Description
前記方法は、観察装置を用いて、
‐試料上の観察対象領域からの発光の少なくとも一部を捕捉し、発光の超臨界角発光成分から生じる発光成分を含む捕捉光信号を得ることと、
‐捕捉光信号にフィルタを適用することにより、選択的に所定の発光成分の振幅を減衰、及び/又は位相を変更してフィルタリング済み光信号を得ることと、
‐フィルタリング済み光信号を、試料上の観察対象領域の画像域に変換することと、を可能とする方法であって、
前記方法は、
‐フィルタリング済み光信号の変調を可能にし、変調において、発光の臨界角発光成分又は亜臨界角発光成分から生じる発光成分を通すことにより、試料上の同一の観察対象領域の画像域が得られ、発光の超臨界角発光成分から生じた捕捉光信号の発光成分のうち少なくとも一部が、変調の対象となることと、
‐複数の画像域を結合することにより試料の有効画像域が少なくとも一つ生成され、結合によって、変調に係る複数の画像域間の差異を強調することと、を特徴とする、方法。
‐観察装置を用いて得られ、結合によって、試料の有効画像域を生成する複数の画像域は、捕捉光信号に連続的にフィルタを適用することで連続的に得られる。
‐本方法には、次の手順が含まれてよい。
a)観察装置及び複数のフィルタを用いて、試料上の同一の観察対象領域から、複数の画像域を取得する手順であって、各フィルタは、複数の画像域のうち1つの画像域を取得するためにそれぞれ用いられ、複数のフィルタは、
+複数のフィルタのうち1つのフィルタが、フィルタリング済み光信号のうち、発光の超臨界角発光成分から生じる発光成分を通し、
+複数のフィルタの全てが、発光の臨界角発光成分又は亜臨界角発光成分から生じる捕捉光信号の発光成分を通し、かつ、発光の臨界角発光成分又は亜臨界角発光成分から生じる捕捉光信号の発光成分に対して、互いに略同一の作用を持ち、
+複数のフィルタのうち少なくとも2つのフィルタが、発光の超臨界角発光成分から生じる捕捉光信号の発光成分のうち少なくとも一部の振幅又は位相に対して、互いに異なる作用を持つように構成される。
b)手順a)で取得した複数の画像域を結合する演算によって、試料の有効画像域を生成し、試料の複数の画像域の画像域同士の差異を強調する。
‐本方法には、次の手順が含まれてよい。
a)観察装置及び2つのフィルタを用いて、試料上の同一の観察対象領域から、少なくとも2つの画像域を取得する手順であって、各フィルタは、2つの画像域のうち1つの画像域を取得するためにそれぞれ用いられ、2つのフィルタは、
‐2つのフィルタのうち1つのフィルタが、前記フィルタリング済み光信号のうち、発光の超臨界角発光成分から生じる発光成分を通し、
‐他方のフィルタが、発光の臨界角発光成分又は亜臨界角発光成分から生じる、前記捕捉光信号の発光成分に対し、一のフィルタと略同一の作用を持ち、発光の超臨界角発光成分から生じる、捕捉光信号の発光成分の少なくとも一部について、一のフィルタより著しく振幅を減衰させるように構成される。
b)手順a)で取得した2つの画像域を結合する演算によって、試料の有効画像域を生成する手順であって、演算は、試料の2つの画像域間の代数差を含む。
‐用いられるフィルタは、発光の臨界角発光成分及び亜臨界角発光成分から生じる捕捉光信号の発光成分の全て又は一部の振幅を部分的に減衰させる作用も有する。
‐前記発光の発光成分から生じる捕捉光信号の、同じ角度θを形成する複数の発光成分は、同一のフィルタで、略同様に、振幅の減衰又は位相の変更がなされる。
‐試料の示す、観察対象となる現象が、所定の特性時間を有する場合、特性時間の半分以下の間隔で、連続して複数の画像域を取得する。
‐試料の画像域のうち1つが、フィルタリング済み光信号のうち、発光の超臨界角発光成分から生じる捕捉光信号の全ての発光成分を、振幅を減衰させずに通すニュートラルフィルタを用いて取得され、試料の他方の画像域は、フィルタリング済み光信号のうち、発光の超臨界角発光成分から生じる捕捉光信号の全ての発光成分を相殺するトータルフィルタを用いて取得される。
‐観察装置は、フルフィールドの液浸レンズを含み、前記フィルタは、油浸レンズの後焦点面及び/又は後焦点面の共役面に位置する。
‐フィルタは絞りを備え、絞りは、開放位置においては発光の超臨界角発光成分から生じる捕捉光信号の全ての発光成分を通すことができ、又、その絞りの程度に応じて、絞りの程度に関連する限界値よりも大きい角度θを有する、発光の超臨界角発光成分から生じる捕捉光信号の発光成分をマスキングできる。
‐観察対象の試料は生物試料である。
Claims (12)
- 屈折率nLを有する媒質(11)中の試料(10)の発光(14,15)を観察する方法であって、前記試料は、nLよりも高い屈折率nsを有する透明の支持体(20)の表面(20a)上に載置され、前記発光は複数の発光成分を含み、該発光成分は所定の振幅及び位相を有し、前記支持体に対して配向され、かつ、前記表面(20a)に対し垂直方向(20b)に角度θを形成する発光成分であり、該複数の発光成分のうち、一方は、角度θが臨界角θc=arcsin(nL/ns)より真に大きい超臨界角発光成分であり、他方は、それぞれ角度θが臨界角θcより小さい又は臨界角θcと等しい、臨界角発光成分又は亜臨界角発光成分であり、前記方法は、観察装置(100)であって、
‐前記試料上の観察対象領域からの発光の少なくとも一部を捕捉し、前記発光の前記超臨界角発光成分から生じる発光成分を含む捕捉光信号を得ることと、
‐該捕捉光信号にフィルタ(170)を適用することにより、選択的に所定の発光成分の振幅を減衰及び/又は位相を変更してフィルタリング済み光信号を得ることと、該フィルタリング済み光信号を、前記試料上の前記観察対象領域の画像域に変換することと、が可能な観察装置(100)を用い、
前記方法は、
‐前記フィルタリング済み光信号の変調を可能にし、該変調において、該発光の臨界角発光成分又は亜臨界角発光成分から生じる発光成分を通すことにより、前記試料上の同一の観察対象領域の画像域(6a、6b)が得られ、前記発光の前記超臨界角発光成分から生じた前記捕捉光信号の前記発光成分のうち少なくとも一部が、前記変調の対象となることと、
‐複数の画像域(6a、6b)を結合することにより前記試料の有効画像域(6c)が少なくとも一つ生成され、該結合により、前記変調に係る前記複数の画像域(6a、6b)間の差異を強調することと、を特徴とする、方法。 - 請求項1に記載の方法であって、前記変調の対象となる前記捕捉光信号の前記発光成分は、角度θが、試料のうち調査対象となる深さの範囲に対応する所定の範囲内である、前記発光の前記超臨界角発光成分から発せられることを特徴とする、方法。
- 請求項1又は2に記載の方法であって、前記観察装置(100)を用いて得られる、結合されることによって試料の有効画像域(6c)を生成する前記複数の画像域(6a、6b)は、前記捕捉光信号に連続的にフィルタ(170)を適用することで連続的に得られることを特徴とする、方法。
- 請求項1から3のいずれかに記載の方法であって、
a)前記観察装置(100)及び複数のフィルタ(170)を用いて、前記試料上の同一の観察対象領域から、複数の画像域を取得する手順であって、各フィルタは、該複数の画像域のうち一の画像域を取得するためにそれぞれ用いられ、前記複数のフィルタは、
‐該複数のフィルタのうち一のフィルタが、前記フィルタリング済み光信号のうち、前記発光の前記超臨界角発光成分から生じる発光成分を通し、
‐該複数のフィルタの全てが、前記発光の前記臨界角発光成分又は前記亜臨界角発光成分から生じる、前記捕捉光信号の発光成分を通し、かつ、前記発光の前記臨界角発光成分又は前記亜臨界角発光成分から生じる前記捕捉光信号の前記発光成分に対して、互いに略同一の作用を持ち、
‐該複数のフィルタのうち少なくとも2つのフィルタが、発光の超臨界角発光成分から生じる捕捉光信号の発光成分のうち少なくとも一部の振幅又は位相に対して、互いに異なる作用を持つように構成される、手順と、
b)手順a)で取得した複数の画像域を結合する演算によって、前記試料の有効画像域を生成し、前記試料の前記複数の画像域の画像域同士の差異を強調する手順と、を含むことを特徴とする方法。 - 請求項1から4のいずれかに記載の方法であって、
a)前記観察装置と2つのフィルタとを用いて、前記試料上の同一の観察対象領域から、少なくとも2つの画像域を取得する手順であって、各フィルタは、該2つの画像域のうち一の画像域を取得するためにそれぞれ用いられ、前記2つのフィルタは、
‐該2つのフィルタのうち一のフィルタが、前記フィルタリング済み光信号のうち、発光の超臨界角発光成分から生じる発光成分を通し、
‐他方のフィルタが、発光の臨界角発光成分又は亜臨界角発光成分から生じる捕捉光信号の発光成分に対し、前記一のフィルタと略同一の作用を持ち、発光の超臨界角発光成分から生じる捕捉光信号の発光成分の少なくとも一部について、前記一のフィルタより著しく振幅を減衰させるように構成される、手順と、
b)手順a)で取得した前記2つの画像域を結合する演算によって、前記試料の前記有効画像域を生成する手順であって、前記演算は、前記試料の前記2つの画像域間の代数差を含む、手順と、を含むことを特徴とする、方法。 - 請求項1から5のいずれかに記載の方法であって、用いられる前記フィルタは、前記発光の臨界角発光成分と亜臨界角発光成分とから生じる前記捕捉光信号の発光成分の全て又は一部の振幅を部分的に減衰させる作用を更に有することを特徴とする、方法。
- 請求項1から6のいずれかに記載の方法であって、前記発光の発光成分から生じる捕捉光信号の、同じ角度θを形成する複数の発光成分は、同一のフィルタで、略同様に、振幅の減衰又は位相の変更がなされることを特徴とする、方法。
- 請求項1から7のいずれかに記載の方法であって、観察対象となる前記試料上の現象が所定の特性時間を有する場合、該特性時間の半分以下の間隔で、連続して複数の画像域を取得することを特徴とする方法。
- 請求項1から8のいずれかに記載の方法であって、
‐前記試料の前記複数の画像域のうち一の画像域は、前記フィルタリング済み光信号のうち、発光の超臨界角発光成分から生じる捕捉光信号の全ての発光成分を、振幅を減衰させずに通すニュートラルフィルタを用いて取得され、
‐前記試料の前記複数の画像域のうち他方の画像域は、前記フィルタリング済み光信号のうち、発光の超臨界角発光成分から生じる捕捉光信号の全ての発光成分を相殺するトータルフィルタを用いて取得されることを特徴とする方法。 - 請求項1から9のいずれかに記載の方法であって、前記観察装置(100)は、フルフィールドの油浸レンズ(110)を含み、前記フィルタ(170)は、該油浸レンズの後焦点面(400)及び/又は該後焦点面の共役面(420)に位置することを特徴とする、方法。
- 請求項10に記載の方法であって、前記フィルタ(170)は絞り(176)を含み、該絞りは、開放位置であるとき発光の超臨界角発光成分から生じる捕捉光信号の全ての発光成分を通すことができ、又、前記絞り(176)の程度に応じて、該程度に対応する限界値よりも大きい角度θを有する、発光の超臨界角発光成分から生じる捕捉光信号の発光成分をマスキングできることを特徴とする、方法。
- 請求項1から11のいずれかに記載の方法であって、観察対象となる前記試料は生物試料であることを特徴とする方法。
