Description
a)コラーゲンと糖を反応させた糖化コラーゲン対照群と、コラーゲン、糖及び皮膚弾力改善効能を評価しようとする候補物質を反応させた糖化コラーゲン実験群とを製造し、対照群及び実験群それぞれに線維芽細胞を混合し、ゲルを作る段階;b)培養挿入体を利用して3次元的形態を構築する段階、c)上記b)段階の培養挿入体の内部にa)段階で作った対照群及び実験群のゲルをそれぞれ入れ、培養し、真皮模倣体を製造する段階;d)製造した模倣体の形態を3次元にイメージ化する段階;及びe)上記d)段階のイメージ化結果から弾力変化を評価する段階。
本発明において使用する真皮模倣体は、下記の方法で製造することができる
コラーゲンとリボースを反応させて、コラーゲン糖化(glycation)を誘導した後、コラーゲンゲルと真皮模倣体をそれぞれ作って、コラーゲン糖化が皮膚に与える影響を評価した。
図2の1)は、コラーゲンゲルが正常に収縮しない2次元的結果を示している。これは、リボース処理によってコラーゲンゲルが収縮せず、まるで横に広がったように見える。
皮膚弾力改善に効果がある物質をスクリーニングするために、本発明による上記参考例1の真皮模倣体を利用した。
Claims (2)
- a)コラーゲンと糖を反応させた糖化コラーゲン対照群と、コラーゲン、糖及び皮膚弾力改善効能を評価しようとする候補物質を反応させた糖化コラーゲン実験群とを製造し、対照群及び実験群それぞれに線維芽細胞を混合し、ゲルを作る段階;
b)培養挿入体を利用して3次元的形態を構築する段階;
c)上記b)段階の培養挿入体の内部にa)段階で作った対照群及び実験群のゲルをそれぞれ入れて培養し、真皮模倣体を製造する段階;
d)製造した模倣体の形態を3次元にイメージ化する段階;及び
e)上記d)段階のイメージ化結果から弾力変化を評価する段階
を含む真皮模倣体を利用した皮膚弾力改善効能評価方法。 - 請求項1に記載の皮膚弾力改善効能評価方法を利用した皮膚弾力を改善させる化粧料のスクリーニング方法。
