Description
本出願は、2009年3月23日出願の米国仮出願第61/162、619号の出願日遡及の特典を主張し、参照によりその全体を本明細書に包含する。
本発明は、複合糖質、例えば、複合糖質ワクチンに関する。
これらの研究のほとんどは翻訳後にグリコシル化されたタンパク質のプロセッシングと提示に関し検討している。これらのエピトープは、ペプチドに結合した単純な単糖類または二糖類であるように見える。MHC−Iは、単なるペプチド部位ではなく糖ペプチドのタイプのエピトープに結合することが明らかになっている。さらに、T細胞受容体(TCR)のプロセッシングされた糖ペプチドへの結合は、MHCに結合しているグリカンおよびペプチドの両方により形成されたエピトープとTCRの接触に依存することが示された。Haurum et al.、J Exp Med、180:739−744(1994)。例えば、Holmdahlとその共同研究者は、II型コラーゲン(CII)のT細胞認識において、エピトープグリコシレーションが重大な役割を果たすことを示した。Corthay et al.、Eur J Immunol、28:2580−2590(1998);Dzhambazov et al.、Eur J Immunol、35:357−366(2005);Michaelson et al.、Eur J Immunol、22:1819−1825(1992)。これらの研究者は、ヒトとラットの健常な関節軟骨で免疫優性T細胞エピトープがO‐グリコシル化されていることを見つけた。腫瘍抗原ムチン様糖タンパク質1(MUC1)由来の糖ペプチドエピトープの研究が特に重要である。HanishおよびNinkovic、同上。腫瘍細胞においては、タンパク質グリコシレーションによりCD4+T細胞により認識される腫瘍特異的糖ペプチドエピトープが形成される。Vlad et al.(2002)、同上;Werdelin et al.、Proc Natl Acad Sci USA、99:9611−9613(2002)。グリコシレーションパターンの変化は、T細胞のエピトープ認識を変化させることができる。HanishおよびNinkovic、同上。

(1)ペプチド単位部であって、
(i)MHC−II結合配列であって、50アミノ酸以下、例えば、40アミノ酸以下、30アミノ酸以下、20アミノ酸以下、または15アミノ酸以下、例えば、約8〜15アミノ酸の結合配列;
(ii)1つまたは複数のリジン残基であって、好ましくは、末端、例えば、C末端、に1つのリジンがあり、内部リジンが無い(一部の実施形態では、各ペプチドは単一のリジン残基をC末端に有する)リジン残基;および
任意選択として(iii)MHC−II結合配列とリジン残基を結合する1つまたは複数のアミノ酸;から構成されるペプチド単位部;および
(2)オリゴ糖であって、好ましくは、約5〜20kDa、例えば、約10〜15kDa、例えば、約10kDaまたは15kDaの平均分子量を有し、ペプチド単位に対するオリゴ糖の比率が約1:1となるようにリジン残基に直接結合し、糖化ペプチドがペプチド単位のC末端で切断可能はリンカー、例えば、酸不安定配列またはプロテアーゼ認識配列、例えば、カテプシンまたはカスパーゼ認識配列、を介して、生体適合性ナノ粒子に結合しているオリゴ糖、を含む。
1)炭水化物、切断可能リンカーにより結合された反復T細胞エピトープ、例えば、OVApを含むポリペプチド構築物に共有結合で結合した、例えば、約5〜20kDa、例えば、10〜15kDa、の炭水化物(B細胞受容体に効果的に架橋し、B細胞を活性化するために必要であれば、さらに小さいまたは大きい)。これは、多くの糖化ペプチド残基を生成する酸性のエンドソーム中のポリペプチドの解重合を可能にする。
2)炭水化物、切断可能リンカーにより結合された複数の異なるT細胞ペプチドエピトープを含むポリペプチド構築物に共有結合で結合した、例えば、約5〜20kDa、例えば、10〜15kDa、の炭水化物(B細胞受容体に効果的に架橋し、B細胞を活性化するために必要であれば、さらに小さくまたは大きくてもよい)。これには、より大きなT細胞レパートリーの動員ができるようにすべきである。
3)T細胞エピトープ、例えば、OVAextpep、の1つの末端に切断可能なリンカーで共有結合した、約5〜20kDa、例えば、10〜15kDaの炭水化物(MHC−II結合多糖のサイズ)を、例えば、含む複合糖質ワクチンの糖化ペプチド抗原エピトープでコートした生体高分子ベースのナノ粒子。
I:炭水化物成分:目的の病原体または腫瘍由来の多糖またはオリゴ糖、任意選択として、約5〜20kDa、例えば、10〜15kDaの平均分子量を有するように処理され、またはB細胞受容体に効果的に架橋し、B細胞を活性化するために必要に応じさらに小さくまたは大きく;さらに、
II:少なくとも1つのペプチド単位であって、MHC−II結合配列およびリジン、好ましくは1つの末端に1つのリジンを含み、このリジンに炭水化物が直接結合しているペプチド単位。
本明細書記載の組成物は、免疫応答が求められる任意の多糖、例えば、任意の病原体、例えば、ウイルス、細菌、真菌、または原虫類、から得られた多糖;糖タンパク質;または細胞、例えば、腫瘍細胞、を使って作ることができる。「多糖」は、通常、グリコシド結合により結合した単糖残基で構成される任意の直鎖または分岐高分子を意味し、従ってオリゴ糖を含む。目的の炭水化物を特定し、入手する方法は、当技術分野で既知である。例えば、Dormitzer et al.、Trends Biotechnol.26(12):659−667(2008)を参照。
別の実施形態では、天然のまたは完全な多糖が使用可能である。
通常、MHC−II結合配列は、T細胞エピトープ、例えば、CD4+Tヘルパー細胞エピトープ(Etlinger et al.、Science 249:423−425(1990))を含むペプチドである。T細胞ヘルプを誘導できる多くのCD4+エピトープは、当技術分野で既知である。例えば、Etlingeretal.、1990、同上;Valmori et al.、J.Immunol.149:717−721(1992);Saddetal.、Immunology 76:599−603(1992);Kumar et al.、J.Immunol.148:1499−1505(1992);Kaliyaperumal et al.、Eur J Immunol 25:3375−3380(1995);De Velascoetal.、Infect.Immun.63:961−968(1995);Falugietal.、Eur.J.Immunol.31:3816-3824(2001);およびBixer et al.、Adv.Exp.Med.Biol V:175−180(1989))を参照。
一部の実施形態では、MHC結合成分は、アミノ酸配列よりはむしろ、小分子、ポリマー、またはペプチド類似体である。この実施形態では、複合体ワクチンは、タンパク質またはペプチドでないキャリア分子と結合した細菌、ウイルス、寄生生物または腫瘍由来の多糖またはオリゴ糖を含んでもよい。当分野の技術であれば、MHC−II結合部位に合う適切な大きさの任意の分子を選択、使用し、静電気的な、疎水性の、ファンデルワールスの、または他の既知の2分子を相互に結合させる化学力によってその部位に結合することが可能であろう。1つの例が双性イオン分子、例えば、ポリサッカライドA(PSA)のような双性イオン炭水化物である。これらは、MHC−IIへの結合を可能にする双性イオン電荷または疎水性特性等の物理化学的特性を有する合成分子であってもよい。誘導された免疫応答の標的になるべき多糖またはオリゴ糖は、このキャリア分子に結合される。キャリアがペプチドに代わりアンカーとして作用してMHC−IIに結合し、本出願に記載された糖化ペプチドと類似の方式でT細胞受容体による標的多糖またはオリゴ糖の認識が可能となろう。これらの新規糖化キャリア分子は、ヘルパー機能および抗体産生に必要なT細胞応答を誘導することが可能となろう。
本明細書記載の糖化ペプチドに切断可能リンカーを介して結合したナノ粒子も、本発明に従って作ることができる。従って、本発明は、さらに生体適合性ナノ粒子に結合した糖化ペプチドを含む組成物を含み、任意選択でそのナノ粒子を経由してAPC、例えば、DCやB細胞を標的にする共存抗体を一緒に含んでもよい。MHC結合配列を含むペプチド単位がナノ粒子に結合している場合の実施形態では、各ペプチドとナノ粒子の間に切断可能成分が存在することになる。代表的ナノ粒子には、Xu et al.、Macromol.Biosci.7:968-974(2007)に記載されたものが含まれる。
ナノ粒子を調製可能な種々の方法があるが、いずれの方法でも、結果はナノ粒子を結合成分に結合するために使用可能な官能基を有したナノ粒子でなければならない。
本明細書記載の複合糖質で、ペプチドを、例えば、還元アミノ化を使って、共有結合で直接炭水化物に結合するのが好ましい。他の方法は当技術分野で既知である。例えば、Vliegenthart et al.、FEBS Letters 580:2945-2950(2006)、および引用文献を参照。化学的リンカー、すなわち、ヘキサン酸、アジピン酸、またはその他の二官能性カップリング分子、を使わないのが好ましい。例えば、Jones、 Anaisda Academia Brasileirade Ciencias 77(2):293−324(2005)を参照。
B群連鎖球菌(GBS)複合糖質:GBSは、新生児の重篤な細菌感染の主要原因である。Schuchat、Lancet、353:51−56(1999)。症例の約80%で、病原体の直接母子感染により新生児のGBS感染が起こり、この病原体は健全な女性の25〜40%の肛門生殖器粘膜にコロニー形成する(上記Schuchat(1999)論文で解説されている)。出産前に適切な抗生物質を全てのGBS陽性の女性に投与すべきである(GBS疾患発生率を劇的に減らす処置)とする疾病管理予防センターの勧告にも拘わらず、米国だけでも、GBSは、いまでも新生児の年間2500例の感染と100例の死亡の原因となっている。いずれにせよ、この抗生物質治療を提供するという政策は、抗生物質抵抗の恐れのために、長期的には継続できそうもない。従って、通常、有効なワクチン接種が長期にわたるGBS疾患の発生率を減らす唯一の手段であると考えられている。
本明細書記載の治療に有効な量の1つまたは複数の組成物(すなわち、有効な(治療用)試薬として、単独またはナノ粒子に結合して、本明細書記載の複合糖質を含む)は、患者、例えば、ヒト、への投与に適した医薬組成物に組み込むことができる。このような組成物は、通常、組成物および薬剤的に許容可能なキャリアを含む。本明細書で使われる用語の「薬剤的に許容可能なキャリア」は、医薬品の投与に適合する、任意かつ全ての溶媒、分散媒、被覆材、抗菌および抗真菌剤、等張吸収遅延剤、等、を含むことが意図されている。薬剤的活性物質のためのこのような媒質と試薬の使用は既知である。従来の媒質または試薬がその活性化合物と不適合である場合を除き、このような媒質は本発明の組成物に使用可能である。補充性活性化合物、例えば、リガンド分解抑制剤、もまた、組成物に組み入れることができる。
本明細書には、また、例えば、哺乳類動物、例えば、ヒトまたは非ヒト哺乳類動物の患者の免疫応答を誘導する方法も提供されている。本明細書記載の複合糖質は、例えば、ワクチンとしての使用のために、未変性多糖自体に対する免疫応答を誘導するのに特に有効である。従って、この方法には、免疫保護が必要な患者の特定、および患者の免疫応答を誘導するのに十分な量の本明細書記載の複合糖質の患者への投与が含まれる。一部の実施形態では、方法には、患者への1回または複数回の追加の投与、例えば、追加の接種が含まれる。

複合糖質がどのようにしてT細胞を活性化するかについて、現在受け入れられているパラダイムは、炭水化物が非常に強力な共有結合によりタンパク質に結合(多くの場合二級アミン)しているにしても、T細胞にペプチドが提示され、認識されるということだけである。この古典的仮説では、APC内の多糖に何が起きているか、またはエンドソーム中でどのようにして炭水化物とタンパク質を分解し、正しいペプチドをT細胞に提示するか、説明できない。しかし、特定のCPS−例えば、総腸管グラム陰性偏性嫌気性菌バクテロイデスフラジリスの多糖A(PSA)−は、Toll様受容体(TLR)を介して自然免疫系を活性化し、獲得免疫系と連携して自然免疫系がこれらの分子に応答して作用する。Wang et al.、J Exp Med、203:2853−2863(2006).PSAは、抗原提示細胞(APC)によりプロセッシングされ、主要組織適合性クラスII(MHCII)経路を介してCD4+T細胞に提示される。結果的にT細胞が活性化される。Cobb et al.、Cell、117:677−687(2004)。本発明者等は、糖化ペプチドは、正しくプロセッシングされておれば、提示専門細胞APCによりT細胞に提示されることが可能である、との仮説を立てた。この新しい概念は、複合体ワクチンおよび糖タンパク質含有ウイルスおよび細菌による感染に関し、古典的教示に反する。仮説が探究の価値があるか否かを判定するために、GBSIIIを使って実験を行った。このGBSIIIの反復ユニット構造をモデル多糖として図1に示す。GBSIIIは、古典的T細胞非依存的抗原であると考えられる。Guttormsen et al.、Infect Immun、67:6375−6384(1999)。マウスでは、キャリアタンパク質に共有結合で結合する場合のみに、GBSIIIを使った免疫化により有意な量のIgG抗体が誘導された(図1には、複合化に使われる代表的化学反応を示す)。
MHCII結合糖化ペプチドがAPC表面に提示されるか否かを判定するために、免疫共沈降(co−IP)、フローサイトメトリー、およびウェスタンブロット実験を行った。最初、PSAおよびN−アセチル化PSAのように、純粋なGBSIIIがAPCのエンドソーム内で解重合されるか否かを評価した。(Cobb et al.(2004)、同上を参照)、(Duan et al.、Proc.Natl.Acad.Sci.U.S.A.105(13):5183−5188(2008))にあるPSAに関する報告と同じ方法を使って、放射標識したGBSIIIをラージB細胞と共に18時間インキュベートした。次に、ミクロソームをラージ細胞から単離し、エンドソーム内のGBSIII分子の大きさ(大きさにばらつきがあり、平均で約15kDaであるが、カラムの総容積にいくつかの物質が含まれている)が元の分子(平均で150kDa;カラムの空隙容積に溶出)よりも有意に小さいことを確認した(図2a)。co−IP実験(図2a)の次に、PSAの場合の報告(Cobb et al.(2004)、同上を参照)と類似の実験を行った。表面免疫複合体をmAbとMHCIIの複合体と沈降させた。細胞を純粋な非複合多糖とインキュベートした場合、予想通り、MHCIIの存在下、GBS炭水化物は細胞表面に認められなかった。これらのデータにより、複合GBSIIIはエンドソーム内で解重合されるが、純粋な炭水化物はMHCII分子にロードされ得ず、従って細胞表面に提示できないことが明らかである。PSA−MHCII結合への化学反応のキーは双生イオン電荷モチーフであり、これは、非複合GBSIII(他の炭水化物と同様に)では欠けている。3Hデキストランは同様にエンドソーム内でプロセッシングされることがわかった。これらの知見は、多糖にT細胞非依存性を与える少なくとも1つの機序が、必ずしもAPCが多糖をMHC−IIにロードできる大きさに解重合できないことではなく、MHCIIに結合できないことであることを示唆している。
GBSIII−OVAのエンドソームプロセッシングの動力学に関する予備調査も行った。3時間、6時間、または18時間のGBSIII[3H]−OVAとのインキュベーション後、ラージ細胞のエンドソームを集めた(図2B)。結果は、プロセッシングは3時間で開始され、18時間までに完了することを示唆している。
この実施例は、特定のペプチド(OVAp)を認識するCD4+T細胞受容体(TCR)が、そのペプチドが莢膜多糖(GBSIII)と複合化された場合、同じペプチドを認識しなかったことを実証する実験について記載する。この実験は、糖化ペプチドエピトープの炭水化物部が特定のTCRにより特異的に認識されうるという仮説を裏付ける証拠を集めるため行った。GBSIII−OVApがOVAp特異的T細胞クローンならびにOVAp単独によるT細胞増殖を誘導しないことを示すことにより、III−OVApがペプチド特異的T細胞により認識されることができない糖化ペプチドエピトープを有するということが示唆されることになろう。
T細胞は、特定のMHCII結合炭水化物(ZPS)を認識し、この認識が生物学的に重要な結果をもたらす。MHCII結合に必要なZPSの特有で重要な化学特性は、双性イオン電荷モチーフであり、これにより静電的ZPSMHCII結合が可能となる。非複合化GBSIIIおよびほぼ全ての他の炭水化物は、双性イオン電荷モチーフが欠如しており、代わりに、その反復ユニット構造の一部として、負に帯電した基のみか、または帯電基が全くないかである。これらの知見は、多糖にT細胞非依存性を与える少なくとも1つの機序は、MHCIIに結合できないことであり、必ずしもCD4+T細胞のαβTCRが炭水化物を認識できないことではないことを示唆している。
次に、T細胞応答が1つのキャリアペプチドの方向にのみ向いている複合糖質を構築した。完全長タンパク質との特定の複合体は、複合後、さらに免疫原性の強いペプチドを提示し、単独の場合のタンパク質キャリアにより提示されたペプチドより免疫原性の強いペプチドに対する強化免疫応答が得られる可能性があった。単一のペプチドT細胞エピトープを含むGBSIII−OVApを抗原として使用した。OVAp特異的TCR遺伝子導入(DO11.10)マウスおよび野生型(BALB/c)マウスの両方から、各マウス株をGBSIII−OVApで2週おき3回免疫した後、リンパ細胞を採取した。T細胞増殖実験では、野生型(BALB/c)マウス由来照射APCを、免疫BALB/cマウス(図7A)または免疫DO11.10マウス(図7B)由来のCD4+T細胞と共に同時インキュベートした。iAPC/CD4+T−細胞混合物をインビトロで3つの抗原:GBSIII−OVAp(10μg);GBSIII(7.5μg、GBSIII−OVApの10−μg用量中のGBSIII含量に相当);およびOVAp(2.5μg、GBSIII−OVApの10−μg用量中のOVAp含量に相当)で刺激した。
多糖特異的抗体(IgG)の応答がペプチドエピトープの認識によるものか、または炭水化物エピトープの認識によるものなのかを判定するため、DO11.10OVAp特異的TCR遺伝子導入マウスおよびGBSIII−OVApで免疫した野生型BALB/cマウスを使って、GBSIII特異的IgG力価を測定した。DO11.10遺伝子導入マウスは、GBSIII特異的IgGを産生しなかったが、一方野生型マウスは、GBSIII特異的IgGの高い力価を示した(図8A)。さらに、DO11.10マウスおよび野生型マウスは、同等量のGBSIII特異的IgMを生じ、この結果は、DO11.10マウスにおけるIgG産生の欠如が、少ない数のGBSIII特異的B細胞によるものではないことを示している(Fig.8A)。これらの結果は、ヘルパーT細胞のTCRによるGBSIIIの認識がB細胞による多糖特異的IgG分泌を誘導するという仮説を裏付けるものである。DO11.10マウスは、複合糖質ワクチンの炭水化物エピトープを認識するT細胞レパートリーが欠如しているので、DO11.10T細胞は、B細胞の多糖特異的IgG分泌を誘導できなかった。野生型およびDO11.10マウスをOVAp(無多糖)で免疫した場合、DO11.10マウスが、より高い力価のOVAp特異的IgGを産生した(図8C)。後者の実験により、DO11.10マウスのIgG産生能力の欠如によって、GBSIII特異的抗体産生に失敗したことの説明はできないことを示した。
データは、PSAやGBSIII等の細菌性CPSが、エンドソーム中でより小さい分子にプロセッシングされうることを示唆している。これらの炭水化物は、分解された形でAPC表面上を行き交い、そこで検出されてMHCIIに結合するように思われる。既発表研究では、生物学的に重要な結果と共に、T細胞が特定の多糖(ZPS)に応答することが明確に示されている。Mazmanian et al.、Cell、122:107−118(2005);Ruiz−Perez et al.、PNAS、102:16753−16758(2005);Wang et al.、J Exp Med、203:2853−2863(2006)。次に、MHCIIにより提示された、ペプチドを介してMHCIIに結合した炭水化物(糖化ペプチド)にT細胞が応答するか否かを判定した。再度、仮説は、複合糖質または糖タンパク質をプロセッシングする場合、タンパク質由来ペプチドエピトープを提示するのと同様に、APCが糖化ペプチドをT細胞に対し提示するということであった。
マウスをIII−OVAp19(OVAp19は19アミノ酸を意味し、OVAの17aaTCR依存抗原エピトープ、OVAp、の各末端に付加した2つのリジン基を含む)で免疫した場合、GBSIII特異的IgGの分泌が観察されなかった。GBSIII−OVApがIgM−IgGスイッチを誘導できなかったことに対する推定される理由は、III−OVApの設計に関係のあることかもしれない。この構築物中のペプチドは、NおよびC末端の両方から多糖に共有結合で結合している。この両側の結合は、高度架橋複合体中にペプチドを捕捉し、エンドソーム中でMHCII分子によるペプチドの認識を最小化する可能性がある。(図10B)。GBSIII−OVA延長ペプチド複合体(III−OVAextpep)を設計し合成した(図10C)。ここで、OVAp(323−339)をC末端上で4アミノ酸で延長し、リジンをC末端残基とした。N末端のアミノ官能性は、N末端アミンのアセチル化によりブロックした。延長OVApを設計し、合成した:(N−アセチル)−ISQAVHAAHAEINEAGRESGK。このペプチドは、非常に均一な単鎖構築物をもたらすC末端リジン基を介してGBSIIIとのみ複合できる。また、延長部分(ESGK)は、ペプチドと糖質の間に空隙を生成し、MHCII分子のペプチドに対するアクセスを可能にする。この複合体は、標的特異的であるように設計され(タンパク質特異的免疫応答を誘発するタンパク質キャリアが無い)、また、この構築物は、高度に架橋したタンパク質複合体に比べ分子当たり1オーダー多い抗原エピトープを含む(タンパク質多糖複合体の場合の1〜2エピトープとは対照的に、多糖の10反復単位毎に1ペプチドがあり10〜15糖化ペプチドエピトープになる)。
糖化ペプチドの炭水化物部のCD4+T細胞による認識が、複合糖質に対する体液性免疫応答の誘導における主要因子であるか否かを判定するため、BALB/cマウスをGBSIII−OVAで初回刺激し、GBSIIIOVAまたはGBSIII−TTで追加免疫した。GBSIII特異的IgGの血清中レベルを測定した(図14)。3群のマウスを対照として使った:(1)GBSIII−TTで初回刺激と追加免疫したマウス、(2)GBSIIIで初回刺激し、GBSIII−TTで追加免疫したマウス、および(3)GBSIII−OVAで初回刺激し、GBSIIIで追加免疫したマウス。CD4+ヘルパーT細胞によるペプチド認識が、B細胞による多糖特異的IgG分泌を誘発することが仮定されている。従って、GBSIII−OVAで初期刺激しGBSIII−TTで追加免疫したマウスに比べ、GBSIII−OVAで初回刺激と追加免疫したマウスGBSIIIに対する血清IgGの方がより高い力価をしめすことが期待されよう。すなわち、初回刺激キャリアタンパク質からのペプチドに対するT細胞の記憶のために、同じキャリアタンパク質による初回刺激と追加免疫の方が異種のキャリアによる追加免疫よりも有意に多くのT細胞ヘルプを誘発すると推定できるであろう。あるいは、糖化ペプチドの炭水化物部に対するT細胞記憶の状態で、GBSIII特異的IgG力価は、複合体中の同じキャリアタンパク質による追加免疫に依存しないであろう。図14Aに示すように、GBSIII−OVA初期刺激マウスのGBSIII−TTによる追加免疫は、GBSIII−OVAによる初回刺激と追加免疫と類似のGBSIII特異的IgG分泌を誘導した(p>0.05、ns)。この結果は、炭水化物特異的免疫応答を誘導するT細胞ヘルプは、ペプチド認識ではなく、炭水化物認識を経由して取り込まれることを示している。初回刺激後のGBSIII特異的IgG力価を図14Bに示す。
これらの実験では、MHC結合成分は双性イオン炭水化物、すなわち、多糖A(PSA)である。PSAは、双性イオン電荷を有し、MHCIIに結合する。誘導された免疫応答の標的になる多糖またはオリゴ糖がこのキャリア分子に結合する。PSAキャリアはペプチドに代わりMHCIIに結合するためのアンカーの役割をし、標的多糖またはオリゴ糖は、本出願に記載の糖化ペプチドの場合と類似の方法により、T細胞受容体による認識に使用可能となる。これらの糖化キャリア分子は、ヘルパー機能と抗体産生に必要なT細胞応答を誘導する。
図15Bに示すように、これらの構築物は、非常に高いT細胞増殖を誘導した。
本発明をその詳細な説明と合わせて記載したが、これまでの記述は説明を意図しているもので、本発明の範囲を限定するものではなく、添付の請求項により規定されることは理解されるべきである。他の態様、特徴、および変更は、以下の請求項の範囲に含まれる。
Claims (24)
- 糖化ペプチド複合体を調製する方法であって:
多糖の集団を取得するステップ;
任意選択で、多糖の集団を処理して約5〜20kDaの平均分子量を有するオリゴ糖の集団を作るステップ;
オリゴ糖の集団を、切断可能成分を使って相互に結合された複数の反復ペプチド単位を含むポリペプチドと接触させるステップであって、各ペプチド単位が:
(i)MHC-II結合配列;
(ii)1つまたは複数のリジン残基;および任意選択として、
(iii)MHC-II結合配列とリジン残基を結合する1つまたは複数のアミノ酸;から構成されるステップ;
オリゴ糖を直接リジン残基に結合させるために十分な条件の下、
それにより糖化ペプチド複合体を調製するステップ;を含む方法。 - 糖化ペプチド/ナノ粒子複合体を調製する方法であって:
多糖の集団を取得するステップ;
任意選択で、多糖の集団を処理して約5〜20kDaの平均分子量を有するオリゴ糖の集団を作るステップ;
オリゴ糖の集団を、ペプチド単位の集団と接触させるステップであって、各ペプチド単位が:
(i)MHC-II結合配列;
(ii)1つまたは複数のリジン残基;および任意選択として、
