Abstract
Description
本発明は、一般的にオキサゾールおよびチアゾール化合物およびその薬学的に許容される塩類、エステル類またはプロドラッグに関する。本発明は、さらに、薬学的に許容される担体と一体となった当該化合物の組成物、当該化合物の使用、その製造および関連中間体に関する。
キネシン類は、微小管に結合し、機械力を発生させるためにアデノシン三リン酸を使用するモータータンパク質である。キネシン類は、約350アミノ酸残基を有するモータードメインにより特徴付けられる。数種のキネシンモータードメインの結晶構造が解析されている。
一つの態様において、本発明は置換オキサゾールおよびチアゾール化合物およびその薬学的に許容される塩類、エステル類またはプロドラッグ、その製造、医薬組成物およびKSP仲介疾患の処置のための使用に関し、ここで、該化合物は式(I):
R1はアルキル、分枝鎖アルキルおよび置換アルキルから成る群から選択され;
R2は水素、アルキルおよび置換アルキルから成る群から選択され;

本発明の化合物は容易に入手可能な出発物質から、下記の一般的方法および手順を使用して製造できる。特記しない限り、出発物質は市販されており、当分野で周知である。典型的なまたは好ましい工程条件(すなわち、反応温度、時間、反応物のモル比、溶媒、圧力)が記載されているとき、特記しない限り、他の工程条件も使用できることは当然である。最適な反応条件は、使用する特定の反応物または溶媒により変わるが、当該条件は、日常的な最適化方法により当業者が決定できる。

上記のとおり、ここに記載する化合物は、上に記載する多様な医薬送達系で使用するのに適当である。加えて、投与された化合物のインビボ血清半減期を延長するために、化合物をカプセル封入してよく、リポソームの内腔に導入してよく、コロイドとして製造してよく、または本化合物の長い清半減期を提供する他の慣用の技術を用い得る。例えば、Szoka, et al.、米国特許番号4,235,871、4,501,728および4,837,028(この各々を引用により本明細書に包含させる)に記載のとおり、多様な方法がリポソームの製造に利用可能である。

(Denver, Colorado, USA)から購入した。ヒトKSPのモータードメイン(Eg5、KNSL1)をクローン化し、発現させ、95%を超える均一性まで精製した。Biomol GreenをAffinity Research Products Ltd.(Matford Court, Exeter, Devon, United Kingdom)から購入した。微小管およびKSPモータータンパク質(すなわち、KSPモータードメイン)を、アッセイ緩衝液(20mM Tris−HCl(pH7.5)、1mMMgCl2、10mM DTTおよび0.25mg/mL BSA)中、最終濃度35μg/mL微小管および45nM KSPまで希釈した。微小管/KSP混合物を37℃で10分間プレインキュベートし、KSPの微小管への結合を促した。
KSP阻害剤で処置した腫瘍細胞株における細胞増殖阻害
細胞を、96ウェルプレートに約500細胞/96ウェルプレートウェルの密度で播種し、24時間増殖させる。次いで、細胞を種々の濃度の化合物で72時間処理する。100μlのCellTiter-Glo(登録商標)溶液を添加する。CellTiter-Glo(登録商標)アッセイは、細胞溶解後にウェルに存在するATPの量を測定する;遊離されたATPは、酵素ルシフェラーゼおよびその基質ルシフェリンを含む酵素反応に使用される。発光の量はATPの量に比例し、それはさらにウェル中の生存細胞数に比例する。(Promega product catalog #G7573, CellTiter-Glo(登録商標) Luminescent Cell Viability Assay参照)。次いで、細胞を暗所で30分間インキュベートする。発光の量を、ウェルあたりの細胞数と相関するWallac Triluxプレートリーダーを使用して各ウェルで測定する。DMSO(0.5%)のみを入れたウェル中の生存細胞数は0%阻害の指標として働き、細胞なしのウェルを細胞増殖の100%阻害として働く。50%増殖阻害(GI50)を生じる化合物濃度を、連続72時間化合物暴露後に、ログ変換した用量値対細胞数(対照のパーセント)のシグモイド用量−応答曲線からグラフ的に決定する。
使用した細胞株を下に上げる。
細胞増殖アッセイを上記のとおり行う。
クローン形成性軟寒天アッセイプロトコール
ヒト癌細胞を、6ウェルプレートに3×105細胞/ウェルで播種する。翌日、ある濃度の目的の化合物を各ウェルに添加する。24時間および48時間インキュベーション後、細胞を回収し、洗浄し、計数する。以下の工程をMultimek 96ロボットを使用して行う。ウェル辺り500個の生存細胞を、細胞がウェル底に接着することを防止するためにPolyHemaでコートした96ウェルプレートに播種する。アガロース(3%原液)を融解し、温媒体で希釈し、細胞に最終濃度0.5%で添加する。軟寒天が固化した後、プレートを37℃で6日間インキュベートする。アラマーブルー色素を細胞に添加し、プレートをさらに6時間インキュベートする。光学密度の変化をTecanプレートリーダーで測定し、それは軟寒天に形成したコロニー数と相関する。癌性細胞は本寒天上で増殖でき、故に光学密度の増加を示す。光学密度読み取り値の減少は、癌細胞が阻害されていることを意味する。本発明の化合物は光学密度の減少を示すことが企図される。
Claims (27)
- 式(I):
R1はアルキル、分枝鎖アルキルおよび置換アルキルから成る群から選択され;
R2は水素、アルキルおよび置換アルキルから成る群から選択され;
R3は−L1−A1であり、ここで、L1は−C(O)−、−C(S)−、−S(O)−および−S(O)2−から成る群から選択され、A1はアルキル、置換アルキル、アルコキシ、置換アルコキシ、アリール、置換アリール、ヘテロアリール、置換ヘテロアリール、シクロアルキル、置換シクロアルキル、ヘテロシクロアルキル、置換ヘテロシクロアルキルおよびNR8R9から成る群から選択され;
R4は水素、アルキル、置換アルキル、アルケニル、置換アルケニル、アルキニルおよび置換アルキニルおよび場合により置換されていてよいピロリジニルから成る群から選択され;
またはR3およびR4は、それらが結合している窒素原子と一体となって5〜7員ヘテロシクロアルキルまたは置換ヘテロシクロアルキル基を形成し、ここで、場合により1個の炭素環原子はO、SまたはNR11から成る群から選択されてよく;
XはOまたはSであり;
R6はシクロアルキル、ヘテロシクロアルキル、アリールおよびヘテロアリールから成る群から選択され、この全ては場合により−(R10)mで置換されていてよく、ここで、R10はここに定義したとおりであり、mは1、2、3または4であり、mが2、3または4であるとき、各R10は同一でも異なってもよく;
R7は−L2−A2であり、ここで、L2はC1−C5アルキレンであり、A2はアリール、置換アリール、ヘテロアリール、置換ヘテロアリール、シクロアルキル、置換シクロアルキル、ヘテロシクロアルキルおよび置換ヘテロシクロアルキルから成る群から選択され、ただし、R7はXに結合しておらず;
R8は水素およびアルキルから成る群から選択され;
R9は水素、ヒドロキシ、アルキル、置換アルキル、アリール、置換アリール、ヘテロアリール、置換ヘテロアリール、シクロアルキル、置換シクロアルキル、ヘテロシクロアルキルおよび置換ヘテロシクロアルキルから成る群から選択され;
またはR8およびR9は、それに結合した窒素原子と一体となってヘテロシクロアルキルまたは置換ヘテロシクロアルキルを形成し;
R10はシアノ、アルキル、置換アルキル、アルケニル、置換アルケニル、アルキニル、置換アルキニル、−CF3、アルコキシ、置換アルコキシ、ハロおよびヒドロキシから成る群から選択され;
R11は水素、アルキル、置換アルキル、−SO2アルキルおよび−SO2置換アルキルから成る群から選択される。〕
の化合物またはその薬学的に許容される塩。 - R1がアルキルである、請求項1に記載の化合物。
- R2がC1−4アルキルまたはHである、請求項2に記載の化合物。
- R2がHである、請求項3に記載の化合物。
- R1がi−プロピルまたはt−ブチルである、請求項4に記載の化合物。
- R3が−L1−A1から選択され、ここで、L1が−C(O)−、−C(S)−、−S(O)−および−S(O)2−から成る群から選択され、A1がアルキルおよび置換アルキルから成る群から選択される、請求項5に記載の化合物。
- R3が−L1−A1から選択され、ここで、L1が−C(O)−であり、A1が置換アルキルである、請求項6に記載の化合物。
- 請求項R4が水素、アルキルおよび置換アルキルから成る群から選択される、請求項7に記載の化合物。
- R4が置換アルキルまたは−CH2−フルオロピロリジニルである、請求項8に記載の化合物。
- R3が−L1−A1から選択され、ここで、L1が−C(O)−であり、A1が−CH(CH3)OHである、請求項9に記載の化合物。
- R4が−(CH2)2−CH(CH2F)NH2または−CH2−3−フルオロピロリジニルである、請求項10に記載の化合物。
- R6がアリールおよびヘテロアリールから成る群から選択され、その全てが場合により−(R10)mで置換されていてよく、ここで、R10が上に定義したとおりであり、mが1、2、3または4であり、mが2、3または4であるとき、各R10が同一でも異なってもよく;
R7が−L2−A2であり、ここで、L2がC1−C2アルキレンであり、A2がアリールおよびヘテロアリールから成る群から選択される、

- R7が−CH2−フェニルおよびR6が1,4−ジフルオロ−フェニルである、請求項14に記載の化合物。
- 治療有効量の請求項1に記載の化合物および薬学的に許容される担体を含む、医薬組成物。
- さらに少なくとも1種の付加的癌処置剤を含む、請求項18に記載の組成物。
- 付加的癌処置剤がイリノテカン、トポテカン、ゲムシタビン、イマチニブ、トラスツマブ、5−フルオロウラシル、ロイコボリン、カルボプラチン、シスプラチン、ドセタキセル、パクリタキセル、テザシタビン、シクロホスファミド、ビンカアルカロイド、アントラサイクリン系、リツキシマブおよびトラスツマブから成る群から選択される、請求項19に記載の組成物。
- 哺乳動物患者における、少なくとも一部KSPにより介在される障害の処置方法であって、当該処置を必要とする哺乳動物患者に治療有効量の請求項1に記載の化合物または請求項18に記載の組成物を投与することを含む、方法。
- 障害が細胞増殖性疾患である、請求項21に記載の方法。
- 細胞増殖性疾患が癌である、請求項22に記載の方法。
- 癌が肺および気管支;前立腺;乳;膵臓;結腸および直腸;甲状腺;胃;肝臓および肝内胆管;腎臓および腎盂;膀胱;子宮体;子宮頸;卵巣;多発性骨髄腫;食道;急性骨髄性白血病;慢性骨髄性白血病;リンパ性白血病;骨髄球性白血病;脳;口腔および咽頭;喉頭;小腸;非ホジキンリンパ腫;黒色腫;および絨毛結腸腺腫から成る群から選択される、請求項23に記載の方法。
- 付加的癌処置剤がイリノテカン、トポテカン、ゲムシタビン、イマチニブ、トラスツマブ、5−フルオロウラシル、ロイコボリン、カルボプラチン、シスプラチン、ドセタキセル、パクリタキセル、テザシタビン、シクロホスファミド、ビンカアルカロイド、アントラサイクリン系、リツキシマブおよびトラスツマブから成る群から選択される、請求項19に記載の方法。
