Abstract
Description
この出願は、2010年8月27日に出願された米国仮特許出願第61/377,665号からの優先権を主張する。
本発明は、多能性幹細胞に由来する心筋細胞前駆細胞および心筋細胞をSIRPAの発現に基づいて濃縮するための方法に関する。
ヒト胚性幹細胞(hESC)およびヒト人工多能性幹細胞(hiPSC)が培養下で心臓血管細胞を発生させる潜在能力は、これらの系統(lineages)の特異化、拘束および成熟を支配する細胞間相互作用および分子制御因子を研究するための強力なモデル系、ならびに薬物試験および再生医学のストラテジーのためのヒト心筋細胞の独特かつ無制限の供給源を提供する1−4(非特許文献1〜非特許文献4)。しかしながら、混入する細胞型が、インビトロでは薬物反応および他の機能特性に影響し得、インビボでは移植後の異常な成長および奇形腫形成のリスクを高め得るので、この注目すべき潜在能力の実用化は、高度に濃縮された心筋細胞集団を再現可能に生成できる技術次第である5(非特許文献5)。ヒト多能性幹細胞(hPSC)は、最適な心臓の条件下で誘導されると、効率的に分化することにより、心筋細胞、平滑筋細胞、線維芽細胞および内皮細胞を含む混合された心臓血管集団を作製する3(非特許文献3)。所与のいずれのhPSC株の場合も、心筋細胞はその集団の最大70%に相当し得るが、この系統を生成する効率は、種々の幹細胞株間で大幅に異なる。誘導条件のさらなる操作は、未だ、幅広いhPSC株から純粋な心筋細胞集団を作製するためのストラテジーをもたらしていない。
ある態様において、心筋細胞および心筋細胞前駆細胞について細胞の集団を濃縮する方法が提供され、その方法は、心筋細胞および心筋細胞前駆細胞が単離される予定の細胞の集団を提供する工程;およびその集団からSIRPAを発現している細胞を単離する工程を包含し;ここで、その細胞の集団は、心筋細胞および心筋細胞前駆細胞に分化するように誘導されたヒト多能性幹細胞の集団を含む。
ヒト心筋細胞に特異的な細胞表面マーカーを同定するためのハイスループットフローサイトメトリースクリーニングの使用が本明細書中に記載される。ここで、本発明者らは、細胞表面レセプターSIRPAが、hPSCに由来する心筋細胞上ならびにヒト胎児心筋細胞上に発現されることを報告する。本発明者らは、SIRPAに対する抗体を用いた細胞選別を使用して、最大98%の心筋細胞からなる集団をhPSC分化培養物から単離することが可能であることを実証する。
HPSCの維持および分化
HPSCを、記載されているように26維持した。胚様体(EB)を、以前に記載されているように2,3心臓血管系統に分化させた(図1a)。簡潔には:0日目(d0)にEBを作製し、分化の初日にわたって(d0〜d1)BMP4(1ng/ml)を加えた。d1に、EBを回収し、誘導培地(塩基性線維芽細胞成長因子(bFGF;2.5ng/ml)、アクチビンA(6ng/ml)およびBMP−4(10ng/ml))に再懸濁した。その培地をd4に交換し、血管内皮成長因子(VEGF;10ng/ml)およびDKK(150ng/ml)を補充した。培地を再度d8に交換し、VEGF(20ng/ml)およびbFGF(10ng/ml)を補充した。EBを、この実験全体にわたってStemPro−34(Invitrogen)において培養した。培養物を、d0〜d12は5%CO2、5%O2、90%N2環境において維持し、次いで、残りの培養期間は5%CO2/大気環境に移した。
5日目〜12日目のEBに対する解離手順:hPSC分化実験から作製されたEBを、0.25%トリプシン/EDTAを用いて解離した。13日目以降のEBおよびヒト胎児組織に対する解離手順:hPSC分化培養物から得られたEBを、Hanks溶液(NaCl 136mM、NaHCO3 4.16mM、NaPO4 0.34mM、KCl 5.36mM、KH2PO4 0.44mM、デキストロース5.55mM、Hepes 5mM)中のII型コラゲナーゼ(1mg/ml;Worthington,LS004176)において、静かに振盪しながら一晩室温でインキュベートした28。翌日、その細胞懸濁液に、等量の解離溶液(Hanks溶液中:タウリン(taurin)、10mM、EGTA 0.1mM、BSA 1mg/ml、II型コラゲナーゼ1mg/ml)を加え、完全に解離するためにEBを静かにピペッティングした。細胞を遠心分離し(1000rpm,5分)、濾過した。分化の40日目を過ぎたEBの場合、単一の細胞に完全に解離するために、0.25%トリプシン/EDTAによるさらなる処理が必要な場合がある。
免疫染色は、以下の1次抗体:ウサギ抗心臓トロポニンI(Abcam;1:100)、マウス抗SIRPA(BioLegends;1:100)を使用して、以前に記載されたように13行った。使用された二次抗体は:ヤギ抗マウスIgGCy3(Jackson ImmunoResearch;1:400)、ロバ抗マウスIgG−Alexa488(Invitrogen;1:400)だった。核を対比染色するためにDAPIを使用した。ミトコンドリアを染色するためにMito Tracker Red(Invitrogen)を使用した。染色された細胞は、蛍光顕微鏡(Leica CTR6000)を使用して可視化し、Leica Application Suiteソフトウェアを使用して画像を得た。
RNAqueous−Micro Kit(Ambion)を用いて全RNAを調製し、RNaseフリーDNase(Ambion)で処理した。Superscript III逆転写酵素(Invitrogen)とともにランダムヘキサマーおよびOligo(dT)を使用して、500ng〜1μgのRNAをcDNAに逆転写した。以前に記載されたように13、QuantiFast SYBR Green PCR Kit(Qiagen)を使用し、MasterCycler EP RealPlex(Eppendorf)において、QPCRを行った。発現レベルをハウスキーピング遺伝子TATAボックス結合タンパク質(TBP)に対して正規化した。サンプル間の正規化のためのTBPに加えて、ゲノムDNAをDNA標準物質として使用した。ゲノムDNAに存在する標的遺伝子のコピー数は、直接計算され得る(ヒトゲノムサイズ:2.7×109bp(=1.78×1012ダルトン)は、単一コピー遺伝子の6.022×1023コピーに相当し;1μgのゲノムDNAは、単一コピー遺伝子の3.4×105コピーに相当する)。RT−qPCRグラフのY軸は、TBPのコピー数で除した目的の遺伝子のコピー数であり、ゆえに、実験間を比較し得る任意単位であるが絶対単位である。
hESC由来心筋細胞上に発現される新規マーカーの同定
HES2 hESC株は、適切な濃度のアクチビンAおよびBMP4で誘導されたとき(図1a)、効率的かつ再現可能に分化して、心臓血管系統細胞を発生する2,3。分化培養物の動態解析から、BRACHYURY(T)の発現によって定義される原始線条様の集団(2〜4日目)から、初期中胚葉の発生(MESP1;3〜4日目)ならびにNKX2−5およびISLET1(ISL1)陽性心臓前駆体の出現(4〜8日目)への段階的な発生の進行が明らかになった。MYH6(αMHC)、MYH7(βMHC)およびMYL7(MLC2a)のアップレギュレーションならびにその後のMYL2(MLC2v)の発現と一致して(図1b)、分化の9〜12日目に初めて収縮する心筋細胞が検出された。hESCに由来する集団における心臓特異的遺伝子のいくつかの発現レベルは、胎児および成人の心臓組織において見られるレベルよりもかなり低かった。低レベルのNEUROD1およびFOXA2の発現は、それらの培養物に、神経外胚葉または内胚葉に由来する相当な数の細胞が混入していなかったことを示す。心筋細胞の発生をリアルタイムでモニターすることを可能にするために、本発明者らは、上記プロトコルを、HES3 hESCのNKX2−5遺伝子座に挿入されたEGFP cDNAを含むNKX2−5−GFPレポーターhESC株に適用した(Elliottら、投稿済み)。最初のNKX2−5−GFP+細胞は、分化の7〜8日目に発生した。NKX2−5−GFP+集団のサイズは、時間と共に増大し、12〜20日目に最大に達した(図8)。NKX2−5−GFP ESCに由来する胚様体(EB)の落射蛍光下での解析によって、それらの細胞の大部分において核のGFP発現が確認された。NKX2−5−GFP発現の動態が、HES2培養物におけるNKX2−5発現の開始と密接に対応することから、両方のhESC株由来の心臓の特異化(specification)が、分化の6〜8日目に起こると示唆される(図1b、図8)。20日目の培養物における高い割合のNKX2−5−GFP+細胞は、使用された分化プロトコルが、このhESC株からの心筋細胞の発生を効率的に促進することを実証する。
心筋細胞の濃縮された集団を作製するためにSIRPA表面レセプターの発現を使用することができるか否かを評価するために、SIRPA陽性(SIRPA+)およびSIRPA陰性(SIRPA−)の画分を、分化の8、12および20日目におけるHES2由来EBから細胞選別によって単離し、細胞内フローサイトメトリーによって心臓トロポニンT(cTNT)の発現について解析した(図3a)。選別前(未選別,PS)集団の解析は、cTNTの発現が、分化中の対応する段階において、SIRPAの発現と密接に対応することを実証した(PS:d8、d12、d20)。選別後、各段階からのSIRPA+画分は、cTNT+心筋細胞について高度に濃縮された一方で、SIRPA−画分では、これらの細胞が枯渇していた。SIRPA−画分中に存在する少数のcTNT+細胞が、選別手順からの夾雑物であるのか、または真のSIRPA陰性心筋細胞に相当するのかは、不明である。複数の実験におけるFACSベースの分離は、有意に濃縮された心筋細胞集団を再現可能にもたらした(SIRPA+:8日目(95.2%±1.9)、12日目(94.4%±1.7)、20日目(89.6%±3.6);SIRP−:8日目(13.0%±2.1)、12日目(14.3%±3.9)、20日目(15.7%±6.0))(図3b)。SIRPA+およびSIRPA−の選別された集団の純度ならびに選別手順からの細胞回収の効率は、図15および図20(表1)にまとめられている。
SIRPAの発現が、他のhPSC由来集団中の心臓系統を特徴付けるか否かを決定するために、本発明者らは次に、2つの異なるhiPSC株であるMSC−iPS1(Y2−1としても知られる)および38−2から作製したEBを解析した13,14。cTNT+細胞の割合によって実証されるように、両方の株からの心臓分化の効率は低かった(MSC−iPS1:12.2%±5.6、38−2:26.7%±5.7;図4a)。同様の低レベルのSIRPA発現が、両方のEB集団において検出された。両方のiPSC株由来のSIRPA+細胞のFACSは、cTNT+心筋細胞について有意に濃縮された集団をもたらした(SIRPA+:MSC−iPS1(67.0%±3.6)、38−2(71.4%±3.8);SIRPA−:MSC−iPS1(4.9%±2.1)、38−2(6.2%±0.9))(図4a,b)。これらのSIRPA+集団は、対応するSIRPA−細胞よりも有意に高いレベルのNKX2−5、MYH6、MYH7、MYL2およびMYL7を発現した。hESC由来細胞を用いたときに観察されたように、DDR2、PDGFRB、THY1およびNEURODを含む非筋細胞マーカーは、SIRPA−画分に分離した(図4b、cおよびデータ示さず)。これらのデータは、このプロトコルを用いたときの、心臓系統に効率的に分化しない細胞株を含む一連の多能性幹細胞株から濃縮された心臓集団をもたらすためのこのマーカーの有用性を明らかに実証するものである。
SIRPAが、初代ヒト心筋細胞上で発現されるか否かを決定するために、本発明者らは次に、RT−qPCRによって胎児(妊娠18〜20週)および成人の心臓組織の発現パターンを解析した。図5aに示されるように、SIRPA転写物は、すべての胎児由来心臓組織(左心房(LA)および右心房(RA)細胞、左心室(LV)および右心室(RV)細胞、心尖部(AP)ならびに房室接合部(AVJ))において、20日目のhESC由来細胞に見られるレベルに匹敵するレベルで検出された(図3a)。SIRPAは、未分化なhESC(d0)またはコントロールHEK(ヒト胎児腎)細胞では発現されなかった。胎児の心臓と同様に、成人の心臓においてもSIRPAの発現が検出されたことから、その発現が、ヒト心臓発生の種々の段階において心筋細胞を特徴付けると示唆される。高レベルのSIRPAが、成人の脳および肺において検出され(図18a)、多くの他の組織では低レベルで見られた。これらの低レベルは、このレセプターを発現すると知られる組織マクロファージの存在を反映し得る15,16。SIRPAリガンドであるCD47が、ほとんどの組織において発現されたことから、以前の研究において記載されたパターンが裏付けられた(図18b)15。免疫蛍光染色は、SIRPAが、胎児の心室細胞の表面膜上に局在するが、Mito Tracker Redで同時染色されなかったことによって示唆されるようにミトコンドリア膜などの他の膜画分上には存在しないことを示した(図5b)。フローサイトメトリー解析から、すべての胎児心臓組織における高い割合のSIRPA+細胞がそれぞれの画分中のcTNT+細胞のパーセンテージと相関するレベルで明らかになった(図5c、d)。
最近、本発明者らは、KDRとPDGFRAとの同時発現が、BMP4およびアクチビンAで処理した後の心臓中胚葉の誘導をモニターする信頼性のある方法を提供することを報告した2(図6a)。この研究は、KDR+PDGFRA+集団の誘導が、心筋細胞集団の発生において必要不可欠な最初の工程であることを示したが、すべてのKDR+PDGFRA+集団が分化して心臓系統細胞を生じるわけでなかった(このタイプの集団の例:30ng/mlのBMP4および外来性のアクチビンAなし(A0)によって誘導されたもの)。SIRPAが、分化中の集団の心臓になる潜在能力を初期段階においてより正確に予測し得るか否かを決定するために、本発明者らは、種々の濃度のアクチビンAおよびBMP4を用いて誘導された9日目のEBにおいてその発現をモニターした(図6b)。同じ集団を、5日目にKDRおよびPDGFRAの発現について(図6a)ならびに20日目にcTNTの発現について(図6c)評価した。5日目におけるKDR+PDGFRA+集団のサイズと20日目におけるcTNT+細胞の割合との間に相関はほとんどなかったが、9日目における最も大きいSIRPA集団を含む培養物(アクチビンA 6ng/ml、BMP4 10ng/ml)は、その後の時点において最大数のcTNT+細胞を含んだ。SIRPAの発現は、試験されたほとんどの条件に対するcTNTの出力(output)とよく相関し、最も高いレベルのSIRPAは、20日目における最も高い心筋細胞の発生を予測した(図6d)。これらのデータは、9日目におけるSIRPAの発現が、心筋細胞になる潜在能力の信頼性のある指標であること、ならびに同様に、hPSCが心臓系統に定方向分化するための誘導プロトコルをモニターするためおよび最適化するために使用され得ることを実証する。
本発明者らのフローサイトメトリースクリーニングは、心筋細胞を認識する抗体に加えて、分化培養物中の非筋細胞集団を標識する抗体のパネルも同定した。抗CD90(THY1、線維芽細胞上で発現される)、抗CD31(PECAM1、内皮細胞上で発現される)、抗CD140B(PDGFRB、平滑筋細胞上で発現される)および抗CD49A(INTEGRIN1A)を含むこの抗体のセットはすべて、20日目のHES2由来EBの種々の割合のSIRPA−集団を認識した(図7a/d)。これらの抗体の組み合わせは、培養物中の非筋細胞(SIRPA−)の大部分を特徴付けた(図7c,選別前)。非筋細胞マーカーを発現している細胞を枯渇させることによって心筋細胞について濃縮することが可能であるか否かを決定するために、本発明者らは、これらの抗体を組み合わせ、20日目のEBを系統陽性(LIN+)および系統陰性(LIN−)画分に選別した(図7b)。このアプローチは、いかなる結合した抗体または磁気ビーズも含まない濃縮された集団を作製する際に有利である。予想どおり、LIN−集団は、SIRPA+細胞について有意に濃縮された一方で、LIN+集団では、心筋細胞が枯渇していた(図7c/e)。LIN−およびLIN+細胞に対するFACS後の細胞回収の効率は、図21(表2)にまとめられている。遺伝子発現解析によって、PECAM1、PDGFRB、THY1およびDDR2を含む非筋細胞特異的遺伝子が、LIN+画分において主に発現される一方で、心臓遺伝子発現が、LIN−画分に限定されることが明らかになった(図7f)。LIN−画分は、ゼラチンコートされたディッシュ上に平板培養されるか、または細胞塊として再凝集されると、高い割合の収縮する心筋細胞を含む集団を作製した(データ示さず)。同じ抗体の系統カクテルもまた、iPSC(MSC−iPS1)由来の20日目のEB集団の非筋細胞(SIRPA−)画分を標識したことから(図19)、この枯渇アプローチが、変動する分化効率を有する種々のPSC株に適用され得ることが示唆される。
Claims (30)
- 細胞の集団を心筋細胞および心筋細胞前駆細胞について濃縮する方法であって:
(a)心筋細胞および心筋細胞前駆細胞が単離される予定の該細胞の集団を提供する工程;ならびに
(b)該集団からSIRPAを発現している細胞を単離する工程;
を包含し、ここで、該細胞の集団は、心筋細胞および心筋細胞前駆細胞に分化するように誘導されたヒト多能性幹細胞の集団を含む、方法。 - 前記ヒト多能性幹細胞が、胚性幹細胞である、請求項1に記載の方法。
- 前記ヒト多能性幹細胞が、人工多能性幹細胞である、請求項1に記載の方法。
- 前記ヒト多能性幹細胞が、細胞分化を誘導するのに有効な量の少なくとも1つの誘導因子に曝露される、請求項1〜3のいずれか1項に記載の方法。
- 前記少なくとも1つの誘導因子の量が、前記集団中のSIRPA濃度に基づいて最適化される、請求項4に記載の方法。
- 前記少なくとも1つの誘導因子が、サイトカインを含む、請求項4および5のいずれか1項に記載の方法。
- 前記少なくとも1つの誘導因子が、好ましくは40ng/mlまでの濃度、さらに好ましくは約6ng/mlまたは約30ng/mlの濃度の、アクチビンAを含む、請求項4〜6のいずれか1項に記載の方法。
- 前記少なくとも1つの誘導因子が、好ましくは40ng/mlまでの濃度、さらに好ましくは約10ng/mlの濃度の、骨形成タンパク質4を含む、請求項4〜7のいずれか1項に記載の方法。
- 前記ヒト多能性幹細胞が、さらに骨形成タンパク質インヒビターに曝露される、請求項4〜8のいずれか1項に記載の方法。
- 前記骨形成タンパク質インヒビターが、Dorsomorphin、Nogginおよび可溶性骨形成タンパク質レセプターからなる群から選択される、請求項9に記載の方法。
- 前記ヒト多能性幹細胞が、さらにVEGF、DKKおよびbFGFのうちの少なくとも1つに曝露される、請求項4〜10のいずれか1項に記載の方法。
- 前記ヒト多能性幹細胞が、約1日間〜約5日間、好ましくは約3日間、前記誘導因子に曝露される、請求項4〜11のいずれか1項に記載の方法。
- 前記ヒト多能性幹細胞の誘導の開始と前記SIRPAを発現している細胞を単離する工程との間の時間が、約5日間〜約45日間、好ましくは約8日間〜約25日間である、請求項1〜12のいずれか1項に記載の方法。
- 前記SIRPAを発現している細胞が、該細胞によるNkx2.5発現の開始時点ごろに現われる、該細胞によるSIRPAの発現の開始後に単離される、請求項1〜13のいずれか1項に記載の方法。
- SIRPA細胞表面抗原を有する前記細胞が、該細胞によるNkx2.5発現の開始時と、該細胞による収縮および心臓特異的構造タンパク質発現の開始時との間に単離される、請求項1〜13のいずれか1項に記載の方法。
- CD90、CD31、CD140BおよびCD49Aのうちの少なくとも1つを発現している細胞を前記集団から枯渇させる工程をさらに包含する、請求項1〜15のいずれか1項に記載の方法。
- CD90、CD31、CD140BおよびCD49Aのうちの1つを発現している前記細胞が、対応する抗体を用いて枯渇される、請求項16に記載の方法。
- 前記SIRPAを発現している細胞が、SIRPA特異的リガンドを用いて単離される、請求項1〜17のいずれか1項に記載の方法。
- 前記SIRPAを発現している細胞が、抗SIRPA抗体もしくは抗体フラグメントまたは抗体様分子、好ましくは抗SIRPA抗体を用いて単離される、請求項18に記載の方法。
- 前記SIRPAを発現している細胞が、磁気ビーズを用いて単離される、請求項18または19に記載の方法。
- 前記SIRPAを発現している細胞が、フローサイトメトリーを用いて単離される、請求項1〜20のいずれか1項に記載の方法。
- 前記集団中の細胞が、SIRPA遺伝子座の制御性調節エレメントに作動可能に連結されたレポーター遺伝子を含み、ここで、該レポーター遺伝子は、SIRPAを発現する細胞において発現され、該SIRPAを発現している細胞を単離する工程は、該レポーター遺伝子を発現している細胞を単離する工程を包含する、請求項1〜17のいずれか1項に記載の方法。
- 前記レポーター遺伝子が、細胞傷害性因子に対する耐性を付与する、請求項22に記載の方法。
- 前記レポーター遺伝子が、細胞表面タグである、請求項22に記載の方法。
- 前記細胞の濃縮された集団が、心筋細胞および心筋細胞前駆細胞を少なくとも60%、好ましくは少なくとも90%、さらに好ましくは98%含む、請求項1〜24のいずれか1項に記載の方法。
- 請求項1〜24に記載の方法のいずれか1つを用いて得られた心筋細胞および心筋細胞前駆細胞の濃縮された集団。
- 心筋細胞および心筋細胞前駆細胞について濃縮された細胞の単離された集団であって、該細胞の集団は、心筋細胞および心筋細胞前駆細胞を少なくとも60%、好ましくは少なくとも90%、さらに好ましくは98%含む、集団。
- 細胞の集団から心筋細胞および心筋細胞前駆細胞を単離するためのSIRPAの使用であって、ここで、該細胞の集団は、心筋細胞および心筋細胞前駆細胞に分化するように誘導されたヒト多能性幹細胞の集団を含む、使用。
- 細胞の集団を心筋細胞および心筋細胞前駆細胞について枯渇させる方法であって:
(a)心筋細胞および心筋細胞前駆細胞が枯渇される予定の該細胞の集団を提供する工程;ならびに
(b)該集団からSIRPAを発現している細胞を枯渇させる工程;
を包含し、ここで、該細胞の集団は、心筋細胞、心筋細胞前駆細胞および非心筋細胞に分化するように誘導されたヒト多能性幹細胞の集団を含む、方法。 - 細胞の集団を心筋細胞および心筋細胞前駆細胞について濃縮する方法であって:
(a)心筋細胞および心筋細胞前駆細胞が単離される予定の該細胞の集団を提供する工程;ならびに
(b)CD90、CD31、CD140BおよびCD49Aのうちの少なくとも1つを発現している細胞を該集団から枯渇させる工程;
を包含し、ここで、該細胞の集団は、心筋細胞および心筋細胞前駆細胞および非心筋細胞に分化するように誘導されたヒト多能性幹細胞の集団を含む、方法。
