。次いで、実験曲線を、BIAevaluationソフトウェア3.2(BIAcore Inc)を使用して1:1Langmuir結合モデルに当てはめた。1:1の当てはめの後に得られる会合速度Kon、解離速度Koffおよび解離定数Kdを表2に示す。
gp120の構造上の立体配座に対する、ジスルフィド結合によって層間可動性を固定する影響を特徴付けるために、野生型(変化なし)および層1−2ジスルフィド連結変異体(V59CとS109C;L1−SS−L2)由来のタンパク質を精製し、表面プラズモン共鳴(Surface Plamson Resonance)(SPR)アッセイによって特徴付けた。このアッセイでは、異なる立体配座のgp120に結合するリガンドをCM5チップ上に捕捉し(方法を参照のこと)、野生型または変異体の可溶性gp120への結合を比較した。CD4が結合したgp120の立体配座に選択的に結合するCD4誘導型抗体である17b抗体と変異体gp120との結合は、野生型への結合と比べて有意な量だけ増大した。ジスルフィド結合によって層1と層2との相互作用が安定化されると、17bへの結合の親和性が5倍超増大した。興味深いことに、この親和性の増大のほとんどすべては、会合速度の増大に由来し(表2を参照のこと)、これは、CD4が結合した立体配座でのgp120の固定を裏付ける。しかしながら、層1と層2とのジスルフィド結合のみによって安定化されたgp120では、なおも相当量の可撓性が残っており、これは、可溶性CD4への結合において特に明らかである。
1.CD4への結合:gp120は、24.5nM(2.48e−8M)のKd(解離定数)で可溶性CD4(sCD4)に結合したが、安定化されたgp120(gp120 L1−SS−L2)は、4分の1の親和性(Kd=93.1nM/9.31e−8M)で結合した。この4倍の差異は、CD4が結合した立体配座でそのタンパク質を「ロック」したことが原因の、sCD4へのジスルフィド安定化されたgp120のより低い会合速度(Kon)にもっぱら起因した[行2および3を比較のこと]
2.b12への結合:本発明者らは、同様の傾向を観察したが、gp120とgp120 L1−SS−L2との間に≦7倍のKdのより大きな差異があった[行4と5とを比較のこと]
3.17b(+/−sCD4)への結合:本発明者らは、gp120がsCD4の非存在下ではmAb 17bに効率的に結合しないことを承知している。しかしながら、sCD4の存在下において[行6]、gp120は、0.26nM(2.62e−10M)というナノモル濃度未満の親和性で結合した。重要かつ決定的なことに、ジスルフィド安定化されたgp120(gp120 L1−SS−L2)は、0.44nM(4.42e−10M)という、CD4結合状態のgp120(gp120+CD4)と類似の親和性でsCD4の非存在下においてmAb 17bに結合した[行7]。安定化されたgp120にsCD4が加えられると、親和性は、20倍改善し、96pM[9.62e−11M,行7]という解離定数を示す。
さらに安定化された各ポリペプチドが生成されたら、その各ポリペプチドは、タンパク質発現のレベル、適切な折り畳みおよびリガンド結合について評価され得る。これらは、以前に安定化された構造および保存された結合部位の露出を増強するために開発された他の改変されたgp120と比較され得る。gp120上の保存された結合部位の安定した露出を達成したそれらのタンパク質は、候補免疫原として、小規模の動物治験においてさらに解析される。
Carbopol(登録商標)971P NF(この研究ではCarbopol 971Pと称される)を粉末としてLubrizolから購入し、次いで、滅菌条件下において水に再懸濁することにより、0.5%の均質な(homogenous)低粘度の懸濁物を生成した。その懸濁物を、さらに使用するまで4℃で保存した。gp140タンパク質と0.5%(w/v)Carbopol971Pとの1:1(v/v)混合物(pH≧3.0)をすべてのインビトロ評価のために生成した。動物に投与するために、まず、gp140と0.5%(w/v)Carbopol971Pとの1:1(v/v)混合物を生成し、gp140タンパク質−Carbopol971P複合体を30分間インキュベートした後、等体積のMF59を加えることにより、0.125%(w/v)で投与されるCarbopol971P懸濁物の最終濃度を維持した。インビボ研究用のすべてのCarbopol971P懸濁物に対して、エンドトキシンレベルを、Endosafe(登録商標)カートリッジおよびEndosafe(登録商標)−PTSTM分光光度計(Charles River Laboratories International,Inc.,Wilmington,MA)を用いて計測した。
免疫化研究を、USDAから許諾を得た(No.93−R−0260)およびNIHからのPublic Health Service(PHS)Assuranceを有する(No.A3404−01)研究施設であるJosman LLC(Napa,CA)において行った。3つの群のニュージーランドシロウサギ(1群あたり5匹の若年成体の雌)をこの研究では使用した。MF59単独、Carbopol971P単独またはCarbopol971P+MF59のいずれかでアジュバント化した(adjuvanted)SF162 gp140タンパク質をウサギに免疫化した。4回の免疫化を、0、4、12および24週目に殿筋(1つの免疫化あたり2つの部位)の筋肉内に行った。各免疫化におけるタンパク質投与量は、50μgだった。最初の免疫化の前(事前採血)および各免疫化の後の様々な時点(2wp2、2wp3、2wp4、4wp4および15wp4)において回収された血液から血清サンプルを調製し、結合および中和について解析した。
本発明者らは、CarbopolおよびMF59をアジュバントとして用いる、タンパク質のみ(25μg)の免疫化のウサギ研究を行った。野生型gp120およびgp140を、それぞれgp120 L1−SS−L2(配列番号4)およびgp140 L1−SS−L2(V59CおよびS109C)と比較した。下記の表3に示されるような免疫化を行った。
本発明者らは、TZM−blアッセイにおいて偽ウイルス(交差サブタイプ)の中和に対する2wp3血清も解析した。以前に記載されているように[46、47]、TZM−bl細胞[Dr.John C.Kappes,Dr.Xiaoyun Wu and Tranzyme,Inc.(Durham,NC)]において、偽ウイルスを使用する十分に標準化されたアッセイおよびルシフェラーゼレポーター遺伝子アッセイを使用して、ウイルス中和力価を計測した。簡潔には、合計200 TCID50偽ウイルス/ウェルを、希釈された血清サンプルに加え、37℃で1時間インキュベートした。インキュベーションの後、DEAE−デキストラン含有培地中の10,000細胞/ウェルを加え、37℃で48時間インキュベートした。DEAE−デキストランの最終濃度は、10μg/mlだった。48時間のインキュベーションの後、供給業者(Promega)が説明するようにBright Glo基質溶液を使用するルミネセンスの測定のために、100μlの細胞を96ウェル黒色固体プレート(Costar)に移した。中和力価は、バックグラウンドRLUを減算した後、ウイルスコントロールウェルと比べて相対ルミネセンス単位(RLU)を50%減少させた希釈度である。以前に報告されたように[Montefiori,D.C.,Measuring HIV neutralization in a luciferase reporter gene assay..HIV Protocols:Second Edition ed.G.V.K.Vinayaka R.Prasad,eds.Vol.485.2009:Humana Press.395−405]、FuGENE−6 HD(Roche Applied Sciences,Indianapolis,IN)を使用して、env欠損HIV−1骨格ベクター(pSG3Δenv)と組み合わせて、HIV−1分離株のパネル由来の分子的にクローニングされた完全長gp160env遺伝子を含む発現プラスミドで293T細胞をコトランスフェクションすることによって、HIV−1 Env偽ウイルスを調製した。48時間後、偽ウイルスを含む細胞培養上清を0.45μmフィルターで濾過し、使用するまで−80℃で保管した。
本発明者らは、2wp3血清を使用して、上記4つの免疫原によって免疫化されたときに産生された抗体のエピトープ特異性を評価した。本発明者らが検討した4つのエピトープは、V3ループ、V1V2ループ、CD4結合部位(CD4BS)およびCD4誘導性/誘導型(CD4i;CD4結合)部位だった。MF59でアジュバント化したgp140タンパク質、Carbopol971Pでアジュバント化したgp140タンパク質またはCarbopol971P+MF59でアジュバント化したgp140タンパク質を免疫化された動物由来の血清中のエンベロープ特異的な総抗体価を、先に記載されたように[52]、SF162 gp140タンパク質を使用する標準的なELISAアッセイによって定量した。抗体アビディティー指数の測定を、他で[52]説明されているようなチオシアン酸アンモニウム(NH4SCN)置換ELISAを使用して行った。
Claims (23)
- CD4結合エピトープとCD4結合部位エピトープの両方を同時に露出する立体配座で安定化されたHIV gp120ポリペプチドまたは可溶性gp140ポリペプチドを含む単離されたポリペプチド。
- 前記ポリペプチドが、内側ドメインの層の間の層間接触によって安定化されている、請求項1に記載の単離されたポリペプチド。
- 前記ポリペプチドが、層1と層2との間の層間接触によって安定化されている、請求項1または請求項2に記載の単離されたポリペプチド。
- 前記ポリペプチドが、層2と層3との間の層間接触によって安定化されている、前述のいずれかの請求項に記載の単離されたポリペプチド。
- 前記層間接触が、ジスルフィド結合によってもたらされる、前述のいずれかの請求項に記載の単離されたポリペプチド。
- 前記ジスルフィド結合が、配列番号2における1つ以上のアミノ酸対:V59CとS109C、V95CとW465C、V95CとR462C、V95CとL469Cおよび/またはH99CとR462Cと等価な位置のシステイン残基間に形成される、請求項5に記載の単離されたポリペプチド。
- 前記安定化されたHIV gp120ポリペプチドまたは可溶性gp140ポリペプチドが、配列番号2に示されるような内側ドメイン配列と少なくとも90%の同一性を有するアミノ酸配列を含み、そして配列番号2のV59とS109、V95とW465、V95とR462、V95とL469および/またはH99とR462と等価な位置に1つ以上の天然に存在しないシステイン残基対を含む、前述のいずれかの請求項に記載の単離されたポリペプチド。
- 前記安定化されたHIV gp120ポリペプチドまたは可溶性gp140ポリペプチドが、配列番号2に示されるような配列と少なくとも90%の同一性を有するアミノ酸配列を含み、そして配列番号2のV59とS109、V95とW465、V95とR462、V95とL469および/またはH99とR462と等価な位置に1つ以上の天然に存在しないシステイン残基対を含む、前述のいずれかの請求項に記載の単離されたポリペプチド。
- 前記安定化されたHIV gp120ポリペプチドまたは可溶性gp140ポリペプチドが、配列番号2におけるW90とE268、I103とQ413に対応する位置に1つ以上の天然にコードされないシステイン対を含む、前述のいずれかの請求項に記載の単離されたポリペプチド。
- 前記安定化されたHIV gp120ポリペプチドが、配列番号4、6、8、10、12、14、16、18、20または22に示されるようなアミノ酸配列を含む、請求項7または請求項8に記載の単離されたポリペプチド。
- 前述のいずれかの請求項に記載の単離されたポリペプチドの免疫原性フラグメントであって、ここで、該免疫原性フラグメントは、HIV gp120のCD4結合エピトープおよびCD4結合部位エピトープの両方を含み、かつそれらを露出する、免疫原性フラグメント。
- 請求項1〜10のいずれか1項に記載の単離されたポリペプチドまたは請求項11に記載の免疫原性フラグメントおよびさらなるポリペプチドを含む融合タンパク質。
- 前記さらなるポリペプチドが、インフルエンザ由来のHAポリペプチドである、請求項12に記載の融合タンパク質。
- 3つのサブユニットを含む三量体ポリペプチドであって、ここで、各サブユニットは、独立して、請求項1〜10のいずれか1項に記載の単離されたポリペプチド、請求項11に記載の免疫原性フラグメントおよび請求項12または請求項13に記載の融合タンパク質からなる群より選択される、三量体ポリペプチド。
