1.技術分野
本発明は概ね、ヘプシジン活性を示すペプチドに関する。

A1、A1〜A2、A10、またはそれらの組み合わせが場合により存在せず、但し、そのペプチドが表1に記載のペプチドの1つではない。いくつかの実施形態において、改変ミニヘプシジンは、ジスルフィド結合により環状構造を形成する。いくつかの実施形態において、N末端アミノ酸は遊離アミンである。いくつかの実施形態において、N末端アミノ酸は、例えば、アセチル基でブロックされたアミンである。いくつかの実施形態において、A6および/またはA8はリジン誘導体、例えば、N−ε−ジニトロフェニル−リジン、N−ε−メチル−リジン、N,N−ε−ジメチル−リジン、およびN,N,N−ε−トリメチル−リジンである。
Claims (18)

A9は、Phe、D−Phe、Leu、D−Leu、Ile、D−Ile、Tyr、D−Tyr、Trp、D−Trp、Phe−Ra、D−Phe−Ra、Dpa−Ra、D−Dpa−Ra、Trp−Ra、bhPhe−Ra、またはその代用物として通常使用される非天然アミノ酸であって、結合するRaがあってもよく、またはなくてもよい、非天然アミノ酸、例えば、PheF5、N−MePhe、ベンジルアミド、2−アミノインダン、bhPhe、Dpa、Bip、1Nal、2Nal、bhDpa、およびシクロヘキシルアラニン、好ましくはbhPheおよびbhPhe−Raであり、Raがパルミトイル−PEG−であり、PEGがPEG11またはミニPEG3、パルミトイル−PEG−PEGであり、PEGがPEG11またはミニPEG3、ブタノイル(C4)−PEG11−、オクタノイル(C8、カプリル酸)−PEG11−、パルミトイル(C16)−PEG11−、またはテトラコサノイル(C24、リグノセリン酸)−PEG11−であり、
A10は、Cys、D−Cys、Ser、D−Ser、Ala、D−Ala、または非天然アミノ酸、例えば、Ida、Ida(NHPal)Ahx、およびIda(NBzl2)Ahxであり、
カルボキシ末端アミノ酸がアミドまたはカルボキシ形態であり、
少なくとも1個のスルフィドリルアミノ酸が前記配列内のアミノ酸の1つとして存在し、
A1、A1〜A2、A10、またはそれらの組み合わせが場合により存在せず、但し、前記ペプチドが表1に記載のペプチドの1つではない、
を有する単離されたペプチド。 - 前記ペプチドが以下、
a)A1=N−MeAsp、Ida、またはIda(NHPal)、
b)A5=bhPro、
c)A6=D−Val、D−Leu、Lys、D−Lys、Arg、D−Arg、Ach、bhArg、またはN−MeArg、
d)A7=Cys(S−S−Pal)、Cys(S−S−システアミン−Pal)、Cys(S−S−Cys−NHPal)、またはCys(S−S−Cys)、および/または
e)A8=D−Val、D−Leu、Lys、D−Lys、Arg、D−Arg、Ach、bhArg、またはN−MeArg
の少なくとも1つを含有する、請求項1に記載のペプチド。 - i)A1がIdaであり、A9がPheであるときに、A10がAhx−Ida(NHPal)でなく、
ii)A1がIdaであり、A9がbhPhe−Rbでないときに、RbがS−(パルミトイル)チオグリコール酸−PEG−であり、
iii)A4がD−Pheであり、A7がD−Cys(S−S−tBut)ではなく、A9がD−Trp−Rcではないときに、Rcがブタノイル−PEG11−、オクタノイル−PEG11−、パルミトイル−PEG11−、またはテトラコサノイル−PEG11−であり;
iv)A1がIdaであり、A9がbhPhe−Rdであるときに、Rdがパルミトイル−PEG−ミニPEG3−であり、A6およびA8がともにD−Argでなく、またはともにbhArgではない、
請求項1または請求項2に記載のペプチド。 - A1がD−Asp、N−MeAsp、Ida、またはIda(NHPal)であり、
A2がThrまたはD−Thrであり、
A3がHisまたはD−Hisであり、
A4がDpaまたはD−Dpaであり、
A5がPro、D−Pro、bhPro、またはOicであり、
A6がIle、D−Ile、Arg、D−Val、D−Leu、Ach、またはN−MeArgであり、
A7がCys、D−Cys、Cys(S−S−Pal)、Cys(S−S−システアミン−Pal)、Cys(S−S−Cys−NHPal)、またはCys(S−S−Cys)であり、
A8がIle、D−Ile、Arg、D−Val、D−Leu、Ach、またはN−MeArgであり、
A9がPhe、D−Phe、Dpa、D−Dpa、Trp、D−Trp、bhPhe、Phe−Ra、D−Phe−Ra、Dpa−Ra、D−Dpa−Ra、Trp−Ra、bhPhe−Raであり、Raがパルミトイル−PEG−であり、PEGがPEG11またはミニPEG3、パルミトイル−PEG−PEGであり、PEGがPEG11またはミニPEG3、ブタノイル(C4)−PEG11−、オクタノイル(C8、カプリル酸)−PEG11−、パルミトイル(C16)−PEG11−、またはテトラコサノイル(C24、リグノセリン酸)−PEG11−であり、
A10が、存在する場合、Ida(NHPal)AhxまたはIda(NBzl2)Ahxである、
請求項1〜請求項3のいずれか1項に記載のペプチド。 - 前記ペプチドが、PR42’、PR47、PR48、PR49、PR50、PR51、PR52、PR53、PR56、PR57、PR58、PR59、PR60、PR61、PR63、PR65、PR66、PR67、PR68、PR69、PR70、PR71、PR72、PR73、PR74、およびPR82、好ましくはPR47、PR48、PR49、PR50、PR51、PR52、PR53、PR56、PR57、PR58、PR59、PR60、PR61、PR63、PR65、PR66、PR67、PR68、PR69、PR70、PR71、PR72、PR73、PR74、およびPR82からなる群から選択される、請求項1〜請求項4のいずれか1項に記載のペプチド。
- 前記ペプチドがヘプシジン活性を示す、請求項1〜5のいずれか1項に記載のペプチド。
- 前記ペプチドがフェロポルチンと結合する、請求項1〜5のいずれか1項に記載のペプチド。
- 請求項1〜7のいずれか1項に記載の少なくとも1つのペプチドを含む組成物。
- 前記フェロポルチンと請求項1〜7のいずれか1項に記載の少なくとも1つのペプチドまたは請求項8に記載の組成物を接触させることを含む、フェロポルチンと結合し、フェロポルチン内在化および分解を誘発する方法。
- 請求項1〜7のいずれか1項に記載の少なくとも1つのペプチドまたは請求項8に記載の組成物を対象に投与することを含む、前記対象の鉄代謝の疾患を治療する方法。
- 前記鉄代謝の疾患が鉄過負荷疾患である、請求項10に記載の方法。
- 試薬、デバイス、説明資料、またはそれらの組み合わせをともに梱包された、請求項1〜7のいずれか1項に記載の少なくとも1つのペプチドまたは請求項8に記載の組成物を含むキット。
- フェロポルチンまたは抗体と結合する請求項1〜7のいずれか1項に記載の少なくとも1つのペプチドを含む複合体。
- 治療を必要とする対象の鉄代謝の疾患を治療し、かつ/または鉄量を低下させるための医薬品の製造のための請求項1〜7のいずれか1項に記載の1つ以上のペプチドまたは請求項8に記載の組成物の使用。
- 治療を必要とする対象の鉄代謝の疾患を治療し、かつ/または鉄量を低下させるときに使用される、請求項1〜7のいずれか1項に記載の1つ以上のペプチドまたは請求項8に記載の組成物。
- 治療を必要とする対象に鉄代謝の疾患を治療し、かつ/または鉄量を低下させるための医薬品の製造のためであり、前記医薬品が単回の1日用量として、または分割の1日用量として有効1日用量にて投与されるよう調製される、請求項1〜7のいずれか1項に記載の1つ以上のペプチドまたは請求項8に記載の組成物。
- 前記有効1日用量が、約10〜500μg/kg/日であり、前記医薬品が皮下注射用に配合される、請求項16に記載の使用。
- 前記有効1日用量が約10〜1000μg/kg/日であり、前記医薬品が、経口、経肺または粘膜投与用に配合される、請求項16に記載の使用。
