Abstract
Description
- 4種の主要分子:ケルセチン、エモジン、4,5,6,7−テトラブロモ−1H−ベンゾトリアゾール(TBB)及びその誘導体並びに5−オキソ−5,6−ジヒドロインドロ−(1,2−a)キナゾリン−7−イル)酢酸を含むATP競合的阻害剤(Duncanら、Biochimica et Biophysica Acta、2008、1784、33〜47)。CK2の触媒活性を特異的に阻害できるこれらの分子としては、CK2に対してより顕著な選択性を示すTBB、5,6−ジクロロ−1−β−D−リボフラノシルベンズイミダゾール(DRB)の誘導体(Sarnoら、FEBS lett.、2001、496、44〜48;Duncanら、Molecular and Cellular Proteomics、2008、7、1077〜1088)を挙げることができる。しかし、TBB及び他のキナーゼ基質類似体は、細胞ATPを用いて他の既知又は未知タンパク質の活性を阻害できるが;このような製品の特異性は確実でないので、それらの使用はインビボでは除外されている;
− 米国特許出願公開第2002/147163号及び米国特許第6,455,307号に記載されている、CK2のαサブユニットに対するアンチセンスオリゴヌクレオチド。しかし、インビトロで立証されているCK2の阻害は、部分的且つ一過性であって、非常に高用量のアンチセンスオリゴヌクレオチド(細胞の感受性に応じて数十又は数百μg/mL)を必要とする;
− 国際公開WO2005/005632に開示されているsiRNA。
− 環Cは、ピロール複素環又はピラジン複素環であり、
− R1は、−H又は炭素数1〜6のアルキル基であり、R1は、環Cがピロール複素環である場合にのみ存在し、
− R2は、−H、二重結合によって環Dに連結されている酸素原子、ハロゲン原子、1,2−エタンジオキシ基(−O−CH2−CH2−O)、1,2−エタンジチオ基(−S−CH2−CH2−S−)、フェニル基、ベンジル基、−OR、−SR、−NHR若しくは−N(R)2基(式中、Rは−H又は炭素数1〜6のアルキル基である)であり、
− R3は、R2が二重結合によって環Dと連結されている酸素原子、1,2−エタンジオキシ基又は1,2−エタンジチオ基である場合にのみ存在し、R3は水素原子又は炭素数1〜6のアルキル基を表し、
− R4及びR5は、同一であるか異なり、水素原子、炭素数1〜6のアルキル基、又は炭素原子3及び4を介して環Dと縮合されている置換若しくは非置換アリール環を表し、
− R6及びR7は、水素原子、又は炭素原子6及び7を介して環Bと縮合されているアリール環を表し、前記アリール環は少なくとも1つの−OH又は−NHR’基(式中、R’は、−H又は炭素数1〜6のアルキル基である)によって置換されており、
− R8及びR9は、水素原子、又は炭素原子8及び9を介して環Bと縮合されているアリール環を表し、前記アリール環は少なくとも1つの−OH又は−NHR’基(式中、R’は、−H又は炭素数1〜6のアルキル基である)によって置換されており、

・ R6及びR7が少なくとも1つの−OH若しくは−NHR’基によって置換されているアリール環を表す場合には、R8及びR9は水素原子であり、

(i)化合物III又はVIIIを得るための、化合物IとIIとの又はIとVIIとの縮合と
(ii)化合物IV又はIXを得るための、得られたヒドラゾンIII又はVIIIの熱的フィッシャーインドール化(thermal Fischer indolization)によるインドール化と
(iii)木炭上パラジウムの存在下における化合物IV又はIXの芳香族化による、化合物V又はXの直接生成と

式(I)の化合物1及び2を、Bioconjugate Chem.、2003、14、120〜135に記載され且つ公表された手順に従って、合成した。
式(I)の化合物の作用機序を特徴付けるために、それらを、CK2、c−kit及びトポイソメラーゼII活性並びにそれらのDNA挿入能について試験した。
− CK2活性に対する式(I)の化合物の作用を、Prudentら(Prudent,R.ら(2008)Mol Cell Biochem 316(1−2):71〜85)に従って測定する:指示濃度の化合物3μL、3μLのCK2α(36ng)、及び100μMのペプチド基質(RRREDEESDDEE、配列番号:1)と10mM MgCl2と100μM [γ-32P]-ATPとを含む混合物を含む最終容量18μL中で実施される放射能分析で、化合物を試験した。分析は、室温で5分間実施後に、60μLの4%TCA添加によって終了させた。ペプチド基質への32Pの取り込みを、Filholら(Biochemistry 29:9928〜36(1990))に既に記載されているようにして、測定した。
化合物3及び化合物4の式はそれぞれ、アリール環の−OH基が−OCH3基によって置き換えられた本発明の化合物1及び2の式に対応する。化合物4は具体的には、先行技術において開示された化合物の定義に対応し:化合物6は、Thi My−Nhung Hoangら、Cell Cycle、2009年、第8巻、第5号、765〜772頁に記載されている。
1.材料及び方法
生細胞へのCK2に対する化合物の阻害能力を試験するための細胞アッセイを開発した。
5’−GCTCAAGCTTCGGATTCTGAAGACGACGATACCGCGGGCCCG−3’(配列番号:3)及び
5’−CGGGCCCGCGGTATCGTCGTCTTCAGAATCCGAAGCTTGAGC−3’(配列番号:4)並びに
突然変異誘発2には
5’−GCTCTGAGGAGGTGTCCGAGGTCGACTGGTTCTGAGGGCTCCGT−3’(配列番号:5)及び
5’−CGGGCCCGCGGTATCGTCGTCTTCAGAATCCGAAGCTTGAGC−3’(配列番号:6)
を用いて、Quickchange−Site Directed mutagenesis kit(Stratagene)による引き続く2ラウンドの突然変異後に得られた(Prudentら、Biochim Biophys Acta.2008;1780(12):1412〜20)。
HeLa細胞を、12ウェルプレート中に細胞106個/ウェルで蒔き、リポフェクタミン(lipofectamine)試薬(Invitrogen)を用いてCK2活性レポータープラスミド(pEYFPc1−SβS)をトランスフェクトした。
非処理又はDMSO処理HeLa細胞において、CK2は十分に活性であり、CK2活性レポーターは殆どリン酸化アイソフォームとして検出できた(図2Aの下方のバンドを参照)。
既知のCK2阻害剤は細胞生存率を低下させるので、いくつかの細胞型の生存率を、化合物1及び2による48時間処理後に測定した。
− 細胞:HeLa(ヒトの子宮頚部腺癌)及びU373(ヒトのグリア芽細胞腫)がん細胞株、
− HeLa細胞の増殖阻害を、以下のようにして評価する:Hela細胞を、96ウェルプレートに細胞1.5×104個/ウェルで蒔いた。翌日、培地を、漸増濃度の阻害剤又は対照としての当量のDMSOを含む、新しい培地(10%(v/v)ウシ胎児血清(FBS、BioWest)が補充されたダルベッコ(Dulbecco)培地(Invitrogen Life Technologies,Inc.))に置き換えた。2日後、細胞生存率を、CellTiter−Glo(登録商標)Luminescent Cell Viability Assay(Promega)を用いて製造業者推奨基準に従って測定した。
HeLa細胞の細胞生存率の低下:
化合物1及び2は、低用量であっても、腫瘍細胞の強い増殖阻害を誘発する(図3)。
5μMの化合物1及び化合物2は、48時間の処理後に、これらの細胞株の細胞増殖を減少させる効果がある(図4)。

次に、実施例4において化合物1及び化合物2で予め処理した細胞を、ヨウ化プロピジウムの混和後に細胞周期分析に供する。
分析から、化合物1及び化合物2で処理されたHeLa細胞及びU373細胞が、G2/M期におけるかなりの蓄積によって細胞周期停止を示すことがわかった。HeLa処理細胞ではアポトーシス細胞の付随的増加も観察されるが、U3763細胞ではアポトーシス細胞の付随的増加は観察されず、これらの細胞はアポトーシスに抵抗性である(図5A)。sub−G1、G1、S及びG2/M期におけるHeLa細胞及びU373細胞の比率(%)を、図5Bの表に要約する。

腫瘍細胞の腫瘍形成に対する化合物の作用を、コロニー形成アッセイで試験する。
図6に示すように、5μMの化合物1は、コロニー形成の50%阻害の誘発に十分である。
1.材料及び方法
実験手順は全て、地域の倫理委員会に従った(Comite regional d’ethique pour l’Experimentation animale CREEA、Rhone Alpes−プロトコール番号286)。雌のHarlan胸腺欠損ヌードマウス(週齢6〜8週)の右側腹部に、U373細胞7.5x105個を皮下播種した。腫瘍が±50mm3(体積=長さ×幅×高さ)に達したら、22.5% PEG3350、0.45%BSAに溶解された化合物1(0.14mg/100μL/注射−マウス4匹)又は同一緩衝液に溶解された当量のDMSO(最終濃度10%−マウス3匹)(対照群)を、週に3回(2日毎に)2週間、動物に腹腔内投与した。体重及び腫瘍体積を、週2回測定した。腫瘍体積が約1000mm3となった時点で、実験を終了させた。結果を、平均値±平均値の標準誤差(SEM)として示す。
化合物1が注射されたマウスでは、DMSOが注射されたマウスと比較して、腫瘍発生率が全ての時点で著しく低下したことが観察される(図7A)。最初の化合物注射後15日で、化合物1が注射されたマウスの腫瘍は、DMSOが注射されたマウスの腫瘍の4.7倍小さかった。更に、記録された体重変化は、統計的に有意でなかった。これは、化合物1が、適用用量において忍容性が高かったことを示している(図7B)。
構造−活性相関はまた、いくつかの位置が置換に対して許容性があることを示している。CK2−式(I)の化合物の複合体のX線共構造(co-structure)から、キナーゼと阻害剤との分子相互作用へのより深い洞察が得られる。
CK2−式(I)の化合物の複合体のX線構造において、化合物(I)のフェニル環の−OH基とVal116の窒素骨格(nitrogen backbone)との間に水素結合が形成されていることが観察でき、これは、この位置に遊離−OH基が必要な根拠を示している。

- 医薬品として使用するための、下記式(I):
− 環Cは、ピロール複素環又はピラジン複素環であり、
− R1は、−H又は炭素数1〜6のアルキル基であり、R1は、環Cがピロール複素環である場合にのみ存在し、
− R2は、−H、二重結合によって環Dに連結されている酸素原子、ハロゲン原子、1,2−エタンジオキシ基、1,2−エタンジチオ基、フェニル基、ベンジル基、−OR、−SR、−NHR、−N(R)2基(式中、Rは−H又は炭素数1〜6のアルキル基である)であり、

- R2がハロゲン原子であることを特徴とする、請求項1から4に記載の化合物。
- R2が塩素原子であることを特徴とする、請求項5に記載に記載の化合物。
- R4が水素原子であることを特徴とする、請求項1から6に記載の化合物。
- R5がメチル基であることを特徴とする、請求項1から7に記載の化合物。
- 請求項1から10のいずれか一項に記載の式(I)の化合物を、薬学的に許容される賦形剤又は担体と共に含むことを特徴とする医薬組成物。
- がん;自己免疫疾患及び炎症性疾患;感染性疾患;糖尿病;血管新生関連障害;網膜症及び心肥大から選択される障害及び/又は疾患の予防及び/又は治療を目的とする医薬品の調製への、請求項1から10のいずれか一項に記載の化合物の使用。
