Abstract
Description
本発明の発明者らは、i)ビタミンKの還元型及び/又はii)ビタミンKアナログの還元型及び/又はiii)ビタミンK前駆体の還元型を含むリストより選択される1つ又はそれ以上の化合物をビタミンK依存性タンパク質の細胞培養培地に補うことによって、同量の同一しかし非還元型のそれぞれのビタミンK及び/又はビタミンKアナログ及び/又はビタミンK前駆体を細胞培養培地に添加する場合に得られる発現収率と比較して、それぞれの活性ビタミンK依存性タンパク質の発現収率が顕著に増加し得ることを、驚くべきことに見出した。
記載の実施態様は本発明の好ましい実施態様を示す一方、本発明の要旨から逸脱することなく、改変が当業者によって考えられることが理解される。従って、本発明の範囲は、添付の特許請求の範囲によってのみ決定される。

懸濁細胞灌流プロセスにおいて、細胞を、動物由来の成分を好ましくは欠く培養培地を含有する種培養容器中へ接種し、細胞が最低限の密度に達するまで増殖させる。続いて、増殖した種培養物を、動物由来の成分を欠く培養培地を含有する大量培養容器へ移し、少なくとも所定の細胞密度が達成されるまで増殖させる。

2.1 単純なバッチプロセス:
単純なバッチプロセスにおいて、細胞を、動物由来の成分を欠く培養培地を含有する種培養容器中へ接種し、細胞が最低限の密度に達するまで増殖させる。続いて、増殖した種培養物を、動物由来の成分を好ましくは欠く培養培地を含有する大量培養容器へ移す。次いで、培地中の栄養素が使い尽くされるまで、培養容器を操作する。
前述したように、単純なバッチプロセスは、培養容器に細胞を接種すること、及び培地中の栄養素が使い尽くされるまでタンクを操作することからなる。これのようなバッチプロセスは、タンクへ栄養素の濃縮溶液を供給することによって延長され得る。これは、プロセス時間を延長し、培養容器内のそれぞれのビタミンK依存性タンパク質の産生の増加を最終的にもたらす。
単純なドローフィルプロセスは、反復バッチ発酵に非常によく似ている。バッチ発酵においては、細胞を培養容器中において増殖させ、培地を実行の終了時に採取する。ドローフィルプロセスにおいては、培養容器を、栄養素が使い尽くされる前に採取する。容器から内容物の全部を取り出す代わりに、タンク容量の一部のみを取り出す(一般的にタンク容量の80%)。
2つのcDNAがFIX由来のリンカー配列によって分離された状態で、ヒト凝固第IX因子cDNAをヒトアルブミンcDNAに対してインフレーム5'にクローニングした。FIXとアルブミンとの間のこのリンカー配列は、FIX活性化に関与する内因性FIX配列に由来し、従って、FIXを活性化する同一の酵素(FXIa又はFVIIa/TF)による融合タンパク質の切断を可能にする。
rIX−FP−PACE/フーリン構築物をCHOK1SV細胞中へエレクトロポレーションし、市販のグルタミンフリーの培地を使用して96ウェルプレート中においてコロニーを選択した。次いで、得られたコロニーを、FIX発現及び活性についてスクリーニングし、所望の発現/活性プロフィールを有する2つのクローン(A2及びA5)をさらなる開発のために選択した。
手順は、亜鉛粉末の触媒添加及びアスコルビン酸の水溶液を使用するメナジオン重亜硫酸ナトリウム(MSB)のインサイチュ還元を含んだ。得られた濾過反応混合物は、アスコルビン酸及び微量のZnの存在下で、MSB及び2つの対応の異性体還元生成物(図3において番号2及び番号3として示される;還元型ビタミンK)を含有した。この濾液混合物を、最終生成物混合物として使用した。
Tatchell, B.S. Furnis. [year, editor]に従って作製した。簡潔には、50mLポリプロピレンチューブ中の亜鉛粉末40gヘ、10% v/v塩酸15mLを添加した。混合物を振盪し、2分間静置した。混合物を500mL Pyrexビーカーへ移し、Milli Q水で数回洗浄した。これはまた、微粒子を除去するためにMilli Q水での大量のエルトリエーションを必要とした。リンスされた活性化亜鉛を、Milli Q水中に懸濁し、フィルター装置へ移し、ここで、それは0.45μm Durapore(登録商標)GV膜上に捕らえられた。亜鉛を50mL HPLC等級アセトニトリルで数回洗浄し、その後、1.5時間60℃でロータリーエバポレーターにて乾燥した。乾燥された活性化亜鉛を、アルゴン下でPyrexボトル中に保存した。
エレクトロスプレーイオン化質量分析(ESI−MS)を備える逆相HPLCによって、還元MSBをアッセイした。
ヒトアルブミンを有する組換え第IX因子融合タンパク質を発現するCHO細胞株を、実施例1及び2に詳述したように作製した。活性分子の発現を増加させる方法を調べるために、培養物へのメナジオン重亜硫酸ナトリウム(MSB)及び還元MSB(rMSB)の添加の効果を評価する多数の実験を行った。この研究の目的は、MSB又はrMSBのいずれか添加が活性分子の発現を増加させることができるかどうか又は発現される抗原性物質の量を減少させることができるかどうかを確認することであった。活性の増加は、ビタミンK3の利用可能性によって駆動される、分子上のグルタミン酸残基のγ−カルボキシル化の増加に起因すると仮定して、分子の活性を特定の第IX因子アッセイキットによって評価した。
培地組成
増殖のために使用した培地は、CHO細胞中での組換えタンパク質の産生のために特別に設計された市販の無血清の化学的に規定された培地であった:「培地A」。クローンは、Lonzaグルタミンシンセターゼ(GS)システムを利用し、グルタミンの非存在下で継代し、25uMメチオニンスルホキシイミン(MSX)によって選択を維持した。生産性測定のために使用した、最後の培養段階において、グルタミン及びMSXを両方とも培地に添加しなかった。
グルコース濃度が2g/Lの下限値に達したらグルコース100g/Lのボーラス添加を培養物に対して行い、いったん下限値濃度が達成されたら、4g/Lの上限量へ残りの培地濃度を増加させるように供給を誘導した。
リアクターは、撹拌のための単一のRushtonインペラー及びリングスパージャーを備える、Sartorius/ Braun 5L皿型底ガラスリアクターであった。特に指定のない限り、全てのリアクター培養を下記の条件下で行った。
体積:合計4リットルで開始(3.2リットル培地及び0.8リットル接種物);
温度:37℃、接種から採取まで;
通気:0.025vvmの最大速度で、溶存酸素(D.O.)セットポイントに応答して散布される、期間について0.0375毎分空隙容量vvmでのヘッドスペースエア;
D.O.:40%飽和のセットポイント;
酸素:0.075vvmの最大流量でのD.O.セットポイントに応答して散布される;
撹拌:100rpmの一定速度で;
pH:7.00±0.05の単一のセットポイント;
塩基:2M NaOH;
酸:100% CO2;
接種物:3×105〜5×105細胞/mLの開始細胞密度。
Innovartis Cedex自動細胞カウンターを利用して製造業者の使用説明書に従って、細胞カウントを行った。Yellow Springs Instruments (YSI) 7100アナライザーを利用して製造業者の使用説明書に従って、グルコース、ラクテート、アンモニア及びグルタミンアッセイを行った。Siemens(以前はBayer)製のRapidLab 248血液ガス分析装置を使用して製造業者の使用説明書に従って、溶存二酸化炭素濃度のオフライン測定を行った。第IX因子抗原の定量化のための内部手順ELISAを使用して、抗原アッセイを行った。“BIOPHEN FIX”発色FIXテストを使用して、第IX因子活性アッセイを行った。全てのアッセイについて接種時から毎日、サンプリングを行った。
フラスコ生産性アッセイを下記に説明するように行った。
実験1:振盪フラスコ中におけるFIX発現の比較
実験1を行い、フラスコ形式でのMSBの添加と比較しての培養パフォーマンスに対する還元MSBの効果を評価した。この実験において調べたテスト条件の概要を表1に示す。細胞を標準フラスコ生産性アッセイへ供し、細胞密度、第IX因子抗原及び第IX因子活性についてのアッセイを行った。このフラスコ生産性アッセイから得られた結果の概要を表2及び表3に示す。
リアクターフェドバッチ条件下で、rIX−FP CHO−K1クローンを標準条件下で培養した。この実験は、MSB添加又は還元MSB添加のいずれかの下での、細胞増殖、抗原産生、及び細胞からの活性rIX−FP産生を評価した。この実験において利用した供給は、下記のアミ
ノ酸が補われたグルコースベースのもの(100g/L)であった:
アスパラギン 21.4g/L;
システイン 5.6g/L;
アスパラギン酸 5g/L。
CHO−K1クローンA5を、2つの培地、培地Aと、CHO細胞中での組換えタンパク質の産生のために特別に設計されたさらに別の市販の無血清の化学的に規定された培地:培地Cとを用いて、延長ドローフィル条件下で培養した。培地のパフォーマンスを比較する一方で、MSB又は還元MSBの添加の効果を評価した(表5)。前の実験と同様に、この研究からのアウトプットは、細胞の増殖パラメーター並びに活性rIX−FP分子及びrFIX−FP抗原の産生であった。
細胞増殖及び生存能力に対する濃度増大のMSB及び還元MSBの効果を試験するために、さらなる研究を行った。MSB添加又は還元MSB添加を除いて、8個のリアクターを同一のパラメーター下で培養した。前の実験と同様に、還元MSBの濃度は計算値であり、真の濃度は計算値よりも低い場合がある。使用した培地は、実験1に記載されるような培地Aであった。添加を毎日細胞当たりに基づいて増加させ(表7)、細胞増殖に対する効果をモニタリングした。リアクターをドローフィル様式で行い、第1サイクルは6日間の期間を有し、第2サイクルは4日間の期間を有した。
この研究は、細胞増殖及び活性第IX因子融合タンパク質分子の生産性に対するMSB及び還元MSB添加のパフォーマンスを評価するために行った。最初に、さらなる研究を支援するためのデータを得るために、CHO−Sクローンを用いてフラスコ実験を行った。この実験において、データを72時間及び96時間の両方の時点で集めた。72時間で、増加した活性収率が、還元MSB添加で達成された。抗原性収率もまた、MSB添加と比較して増加し;しかし、最も高い比率が、還元MSB添加によって維持された。この場合において、還元MSBの単回添加は、MSBの二重添加と比較して増加したしかし同等の活性収率を与えた。96時間の時点で集めたデータは、還元MSB添加について活性の僅かな増加しか示さなかった。しかし、全ての還元MSB添加フラスコが、MSB添加と比較して等価の又はより高い比率を維持した。全体的にみて、増加した比率及び等価の又は増加した活性発現が、MSB添加と比較して還元MSB添加から得られた。
Thromb. Haemost. 2008 Apr;99(4):659−67に記載されるように、アルブミン融合第VII因子をクローニングし、CHO−S細胞中において発現させた。
Claims (22)
- 1つ又はそれ以上のビタミンK依存性タンパク質を発現する真核細胞の発酵方法であって、
i)ビタミンKの還元型及び/又は
ii)ビタミンKアナログの還元型及び/又は
iii)ビタミンK前駆体の還元型
を含むリストより選択される1つ又はそれ以上の化合物を、発酵プロセスの前及び/又は間に細胞培養培地に添加する、上記方法。 - ビタミンK依存性凝固因子が、FIX、FVII、FX、FII、プロテインC、プロテインS、プロテインZ、オステオカルシン、石灰化抑制マトリックスGlaタンパク質(MGP)又は細胞増殖調節増殖停止特異的遺伝子6タンパク質(Gas6)からなるリストより選択される、請求項1に記載の方法。
- ビタミンK依存性凝固因子がFIX又はFVIIである、請求項2に記載の方法。
- 還元ビタミンKアナログが還元型メナジオン重亜硫酸塩である、請求項1〜3のいずれか1項に記載の方法。
- 発現をバイオリアクター中で行う、請求項1〜4のいずれか1項に記載の方法。
- 発酵をドローフィル様式で操作する、請求項1〜5のいずれか1項に記載の方法。
- 発酵をバッチ様式で操作する、請求項1〜5のいずれか1項に記載の方法。
- 発酵を灌流様式で操作する、請求項1〜5のいずれか1項に記載の方法。
- 懸濁状態の細胞を使用して発酵を行う、請求項1〜8のいずれか1項に記載の方法。
- 接着細胞を用いて発酵を行う、請求項1〜8のいずれか1項に記載の方法。
- i)ビタミンKの還元型及び/又は
ii)ビタミンKアナログの還元型及び/又は
iii)ビタミンK前駆体の還元型
を含むリストより選択される1つ又はそれ以上の化合物の使用であって、該化合物を細胞培養培地に添加することによる細胞培養における1つ又はそれ以上の機能性ビタミンK依存性タンパク質の発現のための、上記使用。 - 同量の同一しかし非還元型のそれぞれのビタミンK及び/又はビタミンKアナログ及び/又はビタミンK前駆体をそれぞれの還元型と同一の様式で細胞培養培地に添加する場合に得られる同一の機能性ビタミンK依存性タンパク質の発現収率と比較して、それぞれの機能性ビタミンK依存性タンパク質の発現収率が増加する、請求項11に記載の使用。
- ビタミンK依存性タンパク質の抗原レベル発現に対する活性の比率が増加する、請求項11又は12に記載の使用。
- ビタミンK依存性タンパク質の抗原レベル発現に対する活性の比率が、少なくとも15%増加する、請求項11〜13のいずれか1項に記載の使用。
- ビタミンK依存性凝固因子が、FIX、FVII、FX、FII、プロテインC、プロテインS、プロテインZ、オステオカルシン、石灰化抑制マトリックスGlaタンパク質(MGP)又は細胞増殖調節増殖停止特異的遺伝子6タンパク質(Gas6)からなるリストより選択される、請求項11〜14のいずれか1項に記載の使用。
- ビタミンK依存性凝固因子がFIX又はFVIIである、請求項11〜15のいずれか1項に記載の使用。
- 還元型ビタミンKアナログが還元型メナジオン重亜硫酸塩である、請求項11〜16のいずれか1項に記載の使用。
- 発現をバイオリアクター中で行う、請求項11〜17のいずれか1項に記載の使用。
- 発酵をドロー−フィル様式で操作する、請求項11〜18のいずれか1項に記載の使用。
- ビタミンK及び/又はビタミンKアナログ及び/又はビタミンK前駆体の還元型で補足されたビタミンK依存性タンパク質の発酵用の培地。
- 還元型メナジオン重亜硫酸塩で補足された、請求項20に記載の培地。
- 発酵における細胞の生存能力を増強するための細胞培養培地中のビタミンK及び/又はビタミンKアナログ及び/又はビタミンK前駆体の還元型の使用。
