Abstract
【解決手段】線条体においてプロサイモシンα遺伝子発現不全非ヒト哺乳動物であって、対応する野生型動物と比較して、
(1)脳虚血処置に対して脆弱である、
(2)運動性が劣る、
(3)加齢によって、上記(1)および(2)の症状を自然発症する、および
(4)ドパミンD2受容体アゴニスト、ドパミン代謝阻害剤、NMDA受容体拮抗剤またはドパミンD1受容体アゴニストによって上記(1)、(2)および(3)の症状が改善する、
ことを特徴とする動物など。

Description
ハンチントン病の症状のうちジストニアについては、経験的にL−DOPA製剤が投与されてきた(非特許文献4)。しかしながら、L−DOPA製剤は有効性に優れる反面、作用持続時間が短く、長期使用で効果の変動(wearing off,on−off)やジスキネジアなどの症状を生じるため患者のQOL低下の原因となる。そのため、L−DOPA製剤に次ぐ効果を有し、作用持続時間が長いドパミンアゴニストが注目されるようになった。現在、我が国では6種類のドパミンアゴニストが臨床で使用されているが、なかでもプラミペキソールはハンチントン病における筋剛直に対し治療効果を示すとの報告があり(非特許文献5)、有効性の確立に期待が集まっている。
本発明者は、これらの知見に基づいてさらに研究を重ねた結果、本発明を完成するに至った。

また、哺乳動物以外にもニワトリなどの鳥類を、本発明で対象とする「非ヒト哺乳動物」と同様の目的に用いることができる。
ES細胞は胚盤胞期の受精卵の内部細胞塊(ICM)に由来し、インビトロで未分化状態を保ったまま培養維持できる細胞をいう。ICMの細胞は将来、胚本体を形成する細胞であり、生殖細胞を含むすべての組織の基になる幹細胞である。ES細胞としては、既に樹立された細胞株を用いてもよく、また、EvansとKaufmanの方法(ネイチャー(Nature)第292巻、154頁、1981年)に準じて新しく樹立したものでもよい。例えば、マウスES細胞の場合、現在、一般的には129系マウス由来のES細胞が使用されているが、免疫学的背景がはっきりしていないので、これに代わる純系で免疫学的に遺伝的背景が明らかなES細胞を取得するなどの目的で、例えば、C57BL/6マウスやC57BL/6の採卵数の少なさをDBA/2との交雑により改善したBDF1マウス(C57BL/6とDBA/2とのF1)から樹立されるES細胞なども良好に用いることができる。BDF1マウスは、採卵数が多く、かつ卵が丈夫であるという利点に加えて、C57BL/6マウスを背景に持つので、これ由来のES細胞は疾患モデルマウスを作製したとき、C57BL/6マウスと戻し交雑することでその遺伝的背景をC57BL/6マウスに代えることが可能である点で有利に用いられ得る。
また、第二次セレクションは、例えば、G−バンディング法による染色体数の確認等により行うことができる。得られるES細胞の染色体数は正常数の100%が望ましい。
実質的に同一のアミノ酸配列を有する蛋白質としては、前記アミノ酸配列を含み、該アミノ酸配列からなる蛋白質と実質的に同質の活性を有する蛋白質が好ましい。実質的に同質の活性としては、例えば、loxP配列結合作用、loxPに挟まれたDNA配列の切り出し作用などが挙げられる。実質的に同質とは、それらの活性が定性的に同等であることを示す。したがって、loxP配列結合作用、loxPに挟まれたDNA配列の切り出し作用などの活性が同等であることが好ましいが、これらの活性の程度(例、約0.01〜100倍、好ましくは約0.5〜20倍、より好ましくは約0.5〜2倍)や蛋白質の分子量などの量的要素は異なっていてもよい。また、Cre蛋白質をコードするDNAは、GenBank accession No.NC_005856.1として登録されている塩基配列に含まれるDNAが用いられ得る。
用いる非ヒト哺乳動物の齢や飼育条件等は動物種によってそれぞれ異なるが、例えばマウス(好ましくはC57BL/6J(B6)などの近交系マウス、B6と他の近交系とのF1など)を用いる場合は、雌が約4〜約6週齢、雄が約2〜約8ヶ月齢程度のものが好ましく、また、約12時間明期条件(例えば7:00−19:00)で約1週間飼育したものが好ましい。
通常、F0動物は相同染色体の一方にのみ導入DNAを有するヘテロ接合体として得られる。また、個々のF0個体は相同組換えによらない限り異なる染色体上にランダムに挿入される。相同染色体の両方に導入DNAを有するホモ接合体を得るためには、F0動物と非トランスジェニック動物とを交雑してF1動物を作製し、相同染色体の一方にのみ導入DNAを有するヘテロ接合体の兄妹同士を交雑すればよい。1遺伝子座にのみ導入DNAが組み込まれていれば、得られるF2動物の1/4がホモ接合体となる。
プロサイモシンαをコードするポリヌクレオチドの塩基配列に、相補的もしくは実質的に相補的な塩基配列またはその一部を有するアンチセンスDNAとしては、プロサイモシンαをコードするポリヌクレオチドの塩基配列に相補的もしくは実質的に相補的な塩基配列またはその一部を含有し、該ポリヌクレオチドの発現を抑制し得る作用を有するものであれば、いずれのアンチセンスDNAであってもよい。
特に、プロサイモシンαをコードするポリヌクレオチドの相補鎖の全塩基配列のうち、(a)翻訳阻害を指向したアンチセンスDNAの場合は、プロサイモシンα蛋白質のN末端部位をコードする部分の塩基配列(例えば、開始コドン付近の塩基配列など)の相補鎖と約70%以上、好ましくは約80%以上、より好ましくは約90%以上、最も好ましくは約95%以上の相同性を有するアンチセンスDNAが、(b)RNaseHによるRNA分解を指向するアンチセンスDNAの場合は、イントロンを含むプロサイモシンαをコードするポリヌクレオチドの全塩基配列の相補鎖と約70%以上、好ましくは約80%以上、より好ましくは約90%以上、最も好ましくは約95%以上の相同性を有するアンチセンスDNAがそれぞれ好適である。
さらに、本発明のアンチセンスDNAは、プロサイモシンαのmRNAもしくは初期転写産物とハイブリダイズして蛋白質への翻訳を阻害するだけでなく、二本鎖DNAであるプロサイモシンα遺伝子と結合して三重鎖(トリプレックス)を形成し、RNAの転写を阻害し得るものであってもよい。あるいはDNA:RNAハイブリッドを形成してRNaseHによる分解を誘導するものであってもよい。
(1)脳虚血処置に対して脆弱である、
(2)運動性が劣る、
(3)加齢によって、上記(1)および/または(2)の症状を自然発症する、および
(4)ドパミンD2受容体アゴニスト、ドパミン代謝阻害剤、NMDA受容体拮抗剤またはドパミンD1受容体アゴニストによって上記(1)、(2)または(3)の症状が改善する、
を有する。これらの表現型は、従来公知のハンチントン病モデルマウスとしてのハンチンチントランスジェニックマウスやハンチンチンノックインマウスの表現型と一致する部分もあるが、それらのマウスは、線条体全体の神経細胞が傷害されているため、上記の表現型の進行が早く、より重篤であり、実際のハンチントン病の病態を正確に再現していない。また、本発明の線条体においてプロサイモシンα遺伝子発現不全非ヒト哺乳動物は、運動性異常を示すことからジストニア(dystonia)症状を示すモデルマウスの表現型とも一致する。ここでジストニアは、虚血性疾患に由来して発症するもの(二次性ジストニア)であってよく、虚血性疾患としては脳梗塞(脳血栓、脳塞栓)が挙げられる。
C57BL/6J系雄性マウス(野生型マウス)に加え、線条体に特異的に発現するGng7のプロモーターにより制御された相同組換え酵素(Cre)を発現するTGマウス(Gng7−Cre(+/−)雄性マウス)、さらにはGng7−Cre(+/−)とFloxedプロサイモシンα遺伝子組み換えマウスを交配させたコンディショナル遺伝子欠損マウス(Gng cre/+;ProTa flox/flox雄性マウス)を実験に用いた。両マウスは若齢マウスとして8−16週齢、高齢マウスとして20−30週齢ともに体重:20−35gの個体を使用した。飼育条件は恒温(22±2℃)、恒湿(55±5%)とし、一般実験用の固形飼料(MF、オリエンタル酵母、東京)、水道水を自由に摂取させた。すべての実験は長崎大学動物実験規則で定める方法に準じて行った。Gng7−Cre(+/+)雄性マウスは東京大学大学院 医学系研究科分子神経生物学教室三品昌美教授より、Floxedプロサイモシンα遺伝子組み換えマウスは大塚製薬株式会社から譲渡された。
マウスプロサイモシンα遺伝子の野生型アリルの構造は図1上段に示すとおりである。また、Floxedプロサイモシンα遺伝子組み換えマウスの有するFloxedアリルは、エキソン1と2の間、およびエキソン3と4の間にloxP配列が相互にタンデムになるようにそれぞれ挿入されており(図1、中段)、Cre蛋白質がloxP配列に作用することによって、2つのloxP配列に挟まれたエキソン2および3を含む配列が欠失する(図1、下段)。なお、Cre蛋白質は、線条体特異的発現プロモーターであるGng7プロモーター下流に連結され、上記のコンディショナルマウスは線条体において特異的にプロサイモシンα遺伝子が機能不全となる。本実施例においては、特に別途記載が無い限り、WTは野生型マウス(C57BL/J6)を、GgはGng7−Cre(+/−)マウスつまりGng7プロモーター下流にcre遺伝子をヘテロ接合型となるように挿入したマウスを、GgFFはGng7発現領域である大脳基底核の線条体領域特異的にcre−loxPシステムでプロサイモシンαを欠損させたマウスを示す。
8−16週齢のWT、GgおよびGgFFマウスを15分間左中大脳動脈を閉塞することで、一過性の脳虚血状態を引き起こした軽度脳卒中モデルの病態を評価した(図2、3)。GgFFマウスの対照群にはGgマウスを用いた。
tMCAO処置後の一過性中大脳動脈閉塞モデルの生存率を経時的に評価した結果、WTマウスでは、生存率は5日間にわたり無変化であった(図2B)。また、Ggマウスでも4日目において90%以上の生存率を示した(図3B)。しかし、GgFFマウスでは生存率を大幅に低下させ、処置後2日において、すでに生存率60パーセント以下に低下した(図3B)。Survival ratio(%)は、tMCAO実施前に測定した値を100%とし日数経過による生存率を百分率で示した。
また、tMCAO処置後の一過性中大脳動脈閉塞モデルの運動機能障害を経日的に評価した結果、WTマウスとGgマウスでは微弱な運動障害しか観察されなかった(図2C、3C)。しかし、GgFFマウスでは4ポイントにも及ぶ障害が観察された(図3C)。本願の実施例において、Clinical scoreは次に示す1〜5の臨床スコアで表される。1:右前肢の麻痺、2:一方向性の動き、3:体勢を保てず一方向に傾く、4:自発運動の消失、5:死。
さらに、tMCAO処置後の一過性中大脳動脈閉塞モデルの運動機能を経日的に評価した結果、GgFFマウスはGgマウスに比して、滞在時間が短くなる傾向が見られた(図3D)。ロータロッドの回転速度は、脳卒中病態モデルマウスで実施可能な6rpmにて行った。
実施例2と同様に、8−16週齢のWT、GgおよびGgFFマウスを15分間左中大脳動脈を閉塞することで、一過性の脳虚血状態を引き起こした軽度脳卒中モデルに対して、プラミペキソールによる生存率、運動機能障害の改善効果を評価した(図4、5)。15分間のtMCAO処置を施したGgFFマウスに0.01mg/kgプラミペキソールを腹腔内投与した際の症状改善効果を経過観察した結果をsurvival ratio、Clinical scoreおよびLatency to fallで示す。対照群には同じく15分間のtMCAO処置を施したWTマウス、Ggマウスを用いた。PPXはプラミペキソールを示す。
その結果、WTマウスでは投与群と非投与群に差は見られなかったが、GgFFマウスでは、投与群において生存率が75パーセントに改善した(図4B)。また、運動機能障害についても、GgFFマウスの投与群において顕著に改善した(図5C)。さらに、運動機能について、6rpmと12rpmの両条件下で、WTマウスに比較してGgFFマウスの運動機能は低下していたが、プラミペキソールの腹腔内投与により改善が認められた(図5B、C)。
実施例2と同様に、8−16週齢のWT、GgおよびGgFFマウスを15分間左中大脳動脈を閉塞することで、一過性の脳虚血状態を引き起こした軽度脳卒中モデルに対して、セレギリンによる生存率、運動機能障害の改善効果を評価した(図6、7)。15分間のtMCAO処置を施したGgFFマウスに10mg/kgセレギリンを腹腔内投与した際の症状改善効果を経過観察した結果をsurvival ratio、Clinical scoreおよびLatency to fallで示す。対照群には同じく15分間のtMCAO処置を施したWTマウス、Ggマウスを用いた。
その結果、GgFFマウスでは、投与群において生存率が顕著に改善した(図6B)。また、運動機能障害についても、GgFFマウスの投与群において顕著に改善した(図6C)。さらに、運動機能について、6rpmと12rpmの両条件下で、非投与群に比べて、GgFFマウスの運動機能は、セレギリンの腹腔内投与により改善が認められた(図7B、C)。
大脳基底核の線条体領域特異的にプロサイモシンαを欠損したマウスで週齢依存的なClasping reflex(抱擁反射)症状の発症ならびに運動機能の低下に関する評価を行った(図8)。Clasping scoreは異常がない場合0、前肢同士もしくは後肢同士を結ぶ行動を1、前後肢を結び抱え込む行動を2として判定した。その結果、WTマウスおよびGgマウスは異常が確認できなかったが、10週齢および20週齢のGgFFマウスのClasping scoreは、有意に高いことが確認できた(図8A、B、C)。
また、運動機能の評価について、マウスを回転式のロッド上で歩行させ、ロッドに滞在できる時間を測定(ロータロッド法)することで評価した。ロッド上を歩行するには全身の各部位をコントロールしバランスをとる必要があるため、本試験は協調運動性の評価法として用いられている。マウス用ロータロッド(MK−610A;室町器械)を最大滞在時間を60秒間、ロッドの回転速度を30rpm、40rpmの条件下で試験を行った。本実施例において本試験を行う前には、20rpmの回転速度で60秒間の訓練試行を1時間の間隔をあけて1日につき4回、3日間連続で行った。30rpmはロータロッド回転速度1分毎30回転すること、40rpmは1分毎40回転することを示す。その結果、GgFFマウスのロータロッド上での滞在時間は、WTマウスおよびGgマウスに比して有意に短いことが確認できた(図8D、E)。
Clasping reflexおよび運動機能低下を示すGgFFマウスおよびコントロールとしてのGgマウスに対し、ドパミンD2受容体のアゴニストであるプラミペキソール0.01mg/kgを腹腔内に投与し、急性(24時間以内)の症状改善効果を評価した(図9)。Preはプラミペキソール投与前、1hrはプラミペキソール投与1時間後を、以下5hrは5時間後、24hrは24時間後を示す。なお、本願実施例で用いられるプラミペキソールは塩酸プラミペキソール(ビ・シフロール 0.125mg錠、日本ベーリンガーインゲルハイム)を乳棒、乳鉢を用いて粉砕し、生理食塩水に溶解した後、水酸化ナトリウム水溶液でpHを7.0に調整して用いた。各種マウスへは27ゲージの注射針を用いて腹腔内に投与した。投与5時間後において、GgFFマウスの上記の症状が緩和される傾向が観察された。
また、GgFFマウスおよびGgマウスに対し、慢性的にプラミペキソールを0.01 mg/kg腹腔内投与した場合の症状改善効果についても、プラミペキソールの投与方法以外は原則的に上記と同様の方法に従って評価した(図10)。Pre値はプラミペキソール投与前の測定値であり、プラミペキソールは投与1日目から7日目まで毎日投与し、投与してから24時間後に投与1日後の値を測定、以下1日おきに7日目までclasping scoreの判定ならびにロータロッドによる運動機能評価を行った(図10A)。その結果、GgFFマウスのClasping scoreは、投与前に比して経日的に改善する傾向が確認できた(図10B)。また、GgFFマウスのロータロッド上での滞在時間は、投与前に比して経日的に長くなる傾向が確認できた(図10C、D)。
GgFFマウスに対し、ドパミン代謝に関わるMAO−Bの阻害剤であるセレギリン10mg/kgを腹腔内に投与し、急性(24時間以内)の症状改善効果を評価した(図11)。セレギリンは、モノアミン酸化酵素阻害剤(MAOI)のひとつで、選択的にMAO−Bを阻害しドーパミンの分解を防ぐことで、結果的に脳内ドーパミン量を増加させる薬剤である。Preはセレギリン投与前、1hrはセレギリン投与1時間後を、以下5hrは5時間後、24hrは24時間後を示す。その結果、GgFFマウスのClasping scoreは、投与前に比して改善する傾向が確認できた(図11B)。また、GgFFマウスのロータロッド上での滞在時間は、投与前に比して経時的に長くなる傾向が確認できた(図11C、D)。
GgFFマウスに対し、グルタミン酸過剰による細胞死を抑制するNMDA受容体拮抗作用を有するメマンチン10mg/kgを腹腔内に投与し、急性(24時間以内)の症状改善効果を評価した(図12)。Preはメマンチン投与前、1hrはメマンチン投与1時間後を、以下5hrは5時間後、24hrは24時間後を示す。投与24時間後において、GgFFマウスの運動機能は最も回復が見られた。
GgFFマウスに対し、慢性的にメマンチンを10mg/kg腹腔内投与した場合の症状改善効果を示す。Pre値はメマンチン投与前の測定値であり、メマンチンは投与1日目から7日目まで毎日投与し、投与してから24時間後に投与1日後の値を測定、以下1日おきに7日目までclasping scoreの判定ならびにロータロッドによる運動機能評価を評価した(図13)。Pre値はメマンチン投与前の測定値であり、メマンチンは投与1日目から7日目まで毎日投与し、投与してから24時間後に投与1日後の値を測定、以下1日おきに7日目までclasping scoreの判定ならびにロータロッドによる運動機能評価を行った(図13A)。結果として、慢性投与によって顕著に症状が回復することが確認できた。
週齢20−30のGgFFおよびGgの耳片を50μl Extraction buffer(20×SSC 500μl,500mM Tris−HCL pH8.0 200μl,500mM EDTA ph8.0 400μl,10%SDS 1.0ml,1mg/ml ProteinaseK 500μl,DW 7.4ml)に入れ、60℃でOvernightすることによりDNAを抽出した。MangoMix 2xマスターミックス(BIOLINE)を用い、遺伝子型をPCR法により判定した(図14)。プライマーとPCR条件は以下のものを用いた。

週齢20−30のGgFFマウスまたはGgマウスについて、ペントバルビタール麻酔下で開腹開胸後、心臓の右心耳に切れ目をいれ、左心室からK+ free PBSを灌流、脱血させた後、4%パラホルムアルデヒド/0.1M PBで灌流固定した。脳を摘出し、25%sucrose/K+ free PBSに浸漬して4℃で一晩放置した。O.C.T. Compound(Sakura)で包埋し、エタノール/ドライアイスにより、組織を急速凍結させた。クリオスタット(CM1900、ライカマイクロシステムズ株式会社)を用いて、厚さ30 μmの脳切片を作製した。組織切片をPBST(0.1%TritonX−100 in K+ free PBS)で洗浄後1% H2O2に30分置換し、内因性ペルオキシダーゼを分解した。PBSTで洗浄後(以後毎ステップPBST洗浄表記なし)、ブロッキング反応として2%BSA/2%anti−mouse serum(cappel 55435、MP Biomedicals)/PBST溶液で室温にて1時間反応させた。1%BSA/PBST溶液にて1:1000に希釈した抗プロサイモシンα抗体(NT 2F11、ALEXIS Enzo Life Sciences)を4℃にてover night後、Biotinylated α−mouse IgG(1:500 Zymax)in 1%BSA/PBSTに1時間置換した。次に30分前に作成したvectastain ABC液(A液15μl、B液15μl in 1%BSA/PBST 2000μl)に1時間置換しシグナル増強を行った。20mg/mL DAB solutionを10mlのPBSに溶解し1%CoCl2 250μL,1%NiSO4 200μLを少量ずつ加えながら混ぜ3.3μlの30%H2O2(Wako)を入れた溶液に約5分置換し、シランコートされたスライドグラス(松波硝子工業株式会社)に張り付け風乾後アルコール脱水、キシレン透徹を行い、Permount(Fisher Chemicals)で封入後、蛍光位相差顕微鏡(BZ−8000、株式会社キーエンス)で観察した。蛍光染色時は30μm切片作製、PBSTで洗浄後、50%MeOH 10分、100%MeOH 10分浸漬し、PBSTで洗浄後にブロッキング反応を行った。二次抗体にはalexa fluor 488 anti−mouse IgG(1:300 Molecular probe)in 1%BSA/PBSTで2時間反応させた。
本発明者は、Gng7は主に線条体に発現が多く、海馬、小脳にも発現していることをReal−time PCRによってこれまでに確認している。週齢20−30のGgFFマウスの30μm脳切片を用いてプロサイモシンαの免疫組織化学を行った所、線条体、海馬、小脳での顕著な発現低下を観察した(図15A,B)。DAB染色および蛍光染色の20倍視野の写真からは、線条体で7〜8割の細胞でプロサイモシンαが欠損していること、歯状回の内側ではGgFFマウスでも発現が残っていること、小脳ではプルキンエ細胞層ではプロサイモシンα発現が残っており、主に顆粒細胞層でプロサイモシンα欠損が起こっていることが分かった(図15C)。
また、線条体におけるプロサイモシンαの発現をイムノブロット法で確認した。週齢20−30のGgFFマウスまたはGgマウスを断頭したのち氷上でマウスの脳組織を摘出しPBSで洗浄した。マウス用ブレインスライサ(室町機械)で500μmの厚さにスライスしカミソリで線条体、海馬、小脳の各部位を切り出し、SDS sample buffer(500mM Tris−HCl(pH6.8)5ml,10%SDS 10ml,100%glycerol 5mlをMQで50mlにメスアップ)100μlの入ったエッペンチューブに入れる。Bioruptor(Cosmo Bio)で超音波破砕した後、15000rpmにて10分遠心後上清を回収しDC Protein Assay Regent(BIO−RAD laboratories)を用いてタンパク定量を行った。1レーン当たり20μgのタンパク質を15%SDS−PAGEで30mA,300Vの条件でDyeがゲル下端に来るまで泳動を行う。ゲルのタンパク質をニトロセルロースメンブレンにセミドライ式で100mA,30Vの条件で90分間転写する。5%スキムミルク,2%Fetal bovine serum/TBST(TBS,0.1%Tween20)で1時間ブロッキングを行った。一次抗体をブロッキングに用いたBufferにて1000倍に希釈し一晩反応させる。TBSTで3回洗浄後、HRP標識した2次抗体をブロッキングに用いたBufferにて2000倍に希釈し2時間反応させた。TBSTで3回洗浄後した。Super Signal West Pico Chemiluminesent substrateおよびSuperSignal West Dura Extended Duration Substrate(Thermo scientific)にて化学発光させシグナルを検出した。一次抗体には抗プロサイモシンα抗体(1:1000; ALEXIS)および抗β−tubulin抗体(1:1000; Santa Cruz)を用い、二次抗体にはHRP標識抗マウスIgG抗体(1:2000;Promega)およびHRP標識抗ラビットIgG抗体(1:2000;Promega)を用いた。
その結果、Western blotでも線条体、海馬、小脳でのプロサイモシンα発現減少の結果を得た(図15D)。
本発明者はこれまでにプロサイモシンαが虚血ストレス時に神経細胞を保護する作用を有することをin vitro,in vivoともに明らかとしてきた。そこで前記実施例10のGgFFマウスにおいてプロサイモシンαの発現低下していた領域において、神経細胞の形態や細胞数に変化があるかをNissl染色により確認した。Nissl染色は神経細胞を可視化する標準的な組織学的方法として用いられており、神経細胞形質および樹状突起中の粗面小胞体由来のリボソームRNAから構成されるNissl物質を染色する。損傷を受けた場合、神経細胞体内に再分布し、神経細胞死のマーカーとしても用いられている。免疫染色と同様に30μmの脳切片を作製し、シランコートされたスライドグラスに張り付け風乾後、PBSTで洗浄した。MQに10秒程度浸漬後、水浴で37℃に温めておいたcresyl violet溶液(2.5g cresylecht violet,300ml H2O,30ml 1M NaOAc,170ml 1M AcOHをmixし7日間スターラーで撹拌)に15〜30分浸漬し染色する。MQで洗浄後、アルコール脱水、キシレン透徹を行い、Permountで封入後、顕微鏡(Keyence)で観察した。
その結果、GgFFマウスでは線条体、海馬、小脳において顕著な神経細胞死や脱落、神経細胞層の配列の乱れは観察されなかった(図16A,B,C)。
20−30週齢のマウスを用いて新規環境下での自発運動機能をOpen field testにて評価した。約50lxの部屋に試験30分前にマウスを入れておき環境に適応させた。縦70cm×横70cm×高さ30cmのアクリル箱の隅からマウスを入れ30分間、Video tracking system(Muromachi Kikai)を用いてPCにて解析を行った。箱を8×8に分画した際の中心4×4区画での行動時間を不安様行動の指標として測定した。Open field試験において、20−30週齢のGgFFマウスは顕著な自発行動量の低下を認めた(図17A,B)。また行動速度にも低下がみられ、中心滞在時間の低下も観察された(図17C,D)。
これらの結果より、GgFFマウスは週齢依存的な運動機能障害または、不安様行動の促進がみられることが明らかとなった。
さらに、20−30週齢のGgFFマウスは不安行動が促進しているか否かを調べるため、他の不安行動の試験であるMarble burying testおよびNovelty induced hypophagia testを行った。
Marble burying testは、無害なガラス玉を床敷きで覆い隠そうとするマウスの行動が、不合理と認識しつつ繰り返される強迫性障害患者の脅迫行為と見かけ上類似していることから、強迫性障害に関連した不安様行動試験として認知されつつある試験である。抗不安薬の投与により隠すビー玉の数が低下することが報告されている。縦28cm×横45cm×高さ20cmのアクリルケージに床敷を5cm入れ、17mmの青色ビー玉を等間隔に5×5個置く。ケージを薄暗い所に置き、マウスをケージに入れた。30分後マウスを回収し、2/3以上床敷に埋められたビー玉の数をカウントした。この試験においてGgFFマウスが隠したビー玉の数は、Ggマウスと有意な差はなかったが、若干の低下傾向にあり、運動機能低下が観察された(図18A,B)。
Novelty induced hypophagia testは、コンデンスミルクをHomecage(HC)で飲む群と、新規環境(Novelty;N)で飲む群で、飲みに行くまでの時間と飲んだミルクの量を比較し、不安行動を評価する試験である。不安行動が亢進しているマウスは新規環境でミルクを飲みに行くまでの時間が増加し、飲む量も減少するとされている。まずグループ飼いしたマウスを約50lxの薄暗い部屋に入れ、5mlの25%コンデンスミルクを30分間自由に摂取できるようにした。これを1日1回3日間繰り返して、ミルクの味を覚えさせた。4日目は、3日間と同様に薄暗い部屋のHomecageでミルクを飲めるマウス(Homecage群)と、1000lxの明るく照らされた床敷のない透明ケージでミルクを飲めるマウス(Novelty群)の2群に分けて実験を行った。どちらの群も1匹ずつ30分間試行し、ミルクを初めて飲むまでの時間と、5分毎のミルクを飲んだ量を測定した。この試験においてGgFFマウスはHC群、N群ともにミルクを飲みに行くまでの時間、飲む量のどちらも有意な差はなかった(図18C,D)。
これらの結果からGgFFマウスは不安様行動の亢進はみられないことが明らかとなった。
これまでの試験において20−30週齢のGgFFマウスは自発行動量の低下がみられることが明らかとなった。そこで、GgFFマウスの運動機能を詳細に調べるため若齢マウスと同様にロータロッド試験、さらにStationary thin rod試験、Footprint試験を行った。
ロータロッド試験においてGgFFマウスはGgマウスと比べ、20rpmの低速で回るロータ上を歩くことを覚える訓練1回目から落ちる時間が顕著に早かった。またGgFFマウスは訓練3日間の間に少しずつ上達は見られるが、Ggマウスと同等の運動機能にはならなかった(図19A)。4日目により高速の30rpmで試験したところ、訓練の結果と同様にロータ上から落下するまでの時間は短かった(図19B)。
さらに、バランス感覚を調べるため、止まった細い棒上から落ちるまでの時間を調べるStationary thin rod試験を行った。1.5cm diameter,50cm longの滑らかなステンレス棒を地面から40cmの高さに水平に設置し、マウスの怪我防止のため、マウスを棒の中心に乗せ、落下するまでの時間を測定した。最大滞在時間を60秒とし、1時間の間隔をあけて6試行行った。その結果、GgFFマウスはGgマウスと同様に6回の試行において徐々に棒上に乗れるようになり、有意な差はなかった(図19C)。
また歩行機能を調べるFootprint試験では、マウスの前肢および後肢足底をそれぞれ赤および黒インクで塗布し、幅48mm、長さ650mm、高さ230mmの通路上を歩行させた。その際、マウスが暗い場所を好む性質を利用し、後ろからライトで照らして前進させた。歩き始めと歩き終わりを除いた3歩の平均値で以下のパラメータを解析し、歩行機能を評価した。その結果、GgFFマウスに異常な歩行様式はみられず、歩幅(Stride length:片前肢から同前肢までの距離で左右の平均)、前肢両間の距離(Forepaw base:左右の前肢間の距離)、後肢両間の距離(Hindpaw base:左右の後肢間の距離)、前後肢のかぶり(Forepaw/Hindpaw overlap:片前肢と後肢の距離で左右の平均)の4項目すべてでGgマウスと有意な差はなかった(図19D)。
これらの結果より、20−30週齢のGgFFマウスは通常の歩行やバランスといった顕著な運動機能障害はないが、ロータロッド試験のような高度な運動協調性に異常があることが示唆された。
これまでの結果からGgFFマウスは運動機能障害を自然発症することが明らかとなった。そこで、GgFFマウスと同様に線条体が病態となっているパーキンソン病で用いられているドパミンアゴニストを用いてGgFFマウスの運動機能障害が治療できるか検討した。
D1アゴニストであるSKF38393 30mg/kgの腹腔内投与5時間後では、ロータロッド試験における運動機能障害の有意な改善を認めた(図20A)。しかしD2アゴニストのパラミペキソールの投与では最大容量の1mg/kgでも改善効果はみられなかった(図20B)。
この結果から、Gng7発現細胞でプロサイモシンαが欠損することはドパミン作動性神経細胞のバランスになんらかの影響を与えていることが示唆された。
さらに、GgFFマウスに関して、以下の点についても検証を行った。
(1)体重
体重に関しては若齢において有意な差は確認されなかったが、週齢を重ねるごとに体重増加の程度が対照群と比較し低下し、29週齢まででGgマウスと差はないが、30−39週齢で有意な体重の低下がみられた(図21A)。
(2)握力
筋力の評価としてWirehang testを行った。1cm角の格子状の網の上にマウスを乗せ、上下逆さにして30cmの高さに固定する。マウスが落ちるまでの時間を測定し、最大180秒とした。握力を測定するWirehang試験に関してはGgマウスがほぼ最大潜時の180秒間落ちないのに対して、GgFFマウスは20秒程度落下が早かった(図21B)。
(3)うつ様症状
うつ様症状の評価にはTail suspension testおよびPorsolt forced swim testを行った。
Tail suspension testでは、マウスの尾を木の棒の先にテープで固定し、高さ30cmのところに吊るした。無動になるまでの時間と1分毎の無動時間を6分間測定した。その結果、GgFFマウスは全体6分間を通して有意な無動時間の低下を示し、前半3分間は特に顕著であった(図21C)。
また、Porsolt forced swim testでは、直径20cm高さ30cmのアクリル円柱状容器に25±1℃の水を20cm入れた。マウスをその容器に入れ、無動になるまでの時間と1分毎の無動時間を6分間測定した。測定後マウスを回収し、キムタオルで水分をふきとった。その結果、GgマウスとGgFFマウスに有意な差はなかった(図21D)。
(4)社会性
自己の周辺環境への配慮や社会性を調べるため、Nest building testを行った。明暗サイクル(明期8:00〜20:00/暗期20:00〜8:00)の暗期1時間前(19:00)にマウスを1匹ずつ大ケージに分け、Nestlet 2.5g(Ancare)を入れた。翌朝巣の形状を以下の基準でスコアリングし、未使用のNestletの量を量った。
Score1.Nestletの90%が未使用。
Score2.Nestletの50−90%が未使用。
Score3.Nestletの50%以上使用。隅に90%以上Nestletを集めていない。巣らしきものがない。
Score4.Nestletの90%以上使用し、隅に集めている。平坦な巣がある。
Score5.Nestletの90%以上使用し、くぼみのある完全な巣がある。
その結果、巣として使わなかったNestletsの量と巣の形状のスコアリングは、GgFFマウスはどちらの項目でもGgマウスと差がなく、社会性に異常はないことが明らかとなった(図21E)。
(5)学習機能
学習機能の評価にはStep through testを用いた。1日目の電流を覚えさせる訓練ではGgマウスとGgFFマウスに有意な差はなかったことから、両群に同じ条件付けができたと考えられる。しかし2日目の試験ではGgFFマウスは電流がかかる暗箱にGgマウスよりも早く入ってしまい、学習機能の低下が示唆された(図21F)。
本出願は、日本で出願された特願2011−119651(出願日:平成23年5月27日)を基礎としており、その内容はすべて本明細書に包含されるものとする。
Claims (14)
- 線条体においてプロサイモシンα遺伝子発現不全非ヒト哺乳動物であって、対応する野生型動物と比較して、
(1)脳虚血処置に対して脆弱である、
(2)運動性が劣る、
(3)加齢によって、上記(1)および(2)の症状を自然発症する、および
(4)ドパミンD2受容体アゴニスト、ドパミン代謝阻害剤、NMDA受容体拮抗剤またはドパミンD1受容体アゴニストによって上記(1)、(2)および(3)の症状が改善する、
ことを特徴とする動物。 - G蛋白質γ7サブユニット遺伝子プロモーターにより発現制御されたcre遺伝子を有し、かつloxP配列に挟まれたプロサイモシンα遺伝子をホモ接合型で有する、請求項1記載の動物。
- ドパミンD2受容体アゴニストがプラミペキソール、ペルゴリド、カベルゴリン、タリペキソールまたはロビニロールである、請求項1に記載の動物。
- ドパミン代謝阻害剤がセレギリン、エンタカルボン、ドアマンタジン、L−ドーパ、ドロキシドパまたはゾニサミドである、請求項1に記載の動物。
- NMDA受容体拮抗剤がメマンチンまたはCP−101606である、請求項1に記載の動物。
- ドパミンD1受容体アゴニストがSKF38393である、請求項1に記載の動物。
- 動物がマウスまたはラットである、請求項1に記載の動物。
- 請求項1記載の動物に試験化合物を適用し、(1)脳虚血処置に対する生存率および/または(2)運動性を測定することを特徴とする、神経変性疾患または虚血性疾患由来のジストニアの治療/予防薬のスクリーニング方法。
- 請求項1記載の動物であって、週齢が進んだ該動物に試験化合物を適用し、(1)生存率および/または(2)運動性を測定することを特徴とする、神経変性疾患または虚血性疾患由来のジストニアの治療/予防薬のスクリーニング方法。
- 試験化合物がドパミンD2受容体アゴニストである、請求項8または9に記載のスクリーニング方法。
- 試験化合物がドパミン代謝阻害剤である、請求項8または9に記載のスクリーニング方法。
- 試験化合物がNMDA受容体拮抗剤である、請求項8または9に記載のスクリーニング方法。
- 試験化合物がドパミンD1受容体アゴニストである、請求項8または9に記載のスクリーニング方法。
