【解決手段】ジフテリア毒素(DT)の変異である、CRM197のRドメインを含むポリペプチドであって、前記Rドメインの391位、460位、466位、467位、468位、472位、507位及び520位からなる群から選ばれる少なくとも1個のアミノ酸が置換又は欠失され、CRM197のAドメイン、Tドメイン及びRドメインを含むCRM197改変体であるポリペプチド、及び、該ポリペプチドを含む抗腫瘍剤。

Description
項1. CRM197のRドメインを含むポリペプチドであって、前記Rドメインの391位、460位、466位、467位、468位、472位、507位及び520位からなる群から選ばれる少なくとも1個のアミノ酸が置換されたポリペプチド。
項2. 前記Rドメインの391位、460位、466位、467位、468位及び472位からなる群から選ばれる少なくとも1個のアミノ酸が置換された、項1に記載のポリペプチド。
項3. CRM197のRドメインにおいて、H391K、R460H、G466D、D467A、V468T、R472K、D507N、H520Kからなる群から選ばれる少なくとも1種を含む項1又は2に記載のポリペプチド。

材料と方法
試薬
組換えHB-EGF及びTGF-αはR&D Systems (MN, U.S.A.)から入手した。ヘパリン結合ドメインを欠失したC-末端His-タグ化HB-EGFを文献(Takazaki, R., et al., J Biol Chem, 2004. 279(45): p. 47335-43)に記載のように精製した。
DER細胞は外因性ヒトEGFR cDNAを骨髄由来リンパ芽球性32D細胞に導入することにより確立した[Iwamoto, R., K. Handa, and E. Mekada, J Biol Chem, 1999. 274(36): p. 25906-12.]。DER細胞は既報のように維持した。Buffaloラット肝(BRL)細胞及び外因性ヒトHB-EGFを発現するBRL細胞(BRLH)を既報のように確立し、維持した(Mizushima, H., et al., J Cell Sci, 2009. 122(Pt 23): p. 4277-86)。
PCRにより増幅した全Rドメイン(残基380-535)のcDNAをpGEX-3ベクター(GE Healthcare)のBamHI-EcoRI部位に挿入し、E.coli JM109を形質転換した。単一コロニーを50μg/mlアンピシリンを含有する40 mlのLB培地(LB-amp)に接種し、37℃で終夜増殖した。プレ培養培地を500 mlの新鮮なLB-ampに接種し、37℃で細菌の対数増殖期に到達するまで増殖した。これに0.4 mMイソプロピルβ-D-1-チオガラクトピラノシド(IPTG)を加えて37℃で2時間培養し、培養液を遠心分離し、大腸菌を集めた。0.2 mMフェニルメチルスルホニルフルオライド(PMSF)を含有する溶液A(20 mM Tris-HCl, pH8.0, 30 mM NaCl, 10mM EDTA)で洗浄した大腸菌を再び遠心分離により集め、リゾチーム(40 μg/ml)を含有する溶液Aで氷上1時間溶解した。細菌溶解物にTriton-X100(0.53%)及びNaCl(100 mM)を加えて15分間氷上でインキュベートし、さらに0.2 mM PMSFの存在下で20秒間(氷上で2回)超音波処理した。遠心分離により、上清をグルタチオン-セファロース4Bゲル(GE Healthcare)と4℃で終夜インキュベートした。ゲルを溶液Aで洗浄し、結合したタンパク質を溶離バッファー(100 mM Tris-HCl, pH8.0, 6 mMグルタチオン)で溶離した。
RドメインのcDNAをpThioHisベクター(Invitrogen, CA, U.S.A.)のXhoI-EcoRI部位にクローニングし、E.coli JM109を形質転換した。ThioHis-融合タンパク質を製造元の指示書に従いHis-Patch ThioFusionTM発現システム(Invitrogen)を用いて精製した。
RドメインcDNAをpMAL-p2ベクター(New England BioLabs Inc.MA, U.S.A)のBamHI-PstI部位にクローニングし、E.coli JM109を形質転換した。PCRに使用したプライマーは以下のものである(クローニング用の制限酵素部位は下線で示す):
Fw, 5'-GCAGGATCCCCCGCGTATTCTCCGGGG-3',
Rv, 5'-CGCTGCAGTCAGCTTTTGATTTCTAG-3'.
さらにRドメインをMBPから分離するために、PreSicission Protease(GE Healthcare)認識配列(LEVLFQ/GP)をコードするオリゴヌクレオチドをpMAL-p2ベクターのBamHI部位に挿入した。
MBP-Rドメイン変異体をPCRを用いた部位特異的突然変異により構築した。使用したオリゴヌクレオチドを表1に列挙する。MBP-Rドメイン変異体をコードするプラスミッドをE.coli JM109に導入し、形質転換した。MBP-Rドメイン変異体は上記の方法で精製した。
DER細胞(1×104細胞)を、種々の濃度のMBP-Rドメインタンパク質の存在下又は非存在下に40時間10%牛胎児血清(FBS)、組換えHB-EGF(10 ng/ml)、ヘパリン(10 μg/ml), ペニシリンG(100U/ml), ストレプトマイシン(100 μg/ml)を含有するRPMI1640/10% FCS培地で培養した。MBP-Rドメインタンパク質の濃度は0.22, 0.67, 2, 6.1, 18.5, 55.5及び167μMであった。培養は96-ウェルプレート(100 μl培地/ウェル)を用いて行った。DER細胞数を細胞カウント試薬SF(Nacalai Tesque, Japan)を用い製造元のマニュアルに従い測定した。全ての実験は3回行い、データは平均±s.dで示す。
氷冷ウシI型コラーゲン(Nitta Gelatin; 3 mg/ml), 2.2%(w/v)NaHCO3, 0.2 M HEPES及び50 mM NaOHを含む再構成バッファー、10X DMEM-F12(1:1)培地及び血清フリーDMEM(5×106 細胞/ml)に懸濁した細胞を8:1:1:0.1比で混合し、24-ウェルディッシュ(0.5 ml/ウェル)に既報のように注いだ(Mizushima, H., et al., J Cell Sci, 2009. 122(Pt 23): p. 4277-86)。37℃で30分のインキュベート後、コラーゲンゲルをCRM197, MBP-R(WT)、MBP-R(変異体)のいずれかを含有する1 ml DMEM-FBSで覆い、1週間培養した。細胞数をカウントするために、コラーゲンゲルを溶解するまでHBS(+)(10 mM HEPES, pH7.2, 140 mM NaCl, 4 mM KCl, 1.8 mM CaCl2及び1 mM MgCl2)中37℃で0.5 mlの0.5%(w/v)細菌コラゲナーゼ(Invitrogen, CA, U.S.A.)とインキュベートし、細胞を遠心分離により集めた。細胞を固定し、クリスタルバイオレット溶液(0.1%クリスタルバイオレット, 0.1 Mクエン酸)で染色し、核の数をボルテックス後に顕微鏡下でカウントした。全ての実験は3回行い、データは平均±s.dで示す。
Biacore T200及びセンサーチップNTA(GE Healthcare)をこの分析に使用した。HB-EGF及びRドメイン変異体のカップリング定数を製造元の指示書に従い測定した。Ni2+を10μl/mlの流量で60秒間センサーチップ上で捕捉し、続いてヘパリン結合ドメインを欠失したC-末端His-タグ化組換えHB-EGF(Δ・・-EGF-His)を180秒間捕捉した。このセンサーチップに種々の濃度のCRM197, MBP-R(WT)又はMBP-R(変異体)を30μl/mlの流量で600秒間作用させ結合反応を行った。次に解離を30μl/mlの流量で600秒間モニターした。KD及びKA値をBiacore Software(GE Healthcare)を用いて計算した。
CRM197遺伝子及びそのプロモーターをC. diphtheriae C7hm723(β197)株(Kanei, C., T. Uchida, and M. Yoneda, Infect Immun, 1977. 18(1): p. 203-9.)のゲノムDNAからPCRにより得た。使用したプライマーを以下に示す(クローニング用の制限酵素部位は下線で示す):
Fw, 5'-GCGAGATCTTTAGGATAGCTTTACCTAATTATTTTATG-3',
Rv, 5'-CGCCATGCGGCCGCTCAGCTTTTGATTTCAAAAAATAGCG-3'.
増幅したDNAをpK-PIMベクター(Oram, M., Woolston, J. E., Jacobson, A. D., Holmes, R. K. & Oram, D. M. Bacteriophage-based vectors for site-specific insertion of DNA in the chromosome of Corynebacteria. Gene 391, 53-62, (2007).)のBglII-NotI部位に挿入しした。CRM197(R460H)変異体をpK-PIM/CRM197を用いた部位特異的突然変異により構築した。DH10B/pK-PIM, HB101/prk2013及びC. diphtheriaeをそれぞれ37℃で終夜プレ培養し、次にDH10B:HB101:C. diphtheriaeC7(-)=1:1:8の比で混合した。細菌混合物のペレットをBrain Heart Infusion (BHI)寒天プレートに加え、30℃でインキュベートした。細菌混合物をBHI培地で希釈し、ナリジクス酸(20 μg/ml)及びカナマイシン(5 μg/ml)を含有する複合BHI寒天プレートで30℃で16時間インキュベートした。さらに分離したコロニーを20 μg/mlナリジクス酸及び5μg/mlカナマイシンを加えたBHI培地で培養し、pK-PIMを組み込まれたジフテリア菌を得た。
CRM197及びCRM197変異体は既報のように調製及び精製した(Uchida, T., A.M. Pappenheimer, Jr., and R. Greany, J Biol Chem, 1973. 248(11): p. 3838-3844)。
CRM197の組換えRドメインの調製及びHB-EGF-阻害活性
3種類の発現-精製タグ(GST, TRX及びMBP)と融合したCRM197のRドメインを構築し、大腸菌で発現した。GST-R(WT), TRX-R(WT)及びMBP-R(WT)と名付けられたこれらの融合タンパク質は標準的手順又は製造元の指示書に従い調製及び精製した。MBP-融合タンパク質は、構築された融合タンパク質の中で純度及び安定性が最もよかったので、MBP-Rドメインをさらなる検討に使用した。
HB-EGF細胞増殖活性に対するRドメイン変異体の阻害活性を評価するために、DER細胞を一定量のHB-EGFと種々の濃度のRドメイン変異体タンパク質存在下に培養した。代表的データを図3及び図4に示す。MBP-R変異体のID50値を表2に列挙する。
HB-EGFによる細胞増殖作用は3D-培養条件で特に顕著に表れる(Mizushima, H., et al., J Cell Sci, 2009. 122(Pt 23): p. 4277-86.)。そこで、本発明者はコラーゲンゲルを用いた3D-培養条件下でBRL細胞を培養し、BRL細胞のHB-EGF-依存性増殖に対する4つのRドメイン変異体の阻害活性を調べた。4つのRドメイン変異体、MBP-R(H391K)、MBP-R(G466D/D467A/V468T), MBP-R(R460H), MBP-R(R472K)は、MBP-R(WT)よりも効果的に組換えHB-EGFにより誘導されるBRL細胞増殖を抑制した(図6A-C, 表2)。さらに、これらの変異体はMBP-R(WT)と比較して外因性プロHB-EGFを過剰発現するBRLH細胞の増殖をより強く抑制した(図6D, E, 表4)。これらの結果は、MBP-R(WT)と比較した4つのRドメイン変異体のより高い阻害活性は、特定の細胞型又は培養条件に特異的ではないことを示す。
4つのRドメイン変異体が野生型Rドメインと比較してHB-EGFに対する増大した親和性を得ているかを調べるために、該変異体の親和性定数を製造元の指示書に従いBiacoreを用いて求めた。センサーチップ上に捕捉される組換えHB-EGFに対するMBP-Rドメインタンパク質の結合を測定し、結合速度定数及び解離速度定数から親和性定数を計算した。タンパク質の親和性定数値を以下に示す:CRM197, 9.22×108 M-1; MBP-R(R460H), 6.09×108 M-1; MBP-R(G466D/D467A/V468T), 4.44×108M-1; MBP-R(R472K), 4.42×108 M-1; MBP-R(H391K), 3.17×108 M-1; MBP-R(WT), 2.99×108 M-1 (表 4)。親和性定数値はID50値とほぼ相関する。これらの結果は、HB-EGFに対する増大した親和性が4つの変異体の阻害活性の原因であることを示す。
MBP-R(WT)は全長CRM197より低い細胞増殖阻害活性を示し、これは全分子構造が完全な阻害作用を示すのに必要であることを示唆する。従って、本発明者はC. diphtheriae株においてR460H変異を有する全長CRM197を上記のように再構成及び調製し、CRM197の阻害活性と比較した。CRM197(R460H)はDER細胞のHB-EGF-依存性増殖を野生型CRM197より効果的に阻害した(図7)。CRM197及びCRM197(R460H)のID50値は各々1.02 nM及び0.72 nMであった(表5)。
Claims (6)
- CRM197のRドメインを含むポリペプチドであって、前記Rドメインの391位、460位、466位、467位、468位、472位、507位及び520位からなる群から選ばれる少なくとも1個のアミノ酸が置換されたポリペプチド。
- 前記Rドメインの391位、460位、466位、467位、468位及び472位からなる群から選ばれる少なくとも1個のアミノ酸が置換された、請求項1に記載のポリペプチド。
- CRM197のRドメインにおいて、H391K、R460H、G466D、D467A、V468T、R472K、D507N、H520Kからなる群から選ばれる少なくとも1種を含む請求項1又は2に記載のポリペプチド。
- 前記ポリペプチドがCRM197のAドメイン、Tドメイン及びRドメインを含むCRM197改変体である、請求項1〜3のいずれか1項に記載のポリペプチド。
- 請求項1〜4のいずれかに記載のポリペプチドを含む抗腫瘍剤。
- 請求項1〜4のいずれかに記載のポリペプチドに抗腫瘍剤が結合されている、請求項5に記載の抗腫瘍剤。
