変異体LIGHT送達システムまたは等価の送達システムの送達を可能にするために、変異体LIGHTを腫瘍標的化薬剤、例えば腫瘍特異的抗体にカップリングするまたは結合体化させることができる。例えば、LIGHTまたは変異体LIGHTに結合体化させた腫瘍特異的抗体を使用して、腫瘍部位に融合タンパク質を選択的に送達することができる。加えて、腫瘍特異的抗体を、ウイルス送達システムまたはリポソームビヒクル送達システムとカップリングするように設計することができる。変異体LIGHTを発現するおよび腫瘍標的化薬剤を内部に持つ送達ビヒクルは、先ず、特定の腫瘍細胞に標的化し、その後、その腫瘍細胞を、その細胞の表面で変異体LIGHTを発現するように形質転換させることになる。腫瘍細胞の表面でのこの標的化された変異体LIGHT発現は、その腫瘍の周囲の間質細胞上のケモカインを誘導して、T細胞を誘引し、T細胞のプライミングを開始させることになる。かかる処置は、固形腫瘍を含めて、すべての腫瘍に適している。4T1、MC38、B16および肥満細胞腫をAd−LIGHTで処置し、それらは、原発性および/または続発性腫瘍の低減を示した。したがって、抗体−LIGHTを使用して、様々な腫瘍に、特に、遠隔部位への原発性腫瘍細胞移動の結果として形成する転移に、標的化させることができる。例えば、全身注射により、抗her2/neu抗体とLIGHTは、her2/neuを発現する転移性腫瘍にLIGHTを運び、その後、局所免疫応答を発生させて腫瘍を一掃することができる。したがって、前記融合タンパク質を任意の全身および局所経路によって送達することができ、前記融合タンパク質は、抗体または別の薬剤の腫瘍抗原に対する特異性のため、より多く腫瘍に局在することになる。
LIGHT、LTβRおよびHVEMの受容体に結合する抗体−LIGHTの能力を、LTβR−IgおよびHVEM−Igをそれぞれ用いてフローサイトメトリーによって判定する。抗体−LIGHTの機能活性を、先ず、最適未満の用量のプレート結合抗CD3の存在下でT細胞を共刺激するその能力についてin vitroで試験する。抗体−LIGHTの機能性は、抗CD28のものに匹敵するようである。
抗体−LIGHT融合タンパク質または結合体の重要な有用性は、かかる標的化薬剤が、免疫系を有効に刺激しない少数の転移性腫瘍細胞または残留癌細胞の一掃に非常に効くことができることである。抗体−LIGHTと、アデノウイルス発現LIGHT、すなわちAd−LIGHTとを含む併用療法を試験する。
乳癌および結腸癌患者の五分の一は、Her2/neuを発現する。一般に、Her2に対する抗体は、これらの腫瘍の成長を減速させるが、それらを根絶しない。LIGHTとカップリングさせた抗Her2/neu抗体は、LIGHTを転移性腫瘍部位に標的化させる。前記抗Her2/neu抗体は、腫瘍成長を減速させ、アポトーシスを誘導し、それにより、そのカップリングされたLIGHTは、T細胞のLIGHT媒介動員および活性化が腫瘍内部で起こるように誘導することが可能になる。加えて、LIGHTはまた、FcR+ 細胞を動員して抗neu抗体の治療効果を強化する。ある実験モデルでは、LIGHTと連結させた抗体の10μgほども低い用量によりマウスにおいて腫瘍の成長が減速された。他のより低いまたはより高い用量が企図される。抗Her2/neu抗体−LIGHTは、乳癌転移のための新規処置剤である。図2は、Adv−mmlitがneu+ N202腫瘍成長を阻害することを示す。
腫瘍特異的抗体−LIGHT融合タンパク質または結合体を、抗腫瘍剤、例えばドキソルビシン、パクリタキセル、ドセタキセル、シスプラチン、メトトレキサート、シクロホスファミド、5−フルオロウリジン、ロイコボリン、イリノテカン(CAMPTOSAR(商標)もしくはCPT−11もしくはカンプトテシン−11もしくはカンプト)、カルボプラチン、フルオロウラシル・カルボプラチン、エダトレキサート、ゲムシタビン、もしくはビノレルビン、またはこれらの組み合わせなどと、さらにカップリングさせることができる。これらの薬物を別々に投与することができ、または抗体−LIGHT融合タンパク質または結合体との結合体化またはカップリングによって併用投与することができる。
抗neu抗体とLIGHT.TUBOの相乗作用。TUBOは、BALB−neuTマウスからの自然発生乳腺腫瘍から産生されるクローン化細胞系であり、細胞膜上でHER−2タンパク質を高度に発現する。この腫瘍系統は、in vitroおよびin vivoでの抗neu抗体処置に対して感受性である。しかし、腫瘍が十分に定着すると、抗体またはLIGHTいずれかのみの効果は、減少される。抗neu細胞を中断すると、3〜4週間以内にTUBO細胞は回復することができる。この2つの間に相乗作用があるかどうかを判定するために、TUBO細胞を18日間、定着させ、その後、ad−LIGHTと抗neu抗体の両方で週1回、3週間にわたって処置した。印象的に、この併用で検出できる癌はなく、その一方でいずれかの単独処置では腫瘍が漸進的に成長する(図4〜5)。前記併用処置を投与したものを除き、各群の5匹のマウスすべてが腫瘍を有した。
sc−Fv−LIGHTを発現させるために、scFV−58LIGHT(アミノ酸位置58−240を有するLIGHT断片)およびscFV−85LIGHT(81−84のプロテアーゼ部位を迂回してアミノ酸位置85−240を有するLIGHT断片)を構築した。抗Flag抗体は、非常に特異的かつ感受性であるので、ウエスタンブロット法に従ってFlag tapを前記LIGHT断片に取り付けた。かかるプラスミドを293細胞系にトランスフェクトした。それらの細胞を1週間後に回収し、溶解産物を調製し、抗flag抗体でブロットした。抗Flagウエスタンブロットの可視化により、scFV−85LIGHT発現の発現が、scFV−58LIGHTより高度であることが証明される。
腫瘍標的化抗体−LIGHT免疫サイトカインを産生させるために、次の工程を踏んだ:
a.LIGHTを安定性/親和性増加のために作出した;
b.scFV(neu)−LIGHT融合タンパク質を生産のために産生させた;および
c.そのscFV(neu)−LIGHT融合タンパク質をin vitroおよびin vivoで試験した。
ヒトLIGHTをその作出のプラットホームとして使用した。
ヒトLIGHTは、マウスLIGHTより安定しているようであり(YDおよび事前発現);および
ヒトLIGHTは、ネズミ受容体と(弱く)交差反応する。
a.ネズミおよびヒトLTbRおよびHVEMへの同等の結合、および可能であれば、DcR3への結合減少;および
b.生産を容易にするための改善された発現/安定性。
抗体−LIGHTの融合タンパク質の産生。標準的なプロトコルを用いて、腫瘍抗原または腫瘍細胞に高親和性で結合する抗体へのLIGHTの特異的標的化を可能にするヘテロミニボディと称する組換え抗体構築物を開発した。
核酸配列によってコードされたアミノ酸配列を有する変異体LIGHT分子であって、
i)野生型ヒトLIGHTと比較して増大した親和性でLIGHTのネズミ受容体に結合し、かつ
ii)野生型ヒトLIGHTと少なくとも同じ親和性でLIGHTのヒト受容体に結合する、

原発性腫瘍の成長および癌転移を低減させる方法であって、上記方法は:(a)腫瘍特異的抗体を含む医薬組成物を投与する工程;(b)LIGHTタンパク質をコードする核酸分子を腫瘍部位に導入する工程であって、上記LIGHTは、ヒトLIGHTタンパク質またはその変異体もしくは断片であるか、あるいはプロテアーゼ耐性である、工程;(c)腫瘍細胞の表面で上記LIGHTタンパク質またはその断片を安定して発現させる工程;および(d)上記腫瘍の成長および癌転移を、上記腫瘍に対する腫瘍特異的T細胞の活性化を刺激することによって低減させる工程を含む、方法。
LIGHTの実施形態、例えば、腫瘍に対する標的化薬剤に連結、融合または結合体化された変異体LIGHTタンパク質、ペプチド、またはそれらの断片、での腫瘍への標的化は、腫瘍の成長を低減させ、転移(微小転移を含む)も低減させる。標的化薬剤は、抗体を含む。さらに、腫瘍抗原に対する抗体に連結されたサイトカインは、微小転移に対して有用である。
Claims (39)
- 核酸配列によってコードされたアミノ酸配列を有する変異体LIGHT分子であって、前記配列は、
i)野生型ヒトLIGHTと比較して増大した親和性でLIGHTのネズミ受容体に結合し、かつ
ii)野生型ヒトLIGHTと少なくとも同じ親和性でLIGHTのヒト受容体に結合する能力を、前記分子に付与し、
ここで、前記能力は、LIGHTの細胞外ドメインのコーディング配列内の変異に起因する、変異体LIGHT分子。 - 前記細胞外ドメインが、出現順に、それぞれ、配列番号6−8、8−10、10、4、3、5、5、11−13、12、14−17、20、19−23、45、20、および25と指定されるアミノ酸配列から成る群より選択される、請求項1に記載の変異体LIGHT分子。
- 変異体LIGHTタンパク質、ペプチド、タンパク質またはペプチドの断片、および変異体LIGHTをコードする核酸分子の発現産物から成る群より選択される、請求項1に記載の変異体LIGHT分子。
- ヒトLIGHT配列に由来するアミノ酸配列を含む、請求項1に記載の変異体LIGHT分子。
- 請求項1に記載の変異体LIGHT分子を含む免疫学的組成物。
- 請求項1に記載の変異体LIGHT分子に連結、結合体化または融合された腫瘍特異的薬剤を含む医薬組成物であって、前記組成物は、腫瘍細胞に対して細胞傷害性のTリンパ球を刺激する、医薬組成物。
- 前記薬剤が、前記変異体LIGHT分子に化学的に結合体化されている、請求項8に記載の組成物。
- 前記薬剤が、ヒト化モノクローナル抗体である、請求項8に記載の組成物。
- 前記薬剤が、キメラ抗体である、請求項8に記載の組成物。
- 前記薬剤が、ヘテロミニボディである、請求項8に記載の組成物。
- 前記薬剤が、一本鎖抗体である、請求項8に記載の組成物。
- 前記薬剤が、腫瘍抗原を認識するために十分な抗体断片である、請求項8に記載の組成物。
- 前記変異体LIGHT分子が、細胞傷害性T細胞を刺激するために十分な細胞外ドメインのセクションを含む、請求項8に記載の組成物。
- 前記セクションが、LIGHTの約100〜150個のアミノ酸をヒトLIGHTアミノ酸の順序で含む、請求項15に記載の組成物。
- 前記セクションが、ヒトLIGHTタンパク質の約85〜240位からのアミノ酸配列を含む、請求項15に記載の組成物。
- 変異体LIGHTが、プロテアーゼ認識配列EQLI(残基81〜84)(配列番号1)に変異を含み、それによって前記プロテアーゼ認識配列が不活性化される、請求項8に記載の組成物。
- 変異体LIGHTが、欠失を含む、請求項8に記載の組成物。
- 前記変異体LIGHTが、アミノ酸配列
- m4−14、m4−7およびm4−16と称する変異体LIGHT細胞外ドメイン配列。
- 原発性腫瘍の成長の低減に使用するためのまたは癌転移の処置のための、請求項1に記載の変異体LIGHT分子に連結、結合体化または融合された腫瘍特異的薬剤を含む医薬組成物。
- 出現順に、それぞれ、配列番号6−8、8−10、10、4、3、5、5、11−13、12、14−17、20、19−23、45、20、および25と指定されるアミノ酸配列から成る群より選択されるヒト(hLIGHT)変異体分子。
- 請求項23に記載の分子とHer−2に対する抗体とを含む融合タンパク質。
- 腫瘍細胞を特異的に認識する薬剤との組み合わせでプロテアーゼ耐性LIGHTタンパク質の断片を含む組成物。
- 前記薬剤が、腫瘍抗原に対する抗体である、請求項25に記載の組成物。
- 前記薬剤が、腫瘍表面受容体を結合するリガンドである、請求項25に記載の組成物。
- LIGHT分子と連結、融合または結合体化された腫瘍抗原を認識するアミノ酸領域を含むキメラタンパク質であって、前記LIGHT分子が、腫瘍環境でプロテアーゼ消化に対して耐性であるか、または変異体LIGHT分子である、キメラタンパク質。
- 請求項1に記載の変異体LIGHT分子を患者中の腫瘍に送達するための方法であって、前記方法は:(a)前記分子を必要とする患者に、前記分子をコードする核酸分子を有するアデノウイルスを投与する工程、または;(b)前記分子を必要とする患者に、前記分子を有するナノ粒子を投与する工程;または(c)前記分子を必要とする患者への前記分子の直接移入(direct transfer)を含む、方法。
- 原発性腫瘍の成長または癌転移を低減させる方法であって、前記方法は:(a)LIGHTポリペプチド変異体または断片に連結、結合体化または融合された腫瘍特異的薬剤を含む、請求項8に記載の医薬組成物を投与する工程であって、ここで、前記組成物は、細胞傷害性Tリンパ球の生産を刺激するために十分なものである、工程;および(b)前記原発性腫瘍の成長または癌転移を、前記腫瘍に対する腫瘍特異的T細胞の活性化を刺激することによって低減させる工程を含む、方法。
- 前記薬剤が、表面腫瘍抗原を認識する抗体である、請求項30に記載の方法。
- 前記薬剤が、HER2、HER4、HERB、EGFR、STEAP、c−MET、アルファフェトプロテイン(AFP)、癌胎児抗原(CEA)、CA−125、MUC−1;rasまたはp53の異常産物;およびDcR3から成る群より選択される腫瘍抗原に特異的な抗体である、請求項30に記載の方法。
- 前記薬剤が、前記LIGHT断片に化学的に結合体化されているか、または前記LIGHT断片に組換え融合されている抗体である、請求項30に記載の方法。
- 前記LIGHT断片が、ヒトLIGHTタンパク質のアミノ酸85−240を含むLIGHTの細胞外ドメインを含む、請求項30に記載の方法。
- 前記医薬組成物が、静脈内投与される、請求項30に記載の方法。
- 前記癌転移が、ケモカイン、接着分子、およびナイーブT細胞をプライミングするための共起刺激分子の少なくとも1つの生産を刺激することにより低減される、請求項30に記載の方法。
- 前記癌が、乳癌、肺癌、前立腺癌、結腸癌、腎癌、肝臓癌、白血病および皮膚癌から成る群より選択される、請求項30に記載の方法。
- 化学療法薬または放射線療法を投与する工程をさらに含む、請求項30に記載の方法。
- 腫瘍の成長および癌転移を低減させる方法であって、前記方法は:(a)請求項8に記載の組成物を投与する工程;または(b)LIGHTをコードする核酸分子もしくはその断片を個体の腫瘍部位に導入する工程であって、前記LIGHTは、細胞外ドメインに変異を含むか、プロテアーゼ耐性であり、かつLIGHTは、腫瘍細胞表面に安定して存在する、工程;および(c)前記原発性腫瘍の成長・癌転移を、前記腫瘍に対する腫瘍特異的T細胞の活性化を刺激することにより低減させる工程を含む、方法。
