4−SNPは成分に対して中程度の影響を有する。
5(最も高い影響因子)−SNPは成分にかなりの効果がある。
・第1レベルの相互作用:成分と直接的に相互作用する第1のタンパク質(化学的相互作用または成分修飾の原因である受容体または酵素)この標的が機能していない場合、成分の代謝に重大な影響。
・第2レベルの相互作用:成分の代謝に関与するカギとなる分子。この標的が機能していない場合、成分の代謝にある程度の影響。
・第3レベルの相互作用:成分の代謝に関与する分子であるが、この標的が機能していない場合、成分の代謝にわずかな影響。
この方法の重要な態様の1つは、正しい遺伝子型を標的にするために特定のプライマーを設計することである。DNAを増幅するために、ポリメラーゼ連鎖反応(PCR)、SNAP、またはLAPMアッセイなどの複数の標準的な方法を使用することができる。これらの技術はすべて正確なプライマーの選択に基づく。正しいプライマーを選択するために用いられるパラメータが知られているが、最終的な結末と設計されるプライマーの有効性は新しい。少数のSNPのために設計されたプライマーが以下に記載されている。プライマーは、プライマー長、プライマー中の末端ヌクレオチド、適切なGC量、およびTmを含む、多くの基準にしたがって選択された。

Claims (22)
- 個人に対する化粧品および/または栄養化粧品および/またはスキンケア製品のセットの適合性を評価する方法であって、
前記方法は、
あらかじめ決められたセットの単一ヌクレオチドの位置で一塩基多型の存在または不在を確認するために、個人の遺伝物質のサンプルを試験する工程、
単一ヌクレオチドの位置での一塩基多型の存在または不在にしたがって、それぞれの位置での1以上の重み付けを特定する工程、および、
前記の製品のそれぞれに関して製品スコアを測定するために用いられる単一ヌクレオチド位置の重み付けを用いる工程であって、前記スコアが個体に対する製品の適合性を示している、工程、
を含む、方法。 - 試験する工程は、一塩基多型が所定の単一ヌクレオチド位置に存在する場合に、一塩基多型がヘテロ接合またはホモ接合の変異型で存在しているかどうかを測定する工程を含み、それぞれの位置について重み付けを特定する前記工程は、ヘテロ接合の変異型に様々な重み付けを適用する工程を含んでいる、請求項1に記載の方法。
- 所定の単一ヌクレオチド位置について、一塩基多型が存在しない場合(野生型)、その位置には比較的高い重み付けが与えられ、一塩基多型がホモ接合体の変異型で存在している場合、その位置には比較的低い重み付けが与えられ、および、一塩基多型がヘテロ接合の変異型で存在している場合、中程度の重み付けが与えられる、請求項2に記載の方法。
- 位置に適用される重み付けは、内部に単一ヌクレオチドの位置が見られる発現した遺伝子と、有効成分との相互作用の値に依存する、請求項1乃至3のいずれか1つに記載の方法。
- 機能の欠損を示す一塩基多型が存在する場合、1つの位置には比較的低い重み付けが与えられ、機能の獲得を示す一塩基多型が存在する場合、その位置には比較的高体重が与えられる、請求項4に記載の方法。
- 単一ヌクレオチドの位置の重み付けを使用する工程は、あらかじめ定められた有効な製品の成分のセットのそれぞれを前記単一ヌクレオチド位置の1つ以上に関連付ける工程、
成分スコアを決定するために、それぞれの有効な製品の成分に関連する単一ヌクレオチドの位置について位置の重み付けを組み合わせる工程、および、
所定の製品に関して、製品内の有効成分を特定し、関連する成分スコアを用いて前記製品のスコアを決定する工程を含む、請求項1乃至5のいずれか1つに記載の方法。 - 製品のスコアを決定する前記工程は、製品スコアが、あらかじめ定められた閾値スコアを超える製品スコアを有している製品内の有効成分の数を特定する工程と、その製品内の有効成分の総数の一部または割合として、その数を表す工程を含む、請求項6に記載の方法。
- それぞれの位置について1つ以上の重み付けを特定する工程は、異なる成分について異なる重み付けを決定する工程を含む、請求項1乃至7のいずれか1つに記載の方法。
- 個体について定められたライフスタイルおよび/または外部の要因に従って、重み付けおよび/またはスコアを修正する工程を含む、請求項1乃至8のいずれか1つに記載の方法。
- 成分の投与および/または製品の組成に従って、重み付けおよび/またはスコアを修正する工程を含む、請求項1乃至9のいずれか1つに記載の方法。
- 個体に合わせた化粧品、栄養化粧品、および/または、スキンケア製品を製造する方法であって、
前記方法は、
あらかじめ定められたセットの単一ヌクレオチドの位置で一塩基多型の存在または不在を特定するために、個体について遺伝物質のサンプルを試験する工程、
前記位置での一塩基多型の存在または不在にしたがって、それぞれの位置の重み付けを特定する工程、
あらかじめ定められたセットの有効な製品の成分の各々を、前記単一ヌクレオチド位置の1つ以上に関連付ける工程、
成分スコアを決定するために、それぞれの有効な製品の成分に関連する単一ヌクレオチドの位置について位置の重み付けを組み合わせる工程、
成分スコアを用いて有効な製品の成分のサブセットを選択する工程、および、
前記個体用の製品を製造するために、成分のサブセットを混合する工程、
を含む、方法。 - 化粧品、栄養化粧品、および/または、スキンケア製品に用いられる1つの成分または成分の組み合わせの有効性に影響を及ぼす、1つ以上の一塩基多型SNPを特定する方法であって、ユーザーに対する製品の適合性を試験するために使用することができる、方法であって、
前記方法は、
1つの成分または成分の組み合わせによって影響を受ける1つ以上の生物学的経路に関連する1つの遺伝子または遺伝子の組み合わせを特定する工程、
それぞれの遺伝子に関して、前記遺伝子内に存在し得るSNPを特定する工程、および、
成分の影響を受ける1つ以上の生物学的経路の能力に大きな影響を及ぼすSNPを特定するために、前記工程で特定されたSNPを評価する工程
をさらに含む、方法。 - 評価する工程は、特定されたSNPの多くの特徴および/または効果を考慮に入れ、特徴の1つは、ユーザーの母集団またはユーザーの母集団のサブグループ内におけるSNPの広まりであり、あらかじめ定められた閾値の普及率よりも大きな普及率を有しているSNPには、前記閾値よりも普及率が低いSNPに比べて高い評価が割り当てられる傾向がある、請求項12に記載の方法。
- 評価する工程の際に考慮に入れられるさらなる特徴は、マイナー対立遺伝子頻度、機能母集団型、ヘテロ接合頻度、および、生物学的経路の1つ以上を含む、請求項13に記載の方法。
- 前記方法はさらに、成分の影響を受ける1つ以上の生物学的経路の能力に大きな影響を及ぼす特定されたSNPの情報を、SNPが関連付けられる成分と一緒に、マッピングする工程と、
ユーザーに対する製品の適合性を試験する際にマッピングされた情報を参照することができるように、マッピングされた情報をデータベースに保存する工程を含む、請求項12乃至14のいずれか1つに記載の方法。 - 前記方法はさらに、特定されたSNPが関連付けられる成分を含む、化粧品、栄養化粧品、および/または、スキンケア製品に、特定されたSNPをマッピングする工程と、
ユーザーのために製品を選択する際に、マッピングされた情報を参照することができるように、マッピングされた情報をデータベースに保存する工程を含む、請求項12乃至15のいずれか1つに記載の方法。 - 1つの成分または成分の組み合わせは、1つの成分または成分の組み合わせの影響を受ける生物学的経路またはそれぞれの生物学的経路に関連する遺伝子またはそれぞれの遺伝子の内部に、1以上の特定されたSNPが存在するため、有効性を減少させたことが分かる、請求項12乃至16のいずれか1つに記載の方法。
- 1以上の特定されたSNPが、成分によって修正される1以上の遺伝子中に欠陥を形成するため、1つの成分または成分の組み合わせは、1以上の特定されたSNPの存在によってその有効性を増加させたことがわかる、請求項12乃至17のいずれか1つに記載の方法。
- 消費者のために化粧品、栄養化粧品、または、スキンケア製品を選択する方法であって、
前記方法は、
請求項12乃至18のいずれか1つの方法を駆使して特定されたSNPを検知するために、消費者から得られた生体サンプルを試験する工程と、
検知されたSNPに基づいて、広範な利用可能な製品から化粧品、栄養化粧品、または、スキンケア製品を選択する工程、
を含む、方法。 - 製品の選択は、一緒に作用する2つ以上の成分の任意の相乗効果も考慮に入れられる、請求項19に記載の方法。
- 消費者から得られた生体サンプルを試験する工程は、検知されるSNPを増幅するために選択されたプライマーを使用する工程を含む、請求項19または20のいずれか1つに記載の方法。
- プライマーは多くの基準によって選択され、基準は、プライマー長、プライマー中の末端ヌクレオチド、適切なGC(グアニン−シトシン)量、およびTmを含む、請求項21に記載の方法。
